  • BACKGROUND Nitrate is one of the most common chemical contaminants of groundwater, and it is an important unqualified factor of rural groundwater in Yantai. In order to assess the risk of exposure to drinking water nitrate for adults and juveniles, in recent years, we monitored the nitrate concentrations in rural drinking water,a model was also used to assess the human health risk of nitrate pollution in groundwater. METHODS From the year 2015 to 2018, the drinking water in rural areas of Yantai was tested according to the "Sanitary Standard for Drinking Water" (GB5749-2006). The principal component analysis was used to analyze the relationship between groundwater chemicals and nitrate. The model was used to assess human health risks of groundwater nitrate through the drinking water and skin contact. RESULTS A total of 2348 samples were tested during the year 2015-2018.Nitrate and total dissolved solids, total hardness, chloride are all relevant, the above indicators may come from the same source of pollution; The median nitrate content (CEXP50) was 17.8 mg / L; the risk of exposure in each group was ranked as Juveniles > Adult female > Adult male;the median health risk (HQ50) for minors and adults exceed 1. CONCLUSIONS The concentrations of nitrate is stable and does not change over time. The high concentration of nitrate in rural areas of Yantai may be the result of the interaction of fertilizers and geological factors. The risk of exposure to nitrate in juveniles and adults is above the limit, so it is necessary to be on the alert for the high levels of nitrate.BACKGROUND Echinococcosis is a zoonotic parasitic disease causing serious health problems in both humans and animals in different endemic regions across the world. There are two different forms of human echinococcosis Cystic Echinococcosis (CE) and Alveolar Echinococcosis (AE). CE is caused by the larval stage of Echinococcus granulosus sensu lato and AE by the larval stage of Echinococcus multilocularis. Geographically, CE is universally distributed, while AE is prevalent in the northern hemisphere. Although the disease is endemic in neighboring countries (China, Iran and India) of Pakistan, there are limited reports from that country. Besides, there are no comprehensive data on the genotyping of Echinococcus species in humans based on sequence analysis. This study aimed to detect the presence of human CE and to identify Echinococcus spp. in human isolates through genetic characterization of hydatid cysts in the Punjab Province of Pakistan. METHODS Genetic analysis was performed on 38 human hydatid cyst samples collected from patients with echinococcosis using mitochondrial cytochrome c oxidase subunit 1 (cox1), cytochrome b (cytb) and NADH subunit 1 (nad1). Patient data including age, epidemiological history, sex, and location were obtained from hospital records. RESULTS According to the sequence analysis we detected E. granulosus sensu stricto (n = 35), E. canadensis (G6/G7) (n = 2), and E. multilocularis (n = 1). Thus, the majority of the patients (92.1%, 35/38) were infected with E. granulosus s.s. This is the first molecular confirmation of E. canadensis (G6/G7) and E. multilocularis in human subjects from Pakistan. CONCLUSIONS These findings suggested that E. granulosus s.s. is the dominant species in humans in Pakistan. In addition, E. canadensis (G6/G7) and E. multilocularis are circulating in the country. Further studies are required to explore the genetic diversity in both humans and livestock.BACKGROUND Q fever (Coxiella burnetii infection) has been associated with adverse perinatal outcomes. METHODS Between January 9 and July 24, 2014, within the level-4 maternity of Saint Pierre hospital and the level-1 maternity of Le Tampon, we proposed to screen all parturient women for Coxiella burnetii serology. Seropositivity was defined using indirect immunofluorescence for a dilution of phase 2 IgG titre ≥164. https://www.selleckchem.com/CDK.html Further dilutions were chosen to discriminate recent or active infections from past or prevalent infections ( 90% of positive serologies corresponded to past infections or false positives. Seropositivity was associated with none of the abovementioned adverse perinatal outcomes, whether in unpaired or matched analyses on propensity score. CONCLUSION The magnitude and the pattern of seroprevalence suggest that Q fever is endemic on Reunion island. In this context, we found no significant contribution of prevalent Coxiella burnetii infection to adverse pregnancy outcomes. Although reassuring, these data put in our endemic context, with a previously demonstrated increased risk of incident Q fever associated miscarriage, encourage us to protect pregnant women against the risk of new infection, periconceptional or early in pregnancy.BACKGROUND KwaZulu-Natal (KZN) Initiative for breastfeeding support (KIBS) was a multipronged intervention to support the initiation and sustaining of breastfeeding, implemented between 2014 and 2017. We present results of two surveys conducted before and after KIBS implementation to assess changes in infant feeding practices in KZN over this time period. METHODS Two cross-sectional surveys were conducted in primary health care clinics. Multistage stratified random sampling was used to select clinics and participants. Sample size was calculated to provide district estimates of 14-week exclusive breastfeeding (EBF) rates at baseline (KIBS1), and provincial estimates at endpoint (KIBS2). At KIBS1 the sample required was nine participating clinics in each of 11 districts (99 clinics) with 369 participants per district (N = 4059), and at KIBS2 was 30 clinics in KZN with 30 participants per clinic (N = 900). All caregivers aged ≥15 years attending the clinic with infants aged 13- less then  16 weeks were eligibleted odds ratio [AOR] 3.76; 95% CI 3.1-4.6), as were HIV positive mothers (AOR 2.1; 95% CI 1.7-2.6). CONCLUSION Despite improvements to exclusive breastfeeding, failure to initiate and sustain breastfeeding is a challenge to achieving optimal breastfeeding practices. Interventions are required to address these challenges and support breastfeeding particularly among working mothers and HIV positive mothers.

  • Background Cytochrome P-450 4A11 (CYP4A11) and peroxisome proliferator-activated receptor-α (PPARα) are expressed at high levels in renal proximal tubules, and upregulation of CYP4A11 protein levels is known to be influenced by PPAR agonists. The goal of this study was to evaluate the clinicopathological role of CYP4A11 expression in renal cell carcinoma (RCC). Methods We performed immunohistochemical analysis of CYP4A11, CYP4A22 and PPARα and correlated the results with the clinicopathological features of RCC (n=139). Reverse transcription digital droplet polymerase chain reaction (RT-ddPCR) against CYP4A11 and CYP4A22 was also performed. Results CYP4A11 mRNA expression levels were higher in non-neoplastic kidney tissues than in matched tumor tissues in 12 matched pairs of freshly frozen primary clear-cell RCC (ccRCC) and nontumor tissue (p=0.002). Immunohistochemical staining showed that CYP4A11 expression was significantly lower in ccRCC than in non-ccRCCs, including papillary, chromophobe, and unclassified RCCs (p less then 0.001). CYP4A11 expression was associated with PPARα expression, males and high nuclear histologic grades (p=0.001, p=0.018 and p less then 0.001). Univariate and multivariate analyses revealed that CYP4A11 expression was correlated with short overall survival (p=0.007 and p=0.010). Conclusion These findings suggest that CYP4A11 expression is a potential poor prognostic factor of RCC. Our study was conducted to study the mechanism underling p16 mediated promoting effect of cell proliferation in cervical cancer cell lines. CCK8 assay and EdU incorporation were conducted to evaluate cell proliferation. Loss-of-function assay was used to silence p16 in Ca Ski and SiHa cells. Next, western blot, qPCR, RNA silencing, luciferase activity assay, run-on assay, mRNA stability assay, RNA immunoprecipitation, co-immunoprecipitation Immunofluorescence were performed to examine the interaction between CDK6, HuR, and IL1A mRNA in p16 mediated proliferation promoting effect. Our results showed that (1) Silencing p16 inhibited the proliferation of cervical cancer cells by decreasing the half-life of IL1A mRNA in CDK6 dependent manner; (2) The stabilization of IL1A mRNA was regulated by HuR which could be inactivated by p16/CDK6 mediated phosphorylation at Ser202; (3) IL1A mediated the oncogenic activity of p16 in cervical carcinoma cell lines. Clinical significance of the various miR-34 family members has not been studied in ovarian cancer. In 228 ovarian cancers and in 19 non-neoplastic fallopian tube samples we analysed miR-34 a/****expression in relation to clinicopathological characteristics and clinical outcome. We found significantly lower levels of miR-34 a/****in ovarian cancers as compared to control-tissues (P=0.002, P less then 0.001, P less then 0.001, respectively). Expression of miR-34 ****revealed an inverse correlation with BRCA1/2 mRNA-expression (BRCA1 miR34 ****P=0.002 each; BRCA2 miR-34 ****P less then 0.001 each), the same was true for miR-34a and BRCA2 mRNA-expression (P less then 0.001). The miR-34 family expression was found to be significantly lower in type 2 in comparison to type 1 cancers (P less then 0.001) and in TP53-mutated compared with TP53-wild-type ovarian cancers (P less then 0.001, P=0.002, P=0.004, respectively). When low grade serous ovarian cancers were compared with high grade serous cancers the respective miR-34 a/****expression was 2.6-, 40.8- and 32.3-fold higher. The expression of each of the miR-34 family members was revealed to be of independent prognostic relevance regarding progression free survival (PFS); miR-34a HR 0.6, P=0.033; miR-34b HR 0.2, P=0.001 and miR-34c HR 0.3, P=0.002, respectively). For overall survival (OS) independency of the prognostic value was confined to miR-34b (HR 0.4, P=0.016) and miR-34c (HR 0.6, P=0.049). The independency of the prognostic value of our identified thresholds was confirmed for PFS for miR-34c in a publicly available dataset (NCBI Gene Expression Omnibus GSE73582). Our findings suggest that downregulation of miR-34 family is a crucial part in ovarian cancer development. Several previous studies have described estrogen receptors (ER), progesterone Receptor (PR) and phosphatase and tensin homolog (PTEN) are associated with clinicopathological factors and prognosis in EC patients. However, during EC patients follow-up, we found that some EC patients with down-regulation of PTEN, but up-regulation of ER or PR , and some EC patients with down-regulation of ER or PR, but up-regulation of PTEN also had a poor prognosis. Therefore, to reveal the prognosis of EC patients with different phenotypes based on PTEN, ER and PR expression, 120 cases formalin-fixed paraffin-embedded EC tissues and 543 cases uterine corpus endometrial carcinoma (UCEC) patients from the cancer genome atlas (TCGA) UCEC datasets were analyzed. Results showed that EC tissues can be classified to PTENLERLPRL, PTENHERLPRL, PTENHERHPRH, PTENLERHPRH, PTENHERHPRL, PTENHERLPRH, and PTENLERHPRL phenotypes basing on IHC analysis.

Results The median hospital stay of COVID-19 patients was 6 days. Serum albumin (Alb) level was lower in patients with COVID-19 confirmed on admission than in patients with suspected COVID-19 (40.08 g/L vs 42.50 g/L, P = .016), while the level of aspartate aminotransferase (AST) was higher (23 U/L vs 18 U/L, P = .005). Abnormal results of liver blood tests in patients with COVID-19 included increased levels of alanine transaminase (ALT) (21.2%, 14 patients), AST (15.2%, 10 patients), and gamma-glutamyl transpeptidase (GGT) (22.7%, 15 patients). After 5-10 days of treatment, levels of Alb and AST in COVID-19 patients were significantly decreased (P .05). In addition, only levels of AST were positively correlated with the duration of hospital stay (r = .334, P = .007). Incidence rate ratios (IRRs) were estimated from Poisson regression (Period 1 reference) adjusting for hospital setting, sex, age, Privigen indication, dose, and first use. Results Crude incidence rates of HA were 1.49 per 10,000 person-days in Period 1 (38 HA, 9439 patients), 1.01 in Period 2 (20 HA, 7710 patients), and 0.14 in Period 3 (3 HA, 7759 patients). Adjusted IRR for HA in Period 2 was 0.71 (95% confidence interval [CI], 0.41-1.23), and in Period 3 was 0.10 (0.03-0.33) compared with Period 1. The concentration-QTc relationship assessed on placebo-adjusted change from baseline for Fridericia-corrected QT (ΔΔQTcF) was the primary end point. ΔΔQTcF values of 4.5, 12.3, and 19.8 ms for the 20-, 60-, and 140-mg doses were observed at the corresponding trazodone peak plasma concentrations. A true-positive finding was defined as the presence of a pathology-confirmed cancer within 12 months. Performance benchmarks included sensitivity and specificity, examined per quartile of radiologists' pes showed a wide range of performance differences between high-volume readers. However, transfection methods suitable for open field applications are still desirable as a cheaper alternative. Biosafety concerns related to the use of recombinant agrobacteria in an industrial transfection process include possible transformation or transfection of unintended hosts or spread of the genetically modified agrobacteria in the environment. In this paper, we explored a novel biocontrol approach resulting in greater biosafety of the transient expression process in plants. Our proposed solution involves inducible expression of Agrobacterium tumefaciens toxin PemK and antitoxin PemI that provides for strictly regulated T-DNA transfer from agrobacteria to plants. We also identified several other toxins from putative Agrobacterium toxin-antitoxin modules and demonstrate their potential usefulness in the control of Agrobacterium tumefaciens as a DNA vector.

  • The frontline treatment options for patients with metastatic renal cell carcinoma (****) are evolving rapidly since the approval of combination immunotherapies by the U.S. Food and Drug Administration (USFDA) and the European Medicines Agency (EMA). In particular, in combination with vascular endothelial growth factor receptor (VEGFR) tyrosine-kinase inhibitors (TKIs), immune checkpoint inhibitors (ICIs) have significantly improved the outcome of patients with **** compared to TKI monotherapy. Here, we review the preclinical data supporting the combination of ICIs with VEGFR TKIs. The VEGF-signaling inhibition could ideally sustain immunotherapy through a positive modulation of the tumor microenvironment (TME). Antiangiogenetics, in fact, with their inhibitory activity on myelopoiesis that indirectly reduces myeloid-derived suppressor cells (MDSCs) and regulatory T cells' (Tregs) frequency and function, could have a role in determining an effective anti-tumor immune response. Particularly, the role of NK cells has been argued in either elimination of target tumor cells or escape of tumor cells from immune surveillance. Therefore, an NK cell activation approach has been explored. The immune balance at different stages of both diseases is explored in light of disease progression. Furthermore, the tenure length of officials plays an inverted U-shaped role in regulating the impact of environmental regulations on enterprise green innovation. Resting whole saliva was collected with the drainage technique, followed by the measurement of 11 biomarkers. RESULTS The salivary flow was higher in patients with BMS. Among all the biomarkers studied, significantly higher levels of alpha-amylase, immunoglobulin A (IgA), and macrophage inflammatory protein-4 (MIP4) and lower levels of uric acid and ferric reducing activity of plasma (FRAP) were observed in the saliva of patients with BMS and the controls (p less then 0.05 in all cases).

Here, we review the preclinical data supporting the combination of ICIs with VEGFR TKIs. The VEGF-signaling inhibition could ideally sustain immunotherapy through a positive modulation of the tumor microenvironment (TME). At the same time, morphine induces the activation of three arms of unfolded protein response (UPR) inositol-requiring enzyme 1α/X-box binding protein 1 (IRE1α/XBP1), protein kinase RNA-like ER kinase/eukaryotic initiation factor 2 subunit alpha (PERK/eIF2α), and activating transcription factor 6 (ATF6). Notably, we found that inhibition on either IRE1α/XBP1 or ATF6, but not on PERK/eIF2α could attenuate the development of OIH. Consequently, ER stress induced by morphine enhances PKA-mediated phosphorylation of NMDA receptor subunit 1(NR1) and leads to OIH. We further showed that heat shock protein 70 (HSP70), a molecular chaperone involved in protein folding in ER, is heavily released from spinal neurons after morphine treatment upon the control of KATP channel. Glibenclamide, a classic KATP channel blocker that inhibits the efflux of HSP70 from cytoplasm to extracellular environment, or HSP70 overexpression in neurons, could markedly suppress morphine-induced ER stress and hyperalgesia. Brg1 interacted with SREBP2 and was recruited by SREBP2 to the cholesterogenic gene promoters. Reciprocally, Brg1 deficiency dampened the occupancies of SREBP2 on target promoters likely through modulating H3K9 methylation on the cholesterogenic gene promoters. Mechanistically, Brg1 recruited the H3K9 methyltransferase KDM3A to co-regulate pro-cholesterogenic transcription. Automated ECG measurements from raw data were determined. The primary outcome was incident atrial fibrillation (AF). Results A total 59 mitral stenosis patients were included (age 59 [54-65] years, 13 (22%) males). New onset AF was observed in 27 patients. Age (odds ratio [OR] 1.08 [1.01-1.16], P = 0.017), systolic blood pressure (OR 1.03 [1.00-1.07]; P = 0.046), mean P-wave area in V3 (odds ratio 3.97 [1.32-11.96], P = 0.014) were significant predictors of incident AF. On multivariate analysis, age (OR 1.08 [1.00-1.16], P = 0.037) and P-wave area in V3 (OR 3.64 [1.10-12.00], P = 0.034) remained significant predictors of AF. Receiver-operating characteristic (ROC) analysis showed that the optimum cut-off for P-wave area in V3 was 1.45 Ashman units (area under the curve 0.65) for classification of new onset AF. A decision tree learning model with individual and non-linear interaction variables with age achieved the best performance for outcome prediction (accuracy = 0.84, precision = 0.84, recall = 0.83, F-measure = 0.84). Conclusion Atrial electrophysiological alterations in mitral stenosis can detected on the electrocardiogram. Age, systolic blood pressure, and P-wave area in V3 predicted new onset AF. The definition of the Moho in the Iranian region is of considerable importance due to the geological complexity of the area also characterized by tectonic and orogenic events of particular uniqueness. We applied the collocation method to Moho recovery using the gravity data generated by GOCO03S model reduced by topography/bathymetry, sediment and consolidated crust effects from CRUST1.0. These data have been complemented with seismic Moho depth estimates. A compilation of 213-points seismic depth has been collected over Iran and used in the integrated gravimetric-seismic inversion. Among them, 140 seismic points have been selected completely random and included as data in the integrated collocation approach for Moho depth estimation. The 73 remaining seismic points have been used as checking points for validating the estimated Moho. In the first run, gravity data only have been considered to collocation Moho recovery. When comparing this gravimetric solution with the 73 seismic checking points, a standard deviation of 6.2 km was found. In case of considering the regional seismic depths into the collocation approach, the standard deviation of the residuals between our results and seismic checking Moho depths improved to 4.9 km. It must be stated that, even in the integrated inversion, a significant discrepancy between the seismic and the integrated gravimetric-seismic Moho is present in the South Caspian Basin. Significant advances have been achieved in the diagnosis, management, and treatment of LSDs patients. In terms of approved therapies, these include enzyme replacement therapy (ERT), substrate reduction therapy, hematopoietic stem cell transplantation, and pharmacological chaperone therapy. In this review, we summarize the Colombian experience in LSDs thorough the evidence published. We identified 113 articles published between 1995 and 2019 that included Colombian researchers or physicians, and which were mainly focused in Mucopolysaccharidoses, Pompe disease, Gaucher disease, Fabry disease, and Tay-Sachs and Sandhoff diseases. The adsorption sequence, carried out at various ionic strengths, showed that the adsorption mechanism onto the Gh particles is both electrostatic and specific at pH below the pHpzc, while at a pH range greater than the pHpzc the mechanism is electrostatic in nature. The total number of surface sites, determined using the graphical extrapolation method, was 11OH/nm2. Several studies have suggested that polyphenols could also bind to cellular proteins and modulate their activity, however, the mechanisms of action underlying their beneficial effects are complex and are not fully understood. The aim of this work was to characterize phenolic compounds present in blue corn and black bean extracts as well as identify their potential interactions with target proteins involved in diabetes pathogenesis using in silico approach. Total polyphenols content of both blue corn and black beans was identified using UPLC-ESI/qTOF/MS and quantified by colorimetric assays. In this work we identified twenty-eight phenolic compounds in the extracts, mainly anthocyanins, flavonols, hydroxycinamic acids, dihydroxybenzoic acids, flavones, isoflavones, and flavanols.

The definition of the Moho in the Iranian region is of considerable importance due to the geological complexity of the area also characterized by tectonic and orogenic events of particular uniqueness. We applied the collocation method to Moho recovery using the gravity data generated by GOCO03S model reduced by topography/bathymetry, sediment and consolidated crust effects from CRUST1.0. These data have been complemented with seismic Moho depth estimates. A compilation of 213-points seismic depth has been collected over Iran and used in the integrated gravimetric-seismic inversion. Among them, 140 seismic points have been selected completely random and included as data in the integrated collocation approach for Moho depth estimation. The 73 remaining seismic points have been used as checking points for validating the estimated Moho. In the first run, gravity data only have been considered to collocation Moho recovery. When comparing this gravimetric solution with the 73 seismic checking points, a standard deviation of 6.2 km was found. In case of considering the regional seismic depths into the collocation approach, the standard deviation of the residuals between our results and seismic checking Moho depths improved to 4.9 km. It must be stated that, even in the integrated inversion, a significant discrepancy between the seismic and the integrated gravimetric-seismic Moho is present in the South Caspian Basin. Low quality of CRUST1.0 could explain this inconsistency in this area. © 2020 The Author(s).Lysosomal storage diseases (LSDs) are a group of about 50 inborn errors of metabolism characterized by the lysosomal accumulation of partially or non-degraded molecules due to mutations in proteins involved in the degradation of macromolecules, transport, lysosomal biogenesis or modulators of lysosomal environment. Significant advances have been achieved in the diagnosis, management, and treatment of LSDs patients. In terms of approved therapies, these include enzyme replacement therapy (ERT), substrate reduction therapy, hematopoietic stem cell transplantation, and pharmacological chaperone therapy. In this review, we summarize the Colombian experience in LSDs thorough the evidence published. We identified 113 articles published between 1995 and 2019 that included Colombian researchers or physicians, and which were mainly focused in Mucopolysaccharidoses, Pompe disease, Gaucher disease, Fabry disease, and Tay-Sachs and Sandhoff diseases.   • Increased prevalence of antibiotic resistance in skin and soft tissue infections is a concerning public health challenge currently facing medical science. A combinatory, broad spectrum biocidal antiseptic has been developed ("ASP") as a topically applied solution to potential resistant and polymicrobial infected wounds that may be encountered in this context. The ASP-105 designate was evaluated in vitro by determining the minimal inhibitory concentration (MIC) and minimal bactericidal concentration (MBC), against different strains of methicillin-resistant Staphylococcus aureus (MRSA), resulting estimates of which approximated the positive control (bacitracin). To evaluate in vivo microbicide efficacy, we utilized a murine full thickness wound model to study bacterial infection and wound healing kinetics. **** were experimentally wounded dorsally and infected with bioluminescent MRSA. However, the community assembly of endophytic fungi in ectomycorrhizae is less investigated at a large spatial scale. Here, we examined the endophytic fungal communities in ectomycorrhizae of 22 species in four genera belonging to Betulaceae in Chinese forest ecosystems, using Illumina Miseq sequencing of internal transcribed spacer 2 amplicons. The relative contribution of host phylogeny, climate and soil (environmental filtering) and geographic distance (dispersal limitation) on endophytic fungal community was disentangled. https://www.selleckchem.com/products/azd9291.html In total, 2,106 endophytic fungal operational taxonomic units (OTUs) were obtained at a 97% sequence similarity level, dominated by Leotiomycetes, Agaricomycetes, Eurotiomycetes, and Sordariomycetes. In this work, we studied the use of six lactic acid bacteria (LAB), previously selected for their properties as bioprotective agents, for salmon dill gravlax biopreservation. Naturally contaminated salmon dill gravlax slices, with a commercial shelf-life of 21 days, were purchased from a French industrial company and inoculated by spraying with the protective cultures (PCs) to reach an initial concentration of 106 log CFU/g. PC impact on gravlax microbial ecosystem (cultural and acultural methods), sensory properties (sensory profiling test), biochemical parameters (pH, TMA, TVBN, biin to extend the sensory quality, even beyond 25 days. In this study, we investigated the growth character and cariogenic virulence during the recovery of Streptococcus mutans drug-tolerant persister cells induced by a novel quaternary ammonium dimethylaminododecyl methacrylate (DMADDM). A remarkable lag phase was observed in S. mutans persisters when regrew at the first 24 h compared to normal cells. During the entire recovery state, persisters are metabolically active to increase the production of both water-soluble and water-insoluble glucan. The shortage of cell number in persisters resulted in the decrease of lactic acid production, but persisters gradually recovered th prolonged lag phase and the involvement of quorum-sensing system in the regrowth of S. mutans persisters may provide the potential targets. In this study, 323 bacterial strains were isolated from the rhizosphere of severely diseased B. napus in Dangyang county, Hubei province, China. Antagonistic strains were first identified based on dual culture inhibition zones with Fusarium oxysporum and Magnaporthe oryzae. These were then further screened in germination inhibition and viability assays of resting spores of P. brassicae. Finally, eight of the antagonistic strains were found to significantly reduce the disease severity of clubroot by more than 40% under greenhouse conditions, and two strains, F85 and T113, were found to have efficacy of more than 80%. Root hair infection experiments showed that F85 and T113 can inhibit early infection of root hairs, reduce the differentiation of primary plasmodia of P. brassicae, and inhibit formation of secondary zoosporangia. Based on sequence analysis of 16S rDNA gene, gyrA gene and 22 housekeeping genes as well as carbon source utilization analysis, the F85 was identified as Bacillus velezensis and T113 as Bacillus amyloliquefaciens.

  • Graphical abstract.The current study was performed to evaluate the antiproliferative and apoptosis-inducing potential of n-hexane fraction from Cassia fistula L. (Caesalpinioideae) fruits. The antiproliferative property of the fraction was determined by MTT assay against cancer cell lines including HeLa, MG-63, IMR-32, and PC-3 with GI50 value of 97.69, 155.2, 143, and 160.2 μg/ml respectively. The fraction was further explored for its apoptotic effect using confocal, SEM, and flow cytometry studies in HeLa cells. It was observed that the treatment of fraction revealed fragmentation of DNA, chromatin condensation, membrane blebbing, and formation of apoptotic bodies in a dose-dependent manner. The fraction also showed a remarkable increase in the level of ROS, mitochondrial depolarization and G0/G1 phase cell cycle arrest, and induction in the phosphatidylserine externalization analyzed using Annexin V-FITC/PI double staining assay in HeLa cells. Kaempferol, Ellagic acid, and Epicatechin are the major phytoconstituents present in the fraction as revealed by the HPLC. The treatment of n-hexane fraction showed downregulation in the gene expression of Bcl-2 and upregulation in the expression level of p53, Bad, and caspase-3 genes analyzed using semi-quantitative RT-PCR in HeLa cells. These results suggest that n-hexane fraction from C. fistula inhibited the proliferation of cervical cancer cells efficiently by the induction of apoptosis. We hypothesize that an optimum ratio of dairy manure to synthetic fertilizers leads to large nitrogen use efficiency (NUE) and small yield-scaled N2O emissions. The aims of this study were to (1) quantify the variations in the crop yields and N2O emissions from winter wheat-summer maize cropping systems in Northwest China, (2) determine the responses of the NUE and yield-scaled N2O emission to the ratio of organic materials to synthetic fertilizers, and (3) evaluate the relationship between the NUE and yield-scaled N2O emissions. Field measurements were conducted within long- and short-term fertilization experiments between the years of 2014 and 2016. Treatments included synthetic fertilizers, synthetic fertilizers plus crop residues, and synthetic fertilizers plus dairy manure at both sites. The annual grain yields and N2O emissions varied from 13.3 to 18.0 Mg ha-1 and from 1.3 to 3.6 kg N ha-1, respectively, across the treatments. The yield-scaled N2O emissions related negatively to the NUE, suggesting that agronomic aims of improving NUE are an effective approach to mitigate N2O emissions. The ratio of organic materials to synthetic fertilizers was not a significant limit on the NUE and yield-scaled N2O emissions. We conclude that organic amendments appeared to play a minor influence on the promotion of the NUE and N2O mitigation.Exploring fiscal reform's pollution effect draws increasing attention. Our paper brings a new perspective, i.e., the vertical fiscal imbalance (VFI) perspective, to study such issue. With the panel data of China's provincial level from 1999 to 2016, we reveal whether and how VFI affects environmental pollution. We find that VFI significantly enhances the environmental pollution level of China. As the VFI increases, environmental pollution effect caused by VFI appears to be aggravated. According to the transmission path check, we confirm in our paper that VFI can indirectly lead to environmental degradation by affecting the two channels environmental regulation and industrial transformation. Further, through the sub-sample regressions, we find that the impacts of VFI on environmental pollution as well as the transmission paths are regionally heterogeneous. Finally, we believe that using the VFI as a variable to explore fiscal reform's effect on environment is of rationality, and for a better environmental quality, VFI should be avoided.The deposition of toxic metals in the ecosystem contributes to the exposure and bioaccumulation of metals in the food chain, thus affecting human health. This study aimed to access the distribution of metal pollution emitted from automobiles in the dust, soil, and plant samples collected from the roadsides of national highways. Furthermore, metals were also determined in fuels and vehicular emissions. High contents of Pb, Cd, Cr, Cu, Fe, Mn, and Zn were found in roadsides as compared to control (35 km away from roads). The comparison among plants indicated that Calotropis procera and Rumex dentatus contained significantly higher metals than other plants. The concentrations of Pb, Cd, Cr, and Fe in plants were above the safe limits of the WHO/FAO. Significant and positive correlations were found between Cr, Cu, Fe, Pb, and Zn in the dust and soil samples and Cd and Cr in dust and plants. According to the results of the PCA analysis, all metals formed the first two components explaining 89.5% of the total variance. The source of these metals was attributed to automobile exhaust and dust depositions. The findings of the present study suggest that roadside plants are heavily infested with heavy metals due to vehicular smoke pollution, so the consumption of vegetation facing vehicular pollution may lead to certain physiological disorders and diseases.     Graphical abstract.The current study was performed to evaluate the antiproliferative and apoptosis-inducing potential of n-hexane fraction from Cassia fistula L. (Caesalpinioideae) fruits. The antiproliferative property of the fraction was determined by MTT assay against cancer cell lines including HeLa, MG-63, IMR-32, and PC-3 with GI50 value of 97.69, 155.2, 143, and 160.2 μg/ml respectively. The fraction was further explored for its apoptotic effect using confocal, SEM, and flow cytometry studies in HeLa cells. It was observed that the treatment of fraction revealed fragmentation of DNA, chromatin condensation, membrane blebbing, and formation of apoptotic bodies in a dose-dependent manner. The fraction also showed a remarkable increase in the level of ROS, mitochondrial depolarization and G0/G1 phase cell cycle arrest, and induction in the phosphatidylserine externalization analyzed using Annexin V-FITC/PI double staining assay in HeLa cells. Kaempferol, Ellagic acid, and Epicatechin are the major phytoconstituents present in the fraction as revealed by the HPLC. The treatment of n-hexane fraction showed downregulation in the gene expression of Bcl-2 and upregulation in the expression level of p53, Bad, and caspase-3 genes analyzed using semi-quantitative RT-PCR in HeLa cells. These results suggest that n-hexane fraction from C. fistula inhibited the proliferation of cervical cancer cells efficiently by the induction of apoptosis. We hypothesize that an optimum ratio of dairy manure to synthetic fertilizers leads to large nitrogen use efficiency (NUE) and small yield-scaled N2O emissions. The aims of this study were to (1) quantify the variations in the crop yields and N2O emissions from winter wheat-summer maize cropping systems in Northwest China, (2) determine the responses of the NUE and yield-scaled N2O emission to the ratio of organic materials to synthetic fertilizers, and (3) evaluate the relationship between the NUE and yield-scaled N2O emissions. Field measurements were conducted within long- and short-term fertilization experiments between the years of 2014 and 2016. Treatments included synthetic fertilizers, synthetic fertilizers plus crop residues, and synthetic fertilizers plus dairy manure at both sites. The annual grain yields and N2O emissions varied from 13.3 to 18.0 Mg ha-1 and from 1.3 to 3.6 kg N ha-1, respectively, across the treatments. The yield-scaled N2O emissions related negatively to the NUE, suggesting that agronomic aims of improving NUE are an effective approach to mitigate N2O emissions. The ratio of organic materials to synthetic fertilizers was not a significant limit on the NUE and yield-scaled N2O emissions. We conclude that organic amendments appeared to play a minor influence on the promotion of the NUE and N2O mitigation.Exploring fiscal reform's pollution effect draws increasing attention. Our paper brings a new perspective, i.e., the vertical fiscal imbalance (VFI) perspective, to study such issue. With the panel data of China's provincial level from 1999 to 2016, we reveal whether and how VFI affects environmental pollution. We find that VFI significantly enhances the environmental pollution level of China. As the VFI increases, environmental pollution effect caused by VFI appears to be aggravated. According to the transmission path check, we confirm in our paper that VFI can indirectly lead to environmental degradation by affecting the two channels environmental regulation and industrial transformation. Further, through the sub-sample regressions, we find that the impacts of VFI on environmental pollution as well as the transmission paths are regionally heterogeneous.
