  • The MRE11-RAD50-NBS1 (MRN) complex supports the synthesis of damage-induced long non-coding RNA (dilncRNA) by RNA polymerase II (RNAPII) from DNA double-strand breaks (DSBs) by an unknown mechanism. Here, we show that recombinant human MRN and native RNAPII are sufficient to reconstitute a minimal functional transcriptional apparatus at DSBs. MRN recruits and stabilizes RNAPII at DSBs. Unexpectedly, transcription is promoted independently from MRN nuclease activities. Rather, transcription depends on the ability of MRN to melt DNA ends, as shown by the use of MRN mutants and specific allosteric inhibitors. Single-molecule FRET assays with wild-type and mutant MRN show a tight correlation between the ability to melt DNA ends and to promote transcription. The addition of RPA enhances MRN-mediated transcription, and unpaired DNA ends allow MRN-independent transcription by RNAPII. RNA sequencing analysis demonstrates that the retinoic-acid-inducible gene (RIG)-I-like receptor (RLR) and the Toll-like receptor (TLR) signaling pathways are significantly compromised by simulated microgravity (Sμg). TRIM25, an essential E3 for RLR signaling, is inhibited under Sμg, hampering the K63-linked ubiquitination of RIG-I and the following function-induction positive feedback loop of antiviral immune response. These mechanisms provide insights into better understanding of the effects and principles of microgravity on host antiviral immunity and present broad potential implications for developing strategies that can prevent and control viral diseases during space flight.The cyclic GMP-AMP (cGAMP) synthase (cGAS) is a key DNA sensor that initiates STING-dependent signaling to produce type I interferons through synthesizing the secondary messenger 2'3'-cGAMP. In this study, we confirm previous studies showing that cGAS is located both in the cytoplasm and in the nucleus. Nuclear accumulation is observed when leptomycin B is used to block the exportin, CRM1 protein. As a result, leptomycin B impairs the production of interferons in response to DNA stimulation. We further identify a functional nuclear export signal (NES) in cGAS, 169LEKLKL174. Mutating this NES leads to the sequestration of cGAS within the nucleus and the loss of interferon response to cytosolic DNA treatment, and it further determines the key amino acid to L172. Collectively, our data demonstrate that the cytosolic DNA-sensing function of cGAS depends on its presence within the cytoplasm, which is warranted by a functional NES.Ribosome-associated quality control (RQC) relieves stalled ribosomes and eliminates potentially toxic nascent polypeptide chains (NCs) that can cause neurodegeneration. We demonstrate that NEMF, a mammalian RQC2 homolog, modifies translation products of nonstop mRNAs, major erroneous mRNAs in mammals, with a C-terminal tail mainly composed of alanine with several other amino acids. Overproduction of nonstop mRNAs induces NC aggregation and caspase-3-dependent apoptosis and impairs neuronal morphogenesis, which are ameliorated by NEMF depletion. Moreover, we found that homopolymeric alanine tailing at least partially accounts for CAT-tail cytotoxicity. These findings explain the cytotoxicity of CAT-tailed NCs and demonstrate physiological significance of RQC on proper neuronal morphogenesis and cell survival.Although various long noncoding RNAs (lncRNAs) are specifically expressed in activated macrophages, their in vivo functions and mechanisms of action are largely unexplored. In mechanisms, linc-AAM interacts with heterogeneous nuclear ribonucleoprotein L (hnRNPL) via two CACACA motifs, resulting in its dissociation from histone H3 to activate chromatin and facilitate transcription of IRGs. Ogt is highly expressed in HSPCs, and its disruption induces rapid loss of HSPCs with increased reactive oxygen species and apoptosis. In particular, Ogt-deficient hematopoietic stem cells (HSCs) lose quiescence, cannot be maintained in vivo, and become vulnerable to regenerative and competitive stress. Interestingly, Ogt-deficient HSCs accumulate defective mitochondria due to impaired mitophagy with decreased key mitophagy regulator, Pink1, through dysregulation of H3K4me3. Furthermore, overexpression of PINK1 restores mitophagy and the number of Ogt-deficient HSCs. Collectively, our results reveal that OGT critically regulates maintenance and stress response of HSCs by ensuring mitochondrial quality through PINK1-dependent mitophagy.

No other fall-**** good agricultural practices (GAPs) were received, and thus, a lowering of the MRLs for peaches, apples, pears and broccoli is proposed.Regulation (EC) No 396/2005 establishes the rules governing the setting and the review of pesticide maximum residue levels (MRLs) at European level.     We explore the spectrum of comorbidities in psoriatic arthritis (PsA) patients in comparison with other high comorbidity-burden diseases like rheumatoid arthritis (RA) and diabetes mellitus (DM).

    Two hundred and fifteen PsA patients, cross-sectionally collected from two tertiary hospitals, were compared with 215 RA and 215 DM patients (age/sex-matched, similar disease duration). Cardiovascular risk factors [hypertension, current smoking, hyperlipidaemia, obesity (body mass index (BMI) ⩾30)], coronary artery disease (***), stroke, major adverse cardiac events (MACEs; combined *** and stroke), depression, osteoporosis and malignancies were recorded. Odds ratios (ORs) for stroke, *** and **** were adjusted for age, sex, hypertension, smoking, hyperlipidaemia, BMI, glucocorticoids use and those for depression were adjusted for age, sex, disease duration, skin involvement and smoking. Within the PsA group, associations between comorbidities and demographic/clinical features were assessed.

SpA associated with IBD presents with neutrophilic and granulomatous skin disorders, including pyoderma gangrenosum, hidradenitis suppurativa and cutaneous Crohn's disease. Reactive arthritides has a favourable prognosis and may feature keratoderma blenorrhagicum or balanitis circinatum as typical skin manifestations. Immunologically, SpA-associated skin diseases share interleukin (IL)-17 and IL-23 dysregulation but show distinctive genetic and immunological profiles. Therefore, they vary in their treatment responses to targeted therapies with biologicals or small molecules. In this review, we highlight the clinical presentation of skin manifestations in SpA and discuss therapeutic approaches in this interdisciplinary field.

  • In the dry season, most large herbivores--a majority of which are endangered and threatened by anthropogenic pressures--avoided areas with a strong human footprint, while preferring spaces with high land-use diversity. During the floods, such species were pushed out of inundated habitats within the protected area, and they chose to move through woodlands and areas under bamboo cover on private lands, as they migrated to forested refugia on higher ground. Our results show how seasonal environmental constraints shaped by floods determine the internal motivation of animals to risk traversing a human-dominated space to seek refuge, which contextually defines how animals view and navigate the landscape. Such insights underscore the importance of dynamic and adaptive planning, and participatory conservation efforts, to facilitate connectivity in the changing environment and climate of the present Anthropocene.Flooding and other sediment disturbances can lead to increases in sediment resuspension. In this context, it is of central importance to understand the kinetics of release from these sediments and the uptake of pollutants, such as polychlorinated biphenyls (PCBs) and polychlorinated dioxins and furans (PCDD/Fs), into aquatic organisms. In the present study, we parameterized a sediment desorption model based on experimentally determined rapidly-desorbing fractions of dioxin-like chemicals (DLCs). We coupled this desorption model with a physiologically-based toxicokinetic model for rainbow trout. This combined model was used to predict DLC concentrations in the muscle of exposed fish. The performance of this model was evaluated using a previously published dataset on DLC uptake from sediment suspensions during simulated re-suspension events. Predictions generally differed less than 10-fold from measured values, and the model showed a good global coefficient of determination (R2) of 0.95. The root mean squared error (RMSE) for PCBs was 0.31 log units and 0.53 log units for PCDD/Fs. The results of our study demonstrate that the prediction of bioconcentration and related risk to fish resulting from sediment resuspension can be accurately predicted using coupled desorption and toxicokinetic models.Industrialization and urbanization have aggravated the contradiction between environmental protection and economic growth, leading to health issues.     In the dry season, most large herbivores--a majority of which are endangered and threatened by anthropogenic pressures--avoided areas with a strong human footprint, while preferring spaces with high land-use diversity. During the floods, such species were pushed out of inundated habitats within the protected area, and they chose to move through woodlands and areas under bamboo cover on private lands, as they migrated to forested refugia on higher ground. Our results show how seasonal environmental constraints shaped by floods determine the internal motivation of animals to risk traversing a human-dominated space to seek refuge, which contextually defines how animals view and navigate the landscape. In the present study, we parameterized a sediment desorption model based on experimentally determined rapidly-desorbing fractions of dioxin-like chemicals (DLCs). We coupled this desorption model with a physiologically-based toxicokinetic model for rainbow trout. This combined model was used to predict DLC concentrations in the muscle of exposed fish. The performance of this model was evaluated using a previously published dataset on DLC uptake from sediment suspensions during simulated re-suspension events. Predictions generally differed less than 10-fold from measured values, and the model showed a good global coefficient of determination (R2) of 0.95. The root mean squared error (RMSE) for PCBs was 0.31 log units and 0.53 log units for PCDD/Fs. The results of our study demonstrate that the prediction of bioconcentration and related risk to fish resulting from sediment resuspension can be accurately predicted using coupled desorption and toxicokinetic models.Industrialization and urbanization have aggravated the contradiction between environmental protection and economic growth, leading to health issues. Leucocyte-reduced platelet apheresis (LRPA) can reduce the risk of platelet refractoriness and febrile nonhemolytic transfusion reactions (FNHTRs) for transfusion. Accordingly, this study aimed to investigate and compare the platelet metabolic and functional responses between PA performed with Haemonetics and LRPA performed with Trima Accel cell separator.

    The qualities of platelets collected through PA and LRPA were evaluated in terms of visual appearance, morphology, platelet-aggregation changes, metabolic activities, and bacterium-screening test during 5-day storage.     Croatian Institute of Transfusion Medicine (CITM) implemented non-invasive fetal RHD genotyping as a request for targeted antenatal anti-D prophylaxis. The diagnostic performance of in-house RT-PCR method for fetal RHD genotyping and preliminary results are analyzed.

  • Measuring conditional dependence is an important topic in econometrics with broad applications including graphical models. Under a factor model setting, a new conditional dependence measure based on projection is proposed. The corresponding conditional independence test is developed with the asymptotic null distribution unveiled where the number of factors could be high-dimensional. It is also shown that the new test has control over the asymptotic type I error and can be calculated efficiently. A generic method for building dependency graphs without Gaussian assumption using the new test is elaborated. We show the superiority of the new method, implemented in the R package pgraph, through simulation and real data studies.Mucin-type O-glycosylation is one of the most common post-translational modifications of proteins. In humans there are 20 isoforms that are differentially expressed across tissues that serve multiple important biological roles. Using random peptide substrates, isoform specific amino acid preferences have been obtained in the form of enhancement values (EV). These EVs alone have previously been used to predict O-glycosylation sites via the web based ISOGlyP (Isoform Specific O-Glycosylation Prediction) tool. Here we explore additional protein features to determine whether these can complement the random peptide derived enhancement values and increase the predictive power of ISOGlyP. Detecting the stable boundary-layer top depends on the method used (flux- vs. anisotropy-profiles) but both methods are highly correlated. The top of the stable boundary layer, however, mostly deviates from the jet maximum height or the top of the near-surface inversion. The flat-terrain formulations for the boundary-layer height correlate well with the detected top of the stable boundary layer if the near-surface and not the background stratification is used in their formulations; however, they mostly largely overestimate this boundary-layer height. The difference from flat-terrain boundary layers is also shown through the dependence of size of the dominant eddy with height. In katabatic flows the eddy size is semi-constant with height throughout the stable boundary-layer depth, whereas in flat terrain, eddy size varies significantly with height. Analysis of the excess Doppler estimation, retrieved bending angles and impact parameters also confirms the improved performances of the proposed algorithm over OL tracking.This paper analyzes the effects of permanently unbalanced sex ratios in Germany caused by World War II on fertility outcomes over the life cycle. Using Census records linked with individual biography data, our analysis confirms the commonly found short-term pattern of decreased fertility rates due to a stark imbalance of the sex ratio. Yet, the long-term effects of such an imbalance crucially depend on when in the life cycle fertility is evaluated. We find that female cohorts with low sex ratios have fewer children at younger ages and a larger fraction remains childless. While childlessness remains higher throughout their life cycle, mothers from affected cohorts catch up and even overcompensate at later ages with respect to the number of children. Our preferred reading of this result is that with low sex ratios women select themselves into late motherhood according to their fertility preferences. This interpretation is consistent with the finding that women from affected cohorts expand their childbearing period and accept lower quality matches in the marriage market.

Under a factor model setting, a new conditional dependence measure based on projection is proposed. The corresponding conditional independence test is developed with the asymptotic null distribution unveiled where the number of factors could be high-dimensional. It is also shown that the new test has control over the asymptotic type I error and can be calculated efficiently. A generic method for building dependency graphs without Gaussian assumption using the new test is elaborated. The flat-terrain formulations for the boundary-layer height correlate well with the detected top of the stable boundary layer if the near-surface and not the background stratification is used in their formulations; however, they mostly largely overestimate this boundary-layer height. The difference from flat-terrain boundary layers is also shown through the dependence of size of the dominant eddy with height. In katabatic flows the eddy size is semi-constant with height throughout the stable boundary-layer depth, whereas in flat terrain, eddy size varies significantly with height. Using Census records linked with individual biography data, our analysis confirms the commonly found short-term pattern of decreased fertility rates due to a stark imbalance of the sex ratio. Yet, the long-term effects of such an imbalance crucially depend on when in the life cycle fertility is evaluated. We find that female cohorts with low sex ratios have fewer children at younger ages and a larger fraction remains childless. While childlessness remains higher throughout their life cycle, mothers from affected cohorts catch up and even overcompensate at later ages with respect to the number of children. Our preferred reading of this result is that with low sex ratios women select themselves into late motherhood according to their fertility preferences. This interpretation is consistent with the finding that women from affected cohorts expand their childbearing period and accept lower quality matches in the marriage market.

Nonetheless, the toxicity change related to transformation of microplastics has not been considered in most published studies thus far. In the current study, we investigated the cytotoxicity of transformed polystyrene microplastics in hepatocytes. Our results revealed that 500 nm polystyrene microplastics, which were chemically transformed by simulated gastricfluid, exacerbated their adverse effects on SMMC-7721 cells at 20 μg/mL for 24 h treatment, including morphological alteration, membrane damage and increased cell apoptosis via oxidative stress. This exacerbated cytotoxicity could be at least partially explained by the degradation, changed surface charge and altered surface chemistry of these polystyrene microplastics after transformation. This study surveyed buildings in an area of deteriorated dwellings targeted for renewal and determined the status and distribution of ACMs in that area. Asbestos distribution maps were generated and asbestos characteristics were analyzed. In addition, the risk posed by the identified ACMs was assessed using four international methods (the Korean Ministry of Environment, US Environmental Protection Agency, American Society for Testing and Materials, and UK Health and Safety Executive methods), and the results were compared. Notable differences between the assessment results were identified and were found to reflect the specific characteristics of buildings in the study area. As a result, 21 CuAl@p-CN possesses appropriate Cu-O-C configuration, has favorable electron transfer environment for rapid redox behavior of Cu(II)/Cu(I), leading to excellent PMS activation for mirco-polluted veterinary antibiotics elimination.

First, an exploratory qualitative study collected the experiences and views of DM specialist nurses regarding the use of mobile apps in clinical practice. Second, a CDSS tool was developed for the management of patients with DM and CKD. Finally, a randomized controlled trial examined the acceptability and impact of the tool.

First, women are largely responsible for household chores-including cleaning water storage containers-with men as assistants. Second, men described their role in the family as the protector. Meta-analyses were undertaken to determine the effects of higher fibre intake on all-cause and cardiovascular mortality and increasing fibre intake on glycaemic control and a range of cardiometabolic risk factors. For trials, meta regression analyses identified further variables that influenced the pooled findings. Dose response testing was undertaken; Grading of Recommendations Assessment, Development avements in measures of glycaemic control, blood lipids, body weight, and inflammation, as well as a reduction in premature mortality. Second, we explored to what extent behavioural, socioeconomic, and environmental factors explain HIV prevalence at the int, among young adults, micro-epidemics of relatively high HIV prevalence alternate with areas of very low prevalence, clearly illustrating the existence of areas with high levels of transmission. These areas are partially characterised by high economic activity, relatively high socioeconomic status, and risky sexual behaviour. Localised HIV prevention interventions specifically tailored to the populations at risk will be essential to curb transmission. More fine-scale geospatial mapping of key populations,-such as sex workers and migrant populations-could help us further understand the drivers of these areas with high levels of transmission and help us determine how they fuel the generalised epidemics in SSA.
    Study findings uncovered three ways in which gender may influence Zika prevention. First, women are largely responsible for household chores-including cleaning water storage containers-with men as assistants. Second, men described their role in the family as the protector.     Despite procedural skills being recognized as an important component of medical school education, medical students are not confident in their ability to carry out a range of medical procedures. We conducted an institutional needs assessment and used the results to inform the creation of a procedure-based preclinical elective for first- and second-year students.

    We surveyed second-, third-, and fourth-year medical students at Alpert Medical School as well as select program directors to guide selection of a list of procedures to be taught in the elective. We then created an extracurricular 10-week procedural skills course for preclerkship medical students utilizing a hands-on, flipped classroom practice model. Volunteer preceptors were recruited from the Department of Emergency Medicine to participate with a student-to-faculty ratio not exceeding 51. Knowledge and skill acquisition were assessed using a multiple-choice knowledge exam and 4-station practical exam, respectively. Pre- and post-course online sukship procedural course early during medical school is a feasible method of teaching procedural skills to a cohort of learners. A number of adjustments could be made to the course in order to scale up and include a larger cohort of students at our own or another institution.

We explored the feasibility of the Resident Communication Assessment Program (ReCAP) for eliciting patient feedback about resident physician communication in the emergency department (ED).

    This study is a prospective, observational study conducted in the ED of a university-based hospital from December 2018 through April 2019. ReCAP is a program that interviews patients prior to discharge from the ED using the Communication Assessment Tool (CAT). CAT consists of 14 Likert style questions and 3 open-ended questions for patient feedback aboutresidents' communication. Open-text, narrative responses from patients were coded using a modified version of the Completed Clinical Evaluation Report Rating tool.

    We collected data from 42 subjects who completed the CAT, and provided 32 open-text, narrative responses about 20 resident physicians. Patient responses were overwhelmingly positive with 551/588 (94%) CAT responses scoring "Very Good," the highest category. Open-text, narrative comments analyzed using CCERR were unbalanced, favoring residents' strengths rather than areas for improvement. Patient comments offered more examples of strengths than weaknesses, and few subjects provided recommendations to improve resident performance.

    ReCAP represents a feasible method for eliciting patient feedback about resident communication skills in the ED.
