  • Timely detection of liver fibrosis by X-ray computed tomography (CT) can prevent its progression to fatal liver diseases. However, it remains quite challenging because conventional CT can only identify the difference in density instead of X-ray attenuation characteristics. Spectral CT can generate monochromatic imaging to specify X-ray attenuation characteristics of the scanned matter. The reproducible on-chip generation of 360 spheroids of five dimensions on a well-plate format using an integrated microlens technology is demonstrated. While spheroid size-related IC50 values vary up to 160% using the anticancer drugs cisplatin (CIS) or doxorubicin (DOX), reduced CISDOX drug dose combinations eliminate all lung microtumors independent of their sizes. A further application includes optimizing cell seeding ratios and size-dependent compound uptake studies in a perfused BBB model. Generally, smaller BBB-spheroids reveal an 80% higher compound penetration than larger spheroids while verifying the BBB opening effect of mannitol and a spheroid size-related modulation on paracellular transport properties.Manipulation of Ohmic contacts in 2D transition metal dichalcogenides for enhancing the transport properties and enabling its application as a practical device has been a long-sought goal. In this study, n-type tungsten disulfide (WS2 ) single atomic layer to improve the Ohmic contacts of the p-type molybdenum ditelluride (MoTe2 ) material is covered. The Ohmic properties, based on the lowering of Schottky barrier height (SBH) owing to the tunneling barrier effect of the WS2 monolayer, are found to be unexpectedly excellent at room temperature and even at 100 K. The improved SBH and contact resistances are 3 meV and 1 MΩ µm, respectively. The reduction in SBH and contact resistance is confirmed with temperature-dependent transport measurements. This study further demonstrates the selective carrier transport across the MoTe2 and WS2 layers by modulating the applied gate voltage. This WS2 /MoTe2 heterostructure exhibits excellent gate control over the currents of both channels (n-type and p-type). The on/off ratios for both the electron and hole channels are calculated as 107 and 106 , respectively, indicating good carrier type modulation by the electric field of the gate electrode.

  • Comprehensive data is key for evidence-informed policy aiming to improve the lives of persons experiencing different levels of disability. The objective of this paper was to identify the environmental barriers - including physical, social, attitudinal, and political barriers - that might become priorities for cross-cutting policies and policies tailored to the needs of persons experiencing severe disability in Cameroon.

    A secondary analysis of data obtained with the WHO Model Disability Survey was completed in the Bankim Health District (N = 559) using random forest regression to determine and compare the impact of the environmental factors on the experience of disability.

    The physical environment had by far the highest influence on disability, with transportation, toilet of the dwelling, and the dwelling itself being the most important factors. Factors inside one's own home (toilet of the dwelling, and the dwelling itself) were the most important for persons with moderate and severe disability, followed by attitudes of others and issues with accessing health care.

    Our study provides country policy makers with evidence for setting priorities and for the development of evidence-informed policies for the Bankim Health District in Cameroon.

    Recent efforts in the field of nutritional science have allowed the discovery of disease-beating molecules within foods based on the commonality of bioactive food molecules to FDA-approved drugs. The pioneering work in this field used an unsupervised network propagation algorithm to learn the systemic-wide effect on the human interactome of 1962 FDA-approved drugs and a supervised algorithm to predict anticancer therapeutics using the learned representations. Then, a set of bioactive molecules within foods was fed into the model, which predicted molecules with cancer-beating potential.The employed methodology consisted of disjoint unsupervised feature generation and classification tasks, which can result in sub-optimal learned drug representations with respect to the classification task. Additionally, due to the disjoint nature of the tasks, the employed approach proved cumbersome to optimize, requiring testing of thousands of hyperparameter combinations and significant computational resources.To overcome thows interpretability, paving the way to the future of a personalized nutritional science approach allowing the development of nutrition strategies for cancer prevention and/or therapeutics.

    Drugs currently used for controlling onchocerciasis and lymphatic filariasis (LF) are mainly microfilaricidal, with minimal or no effect on the adult worms. For efficient management of these diseases, it is necessary to search for new drugs with macrofilaricidal activities that can be used singly or in combination with existing ones. Daniellia oliveri and Psorospermum febrifugum are two plants commonly used in the local management of these infections in Bambui, a township in the North West Region of Cameroon, but there is currently no documented scientific evidence to support their claimed anthelmintic efficacy and safety. The aim of this study was to provide evidence in support of the search for means to eliminate these diseases by screening extracts and chromatographic fractions isolated from these plants for efficacy against the parasitic roundworms Onchocerca ochengi and Brugia pahangi.

    The viability of O. ochengi adult worms was assessed using the MTT/formazan assay.     Intestinal epithelial cells (IECs) and intestinal epithelial stem cells (IESCs) were harvested from BKS.Cg-Dock7
    +/+Lepr
    /JNju (DM) ****, and the expression of R-Spondin 3 (Rspo3) was detected by RT-qPCR, Western blotting, immunohistochemistry, and immunofluorescence. The role of Rspo3 in the abnormal differentiation of IECs during DM was confirmed by knockdown experiments. Through miRNA expression profiling, bioinformatics analysis, and RT-qPCR, we further analyzed the differentiation-related miRNAs in the IECs from **** with DM.

    Abnormal differentiation of IECs was observed in the **** with DM. The expression of Rspo3 was upregulated in the IECs from the **** with DM. This phenomenon was associated with Rspo3 overexpression. Additionally, Rspo3 is a major determinant of Lgr5+ stem cell identity in the diabetic state. Microarray analysis, bioinformatics analysis, and luciferase reporter assays revealed that microRNA (miR)-380-5p directly targeted Rspo3. Moreover, miR-380-5p upregulation was observed to attenuate the abnormal differentiation of IECs by regulating Rspo3 expression.

Our previous studies showed that N-of-1 trials could reflect the individualized characteristics of traditional Chinese medicine (TCM) syndrome differentiation with good feasibility, but the sensitivity was low. Therefore, this study will use hierarchical Bayesian statistical method to improve the sensitivity and applicability of N-of-1 trials of TCM. Methods/Design. This is a randomized, double-blind, placebo-controlled, three-pair crossover trial for a single subject, including 4-8 weeks of run-in period and 24 weeks of formal trial. In this study, we will recruit a total of 30 participants who are in the stable stage of bronchiectasis. The trial will be divided into three pairs (cycles), and one cycle contains two observation periods. The medications will be taken for three weeks and stopped for one week in the last week of each observation period. The order of syndrome differentiation decoction and placebo will be randomly determined. Patient self-reported symptom score (on a 7-point Likert scale) is the primary outcome. Discussion. Some confounding variables (such as TCM syndrome type and potential carryover effect of TCM) will be introduced into hierarchical Bayesian statistical method to improve the sensitivity and applicability of N-of-1 trials of TCM, and the use of prior available information (e.g., "borrowing from strength" of previous trial results) within the analysis may improve the sensitivity of the results of a series of N-of-1 trials, from both the individual and population level to study the efficacy of TCM syndrome differentiation. Our previous studies showed that N-of-1 trials could reflect the individualized characteristics of traditional Chinese medicine (TCM) syndrome differentiation with good feasibility, but the sensitivity was low. Therefore, this study will use hierarchical Bayesian statistical method to improve the sensitivity and applicability of N-of-1 trials of TCM. Methods/Design. This is a randomized, double-blind, placebo-controlled, three-pair crossover trial for a single subject, including 4-8 weeks of run-in period and 24 weeks of formal trial. In this study, we will recruit a total of 30 participants who are in the stable stage of bronchiectasis. The trial will be divided into three pairs (cycles), and one cycle contains two observation periods. The medications will be taken for three weeks and stopped for one week in the last week of each observation period. The order of syndrome differentiation decoction and placebo will be randomly determined. Patient self-reported symptom score (on a 7-point Likert scale) is the primary outcome. Discussion. No data about the evolution of the immune response after treatment are available. We aim to phenotype the regulatory immune response during natural HW infection and its evolution after treatment.

Diagnosis of coronavirus disease 2019 (COVID-19) is currently established with a polymerase chain reaction (PCR) test by means of oropharyngeal-, nasopharyngeal-, anal-swabs, sputum and blood plasma. However, oral and nasal swabs are more commonly used. This study, therefore, assessed sensitivity and specificity of plasma as a diagnostic in comparison with a combination of oral and nasal swab samples, and the implications for blood transfusion. Oropharyngeal (OP) and nasopharyngeal (NP) swab samples were obtained from 125 individuals suspected to have COVID-19 and stored in viral transport medium (VTM) tubes. Ten millilitres of blood samples in EDTA were also obtained by venepuncture and spun to obtain plasma. Viral RNA was obtained from both swabs and plasma by manual extraction with Qiagen QIAamp viral RNA Mini Kit. Detection was done using a real time fluorescent RT-qPCR BGI kit, on a QuantStudio 3 real-time PCR instrument. Average age of study participants was 41 years, with 74 (59.2%) being male. Out of the 125 individuals tested for COVID-19, 75 (60%) were positive by OP/NP swab. However, only 6 (4.8%) had a positive plasma result for COVID-19 with median Ct value of 32.4. Sensitivity and specificity of RT-PCR SARS-CoV-2 test using plasma was 8% and 100% respectively. There was no false positive recorded, but 69 (55.2%) false negatives were obtained by plasma. SARS-CoV-2 viral RNA was detected, albeit low (4.8%) in plasma. Plasma is likely not a suitable biological sample to diagnose acute SARS-CoV-2 infection. To check for accuracy and precision, measurements of the 87Sr/86Sr ratios and trace element concentrations were then compared in two laboratories that use different analytical methods. Differences in trace element concentrations between locations were not significant and could not, therefore, be used to discriminate between migration pathways. However, the Mekong mainstream, tributaries and distributaries could all be discriminated using Sr isotopes. The 87Sr/86Sr profiles recorded in P. krempfi otoliths showed that there were three contingents with obligate freshwater hatching and variable spawning sites along the Mekong mainstream, from Phnom Penh (Cambodia) to Nong Khai (Thailand) or further. After hatching, the fish migrated more or less rapidly to the Mekong Delta and then settled for most of their lifetime in brackish water. Spawning habitats and migration routes may be threatened by habitat shifts and the increasing number of hydropower dams along the river, especially the contingents born above Khone Falls (Laos). The conservation of P. krempfi, as well as other migratory fish in the Mekong River, requires agreements, common actions and management by all countries along the Mekong River. This study highlighted the importance of using both Sr/Ca and 87Sr/86Sr ratios to understand life history of anadromous fishes as the 87Sr/86Sr ratio in the water was shown to be less effective than the Sr/Ca ratio in identifying movements between different saline areas.

    This study employed a mixed-methods design. Focus group interviews and surveys were administered to a convenience sample (n = 34) of multidisciplinary obstetric teams. Data collected from focus group interviews were used to perform a task and equipment analysis. Survey data were coded and mapped by specialty to identify reported areas of congestion and time spent, and to identify themes related to physical space of the OR and labor and delivery unit.

    Task analysis revealed complex interdependencies between specialties. Thirty task groupings requiring over 20 pieces of equipment were identified. Perceived areas of congestion and areas of time spent in the OR varied by clinical specialty. The following categories emerged as main challenges encountered during an emergency Cesarean 1) size of physical space and equipment, 2) layout and orientation, and 3) patient transport.

  • Blood fatty acid profiles can indicate an animal's wild-type diet composition and fatty acid status, but have not been reported in sea turtles. Newer technologies allow for fatty acid profiles from very small (less than three drops) samples of whole blood. This study examined whole blood fatty acid profiles of presumably healthy, neritic, juvenile, wild green (Chelonia mydas) (n = 9; 6 males, 3 females) and Kemp's ridley (Lepidochelys kempii) (n = 8; 6 males, 2 females) turtles from North Carolina, USA. Saturated fatty acids, which can be synthesized de novo, consisted primarily of 160, although green turtle blood had a higher proportion of 180 (P less then 0.001) than Kemp's ridleys, while Kemp's ridley blood had higher proportions of 170 (P = 0.007), 200 (P = 0.03), 220 (P= 0.002), and 240 (P less then 0.001) as compared with green turtles. Total monounsaturated w7 fatty acids, which can be synthesized de novo or may be diet derived, were higher in Kemp's ridleys and predominantly in the form of 161 and 181w7 fatty acids. https://www.selleckchem.com/products/fdi-6.html Kemp's ridley blood had more than double the relative proportion of 161w7 as compared with green turtles (P= 0.03). Twenty-three healthy C. caretta (housed at the Marine Turtle Research Center) without preexisting ophthalmic disease were enrolled in the study. IOP measurements were obtained by the same ophthalmologist, with the turtle in ventral recumbency between 230 p.m. and 430 p.m., using a rebound tonometer (RT; TonoVet) in dog calibration mode, and after topical anesthesia, an applanation tonometer (AT; Tono-Pen) in both eyes. The average of three readings per instrument was used for analysis. The agreement between the two tonometers was assessed by Bland-Altman analysis and intraclass correlation coefficient (ICC). Moreover, differences in IOP between the two tonometers were analyzed using the Mann-Whitney test. Moderate agreement was found between the two tonometers (ICC, 0.663; 95% confidence interval, 0.206-0.857). The median, Q1, and Q3 IOP obtained with AT (6.2, 4.7, and 9.1 mm Hg) were significantly lower (P = 0.001) than that obtained with RT (9.7, 8.3, and 11.6 mm Hg). It was not possible to obtain an instrument automatically generated mean of four values with AT because of retraction of the globe by the animals, and IOP measurement was unsuccessful in 7 eyes. RT proved to be more feasible because of the light, short-lasting contact with the cornea.Five genetically distinct macropodid marsupial herpesviruses have been reported [Macropodid alphaherpesviruses 1 and 2 (MaHV-1 and -2); Macropodid herpesviruses 3 to 5 (MaHV-3 to -5)]. MaHV-2 was originally isolated from an outbreak of fatal disease in captive quokkas (Setonix brachyurus) that were in contact with other macropodid species. This warranted a survey of the presence of herpesviruses in this threatened and endemic Western Australian (WA) wallaby. Blood samples from 142 apparently healthy quokkas were tested for exposure to MaHV-1 and -2 by serology. Of these 142, 121 [Rottnest Island (RI), n = 93; mainland WA, n = 28] were tested for herpesvirus infection by polymerase chain reaction (PCR). Antibodies to MaHV-1 and -2 were detected in one individual [prevalence, 0.7%; 95% confidence interval (CI), 0.1%-3.2%] from the mainland and none from RI. However, a novel gammaherpesvirus [designated Macropodid herpesvirus 6 (MaHV-6)] was detected by PCR in the blood of 13 of 121 individuals (11%; 95% CI, 6.2-17.2). Infection with MaHV-6 was significantly more prevalent on the mainland (7/28; i.e., 25%) compared with RI (6/93; i.e., 6.45%; difference in sample proportions, 95% CI, 6%-32%; P = 0.015). There was no association (P > 0.05) between infection with MaHV-6 and differences in hematology, blood chemistry, peripheral blood cell morphologies, or on clinical status. There was a significant association between infection with MaHV-6 and the presence of Theileria spp. in blood [odds ratio (OR) = 11.0; 95% CI, 2.31-52.3; P = 0.001] and yeast in the nasal lining (OR = 7.0; 95% CI, 1.54-31.8; P = 0.021), suggesting that quokkas may be more susceptible to infection with these microorganisms if also infected with MaHV-6. MaHV-6 infection may be a catalyst for vulnerability to disease with other infectious agents and may pose a significant threat to other macropods. Newer technologies allow for fatty acid profiles from very small (less than three drops) samples of whole blood. This study examined whole blood fatty acid profiles of presumably healthy, neritic, juvenile, wild green (Chelonia mydas) (n = 9; 6 males, 3 females) and Kemp's ridley (Lepidochelys kempii) (n = 8; 6 males, 2 females) turtles from North Carolina, USA. Saturated fatty acids, which can be synthesized de novo, consisted primarily of 160, although green turtle blood had a higher proportion of 180 (P less then 0.001) than Kemp's ridleys, while Kemp's ridley blood had higher proportions of 170 (P = 0.007), 200 (P = 0.03), 220 (P= 0.002), and 240 (P less then 0.001) as compared with green turtles. Twenty-three healthy C. caretta (housed at the Marine Turtle Research Center) without preexisting ophthalmic disease were enrolled in the study. IOP measurements were obtained by the same ophthalmologist, with the turtle in ventral recumbency between 230 p.m. and 430 p.m., using a rebound tonometer (RT; TonoVet) in dog calibration mode, and after topical anesthesia, an applanation tonometer (AT; Tono-Pen) in both eyes. The average of three readings per instrument was used for analysis. The agreement between the two tonometers was assessed by Bland-Altman analysis and intraclass correlation coefficient (ICC). Moreover, differences in IOP between the two tonometers were analyzed using the Mann-Whitney test. Moderate agreement was found between the two tonometers (ICC, 0.663; 95% confidence interval, 0.206-0.857). The median, Q1, and Q3 IOP obtained with AT (6.2, 4.7, and 9.1 mm Hg) were significantly lower (P = 0.001) than that obtained with RT (9.7, 8.3, and 11.6 mm Hg). Of these 142, 121 [Rottnest Island (RI), n = 93; mainland WA, n = 28] were tested for herpesvirus infection by polymerase chain reaction (PCR). Antibodies to MaHV-1 and -2 were detected in one individual [prevalence, 0.7%; 95% confidence interval (CI), 0.1%-3.2%] from the mainland and none from RI. However, a novel gammaherpesvirus [designated Macropodid herpesvirus 6 (MaHV-6)] was detected by PCR in the blood of 13 of 121 individuals (11%; 95% CI, 6.2-17.2). Infection with MaHV-6 was significantly more prevalent on the mainland (7/28; i.e., 25%) compared with RI (6/93; i.e., 6.45%; difference in sample proportions, 95% CI, 6%-32%; P = 0.015).     A literature search was conducted in PubMed, Web of Science, PsycINFO, and two Chinese academic databases (China National Knowledge Infrastructure and Wanfang) for studies published from December 2019 to September 2020 that reported the prevalence of above mental health problems among children and adolescents. Random-effects meta-analyses were used to estimate the pooled prevalence.

    Twenty-three studies (21 cross-sectional studies and 2 longitudinal studies) from two countries (i.e., China and Turkey) with 57,927 children and adolescents were identified. Depression, anxiety, sleep disorders, and posttraumatic stress symptoms were assessed in 12, 13, 2, and 2 studies, respectively. Meta-analysis of results from these studies showed ths other than China are urgently needed.

  • Sulfanilamide is a common antibacterial drug that is used in clinical practice and in the industry. However, its abuse has emerged as a serious problem in the aquaculture industry, and more predominantly in the poultry industry, where it is administered for therapeutic purposes or as a growth promoter. In this study, a novel technical platform, heat-assisted desorption-dielectric barrier discharge ionization mass spectrometry (HAD-DBDI-MS), was used to detect sulfanilamide in situ. A method for the rapid identification of five typical sulfanilamide drugs sulfamopyridine, sulfamethoxan, sulfamethoxoline, sulfamidine, and sulfamethoxazole was established after optimizing a series of parameters. Secondary mass spectrometry was used to distinguish the sulfanilamide drugs from one another and from other isomers. Our HAD-DBDI-MS method enhanced the sulfanilamide sensitivities by reducing the limits of detection by about 1-2 orders of magnitude compared to those obtained with the DBDI-MS method. As further research, the profiles of chicken feed with added sulfamopyridine standard as well those of commercial feed with added sulfamidine and sulfamethoxazole were obtained using HAD-DBDI-MS respectively. With the HAD-DBDI-MS method, sulfamopyridine could be directly detected in the complex chicken meat matrix. However, satisfactory results were not obtained when direct HAD-DBDI-MS was used for sulfamidine and sulfamethoxazole detection in the feed matrix. The identification ability was improved when methanol was introduced as the assisting solvent. In this method, chicken feed spiked with sulfamidine and sulfamethoxazole standards were pressed into tablets, and methanol was dropped on the tablet surface. The experimental data indicate that HAD-DBDI-MS is suitable for the rapid identification of sulfanilamide drugs in poultry feed and meat products, thus showing potential value as a detection tool for future application.A thermal desorption electrospray ionization-triple quadruple mass spectrometry (TD-ESI-MS/MS) method was developed for the rapid screening of poppy husk in hot pot sauce and soup. The solid surface or liquid sample was directly touched by a simple metal probe. The analytes collected on the probe were desorbed and ionized using a TD-ESI source, after which the analyte ions were detected by MS/MS in multiple reaction monitoring (MRM) mode. The results were compared with those of colloidal gold card rapid detection and verified by high performance liquid chromatography coupled with triple quadrupole mass spectrometry (HPLC-MS/MS). The instrument gave the best response with the highest sensitivity under the following conditions thermal desorption temperature, 260 ℃; injection solvent, 0.1% formic acid aqueous solution containing 10 mmol/L formic ammonium-acetonitrile (11, v/v); flow rate, 200 μL/h. The PCBs were extracted from aquatic product samples using a dichloromethane-n-hexane (11, v/v) mixture at 100 ℃ with two extraction cycles. The extracting solution was purified twice with 0.5 mL concentrated sulfuric acid. After concentration to a constant volume, the target compounds were detected by GC-HRMS and quantified by the isotope internal standard method. Under the optimized conditions, the relative standard deviations (RSDs) of the mean relative response factor (RRF) for the 32 PCBs in the range of 0.1-20 μg/L were less than 15%. The limits of quantification were 0.3-1.9 ng/kg. At three spiked levels (5, 20, and 50 ng/kg) in grass carp and sea bass, the recoveries of the 32 PCBs were between 71.9% and 119.0%, with the RSDs varying from 3.5% to 19.6%. The excitation and emission wavelengths were 326 nm and 432 nm, respectively. An external standard method was used for quantification. The results showed that the linearities of vitamin K1 and vitamin K2 was in the ranges of 0.0025-2.0 μg/mL and 0.01-2.0 μg/mL, respectively, with correlation coefficients both greater than 0.999. The spiked recoveries were 80.39%-94.39% and the precisions were 0.85%-3.98%. Our HAD-DBDI-MS method enhanced the sulfanilamide sensitivities by reducing the limits of detection by about 1-2 orders of magnitude compared to those obtained with the DBDI-MS method. As further research, the profiles of chicken feed with added sulfamopyridine standard as well those of commercial feed with added sulfamidine and sulfamethoxazole were obtained using HAD-DBDI-MS respectively. With the HAD-DBDI-MS method, sulfamopyridine could be directly detected in the complex chicken meat matrix. However, satisfactory results were not obtained when direct HAD-DBDI-MS was used for sulfamidine and sulfamethoxazole detection in the feed matrix. The identification ability was improved when methanol was introduced as the assisting solvent. In this method, chicken feed spiked with sulfamidine and sulfamethoxazole standards were pressed into tablets, and methanol was dropped on the tablet surface. The experimental data indicate that HAD-DBDI-MS is suitable for the rapid identification of sulfanilamide drugs in poultry feed and meat products, thus showing potential value as a detection tool for future application.A thermal desorption electrospray ionization-triple quadruple mass spectrometry (TD-ESI-MS/MS) method was developed for the rapid screening of poppy husk in hot pot sauce and soup. The solid surface or liquid sample was directly touched by a simple metal probe. The analytes collected on the probe were desorbed and ionized using a TD-ESI source, after which the analyte ions were detected by MS/MS in multiple reaction monitoring (MRM) mode. The results were compared with those of colloidal gold card rapid detection and verified by high performance liquid chromatography coupled with triple quadrupole mass spectrometry (HPLC-MS/MS). The instrument gave the best response with the highest sensitivity under the following conditions thermal desorption temperature, 260 ℃; injection solvent, 0.1% formic acid aqueous solution containing 10 mmol/L formic ammonium-acetonitrile (11, v/v); flow rate, 200 μL/h. The limits of detection (LODs) for papaverine, noscapine, and thebaine in five alkaloids were 2 μg/kg, while those for codeine and morphine were 10 μg/kg in hot pot sauce and 5 μg/kg in soup. The sensitivity of this method was significantly superior to that of the colloidal gold card rapid detection. The method was applied to 50 batches of hot pot sauce and soup. Noscapine, papaverine, thebaine, and morphine were detected in a positive sample of chicken soup, which was consistent with the result of the HPLC-MS/MS method. This method without sample preparation and chromatographic separation is fast, green, and environmentally benign, thus being suitable for the rapid qualitative analysis of poppy husk in food.Gas chromatography (GC)-high-resolution mass spectrometry (HRMS) with accelerated solvent extraction (ASE)-purification simultaneously coupled to the isotope internal standard method is proposed for the determination of 32 polychlorinated biphenyls (PCBs) in aquatic products. Synchronous purification was achieved by adding 2 g of anhydrous sodium sulfate, 1 g of Cleanert Florisil, and 50 g of neutral alumina as the adsorbent to the ASE system. The PCBs were extracted from aquatic product samples using a dichloromethane-n-hexane (11, v/v) mixture at 100 ℃ with two extraction cycles. The extracting solution was purified twice with 0.5 mL concentrated sulfuric acid. After concentration to a constant volume, the target compounds were detected by GC-HRMS and quantified by the isotope internal standard method. Under the optimized conditions, the relative standard deviations (RSDs) of the mean relative response factor (RRF) for the 32 PCBs in the range of 0.1-20 μg/L were less than 15%. The limits of quantification were 0.3-1.9 ng/kg. At three spiked levels (5, 20, and 50 ng/kg) in grass carp and sea bass, the recoveries of the 32 PCBs were between 71.9% and 119.0%, with the RSDs varying from 3.5% to 19.6%. This method effectively reduces the matrix interference and shows high sensitivity, good reproducibility, and stable recovery, thus proving useful for the detection of PCBs in aquatic products.A method for the determination of vitamin K1 and vitamin K2 in modulation milk powder was developed by high performance liquid chromatography (HPLC) coupled with post-column reduction. The samples were dissolved in water, lipase hydrolyzed, saponified with 2.5 mol/L sodium hydroxide solution and ethanol solution, extracted with n-hexane, and dissolved in methanol after concentration. The vitamin K were first separated on an Xbridge C18 column and then on a zinc powder reduction column, and detected using a fluorescence detector. The excitation and emission wavelengths were 326 nm and 432 nm, respectively. An external standard method was used for quantification. The results showed that the linearities of vitamin K1 and vitamin K2 was in the ranges of 0.0025-2.0 μg/mL and 0.01-2.0 μg/mL, respectively, with correlation coefficients both greater than 0.999. The spiked recoveries were 80.39%-94.39% and the precisions were 0.85%-3.98%. However the effects of cannabinoid genotoxicity have not been assessed on whole populations and formal consideration of effects as a broadly acting genotoxin remain unexplored. Our study addressed these knowledge gaps in USA datasets. Cancer data from CDC, drug exposure data from National Survey of Drug Use and Health 2003-2017 and congenital anomaly data from National Birth Defects Prevention Network were used. We show that cannabis, THC cannabigerol and cannabichromene exposure fulfill causal criteria towards first Principal Components of both (A) Down syndrome, Trisomies 18 and 13, Turner syndrome, Deletion 22q11.2, and (B) thyroid, liver, breast and pancreatic cancers and acute myeloid leukaemia, have mostly medium to large effect sizes, are robust to adjustment for ethnicity, other drugs and income in inverse probability-weighted models, show prominent non-linear effects, have 55/56 e-Values > 1.25, and are exacerbated by cannabis liberalization (P = 9.67 × 10-43, 2.66 × 10-15). The results confirm experimental studies showing that cannabinoids are an important cause of community-wide genotoxicity impacting both birth defect and cancer epidemiology at the chromosomal hundred-megabase level.Cancer is the second leading cause of death in the United States. Although screening facilitates prevention and early detection and is one of the most effective approaches to reducing cancer mortality, participation is low-particularly among underserved populations. In a large, preregistered field experiment (n = 7711), we tested whether deadlines-both with and without monetary incentives tied to them-increase colorectal cancer (CRC) screening. We found that all screening invitations with an imposed deadline increased completion, ranging from 2.5% to 7.3% relative to control (ps  less then  .004). Most importantly, individuals who received a short deadline with no incentive were as likely to complete screening (9.7%) as those whose invitation included a deadline coupled with either a small (9.1%) or large declining financial incentive (12.0%; ps = .57 and .04, respectively). These results suggest that merely imposing deadlines-especially short ones-can significantly increase CRC screening completion, and may also have implications for other forms of cancer screening.HER3 is a member of the EGF receptor family and elevated expression is associated with cancer progression and therapy resistance. The scDb-scFv showed increased binding to HER3-expressing cancer cell lines compared to the scDb and consequently more effective T-cell activation and T-cell proliferation. Furthermore, the bivalent binding mode of the scDb-scFv for HER3 translated into more potent T-cell mediated cancer cell killing, and allowed to discriminate between moderate and low HER3-expressing target cells. To further our understanding of biological processes in their early life, transcriptomic analysis of larval developmental stages (nauplius, zoea, mysis, and postlarva) were determined in the black tiger shrimp, Penaeus monodon using next-generation RNA sequencing. Gene clustering and gene enrichment analyses revealed that most of the transcripts were mainly related to metabolic processes, cell and growth development, and immune system. Interestingly, Spätzle and Toll receptors were found in nauplius stage, providing evidence that Toll pathway was a baseline immune system established in early larval stages.

    In the Indian subcontinent, although the prevalence of PP is influenced by the definition used, the most common PP is that of mania.Duck hepatitis A virus serotype 1 (DHAV-1) causes acute inflammatory injury with a very high mortality rate in ducklings, leading to severe economic losses worldwide, especially in mainland China. There is an urgent need to find new treatments to prevent and control infection with DHAV-1. Not only is there a shortage of commercial anti-DHAV-1 drugs, but there are also gaps in the use and protection rates of existing commercial vaccines. We previously found that icariin (ICA), an extract of Epimedium, can reduce the mortality rate of ducklings after DHAV-1 infection, and the effect of ICA after phosphorylation modification (pICA) is more evident. In this study, we used duck embryo hepatocytes (DEHs) to investigate the mechanism of the alleviation of DHAV-1-induced inflammation and oxidative stress by ICA and pICA, and to further study their effects on hepatocyte mitochondrial function, apoptosis and cell cycle. It was found that ICA and pICA can inhibit the negative effects of DHAV-1 on apoptosis and cell cycle progression by stabilizing mitochondrial function, thereby reducing inflammation and ultimately protecting liver cells. The effects of pICA are more beneficial than those of ICA. The results of this study may be useful in the development of a new prophylactic and therapeutic strategy against DHAV-1 and other acute inflammatory diseases.This study established the causes and timing of spontaneous sow deaths in the farrowing units of ten Danish sow herds. Herds participated for seven to 15 months during 2018-19. We received data (production data and detailed information on the sows that died) on a total of 126 sows. Fifty-three sows were necropsied, and tissues were evaluated histopathologically. Twenty-four percent of the sows died 0-5 days postpartum. The main cause of death in the study was liver lobe torsion, which was diagnosed in 22 of 53 necropsied sows (42%). Deaths caused by liver lobe torsions were less often seen during the 0-5 day postpartum period compared to deaths caused by other reasons (P = 0.002). Seven of the necropsied sows (13%) died from endotoxaemic shock from retained foetuses. This cause of death was seen in seven of ten herds. These sows typically died 1-3 days postpartum. Pneumonia accounted for 13% of deaths in the necropsied sows, but the majority of these sows originated from one herd experiencing a respiratory outbreak caused by the introduction of M. hyopneumonia. Less prevalent causes of death in the study were torsion of the intestinal segment (8%), suspected cardiovascular collapse (8%), rupture of blood vessels (uterine and nonuterine) (8%), gastric ulcer (4%), sepsis (2%) and liver abscess (2%). We concluded that liver lobe torsion needs further attention to establish the background of this surprisingly prevalent cause of death. A radial immunodiffusion (RID) test was used to determine the concentration of serum IgG and calves were classified as having FPTI if the IgG concentration was less than 10 mg/mL. Estimation of FPTI was also assessed using two indirect methods using serum total protein (STP) and a brix refractometer. The estimated prevalence of FPTI was found to be 8.7% (43 calves out of 495) by RID with the concentration of IgG ranging between 0 and 6.2 mg/ml. The STP was found to vary from 46 to 96 mg/mL and using a cut-off point of 55 mg/mL the calf level prevalence was estimated as 7.1% (33 calves). Using the brix refractometer, the prevalence was found to be 13.1% (65 calves) with the refractometer reading ranging 6-14% of IgG. In the present study there was no association between calf-level factors (age, sex and breed) and FPTI. There was a higher correlation of the RID test results and the STP results compared to the RID and brix refractometer results. It is concluded that the prevalence of FPTI in dairy calves in the south-west region of Western Australia is low (8.7%) and the brix refractometer is not a reliable indirect method for determining passive transfer of immunity to calves.Meiotic recombination is key to the repair of DNA double-strand break damage, provide a link between homologs for proper chromosome segregation as well as ensure genetic diversity in organisms. The Rad51 gene was amplified using PCR, cloned and sequenced using testicular cDNA from yak and cattle-yak. The STP was found to vary from 46 to 96 mg/mL and using a cut-off point of 55 mg/mL the calf level prevalence was estimated as 7.1% (33 calves). Using the brix refractometer, the prevalence was found to be 13.1% (65 calves) with the refractometer reading ranging 6-14% of IgG. In the present study there was no association between calf-level factors (age, sex and breed) and FPTI. There was a higher correlation of the RID test results and the STP results compared to the RID and brix refractometer results. The Rad51 gene was amplified using PCR, cloned and sequenced using testicular cDNA from yak and cattle-yak.
