Finger was measured by the distance from the proximal crease to the tip by using a digital caliper. The mean age of patients with BPE and non-BPE was 62 ± 8.9 and 61.5 ± 7.1 years respectively. There was statistically significant difference between groups in terms of prostate-specific antigen levels, prostate volumes and international prostate symptom scores. Determining the retention time of insect cuticles is important for understanding the digestive physiology of pangolins, while determining the recovery rate of termites and ants in feces is required to estimate the number of these prey items that are consumed by pangolins. In this study, the authors conducted feeding trials with captive Chinese pangolins (Manis pentadactyla). Sixty grams of the fungus-growing termite Odontotermes formosanus (18,816 individuals) and 15-20 g of the yellow crazy ant Anoplolepis gracilipes (14,400-19,200 individuals) were fed to each pangolin. After feeding, pangolin feces were collected daily for 1 week. The authors also assessed the accuracy of using chromium (III) oxide powder (Cr2 O3 ) as a proxy for determining gut passage time, as has been done in previous studies. The results showed that remaining termite mandibles and ant head capsules in feces peaked at 66 and 90 hr after feeding and their recovery rates were 0.35 ± 0.10 and 0.65 ± 0.04, respectively. In both feeding trials, the retention time of Cr2 O3 was **** shorter than that of the termite mandibles and ant head capsules, indicating that Cr2 O3 is not an appropriate indicator for estimating food retention time of myrmecophagous animals.   • Purpose To examine the effect of dynamic changes in neutrophil-to-lymphocyte ratio (NLR) on tumor response and overall survival (OS) in patients with hepatocellular carcinoma (HCC) undergoing transarterial chemoembolization (TACE). Patients and methods Data from 181 patients with HCC were retrospectively collected. White blood cell, neutrophil and lymphocyte counts, and the NLR were obtained 1-3 days before as well as 3-6 weeks and 3 months after TACE. Patients were divided into two groups at each time point according to the mean value of NLR, and also divided into continuous decrease, fluctuating increase-decrease (I-D), fluctuating decrease-increase (D-I), and continuous increase groups according to the dynamic changes in the NLR. https://www.selleckchem.com/products/bix-01294.html The dynamic changes in blood counts and NLR were analyzed using repeated-measures ANOVA. However, limited studies report metastasis-associated lncRNAs in OS. Our present study aimed to explore the roles of lncRNA LINC00514 (LINC00514) in OS. Materials and methods The LINC00514 expression was measured using qPCR assays in OS tissues and cell lines. The clinical significance of LINC00514 expression in OS patients was analyzed using chi-square test, Kaplan-Meier assays and multivariate analysis. The possible effects of LINC00514 in tumor cellular progression were determined using a series of functional assays. The mechanisms of LINC00514 action were explored through bioinformatics, luciferase reporter assays and RT-PCR assays. The mechanisms involved the upregulation of LINC00514 expression in OS were determined using luciferase reporter and chromatin immunoprecipitation (ChIP) assays. Results We showed that LINC00514 expressions were distinctly upregulated in both OS tissues and cell lines, especially in advanced cases. High levels of LINC0051 were positively correlated with advanced tumor stages, distant metastasis, and reduced survival of patients with OS. Functional experiments indicated that silencing of LINC00514 suppressed the ability of cell growth, colony formation and metastasis, whereas promoted cell apoptosis in vitro. Mechanistic investigation revealed that LINC00514 could directly bind to miR-708 and effectively serve as a ceRNA for miR-708. In addition, LINC00514 was upregulated by the transcription factor SP1. Conclusion Our findings revealed SP1-induced upregulation of LINC00514 as an oncogene in OS through competitively binding to miR-708, suggesting that there are potential diagnostic and treatment values of LINC00514 in OS.Purpose The number of non-responders to treatment among patients with chronic pain (CP) is high, although intensive multimodal treatment is broadly accessible. Patients and methods A latent class analysis (LCA) was carried out in retrospective data from 411 patients with CP of different origins. All patients experienced severe physical and psychosocial consequences and were therefore undergoing multimodal inpatient pain treatment. Self-reported measures of pain (visual analogue scales for pain intensity, frequency, and impairment; Pain Perception Scale), emotional distress (Patient Health Questionnaire, PHQ-9; Generalized Anxiety Disorder Scale, GAD-7) and physical health (Short Form Health Survey; SF-8) were collected immediately after admission and before discharge. Instruments assessed at admission were used as input to the LCA. Resulting classes were compared in terms of patient characteristics and treatment outcome. Results A model with four latent classes demonstrated the best model fit and interpretability. Classes 1 to 4 included patients with high (54.7%), extreme (17.0%), moderate (15.6%), and low (12.7%) pain burden, respectively. Patients in class 4 showed high levels of emotional distress, whereas emotional distress in the other classes corresponded to the levels of pain burden. While pain as well as physical and mental health improved in class 1, only the levels of depression and anxiety improved in patients in the other groups during multimodal treatment. Conclusion The specific needs of these subgroups should be taken into account when developing individualized treatment programs. However, the retrospective design limits the significance of the results and replication in prospective studies is desirable.The continued prevalence of chronic low **** pain (CLBP) is a testament to our lack of understanding of the potential causes, leading to significant treatment challenges. CLBP is the leading cause of years lived with disability and the fifth leading cause of disability-adjusted life-years. No single non-pharmacologic, pharmacologic, or interventional therapy has proven effective as treatment for the majority of patients with CLBP. Although non-pharmacologic therapies are generally helpful, they are often ineffective as monotherapy and many patients lack adequate access to these treatments. Noninvasive treatment measures supported by evidence include physical and chiropractic therapy, yoga, acupuncture, and non-opioid and opioid pharmacologic therapy; data suggest a moderate benefit, at most, for any of these therapies. Until our understanding of the pathophysiology and treatment of CLBP advances, clinicians must continue to utilize rational multimodal treatment protocols. Recent Centers for Disease Control and Prevention guidelines for opioid prescribing recommend that opioids not be utilized as first-line therapy and to limit the doses when possible for fear of bothersome or dangerous adverse effects.

  • It may reflect possible molecular mechanisms of bioluminescence as well as features of bacterial composition. The obtained data expands the existing body of knowledge about the bioluminescence spread among the bacteria of Nordic latitudes and provides complex information that is crucial for their precise identification.We studied the luminescence properties of indole in poly (vinyl alcohol) (PVA) film. The indole molecules are effectively immobilized in this polymer film and display both fluorescence and phosphorescence emission at room temperature. We noticed that the phosphorescence of indole in PVA film can be effectively excited at a longer wavelength than its typical singlet to triplet population route involving intersystem crossing. The maximum of the phosphorescence excitation is about 410 nm which corresponds to the energy of indole's triplet state. Interestingly, the phosphorescence anisotropy excited with the longer wavelength (405 nm) is positive and reaches a value of about 0.25 in contrast to the phosphorescence anisotropy excited within the indole singlet absorption spectrum (290 nm), which is negative. Very different temperature dependences have been observed for fluorescence and phosphorescence of indole in PVA film. While fluorescence depends minimally, the phosphorescence decreases with temperature dramatically. The fluorescence lifetime was measured to be a single component 4.78 ns while the intensity weighted average phosphorescence lifetime with 290 nm and 405 nm excitations were 6.57 and 5.62 ms, respectively. The purpose of this study is to synthesise and evaluate the learning experiences that first-year nursing students had following geriatric patient simulation and practice of clinical patient handover. Qualitative content analysis of survey comments from first-year students (n = 216) at a large university in Norway were performed. Simulation training included systematic patient observation of scenarios based on genuine geriatric cases in nursing homes and practice of clinical patient handover. Content analyses identified four generic categories (1) 'embodying theoretical knowledge'; (2) 'increased awareness about one's self'; (3) 'understanding that collaboration is needed'; (4) 'preparing for future work life'. These themes provide evidence for students integrating geriatric theoretical knowledge with clinical skills as a result of simulation. Analysis of learning experiences shows that geriatric scenario simulation and practice of clinical patient handover are valuable instruments before entering clinical training with geriatric patients, even for novice students. The obtained data expands the existing body of knowledge about the bioluminescence spread among the bacteria of Nordic latitudes and provides complex information that is crucial for their precise identification.We studied the luminescence properties of indole in poly (vinyl alcohol) (PVA) film. The indole molecules are effectively immobilized in this polymer film and display both fluorescence and phosphorescence emission at room temperature. We noticed that the phosphorescence of indole in PVA film can be effectively excited at a longer wavelength than its typical singlet to triplet population route involving intersystem crossing. The maximum of the phosphorescence excitation is about 410 nm which corresponds to the energy of indole's triplet state. Interestingly, the phosphorescence anisotropy excited with the longer wavelength (405 nm) is positive and reaches a value of about 0.25 in contrast to the phosphorescence anisotropy excited within the indole singlet absorption spectrum (290 nm), which is negative. Very different temperature dependences have been observed for fluorescence and phosphorescence of indole in PVA film. While fluorescence depends minimally, the phosphorescence decreases with temperature dramatically. The fluorescence lifetime was measured to be a single component 4.78 ns while the intensity weighted average phosphorescence lifetime with 290 nm and 405 nm excitations were 6.57 and 5.62 ms, respectively. The purpose of this study is to synthesise and evaluate the learning experiences that first-year nursing students had following geriatric patient simulation and practice of clinical patient handover. Qualitative content analysis of survey comments from first-year students (n = 216) at a large university in Norway were performed. Simulation training included systematic patient observation of scenarios based on genuine geriatric cases in nursing homes and practice of clinical patient handover. Content analyses identified four generic categories (1) 'embodying theoretical knowledge'; (2) 'increased awareness about one's self'; (3) 'understanding that collaboration is needed'; (4) 'preparing for future work life'. These themes provide evidence for students integrating geriatric theoretical knowledge with clinical skills as a result of simulation. Analysis of learning experiences shows that geriatric scenario simulation and practice of clinical patient handover are valuable instruments before entering clinical training with geriatric patients, even for novice students. Acquisition of magnetic resonance imaging (MRI) data for quantitative susceptibility mapping, R1 , R2 relaxometry and diffusion imaging was performed at 7 Tesla. Results of volume of interest (VOI)-based analyses of the quantitative data were compared with a voxel-based morphometry (VBM) evaluation. Differences between patients and controls were assessed using the analysis of covariance (ANCOVA; p less then 0.01) with age and sex as covariates, effect size of group differences, and correlations with disease characteristics with Spearman correlation coefficient. For the VBM analysis, a statistical threshold of 0.001 for uncorrected and 0.05 for corrected p-values was used. Statistically significant differences between FRDA patients and controls were found in five out of twelve investigated structures, and statistically significant correlations with disease characteristics were revealed. Moreover, VBM revealed significant white matter atrophy within regions of the brainstem, and the cerebellum. These regions overlapped partially with brain regions for which significant differences between healthy controls and patients were found in the VOI-based quantitative MRI evaluation. It was shown that two independent analyses provided overlapping results. This study aimed to develop and validate deep learning models that automatically classify colorectal lesions histologically on white-light colonoscopy images. Methods White-light colonoscopy images of colorectal lesions exhibiting pathological results were collected and classified into seven categories stages T1-4 colorectal cancer (CRC), high-grade dysplasia (HGD), tubular adenoma (TA), and non-neoplasms. The images were then re-classified into four categories including advanced CRC, early CRC/HGD, TA, and non-neoplasms. Two convolutional neural network models were trained, and the performances were evaluated in an internal test dataset and an external validation dataset. Results In total, 3828 images were collected from 1339 patients. The mean accuracies of ResNet-152 model for the seven-category and four-category classification were 60.2% and 67.3% in the internal test dataset, and 74.7% and 79.2% in the external validation dataset, respectively, including 240 images. In the external validation, ResNet-152 outperformed two endoscopists for four-category classification, and showed a higher mean area under the curve (AUC) for detecting TA+ lesions (0.818) compared to the worst-performing endoscopist. The mean AUC for detecting HGD+ lesions reached 0.876 by Inception-ResNet-v2. Conclusions A deep learning model presented promising performance in classifying colorectal lesions on white-light colonoscopy images; this model could help endoscopists build optimal treatment strategies.Human perinatal stem cells (SCs) can be isolated from fetal annexes without ethical or safety limitations. They are generally considered multipotent; nevertheless, their biological characteristics are still not fully understood. The aim of this study was to investigate the pluripotency potential of human perinatal SCs as compared to human induced pluripotent stem cells (hiPSCs). Despite the low expression of the pluripotent factors NANOG, OCT4, SOX2, and C-KIT in perinatal SC, we observed minor differences in the promoters DNA-methylation profile of these genes with respect to hiPSCs; we also demonstrated that in perinatal SCs miR-145-5p had an inverse trend in comparison to these stemness markers, suggesting that NANOG, OCT4, and SOX2 were regulated at the post-transcriptional level. The reduced expression of stemness markers was also associated with shorter telomere lengths and shift of the oxidative metabolism between hiPSCs and fetal annex-derived cells. Conclusions A deep learning model presented promising performance in classifying colorectal lesions on white-light colonoscopy images; this model could help endoscopists build optimal treatment strategies.Human perinatal stem cells (SCs) can be isolated from fetal annexes without ethical or safety limitations. They are generally considered multipotent; nevertheless, their biological characteristics are still not fully understood. The aim of this study was to investigate the pluripotency potential of human perinatal SCs as compared to human induced pluripotent stem cells (hiPSCs). Despite the low expression of the pluripotent factors NANOG, OCT4, SOX2, and C-KIT in perinatal SC, we observed minor differences in the promoters DNA-methylation profile of these genes with respect to hiPSCs; we also demonstrated that in perinatal SCs miR-145-5p had an inverse trend in comparison to these stemness markers, suggesting that NANOG, OCT4, and SOX2 were regulated at the post-transcriptional level. The reduced expression of stemness markers was also associated with shorter telomere lengths and shift of the oxidative metabolism between hiPSCs and fetal annex-derived cells. Our findings indicate the differentiation ability of perinatal SCs might not be restricted to the mesenchymal lineage due to an epigenetic barrier, but other regulatory mechanisms such as telomere shortening or metabolic changes might impair their differentiation potential and challenge their clinical application.In this paper, we present a new approach to the fusion of Sentinel 1 (S1) and Sentinel 2 (S2) data for land cover mapping. The proposed solution aims at improving methods based on Sentinel 2 data, that are unusable in case of cloud cover. This goal is achieved by using S1 data to generate S2-like segmentation maps to be used to integrate S2 acquisitions forbidden by cloud cover. In particular, we propose for the first time in remote sensing a multi-temporal W-Net approach for the segmentation of Interferometric Wide swath mode (IW) Sentinel-1 data collected along ascending/descending orbit to discriminate rice, water, and bare soil. Using a descriptive design, a study was conducted at seven malls located in the Eastern Province of Saudi Arabia. Two hundred and fifty participants were surveyed using the American Heart Association (AHA) 2015 guidelines to assess CPR and AED knowledge and availability in Saudi malls. The sample mean age was 32.60 years (SD = 10.02), and 87% of participants were working as security personnel. The majority of the participants had not received training about CPR and AED (75.8% and 95.2%, respectively). Common misconceptions are fallen into all categories of CPR and AED knowledge. Correctly answered statements ranged from 7.2% in the compression rate to 24.2% in hand placement. The study results indicated a poor training knowledge of CPR and AED in public settings. Integrating high-quality CPR and AED knowledge within the school and college curricula is a vital need. Besides, large quantities remain unused or expired since not all medications given to the consumers are consumed. Hence, this study could serve as an indicator for the country policy makers concerning pharmaceutical waste management. Objective To assess knowledge, attitude, and disposal practice of unused and expired pharmaceuticals in the community of Adigrat city, Tigray, Ethiopia, 2019. Methodology. A cross-sectional study was conducted among 359 respondents from the residents of Adigrat city. Semistructured questionnaires, whichposal of unused and expired medications, almost half of the sample population were unaware of proper disposal practices. Furthermore, less were inclined to practice proper disposal of unused and expired medications in the city. Therefore, we recommend further studies that focus on how the disposal attitude of the population can influence their knowledge and practice of the disposal of unused and expired medications. Methods The study included the results of a survey of 415 children aged 11-15 years from public schools in Ukraine. We have adapted the standardized questionnaire QAPACE, which allows characterizing the level of adolescent's PA by indicators type, duration, and intensity. Results A sedentary lifestyle is typical for most Ukrainian urban adolescents. Only 30.8% of Ukrainian urban adolescents (45.4% of boys and 21.4% of girls; p less then 0.001) meet the recommended level of moderate-to-vigorous PA (at least 60 minutes per day). The chances to follow the recommended moderate-to-vigorous PA are almost 5 times higher in the group of adolescents exercising at their own wish and not by parents' direction (OR = 4.96; 95% CI 2.77-8.90; p less then 0.001). Girls have a 3.0 higher chance of not adhering to recommendations for duration of PA (OR = 3.01; 95% CI 1.95-4.63). They are 2.5 times more likely to lead a sedentary lifestyle (OR = 2.48; 95% CI 1.54-3.98) than boys. This study assessed the population weighting effects on the estimated PM10 concentrations in Malaysia for years 2000, 2008, and 2013. Estimated PM10 annual mean concentrations with a spatial resolution of 5 kilometres retrieved from satellite data and population count obtained from the Gridded Population of the World version 4 (GPWv4) from the Centre for International Earth Science Information Network (CIESIN) were overlaid to generate the PWEL of PM10 for each state. The calculated PWEL of PM10 concentrations were then classified based on the World Health Organization (WHO) and the national Air Quality Guidelines (AQG) and interim targets (IT) for comparison.

  • The traditional method of retrieving atmospheric ducts is to use the special sensor of weather balloons or rocket soundings to obtain information intelligently, and it is very expensive. Today, with the development of technology, it is very convenient to retrieve the atmospheric ducts from Global Navigation Satellite System (GNSS) phase delay and propagation loss observation data, and then the GNSS receiver on the ground forms an automatic receiving sensor. This paper proposes a hybrid decomposition-based multi-objective evolutionary algorithm with adaptive neighborhood sizes (EN-MOEA/ACD-NS), which dynamically imposes some constraints on the objectives. The decomposition-based multi-objective evolutionary algorithm (MOEA/D) updates the solutions through neighboring objectives, the number of which affects the quality of the optimal solution. Properly constraining the optimization objectives can effectively balance the diversity and convergence of the population. The experimental results from the Congress on Evolutionary Computation (CEC) 2009 on test instances with hypervolume (HV), inverted generational distance (IGD), and average Hausdorff distance ∆2 metrics show that the new method performs similarly to the evolutionary algorithm MOEA/ACD-NS, which considers only the dynamic change of the neighborhood sizes. The improved algorithm is applied to the practical problem of jointly retrieving atmospheric ducts with GNSS signals, and its performance further demonstrates its feasibility and practicability.It is essential to develop a simple and sensitive method to rapidly detect residual fungicides in agricultural products to protect human health. So far, little studies have been reported on potential application of gold nanospheres (AuNSps) as a surface plasmon resonance based sensor for in-situ detection of residual fungicides. Therefore, in this study, we investigated the potential application of AuNSps as a surface plasmon resonance based sensor for in-situ detection of fungicides. AuNSps were successfully synthesized via a seed-mediated method with some modifications. Firstly, gold nanoseeds were made during the reduction of chloroauric acid by trisodium citrate dihydrate (TSC). Then, AuNSps were grown from the seeds by using HAuCl4, TSC and EDTA. AuNSps were subsequently dropped on a glass substrate before covered by thiophanate methyl, a broad-spectrum systemic fungicide. The AuNSps coated glass substrate was subsequently dried in the air for further surface-enhanced Raman spectroscopy (SERS) measurements. Optical properties, shape and size of AuNSps were confirmed by UV-vis spectroscopy, XRD, SEM-EDX and TEM. The results showed that AuNSps were successfully synthesized with the size of 53 nm, and their resonance peak was located at 560 nm. The Raman signal intensity of thiophanate methyl covered on AuNSps is higher than that without AuNSps, indicating SERS effects of AuNSps deposited glass substrate.Partially-stabilized zirconia is used in ceramic crowns due to its excellent mechanical properties and bio-inertness but does not match the natural color and translucency of tooth enamel. To reduce scattering of light and improve translucency, the grain size of zirconia ceramics should be less than the wavelength of visible light (0.4-0.7 μm), and porosity should be eliminated. The aim of the present work was to study the effect of two-step sintering of a commercial powder (Zpex Smile, Tosoh Corp., Tokyo, Japan) on the grain size and translucency of zirconia for use in ceramic crowns. Samples were sintered at a first step temperature (T1) of 1300, 1375 and 1400 °C for 5 min, followed by a decrease to the second step temperature (T2) and holding at T2 for 5-20 h. Samples were also conventionally sintered at 1450 °C for 2 h for comparison. Two-step sintered samples with an almost equal density, smaller grain size and narrower grain size distribution compared to conventionally sintered samples could be sintered. However, the translucency of two-step sintered samples had lower values compared to conventionally sintered samples. This is due to the slightly higher porosity in the two-step sintered samples. The aim of the present work was to study the effect of two-step sintering of a commercial powder (Zpex Smile, Tosoh Corp., Tokyo, Japan) on the grain size and translucency of zirconia for use in ceramic crowns. Samples were sintered at a first step temperature (T1) of 1300, 1375 and 1400 °C for 5 min, followed by a decrease to the second step temperature (T2) and holding at T2 for 5-20 h. Samples were also conventionally sintered at 1450 °C for 2 h for comparison. Two-step sintered samples with an almost equal density, smaller grain size and narrower grain size distribution compared to conventionally sintered samples could be sintered. However, the translucency of two-step sintered samples had lower values compared to conventionally sintered samples. This is due to the slightly higher porosity in the two-step sintered samples. Density and translucency of both conventionally and two-step sintered samples could be increased further by using a ball milled powder.Within Australia, vitamins, minerals, nutritional supplements, essential oils, and homoeopathic and herbal preparations are collectively termed and regulated as Complementary Medicines (CMs) by the Australian Therapeutic Goods Administration (TGA). CMs are predominantly self-selected through a pharmacy, providing pharmacy personnel an opportunity to engage with the public about their CM use. CMs are currently non-scheduled products in Australia. This review aimed to summarize the literature reporting the potential effect on pharmacies if scheduling of CMs was adopted, using codeine as an example. Seven studies were included for analysis. The majority of the literature has explored qualitative studies on the perception and opinion of pharmacists in relation to the up-scheduling of codeine. If CMs were to be up-scheduled, the accessibility of CMs would be limited to the pharmacy providing a role for pharmacy personnel, including both pharmacists and pharmacy technicians, to counsel on CM use. However, careful collaboration and consideration on how such a regulatory change would impact other key-stakeholders, including CM practitioners, requires both a strategic and collaborative approach. Oxidative stress is an imbalance between production and accumulation of oxygen reactive species and/or reactive nitrogen species in cells and tissues, and the capacity of detoxifying these products, using enzymatic and non-enzymatic components, such as glutathione. Oxidative stress plays roles in several pathological processes in the nervous system, such as neurotoxicity, neuroinflammation, ischemic stroke, and neurodegeneration. The concepts of oxidative stress and rare diseases were formulated in the eighties, and since then, the link between them has not stopped growing. The present review aims to expand knowledge in the pathological processes associated with oxidative stress underlying some groups of rare diseases Friedreich's ataxia, diseases with neurodegeneration with brain iron accumulation, Charcot-Marie-Tooth as an example of rare neuromuscular disorders, inherited retinal dystrophies, progressive myoclonus epilepsies, and pediatric drug-resistant epilepsies. Despite the discrimination between cause and effect may not be easy on many occasions, all these conditions are Mendelian rare diseases that share oxidative stress as a common factor, and this may represent a potential target for therapies.

METHODOLOGY A cross-sectional retrospective study was performed. DATA ANALYSIS correlation and lagged cross correlation between time series of Oscillation Niño Index and time series of standardized number of leptospirosis cases; construction of annual cycle of leptospirosis; comparison of changes of number of cases between Neutral, periods with El Niño and Neutral periods with La Niña. RESULTS At the national level, monthly number of cases raised a 25% during La Niña and decreased of 17% during El Niño. At departmental level, increase of cases in both phases of ENSO, depending on the location in the country, was found. At the municipal level, 17 have a rise in the number of cases during La Niña months. Of those, seven presented also an increase of cases during El Niño months and eight have a significant negative correlation with ONI. CONCLUSIONS In Colombia, there exists a relationship between leptospirosis and the excess and lack of rainfall related with ENSO. The contrasting results from each spatial scale, reinforce that leptospirosis is a multidimensional disease with high complex interactions among its determinants. MPB64 assay was used to screen NTM strains and confirmed by Rv0577 amplification. The species were identified by hsp65 sequencing. The clinical records of patients with NTM were retrospectively reviewed. RESULTS Among the 122 identified NTM isolates, the most common strains were Mycobacterium avium complex (MAC, n = 102, 83.6%), Mycobacterium abscessus (n = 9, 7.4%) and Mycobacterium lentiflavum (n = 5, 4.1%). The predominant species among ****were Mycobacterium chimaera (n = 57, 46.7%), followed by Mycobacterium intracellulare (n = 25, 20.5%) and Mycobacterium colombiense (n = 17, 13.9%). A significantly lower percentage of positive acid-fast assay was observed in Mycobacterium colombiense positive patients than in those with Mycobacterium intracellulare and Mycobacterium chimaera. Mycobacterium intracellulare was more frequently isolated in patients from the infectious department than in other ****members. CONCLUSIONS A predominant prevalence of Mycobacterium chimaera in Dongyang of Zhejiang Province was different from other regions in China, indicating that its prevalence has been likely underestimated. The heterogeneity in clinical features, caused by different ****members, required an accurate species identification of the NTM isolated in the primary hospitals. METHODOLOGY Eighteen stool specimens from 11 diarrheal patients and 3 close contacts were collected and tested for 16 enteric bacterial and viral pathogens using real-time PCR methods. Multilocus Sequence Typing (MLST), pulsed-field gel electrophoresis (PFGE) and antimicrobial susceptibility testing were applied to C. jejuni isolates in this outbreak in order to characterize the incident. RESULTS Ten (90.9%) of 11 stool specimens from diarrheal patients were positive for Campylobacter jejuni, confirmed as the cause of outbreak. Eight C. jejuni strains were obtained and produced 2 sequence types (STs) and 3 PFGE patterns. Six of them had the same ST (ST2274) and PFGE pattern (SMA001).

  • Severe acute respiratory syndrome coronavirus-2 (SARS-CoV-2) has been identified as the causal agent of COronaVIrus Disease-19 (COVID-19), an atypical pneumonia-like syndrome that emerged in December 2019. While SARS-CoV-2 titers can be measured by detection of viral nucleic acid, this method is unable to quantitate infectious virions. Measurement of infectious SARS-CoV-2 can be achieved by tissue culture infectious dose-50 (TCID50 ), which detects the presence or absence of cytopathic effect in cells infected with serial dilutions of a virus specimen. However, this method only provides a qualitative infectious virus titer. Plaque assays are a quantitative method of measuring infectious SARS-CoV-2 by quantifying the plaques formed in cell culture upon infection with serial dilutions of a virus specimen. As such, plaque assays remain the gold standard in quantifying concentrations of replication-competent lytic virions. Here, we describe two detailed plaque assay protocols to quantify infectious SARS-CoV-2 using different overlay and staining methods. Each step involves an increasing complexity and should serve a more sophisticated material selection, which serves as an orientation to clinical studies and the final application in patients. Recently, the usefulness of degradation analyses is being discussed. In this context, the aim of this work is to increase the importance of in vitro degradation analysis by using flowing media to move closer to the in vivo situation. In the long term, this should lead to a more sensitive biomaterial characterization as well as to a replacement of time-consuming static or quasi-dynamic incubation experiments. The practicability of the novel chamber is demonstrated in context of a degradation study of silica/collagen/calcium phosphate composites in flowing media with physiological (2.4 mM) and lowered (0.5 mM) calcium ion concentrations. This is done by comparison with static and quasi-dynamic incubation experiments. In order to keep all media regimes comparable to each other, for the dynamic experiment, a flow rate was chosen equivalent to the medium exchange in quasi-dynamic incubation. Under flow-through conditions, there is a clearly decreased tendency to lower the calcium concentration, so that a concentration close to the physiological initial situation can be continuously maintained.Backgroud Associations between sex hormones and diabetic vascular complications was recently studied, but the role luteinizing hormone (LH), playing in diabetic kidney disease (DKD) remains uncertain. We aimed to investigate the relationship of LH and DKD in Chinese men and postmenopausal women with type 2 diabetes mellitus (T2DM). Methods Data were collected from 1775 T2DM men and postmenopausal women in hospital. The odds ratios (OR) and corresponding 95% confidence intervals (CI) in relation to LH quartiles were obtained by multiple logistic regression analysis. Results LH levels were significantly higher in patients with macroalbuminuria than those with microalbuminuria, but not in patients with microalbuminuria than those with normoalbuminuria. Consistently, LH in those with estimated glomerular filtration rate (eGFR) less then 60 mL/min/1.73m2 were significantly higher than those with eGFR≥60 mL/min/1.73m2 . The prevalence of macroalbuminuria was obviously increased for subjects of the fourth quartile of LH vs the first to third quartile (20.4% vs 6.2%, 8.0%, 12.2% in men; 25.3% vs 5.5%, 3.8%, 9.3% in postmenopausal women). Multivariate logistic regression demonstrated that subjects within the highest quartile of LH had higher odds of macroalbuminuria than those within the lowest quartile (OR 4.00, 95%CI 1.87-8.55 for men; OR 9.62, 95%CI 3.42-27.08 for postmenopausal women), independent of age, diabetes duration or other metabolic factors. The area under curve (AUC) for detecting macroalbuminuria based on LH was 0.662 for men, and 0.767 for postmenopausal women. Conclusion High LH levels are positively associated with established DKD among Chinese men and postmenopausal women. Elevated LH may be a promising clinical factor for identifying established DKD. Here, we explored the role of YB-1 in T-cell homeostasis and survival, and the potential contribution of YB-1 in the pathogenesis of systemic lupus erythematosus (SLE). Methods We analyzed the expression of YB-1 and apoptosis by qPCR and FACS of CD4+ T-cells from peripheral blood of healthy donors (n=25) and SLE patients (n=25) ex vivo as well as in vitro after a 6-day-stimulation with αCD3- or αCD3/αCD28-coupled microspheres. YB-1 was overexpressed using lentiviral transduction with GFP-YB-1wt and knock-down using specific shRNA (-3 fold reduction, p less then 0.0001), respectively. Results YB-1 expression was significantly lower in apoptosis-prone and in activated T-cells of patients suffering from SLE compared to those of non-apoptotic and of activated T-cells of healthy volunteers (p=0.001). Knock-down of YB-1 demonstrates that reduced expression of YB-1 in T-cells inevitably led to expression of pro-apoptotic molecules and caspase 3 activation (1.6 fold), and subsequently to apoptosis. Furthermore, we revealed that YB-1 promotes survival pathways involving enhanced protein expression of the kinase Akt (2 fold) and Bcl-2 (3 fold), even when Fas was triggered. YB-1-mediated survival was reversed by Akt and PI3K inactivation. In SLE patients rescue of YB-1 expression strongly promotes survival of T-cells and even prevents cell death in those that were extremely apoptosis-prone. Conclusion Our data show that failure of YB-1 upregulation in T-cells of SLE patients leads to enhanced apoptosis. These findings have implications for diagnostics and possibly new treatment strategies for hematopoietic alterations in SLE.

The area under curve (AUC) for detecting macroalbuminuria based on LH was 0.662 for men, and 0.767 for postmenopausal women. Conclusion High LH levels are positively associated with established DKD among Chinese men and postmenopausal women. Elevated LH may be a promising clinical factor for identifying established DKD. Here, we explored the role of YB-1 in T-cell homeostasis and survival, and the potential contribution of YB-1 in the pathogenesis of systemic lupus erythematosus (SLE). Methods We analyzed the expression of YB-1 and apoptosis by qPCR and FACS of CD4+ T-cells from peripheral blood of healthy donors (n=25) and SLE patients (n=25) ex vivo as well as in vitro after a 6-day-stimulation with αCD3- or αCD3/αCD28-coupled microspheres. YB-1 was overexpressed using lentiviral transduction with GFP-YB-1wt and knock-down using specific shRNA (-3 fold reduction, p less then 0.0001), respectively. Results YB-1 expression was significantly lower in apoptosis-prone and in activated T-cells of patients suffering from SLE compared to those of non-apoptotic and of activated T-cells of healthy volunteers (p=0.001). Knock-down of YB-1 demonstrates that reduced expression of YB-1 in T-cells inevitably led to expression of pro-apoptotic molecules and caspase 3 activation (1.6 fold), and subsequently to apoptosis. Furthermore, we revealed that YB-1 promotes survival pathways involving enhanced protein expression of the kinase Akt (2 fold) and Bcl-2 (3 fold), even when Fas was triggered. YB-1-mediated survival was reversed by Akt and PI3K inactivation. In SLE patients rescue of YB-1 expression strongly promotes survival of T-cells and even prevents cell death in those that were extremely apoptosis-prone. Conclusion Our data show that failure of YB-1 upregulation in T-cells of SLE patients leads to enhanced apoptosis. These findings have implications for diagnostics and possibly new treatment strategies for hematopoietic alterations in SLE.

  • In the following, a mathematical model is outlined and used to compare the predicted efficacies of separate vs. nanocell administration for AAs and HAPs in tumours. The model is experimentally motivated, both in mathematical form and parameter values. Preliminary results of the model are highlighted throughout which qualitatively agree with existing experimental evidence. The novel prediction of our model is an improvement in the efficacy of AA/HAP combination therapies when administered through nanocells as opposed to separately. While this study specifically models treatment on glioblastoma, similar analyses could be performed for other vascularized tumours, making the results potentially applicable to a range of tumour types.Objectives One in five patients does not improve in pain with walking (non-responders) 12 months after total knee arthroplasty (TKA). This longitudinal study investigated a broad range of symptoms before and after TKA and evaluated possible differences in symptom distress between responders and non-responders with regards to pain with walking after TKA. Methods Prior to TKA surgery, 182 patients completed a demographic questionnaire and the Memorial Symptom Assessment Scale-Short Form (MSAS-SF). The MSAS-SF was repeated 12 months following TKA. Clinical data were extracted from medical records. Patients were categorized as responders or non-responders based on their trajectories of pain with walking assessed prior to surgery, on postoperative day 4, at 6 weeks, and at 3 and 12 months. In Arabidopsis, ANTAGONIST OF LIKE HETEROCHROMATIN PROTEIN 1 (ALP1) arose by domestication of the nuclease component of Harbinger class TE and acquired a new function as a component of POLYCOMB REPRESSIVE COMPLEX 2 (PRC2), a histone H3K27me3 methyltransferase involved in regulation of host genes and in some cases TE. It was not clear how ALP1 associated with PRC2, nor what the functional consequence was. Here, we identify ALP2 genetically as a suppressor of Polycomb-group (PcG) mutant phenotypes and show that it arose from the second, DNA binding component of Harbinger transposases. Molecular analysis of PcG compromised backgrounds reveals that ALP genes oppose silencing and H3K27me3 deposition at key PcG target genes. Proteomic analysis reveals that ALP1 and ALP2 are components of a variant PRC2 complex that contains the four core components but lacks plant-specific accessory components such as the H3K27me3 reader LIKE HETEROCHROMATION PROTEIN 1 (LHP1). We show that the N-terminus of ALP2 interacts directly with ALP1, whereas the C-terminus of ALP2 interacts with MULTICOPY SUPPRESSOR OF IRA1 (MSI1), a core component of PRC2. Proteomic analysis reveals that in alp2 mutant backgrounds ALP1 protein no longer associates with PRC2, consistent with a role for ALP2 in recruitment of ALP1. We suggest that the propensity of Harbinger TE to insert in gene-rich regions of the genome, together with the modular two component nature of their transposases, has predisposed them for domestication and incorporation into chromatin modifying complexes.

  • Introduction The ILUVIEN® (fluocinolone acetonide) Clinical Evidence in Portugal (ICE-PT) study is a retrospective, multicenter, observational study evaluating the effectiveness and safety of the FAc implant in patients with diabetic macular edema. Methods Patients included in this study had received the 0.2 µg/day fluocinolone acetonide implant for the treatment of diabetic macular edema and had measurements of visual acuity and retinal thickness assessed by optical coherence tomography for at least 12 months pre- and post-fluocinolone acetonide implant administration, with ⩾2 follow-up visits. Outcomes measured included visual acuity, central foveal thickness, and intraocular pressure. Results There was a significant increase in mean visual acuity compared with baseline at 3, 6, 9, and 12 months post-fluocinolone acetonide in both the overall study population and the pseudophakic subgroup (p less then 0.05 at all time points in both groups). A significant reduction in mean central foveal thickness compared with baseline was seen in the overall study population at 3, 6, 9, and 12 months post-fluocinolone acetonide (p less then 0.05 at all time points). At 12-month post-fluocinolone acetonide, a small but significant intraocular pressure increase of 1.0 mmHg was seen in the overall study population. Conclusion The results of this analysis show that switching from the current standard of care to the fluocinolone acetonide implant leads to beneficial effects in terms of vision and retinal structure in patients with diabetic macular edema and that patients benefited from FAc implant administration, regardless of lens status.Hemolytic uremic syndrome (HUS) may result in thrombotic central nervous system complications. We present a child with diarrhea-associated HUS who developed new-onset focal seizures secondary to cerebral sinovenous thrombosis (CSVT). Her CSVT was treated with low-molecular-weight heparin. The patient's seizures were controlled with levetiracetam, and her HUS was managed supportively with hemodialysis. Repeat imaging nearly 6 months following presentation and initiation of anticoagulation demonstrated cerebral sinus enlargement and persistent intraluminal webbing. Anticoagulation was discontinued after 6 months, and she did not experience long-term gross neurologic sequelae. CSVT is a complication of HUS that has not been previously described. In this report, we summarize the thrombotic central nervous system complications of pediatric HUS.Introduction A lack of uniformity in the choice of outcome measurement in hemophilia care and research has led to studies with incomparable results. We identified a need to define core outcome measures for use in research and clinical care of persons with hemophilia. Objective To move toward a core set of outcome measures for the assessment of persons with hemophilia in research and practice. Methods A modified nominal groups process was conducted with an international group of hemophilia experts, including persons with hemophilia as follows. Step 1 item generation for all potential outcome measures. Step 2 survey where respondents voted on the relative importance and usefulness of each item. Steps 3/4 2-day meeting where attendees voted for items they valued, followed by open discussion and a second round of voting. Step 5 survey where respondents selected their top five items from those with >50% agreement at the meeting. Results The highest ranked items for the pediatric core set (% agreement) are treatment satisfaction (92.7%), joint health (83.3%), a measure of access to treatment (82.5%), a measure of treatment adherence (72.5%), and generic performance based physical function (72.1%). The highest ranked items for the adult core set (% agreement) are total bleeding events (88.1%), EuroQol five dimensions (85.4%), treatment adherence (82.1%), joint health (79.1%), and number/location of bleeds per unit time (78.6%). Conclusion This process generated a list of preferred outcome measures to consider for assessment in persons with hemophilia. The online survey consisted of 36 questions covering demographic information, perceptions and knowledge of gene therapy for hemophilia, and educational preferences. Survey results were summarized using descriptive statistics. Results Of the 5117 survey recipients, 201 responded from 55 countries (4% response rate). We determined the effect of chemotherapy on extracellular vesicle tissue factor (EVTF) activity and the association of plasma EVTF activity with venous thromboembolism (VTE) and survival. Material and methods We enrolled 13 patients with pancreatic and 22 patients with colorectal cancer. Plasma samples were collected during the 85-day study period. Patients were followed for 3 months after the study period. We recorded symptomatic VTE during the study period (3 months) or asymptomatic deep vein thrombosis detected by ultrasound at day 85. We measured EVTF activity before and after chemotherapy. Results and conclusions In the pancreatic cancer group, 2 patients had elevated levels of EVTF activity. One of these patients developed symptomatic VTE and died, and the second patient did not have a VTE but died. Chemotherapy decreased EVTF activity in 2 pancreatic patients with high levels. In the colorectal cancer group, 4 patients developed VTE, but EVTF activity was not elevated in any patient and no patient died.
