Fenbendazole is an antiparasitic medication used for human pinworms, giardia, roundworms, hookworms, and Taenia solium. In the past, it was also a veterinary medication used to treat animal parasites. Recently, it has been repurposed as a chemotherapy agent to treat cancer and other conditions like fibromyalgia. It is used to inhibit the polymerization of tubulin, which forms microtubules that support cytoskeleton structures and give cells shape. It has been shown to have cytotoxic effects on tumors and suppress the expression of RAS-related signaling pathways [1,3].

We tested the effects and mechanisms of fenbendazole in 5-fluorouracil-resistant colorectal cancer (CRC) cells. Compared to albendazole, fenbendazole showed more potent toxicity to 5-FU-resistant SNU-C5 cells and induced apoptosis and cell cycle arrest in both wild-type and resistant cells. It also activated p53 in the resistant cells and caused mitochondrial injury and caspase-3-PARP activation to trigger apoptosis.
