More attention should be paid to the prevention and prompt treatment of diabetic patients, to maintain good glycaemic control especially those who require insulin therapy.Despite evidence to the effect that there is low parental adoption of tick-bite personal protective behavior (PPB) for their children - a population at high risk for tick exposure, very limited information is available on factors associated with parental adoption of PPB. The objective of this study was to identify the most significant factors associated with parental adoption of tick-bite PPB on behalf of a child or children at risk of tick encounters. A cross-section of parents in Indiana, USA whose child had spent time outdoors in tick habitat during the summer were recruited from representative online panels maintained by Qualtrics. Binary logistic regression was used to model determinants of five tick-bite PPBs. Our results revealed that the application of tick repellent (89 %, n = 718) followed by conducting a tick check of the child's body soon after returning from the outdoors (84 %, n = 676) were the PPBs most frequently adopted by parents. Conversely, tucking one's shirt into pants and pants into socks was the least frequently adopted PPB (48 %, n = 386). Compared to other factors evaluated in logistic regression models, parents who reported implementing one or more residential tick control practices were significantly more likely to adopt nearly all five tick-bite PPBs for their children. Additionally, parents who were more worried about their health due to ticks and reported being more likely to avoid the outdoors because of ticks were more likely to adopt at least three PPBs on behalf of their children. To ensure children can most safely engage in outdoor activity, identifying the factors associated with parental adoption of tick-bite preventive behaviors represents an important mechanism in the prevention of tick-borne diseases.Chalcone is a polyphenolic compound found abundantly in natural plant components. The IRE1α sulfonation-induced RIDD degraded miR-23b, enhancing the expression of NOX4. The expression of NOX4 was also upregulated in breast, and prostate cancer tissue. In chalcone-administered **** in vivo, tumor growth was regressed by the consistent mechanisms "NOX4-IRE1α sulfonation-RIDD". Similarly, NOX4 activation and IRE1α sulfonation were also highly increased under severe ER stress conditions. The role of FGF10 in liver IRI was further examined using adeno-associated virus-mediated gene silencing and overexpression. Overexpression of FGF10 alleviated liver dysfunction, reduced necrosis and inflammation, and protected hepatocytes from apoptosis in the early acute injury phase of IRI.

CRISPR/Cas9 (clustered regularly interspaced short palindromic repeats/CRISPR-associated protein 9)-mediated genome editing holds remarkable promise for the treatment of human genetic diseases. However, the possibility of off-target Cas9 activity remains a concern. To address this issue using clinically relevant target cells, we electroporated Cas9 ribonucleoprotein (RNP) complexes (independently targeted to two different genomic loci, the CXCR4 locus on chromosome 2 and the AAVS1 locus on chromosome 19) into human mobilized peripheral blood-derived hematopoietic stem and progenitor cells (HSPCs) and assessed the acquisition of somatic mutations in an unbiased, genome-wide manner via whole genome sequencing (WGS) of single-cell-derived HSPC clones. Bioinformatic analysis identified >20,000 total somatic variants (indels, single nucleotide variants, and structural variants) distributed among Cas9-treated and non-Cas9-treated control HSPC clones. Statistical analysis revealed no significant difference in the number of novel non-targeted indels among the samples. Moreover, data analysis showed no evidence of Cas9-mediated indel formation at 623 predicted off-target sites. The median number of novel single nucleotide variants was slightly elevated in Cas9 RNP-recipient sample groups compared to baseline, but did not reach statistical significance. Structural variants were rare and demonstrated no clear causal connection to Cas9-mediated gene editing procedures. We find that the collective somatic mutational burden observed within Cas9 RNP-edited human HSPC clones is indistinguishable from naturally occurring levels of background genetic heterogeneity.Since the life cycle of a building spans more than 50 years, studies of the environmental impacts in the construction industry have focused on reducing the energy consumption and greenhouse gas emissions during the operation and maintenance phase. The products of the construction industry are assembled using various building materials manufactured outside of the construction site. Consequently, it is essential that the manufactured building materials be transported to the construction site using various types of transportation methods. However, there is a lack of studies that assess the pollutant emissions of road transport while executing a construction project. The purpose of this study is to investigate the changes in the road pollutant emissions when the old diesel vehicles for transporting building materials are replaced according to enhanced pollutant emission regulations. In this study, we found that approximately 89, 64, 77, and 64% of NOx, VOC, PM, and CO, respectively, were emitted during transportation of building materials as a proportion of the emissions during the construction of the structure. A target increased ****range was determined, and participants were instructed to control their ****within the safe range. Gait assessment was conducted three times Baseline, Post-training and one month from the final biofeedback training session. In addition to MFC, step length, step width, double support time and foot contact angle were measured. Se-containing compounds have been found to reduce carcinogenesis of animal models, and dietary supplemental Se might decrease cancer risk. Se is mainly taken up by plant roots in the form of selenate via high-affinity sulfate transporters. Consequently, owing to the chemical similarity between Se and sulfur (S), the availability of S plays a key role in Se accumulation owing to competition effects in absorption, translocation, and assimilation. Moreover, naturally occurring S-containing compounds have proven to exhibit anticancer potential, in addition to other bioactivities. Therefore, it is important to understand the interaction between Se and S, which depends on Se/S ratio in the plant or/and in the growth medium. Brassicaceae (also known as cabbage or mustard family) is an important family of flowering plants that are grown worldwide and have a vital role in agriculture and populations' health. In this review we discuss the distribution and further interactions between S and Se in Brassicaceae and provide several examples of Se or Se/S biofortifications' experiments in brassica vegetables that induced the chemopreventive effects of these crops by enhancing the production of Se- or/and S-containing natural compounds. Extensive further research is required to understand Se/S uptake, translocation, and assimilation and to investigate their potential role in producing anticancer drugs.

Electrical impedance tomography (EIT) is an imaging modality where a patient or object is probed using harmless electric currents. The currents are fed through electrodes placed on the surface of the target, and the data consists of voltages measured at the electrodes resulting from a linearly independent set of current injection patterns. EIT aims to recover the internal distribution of electrical conductivity inside the target. The inverse problem underlying the EIT image formation task is nonlinear and severely ill-posed, and hence sensitive to modeling errors and measurement noise. Therefore, the inversion process needs to be regularized. However, traditional variational regularization methods, based on optimization, often suffer from local minima because of nonlinearity. This is what makes regularized direct (non-iterative) methods attractive for EIT. The most developed direct EIT algorithm is the D-bar method, based on Complex Geometric Optics solutions and a nonlinear Fourier transform. Variants and recent developments of D-bar methods are reviewed, and their practical numerical implementation is explained.Coccolithophores (calcifying phytoplankton) form extensive blooms in temperate and subpolar oceans as evidenced from ocean-color satellites. This study examines the potential to detect coccolithophore blooms with BioGeoChemical-Argo (****Argo) floats, autonomous ocean profilers equipped with bio-optical and physicochemical sensors. We first matched float data to ocean-color satellite data of calcite concentration to select floats that sampled coccolithophore blooms. We identified two floats in the Southern Ocean, which measured the particulate beam attenuation coefficient (cp) in addition to two core ****Argo variables, Chlorophyll-a concentration ([Chl-a]) and the particle backscattering coefficient (bbp). We show that coccolithophore blooms can be identified from floats by distinctively high values of (1) the bbp/cp ratio, a proxy for the refractive index of suspended particles, and (2) the bbp/[Chl-a] ratio, measurable by any ****Argo float. The latter thus paves the way to global investigations of environmental control of coccolithophore blooms and their role in carbon export.Permafrost thaw in Arctic watersheds threatens to mobilize hitherto sequestered carbon. We examine the radiocarbon activity (F14C) of dissolved organic carbon (DOC) in the northern Mackenzie River basin. From 2003-2017, DOC-F14C signatures (1.00 ± 0.04; n = 39) tracked atmospheric 14CO2, indicating export of "modern" carbon. This trend was interrupted in June 2018 by the widespread release of aged DOC (0.85 ± 0.16, n = 28) measured across three separate catchment areas. Increased nitrate concentrations in June 2018 lead us to attribute this pulse of 14C-depleted DOC to mobilization of previously frozen soil organic matter. We propose export through lateral perennial thaw zones that occurred at the base of the active layer weakened by preceding warm summer and winter seasons. Although we are not yet able to ascertain the broader significance of this "anomalous" mobilization event, it highlights the potential for rapid and large-scale release of aged carbon from permafrost.The North Atlantic warming hole (NAWH) is referred to as a reduced warming, or even cooling, of the North Atlantic during an anthropogenic-driven global warming. A NAWH is predicted by climate models during the 21st century, and its pattern is already emerging in observations. Despite the known key role of the North Atlantic surface temperatures in setting the Northern Hemisphere climate, the mechanisms behind the NAWH are still not fully understood. Using state-of-the-art climate models, we show that anthropogenic aerosol forcing opposes the formation of the NAWH (by leading to a local warming) and delays its emergence by about 30 years. In agreement with previous studies, we also demonstrate that the relative warming of the North Atlantic under aerosol forcing is due to changes in ocean heat fluxes, rather than air-sea fluxes. Electrical impedance tomography (EIT) is an imaging modality where a patient or object is probed using harmless electric currents. The currents are fed through electrodes placed on the surface of the target, and the data consists of voltages measured at the electrodes resulting from a linearly independent set of current injection patterns. EIT aims to recover the internal distribution of electrical conductivity inside the target. The inverse problem underlying the EIT image formation task is nonlinear and severely ill-posed, and hence sensitive to modeling errors and measurement noise. Therefore, the inversion process needs to be regularized. However, traditional variational regularization methods, based on optimization, often suffer from local minima because of nonlinearity. This is what makes regularized direct (non-iterative) methods attractive for EIT. The most developed direct EIT algorithm is the D-bar method, based on Complex Geometric Optics solutions and a nonlinear Fourier transform. Variants and recent developments of D-bar methods are reviewed, and their practical numerical implementation is explained.Coccolithophores (calcifying phytoplankton) form extensive blooms in temperate and subpolar oceans as evidenced from ocean-color satellites. This study examines the potential to detect coccolithophore blooms with BioGeoChemical-Argo (BGC-Argo) floats, autonomous ocean profilers equipped with bio-optical and physicochemical sensors. We first matched float data to ocean-color satellite data of calcite concentration to select floats that sampled coccolithophore blooms. We identified two floats in the Southern Ocean, which measured the particulate beam attenuation coefficient (cp) in addition to two core BGC-Argo variables, Chlorophyll-a concentration ([Chl-a]) and the particle backscattering coefficient (bbp). We show that coccolithophore blooms can be identified from floats by distinctively high values of (1) the bbp/cp ratio, a proxy for the refractive index of suspended particles, and (2) the bbp/[Chl-a] ratio, measurable by any BGC-Argo float. The latter thus paves the way to global investigations of environmental control of coccolithophore blooms and their role in carbon export.Permafrost thaw in Arctic watersheds threatens to mobilize hitherto sequestered carbon. We examine the radiocarbon activity (F14C) of dissolved organic carbon (DOC) in the northern Mackenzie River basin. From 2003-2017, DOC-F14C signatures (1.00 ± 0.04; n = 39) tracked atmospheric 14CO2, indicating export of "modern" carbon. This trend was interrupted in June 2018 by the widespread release of aged DOC (0.85 ± 0.16, n = 28) measured across three separate catchment areas. Increased nitrate concentrations in June 2018 lead us to attribute this pulse of 14C-depleted DOC to mobilization of previously frozen soil organic matter. We propose export through lateral perennial thaw zones that occurred at the base of the active layer weakened by preceding warm summer and winter seasons. Although we are not yet able to ascertain the broader significance of this "anomalous" mobilization event, it highlights the potential for rapid and large-scale release of aged carbon from permafrost.The North Atlantic warming hole (NAWH) is referred to as a reduced warming, or even cooling, of the North Atlantic during an anthropogenic-driven global warming. A NAWH is predicted by climate models during the 21st century, and its pattern is already emerging in observations. Despite the known key role of the North Atlantic surface temperatures in setting the Northern Hemisphere climate, the mechanisms behind the NAWH are still not fully understood. Using state-of-the-art climate models, we show that anthropogenic aerosol forcing opposes the formation of the NAWH (by leading to a local warming) and delays its emergence by about 30 years. In agreement with previous studies, we also demonstrate that the relative warming of the North Atlantic under aerosol forcing is due to changes in ocean heat fluxes, rather than air-sea fluxes. The patient had received parenteral iopamidol and oral iohexol five days prior. The patient's chief complaint at the Emergency Department (ED) presentation was shortness of breath and blisters throughout body. Upon arrival, the patient was awake, alert, and oriented with a blood pressure (BP) of 166/68, heart rate (HR) of 117 beats per minute, respiratory rate (RR) of 22 breaths per minute and oxygen saturation of 94% on room air. The features of pneumonia in children with neurologic impairment (NI) resemble those of healthcare-associated pneumonia is defined as pneumonia occurring in the community associated with healthcare risk factors. There are currently no guidelines for the treatment of pneumonia in children with NI. Here, we assessed whether the guidelines applicable for treating pneumonia in adults could be applied to children with NI.
Between 2008 and 2019, we enrolled children with NI who developed pneumonia and were treated in the pediatric ward of Kawasaki Medical School Hospital. We evaluated patient characteristics, the frequency of isolation of multidrug-resistant (MDR) pathogens, and clinical outcomes.
MDR pathogens were more frequently isolated from patients receiving tube feeding (TF) and/or with tracheostomy than from patients without these risk factors. Other risk factors, including a history of antibiotic therapy and methicillin-resistant Staphylococcus aureus isolation, recent hospitalization, residence in a nursing home or extended care facility, and low-dose, long-term macrolide therapy, did not significantly affect the frequency of MDR pathogen isolation. In patients receiving TF and/or with tracheostomy, treatment success was achieved in all cases treated with broad-spectrum antibiotics and 72.2% of cases treated with non-broad-spectrum antibiotics (P=0.007). Conversely, among patients without these risk factors, no such difference was observed.
Our findings indicate that the guideline to select antibiotics for treating pneumonia in children with NI should be simpler and more useful than the current guidelines for adult pneumonia, based on risk factor assessment for MDR pathogens.

The incidence of Clostridioides difficile infection (CDI) has been continuously increasing and thereby became an important issue worldwide. Appropriate diagnosis, management, and infection control are required for patients with CDI. Enzyme immunoassay (EIA) is a widely used standard diagnostic tool for C.difficile-specific glutamate dehydrogenase (GDH) and C.difficile toxins (toxins A and B). However, the sensitivity of EIA in detecting C.difficile toxins has been reported to be relatively low, resulting in CDI underdiagnosis. Therefore, nucleic acid amplification tests (NAAT) are recently developed for higher sensitivity/specificity test.
In this study, a total of 279 stool samples submitted for CDI diagnosis were examined using an independently developed new high-speed polymerase chain reaction (PCR) device (PathOC RightGene, Metaboscreen). In parallel, results were compared with those of definitive diagnosis and conventional diagnostic methods (EIA, real-time PCR) to assess the inspection accuracy.
PathOC RightGene showed high sensitivity (96.7%) and specificity (96.7%). Regarding the measurement time, C.difficile-specific and C.difficile toxin genes were simultaneously detected in approximately 25min for one sample (including the preprocessing and measurement time).

We herein report a first case of prosthetic valve endocarditis due to this organism after cardiovascular surgery. An 82-year-old man presented with repeat episodes of syncope and fever after aortic valve replacement, mitral valve replacement, left atrial appendage closure, and pulmonary vein isolation. Blood cultures maintained in aerobic bottles were repeatedly positive after 90-100 hours, and Gallium scan revealed abnormal accumulations in the sternum and left testis. While colonies formed by culturing the fluid of the parasternal area and blood cultures revealed gram-positive rods, we could not analyze the colony using matrix-assisted laser desorption ionization time of flight mass spectrometry (MALDI-TOF). M. wolinskyi was finally identified on 16S rRNA, hsp65, and rpoB gene sequencing. We treated the patient with multiple antimycobacterial drugs, i.e., amikacin, imipenem, and clarithromycin for 6 weeks, which was changed to oral ciprofloxacin and minocycline for 12 months.

Evidence-based practice (EBP) is a critical framework for supporting clinical decision-making that has been increasingly promoted in occupational therapy over the last 20years. Discipline-specific EBP frameworks and resources have emerged, primarily created by scholars from developed countries. However, EBP has received limited attention in literature published in developing and non-English speaking countries. This study aimed to explore Chilean occupational therapists' perceptions of EBP, factors influencing the adoption of EBP and sources of information used to inform clinical decision-making.
A cross-sectional online survey of Chilean occupational therapists included socio-demographic information, closed questions using a fivepoint Likert scale and open-ended questions. Numerical data were summarised using frequencies and percentages. Cross-tabulations explored relationships between variables. Narrative data from openended responses were analysed deductively using content analysis.
Complete surveys wpational therapy knowledge powerfully guide clinical reasoning, and build local research capacity. Building research partnerships and collaborations with the international scientific community are crucial to the global achievement of EBP.

Herein, the self-assembly behavior of tetraazatetrathia[8]circulenes is disclosed, which is driven by both enthalpy and entropy. Single-crystal X-ray diffraction analysis demonstrated that molecules of these tetraazatetrathia[8]circulenes form face-to-face stacked dimers with a 1D columnar structure owing to the circularly arranged dipole moments. Importantly, concentration- and temperature-dependent 1 H NMR spectra revealed that the formation of self-assemblies of tetraazatetrathia[8]circulenes in chloroform and methanol is favored by both enthalpic and entropic factors. The unique association behavior is due to the presence of sp2 -hybridized nitrogen atoms, which weakly coordinate to the hydrogen atoms of these solvents and reduce the π-electron density of the circulene cores.
Networked identification (ID) scanner technology is a novel approach to collecting licensed venue attendance data, and potentially reducing alcohol-related violence by detecting banned patrons. Using ID scanner data from three Queensland entertainment precincts (Safe Night Precincts; SNPs), we aim to (i) examine patterns in patron attendance to licensed venues; and (ii) examine patterns in the detection of banned patrons.
We conducted descriptive analyses of licensed venue patron entries between 1 October 2017 and 30 June 2019. Scans during high alcohol hours (Friday and Saturday, 2000-0600) were stratified by patron sex and key age groups. We described scans associated with a detected banning notice issued by venues, courts or police across all Queensland SNPs and three key SNPs.
ID scanner data showed similar attendance trends across SNPs. The peak licensed venue entry hour was 2300-2359, with Saturday being the peak day. Approximately two-thirds of scanned patrons were male, and patrons had a mean age of 25.8 years (SD = 8.80). Scanners detected 48 657 attempted entries by banned patrons, with the majority of attempts made by males (85%) aged 18-24 years (61%).
This study adds to the limited evidence on the use of ID scanners in the night-time economy and highlights the feasibility of ID scanner use in this setting. Results demonstrate the value of scanners to passively collect data on patrons, identify banned patrons and assist in the enforcement of bans.

Fragrances are organic compounds with pleasant odors that are widely used in every aspect of our daily life; some fragrance ingredients can cause allergic reactions. Hence, the qualitative and quantitative analysis of fragrance allergens can prevent consumers coming into contact with these compounds. In this study, we evaluated the ability of a dielectric barrier discharge ionization (DBDI) source for analyzing allergens that occur in fragrances.
A home-built liquid-infusion device was used to evaporate the liquid samples. An active capillary plasma ionization source, which is based on a dielectric barrier discharge, was used to ionize the analytes. Mass spectra were acquired in positive ion mode with an LTQ Orbitrap mass spectrometer.
Seven typical fragrance allergens were analyzed in this study. This work introduces a DBDI-MS-based analytical method for detecting and quantifying fragrance allergens. Since DBDI has the advantages of high sensitivity, simple operation and fast analysis time, it is very suitable for the rapid analysis of trace allergens in fragrances, and could easily be used for quality control of consumer products in the cosmetics market.

This study aimed to explore Chilean occupational therapists' perceptions of EBP, factors influencing the adoption of EBP and sources of information used to inform clinical decision-making. A cross-sectional online survey of Chilean occupational therapists included socio-demographic information, closed questions using a fivepoint Likert scale and open-ended questions. Numerical data were summarised using frequencies and percentages. Cross-tabulations explored relationships between variables. Narrative data from openended responses were analysed deductively using content analysis. Single-crystal X-ray diffraction analysis demonstrated that molecules of these tetraazatetrathia[8]circulenes form face-to-face stacked dimers with a 1D columnar structure owing to the circularly arranged dipole moments. Importantly, concentration- and temperature-dependent 1 H NMR spectra revealed that the formation of self-assemblies of tetraazatetrathia[8]circulenes in chloroform and methanol is favored by both enthalpic and entropic factors. The unique association behavior is due to the presence of sp2 -hybridized nitrogen atoms, which weakly coordinate to the hydrogen atoms of these solvents and reduce the π-electron density of the circulene cores. Networked identification (ID) scanner technology is a novel approach to collecting licensed venue attendance data, and potentially reducing alcohol-related violence by detecting banned patrons. Using ID scanner data from three Queensland entertainment precincts (Safe Night Precincts; SNPs), we aim to (i) examine patterns in patron attendance to licensed venues; and (ii) examine patterns in the detection of banned patrons. We conducted descriptive analyses of licensed venue patron entries between 1 October 2017 and 30 June 2019. Scans during high alcohol hours (Friday and Saturday, 2000-0600) were stratified by patron sex and key age groups. We described scans associated with a detected banning notice issued by venues, courts or police across all Queensland SNPs and three key SNPs. ID scanner data showed similar attendance trends across SNPs. The peak licensed venue entry hour was 2300-2359, with Saturday being the peak day. Approximately two-thirds of scanned patrons were male, and patrons had a mean age of 25.8 years (SD = 8.80). Scanners detected 48 657 attempted entries by banned patrons, with the majority of attempts made by males (85%) aged 18-24 years (61%). This study adds to the limited evidence on the use of ID scanners in the night-time economy and highlights the feasibility of ID scanner use in this setting. A home-built liquid-infusion device was used to evaporate the liquid samples. An active capillary plasma ionization source, which is based on a dielectric barrier discharge, was used to ionize the analytes. The analysis indicates that while CWC is strongly correlated with VOD (R2 = 0.62 across all sites), LWs accounts for 61-76% of the diurnal variation in CWC despite being less then 10% of CWC. Ignoring LWs weakens the near-linear relationship between CWC and VOD and reduces the consistency in diurnal variation. The contribution of LWs to CWC variation, however, decreases at longer, seasonal to inter-annual, time scales. Our results demonstrate that diurnal patterns of dew formation and rainfall interception can be an important driver of diurnal variation in CWC and VOD over tropical ecosystems and therefore should be accounted for when inferring plant diurnal water stress from VOD measurements.

In this narrative review, we aimed at examining the association between procedure-related bleeding risk and platelet count in patients with cirrhosis and severe thrombocytopenia in various settings. We updated to 2020 a previously conducted literature search using MEDLINE/PubMed and EMBASE. The search string included clinical studies, adult patients with chronic liver disease and thrombocytopenia undergoing invasive procedures, any interventions and comparators, and haemorrhagic events of any severity as outcome.
The literature search identified 1276 unique publications, and 15 studies met the inclusion criteria and were analysed together with those identified by the previous search. Most of the new studies included in our analysis did not assess the association between post-procedural bleeding risk and platelet count alone in patients with chronic liver disease. Furthermore, some results could have been biased by prophylactic platelet transfusions. A few studies found that severe thrombocytopenia may be predictive of bleeding following percutaneous liver biopsy, dental extractions, percutaneous ablation of liver tumours and endoscopic polypectomy.
Currently available literature cannot support definitive conclusions about the appropriate target platelet counts to improve the risk of bleeding in cirrhotic patients who underwent invasive procedures; moreover, it showed enormous variability in the use of prophylactic platelet transfusions.

It is important to determine whether A1AT can prevent the progress from moderate to severe lung injury and eventually to be used to treat COVID-19 patients with acute respiratory distress syndrome.Background The opioid epidemic has been extensively documented in the United States and Canada, but fewer data are available for Europe. Objective To predict the active components and signaling pathway of GHKGD and to explore its therapeutic mechanism in influenza and to verified it in vivo using network pharmacology. Methods The potential active components and therapeutic targets of GHKGD in the treatment of influenza were hypothesized through a series of network pharmacological strategies, including compound screening, target prediction and pathway enrichment analysis. Based on the target network and enrichment results, a mouse model of influenza A virus (IAV) infection was established to evaluate the therapeutic effect of GHKGD on influenza and to verify the possible molecular mechanism predicted by network pharmacology. Results A total of 116 candidate active compounds and 17 potential tacological characteristics of GHKGD in the treatment of influenza were revealed, and regulation of the RLR signaling pathway during the anti-influenza process was confirmed. This study provides a theoretical basis for the research and development of new drugs from GHKGD.The complexity of pathological mechanisms in Alzheimer's disease (AD) poses significant challenges to the development of corresponding drugs. Symptom-specific pharmacological interventions and alternative treatments provide promising treatment possibilities. Therefore, we considered a combination of selegiline (SEL) and electroacupuncture (EA). Interleukin 6 (IL-6) and tumor necrosis factor alpha (TNFα), known key players in RA pathogenesis, have been associated with reproductive disorders. This study investigates the role of these cytokines in decreased fertility in women with active RA. Preconception cytokine measurements of 61 patients from the PARA-cohort, a prospective study on RA and pregnancy, were studied in relation to time to pregnancy as a measure for fertility. IL-6 levels were higher in patients with a time to pregnancy longer than 1 year (p = 0.016). Survival analysis of patients stratified by high or low serum IL-6 levels, shows a prolonged time to pregnancy in the high IL-6 group (p = 0.045). Univariate cox regression analysis of IL-6 in relation to time to pregnancy as well as multivariate cox regression analysis correcting for age, disease activity, nulliparity, NSAID use and prednisone use were performed, with hazards ratios for log transformed IL-6 of 0.68 (95% CI 0.51-0.93, p = 0.015) and 0.66 (95% CI 0.43-0.99, p = 0.044), respectively. For TNFα, no association with time to pregnancy was found. This study shows that high IL-6, but not TNFα, is associated with decreased fertility in women with RA. This finding provides a rationale to therapeutically target the IL-6 pathway in the time period before pregnancy. More research in the form of large cohort studies on drug safety and the effect of bDMARDS on fertility is needed for implementation of treatment strategies directed at fertility issues in women with RA.

IL-6 levels were higher in patients with a time to pregnancy longer than 1 year (p = 0.016). Survival analysis of patients stratified by high or low serum IL-6 levels, shows a prolonged time to pregnancy in the high IL-6 group (p = 0.045). Exopolysaccharides (EPS) produced by cyanobacteria are complex biomolecules of anionic nature with potential biomedical applications. The addition of acetate ions to the culture medium slightly stimulated EPS production, achieving 1463.1 ± 16.0 mgL-1 (PCC7413) and 1372.1 ± 29.0 mgL-1 (PCC7936). Both EPS presented nine monosaccharide residues and a MW > 1000 kDa. The acetate addition changed the monosaccharide molar percentages. FTIR and DLS results confirmed the anionic nature and the presence of sulfate groups in both EPS, which are determinant features for biomedical applications. Further, the membranes were characterized for their functionality and surface characteristics using ATR-IR, FESEM, AFM, UV-vis spectroscopy and contact angle measurements, respectively. New asymmetric membrane performances in cross flow process were evaluated in terms of pure water flux, NaCl (∼40 %), red brown/organic dye (>98 %) and tannery wastewater flux and rejection (>98 %). With a higher pure water flux (>100 L m-2 h-1) compared to control (40 L m-2 h-1) at 4 bar, membrane showed exceptional antifouling behaviors in comparison to commercial PA membrane. Further, surface characteristics of the membranes before and after rigorous testing were evaluated using AFM micrographs and SEM imaging.A fully biobased benzoxazine monomer, V-fa (using vanillin and furfurylamine) was grafted onto chitosan (CS) at different weight ratios (CXVY) using "grafting to" benign Schiff base chemistry. Incorporation of V-fa onto CS increased the tensile strength and improved chemical resistance of the CS-graft-V-fa films. Reversible labile linkages, expansion of CS galleries and leaching out of phenolic species from biobased polymer films led to an improved antibacterial activity against Staphylococcus aureus, which is ∼125 times higher than the bare CS film, V-fa and oligomeric V-fa. The leached out species from films were analyzed extensively by NMR, FTIR, GPC, ABTS and HRMS analysis. Oxidative-stress seems to be responsible for antibacterial activity. Current work illustrates an attractive synthetic approach and the improved antibacterial performance of biobased CS-graft-poly(V-fa) films which may hold as a potential alternative for wound-healing and implant applications in future.

To explore the effect of mild hypothermia on nerve injury by establishing a rat model of intracerebral hemorrhage (ICH), and to clarify the specific molecular mechanism of mild hypothermia in improving brain injury in ICH rats.
The rat model of ICH was established by collagenase injection. The neurological deficit score (NDS), brain tissue water detection, and Nissl staining were applied to detect the degree of brain injury. ELISA was used to analyze the expression of proinflammatory cytokines and serum nerve injury indexes. Flow cytometry and Western Blot were used to detect neuronal apoptosis.
Mild hypothermia treatment significantly improved the brain injury of the ICH rats and down-regulated the inflammatory response and oxidative stress in the brain tissue. Moreover, mild hypothermia also effectively inhibited IRAK2/NF-κB signaling pathway and thus affect neuronal apoptosis.

Torch-light Eye Screening Test (TEST) is a simple eye screening technique designed for use by opticians to look for common anterior segment eye conditions. The TEACHES-Learning Electronic Module (TEACHES-LEM) is an e-learning platform that was developed to teach opticians to perform TEST. The objective of this study was to compare the effectiveness of TEACHES-LEM with face-to-face training (F2FT) in the training and assessment of knowledge among opticians.
Participants were randomly assigned in this experimental study to receive either the intervention group ((TEACHES-LEM, n=60) or the control group (F2FT, n=57). The conceptual knowledge of TEST was assessed with a 20-item clinical scenario-based multiple choice question (MCQ) test before and after teaching (immediately post-teaching and 1-month post-teaching). The MCQ test was developed by three ophthalmologists to give face validity.
The pre-teaching test scores (TS), indicating prior knowledge, were comparable in both groups (10.02±2.79 versus 10.40±4.17, p=0.563, independent t test). The mean immediate post teaching score for TEACHES-LEM was 13.3±4.01 versus 12.3±3.29 in the F2FT group (p=0.170, independent t test). The mean post 1-month teaching score for TEACHES-LEM and F2FT groups were also comparable, 14.5±4.19 versus 13.4±3.90 respectively (p=0.295, independent t test), indicating non-inferiority of TEACHES to F2FT.
The TEACHES-LEM e-learning tool is as effective as F2FT in teaching opticians to perform TEST. It is an alternative to face-to-face teaching in delivering knowledge and assessment. The obviation for physical contact will make it a useful teaching tool during the COVID-19 pandemic period.

Pleural mesothelioma, a devastating asbestos-associated malignancy, urgently requires a novel effective therapy. Heat shock protein 70 (HSP70), which is synthesized in the cell response to protein damage, is expected to be a new target for antitumor treatment. In addition to its well-known protein refolding function, HSP70 regulates cell proliferation through different pathways, including PI3K/AKT/mTOR, and autophagy in malignant cells. In this study, we attempted to clarify the effects of VER-155008, an HSP70 inhibitor, on pleural mesothelioma.
Human pleural mesothelioma cell lines 211H, H2452 and H28 were cultured with VER-155008, and protein expression, cell proliferation, colony formation, cell cycle, synergistic effect with cisplatin, and autophagy induction were analyzed.
