Materials and methods Colloidal silver nanoparticles (AgNPs) were synthesized by reducing aqueous AgNO3 with aqueous Taxus leaf extract at nonphotomediated conditions, without any catalyst, template or surfactant. The AgNP production was confirmed by ultraviolet-visible (UV-VIS) spectroscopy, scanning electron microscopy (SEM), X-ray diffraction (XRD) and Fourier-transform infrared (FTI) spectroscopy. Materials and methods Colloidal silver nanoparticles (AgNPs) were synthesized by reducing aqueous AgNO3 with aqueous Taxus leaf extract at nonphotomediated conditions, without any catalyst, template or surfactant. The AgNP production was confirmed by ultraviolet-visible (UV-VIS) spectroscopy, scanning electron microscopy (SEM), X-ray diffraction (XRD) and Fourier-transform infrared (FTI) spectroscopy. The MTT assay for human breast cancer cells as well as the DAPI fluorescent staining microscopy tested the biocompatibility and anticancer effects of AgNPs, silver nitrate, and Taxol.   • Blinding is considered an important methodological characteristic in clinical trials to minimise bias and maximise the validity of a trial. Unlike pharmaceutical substances, most herbal medicines have distinctive sensory specifications, including odour and taste, which can be quite challenging when developing a placebo control to match the specific characteristics of herbal substances being examined. The present study was, therefore, designed to evaluate whether the participants could differentiate an active herbal capsule (Ganopoly combination) from a placebo material capsule. The aim of this study was to develop a suitable placebo substance for encapsulation to be used in a future herbal medicine clinical trial.

    The current study was improved upon the previous investigation, and several modifications were made to the placebo substance in order to mimic the herbal substance characteristics. Prior to conducting the study, a refined placebo substance was developed using commonly consumed culinary agents. Steristics.

However, concerns exist about whether the current structure and delivery of GCP training sufficiently prepares clinical investigators and their delegates to conduct clinical trials.

    We conducted qualitative semi-structured interviews with 13 clinical investigators and 10 research sponsors to 1) examine characteristics of the quality conduct of sponsored clinical trials, including critical tasks and concerns perceived as essential for trial quality, 2) identify key knowledge and skills required to perform critical tasks, and 3) identify gaps and redundancies in GCP training and areas of improvement to ensure quality conduct of clinical trials. Data were examined using applied thematic analysis.

    The top three tasks identified as critical for the quality conduct of clinical trials were obtaining informed consent, ensuring protocol compliance, and protecting participants' health and safety. Respondents acknowledged that GCP principles address each of these critical tasks but also described many challenges and burdens of GCP training, including high training frequency and repetitive content. Respondents suggested moving beyond GCP training as a mere check-box activity by making it more effective, engaging, and interactive. They also emphasized that applying GCP principles in a real-world, skills-based environment would increase the perceived relevance of GCP training.

    Our findings indicate that although investigators and sponsors recognize that GCP training addresses tasks critical to the quality conduct of clinical trials, the need for significant improvement in the design, content, and presentation of GCP training remains.

In specimens processed using multifunction cavitation (MFC), the compressive residual stress was high when the processing time was 10 min and the Cr content on the surface was greater than on the surface of an unprocessed specimen. On the other hands, in specimens subjected to water-jet peening (WJP), the compressive residual stress was high when the processing time was 10 min. In the present study, the processing time was selected to be 10 min and all high-temperature corrosion tests were conducted by the coating method. In the case of long-term high-temperature corrosion at 500 °C, the corrosion loss of the ****processed and WJP-processed specimens was small, whereas the corrosion loss of the unprocessed specimen was large.Understanding the motion of debris cloud produced by the anti-satellite test can help us to know the danger of these tests. This study presents the orbit status of 57 fragments observed by the CelesTrak and presented in the NORAD Two-Line Element Sets of India Anti-Satellite Test. There are 10 of these observed fragments have altitudes of the apogee larger than 1000.0km, the maximum one is 1725.7km. We also numerical calculated the number of debris, the results show that the number of debris with the diameter larger than 0.2m is 14, the number of debris with the diameter larger than 0.01m is 6587, and the number of debris with the diameter larger than 0.001m is 7.22×105. The results of the secondary collision of the debris will produced more fragments in the space. We sought to examine outcomes after pulmonary resection with radiotherapy used as a benchmark comparator. METHODS Patients treated at a single institution (2000-2017) with cT1-3N0-2M1 non-small cell lung cancer, 3 or less synchronous metastases, and performance status 0 to 1, and who received comprehensive local consolidative therapy were analyzed according to local consolidative therapy modality for the primary lesion. Progression was analyzed with death as a competing risk. RESULTS Of 88 patients meeting inclusion criteria, 63 (71.6%) received radiotherapy for local consolidative therapy modality for the primary lesion and 25 (28.4%) underwent surgery (lobectomy 20/25 [80.0%], pneumonectomy 3/25 [12.0%], sublobar 2/25 [8.0%]). OBJECTIVE To examine whether there is an association between prehospital transfer distance and surgical mortality in emergency thoracic aortic surgery. METHODS A retrospective cohort study using a national clinical database in Japan was conducted. Patients who underwent emergency thoracic aortic surgery from January 1, 2014, to December 31, 2016, were included. Patients with type B dissection were excluded. A multilevel logistic regression analysis was performed to examine the association between prehospital transfer distance and surgical mortality. In addition, an instrumental variable analysis was performed to address unmeasured confounding. RESULTS A total of 12,004 patients underwent emergency thoracic aortic surgeries at 495 hospitals. Surgical mortality was 13.8%. The risk-adjusted mortality odds ratio for standardized distance (mean 12.8 km, standard deviation 15.2 km) was 0.94 (95% confidence interval, 0.87-1.01; P = .09). Instrumental variable analysis did not reveal a significant association between transfer distance and surgical mortality as well. CONCLUSIONS No significant association was found between surgical mortality and prehospital transfer distance in emergency thoracic aortic surgery cases. Suspected cases of acute thoracic aortic syndrome may be transferred safely to distant high-volume hospitals. OBJECTIVE We hypothesize that segmentectomy is associated with similar recurrence-free and overall survival when compared with lobectomy in the setting of patients with clinical T1cN0M0 non-small cell lung cancer (NSCLC; >2-3 cm), as defined by the American Joint Committee on Cancer 8th edition staging system. METHODS We performed a single-institution retrospective study identifying patients undergoing segmentectomy (90) versus lobectomy (279) for T1c NSCLC from January 1, 2003, to December 31, 2016. Univariate, multivariable, and propensity score-weighted analyses were performed to analyze the following endpoints freedom from recurrence, overall survival, and time to recurrence. RESULTS Patients undergoing segmentectomy were older than patients undergoing lobectomy (71.5 vs 68.8, respectively, P = .02). There were no differences in incidence of major complications (12.4% vs 11.7%, P = .85), hospital length of stay (6.2 vs 7 days, P = .19), and mortality at 30 (1.1% vs 1.7%, P = 1) and 90 days (2.2% vs 2.3%, P = 1). In addition, there were no statistical differences in locoregional (12.2% vs 8.6%, P = .408), distant (11.1% vs 13.9%, P = .716), or overall recurrence (23.3% vs 22.5%, P = 1), as well as 5-year freedom from recurrence (68.6% vs 75.8%, P = .5) or 5-year survival (57.8% vs 61.0%, P = .9). Propensity score-matched analysis found no differences in overall survival (hazard ratio [HR], 1.034; P = .764), recurrence-free survival (HR, 1.168; P = .1391), or time to recurrence (HR, 1.053; P = .7462). CONCLUSIONS In the setting of clinical T1cN0M0 NSCLC, anatomic segmentectomy was not associated with significant differences in recurrence-free or overall survival at 5 years. Scanning electron microscopy imaging revealed that the cilia were absent only over the contact area between the trachea and the wire forming the helical stent. CONCLUSIONS Helical nitinol stents designed to provide radial support for malacic airways were well tolerated in a porcine model, providing for mucus clearance while also enabling atraumatic removal. OBJECTIVES Comprehensive local consolidative therapy led to improved overall survival in oligometastatic non-small cell lung cancer in a recent phase II trial, yet the role of pulmonary resection in ongoing oligometastatic trials is a matter of controversy. We sought to examine outcomes after pulmonary resection with radiotherapy used as a benchmark comparator. METHODS Patients treated at a single institution (2000-2017) with cT1-3N0-2M1 non-small cell lung cancer, 3 or less synchronous metastases, and performance status 0 to 1, and who received comprehensive local consolidative therapy were analyzed according to local consolidative therapy modality for the primary lesion. Progression was analyzed with death as a competing risk. RESULTS Of 88 patients meeting inclusion criteria, 63 (71.6%) received radiotherapy for local consolidative therapy modality for the primary lesion and 25 (28.4%) underwent surgery (lobectomy 20/25 [80.0%], pneumonectomy 3/25 [12.0%], sublobar 2/25 [8.0%]). OBJECTIVE To examine whether there is an association between prehospital transfer distance and surgical mortality in emergency thoracic aortic surgery. METHODS A retrospective cohort study using a national clinical database in Japan was conducted. Patients who underwent emergency thoracic aortic surgery from January 1, 2014, to December 31, 2016, were included. Patients with type B dissection were excluded. A multilevel logistic regression analysis was performed to examine the association between prehospital transfer distance and surgical mortality. In addition, an instrumental variable analysis was performed to address unmeasured confounding. RESULTS A total of 12,004 patients underwent emergency thoracic aortic surgeries at 495 hospitals. Surgical mortality was 13.8%. The risk-adjusted mortality odds ratio for standardized distance (mean 12.8 km, standard deviation 15.2 km) was 0.94 (95% confidence interval, 0.87-1.01; P = .09). Instrumental variable analysis did not reveal a significant association between transfer distance and surgical mortality as well. CONCLUSIONS No significant association was found between surgical mortality and prehospital transfer distance in emergency thoracic aortic surgery cases. Suspected cases of acute thoracic aortic syndrome may be transferred safely to distant high-volume hospitals. OBJECTIVE We hypothesize that segmentectomy is associated with similar recurrence-free and overall survival when compared with lobectomy in the setting of patients with clinical T1cN0M0 non-small cell lung cancer (NSCLC; >2-3 cm), as defined by the American Joint Committee on Cancer 8th edition staging system. METHODS We performed a single-institution retrospective study identifying patients undergoing segmentectomy (90) versus lobectomy (279) for T1c NSCLC from January 1, 2003, to December 31, 2016. Univariate, multivariable, and propensity score-weighted analyses were performed to analyze the following endpoints freedom from recurrence, overall survival, and time to recurrence. RESULTS Patients undergoing segmentectomy were older than patients undergoing lobectomy (71.5 vs 68.8, respectively, P = .02). There were no differences in incidence of major complications (12.4% vs 11.7%, P = .85), hospital length of stay (6.2 vs 7 days, P = .19), and mortality at 30 (1.1% vs 1.7%, P = 1) and 90 days (2.2% vs 2.3%, P = 1). In addition, there were no statistical differences in locoregional (12.2% vs 8.6%, P = .408), distant (11.1% vs 13.9%, P = .716), or overall recurrence (23.3% vs 22.5%, P = 1), as well as 5-year freedom from recurrence (68.6% vs 75.8%, P = .5) or 5-year survival (57.8% vs 61.0%, P = .9). Propensity score-matched analysis found no differences in overall survival (hazard ratio [HR], 1.034; P = .764), recurrence-free survival (HR, 1.168; P = .1391), or time to recurrence (HR, 1.053; P = .7462). CONCLUSIONS In the setting of clinical T1cN0M0 NSCLC, anatomic segmentectomy was not associated with significant differences in recurrence-free or overall survival at 5 years. Further prospective randomized trials are needed to corroborate the expansion of the role of anatomic segmentectomy to all American Joint Committee on Cancer 8th Edition Stage 1A NSCLC.

The outcome of this study could be a useful index for the fish-borne zoonotic trematode epidemiology in the Mekong area. It is important to understand the deformation of the liquid droplets under acceleration for the design and applications of MEMS liquid sensors, as this will affect the performance of the sensors. This paper presents an investigation into the deformation of a mercury droplet in a liquid MEMS sensor under accelerations and reports the relationship between the deformation and the accelerations. The Laminar level set method was used in the numerical process. The geometric model consisted of a mercury droplet of 2 mm in diameter and an annular groove of 2.5 mm in width and 2.5 mm in height. The direction of the acceleration causing the droplet to deform is perpendicular to the direction of gravity. Fabrication and acceleration experiments were conducted. The deformation of the liquid was recorded using a high-speed camera. Both the simulation and experimental results show that the characteristic height of the droplets decreases as the acceleration increases. At an acceleration of 10 m/s2, the height of the droplet is reduced from 2 to 1.658 mm, and at 600 m/s2 the height is further reduced to 0.246 mm. The study finds that the droplet can deform into a flat shape but does not break even at 600 m/s2. Besides, the properties of the material in the domain surrounding the droplet and the contact angle also affect the deformation of the droplet. This work demonstrates the deformation of the liquid metal droplets under acceleration and provides the basis for the design of MEMS droplet acceleration sensors.Developing a bee-friendly alternative to traditional insecticides used within commercial environments can contribute to reductions in pesticide exposure experienced by managed bees. We performed acute contact toxicity studies using fifteen plant powders from seven plant families against a parasitoid pest, Pteromalus venustus, and its host, the Alfalfa leafcutting bee (ALB). Ajwain, cinnamon, clove, cumin, fennel, ******, nutmeg, oregano and turmeric applied at low contact concentrations had sufficient fumigant properties to cause equivalent or higher parasitoid mortality as that obtained with the traditional insecticide. Nutmeg adversely affected adult ALBs at both low and high contact concentrations, thus eliminating it as a candidate. Increasing the contact concentrations did not consistently increase parasitoid control but did increase adverse effects on the ALBs. In addition, the efficacious plant powders significantly reduced the sexual function and fertility of the female parasitoids, a feature not associated with the traditional insecticide.

  • The improvement of cathode performance has always been the bottleneck and research hot spot for microbial fuel cells (****). While converting the majority of organic carbon to biogas (mainly consisting 60%CH4 + 40%CO2), the high-strength anaerobic digestion liquor consists of a high level of nitrogen concentration. The feasibility of utilizing biogas produced in-situ to achieve satisfactory nitrogen removal performance from partially nitrified anaerobic digestion liquor was examined in this study. To this end, a membrane biofilm reactor (MBfR) was used to couple nitrite- or nitrate-dependent anaerobic methane oxidation (n-DAMO) and anammox microorganisms, which was supplied with synthetic biogas and partially nitrified anaerobic digestion liquor (470 mg NH4+-N/L + 560 mg NO2--N/L). The MBfR achieved not only nearly complete nitrogen removal (~99%), but also a practically useful nitrogen removal rate above 1 kg N/m3/d. Due to the acidification caused by excessive CO2 supply from biogas, pH dropping was observed. Two corresponding strategies, i.e., intermittent alkali dosing and intermittent nitrogen gas flushing, were developed to control the pH at neutral. Mass balance based on batch tests and microbial community analysis by 16S rRNA gene amplicon sequencing both showed the joint contribution of anammox bacteria and anaerobic methane oxidizers to the nitrogen removal. This study proved the potential and capacity of MBfR to access complete nitrogen removal from high-strength wastewater by using biogas produced in-situ, thus leading to a significant reduction of external carbon addition in practice.Polychlorinated biphenyls (PCBs) have been reported to pose a severe risk towards human health, and hydroxylated polychlorinated biphenyls (OH-PCBs) were potential substances basis for PCBs' toxicity. For phenotypic analysis of CES1 and CES2 activity, we used the hydrolysis metabolism of 2-(2-benzoyl3-methoxyphenyl) benzothiazole (BMBT) and fluorescein diacetate (FD) catalyzed by human liver microsomes (HLMs) as the probe reactions. Preliminary inhibition screening showed that the inhibition potential of OH-PCBs towards CES1 and CES2 increased with the increased numbers of chlorine atoms in OH-PCBs. Both 2'-OH-PCB61 and 2'-OH-PCB65 showed concentration-dependent inhibition towards both CES1 and CES2. Lineweaver-Burk plots showed that 2'-OH-PCB61 and 2'-OH-PCB65 exerted non-competitive inhibition towards CES1 and competitive inhibition towards CES2. The inhibition kinetics parameters (Ki) were 6.8 μM and 7.0 μM for 2'-OH-PCB61 and 2'-OH-PCB65 towards CES1, respectively. In many mountainous regions forests deliver slope stability, helping to prevent landslides. However, forests are progressively converted into other land uses in many mountainous regions. In this study, we focus on the Colombian Andes, the most populated and deadly landslide-prone part of Colombia. We aim to determine the difference in frequency of landslides from forested and non-forested areas, and subsequently, quantify the potential costs and benefits of protecting forest and of restoring forest from agricultural lands. To that end, we combine economic data with geographical information related to public and private infrastructure, land use, and landslide susceptibility. Analyzing the national landslide database of Colombia, we established that landslides are almost six times (581%) more likely to occur on non-forested lands than on forested lands. From an economic perspective, by preventing landslides, forests provide a net benefit through the provision of slope stability services. Our most conservative estimates indicate it is 16 times more cost-effective to promote forest corridors, via conservation or reforestation along roads by paying farmers and cattle herders their opportunity costs, than for the public to pay the expected value of landslide damage. Our analysis provides strong evidence that vegetated hillsides can provide a cost-effective ecosystem service approach to mitigate economic losses due to landslides in one of the world's most landslide prone areas.Plastic pollution compromises ocean health, with large amounts of plastics continuing to enter marine and coastal environments. All these results will provide a new perspective for elucidation of toxicity mechanism of PCBs and OH-PCBs.Landslides cause billions of dollars (USD) in damage and hundreds of life losses every year in mountainous areas globally, and these effects are exacerbated by climate change and increased human occupation of vulnerable areas. In many mountainous regions forests deliver slope stability, helping to prevent landslides. However, forests are progressively converted into other land uses in many mountainous regions. In this study, we focus on the Colombian Andes, the most populated and deadly landslide-prone part of Colombia. We aim to determine the difference in frequency of landslides from forested and non-forested areas, and subsequently, quantify the potential costs and benefits of protecting forest and of restoring forest from agricultural lands. To that end, we combine economic data with geographical information related to public and private infrastructure, land use, and landslide susceptibility. Analyzing the national landslide database of Colombia, we established that landslides are almost six times (581%) more likely to occur on non-forested lands than on forested lands. From an economic perspective, by preventing landslides, forests provide a net benefit through the provision of slope stability services. Our most conservative estimates indicate it is 16 times more cost-effective to promote forest corridors, via conservation or reforestation along roads by paying farmers and cattle herders their opportunity costs, than for the public to pay the expected value of landslide damage. Our analysis provides strong evidence that vegetated hillsides can provide a cost-effective ecosystem service approach to mitigate economic losses due to landslides in one of the world's most landslide prone areas.Plastic pollution compromises ocean health, with large amounts of plastics continuing to enter marine and coastal environments. Various mitigative engineering solutions are being developed and implemented in response to this threat. While recognising the positive impacts of clean-ups, we highlight two perspectives given little attention to date, which are vital to evaluating the cost-benefit ratio of clean-ups firstly, clean-up efficiency where density and accessibility of litter are key, and secondly, potential negative externalities that implementation of clean-up technologies may have. These principles, catch per unit effort and the impact on non-target species, are well known from fisheries management. We argue they should also be applied in evaluating marine litter removal schemes.

  • Acute aortic dissection was an incidental finding during computed tomography-angiography in 4/15 (27%). Two out of 15 patients (13%) received intravenous thrombolysis, 9/15 (60%) underwent aortic surgery and 10/15 (67%) died. Results The proportion of patients without comorbidity was 24.8%; 31.8% had low comorbidity; 33.5% had moderate comorbidity and 9.9% had high comorbidity. At 12 months, the proportion of poor outcome (dead or dependent mRS ≥3) was 24.8% (no comorbidity), 34.7% (low), 45.2% (moderate) and 59.4% (high). At five years, these proportions were 37.7%, 50.3%, 64.3%, and 81.7%, respectively. There was clustering of cardiovascular conditions and substantial negative effects of dementia, kidney, and heart failure. Conclusion Comorbidity is common and has a strong impact on mortality and functional outcome. Patients and methods Using all incident and recurrent strokes in a population-based cohort (Oxford Vascular Study/OXVASC) with multiple sources of ascertainment as the reference, we determined the temporal trends in sensitivity and positive predictive value of hospital diagnostic codes for identifying acute stroke from 2002 to 2017. Results Of 1883 hospitalised strokes, 1341 (71.2%) were correctly identified by coding. Sensitivity of coding improved over time for all strokes (ptrend = 0.005) and for incident cases (ptrend = 0.002). Of 1995 apparent stroke admissions identified by International Classification of Disease-10 stroke codes (I60-I68), 1588 (79.6%) used the stroke-specific codes (I60-I61/I63-I64). Positive predictive value was higher with the use of specific codes (83.2% vs. 69.2% for all codes) and highest if combined with the first admission only (88.5%), particularly during more recent time periods (2014-2017 = 90.3%). Of 2254 OXVASC incident strokes, 833 (37.0%) were not hospitalised. Sensitivity of coding increased over time for non-disabling stroke (ptrend = 0.001), but not for disabling/fatal stroke (ptrend = 0.40). Conclusions Although accuracy of hospital diagnostic coding for identifying acute strokes improved over the last 15 years, residual insensitivity supports linkage to other sources in large epidemiological studies. However, data on DICER1 involvement in paratesticular sarcomas have not been published. Herein, we analyse a series of 15 paediatric paratesticular sarcomas and describe in detail the case of a male infant with a paratesticular myxoid tumour, considered to be a low-grade sarcoma, who also manifested a cystic nephroma, a classic DICER1 syndrome phenotype. He harboured a pathogenic germline DICER1 variant and different somatic hot-spot mutations in each tumour. The paratesticular tumour showed strong and diffuse expression for WT1 and CD10, an unusual immunophenotype in paediatric sarcomas, but typical of tumours of Müllerian origin. The tumour was postulated to arise from the appendix testis, a Müllerian remnant located in the paratestis. Such an origin would be analogous to other DICER1-associated non-epithelial gynaecological tumours, thought to arise from Müllerian derivatives. These findings point towards a key role of DICER1 in Müllerian-derived structures. Supporting this hypothesis is the fact that the other paratesticular sarcomas from the series were either negative or focally positive for WT1 and for CD10, and none had any DICER1 mutations. In summary, we present the first case of a paratesticular sarcoma associated with DICER1 syndrome, emphasising that paratesticular tumours with an unusual histological appearance may suggest an underlying DICER1 mutation, especially in the presence of a personal or family history of DICER1-associated disease. In this context, DICER1 mutation testing could lead to changes in clinical care including implementation of cancer care surveillance strategies. © 2020 The Authors. The main outcome was the Childhood Sleep Habits Questionnaire (CSHQ). RESULTS Scores on the CSHQ demonstrated clinically significant sleep problems in 29 of the 30 children. When compared with previously reported normative values for typically developing children of the same age, children with 22q11DS had significantly greater sleep problems. Only 30% of children had previously undergone sleep study. While about half of children had tried a medication for sleep, it usually was not felt to be helpful. In contrast, parents reported that behavioral interventions, such as consistent bedtime routine and appropriate sleep environment, were helpful. This is one of the first studies to specifically address sleep problems other than obstructive sleep apnea in children with 22q11DS. CONCLUSIONS The findings suggest children with 22q11DS may have a higher risk of experiencing clinical sleep problems, compared to typically developing children. Improved anatomically mimetic in vitro models of these microvessels are urgently needed because nonhuman vessels and previous models do not accurately recapitulate the microenvironment and architecture of the human microvascular wall. Here, we describe parallel biofabrication of photopatterned self-rolled biomimetic pulmonary arterial microvessels of tunable size and infrastructure. These microvessels feature anatomically accurate layering and patterning of aligned human smooth muscle cells, extracellular matrix, and endothelial cells and exhibit notable increases in endothelial longevity and nitric oxide production. Computational image processing yielded high-resolution 3D perspectives of cells and proteins. Our studies provide a new paradigm for engineering multicellular tissues with precise 3D spatial positioning of multiple constituents in planar moieties, providing a biomimetic platform for investigation of microvascular pathobiology in human disease. Strikingly, SKP1-deficient spermatocytes show sharply reduced MPF activity and fail to enter MI despite treatment with okadaic acid. SKP1-deficient oocytes exhibit desynapsis, chromosome misalignment, and progressive postnatal loss. Therefore, SKP1 maintains synapsis in meiosis of both sexes. Furthermore, our results support a model where SKP1 functions as the long-sought intrinsic metaphase competence factor to orchestrate MI entry during male meiosis. Upon insect cuticle induction, pathogenic fungi undergo marked infection-related morphological differentiation. However, regulatory mechanisms of fungal infection-related morphogenesis are poorly understood. Here, we show that a histone lysine methyltransferase KMT2 in Metarhizium robertsii (MrKMT2) is up-regulated upon cuticle induction. MrKMT2 plays crucial roles in regulating infection-related morphogenesis and pathogenicity by up-regulating the transcription factor gene Mrcre1 via H3K4 trimethylation during mosquito cuticle infection. MrCre1 further regulates the cuticle-induced gene Mrhyd4 to modulate infection structure (appressorium) formation and virulence. Overall, the MrKMT2-MrCre1-MrHyd4 regulatory pathway regulates infection-related morphogenesis and pathogenicity in M. robertsii. These findings reveal that the epigenetic regulatory mechanism plays a pivotal role in regulating fungal pathogenesis in insects, and provide new insights into molecular interactions between pathogenic fungi and insect hosts. Here, we report the nature-inspired fabrication of several porous organic semiconductor-insulator blend films [semiconductor P3HT (p-type polymer), C8BTBT (p-type small-molecule), and N2200 (n-type polymer); insulator PS] by a breath figure patterning method and their broad and general applicability in organic thin-film transistors (OTFTs), gas sensors, organic electrochemical transistors (OECTs), and chemically doped conducting films. Detailed morphological analysis of these films demonstrates formation of textured layers with uniform nanopores reaching the bottom substrate with an unchanged solid-state packing structure. Device data gathered with both porous and dense control semiconductor films demonstrate that the former films are efficient TFT semiconductors but with added advantage of enhanced sensitivity to gases (e.g., 48.2%/ppm for NO2 using P3HT/PS), faster switching speeds (4.7 s for P3HT/PS OECTs), and more efficient molecular doping (conductivity, 0.13 S/m for N2200/PS). The stability and catalytic function of DUSP6 are maintained through conjugation of small ubiquitin-like modifier-1 (SUMO1) and SUMO2/3 at lysine-234 (K234), which is disrupted during oxidation through transcriptional up-regulation of SUMO-deconjugating enzyme, SENP1, causing DUSP6 degradation by ubiquitin-proteasome. deSUMOylation underlies DUSP6 degradation, Drp1-S616 hyperphosphorylation, mitochondrial fragmentation, and apoptosis induced by H2O2 in cultured cells or brain ischemia/reperfusion in ****. Overexpression of DUSP6, but not the SUMOylation-deficient DUSP6K234R mutant, protected cells from apoptosis.
