Using Synthetic Dosage Lethality screening, "sentinel" yeast strains are identified that are sensitive to overexpression of a human disease gene. Variants of the gene can then be functionalized in high-throughput fashion through simple growth assays using either solid or liquid media. Sentinel Interaction Mapping (SIM) has the potential to create functional assays for the large majority of human disease genes that do not have a yeast orthologue. Using the tumour suppressor gene PTEN as an example, we show that SIM assays can provide a fast and economical means to screen a large number of genetic variants.Contrast-enhanced X-ray imaging provides a non-destructive and flexible approach to optimising contrast in soft tissues, especially when incorporated with Lugol's solution (1% I2, 2% KI), a technique currently referred to as diffusible-iodine contrast-enhanced computed tomography (diceCT). The aim of this optimisation procedure is to provide suitable contrast between neural tissue and background tissue(s) of the head, thereby facilitating digital segmentation and volumetric analysis of the central nervous system. Graph theoretical analysis further explicated the organizational principles of the network. The olfactory network exhibits a modular composition of three (i.e., the sensory, limbic, and frontal) subnetworks and demonstrates strong otions, autonomic tone, and feeding, beyond basic sensory perception. However, it is unclear how the neuroanatomy of olfaction is organized in humans to subserve these functions. Functional connectivity analysis of the HCP dataset combined with graph theoretical analysis revealed an optimized large-scale network consisting of three subnetworks-the sensory, limbic, and frontal subnetworks. Distributed across frontal and temporal lobes in well segregated fashion, these olfactory structures are also highly integrated, linked through hub nodes of the amygdala and anterior insula. HIV trans-activator of transcription (Tat) protein is one such neurotoxin that is thought to play a major role in the neuropathogenesis of HAND. The endocannabinoid (eCB) system provides on-demand neuroprotection against excitotoxicity, and exogenous cannabinoids attenuate neurotoxicity in animal models of HAND. Whether this neuroprotective system is altered in the presence of HIV is unknown. Here, we examined the effects of Tat on the eCB system in rat primary hippocampal cultures. Using whole-cell patch-clamp electrophysiology, we measured changes in retrograde eCB signaling following exposure to Tat. Treatment with Tat significantly reduced the magnitude of depolarization-induced suppression of excitation (DSE) in a graded manner over the course of 48 h. Interestingly, Tat did not alter this form of short-term synaptic plasticity at inhibitory terminals. The Tat-induced decrease in eCB signaling resulted from impaired CB1 receptor (CB1R)-mediated presynaptic inhibition of glutamate release. " Additionally, by the end of the workshop, participants had a statistically significant increase in self-perceptions regarding the importance of improving diversity and recognizing biases and stereotypes in graduate education, knowing what to say when interacting with people from different cultures, and the ability to acknowledge bias when mentoring students from groups underrepresented in the biomedical field. This preliminary initiative was successful in the introduction of faculty and staff to the importance of fostering an inclusive academic environment and thereby developing a diverse workforce.Access to knowledge has never been easier in the internet age, and so it is important that students develop skills to discriminate undependable information from reliably investigated research. We have created an exercise that teaches good research practice by exploring the history, ethics, and design of clinical trials. Students apply their understanding of these principles through an assessed systematic review and meta-analysis (SRMA) exercise. Here, a clinically themed hypothesis is tested using a structured literature search in conjunction with an eligibility matrix to map study design, ethics, subject selection, randomization and blinding, methodological standards, study power, and other potential sources of interstudy heterogeneity. Data extracted from selected studies are used to produce a forest plot with an aggregated effect size, confidence range, and measure of interstudy heterogeneity. A funnel plot is then used in conjunction with the eligibility matrix to evaluate study bias tendency, and, in this way, students reflect on the factors that promote disparate conclusion-making among studies with a common research focus. This exercise produced a normally distributed grade-profile across three academic-year cohorts, and comparison of individual exercise grade with year-long aggregated average suggested students who performed less well on conventional assignments engaged successfully with the systematic nature of this assessment. Those opting to use this format for their final-year capstone project also performed above their grade point average from the preceding year. We suggest that SRMA offers a readily applied method for students to quantitatively explore how differences in experimental research practices influence study dependability.Physiology students grapple with large amounts of subject content and hence memorize facts to pass examinations. In parallel, students display limited critical-thinking and creative skills, integration abilities, and/or a deeper engagement with subject content. This study aimed to assess the feasibility of introducing active learning methods (cooperative learning) in a relatively large class to final-year undergraduate physiology students (Bachelor of Science stream) at Stellenbosch University in South Africa. An assignment designed to enhance active and engaged learning was made available to the students (n = 225) during the second week of a 5-wk cardiovascular physiology series of lectures. Students were instructed to freely form working groups (n = 3/group) and the assignment was due by the end of the module. Student groups were expected and encouraged to continuously work on the assignment (outside class time). Three cooperative learning slots were also created during class time, with the lecturer and postgraduate students acting as guides. After the module, students anonymously completed an electronic questionnaire. This study revealed three major findings in terms of implementing cooperative learning in large classes within a South African context, i.e., 1) relatively good reception by students with some indication of group work; 2) it is logistically feasible in relatively large classes, although adequate support is crucial; and 3) additional measures need to be adopted to ensure its success.The relationship between pH, pKa, and degree of local anesthetic ionization is quantified by the Henderson-Hasselbalch equation. As presented in standard textbooks, the effect of pH on the degree of ionization of any particular local anesthetic is not immediately clear due to the x-axis displaying pH - pKa, which requires conversion to pH, based on the pKa for each local anesthetic, a complex process. We present a graphical solution that clarifies the interrelationships between pH, pKa, and degree of ionization by plotting pKa on the x-axis versus the percentage of unionized local anesthetic on the y-axis. In a qualitative, descriptive case study involving 21 participants, we examined the sexual experiences-during childbirth and postpartum-of women who had a waterbirth in a hospital environment. Women discussed their sexuality in the intrapartum and postpartum period after having a waterbirth in public or private hospitals. Two main themes were developed from the data analysis 'women's sexual pleasure during waterbirth' and 'sexuality after childbirth'. Experiences included transcendence during waterbirth, a feeling that it represented a unique experience, and couples' more positive feelings about intimacy. The shared representations of women suggest that waterbirth can be a calming, joyful event for couples. During this process, they felt that it was a loving, shared event. In addition, after childbirth women experienced positive feelings toward their bodies and sexuality (including orgasm), as well as closer intimacy with their partners.

The first aim of this study was to synthesize and characterize reed-based-beads (BBR), an enhanced adsorbent from Tunisian reed. The second purpose was to evaluate and optimize the BBR efficiency for the simultaneous removal of oxytetracycline (OTC) and cadmium (Cd(II)), using central composite design under response surface methodology. The third goal was to elucidate the biosorption mechanisms taking place. It was shown that under optimum conditions (4.19 g L-1 of BBR, 165.54 μmol L-1 of OTC, 362.16 μmol L-1 of Cd(II), pH of 6, and 25.14-h contact time) the highest adsorption percentages (63.66% for OTC and 99.99% for Cd(II)) were obtained. It was revealed that OTC adsorption mechanism was better described by Brouers-Sotolongo fractal equation, with regression coefficient (R2) of 0.99876, and a Person's chi-square (χ2) of 0.01132. The Weibull kinetic equation better explained Cd(II) biosorption (R2 = 0.99959 and χ2 = 0.00194). FTIR and isotherm studies confirmed that the BBR surface was heterogeneous, and that adsorption mechanisms were better described by the Freundlich/Jovanovich equation (R2 = 0.99276 and χ2 = 0.04864) for OTC adsorption, and by the Brouers-Sotolongo model (R2 = 0.9851 and χ2 = 0.77547) for Cd(II) biosorption. Overall results indicate that, at last, the BBR lignocellulosic biocomposite beads could be considered as cost-effective and efficient adsorbent, which could be of socioeconomic and environmental relevance. In vitro fertilization and maturation can improve sheep productivity. The aims of this study were to evaluate the effects of honey bee addition as a supplementation medium on in vitro maturation improvement, gene expression of matured sheep oocytes, and determine the optimum concentration from honey bee for in vitro maturation of sheep oocytes. Cumulus oocyte complexes were obtained from the ovaries of slaughtered female sheep. Grade A and B oocytes were cultured for 24 h in medium without honey bee (control, G1) or medium supplemented with 5% (G2), 10% (G3), or 20% (G4) honey bee. Oocyte maturation rate, glutathione concentration, and the expression of candidate genes (GDF-9, BAX, Cyclin B, C-MOS, IGF1) were determined in the matured oocytes. The maturation rate of sheep oocyte was better in the presence of 5% and 10% honey bee; the mean number of oocytes in metaphase II stage was higher than that in G1 and G4 groups. Glutathione concentration was highest in G2 (10.93 ± 0.57). In general, gene expression levels were similar in G2 and G3, which were greater that in G1 and G4. In conclusion, the optimal concentration of black seeds honey bee that can be added to the maturation medium is 5% to obtain the highest mean MII and glutathione concentration values, and to improve gene expression in in vitro matured sheep oocytes.Thermal barrier coating (TBC) implementations and oxygenated additives are remarkable issues that may decrease the exhaust emissions of engines. This study examines the effect of chromium oxide (Cr2O3) coating and the addition of ethylhexyl nitrate (EHN) on exhaust emissions of a diesel engine. In addition, an artificial neural network (ANN) model was designed which estimates exhaust emissions based on engine speed in order to reduce time, labor, and costs lost in experimental studies, and the performance of the ANN was evaluated. Piston crown and valves of engine were processed with Cr2O3. The E3, E6, and E9 blends were produced by blending 3%, 6%, and 9% (vol.) ratios of 2-ethylhexyl nitrate with diesel fuel. Engine speed was used as input parameter and carbon monoxide (CO), nitrogen oxide (NOX), hydrocarbon (HC), and smoke density were used as output parameters. To evaluate the performance of ANN, error rates, and regression (R) values were considered. Experimental results revealed that CO, HC, and smoke density decreased in the CE whereas NOX values increased compared with the UE. The addition of EHN reduced NOX emission and smoke density, whereas it increased CO and HC emissions. The result showed that ANN model can predict the exhaust emissions at a high accuracy rate. The first aim of this study was to synthesize and characterize reed-based-beads (BBR), an enhanced adsorbent from Tunisian reed. The second purpose was to evaluate and optimize the BBR efficiency for the simultaneous removal of oxytetracycline (OTC) and cadmium (Cd(II)), using central composite design under response surface methodology. The third goal was to elucidate the biosorption mechanisms taking place. It was shown that under optimum conditions (4.19 g L-1 of BBR, 165.54 μmol L-1 of OTC, 362.16 μmol L-1 of Cd(II), pH of 6, and 25.14-h contact time) the highest adsorption percentages (63.66% for OTC and 99.99% for Cd(II)) were obtained. It was revealed that OTC adsorption mechanism was better described by Brouers-Sotolongo fractal equation, with regression coefficient (R2) of 0.99876, and a Person's chi-square (χ2) of 0.01132. The Weibull kinetic equation better explained Cd(II) biosorption (R2 = 0.99959 and χ2 = 0.00194). FTIR and isotherm studies confirmed that the BBR surface was heterogeneous, and that adsorption mechanisms were better described by the Freundlich/Jovanovich equation (R2 = 0.99276 and χ2 = 0.04864) for OTC adsorption, and by the Brouers-Sotolongo model (R2 = 0.9851 and χ2 = 0.77547) for Cd(II) biosorption. Cumulus oocyte complexes were obtained from the ovaries of slaughtered female sheep. Grade A and B oocytes were cultured for 24 h in medium without honey bee (control, G1) or medium supplemented with 5% (G2), 10% (G3), or 20% (G4) honey bee. Oocyte maturation rate, glutathione concentration, and the expression of candidate genes (GDF-9, BAX, Cyclin B, C-MOS, IGF1) were determined in the matured oocytes. The maturation rate of sheep oocyte was better in the presence of 5% and 10% honey bee; the mean number of oocytes in metaphase II stage was higher than that in G1 and G4 groups. Glutathione concentration was highest in G2 (10.93 ± 0.57). In general, gene expression levels were similar in G2 and G3, which were greater that in G1 and G4. This study examines the effect of chromium oxide (Cr2O3) coating and the addition of ethylhexyl nitrate (EHN) on exhaust emissions of a diesel engine. In addition, an artificial neural network (ANN) model was designed which estimates exhaust emissions based on engine speed in order to reduce time, labor, and costs lost in experimental studies, and the performance of the ANN was evaluated. Piston crown and valves of engine were processed with Cr2O3. The E3, E6, and E9 blends were produced by blending 3%, 6%, and 9% (vol.) ratios of 2-ethylhexyl nitrate with diesel fuel. Engine speed was used as input parameter and carbon monoxide (CO), nitrogen oxide (NOX), hydrocarbon (HC), and smoke density were used as output parameters. To evaluate the performance of ANN, error rates, and regression (R) values were considered. Experimental results revealed that CO, HC, and smoke density decreased in the CE whereas NOX values increased compared with the UE. The addition of EHN reduced NOX emission and smoke density, whereas it increased CO and HC emissions. The result showed that ANN model can predict the exhaust emissions at a high accuracy rate. The results showed that TMD subjects with vision disorders that are corrected with standard glasses present EMG values that are significantly higher than those presented by non-TMD subjects with vision disorders and standard glasses. Infact, in TMD subjects, eye correction did not have a positive effect on the stomatognathic or pericranial musculature. Interferon regulatory factor 8 (IRF8), a critical transcription factor, was reported to be involved in the modulation of neuropathic pain. Here, we focused on exploring whether 2 Hz EA stimulation exerts an inhibitory action on spinal IRF8 in SNI rats. Methods In this study, SNI rats were treated with 2 Hz EA once every other day for 21 days. Paw withdrawal threshold (PWT) was applied to determine the analgesic effect of 2 Hz EA on SNI rats. The spinal IRF8 and CX3CRl expressions were detected with qRT-PCR and western blot, and immunofluorescence staining was used to evaluate colocation of IRF8 or CX3CRl with microglial activation marker CD11b in the spinal cord. Results It was found that SNI induced significant elevation of spinal IRF8 and CX3CRl mRNA and protein expression. Additionally, immunofluorescence results showed that SNI elicited the coexpression of IRF8 with CD11b, as well as CX3CRl with CD11b in the spinal cord. Meanwhile, 2 Hz EA treatment of SNI rats not only reduced IRF8 and CX3CRl mRNA and protein expression, but also reversed the coexpression of IRF8 or CX3CRl with CD11b in the spinal cord, along with an attenuation of SNI-evoked mechanical hypersensitivity. Remdesivir is one of the most promising drugs to treat COVID-19 based on the following facts remdesivir has a broad-spectrum antiviral mechanism of action; it demonstrated in vitro activity against SARS-CoV-2 and in vivo efficacy in animal models against the similar coronavirus MERS-CoV; its safety profile has been tested in Ebola patients and in compassionate use in COVID-19 patients. Currently, remdesivir is being investigated in ten randomized controlled trials against COVID-19. The dose regimen of remdesivir is an IV loading dose of 200 mg on day 1 followed by daily IV maintenance doses of 100 mg for 5-9 days. Based on our data analysis, however, remdesivir with IV administration alone is unlikely to achieve excellent clinical efficacy. This analysis is based on the following observations plasma exposures of remdesivir and its active metabolite are unlikely to be correlated with its clinical efficacy; remdesivir and its active metabolites are unlikely to be adequate in the lung to kill the SARS-CoV-2 virus. Even if remdesivir demonstrates benefits in the current randomized controlled trials, its efficacy may be limited. We suggest that a combination of an IV and pulmonary delivery dose regimen should be studied immediately to realize a potentially more effective antiviral therapy against COVID-19. Predictors of DCI-related cerebral infarction (new CT hypodensities developed within the first 3 weeks not related to other causes) were studied. Vasospasm and brain hypoperfusion were studied with CT angiography and CT perfusion (RAPID-software). Among 50 aSAH patients requiring EVD, 21 (42%) developed DCI-related cerebral infarction, while 27 (54%) presented vasospasm. Mean ICP ranged between 2 and 19 mmHg. Remdesivir is one of the most promising drugs to treat COVID-19 based on the following facts remdesivir has a broad-spectrum antiviral mechanism of action; it demonstrated in vitro activity against SARS-CoV-2 and in vivo efficacy in animal models against the similar coronavirus MERS-CoV; its safety profile has been tested in Ebola patients and in compassionate use in COVID-19 patients. Currently, remdesivir is being investigated in ten randomized controlled trials against COVID-19. The dose regimen of remdesivir is an IV loading dose of 200 mg on day 1 followed by daily IV maintenance doses of 100 mg for 5-9 days. Based on our data analysis, however, remdesivir with IV administration alone is unlikely to achieve excellent clinical efficacy. This analysis is based on the following observations plasma exposures of remdesivir and its active metabolite are unlikely to be correlated with its clinical efficacy; remdesivir and its active metabolites are unlikely to be adequate in the lung to kill the SARS-CoV-2 virus. Even if remdesivir demonstrates benefits in the current randomized controlled trials, its efficacy may be limited. We suggest that a combination of an IV and pulmonary delivery dose regimen should be studied immediately to realize a potentially more effective antiviral therapy against COVID-19. The data set contains information on the seed dormancy classification (sensu Nikolaeva et al., 1985; Baskin and Baskin, 2004), dormancy-breaking conditions/germination, and for some of the included species storage behaviour, of seeds of nearly 3000 gymnosperm and angiosperm species from 843 genera and 164 families occurring worldwide. Additionally, the data set contains about ´550 references, many of them unknown to seed scientists, to seed ecology research conducted between 1926 and 1985. The data set should be of value to applied, basic and theoretical plant biologists, ecologists and evolutionary biologists interested in the various aspects of regeneration of plants from seeds. METHODS We studied the pattern of NSCs (i.e., glucose, fructose, sucrose, and starch) in alpine stands of Vaccinium myrtillus (above treeline) across an elevational gradient. In the same plots, we measured key growth traits (i.e., anatomical stem features) and shrub cover, evaluating putative relationships with NSCs. RESULTS Glucose content was positively related with altitude, but negatively related with shrub cover. Sucrose decreased at high altitude and in older populations and increased with higher percentage of vascular tissue. Starch content increased at middle and high elevations and in stands with high shrub cover. The result was a decrease in KM and increase in the rate at low substrate concentrations. An optimized system for paraoxon hydrolysis decreased KM by 11-fold, with a corresponding increase in second-order rate constant. The initial rates of VX and methyl-parathion hydrolysis were also increased by 3.1- and 6.7-fold, respectively. The designed scaffolds not only increased the local substrate concentration, but they also resulted in increased stability and PTE-DNA particle size tuning between 25 and ~150 nm. The aim of this study was to investigate the relationship between the distribution of ABO or Rh blood group antigens and the incidence of myelomeningocele. MATERIAL AND METHODS A retrospective data was reviewed of all myelomeningocele patients operated at our hospital between years 2014 and 2019. Age, sex, delivery method, physical and neurological examination findings, and radiological findings alongside with blood type of each patient were recorded. The data of blood group distribution among the study patients was compared to the data of healthy individuals in the same region. RESULTS Patients with group B and AB showed a higher chance of developing myelomeningocele. Rh-positive blood group was associated with high incidence of myelomeningocele (93.5%), whereas Rh-negative blood group showed least association (6.5%). Rh-positive blood group was also more frequent in patients with myelomeningocele with hydrocephalus and Chiari malformation. CONCLUSION ABO and Rh blood groups have an effect on the development of myelomeningocele under the influence of environmental or genetic factors.

Subsequent mechanistic experiments revealed that Rhein obviously downregulated the phosphorylation levels of PI3K, Akt, p38, and ERK1/2 and simultaneously upregulated the PTEN expression. In addition, Rhein antagonized the increase of TLR4, p-IκBα, and NF-κB. In summary, Rhein suppresses neuroinflammation via multiple signaling pathways (PI3K/Akt, p38, ERK1/2, and TLR4/NF-κB) in LPS-stimulated BV2 microglia cells. This study highlights a natural agent for prevention and treatment of neuroinflammation.
To explore whether Chuang **** Ye (CLY), a traditional Chinese herbal medicine compound, could improve the treatment of idiopathic granulomatous mastitis (IGM) via decreasing inflammatory response.
Herein, 40 patients with IGM who had wounds requiring dressing change were enrolled and randomly divided into two groups the CLY group and the control group. The size of the neoplasm and pain score of patients were followed-up for 4 weeks. Local tissues were taken during dressing change and examined by commercial enzyme-linked immunosorbent assay (ELISA) kits. The levels of inflammatory markers, including interleukin-1

Thai traditional medicine (TTM), using medicinal plants and ancient knowledge, has been used to treat wounds. Eight Thai medicinal plants, most commonly used to treat wounds, were evaluated for their in vitro biological activities such as antioxidation by NBT assay, anti-inflammation by production inhibition of NO, promoting fibroblast cell proliferation, and wound closure activities. Plant materials were extracted with 95% ethanol or distilled water and then concentrated and dried. Statistical analysis of data was done using one-way ANOVA at p value of 0.05. The ethanolic extracts of Garcinia mangostana L., Glycyrrhiza glabra L., and Nigella sativa L. could inhibit the production of superoxide anion with the IC50 values of 13.97 ± 0.38, 28.62 ± 1.91, and 71.54 ± 3.22 μg/ml and nitric oxide with the IC50 values of 23.97 ± 0.91, 46.35 ± 0.43, and 78.48 ± 4.46 μg/ml, respectively. These extracts could promote cell proliferation and accelerate wound recovery at the rate of 2.02 ± 0.03, 2.12 ± 0.03, and 2.65 ± 0.05% per hour, respectively. Three established markers from these three plants were selected according to the selection criteria. Alpha-mangostin, glycyrrhizin, and thymoquinone were found to be the active markers for wound closure activities. The ethanolic extracts of G. mangostana, G. glabra, and N. sativa could scavenge superoxide anion and inhibit the production of nitric oxide; therefore these extracts could assist in surpassing the inflammatory phase and protected the cells surrounding the wound area. Most importantly, these extracts also increased the proliferation and accelerated wound closure, indicating that these plant extracts could be promoting wound healing processes and support the use of TTM.

Studies investigating the effect of acupuncture on DOMS in humans that were published before March 2020 were obtained from eight electronic databases. The affected muscles, groups, acupuncture points, treatment sessions, assessments, assessment times, and outcomes of the included articles were reviewed. The data were extracted and analyzed via a meta-analysis.
A total of 15 articles were included, and relief of DOMS-related pain was the primary outcome. The statistical meta-analysis showed that there were no significant differences between acupuncture and sham/control groups, except for acupuncture for DOMS on day 1 (total SMD = -0.62; 95% CI = -1.12∼0.11,
< 0.05) by comparing with control groups.
Acupuncture for DOMS exhibited very-small-to-small and small-to-moderate effects on pain relief for the sham and no acupuncture conditions, respectively.
Subsequent mechanistic experiments revealed that Rhein obviously downregulated the phosphorylation levels of PI3K, Akt, p38, and ERK1/2 and simultaneously upregulated the PTEN expression. Herein, 40 patients with IGM who had wounds requiring dressing change were enrolled and randomly divided into two groups the CLY group and the control group. The size of the neoplasm and pain score of patients were followed-up for 4 weeks. Local tissues were taken during dressing change and examined by commercial enzyme-linked immunosorbent assay (ELISA) kits. Thai traditional medicine (TTM), using medicinal plants and ancient knowledge, has been used to treat wounds. Eight Thai medicinal plants, most commonly used to treat wounds, were evaluated for their in vitro biological activities such as antioxidation by NBT assay, anti-inflammation by production inhibition of NO, promoting fibroblast cell proliferation, and wound closure activities. Plant materials were extracted with 95% ethanol or distilled water and then concentrated and dried. Statistical analysis of data was done using one-way ANOVA at p value of 0.05. The ethanolic extracts of Garcinia mangostana L., Glycyrrhiza glabra L., and Nigella sativa L. could inhibit the production of superoxide anion with the IC50 values of 13.97 ± 0.38, 28.62 ± 1.91, and 71.54 ± 3.22 μg/ml and nitric oxide with the IC50 values of 23.97 ± 0.91, 46.35 ± 0.43, and 78.48 ± 4.46 μg/ml, respectively. These extracts could promote cell proliferation and accelerate wound recovery at the rate of 2.02 ± 0.03, 2.12 ± 0.03, and 2.65 ± 0.05% per hour, respectively. Three established markers from these three plants were selected according to the selection criteria. Alpha-mangostin, glycyrrhizin, and thymoquinone were found to be the active markers for wound closure activities. The ethanolic extracts of G. mangostana, G. glabra, and N. sativa could scavenge superoxide anion and inhibit the production of nitric oxide; therefore these extracts could assist in surpassing the inflammatory phase and protected the cells surrounding the wound area. Most importantly, these extracts also increased the proliferation and accelerated wound closure, indicating that these plant extracts could be promoting wound healing processes and support the use of TTM. INTRODUCTION/OBJECTIVES Magnetic resonance imaging (MRI) is recommended for evaluation of changes in juvenile spondyloarthropathies (JSpA). To our knowledge, there is no previous prospective study analysing early changes on axial MRI. The objective is to investigate incidence of reparable changes on axial MRI in patients with established JSpA, lasting for less than 6 months. MATERIALS AND METHODS The pilot study included 27 patients with confirmed diagnosis of JSpA examined within 2 years. Prior to imaging, basic demographic and laboratory data and HLA-B27 were collected. Patients filled out a visual analogue scale for pain and a childhood health assessment questionnaire. A paediatric rheumatologist and a paediatric physiatrist examined patients and measured indices of flexion, extension and sagittal flexibility. We found that the number of passages increased across the age. The capacity to discriminate the larger hole decreased as the ratio between the areas increased. No difference in accuracy was found as a function of age. The accuracy of larval zebrafish almost overlaps that found in adults in a previous study, suggesting a limited role of maturation and experience on the ability to estimate areas in this species.We theoretically study the thermoelectric effect in a hybrid device composed by a topological semiconducting nanowire hosting Majorana bound states (MBSs) and a quantum dot (QD) connected to the left and right non-magnetic electrodes held at different temperatures. The electron-electron Coulomb interactions in the QD are taken into account by the non-equilibrium Green's function technique. We find that the sign change of the thermopower, which is useful for detecting the MBSs, will occur by changing the QD-MBS hybridization strength, the direct overlap between the MBSs at the opposite ends of the nanowire, and the system temperature. Large value of 100% spin-polarized or pure spin thermopower emerges even in the absence of Zeeman splitting in the QD or magnetic electrodes because the MBSs are coupled to electrons of only one certain spin direction in the QD due to the chiral nature of the Majorana fermions. In addition to metal chelation, bifunctional chelators have also been found to impact imaging outcomes due to their differences in stability, charge, hydrophilicity, etc. In the present work, a comparative pharmacokinetic evaluation and imaging characteristics were performed between 68Ga-labelled somatostatin analogues (TATE) using NOTA (1,4,7-triazacyclononane-1,4,7-triacetic acid) and DOTA (1,4,7,10-tetraazacyclododecane-1,4,7,10-tetraacetic acid) as bifunctional chelating agents (BFCAs). RESULTS Both 68Ga-NOTA-TATE and 68Ga-DOTA-TATE were obtained with high radiochemical purity. 68Ga-NOTA-TATE demonstrated higher in vitro stability (≥ 99%) than 68Ga-DOTA-TATE (≥ 95%) after 3 h of incubation. The water solubilities (partition coefficients, - 1.76 ± 0.06 vs. - 2.72 ± 0.16) and plasma protein binding rates (12.12% vs. 30.6%) were lower for 68Ga-NOTA-TATE than for 68Ga-DOTA-TATE. Differential pharmacokinetics and comparable tumour affinities (within 1 h) were observed in AR42J tumour-bearing ****. Healthy volunteer imaging studies showed comparable distribution patterns of these two imaging agents. However, the maximum standardized uptake values (SUVmax) of the two tracers varied in each organ. The two PET agents demonstrated almost identical SUVmax values in the kidneys. 68Ga-NOTA-TATE did have a lower SUVmax in most other organs compared with 68Ga-DOTA-TATE, including the liver (4.2 vs. 10.1), potentially due to the lower protein binding rate. CONCLUSION 68Ga-NOTA-TATE and 68Ga-DOTA-TATE demonstrated comparable tumour uptake in an AR42J mouse model. An initial clinical study revealed that 68Ga-NOTA-TATE may have reduced background uptake in the major organs such as the liver. Although the subject numbers were limited, further investigation of 68Ga-NOTA-TATE is warranted for detecting SSTR2-positive neuroendocrine tumours.

Overall, 213 (35.7%) of all suicides occurred within 120 days of discharge. Upon adjustment and compared to the depressive phases, manic phases were significantly associated with a far lower hazard of dying by suicide (HR 0.34, 95% CI 0.21-0.56, p less then 0.001), though mixed phases were not (HR 0.92, 95% CI 0.48-1.73, p = 0.957). In this study, we performed a meta-analysis of the available toxicity data of heavy metals and ciliates to assess (1) the sensitivity of freshwater ciliates to different heavy metals, (2) the relative sensitivity of ciliates in comparison to the standard test species used in ecotoxicological risk assessment, and (3) the difference in sensitivity across ciliate taxa. Our study shows that the tolerance of ciliates to heavy metals varies notably, which is partly influenced by differences in methodological conditions across studies. Ciliates are, in general, sensitive to Mercury > Cadmium > Copper > Zinc > Lead > Chromium. Also, this study shows that most ciliates are more tolerant to heavy metal pollution than the standard test species used in ecotoxicological risk assessments, i.e., Raphidocelis subcapitata, Daphnia magna, and Onchornyncus mykiss. Threshold concentrations derived from toxicity data for these species is expected to confer sufficient protection for the vast majority of ciliate species. After pyrolysis at 450 °C, the bone char was treated by H3PO4 for surface modification and activated by K2CO3 respectively. The prepared materials were characterized by N2 adsorption isotherms, SEM, FT-IR, and XPS. Adsorption/desorption and regeneration behavior of VOCs were also studied. Results showed that H3PO4 modification can effectively accelerate the adsorption process and after K2CO3 activation, a new hierarchical pore structure was found with an ultrahigh total pore volume of 2.807 cm3/g. The specific adsorption capacity for typical VOC reached ∼13.03 mmol/g which is **** higher than literature data under the same condition. Static toluene adsorption test on the prepared activated bone-char revealed that the hierarchical structure has provided abundant adsorption sites and the adsorption behavior can be well described by the pseudo-second-order model. The objective of this study is to present an in-depth investigation of the structure and dynamics of sulfur bacteria (including SRB and SOB) in the water column of shallow freshwater lakes. A cyanobacterial bloom biomass (CBB)-amended mesocosm experiment was conducted in this study, in which water was taken from a shallow eutrophic lake with sulfate levels near 40 mg L-1. Illumina sequencing was used to investigate SRB and SOB species involved in CBB decomposition and the effects of the increases in sulfate input on the water column microbial community structure. The accumulation of dissolved sulfide (∑H2S) produced by SRB during CBB decomposition stimulated the growth of SOB, and ∑H2S was then oxidized **** to sulfate by SOB in the water column. Chlorobaculum sequences (the main SOB species in the study) were significantly influenced by increases in sulfate input, with relative abundance increasing approximately four-fold in treatments amended with 40 mg L-1 sulfate (referred to as 40S) when compared to the treatment without additional sulfate addition (referred to as CU). Additionally, an increase in SOB number was observed from day 26-37, concurrent with the decrease in SRB number, indicating the succession of sulfur bacteria. These findings suggest that biological sulfur oxidation and succession of sulfur bacteria occur in the water column during CBB decomposition in shallow freshwater ecosystems, and the increases in sulfate input stimulate microbial sulfur oxidation.

After pyrolysis at 450 °C, the bone char was treated by H3PO4 for surface modification and activated by K2CO3 respectively. The prepared materials were characterized by N2 adsorption isotherms, SEM, FT-IR, and XPS. Adsorption/desorption and regeneration behavior of VOCs were also studied. Results showed that H3PO4 modification can effectively accelerate the adsorption process and after K2CO3 activation, a new hierarchical pore structure was found with an ultrahigh total pore volume of 2.807 cm3/g. The specific adsorption capacity for typical VOC reached ∼13.03 mmol/g which is much higher than literature data under the same condition. Static toluene adsorption test on the prepared activated bone-char revealed that the hierarchical structure has provided abundant adsorption sites and the adsorption behavior can be well described by the pseudo-second-order model. A cyanobacterial bloom biomass (CBB)-amended mesocosm experiment was conducted in this study, in which water was taken from a shallow eutrophic lake with sulfate levels near 40 mg L-1. Illumina sequencing was used to investigate SRB and SOB species involved in CBB decomposition and the effects of the increases in sulfate input on the water column microbial community structure. The accumulation of dissolved sulfide (∑H2S) produced by SRB during CBB decomposition stimulated the growth of SOB, and ∑H2S was then oxidized back to sulfate by SOB in the water column. Chlorobaculum sequences (the main SOB species in the study) were significantly influenced by increases in sulfate input, with relative abundance increasing approximately four-fold in treatments amended with 40 mg L-1 sulfate (referred to as 40S) when compared to the treatment without additional sulfate addition (referred to as CU). Additionally, an increase in SOB number was observed from day 26-37, concurrent with the decrease in SRB number, indicating the succession of sulfur bacteria. GABA interneurons are an emerging therapeutic target in AD, with further clinical trials urgently warranted.Ischemic stroke is a severe neurological disorder with a high prevalence rate in developed countries. It is characterized by permanent or transient cerebral ischemia and it activates syndrome of pathological events such as membrane depolarization, glutamate excitotoxicity, and intracellular calcium buildup. Carveol is widely employed as anti-inflammatory and antioxidant in traditional Chinese medicine. In the present study, the neuroprotective effects of post-treated carveol were demonstrated against transient middle cerebral artery occlusion (MCAO) induced focal ischemic cerebral injury. Male Sprague Dawley (SD) rats were subjected to two different experimental protocols to determine the dose and effects of carveol, and to demonstrate the underlying role of the nuclear factor E2-related factor (Nrf2) pathway. Our results showed that MCAO induced marked neuronal injury in the ipsilateral cortex and striatum associated with higher inflammatory cytokines expression, along with apoptotic markers such as caspase- antioxidants, eventually ameliorating MCAO-induced neuroinflammation and neurodegeneration.

To explore whether rhythmic auditory stimulation (RAS) could improve motor functions of post-stroke hemiparetic upper extremity.
A prospective, randomized controlled, assessor-blinded pilot study.
Thirty stroke patients were randomly distributed into the RAS group (

= 15). Both groups received regular therapies. The RAS group received additional 30 min of RAS training, while the control group received additional 30 min of regular therapies for 5 days per week for 4 weeks. The Fugl-Meyer Assessment-Upper Extremity (FMA-UE), Wolf Motor Function Test (WMFT), and Barthel Index (BI) were used. The co-activation interval and co-contraction index were calculated from surface electromyography (sEMG) recordings on the affected biceps and triceps during elbow flexion and extension. Assessments were performed before and after the treatments.

Using this information, the brain exerts its influence on both itself and the body to facilitate an appropriate response. The dynamic interplay between the brain and the heart and how external conditions modulate this relationship deserves attention. In high-stress situations, synchrony between various brain regions such as the prefrontal cortex and the heart may alter. This flexibility is believed to facilitate transitions between functional states related to cognitive, emotional, and especially autonomic activity. This study examined the dynamic temporal functional association of heart rate variability (HRV) with the interaction between three main canonical brain networks in 38 healthy male subjects at rest and directly after a psychosocial stress task. A sliding window approach was used to estimate the functional connectivity (FC) among the salience network (SN), central executive network (CEN), and default mode network (DMN) in 60-s windows on time series of blood-oxygen-level dependent (BOLD) signal. FC between brain networks was calculated by Pearson correlation. A multilevel linear mixed model was conducted to examine the window-by-window association between the root mean square of successive differences between normal heartbeats (RMSSD) and FC of network-pairs across sessions. Our findings showed that the minute-by-minute correlation between the FC and RMSSD was significantly stronger between DMN and CEN than for SN and CEN in the baseline session [b = 4.36, t(5025) = 3.20, p = 0.006]. Additionally, this differential relationship between network pairs and RMSSD disappeared after the stress task; FC between DMN and CEN showed a weaker correlation with RMSSD in comparison to baseline [b = -3.35, t(5025) = -3.47, p = 0.006]. These results suggest a dynamic functional interplay between HRV and the functional association between brain networks that varies depending on the needs created by changing conditions.Guillain-Barré syndrome (GBS) is an acute, immune-mediated inflammatory peripheral polyneuropathy that is characterized by flaccid paralysis. A few cases have reported that GBS can be caused by head trauma or neurosurgery, but it has never been associated with intraventricular hemorrhage. Here, we report an uncommon case of fulminant GBS that occurred after spontaneous intraventricular hemorrhage. A 73-year-old woman was admitted to the hospital after sudden unconsciousness and vomiting. A head computed tomography (CT) scan following the incident showed a newly developed intraventricular hemorrhage, which led to an immediate ventriculostomy. After 5 days, the endotracheal tube was removed. Two days later, the external ventricular drainage tube was also removed. At this time, the patient was alert and the neurological examination was normal. However, the patient suddenly presented with acute respiratory failure and bilateral limb weakness 3 days later. An analysis of the patient's cerebrospinal fluid (CSF) revealed that albuminocytologic dissociation was present.

Methods 27 female students (mean age 20.5 ± 1 year) completed 8 weeks of aerobic training that included two exercise sessions of continuous aerobic activity of moderate intensity (running 35-60 min) and one exercise session with interval training of vigorous intensity. Intervention aerobic capacity and lipid profile were examined before and after the exercise. Results Exercise intervention has caused a decrease of low density lipoprotein levels per 9.8% (from 2.52 ± 0.47 to 2.27 ± 0.53 mmol/L; p less then 0.001) and significant improvement of high density lipoprotein (HDL) levels per 22.7% (from 1.29 ± 0.24 to 1.59 ± 0.24 mmol/L; p less then 0.001), total cholesterol/HDL ratio per 17.2 % and aerobic capacity (VO2max) per 3.8 % (43.9 ± 3.7 to 45.56 ± 3.63 mLO2/kg/min). Basal capillary glucose was determined using Abbott FreeStyle Precision Neo in Brescia (n=63), while clinical laboratory reference P-glucose was determined using tubes containing NaF/K3EDTA and liquid NaF/Na2EDTA/citrate. Basal capillary glucose was determined using a Roche Cobas Accu-Chek Inform II in Vicenza (n=76), while P-glucose was determined using tubes containing NaF/K2Ox and NaF/Na2EDTA/citrate in granulated form. Reference P-glucose was determined with a hexokinase method on Dimension Vista systems. Differences between capillary and reference P-glucose were evaluated according to ADA/ISO 151972013 specifications. Results 96.82% and 97.37% of capillary determinations were within specifications when liquid and granular citrate mixture tubes were used, respectively. Conversely, only 73.02% and 80.26% of determinations were within criteria using NaF. Conclusions It's important to know what is the laboratory reference glucose in evaluating glucose meters' accuracy. Methods This study was carried out on a sample of 150 premenopausal women who were referred to the Outpatient Clinic of Family Medicine between 2014-2015. A survey about their socio-demographic characteristics was carried out, and anthropometric parameters were measured. LDL-C, HDL-C, total cholesterol, triglyceride, glucose, insulin, sex hormone binding globulin (SHBG), estradiol, dehydroepiandrosterone sulfate (DHEA-S), total/free testosterone levels were measured in the blood. Body compositions were assessed with a bioelectrical impedance device. For insulin resistance, Homeostasis Model Assessment (HOMA-IR) was calculated. Results In our study, a significant association was found between high glucose, total cholesterol, LDL-C, TG, insulin, insulin resistance and low HDL-C, SHBG, DHEA-S levels with obesity (p0.05). Conclusions In our study, high glucose, total cholesterol, LDL-c, TG, insulin, insulin resistance and low HDL-C, SHBG, DHEA-S levels were associated with obesity. This relationship leads to many diseases, especially diabetes mellitus and cardiovascular disease. Therefore, obesity is a disease that needs to be monitored closely, frequently and treated properly.Background This study aimed to verify whether blood drawn into six different commercial coagulation tubes generated comparable results of thrombin generation. Methods Blood was sequentially collected from 20 healthy subjects into different brand and draw volume 3.2% sodium citrate tubes (4.3 mL Sarstedt, 3.0 mL Greiner, 2.7 mL Becton Dickinson, 2.0 mL Kima, 1.8 mL Sarstedt and 1.0 mL Greiner). Thrombin generation was measured in plasma with the fully-automated ST Genesia analyzer using the weakest trigger (STG-BleedScreen). Results Different values of lag time (LT), time to reach thrombin peak (TP), thrombin peak height (PH) and endogenous thrombin potential (ETP) were commonly found in different tubes. LDL-C, HDL-C, total cholesterol, triglyceride, glucose, insulin, sex hormone binding globulin (SHBG), estradiol, dehydroepiandrosterone sulfate (DHEA-S), total/free testosterone levels were measured in the blood. Body compositions were assessed with a bioelectrical impedance device. For insulin resistance, Homeostasis Model Assessment (HOMA-IR) was calculated. Results In our study, a significant association was found between high glucose, total cholesterol, LDL-C, TG, insulin, insulin resistance and low HDL-C, SHBG, DHEA-S levels with obesity (p0.05). Conclusions In our study, high glucose, total cholesterol, LDL-c, TG, insulin, insulin resistance and low HDL-C, SHBG, DHEA-S levels were associated with obesity. This relationship leads to many diseases, especially diabetes mellitus and cardiovascular disease. Therefore, obesity is a disease that needs to be monitored closely, frequently and treated properly.Background This study aimed to verify whether blood drawn into six different commercial coagulation tubes generated comparable results of thrombin generation. Methods Blood was sequentially collected from 20 healthy subjects into different brand and draw volume 3.2% sodium citrate tubes (4.3 mL Sarstedt, 3.0 mL Greiner, 2.7 mL Becton Dickinson, 2.0 mL Kima, 1.8 mL Sarstedt and 1.0 mL Greiner). Thrombin generation was measured in plasma with the fully-automated ST Genesia analyzer using the weakest trigger (STG-BleedScreen). Results Different values of lag time (LT), time to reach thrombin peak (TP), thrombin peak height (PH) and endogenous thrombin potential (ETP) were commonly found in different tubes.
