A fluorescence aptasensor for the highly specific and sensitive determination of tetrodotoxin was established with tetrodotoxin-aptamer as the recognition unit, berberine as the signal reporter and exonuclease I as the elimination agent for the background. Berberine has a weak fluorescence emission at 540 nm, and it can form the tetrodotoxin-aptamer/berberine complex, resulted in an increased fluorescence. After introducing exonuclease I, it can degrade the single strand oligonucleotides of tetrodotoxin-aptamer into the single nucleotide in the absence of tetrodotoxin, which lead to dramatic fluorescence quenching, and reduce the background signal of sensing system. Once tetrodotoxin is in the presence, tetrodotoxin-aptamer is converted into the stable neck ring conformation, which resists the degradation of exonuclease I and provides a more rigid micro-environment for the excited state of berberine, and then the strong fluorescence is observed. Based on the above properties, an ultrasensitive label-free fluorescence aptasensor for tetrodotoxin is established. METHODS Prospective study of 823 smokers who reported a failed quit attempt in the last 12 months at baseline and ≥1 quit attempt over 6-month follow-up. The input variable was time in months between the end (and in an exploratory analysis, the start) of the most recent failed quit attempt reported retrospectively at baseline and start of the first attempt made during the 6-month follow-up period. RESULTS Success rates for failed quitters who waited less then 3, 3-6, and 6-12 months between their failed quit attempt ending and making a subsequent quit attempt were 13.8%, 17.5%, and 19.0% respectively. After adjustment for covariates, the odds of cessation relative to those who made a subsequent quit attempt within 3 months were 1.42 (95%CI 0.79-2.55) and 1.52 (95%CI 0.81-2.86) for those who waited 3-6 and 6-12 months respectively before trying again. Bayes factors indicated the data were insensitive. The exploratory analysis showed the odds of cessation were 1.55 (95%CI 0.78-3.08), 1.92 (95%CI 0.94-3.92), and 2.47 (95%CI 1.04-5.83) greater for those with an interval of 3-6, 6-12, and 12-18 months respectively than those who tried again within 3 months. CONCLUSIONS While pre-planned analyses were inconclusive, exploratory analysis of retrospective reports of quit attempts and success suggested the likelihood of success of quit attempts may be positively associated with number of months since beginning a prior quit attempt. However, only the longest inter-quit interval examined (12-18 months) was associated with significantly greater odds of quit success relative to a less then 3 month interval in fully adjusted models; all other comparisons were inconclusive. METHOD We drew the data (N = 1515) from the longitudinal multidisciplinary research program LoRDIA (Longitudinal Research on Development In Adolescence), of which 406 reported substance use and were included in the presented analyses. The data were collected via self-report questionnaires in classroom settings at three time points (mean age 13.5, 14.4 and 15.0). We obtained information for childhood maltreatment using the Childhood Trauma Questionnaire-Short Form (CTQ-SF) and data of frequencies of SURNC with a questionnaire scale. RESULTS Estimates from zero-inflated Poisson growth curve model revealed no baseline differences in SURNCs across children reporting none, single, or multiple maltreatment before the age of twelve. However, children experiencing multiple maltreatment displayed a greater increase in the frequency of SURNCs during the transition from early to mid adolescence than did those reporting no maltreatment. These estimates were only partly influenced by the inclusion of frequency of alcohol and substance drug use to the model. CONCLUSIONS These findings imply that children suffering maltreatment are at a higher risk of experiencing SURNCs, a factor known to elevate the risk of substance use disorders later in life, as they transition from early to mid adolescence. Previous literature has consistently reported socio-affective information processing impairments in patients with severe alcohol use disorder (SAUD). Some recent studies have also suggested that these patients might exhibit biases toward stimuli indicating social threat, such as angry or disgusted faces. Such biases have been largely documented in other psychopathological disorders like anxiety, where they play a critical role in the emergence and maintenance of the disorder. A comprehensive understanding of these biases in SAUD would thus deepen the understanding of interpersonal difficulties and relapse-related factors. However, to date, no study has directly explored these biases in SAUD. In order to initiate efforts to address this issue, we first review preliminary evidence supporting the hypothesis of biased processing of social threat in SAUD. Then, we identify possible pathways through which such biases might negatively impact the course of the disorder. Finally, we provide precise recommendations and available materials to develop research in this promising field, and underline the related theoretical and clinical perspectives. Tuberculosis Preventive Therapy (TPT) is widely used in particular among high-risk populations such as close contacts and immunosuppressed people mostly in high-income settings. TPT is widely recommended for high-risk populations including HIV-infected and household contacts globally, but is not widely used. Historical trials on risk groups as well as the general population have documented a marked effect on reductions in incidence of active disease among those treated, as well as on prevalence of latent TB infection (LTBI) in populations where massive roll-out of TPT has previously taken place. This review summarizes the results of large historical trials conducted more than 50 years ago among Inuit and African populations as well as risk groups in the USA and Europe exhibiting similarities with current high-burden populations with current limited use of TPT. The trials demonstrated a 27-95% reduction in incidence of active TB among those receiving preventive treatment compared with placebo, with efficacy depending somewhat on length of treatment but mostly on adherence rates.

To explore the role of Toll-like receptors (TLRs) and interferon (IFN) in the innate immunity against porcine epidemic diarrhea virus (PEDV), we detected the expression of TLR genes in PEDV-infected IPEC-J2 cells by real-time PCR. We also detected the level of interferon α (IFN-α) and interferon γ (IFN-γ) by enzyme-linked immunosorbent assay (ELISA). Results showed that IPEC-J2 cells exhibited a clear pathological change after PEDV infection at 24 h. https://www.selleckchem.com/products/gkt137831.html In addition, TLR7, TLR9 and TLR10 expressions were significantly upregulated in PEDV-infected IPEC-J2 cells at 24 h. Interestingly, the expression patterns of TLR2 and TLR4 were consistent at different stages of PEDV infection. The expression level of TLR3 decreased significantly with the increase of infection time, but the expression levels of TLR5 and TLR8 genes at 6 h and 12 h were significantly lower than those in the control group (p⟨0.01). There were significant correlations among the expression levels of TLR genes (p⟨0.05). Cytokine detection showed that the secretion level of IFN-α in the PEDV-infected group was significantly higher than that in the control group (p⟨0.01), and IFN-γ at 6 h and 12 h after PEDV infection was significantly higher than that in control group (p⟨0.01). Therefore, our results suggest that PEDV infection can induce innate immune responses in intestinal porcine jejunum epithelial cells, leading to changes in the expression of Toll-like receptors, and can regulate the resistance to virus infection by affecting the release levels of downstream cytokines. Intraperitoneal injection constitutes an alternative to inhalant anesthesia. PURPOSE The aim of this study was to identify a safe, reliable, and effective anesthesia and postoperative analgesia protocol for laboratory rats exposed to painful procedures. MATERIAL AND METHODS Twenty-seven female Wistar rats in an ongoing study that required surgery were randomized into groups for three different intraperitoneal anesthesia protocols and three different analgesia regimens. The anesthesia groups were (1) medetomidine + ketamine (MK), (2) ketamine + xylacine (KX), and (3) fentanyl + medetomidine (FM). Three analgesia groups were equally distributed among the anesthesia groups (1) local mepivacaine + oral ibuprofen (MI), (2) oral tramadol + oral ibuprofen (TI), and (3) local tramadol + oral tramadol + + oral ibuprofen (TTI). A core was assigned to measure anesthesia (0-3) and analgesia (0-2) effectiveness; the lower the score, the more effective the treatment. RESULTS The mean MK score was 0.44 versus 2.00 for FM and 2.33 for KX. Mean score for analgesia on the first postoperative day was TTI (4.66) TI (9.13), and MI (10.14). Mean score 48 hours after surgery was TTI (3.4), TI (6.71), and MI (9.5). These differences were statistically significant. CONCLUSION MK was shown to be a reliable, safe, and effective method of anesthesia. The TTI analgesia regimen is strongly recommended in light of these results. ETEC strains expressing F4 fimbriae are frequently associated with post-weaning diarrhea (PWD) and lead to great economic losses in ***** production industry worldwide. The aim of this study was to establish a rapid and effective isothermal amplification method for detection of F4 fimbriae. Loop-mediated isothermal amplification (LAMP), Polymerase spiral reaction (PSR) and cross-priming ampli- fication (CPA) were used to develop and optimize the detection method first time. Subsequently, the specificity and sensitivity of these methods were evaluated, and the clinical samples were detected with these methods. All the F4-positive samples could produce ladder-like amplifica- tions products and lead the chromogenic substrate SYBR Green I produce green fluorescence, while in blank control and negative samples lack of this pattern or remained orange. The sensi- tivity of LAMP and CPA were 10 times higher than PSR method. Meanwhile, these three methods were validated with clinical samples, 7 were found positive, while 125 samples were negative, the testing results were consisted with the real-time PCR method. These findings suggested that the isothermal amplification based on the F4 fimbriae is a rapid, effective and sensitive method under resource constrains. Among the tested essential oils Achillea millefolium, Eugenia caryophyllus and Citrus grandis were the most active (IC50 values of 51.0, 60.3 and 61.3 μg/mL, respectively). The oils from Abies sibirica, Rosmarinus officinalis, Eucalyptus globulus, Cinnamonum zeylanicum, Mentha piperita and Pinus sylvestris were less active (IC50 values of 134.3, 237.3, 239.3, 367.9, 837.5 and 907.3 μg/mL, respectively). The results support the concept that some essential oil constituents may be useful in the clinical management of babesiosis. Fifteen Rahmani growing male lambs (about 5 months old and 23.21±2.75 kg body weight) were randomly allo- cated to three equal groups consisting of 5 animals each. The animals in the first group, served as a control (group C), were fed a basal diet without any supplementation. The lambs in the second and third group were fed the basal diet supplemented with probiotic (group Y) or yeast culture (group YC), respectively. The probiotic consisted of live yeast (Saccharomyces cerevisae) alone, while the yeast culture was composed of Saccharomyces cerevisiae and the media on which it was grown. In group Y and YC, each lamb was supplemented daily with 0.5 g and 7.0 g of live yeast and yeast culture, respectively. Blood samples were collected before feeding the supplements and then every 15 days until the day 90th. Total and differential leucocytic counts, total protein, albumin, IgA, IgG and IgM levels were measured in blood.

in English, Russian Гастроэзофагеальная рефлюксная болезнь (ГЭРБ) возникает в результате рефлюкса содержимого желудка в пищевод, глотку, дыхательные пути и ротовую полость. В последние годы отмечен значительный рост заболеваемости, что оказывает выраженное негативное влияние на качество жизни населения. 'Золотым стандартом' лечения ГЭРБ является антисекреторная терапия - ингибиторы протонной помпы (ИПП), однако до 40% пациентов невосприимчивы к ИПП даже после коррекции дозы. Проанализированы преимущества и недостатки современных способов хирургического лечения ГЭРБ. Чаще всего выполняют следующие антирефлюксные операции лапароскопическую фундопликацию по Nissen (360°), Toupet (270°) или Dor (180°). Кроме того, применяют минимально инвазивные альтернативные методы хирургического лечения, такие как система управления рефлюксом LINX, электрическая стимуляция нижнего пищеводного сфинктера и трансоральная безразрезная фундопликация (transoral incisionless fundoplication), которые менее агрессивны и у которых выраженность побочных эффектов меньше. Пациент направлен на консультацию в РНЦХ им. акад. Б.В. Петровского. По результатам консилиума рекомендовано проведение оперативного вмешательства в два этапа эндопротезирование грудного отдела аорты и затем ревизия системы транспедикулярной фиксации и перемонтаж системы. 14.07.17 выполнено двухэтапное оперативное вмешательство. Послеоперационный период протекал гладко. in English, Russian Гастроэзофагеальная рефлюксная болезнь (ГЭРБ) возникает в результате рефлюкса содержимого желудка в пищевод, глотку, дыхательные пути и ротовую полость. В последние годы отмечен значительный рост заболеваемости, что оказывает выраженное негативное влияние на качество жизни населения. 'Золотым стандартом' лечения ГЭРБ является антисекреторная терапия - ингибиторы протонной помпы (ИПП), однако до 40% пациентов невосприимчивы к ИПП даже после коррекции дозы. Проанализированы преимущества и недостатки современных способов хирургического лечения ГЭРБ. Чаще всего выполняют следующие антирефлюксные операции лапароскопическую фундопликацию по Nissen (360°), Toupet (270°) или Dor (180°). Кроме того, применяют минимально инвазивные альтернативные методы хирургического лечения, такие как система управления рефлюксом LINX, электрическая стимуляция нижнего пищеводного сфинктера и трансоральная безразрезная фундопликация (transoral incisionless fundoplication), которые менее агрессивны и у которых выраженность побочных эффектов меньше. The results show that the thermally populated excited state contributions, while generally reducing the value of [capital Phi, Greek, macron] calculated on the assumption of a thermally isolated ground state, still confirm the room temperature detectability of this property for all ten studied complexes. Trends on the relative sign of dominant contributions are then discussed on the basis of a crystal field model electrostatic potential splitting a ground spin-orbit multiplet, which provides an insight into the properties of the generalized shielding polarizability tensor for open shell species.Metalloproteins are crucial to many biological processes, such as photosynthesis, respiration, and efficient electron transport. Zinc is the most common transition metal found in proteins and is critical for structure, function and stability, however the effects from the electronic properties of a bound zinc ion on electron transfer are not clearly defined. Here, a series of β-strand and 310-helical peptides, capable of binding Zn2+ via suitably positioned His residues, was synthesized and their ability to undergo electron transfer in the presence and absence of Zn2+ studied by electrochemical and computational means. The β-strand peptide was shown to be conformationally pre-organized, with this geometry maintained on complexation with zinc. Electrochemical studies show a significant increase in charge transport, following binding of the zinc ion to the β-strand peptide. In contrast, complexation of zinc to the helical peptide disrupts the intramolecular hydrogen bonding network known to facilitate electron transfer and leads to a loss of secondary structure, resulting in a decrease in charge transfer. These experimental and computational studies reveal an interplay, which demonstrates that bound zinc enhances charge transfer by changing the electronic properties of the peptide, and not simply by influencing secondary structure.We study the relation between the macroscopic viscoelastic properties of aqueous hyaluronan polymer solutions and the molecular-scale dynamics of water using rheology measurements, differential dynamic microscopy, and polarization-resolved infrared pump-probe spectroscopy. We observe that the addition of hyaluronan to water leads to a slowing down of the reorientation of a fraction of the water molecules. Near pH 2.4, the viscosity of the hyaluronan solution reaches a maximum, while the number of slowed down water molecules reaches a minimum. This implies that the water molecules become on average more mobile when the solution becomes more viscous. This observation indicates that the increase in viscosity involves the expulsion of hydration water from the surfaces of the hyaluronan polymers, and a bundling of the hyaluronan polymer chains.The steady state fluorescence anisotropy of carbon dot solutions of different viscosities η and its variation with temperature T has been investigated. The dependence of the anisotropy on T/η is shown to be described by the Perrin equation, which implies that Brownian rotational motion of carbon dots in solution is a basic mechanism of fluorescence depolarization. Peculiarities of the Perrin plot testify that the luminous entity ("fluorophore") responsible for carbon dot fluorescence displays noticeable segmental motions, which are independent of the overall rotational diffusion of the dots. The Perrin model fit to the experimental data yields the effective volumes of the fluorophore VF = 0.35 ± 0.15 nm3 and of the carbon dot as a whole VC = 10.5 ± 1.8 nm3. The rotational motions of the fluorophores are shown to be limited and spectrally dependent. In the Cr2I3Br3 (Cr2I3Cl3) bilayers, the electrostatic potential and electric polarization can be greatly affected by combination modes, which can be attributed to the parallel or antiparallel built-in electric fields between the monolayers. The Cr2I3X3 bilayers show a perpendicular magnetic anisotropy. The magnetic anisotropy energy of the Cr2I3Cl3 bilayer is larger than that of the Cr2I3Br3 bilayer, which can be attributed to the enhanced contribution of the hybridized I px and py orbitals of the Cr2I3Cl3 bilayer. Additionally, the Dzyaloshinskii-Moriya interaction of the Cr2I3Br3 bilayer can also be modulated by the combination modes. Here in our study, the effects of TLR2 antagonism on the neural damage elicited by CIH were examined. METHODS Ortho-vanillin (O-vanillin) is an inhibitor of TLR2. Adult male **** were subjected to 8 h of intermittent hypoxia per day with or without O-vanillin for 28 days. Neuronal damage, the number of microglia, the interaction of TLR2 with its adapter protein myeloid differentiation factor 88 (MYD88), the expressions of inflammatory cytokines, and the oxidative stress were observed. RESULTS O-vanillin inhibited the increased interaction of TLR2 and MyD88, the activation of NFκB, the aggregation of microglia, the overexpression of proinflammatory agents, the elevation of oxidative stress, and hippocampal neuron cell apoptosis induced by CIH. CONCLUSIONS Our experiments indicate that TLR2 antagonism may alleviate the hippocampal neuronal damage caused by CIH via inhibiting neuroinflammation and oxidative stress.Seasonality of respiratory syncytial virus (RSV) infection is an area of limited knowledge. We retrospectively evaluated medical files of 512 hospitalizations in children due to RSV infection from January 2010 to July 2017. In this cohort of patients, there were 96.3% of children below 1 year of age; the median age was 2.8 months. The influence of weather condition during the week of hospitalization (WH) and also the preceding week (WP) on the rate of hospitalizations was also assessed. An overview of morbidity data demonstrates that the epidemic RSV season started from Week 50 of a year and lasts until Week 15 of the following year, with a peak between Week 4 and Week 10. Only 10-20% of patients are diagnosed at the early stage of disease. Recently, the role of novel biomarkers of the neoplastic process in the early detection of colorectal cancer has been widely discussed. In this review, we focused on the three novel biomarkers that are of potential clinical importance in diagnosing and monitoring colorectal cancer. Chitinase 3-like 1 protein, also known as YKL-40, and nestin and testin proteins are produced by colorectal cancer cells. YKL-40 protein is a marker of proliferation, differentiation, and tissue morphogenetic changes. The level of YKL-40 is elevated in about 20% of patients with colorectal cancer. An increased expression of nestin indicates immaturity. It is a marker of angiogenesis in neoplastic processes. Testin protein is a component of cell-cell connections and focal adhesions. The protein is produced in normal human tissues, but not in tumor tissues. Downregulation of testin increases cell motility, spread, and proliferation, and decreases apoptosis. The usefulness and role of these biomarkers, both alone and combined, in the diagnostics of colorectal cancer should be further explored as early cancer detection may substantially improve treatment outcome and patient survival.Practical work in veterinary medicine provides exposure to various allergens which could increase the risk of sensitization. With the ongoing prospective "AllergoVet" study, we are investigating patterns of allergic symptoms and sensitization among veterinary students. Here, we report the results of the introductory self-reported survey on the presence of allergies in the first-year students of veterinary medicine, who had begun their studies in the years 2013-2016. The survey included 553 students who answered a questionnaire (63% response rate, 83% female ratio). The median age was 20 years (IQR 19-22 years). About half of the responders indicated an interest in participating in the prospective study ("candidates"). Those who were uninterested in participating were termed "noncandidates". A doctor's diagnosis of any kind of atopic disease was reported by every fourth student. Hay fever was reported by 71 (13%) and allergic asthma by 38 (6.9%) students. The prevalence of hay fever in "candidates" (n = 294) and "noncandidates" (n = 259) was 13.5% and 12.2%, respectively. Allergic asthma was reported by 9.9% of "candidates" and 3.5% of "noncandidates", the difference being significant (p = 0.003). We conclude that the prevalence of self-reported allergic symptoms in the first-year students of veterinary medicine is similar to that in the general population. Pre-existing allergic asthma may have increased the motivation for the enrolment into the longitudinal "AllergoVet-study".

YKL-40 protein is a marker of proliferation, differentiation, and tissue morphogenetic changes. The level of YKL-40 is elevated in about 20% of patients with colorectal cancer. An increased expression of nestin indicates immaturity. It is a marker of angiogenesis in neoplastic processes. Testin protein is a component of cell-cell connections and focal adhesions. With the ongoing prospective "AllergoVet" study, we are investigating patterns of allergic symptoms and sensitization among veterinary students. Here, we report the results of the introductory self-reported survey on the presence of allergies in the first-year students of veterinary medicine, who had begun their studies in the years 2013-2016. The survey included 553 students who answered a questionnaire (63% response rate, 83% female ratio). The median age was 20 years (IQR 19-22 years). About half of the responders indicated an interest in participating in the prospective study ("candidates"). Those who were uninterested in participating were termed "noncandidates". A doctor's diagnosis of any kind of atopic disease was reported by every fourth student. Hay fever was reported by 71 (13%) and allergic asthma by 38 (6.9%) students. The prevalence of hay fever in "candidates" (n = 294) and "noncandidates" (n = 259) was 13.5% and 12.2%, respectively. Allergic asthma was reported by 9.9% of "candidates" and 3.5% of "noncandidates", the difference being significant (p = 0.003). We conclude that the prevalence of self-reported allergic symptoms in the first-year students of veterinary medicine is similar to that in the general population. Venous abnormalities have long been observed on pathology characterized by irregular narrowing and dilatation with associated venous wall and perivenous infiltrates. Active CNS lesions areCNS venous structures. Characteristic CNS lesions are a secondary manifestation resulting from an inflammatory response to the uncovering of usually privileged CNS antigens. This study aimed to investigate whether the individuals diagnosed with Attention-deficit Hyperactivity Disorder differ from healthy individuals in terms of facial anthropometric measurements. Forty children, diagnosed with Attention-deficit Hyperactivity Disorder, were included in the study as the case group, and forty healthy children were included in the study as the control group. Two photographs were taken from the facial region, and anthropometric measurements were performed using the computer program "Image J" in the computer environment. It was found that a strong relationship between Attention-deficit Hyperactivity Disorder and nasal width, ear length and upper face debt length. The results obtained from the research support the knowledge that there is a close relationship between the forebrain development process and the facial development process during the embryonic development process. Evaluation of the toxicity of Cu/CuO NPs was performed on 2 types of cells human lung normal cell lines (WI-38) and human lung carcinoma cell (A549). To assess the toxicity of the prepared Cu/CuOs NPs, the two cell types were exposed to Cu/CuO NPs for 72 h. The half-maximal inhibitory concentration IC50 of Cu/CuO NPs for both cell types was separately determined and used to examine the cell genotoxicity concurrently with the determination of some oxidative stress parameters nitric oxide, glutathione reduced, hydrogen peroxide, malondialdehyde and superoxide dismutase. RESULTS Cu/CuO NPs suppressed proliferation and viability of normal and carcinoma lung cells. Treatment of both cell types with their IC50's of Cu/CuO NPs resulted in DNA damage besides the generation of reactive oxygen species and consequently the generation of a state of oxidative stress. CONCLUSION Overall, it can be concluded that the IC50's of the prepared Cu/CuO NPs were cytotoxic and genotoxic to both normal and cancerous lung cells. OBJECTIVE Racial differences in BMI increase with education. Weight perception may be an important factor in overweight and obesity in black women. The aim of this study was to determine the mediating role of weight underassessment on race differences in BMI in college graduates compared with non-college graduates. METHODS Weight perception was assessed among respondents to the 2007-2014 National Health and Nutrition Examination Survey (n = 4,871). Those who had BMI-assessed overweight or obesity and self-assessed underweight or about-right weight underassessed their weight. The associations between race and BMI through weight underassessment by college graduate status were determined using a moderated mediation analysis. RESULTS Black women had higher BMI than white women (β = 2.72, SE = 0.28), and disparities were larger in college graduates (β = 3.50, SE = 0.25) compared with non-college graduates (β = 0.78, SE = 0.15). Non-college graduate black women were more likely to underassess their weight (z score = 0.43, SE = 0.05). Indirect associations between race and BMI through weight underassessment were found only among non-college graduates (z score = -0.02, SE = 0.01), but race differences in BMI remained after accounting for weight perception among college graduates and non-college graduates. Eight weeks after the diabetes was induced, the efficacy of DKK3 determined by three independent experiments experiment 1 (DKK3 peptide [5 μg in 20 μL PBS); experiment 2 (DKK3 plasmid DNA with electroporation [10 μg, 40 μg, or 100 μg in 20 μL PBS, respectively]; and experiment 3 (DKK3 adenovirus [1 × 107 , 1 × 108 , 1 × 109 virus particles per 20 μL, respectively]). Erectile function was measured by electrical stimulation of the cavernous nerve one week (for peptide) or two weeks (for genes) after treatment. OBJECTIVES To examine whether and how Dickkopf3 (DKK3), a secreted modulator of the Wnt pathway that known to be involved in endothelial cell repair and vascular progenitor cell migration, restores erectile function in diabetic mice. METHODS Eight-week-old C57BL/6 mice were received intraperitoneal injections of streptozotocin (50 mg/kg for 5 days). Eight weeks after the diabetes was induced, the efficacy of DKK3 determined by three independent experiments experiment 1 (DKK3 peptide [5 μg in 20 μL PBS); experiment 2 (DKK3 plasmid DNA with electroporation [10 μg, 40 μg, or 100 μg in 20 μL PBS, respectively]; and experiment 3 (DKK3 adenovirus [1 × 107 , 1 × 108 , 1 × 109 virus particles per 20 μL, respectively]). Erectile function was measured by electrical stimulation of the cavernous nerve one week (for peptide) or two weeks (for genes) after treatment.
