5, PM10, SO2, CO, and NO2 were positively correlated with hospital visits for depression. The strongest effects all occurred on lag0 (the same) day, and the corresponding excess risks (ERs) were 2.59 (95%CI 0.72, 4.49) for PM2.5, 3.08 (95%CI 1.05, 5.16) for PM10, 3.22 (95%CI 1.16, 5.32) for SO2, 4.38 (95%CI 1.83, 6.99) for CO, and 4.94 (95%CI 2.03, 7.92) for NO2 per IQR increase, respectively. The associations were found to be stronger in the elderly (≥65 years) and cold season. Furthermore, the effects of CO and NO2 remained significant in most two-pollutant models, suggesting that traffic-related air pollutants might be more important triggers of depression. Waste, especially biomass in general, is a large reservoir of nutrients that can be recovered through different technologies and used to produce biofertilizers. In the present study, environmental impacts of the production of microalgae biomass-based phosphate biofertilizer compared to triple superphosphate through life-cycle assessment conducted in the Simapro® software were investigated. The functional unit of the analysis was 163 g of P for both fertilizers. Phosphorus was recovered from a meat processing industry effluent in a high-rate algal pond. Impacts related to the entire biofertilizer chain impacted mainly on climate changes (3.17 kg CO2eq). Microalgae biofertilizer had higher environmental impact than conventional fertilizer in all impact categories, highlighting climate change and terrestrial ecotoxicity. An ideal scenario was created considering that all energy used comes from photovoltaic panels; in the separation step a physical method will be used, without energy expenditure (i.e. gravimetric sedimentation) and; biomass will be dried in a drying bed instead of the thermal drying. In this scenario, the impact of biofertilizer approached considerably those of triple superphosphate. When impacts of biomass cultivation and concentration stages were disregarded, drying step was of great relevance, contributing to increase biofertilizer impacts. More research is needed to optimize the algae production chain and determine the possibility of obtaining higher added value products more environmental attractive. Mercury (Hg) removal by six different living marine macroalgae, namely, Ulva intestinalis, Ulva lactuca, Fucus spiralis, Fucus vesiculosus, Gracilaria sp., and Osmundea pinnatifida was investigated in mono and multi-contamination scenarios. All macroalgae were tested under the same experimental conditions, evaluating the competition effects with all elements at the same initial molar concentration of 1 μmol dm-3. The presence of the main potentially toxic elements (Cd, Cr, Cu, Ni, and Pb) and rare earth elements (La, Ce, Pr, Nd, Eu, Gd, Tb, and Y) has not affected the removal of Hg. Characterizations of the macroalgae by FTIR before and after the biosorption/bioaccumulation assays suggest that Hg was mainly linked to sulfur-functional groups, while the removal of other elements was related with other functional groups. The mechanisms involved point to biosorption of Hg on the macroalgae surface followed by possible incorporation of this metal into the macroalgae by metabolically active processes. Globally, the green macroalgae (Ulva intestinalis, Ulva lactuca) showed the best performances for Hg, potential toxic elements and rare earth elements removal from synthetic seawater spiked with 1 μmol dm-3 of each element, at room temperature and pH 8.5. Metal release from mining wastes is a major environmental problem affecting ecosystems that requires effective, low-cost strategies for prevention and reclamation. The capacity of two strains (UB3 and UB5) of Sporosarcina luteola was investigated to induce the sequestration of metals by precipitation of carbonates in vitro and under microcosm conditions. These strains carry the ureC gene and have high urease activity. Also, they are highly resistant to metals and have the capacity for producing metallophores and arsenophores. SEM, EDX and XRD reveal that the two strains induced precipitation of calcite, vaterite and magnesian calcite as well as several (M2+)CO3 such as hydromagnesite (Mg2+), rhodochrosite (Mn2+), cerussite (Pb2+), otavite (Cd2+), strontianite (Sr2+), witherite (Ba2+) and hydrozincite (Zn2+) in vitro. Inoculation of the mixed culture of UB3+UB5 in tailings increased the pH and induced the precipitation of vaterite, calcite and smithsonite enhancing biocementation and reducing pore size and permeability slowing down the oxidation of residual sulfides. Results further demonstrated that the strains of S. luteola immobilize bioavailable toxic elements through the precipitation and coprecipitation of thermodynamically stable (M2+)CO3, Fe-Mn oxyhydroxides and organic chelates. Colombia is the fourth contributor to the Amazon River Basin (ARB) by surface, and the third by mean annual runoff. The Yahuarcaca Lakes System (YLS), consisting of four large interconnected water bodies situated on the floodplain of Amazon River, was identified as one of the key areas for the conservation of freshwater biodiversity in the Colombian ARB. This review aimed to provide a general overview of YLS, present its environmental and biological features, identify main ecological and health threats, and propose mitigation strategies and future research prospects. A systematic search was performed using various databases. In summary, YLS harbors significant biodiversity and provides a number of ecological services for local communities, encompassing fish and drinking water supply and utilization of the floodplain for agriculture. Ensuring its sustainability requires attention from local and international authorities, collaboration with indigenous communities and future interdisciplinary research. https://www.selleckchem.com/products/elamipretide-mtp-131.html Cities are severely affected by air pollution. Local emissions and urban structures can produce large spatial heterogeneities. We aim to improve the estimation of NO2, O3, PM2.5 and PM10 concentrations in 6 Italian metropolitan areas, using chemical-transport and machine learning models, and to assess the effect on population exposure by using information on urban population mobility. Three years (2013-2015) of simulations were performed by the Chemical-Transport Model (CTM) FARM, at 1 km resolution, fed by boundary conditions provided by national-scale simulations, local emission inventories and meteorological fields. A downscaling of daily air pollutants at higher resolution (200 m) was then carried out by means of a machine learning Random-Forest (RF) model, considering CTM and spatial-temporal predictors, such as population, land-use, surface greenness and vehicular traffic, as input. RF achieved mean cross-validation (CV) R2 of 0.59, 0.72, 0.76 and 0.75 for NO2, PM10, PM2.5 and O3, respectively, improving results from CTM alone.

Maritime surveillance videos provide crucial on-spot kinematic traffic information (traffic volume, ship speeds, headings, etc.) for varied traffic participants (maritime regulation departments, ship crew, ship owners, etc.) which greatly benefits automated maritime situational awareness and maritime safety improvement. Conventional models heavily rely on visual ship features for the purpose of tracking ships from maritime image sequences which may contain arbitrary tracking oscillations. To address this issue, we propose an ensemble ship tracking framework with a multi-view learning algorithm and wavelet filter model. First, the proposed model samples ship candidates with a particle filter following the sequential importance sampling rule. Second, we propose a multi-view learning algorithm to obtain raw ship tracking results in two steps extracting a group of distinct ship contour relevant features (i.e., Laplacian of Gaussian, local binary pattern, Gabor filter, histogram of oriented gradient, and canny descriptors) and learning high-level intrinsic ship features by jointly exploiting underlying relationships shared by each type of ship contour features. Third, with the help of the wavelet filter, we performed a data quality control procedure to identify abnormal oscillations in the ship positions which were further corrected to generate the final ship tracking results. We demonstrate the proposed ship tracker's performance on typical maritime traffic scenarios through four maritime surveillance videos. Background Medical residency is a time of high stress and long working hours, which increase the risk of cardiovascular disease. This study aimed to investigate the autonomic modulation of resident physicians throughout the on-call duty cycle. Methods Spectral analysis of heart rate variability (HRV) was used to compute cardiac parasympathetic modulation (high-frequency power, HF) and cardiac sympathetic modulation (normalized low-frequency power, LF%, and the ratio of LF and HF, LF/HF) of 18 residents for a consecutive 4-day cycle. Results Male residents show reduced cardiac sympathetic modulation (i.e., higher LF/HF and LF%) than the female interns. Medical residents' cardiac parasympathetic modulation (i.e., HF) significantly increased on the first and the second post-call day compared with the pre-call day. In contrast, LF% was significantly decreased on the first and the second post-call day compared with the pre-call day. Similarly, LF/HF was significantly decreased on the second post-call day compared with the pre-call day. LF/HF significantly decreased on the first post-call day and on the second post-call day from on-call duty. Conclusion The guideline that limits workweeks to 80 h and shifts to 28 h resulted in reduced sympathetic modulation and increased parasympathetic modulation during the two days following on-call duty. Background Abdominal adiposity is an important risk factor of chronic cardiovascular diseases, thus the prediction of abdominal adiposity and obesity can reduce the risks of contracting such diseases. However, the current prediction models display low accuracy and high sample size dependence. The purpose of this study is to put forward a new prediction method based on an improved support vector machine (SVM) to solve these problems. Methods A total of 200 individuals participated in this study and were further divided into a modeling group and a test group. Their physiological parameters (height, weight, age, the four parameters of abdominal impedance and body fat mass) were measured using the body composition tester (the universal INBODY measurement device) based on BIA. Intelligent algorithms were used in the modeling group to build predictive models and the test group was used in model performance evaluation. Firstly, the optimal boundary C and parameter gamma were optimized by the particle swarm algorithm. We then developed an algorithm to classify human abdominal adiposity according to the parameter setup of the SVM algorithm and constructed the prediction model using this algorithm. Finally, we designed experiments to compare the performances of the proposed method and the other methods. Results There are different abdominal obesity prediction models in the 1 KHz and 250 KHz frequency bands. The experimental data demonstrates that for the frequency band of 250 KHz, the proposed method can reduce the false classification rate by 10.7%, 15%, and 33% in relation to the sole SVM algorithm, the regression model, and the waistline measurement model, respectively. For the frequency band of 1 KHz, the proposed model is still more accurate. https://www.selleckchem.com/products/ar-c155858.html (4) Conclusions The proposed method effectively improves the prediction accuracy and reduces the sample size dependence of the algorithm, which can provide a reference for abdominal obesity.Weak delivery systems reduce the potential of evidence-supp orted interventions to improve nutrition. We synthesized the evidence for the effectiveness of nutrition-specific intervention delivery platforms for improving nutrition outcomes in low and middle-income countries (LMIC). A systematic literature search for studies published from 1997 to June 2018 resulted in the inclusion of 83 randomized controlled trials (RCTs), quasi-randomized, and controlled before-after studies across a variety of delivery platforms. In this paper, we report on meta-analysed outcomes for community health worker (CHW) home visits and mother/peer group delivery platforms. Compared to care as usual, CHW home visits increased early initiation of breastfeeding (EIBF) (OR 1.50; 95% CI 1.12, 1.99; n = 10 RCTs) and exclusive breastfeeding (EBF) (OR 4.42; 95% CI 2.28, 8.56; n = 9 RCTs) and mother/peer groups were effective for improving children's minimum dietary diversity (OR 2.34; 95% CI 1.17, 4.70; n = 4) and minimum meal frequency (OR 2.31; 95% CI 1.61, 3.31; n = 3). Pooled estimates from studies using both home visit and group platforms showed positive results for EIBF (OR 2.13; 95% CI 1.12, 4.05; n = 9), EBF (OR 2.43; 95% CI 1.70, 3.46; n = 12), and less then 5 wasting (OR 0.77; 95% CI 0.67, 0.89; n = 4). Our findings underscore the importance of interpersonal community platforms for improving infant and young child feeding practices and children's nutritional status in LMICs.

Results Compared with HCs, AD patients exhibited global cerebral atrophy in gray matter volume (GMV) and cortical thickness, including frontal, parietal, temporal, occipital, and insular lobes. In addition, insular GMV was negatively correlated with depressive symptoms. Moreover, SVM-based classification achieved an accuracy of 77.5%, a sensitivity of 70%, and a specificity of 85% by leave-one-out cross-validation. Conclusion GMV of the insula displayed atrophy among AD patients, which is associated with depressive symptoms. Our observations provide a potential neural substrate for analysis to examine the co-occurrence of AD with depressive symptoms. Background With recent technology, multivariate time-series electrocardiogram (ECG) analysis has played an important role in diagnosing cardiovascular diseases. However, discovering the association of wide range aging disease and chronic habit with ECG analysis still has room to be explored. This article mainly analyzes the possible relationship between common aging diseases or chorionic habits of medical record and ECG, such as diabetes, obesity, and hypertension, or the habit of smoking. Methods In the research, we first conducted different ECG features, such as those of reduced binary pattern, waveform, and wavelet and then performed a k-means clustering analysis on the correlation between ECGs and the aforementioned diseases and habits, from which it is expected to find a firm association between them and the best characteristics that can be used for future research. Results In summary, we discovered a weak and strong evidence between ECG and medical records. For strong evidence, most patients with diabetes are always assigned into a specified group no matter the number of classes in the k-means clustering, which means we can find their association between them. For weak evidence, smokers, obesity, and hypertension have less unique ECG feature vector, enabling clustering them into specific groups, so the ECGs might be used to identify smokers, obesity, and hypertension. It is also interesting that we found obesity and hypertension, which are thought to be related to cardiovascular system. However, they are not highly correlated in our clustering analysis, which might indirectly tell us that the impact of obesity and hypertension to our body is various. In addition, the clustering effect of waveform feature is better than the other two methods.Amyloid plaques and neurofibrillary tangles (NFTs) are hallmark lesions of Alzheimer's disease (AD) related to β-amyloid (Aβ) deposition and intraneuronal phosphorylated tau (pTau) accumulation. Sortilin C-terminal fragments (shortened as "sorfra") can deposit as senile plaque-like lesions within AD brains. The course and pattern of sorfra plaque formation relative to Aβ and pTau pathogenesis remain unknown. In the present study, cerebral and subcortical sections in postmortem human brains (n = 46) from aged and AD subjects were stained using multiple markers (6E10, β-secretase 1, pTau, and sortilin antibodies, as well as Bielschowsky silver stain). The course and pattern of sorfra plaque formation relative to Thal Aβ and Braak NFT pathogenic stages were determined. Sorfra plaques occurred in the temporal, inferior frontal and occipital neocortices in cases with Thal 1 and Braak III stages. https://www.selleckchem.com/products/JNJ-26481585.html They were also found additionally in the hippocampal formation, amygdala, and associative neocortex in cases with Thal 2-4 and Braak IV-V. Lastly, they were also found in the primary motor, somatosensory, and visual cortices in cases with Thal 4-5 and Braak VI. Unlike Aβ and pTau pathologies, sorfra plaques did not occur in subcortical structures in cases with Aβ/pTau lesions in Thal 3-5/Braak IV-VI stages. We establish here that sorfra plaques are essentially a cerebral proteopathy. We believe that the development of sorfra plaques in both cortical and hippocampal regions proceeds in a typical spatiotemporal pattern, and the stages of cerebral sorfra plaque formation partially overlap with that of Aβ and pTau pathologies.Similar to specific natural language instructions, intention-related natural language queries also play an essential role in our daily life communication. Inspired by the psychology term "affordance" and its applications in Human-Robot interaction, we propose an object affordance-based natural language visual grounding architecture to ground intention-related natural language queries. Formally, we first present an attention-based multi-visual features fusion network to detect object affordances from RGB images. While fusing deep visual features extracted from a pre-trained CNN model with deep texture features encoded by a deep texture encoding network, the presented object affordance detection network takes into account the interaction of the multi-visual features, and reserves the complementary nature of the different features by integrating attention weights learned from sparse representations of the multi-visual features. We train and validate the attention-based object affordance recognition network on a self-built dataset in which a large number of images originate from MSCOCO and ImageNet. Moreover, we introduce an intention semantic extraction module to extract intention semantics from intention-related natural language queries. Finally, we ground intention-related natural language queries by integrating the detected object affordances with the extracted intention semantics. We conduct extensive experiments to validate the performance of the object affordance detection network and the intention-related natural language queries grounding architecture.3D objects (artifacts) are made to fulfill functions. Designing an object often starts with defining a list of functionalities or affordances (action possibilities) that it should provide, known as functional requirements. Today, designing 3D object models is still a slow and difficult activity, with few Computer-Aided Design (***) tools capable to explore the design solution space. The purpose of this study is to explore shape generation conditioned on desired affordances. We introduce an algorithm for generating voxelgrid object shapes which afford the desired functionalities. We follow the principle form follows function, and assume that object forms are related to affordances they provide (their functions). First, we use an artificial neural network to learn a function-to-form mapping from a dataset of affordance-labeled objects. Then, we combine forms providing one or more desired affordances, generating an object shape expected to provide all of them. Finally, we verify in simulation whether the generated object indeed possesses the desired affordances, by defining and executing affordance tests on it.

Today, designing 3D object models is still a slow and difficult activity, with few Computer-Aided Design (CAD) tools capable to explore the design solution space. Finally, we verify in simulation whether the generated object indeed possesses the desired affordances, by defining and executing affordance tests on it.0 Kommentare 0 Geteilt 1 Ansichten 0 Bewertungen -
Host response to infection is a major determinant of disease severity in Ebola virus disease (EVD), but gene expression programs associated with outcome are poorly characterized. Collaborative Cross (CC) **** develop strain-dependent EVD phenotypes of differential severity, ranging from tolerance to lethality. We screen 10 CC lines and identify clinical, virologic, and transcriptomic features that distinguish tolerant from lethal outcomes. Tolerance is associated with tightly regulated induction of immune and inflammatory responses shortly following infection, as well as reduced inflammatory macrophages and increased antigen-presenting cells, B-1 cells, and γδ T cells. Lethal disease is characterized by suppressed early gene expression and reduced lymphocytes, followed by uncontrolled inflammatory signaling, leading to death. We apply machine learning to predict outcomes with 99% accuracy in **** using transcriptomic profiles. This signature predicts outcomes in a cohort of EVD patients from western Africa with 75% accuracy, demonstrating potential clinical utility. While interferon (IFN) responses are critical for mammalian antiviral defense, induction of antiviral RNA interference (RNAi) is evident. To date, individual functions of the mammalian RNAi and micro RNA (miRNA) effector proteins Argonautes 1-4 (AGO1-AGO4) during virus infection remain undetermined. AGO2 was recently implicated in mammalian antiviral defense, so we examined antiviral activity of AGO1, AGO3, or AGO4 in IFN-competent immune cells. Only AGO4-deficient cells are hyper-susceptible to virus infection. AGO4 antiviral function is both IFN dependent and IFN independent, since AGO4 promotes IFN but also maintains antiviral capacity following prevention of IFN signaling or production. We identified AGO-loaded virus-derived short interfering RNAs (vsiRNAs), a molecular marker of antiviral RNAi, in macrophages infected with influenza or influenza lacking the IFN and RNAi suppressor NS1, which are uniquely diminished without AGO4. Importantly, AGO4-deficient influenza-infected **** have significantly higher burden and viral titers in vivo. Together, our data assign an essential role for AGO4 in mammalian antiviral defense. Human brain organoid systems offer unprecedented opportunities to investigate both neurodevelopmental and neurological disease. Single-cell-based transcriptomics or epigenomics have dissected the cellular and molecular heterogeneity in the brain organoids, revealing a complex organization. Similar but distinct protocols from different labs have been applied to generate brain organoids, providing a large resource to perform a comparative analysis of brain developmental processes. Here, we take a systematic approach to compare the single-cell transcriptomes of various human cortical brain organoids together with fetal brain to define the identity of specific cell types and differentiation routes in each method. Importantly, we identify unique developmental programs in each protocol compared to fetal brain, which will be a critical benchmark for the utility of human brain organoids in the future. Reproductive aging in female mammals is an irreversible process associated with declining oocyte quality, which is the rate-limiting factor to fertility. Here, we show that this loss of oocyte quality with age accompanies declining levels of the prominent metabolic cofactor nicotinamide adenine dinucleotide (NAD+). Treatment with the NAD+ metabolic precursor nicotinamide mononucleotide (NMN) rejuvenates oocyte quality in aged animals, leading to restoration in fertility, and this can be recapitulated by transgenic overexpression of the NAD+-dependent deacylase SIRT2, though deletion of this enzyme does not impair oocyte quality. These benefits of NMN extend to the developing embryo, where supplementation reverses the adverse effect of maternal age on developmental milestones. These findings suggest that late-life restoration of NAD+ levels represents an opportunity to rescue female reproductive function in mammals. Health and survival in old age can be improved by changes in gene expression. RNA polymerase (Pol) I is the essential, conserved enzyme whose task is to generate the pre-ribosomal RNA (rRNA). We find that reducing the levels of Pol I activity is sufficient to extend lifespan in the fruit fly. This effect can be recapitulated by partial, adult-restricted inhibition, with both enterocytes and stem cells of the adult midgut emerging as important cell types. In stem cells, Pol I appears to act in the same longevity pathway as Pol III, implicating rRNA synthesis in these cells as the key lifespan determinant. Importantly, reduction in Pol I activity delays broad, age-related impairment and pathology, improving the function of diverse organ systems. Hence, our study shows that Pol I activity in the adult drives systemic, age-related decline in animal health and anticipates mortality. Innate immune signaling has recently been shown to play an important role in nuclear reprogramming, by altering the epigenetic landscape and thereby facilitating transcription. However, the mechanisms that link innate immune activation and metabolic regulation in pluripotent stem cells remain poorly defined, particularly with regard to key molecular components. https://www.selleckchem.com/products/elamipretide-mtp-131.html In this study, we show that hypoxia-inducible factor 1α (HIF1α), a central regulator of adaptation to limiting oxygen tension, is an unexpected but crucial regulator of innate immune-mediated nuclear reprogramming. HIF1α is dramatically upregulated as a consequence of Toll-like receptor 3 (TLR3) signaling and is necessary for efficient induction of pluripotency and transdifferentiation. Bioenergetics studies reveal that HIF1α regulates the reconfiguration of innate immune-mediated reprogramming through its well-established role in throwing a glycolytic switch. We believe that results from these studies can help us better understand the influence of immune signaling in tissue regeneration and lead to new therapeutic strategies. RNA decay is crucial for mRNA turnover and surveillance and misregulated in many diseases. This complex system is challenging to study, particularly in mammals, where it remains unclear whether decay pathways perform specialized versus redundant roles. Cytoplasmic pathways and links to translation are particularly enigmatic. By directly profiling decay factor targets and normal versus aberrant translation in mouse embryonic stem cells (mESCs), we uncovered extensive decay pathway specialization and crosstalk with translation. XRN1 (5'-3') mediates cytoplasmic bulk mRNA turnover whereas SKIV2L (3'-5') is universally recruited by ribosomes, tackling aberrant translation and sometimes modulating mRNA abundance. Further exploring translation surveillance revealed AVEN and FOCAD as SKIV2L interactors. AVEN prevents ribosome stalls at structured regions, which otherwise require SKIV2L for clearance. This pathway is crucial for histone translation, upstream open reading frame (uORF) regulation, and counteracting ribosome arrest on small ORFs.

5. In addition to the aspects mentioned above, a possibility to manipulate the aggregation downstream of the reactor is to be presented as well. We found that applying electrical charges to the aerosol particles (in opposite polarity) can significantly foster aggregation.A single crystal chemical vapor deposition diamond-based microdosimeter prototype featuring an array of micro-sensitive volumes (μSVs) and surrounded by a so-called guard ring (GR) electrode has been fabricated using various microfabrication techniques available at Diamond Sensors Laboratory of CEA, Saclay. The GR microdosimeter was irradiated by a raster scanning method with 2 MeV proton microbeams. The charge transport properties of the GR sensor were determined with sub-micron spatial resolution by measuring the charge collection efficiency (CCE), the μSV geometry, and the pulse-height spectra. The response of the microdosimeter showed a well-defined and homogeneously active μSV. Appropriate biasing of the μSV structures led toward a full CCE for protons with lineal energies of ∼46 keV/μm. This shows the GR microdosimeter's great potential for applications in microdosimetry in clinical beam conditions. We have designed and prototyped the process steps for the batch production of large-area micro-channel-plate photomultipliers (MCP-PMT) using the "air-transfer" assembly process developed with single LAPPDTM modules. We have also synthesized a photocathode in a large-area low-aspect-ratio volume and have shown that the micro-channel plates recover their functionality after cathode synthesis. These steps inform a design for a multi-module batch facility employing dual nested low-vacuum and ultra-high-vacuum systems in a small-footprint. Accurate measurements of thermal conductivity at high temperatures are required for the modeling and design of these systems, but commonly employed time-domain measurements can have errors from convection, corrosion, and ambient temperature fluctuations. Here, we describe the development of a frequency-domain hot-wire technique capable of accurately measuring the thermal conductivity of solid and molten compounds from room temperature up to 800 °C. By operating in the frequency-domain, we can lock into the harmonic thermal response of the material and reject the influence of ambient temperature fluctuations, and we can keep the probed volume below 1 µl to minimize convection. The design of the microfabricated hot-wire sensor, electrical systems, and insulating wire coating to protect against corrosion is covered in detail. Furthermore, we discuss the development of a full three-dimensional multilayer thermal model that accounts for both radial conduction into the sample and axial conduction along the wire and the effect of wire coatings. The 3D, multilayer model facilitates the measurement of small sample volumes important for material development. A sensitivity analysis and an error propagation calculation of the frequency-domain thermal model are performed to demonstrate what factors are most important for thermal conductivity measurements. Finally, we show thermal conductivity measurements including model data fitting on gas (argon), solid (sulfur), and molten substances over a range of temperatures. We present a novel and thorough simulation technique to understand image charge generated from charged particles on a printed-circuit-board detector. Automated deposition of a fresh laser-target film before each illumination leads to good repeatability of the LIPP and thus allows for the detection of time-resolved changes in the space-charge distribution over many hours. We describe and discuss the experimental setup and its features, compare the repeatability of the LIPP measurements on the same sample without and with re-preparation of the test cell, and present the time-resolved evolution of the space-charge profile in a two-layer arrangement of a silicone-grease and a silicone-elastomer film as an example. Finally, the temperature dependence of the space-charge evolution during polarization under high voltage and during depolarization in short circuit is shown. Possible uses and future developments of the new LIPP approach are also discussed.

Conclusions Conventional TMS and particularly triple-stimulation technique studies are helpful in disclosing upper motor neuron dysfunction in ALS. The results of this study might favor the cortical hypothesis for split hand in ALS, but they revealed no significant indication for upper motor neuron dysfunction in postpoliomyelitis syndrome.Purpose Research has demonstrated distinct patterns of electroencephalography (EEG) coherence with attention-deficit/hyperactivity disorder (ADHD) in children; however, few studies have examined coherence measures in adults with ADHD. Further, specific research examining EEG coherence within the subgroup of young adults in post-secondary education is absent from the literature. Methods The current study investigated the utility of quantitative EEG in predicting ADHD behavioral symptoms in 35 college students with a documented ADHD diagnosis and 35 control students. Results Increased interhemispheric frontal beta-wave coherence was found in individuals with ADHD. Logistic regression of principle components of quantitative EEG coherence metrics predicted ADHD group membership. Conclusions Suggestions for improving ADHD identification in college populations using quantitative EEG are discussed.Purpose Comprehensive evaluation of the upstream sensory processing in diabetic symmetrical polyneuropathy (DSPN) is sparse. The authors investigated the spinal nociceptive withdrawal reflex and the related elicited somatosensory evoked cortical potentials. They hypothesized that DSPN induces alterations in spinal and supraspinal sensory-motor processing compared with age- and gender-matched healthy controls. Methods In this study, 48 patients with type 1 diabetes and DSPN were compared with 21 healthy controls. Perception and reflex thresholds were determined and subjects received electrical stimulations on the plantar site of the foot at three stimulation intensities to evoke a nociceptive withdrawal reflex. Electromyogram and EEG were recorded for analysis. Results Patients with DSPN had higher perception (P less then 0.001) and reflex (P = 0.012) thresholds. Fewer patients completed the recording session compared with healthy controls (34/48 vs. 21/21; P = 0.004). Diabetic symmetrical polyneuropathy reduced the odds ratio of a successful elicited nociceptive withdrawal reflex (odds ratio = 0.045; P = 0.014). Diabetic symmetrical polyneuropathy changed the evoked potentials (F = 2.86; P = 0.025), and post hoc test revealed reduction of amplitude (-3.72 mV; P = 0.021) and prolonged latencies (15.1 ms; P = 0.013) of the N1 peak. Conclusions The study revealed that patients with type 1 diabetes and DSPN have significantly changed spinal and supraspinal processing of the somatosensory input. https://www.selleckchem.com/products/Rapamycin.html This implies that DSPN induces widespread differences in the central nervous system processing of afferent A-δ and A-β fiber input. These differences in processing may potentially lead to identification of subgroups with different stages of small fiber neuropathy and ultimately differentiated treatments.Purpose Epileptic seizures (ES) and psychogenic nonepileptic seizures (PNES) are difficult to differentiate when based on a patient's self-reported symptoms. This study proposes review of objective data captured by a surface electromyography (sEMG) wearable device for classification of events as ES or PNES. This may help clinicians accurately identify ES and PNES. Methods Seventy-one subjects were prospectively enrolled across epilepsy monitoring units at VA Epilepsy Centers of Excellence. Subjects were concomitantly monitored using video EEG and a wearable sEMG epilepsy monitor, the Sensing Portable sEmg Analysis Characterization (SPEAC) System. Three epileptologists independently classified ES and PNES that contained upper extremity motor activity based on video EEG. The sEMG data from those events were automatically processed to provide a seizure score for event classification. After brief training (60 minutes), the sEMG data were reviewed by a separate group of four epileptologists to independently classify events as ES or PNES. Results According to video EEG review, 17 subjects experienced 34 events (15 ES and 19 PNES with upper extremity motor activity). The automated process correctly classified 87% of ES (positive predictive value = 88%, negative predictive value = 76%) and 79% of PNES, and the expert reviewers correctly classified 77% of ES (positive predictive value = 94%, negative predictive value = 84%) and 96% of PNES. The automated process and the expert reviewers correctly classified 100% of tonic-clonic seizures as ES, and 71 and 50%, respectively, of non-tonic-clonic ES. Conclusions Automated and expert review, particularly in combination, of sEMG captured by a wearable seizure monitor (SPEAC System) may be able to differentiate ES (especially tonic-clonic) and PNES with upper extremity motor activity.Introduction Inflammatory myopathies are treated with glucocorticoids and other immunosuppressive medications. Intravenous immunoglobulin (IVIG) is increasingly used for refractory or severe cases; however, the evidence for their effectiveness is limited. We assessed effectiveness and safety of IVIG when used with other immunomodulatory agents in the treatment of inflammatory myopathies. Methods This study reviewed records of patients diagnosed with dermatomyositis or polymyositis and treated with IVIG, from 2009 through 2016 in 1 tertiary medical center. Mixed-effects general linear regression models were applied to determine effectiveness of treatment on muscle strength, creatinine phosphokinase levels, and steroid dosage. Results Twenty-three patients with dermatomyositis/polymyositis treated with IVIG were followed up for a mean of 31 (SD, ±25) months. During this period, a significant improvement in muscle strength was demonstrated, with a mean increase of 0.92 Medical Research Council scale points (β = 0.14; confidence interval [CI], 0.136-0.149; p less then 0.0001), a significant reduction of creatinine phosphokinase levels and steroid dosage with a mean decrease of 1140 IU/L (β = -0.274; CI, -0.354 to -0.195; p less then 0.0001), and 36 mg/d (β = -0.008; CI, -0.011 to -0.006; p less then 0.0001), respectively. Overall, remission was observed in 10 patients (43.5%), and partial remission in 6 patients (26%), whereas 1 patient (17%) remained refractory to treatment, and 6 patients (27%) were lost to follow-up. Conclusions The majority of patients with inflammatory myopathies experienced a clinical and laboratory improvement during IVIG treatment. In addition, a steroid-sparing effect was noticed in most patients. These results encourage the use of IVIG as an alternative treatment option for patients with limited responsiveness to conventional methods.

These results encourage the use of IVIG as an alternative treatment option for patients with limited responsiveness to conventional methods.0 Kommentare 0 Geteilt 1 Ansichten 0 Bewertungen -
Finally, we validated a role for SP3 and MAZ in the formation of hemogenic endothelium. Our data and computational analyses provide a useful resource for uncovering regulators of HSC formation.Polymerases and exonucleases act on 3' ends of nascent RNAs to promote their maturation or degradation but how the balance between these activities is controlled to dictate the fates of cellular RNAs remains poorly understood. Here, we identify a central role for the human DEDD deadenylase TOE1 in distinguishing the fates of small nuclear (sn)RNAs of the spliceosome from unstable genome-encoded snRNA variants. We found that TOE1 promotes maturation of all regular RNA polymerase II transcribed snRNAs of the major and minor spliceosomes by removing posttranscriptional oligo(A) tails, trimming 3' ends, and preventing nuclear exosome targeting. In contrast, TOE1 promotes little to no maturation of tested U1 variant snRNAs, which are instead targeted by the nuclear exosome. These observations suggest that TOE1 is positioned at the center of a 3' end quality control pathway that selectively promotes maturation and stability of regular snRNAs while leaving snRNA variants unprocessed and exposed to degradation in what could be a widespread mechanism of RNA quality control given the large number of noncoding RNAs processed by DEDD deadenylases.Transcription by RNA polymerase II (RNAPII) is a dynamic process with frequent variations in the elongation rate. However, the physiological relevance of variations in RNAPII elongation kinetics has remained unclear. Here we show in yeast that a RNAPII mutant that reduces the transcription elongation rate causes widespread changes in alternative polyadenylation (APA). https://www.selleckchem.com/products/JNJ-26481585.html We unveil two mechanisms by which APA affects gene expression in the slow mutant 3' UTR shortening and gene derepression by premature transcription termination of upstream interfering noncoding RNAs. Strikingly, the genes affected by these mechanisms are enriched for functions involved in phosphate uptake and purine synthesis, processes essential for maintenance of the intracellular nucleotide pool. As nucleotide concentration regulates transcription elongation, our findings argue that RNAPII is a sensor of nucleotide availability and that genes important for nucleotide pool maintenance have adopted regulatory mechanisms responsive to reduced rates of transcription elongation. The Caribbean was one of the last regions of the Americas to be settled by humans, but how, when, and from where they reached the islands remains unclear. We generated genome-wide data for 93 ancient Caribbean islanders dating between 3200-400 cal. BP and find evidence of at least three separate dispersals into the region, including two early dispersals into the Western Caribbean, one of which seems connected to radiation events in North America. This was followed by a later expansion from South America. We also detect genetic differences between the early settlers and the newcomers from South America with almost no evidence of admixture. Our results add to our understanding of the initial peopling of the Caribbean and the movements of Archaic Age peoples in the Americas.Hot-carriers in plasmonic nanostructures, generated via plasmon decay, play key roles in applications like photocatalysis and in photodetectors that circumvent band-gap limitations. However, direct experimental quantification of steady-state energy distributions of hot-carriers in nanostructures has so far been lacking. We present transport measurements from single-molecule junctions, created by trapping suitably chosen single molecules between an ultra-thin gold film supporting surface plasmon polaritons and a scanning probe tip, that can provide quantification of plasmonic hot-carrier distributions. Our results show that Landau damping is the dominant physical mechanism of hot-carrier generation in nanoscale systems with strong confinement. The technique developed in this work will enable quantification of plasmonic hot-carrier distributions in nanophotonic and plasmonic devices. Background Population mean GFR is lower in older age, but it is unknown whether healthy aging is associated with preserved rather than lower GFR in some individuals. Methods We investigated the cross-sectional association between measured GFR, age, and health in persons aged 50-97 years in the general population through a meta-analysis of iohexol clearance measurements in three large European population-based cohorts. We defined a healthy person as having no major chronic disease or risk factors for CKD and all others as unhealthy. We used a generalized additive model to study GFR distribution by age according to health status. Results There were 935 (22%) GFR measurements in persons who were healthy and 3274 (78%) in persons who were unhealthy. The mean GFR was lower in older age by -0.72 ml/min per 1.73 m2 per year (95% confidence interval [95% CI], -0.96 to -0.48) for men who were healthy versus -1.03 ml/min per 1.73 m2 per year (95% CI, -1.25 to -0.80) for men who were unhealthy, and by -0.92 ml/min per 1.73 m2 per year (95% CI, -1.14 to -0.70) for women who were healthy versus -1.22 ml/min per 1.73 m2 per year (95% CI, -1.43 to -1.02) for women who were unhealthy. For healthy and unhealthy people of both sexes, both the 97.5th and 2.5th GFR percentiles exhibited a negative linear association with age. Conclusions Healthy aging is associated with a higher mean GFR compared with unhealthy aging. However, both the mean and 97.5 percentiles of the GFR distribution are lower in older persons who are healthy than in middle-aged persons who are healthy. This suggests that healthy aging is not associated with preserved GFR in old age. Objectives Although persistent human papillomavirus (HPV) infection is a major cause of cervical squamous intra-epithelial neoplasia, the relationship between vaginal microbiota and different grades of squamous intra-epithelial neoplasia is not well established. We explored the possible relationship between the vaginal microbiota and the progression of cervical squamous intra-epithelial neoplasia. Methods We evaluated 69 women who attended the Obstetrics and Gynecology Hospital of Fudan University. The vaginal bacterial composition of three groups of women was characterized by deep sequencing of bar-coded 16S rRNA gene fragments (V3-4) using Illumina MiSeq. Exclusion criteria were any previous hysterectomy, history of cervical or other lower genital cancer, and/or destructive therapy of the cervix. Women who had autoimmune disorders, who were HIV positive, who received antibiotics within 15 days of sampling, or who had engaged in sexual intercourse or douching within 48 hours prior to sampling were also excluded.

Many of these newly-born cells differentiated into active neurons and glia. LacZ-expressing V-SVZ NSPCs also showed extensive migration towards the non-neurogenic regions ipsilateral to the lesion, and expressed the neuronal marker NeuN. https://www.selleckchem.com/products/avelestat-azd9668.html NeuroD1+/GFP+ V-SVZ NSPCs almost differentiated into neurons in the peri-infarct regions. Conclusion HI promotes the establishment of a substantial number of new neurons in non-neurogenic regions, suggesting intrinsic repair mechanisms of the brain, by controlling the behavior of endogenous NSPCs. The activation of NeuroD1 expression may improve the therapeutic potential of endogenous NSPCs by increasing their neuronal differentiation in HI.Purpose Cardiac power (CP) index is a product of mean arterial pressure (MAP) and cardiac output (CO). In aortic stenosis, however, MAP is not reflective of true left ventricular (LV) afterload. We evaluated the utility of a gradient-adjusted CP (GCP) index in predicting survival after transcatheter aortic valve replacement (TAVR), compared to CP alone. Materials and methods We included 975 patients who underwent TAVR with 1 year of follow-up. CP was calculated as (CO×MAP)/[451×body surface area (BSA)] (W/m²). GCP was calculated using augmented MAP by adding aortic valve mean gradient (AVMG) to systolic blood pressure (CP1), adding aortic valve maximal instantaneous gradient to systolic blood pressure (CP2), and adding AVMG to MAP (CP3). A multivariate Cox regression analysis was performed adjusting for baseline covariates. Receiver operator curves (ROC) for CP and GCP were calculated to predict survival after TAVR. Results The mortality rate at 1 year was 16%. The mean age and AVMG of the survivors were 81±9 years and 43±4 mm Hg versus 80±9 years and 42±13 mm Hg in the deceased group. The proportions of female patients were similar in both groups (p=0.7). Both CP and GCP were independently associated with survival at 1 year. The area under ROCs for CP, CP1, CP2, and CP3 were 0.67 [95% confidence interval (CI), 0.62-0.72], 0.65 (95% CI, 0.60-0.70), 0.66 (95% CI, 0.61-0.71), and 0.63 (95% CI 0.58-0.68), respectively. Conclusion GCP did not improve the accuracy of predicting survival post TAVR at 1 year, compared to CP alone.Purpose Gastric cancer (GC) has a very poor prognosis when diagnosed at a late stage. Acyl-CoA thioesterase 7 (ACOT7) is a major isoform of the acyl coenzyme family that catalyzes the hydrolysis of fatty acyl-CoAs into unesterified free fatty acid and coenzyme A. The purpose of this study was to investigate the expression levels of ACOT7 in GC and mechanisms related therewith. Materials and methods Screening of systematic biology studies revealed ACOT7 as a key gene in GC, as well as involvement of the long non-coding RNA NMRAL2P in ACOT7 expression. In this study, GC tissues and adjacent tissue samples were obtained from 10 GC patients at the Department of Gastrointestinal Surgery. GES1 and SGC-7901 cells were collected and treated to silence ACOT7 and overexpress NMRAL2P. The expressions of ACOT7 and NMRAL2P were detected by real-time quantitative PCR and Western blot. Additionally, cell proliferation, apoptosis, migration, and invasion were examined. Results ACOT7 was upregulated in gastric tumor tissues and GC cell lines. ACOT7 gene silencing induced a less malignant phenotype and was closely correlated to reduced cell proliferation and migration, altered cell cycle, and increased apoptosis. Furthermore, NMRAL2P was downregulated in tumor tissues and GC cell lines. NMRAL2P overexpression induced a more malignant phenotype and significantly inhibited the expression of ACOT7. Importantly, NMRAL2P indirectly methylated ACOT7 by binding to DNMT3b, thereby suppressing ACOT7 expression. Conclusion NMRAL2P activation suppresses ACOT7 expression in GC. Thus, ACOT7 could be a promising target for the treatment of GC.Purpose MicroRNAs play key regulatory roles in the tumorigenesis of hepatitis B virus-related hepatocellular carcinoma (HBV-HCC). This study aimed to explore the regulatory effects of microRNA-98-5p (miR-98-5p) on the proliferation, migration, invasion, and apoptosis of HBV-HCC cells, as well as the underlying mechanisms involving nuclear factor-κB-inducing kinase (NIK). Materials and methods The expressions of miR-98-5p and NIK in HBV-HCC tissues and cells, and the level of HBV DNA in HBV-HCC cells were measured by quantitative real-time polymerase chain reaction (qRT-PCR). The proliferation, migration, invasion, and apoptosis of HBV-HCC cells were analyzed by cell counting kit-8, wound healing, transwell, and flow cytometry assay, respectively. The targeting relationship between miR-98-5p and NIK was predicted by StarBase3.0 and verified by dual-luciferase reporter assay. HBV-HCC xenograft tumor model was constructed in **** to observe the tumor growth in vivo. Results The expression of miR-98-5p was declined in HBV-HCC tissues and cells. Overexpression of miR-98-5p markedly reduced the level of HBV DNA; inhibited the proliferation, migration, and invasion; and promoted the apoptosis of HBV-HCC cells. NIK was a target of miR-98-5p. Overexpression of miR-98-5p markedly decreased the protein expression of NIK in MHCC97H-HBV cells. NIK reversed the tumor-suppressing effect of miR-98-5p on HBV-HCC cells. Furthermore, overexpression of miR-98-5p significantly inhibited the xenograft tumor growth and decreased the expression of NIK in ****. Conclusion MiR-98-5p inhibits the secretion of HBV, proliferation, migration, and invasion of HBV-HCC cells by targeting NIK.Purpose Allogeneic hematopoietic stem cell transplantation (HSCT) with optimal conditioning has helped better long-term survival in acute lymphoblastic leukemia (ALL). This study investigated the efficacy and safety of reduced-intensity conditioning (RIC) with busulfan and fludarabine in adult ALL patients unfit for myeloablation. Materials and methods Records of 78 patients who underwent HSCT with RIC consisting of 3.2 mg/kg/day of busulfan for 2 or 3 days and 30 mg/m²/day of fludarabine for 5 or 6 days were analyzed. Results The median age at diagnosis was 49 years. Over a median follow-up of 22 months, 2-year estimates of relapse-free survival (RFS) and overall survival were 57.4% and 68.7%, respectively. Multivariate analysis showed a trend of improved RFS in patients with chronic graft-versus-host disease (GVHD) (hazard ratio, 0.53; 95% confidence interval, 0.26-1.08; p=0.080). The cumulative incidences of relapse and non-relapse mortality were 42.9% and 19.6%, respectively and one case of central nervous system relapse was noted.

Such clonality could be exploited in the search for early patient-specific therapeutic solutions. The association of glycated hemoglobin (HbA1c) with venous thromboembolism (VTE) and death in the elderly is unknown. In the SWEETCO 65+ study we analyzed prospectively a Swiss Cohort of Elderly Patients with Venous Thromboembolism (SWITCO 65+). 888 patients were enrolled for the SWEETCO 65+ analysis. HbA1c was determined at baseline and divided into three categories (HbA1c 6.5%, diabetic range). Median follow-up was 2.5 years. The primary endpoint was recurrent VTE. Secondary endpoints included all-cause mortality and major bleeds. The total prevalence of diabetes was 22.1%. The risk of recurrent VTE was similar in patients with HbA1c with pre-diabetes (adjusted subhazard ratio (aSHR) 1.07 [0.70 to 1.63]) and diabetes (aSHR 0.73 [0.39 to 1.37]) as compared to those with a HbA1c in the normal range. However, a HbA1c ≥ 6.5% (median IQ range 7.0 [6.70;7.60]) was significantly associated with a higher risk of all-cause mortality (adjusted hazard ratio [aHR] 1.83 [1.21 to 2.75]). In summary we found no association between HbA1c and major bleeding. Elevated HbA1c levels are not associated with recurrent VTE but with increased all-cause mortality in an elderly population with acute VTE.One of the most exciting applications of metaparticles and metasurfaces consists in the magnetic light excitation. However, the principal limitation is due to parasitic extra multipoles of electric family excited in magnetic dipole meta-particles characterized by a radiating nature and corresponding radiating losses. In this paper, we propose the "ideal magnetic dipole" with suppressed additional multipoles except of magnetic dipole moment in the scattered field from a cylindrical object by using mantle cloaking based on metasurface and on anapole concept. The considered metasurface consists of a periodic width modulated microstrip line, with a sinusoidally shaped profile unit cell printed on a dielectric substrate.Random lasers consisting of slab waveguides with two-dimensional disordered wrinkling patterns that act as scattering resonators are reported. As active material 2,2',7,7'-tetraphenyl-9,9'-spirobifluorene is used which is sandwiched between an oxidized silicon wafer and a cladding with higher glass transition temperature. Wrinkles with tailorable periodicity have been induced by thermal annealing. Photopumping experiments show the transition from amplified spontaneous emission to a multiple peak laser spectrum with linewidths as low as 0.1 nm, demonstrating the applicability of this approach for random laser design. The binary blend materials containing the modified recycled rubber powder with maleic a hydride modified polypropylene thermoplastic elastomer were prepared by dynamic vulcanization and blended with a variety of additives such as activated agent, accelerator, solubilizer, and the crosslinking agent. The thermal properties and mechanical properties including tensile strengths and impact strengths of pristine rubber, polypropylene and their corresponding binary blends were investigated. Besides, the effects of the amount of rubber powder, polypropylene, crosslinking agent, accelerator, activator, and solvent were studied and the microstructures of the pristine rubber, pristine polypropylene, and their corresponding binary blends were observed by scanning electron microscopy. It was found that the compatibilizer could effectively disperse the size of 120 mesh of recycled rubber powder into the polypropylene in the same manner and the homogeneous tear section of the rubber/polypropylene thermoplastic elastomer was obtained. The results on the effects of additives on mechanical and morphological properties of recycled rubber/polypropylene binary blends guide the rational design of novel polymeric composites from recycled polymeric materials. To investigate the incidence and risk factors of hepatitis B virus (HBV) reactivation in HBV surface antigen (HBsAg)-/ HBV core antibody (HBcAb)+ patients who underwent rituximab (RTX) therapy for rheumatoid arthritis (RA). From January 2000 through December 2017, a total of 134 RA patients with various HBV serostatuses who received RTX at Dalin Tzu Chi Hospital were screened. Finally, 50 HBsAg-/HBcAb+ patients were enrolled in this retrospective study. https://www.selleckchem.com/products/muramyl-dipeptide.html Baseline characteristics, comedications, and the occurrence of HBV reactivation were recorded. Four HBsAg-/HBcAb+ RA patients (8%; 4/50) experienced HBV reactivation after treatment with RTX. Hepatitis flare-up occurred in 2 of these 4 patients, with a fatal outcome in one. HBV reactivation occurred approximately 1-4 years after the first dose of RTX and 0.5-1.5 years after the last one. In HBsAg-/HBcAb+ patients, HBV reactivation was significantly more common in those who were HBV surface antibody (HBsAb)- at baseline than in those who were HBsAb+ (30% vs 4%; p = 0.02). A history of adalimumab use was associated with HBV reactivation (100% vs 39%; p = 0.02). A moderate risk of HBV reactivation was observed in HBsAg-/HBcAb+ RA patients receiving RTX therapy. The reactivation may induce acute hepatitis and even death. To reduce the risk of HBV reactivation, regular monitoring of liver function is insufficient; monitoring of viral load and HBsAg or prophylaxis with antiviral therapy should be considered.Sustaining an accurate, timely, and global tsunami forecast system remains a challenge for scientific communities. To this end, various viable geophysical monitoring devices have been deployed. However, it is difficult to implement new observation networks in other regions and maintaining the existing systems is costly. This study proposes a new and complementary approach to monitoring the tsunami using existing moving platforms. The proposed system consists of a radar altimeter, Global Navigation Satellite Systems receiver, and an adequate communication link on airborne platforms such as commercial airplanes, drones, or dedicated high-speed aircraft, and a data assimilation module with a deterministic model. We demonstrated, through twin-data experiment, the feasibility of the proposed system in forecasting tsunami at the Nankai Trough of Japan. Our results demonstrated the potential of an airborne tsunami observation as a viable future technology through proxy observations and rigorous numerical experiments.
