Few studies have measured the effect of genetic factors on dementia and cognitive decline in healthy older individuals followed prospectively. We studied cumulative incidence of dementia and cognitive decline, stratified by APOE genotypes and polygenic risk score (PRS) tertiles, in 12,978 participants of the ASPirin in Reducing Events in the Elderly (ASPREE) trial. At enrolment, participants had no history of diagnosed dementia, cardiovascular disease, physical disability or cognitive impairment. Dementia (adjudicated trial endpoint) and cognitive decline, defined as a >1.5 standard deviation decline in test score for either global cognition, episodic memory, language/executive function or psychomotor speed, versus baseline scores. We review the current knowledge of don't-eat-me signaling in normal physiology and disease contexts where aberrant don't-eat-me signaling contributes to pathology.
Glycated albumin (GA) is a biomarker for short-term (2-3 weeks) glycaemic control. However, the predictive utility of GA for diabetes and prediabetes is largely uncharacterised. We aimed to investigate the relationships of baseline serum GA levels with incident diabetes and prediabetes.
This was a longitudinal cohort study involving 516 subjects without diabetes or prediabetes at baseline. Blood glucose levels were observed during follow-up. Hazard ratios (HRs) with 95% confidence intervals (CIs) were calculated using COX proportional hazard models. Receiver operating characteristic curves and areas under the curves (AUCs) were used to evaluate the discriminating abilities of glycaemic biomarkers and prediction models.
During a 9-year follow-up, 51 individuals (9.88%) developed diabetes and 92 (17.83%) prediabetes. Unadjusted HRs (95% CI) for both diabetes and prediabetes increased proportionally with increasing GA levels in a dose-response manner. Multivariable-adjusted HRs (95% CI) for diabetes were significantly elevated from 1.0 (reference) to 5.58 (1.86-16.74). However, the trend was no longer significant for prediabetes after multivariable adjustment. AUCs for GA, fasting blood glucose (FBG) and 2-h postprandial blood glucose (2h-PBG) for predicting diabetes were 0.698, 0.655 and 0.725, respectively. The AUCs for GA had no significant differences compared with those for FBG (p=0.376) and 2h-PBG (p=0.552). Replacing FBG or 2h-PBG or both with GA in diabetes prediction models made no significant changes to the AUCs of the models.

Although parent interventions are recommended as a frontline treatment approach for children with attention deficit hyperactivity disorder (ADHD), a number of practical and situational barriers can impact accessibility and availability. Self-help parent interventions offer a potential alternative to therapist-led interventions when barriers prevent access to face-to-face treatment. This qualitative study aims to explore participant views and acceptability of self-help parent interventions.
Semi-structured interviews were conducted with parents/carers of 12 children (age 6-10years) with ADHD who received the self-help version of the New Forest Parenting Programme (NFPP-SH) as part of a randomised controlled trial. Thematic analysis (as proposed by Braun and Clarke) was used to analyse the data.
Overall, participants had favourable views of the self-help intervention but also experienced some barriers to treatment adherence. Six key themes were identified in parent interviews related to parental desire to learn more; acquisition of new skills; the flexibility of the intervention; self-help intervention vs.

Histological evaluation of human cortical tissues (5x5x1 mm; n = 30; 3 pieces per patient) xenografted for 6 weeks and juvenile murine ovaries cultured for 4 days (n = 9) or allografted for 2 weeks (n = 48) showed no differences in the distribution of resting or growing follicles in S1P-treated ovarian tissues compared to controls. Collectively, S1P increased Ccn2/CCN2 gene expression in isolated preantral follicles and ovarian tissue from **** and human, but it did not promote follicle activation or growth in vivo. Our earlier studies on Plasmodium falciparum metacaspase-2 (PfMCA-2) revealed that the caspase inhibitor, Z-FA-FMK efficiently inhibited PfMCA-2 activity and, expression, and significantly blocked in vitro progression of the parasite developmental cycle via apoptosis-like parasite death. Building on these findings, we synthesized a set of novel inhibitors based on structural modification of Z-FA-FMK with the amides of piperic acid and investigated their effect on PfMCA-2. One of these analogues, SS-5, specifically inhibited the activity and expression of PfMCA-2. The activities of some other known malarial proteases (falcipains, plasmepsins, & vivapain), and human cathepsins-B, D and L, and caspase-3 and -7 were not inhibited by SS-5. SS-5 blocked the development of P. falciparumin vitro (IC50 1µM) and caused prominent morphological distortions. Incubation with SS-5 led to persistent parasite oxidative stress accompanied by depolarization of mitochondrial potential and accumulation of intracellular Ca2+. SS-5 also inhibited the development of P. berghei in a murine model. Our results suggest that the inhibition of PfMCA-2 results in oxidative stress, leading to apoptosis-like parasite death. Thus, SS-5 offers a starting point for optimization of new antimalarials, and PfMCA-2 could be a novel target for antimalarial drug discovery. Observations National Institute of Neurological Disorders and Stroke Network for Excellence in Neuroscience Clinical Trials (NeuroNEXT) was established in 2011 and renewed in 2018 with the goal of being an efficient network to test between 5 and 7 promising new agents in phase II clinical trials. A clinical coordinating center, data coordinating center, and 25 sites were competitively chosen. Common infrastructure was developed to accelerate timelines for clinical trials, including central institutional review board (a first for the National Institute of Neurological Disorders and Stroke), master clinical trial agreements, the use of common data elements, and experienced research sites and coordination centers. During the first 7 yearss were given at regional, national, and international meetings. Conclusions and Relevance NeuroNEXT initiated 8 studies, successfully enrolled participants at or ahead of schedule, collected high-quality data, published primary results in high-impact journals, and provided mentorship, expert statistical, and trial management support to several new investigators. Partnerships were successfully created between government, academia, industry, foundations, and patient advocacy groups. Clinical trial consortia can efficiently and successfully address a range of important neurologic research and therapeutic questions.Importance The rate of opioid-related emergency department (ED) visits and inpatient hospitalizations has increased rapidly in recent years. Medicaid expansions have the potential to reduce overall opioid-related hospital events by improving access to outpatient treatment for opioid use disorder. Objective To examine the association between Medicaid expansions and rates of opioid-related ED visits and inpatient hospitalizations. Design, Setting, and Participants A difference-in-differences observational design was used to compare changes in opioid-related hospital events in US nonfederal, nonrehabilitation hospitals in states that implemented Medicaid expansions between the first quarter of 2005 and the last quarter of 2017 with changes in nonexpansion states. All-payer ED and hospital discharges from 45 states in the Healthcare Cost and Utilization Project FastStats were included. Exposures State implementation of Medicaid expansions between 2005 and 2017. Participant response rates were 46 of 65 (71%) and 40 of 46 (87%) for rounds one and two, respectively, with participants representing, researchers, consumers, and industry partners from 15 countries over five continents. Seven measurement tools were identified for the core outcome set of patients on extracorporeal membrane oxygenation.
This study has identified appropriate measurement instruments and the timing of measurement to include in the core outcome set for research evaluating patients receiving extracorporeal membrane oxygenation. This was an important final step to standardize and synthesize research efforts internationally.

Hyperbaric oxygen therapy (HBO2) is recommended for symptomatic patients within 24-hour postcarbon monoxide poisoning. Introduction Although educational attainment and income are protective against tobacco use, Marginalization-related Diminished Returns (MDRs) theory posits that the protective effects of socioeconomic status (SES) indicators may be smaller for marginalized groups than mainstream social groups. We aimed to compare the effects of educational attainment and income on current smoking status of Chinese American and European American adults. Methods Data came from the National Health Interview Survey (NHIS - 2015). A total number of 28081 individuals entered our analysis. This included 420 Chinese Americans and 27661 European Americans. The independent variables were educational attainment (years of schooling) and annual income. The dependent variable was current established smoking status. Age, gender, region, and marital status were covariates. Ethnicity (Chinese American vs. European American) was the moderator. Results Overall, educational attainment and income were inversely associated with the odds of current established smoking. Ethnicity showed a significant interaction with income but not educational attainment. This finding suggested that the protective effect of income, but not educational attainment, on odds of current established smoking might be smaller for Chinese Americans than European Americans. Conclusions While educational attainment reduces the odds of current established smoking, high-income Chinese Americans remain at high risk of smoking due to a phenomenon called MDRs. In fact, high income is associated with greater smoking prevalence in Chinese Americans, rather than less. Methods In this retrospective, cross-sectional diagnostic study, 2 observers randomly assessed the conventional anteroposterior shoulder radiographs of 280 subjects with rotator cuff imaging for the (1) acromiohumeral (AH) interval, (2) upward migration index (UMI), (3) glenohumeral center-to-center measurement (GHCC), (4) glenohumeral arc measurement (GHa), and (5) scapular spine-humeral head center method (SHC). Reliability was assessed by means of relative consistency (intraclass correlation coefficient) and absolute consistency. Discriminative accuracy for detecting a rotator cuff tear was calculated. Results Relative consistency (intraclass correlation coefficient) for the AH interval, UMI, GHCC, GHa, and SHC was 0.961, 0.913, 0.806, 0.924, and 0.726, respectively. The AH interval had the highest absolute consistency with a random residual measurement error of 0.58 mm compared with 1.0-3.2 mm for the other measurements. The discriminative accuracy of the AH interval did not significantly differ from that of the UMI (-0.010; 95% confidence interval [CI], -0.042 to 0.022; P = .545) but was significantly better than that of the GHCC (0.112; 95% CI, 0.043-0.181; P = .001), GHa (0.074; 95% CI, 0.009-0.139; P = .027), and SHC (0.178; 95% CI, 0.100-0.256; P less then .001). In an old climatically buffered infertile landscape (OCBIL) in Brazil, we previously characterized different strategies for foliar water uptake (FWU). It is possible that photosystem II tolerance to heat and excessive light intensity varies among species with different FWU capacities.
The relationship between FWU, photoinhibition, and thermotolerance was investigated in seven species from this ecosystem.
The species with slow water absorption and high water absorption are those that presented less photoinhibition. Contrastingly, the species that have fast and low water absorption presented greater thermotolerance when their leaves are totally hydrated. However, when there is greater leaf dehydration, the most thermotolerant species were those with slow but high water absorption.

The virus was detected in pig's blood (3.4%; 95% CI 1.1-7.8) and liver (1.0%; 95% CI 0.0-5.0) with no significant differences observed in the frequency of its occurrence between the two by-products (t = 1.33; p = 0.182 > 0.05); however PAdV was detected more frequently in pig's blood than in liver (t = 4.65; p = 0.000 0.05 for pAdV). It is possible that photosystem II tolerance to heat and excessive light intensity varies among species with different FWU capacities. The relationship between FWU, photoinhibition, and thermotolerance was investigated in seven species from this ecosystem. The species with slow water absorption and high water absorption are those that presented less photoinhibition. Contrastingly, the species that have fast and low water absorption presented greater thermotolerance when their leaves are totally hydrated. However, when there is greater leaf dehydration, the most thermotolerant species were those with slow but high water absorption. Nephrotoxicity remains a major long-standing concern for colistin, and it is critical to find agents that can prevent it. The present study aims to investigate the effect of vitamin E on the prevention of colistin-induced nephrotoxicity based on its antioxidant and free radical scavenging properties.
A randomized clinical trial was designed for 52 patients taking colistin. These patients were categorized into two groups of equal size, receiving colistin or colistin plus vitamin E (α-Tocopherol). Vitamin E with doses of 400 units was administrated daily either orally or by a nasogastric tube if needed. The incidence of Acute Kidney Injury (AKI) and its duration was recorded based on RIFLE criteria.
The Incidence of AKI based on RIFLE criteria was 42.3% and 46.2% in intervention and control groups, respectively. The analysis showed no significant difference in the prevalence of AKI for the two groups (P=0.78). There was no significant difference in the duration of AKI neither (P=0.83).

The aim of this study was to monitor the development of drug-resistant bacteria isolated from acute uncomplicated cystitis (AUC) and to evaluate methodology of the survey conducted by collecting only clinical data.
We enrolled female patients at least 16 years of age diagnosed with AUC in 2018. Patient information including age, menopausal status, and results of bacteriological examination were collected and analyzed regardless of bacterial identification, antimicrobial susceptibility testing or extended-spectrum β-lactamase (ESBL) detection method.
A total of 847 eligible cases were collected. Escherichia coli (E.coli) was the most frequently isolated bacterial species at about 70%, with proportions of fluoroquinolone-resistant E.coli (QREC) and ESBL-producing E.coli isolates at 15.6% and 9.5% of all E.coli isolates, respectively. The proportion of Staphylococcus saprophyticus (S.saprophyticus) was significantly higher in premenopausal women. Regarding the drug susceptibility of E.coli, isolates from EIt is expected to be continuously performed as an alternative survey to conventional one collecting bacterial strains.
Our study aimed to evaluate the cytokine levels in pediatric chronic non-bacterial osteomyelitis (CNO) patients and compare these with other immune-mediated diseases and healthy controls.
In this prospective study, we included 42 children with CNO, 28 patients with non-systemic juvenile idiopathic arthritis (JIA), 17 children with insulin-dependent diabetes mellitus (IDDM), and 30 healthy age-matched controls. In each of the CNO patients and comparison groups, the levels of 14-3-3-η protein, S100A8/A9 protein, interleukin-4 (IL-4), interleukin-17 (IL-17), interleukin-18 (IL-18), interleukin-1β (IL-1β), tumor necrosis factor-α (TNF-α) were measured by ELISA assay.
All studied cytokines in the CNO patients were significantly higher than controls, and IDDM, 14-3-3-η protein, IL-18, IL-4, IL-17, IL-1β, and TNF-α were less than in JIA patients. In the discriminant analysis, ESR, 14-3-3 protein, S100A8/A9, IL-18, IL-4, and TNF-α can discriminate CNO from JIA, and 14-3-3 protein, S100A8/A9, IL-18, IL-17, IL-4, and TNF-α can distinguish CNO from other diseases and HC.

Roux-en-Y gastric bypass (RYGB) has been widely used for type 2 diabetes (T2D) patients with overweight or obesity. However, the long-term outcomes of RYGB versus medical therapy have not been well compared.
To evaluate the long-term outcomes of RYGB versus medical therapy for patients with T2D.

Four electronic databases-PubMed, EMBASE, the Cochrane Library, and ClinicalTrials.gov-were searched for articles published through February 2021. Eligible studies were randomized controlled trials.
Of 7 randomized controlled trials (15 articles), 477 patients were included 239 were randomly divided into RYGB groups and 238 to medical therapy groups. Statistically higher rates of T2D remission were observed in RYGB groups at 1 year (relative risk [RR], 18.01; 95% confidence interval [CI], 4.53- 71.70; P < .0001), 3 years (RR, 29.58; 95% CI, 5.92-147.82; P < .0001), and 5 years (RR, 16.92; 95% CI, 4.15-69.00; P < .0001). Meanwhile, statistically higher rates of achieving the American Diabetes Association's (ADA's) treatment goal were observed in RYGB groups at 1 year (RR, 3.99; 95% CI, 1.01-15.82; P = .05), 2 years (RR, 2.98; 95% CI, 1.62- 5.48; P = .0004), 3 years (RR, 3.16; 95% CI, 1.33-7.49; P = .009), and 5 years (RR, 6.18; 95% CI, 1.69-22.68; P = .006).

The Fujiwara-kyo Osteoporosis Risk in Men (FORMEN) study was launched to investigate risk factors for osteoporotic fractures, interactions of osteoporosis with other non-communicable chronic diseases, and effects of fracture on QOL and mortality.
FORMEN baseline study participants (in 2007 and 2008) included 2012 community-dwelling men (aged 65-93 years) in Nara prefecture, Japan. Clinical follow-up surveys were conducted 5 and 10 years after the baseline survey, and 1539 and 906 men completed them, respectively. Infants suffer from a severe epileptic encephalopathy known as West syndrome (WS). Treatment with adrenocorticotropic hormone (ACTH) indicates the involvement of the gut-brain axis in WS. Several pieces of evidence show the communication of the gut microbiota (GM) with the brain via the hypothalamic-pituitary-adrenal axis (HPA axis) and blood cytokines. This study aimed at (1) determining the GM diversity in infants having WS and (2) comparing the results of infants having WS with those of the healthy infants and also in the patients with WS before and after the ACTH therapy.
In this study, 29 infants with WS and 29 healthy infants aged 3-13 months were recruited. Fecal samples were collected, and DNA was extracted and sequenced on the Illumina MiSeq platform. Kruskal-Wallis rank-sum test was used to analyze the between-group differences in the Chao1 index, Shannon index, and the abundances of GM at different taxonomy levels. R software was used to plot the graphs. The top five dominant GM genera between patients with WS and healthy infants showed no significant differences. However, the relative abundance of genus Akkermansia was observed to be significantly (P = 0.011) higher in the BT group than in the HC group and AT group. After 2 weeks of ACTH therapy, the relative abundance of Akkermansia significantly (P = 0.003) decreased.

Tribenuron methyl (TBM) is an herbicide that inhibits sulfonylurea acetolactate synthase (ALS) and is one of the most widely used broad-leaved herbicides for crop production. However, soil residues or drifting of the herbicide spray might affect the germination and growth of rapeseed, Brassica napus, so it is imperative to understand the response mechanism of rape to TBM during germination. The aim of this study was to use transcriptome analysis to reveal the gene responses in herbicide-tolerant rapeseed to TBM stress during seed germination.
2414, 2286, and 1068 differentially expressed genes (DEGs) were identified in TBM-treated resistant vs sensitive lines, treated vs. control sensitive lines, treated vs. control resistant lines, respectively. GO analysis showed that most DEGs were annotated to the oxidation-reduction pathways and catalytic activity. KEGG enrichment was mainly involved in plant-pathogen interactions, α-linolenic acid metabolism, glucosinolate biosynthesis, and phenylpropanoid biosynthesis. Based on GO and KEGG enrichment, a total of 137 target genes were identified, including genes involved in biotransferase activity, response to antioxidant stress and lipid metabolism. Biotransferase genes, CYP450, ABC and GST, detoxify herbicide molecules through physical or biochemical processes. Infants suffer from a severe epileptic encephalopathy known as West syndrome (WS). Treatment with adrenocorticotropic hormone (ACTH) indicates the involvement of the gut-brain axis in WS. Several pieces of evidence show the communication of the gut microbiota (GM) with the brain via the hypothalamic-pituitary-adrenal axis (HPA axis) and blood cytokines. This study aimed at (1) determining the GM diversity in infants having WS and (2) comparing the results of infants having WS with those of the healthy infants and also in the patients with WS before and after the ACTH therapy. In this study, 29 infants with WS and 29 healthy infants aged 3-13 months were recruited. Fecal samples were collected, and DNA was extracted and sequenced on the Illumina MiSeq platform. Kruskal-Wallis rank-sum test was used to analyze the between-group differences in the Chao1 index, Shannon index, and the abundances of GM at different taxonomy levels. R software was used to plot the graphs. The top five dominant GM genera between patients with WS and healthy infants showed no significant differences. However, the relative abundance of genus Akkermansia was observed to be significantly (P = 0.011) higher in the BT group than in the HC group and AT group. Tribenuron methyl (TBM) is an herbicide that inhibits sulfonylurea acetolactate synthase (ALS) and is one of the most widely used broad-leaved herbicides for crop production. However, soil residues or drifting of the herbicide spray might affect the germination and growth of rapeseed, Brassica napus, so it is imperative to understand the response mechanism of rape to TBM during germination. The aim of this study was to use transcriptome analysis to reveal the gene responses in herbicide-tolerant rapeseed to TBM stress during seed germination. 2414, 2286, and 1068 differentially expressed genes (DEGs) were identified in TBM-treated resistant vs sensitive lines, treated vs. control sensitive lines, treated vs. control resistant lines, respectively. GO analysis showed that most DEGs were annotated to the oxidation-reduction pathways and catalytic activity. KEGG enrichment was mainly involved in plant-pathogen interactions, α-linolenic acid metabolism, glucosinolate biosynthesis, and phenylpropanoid biosynthesis. Based on GO and KEGG enrichment, a total of 137 target genes were identified, including genes involved in biotransferase activity, response to antioxidant stress and lipid metabolism. Biotransferase genes, CYP450, ABC and GST, detoxify herbicide molecules through physical or biochemical processes. Antioxidant genes, RBOH, WRKY, CDPK, MAPK, CAT, and POD regulate plant tolerance by transmitting ROS signals and triggering antioxidant enzyme expression. Here, we present data on the pH-dependent kinetics of azanone reactions with the low molecular thiols glutathione and N-acetylcysteine, as well as with important serum proteins bovine serum albumin and human serum albumin. The competition kinetics method used is based on two parallel HNO reactions with RSH/RS- or with O2. The results provide evidence that the reaction of azanone with the anionic form of thiols (RS-) is favored over reactions with the protonated form (RSH). The data are supported with quantum mechanical calculations. A comprehensive discussion of the HNO reaction with thiolates is provided.We analyzed the precipitation chemistry for a maritime region in northern Germany (Schleswig-Holstein) from 1997 to 2017 in order to reveal temporal and spatial patterns and to evaluate the role of meteorological factors relative to emission reductions in Germany and Europe. Therefore, we applied several statistical methods such as time series decomposition, principal component, and redundancy analysis. We extracted two main groups (i) a marine group (Cl, Na, Mg) that was related to natural processes like sea spray input and (ii) an anthropogenic group (Pb, Cd, As, Zn, and nitrogen species) with a terrestrial subgroup (Fe, Al, Mn), which were both related to emissions. These groups were valid for the spatial, seasonal, and annual trend data. Other elements, like Ca, K, total P, and sulfate, were influenced by natural and anthropogenic processes. The seasonal variation of ammonium deposition was caused primarily by ammonia emissions and ancillary by precipitation. Most heavy metals as well as sulfate, nitrate, and ammonium showed decreasing trends in concentrations and deposition fluxes. Only Hg did not show any trend. The decreasing depositions of sulfate and total nitrogen were correlated to emission reductions in Germany. The deposition of most heavy metals was influenced by emission reductions on European scale and meteorological factors such as wind speed and humidity. Hg did not show any correlation with the emission time series in Europe. Instead, it was correlated to the NAO index and wind, implying that global emissions and transport pathways determine the temporal development of Hg depositions. The results are shown that rutin has the best inhibition potentials among the studied molecules with high binding energy - 8.04 kcal/mol, and it is followed by myricetin, quercetin, caffeic acid phenethyl ester and hesperetin. However, the reference molecule has binding energy of - 7.24 kcal/mol. In conclusion, the high potential of flavonoids in ethanolic propolis extracts to bind to ACE-II receptors indicates that this natural bee product has high potential for COVID-19 treatment, but this needs to be supported by experimental studies.
The aim of this study was to evaluate the impact of the COVID-19 lockdown occurred in Italy from March 9th to May 18th, 2020 on anthropometric parameters and glycemic control in patients with type 2 diabetes mellitus (T2DM).
One hundred twenty-eight consecutive patients with T2DM (median age 70 years, 74 males) were retrospectively evaluated at the end of the lockdown period. Data on metabolic control were collected at different time within three months before the lockdown (visit 0) and within the first six weeks after it (visit 1).
During the lockdown, a significant increase in body weight (from 79.7 ± 18.7 kg to 81.4 ± 19.4 kg, p < 0.001), body mass index (BMI, from 29.5 ± 6 kg/m
to 30.1 ± 6.3 kg/m
, p < 0.001), waist circumference (from 103.8 ± 13 cm to 105 ± 13.6 cm, p < 0.001), fasting plasma glucose (FPG; from 138.1 ± 29.4 mg/dL to 146.6 ± 36.4 mg/dL) and glycated hemoglobin (HbA1c; from 7 ± 0.8 to 7.3 ± 0.9%, p < 0.001) was observed. With the regulation of perfluorooctanesulfonate (PFOS), 62 chlorinated polyfluoroalkyl ether sulfonate (62 Cl-PFESA) has been used as a potential PFOS alternative in electroplating. In this study, the uptake, translocation and phytotoxicity of PFOS and 62 Cl-PFESA in mung bean (Vigna radiata (Linn.) Wilczek.) were investigated. The uptake kinetics of PFOS and 62 Cl-PFESA fit the Michaelis-Menten equation well, suggesting that the uptake is a carrier-mediated process. The root concentration factor (RCF) of 62 Cl-PFESA (34.55 mL g-1 dw) was 1.27 times that of PFOS (27.11 mL g-1 dw), and the translocation factor (TF) of 62 Cl-PFESA (0.177) was 1.07 times that of PFOS (0.165). Exposure to 62 Cl-PFESA and PFOS both resulted in the inhibition of mung bean seedling development. Treatment with 62 Cl-PFESA and PFOS led to the concentration-dependent elevation of malondialdehyde (MDA), carbonyl groups, and phosphorylated histone H2AX (γ-H2AX) levels in mung bean roots. The MDA and carbonyl group contents induced by 62 Cl-PFESA were 1.10-1.35 and 1.03-1.14 times, respectively, those of PFOS. The hydroxyl free radical (·OH) levels in mung bean roots after exposure to PFOS and 62 Cl-PFESA were elevated significantly, and the ·OH levels induced by 62 Cl-PFESA were higher than those induced by PFOS. Hydroxyl free radical levels were positively correlated with the MDA and carbonyl group contents in mung bean roots (p less then 0.05). The dynamic changes in some antioxidative enzyme activities in mung bean seedlings were determined, including peroxidase (POD), superoxide dismutase (***), and catalase (CAT). The results demonstrated the phytotoxicities of 62 Cl-PFESA and PFOS to mung bean in the early developmental stage. 62 Cl-PFESA is more harmful to mung beans than PFOS. The production of hydroxyl radical is the mechanism that causes the toxicity of PFOS and 62 Cl-PFESA toward plants.Medical image segmentation is a typical task in medical image processing and critical foundation in medical image analysis. U-Net is well-liked in medical image segmentation, but it doesn't fully explore useful features of the channel and capitalize on the contextual information. Therefore, we present an improved U-Net with residual connections, adding a plug-and-play, very portable channel attention (CA) block and a hybrid dilated attention convolutional (HDAC) layer to perform medical image segmentation for different tasks accurately and effectively, and call it HDA-ResUNet, in which we fully utilize advantages of U-Net, attention mechanism and dilated convolution. In contrast to the simple copy splicing of U-Net in the skip connection, the channel attention block is inserted into the extracted feature map of the encoding path before decoding operation. Since this block is lightweight, we can apply it to multiple layers in the backbone network to optimize the channel effect of this layer's coding operation. In addition, the convolutional layer at the bottom of the "U"-shaped network is replaced by a hybrid dilated attention convolutional (HDAC) layer to fuse information from different sizes of receptive fields. The proposed HDA-ResUNet is evaluated on four datasets liver and tumor segmentation (LiTS 2017), lung segmentation (Lung dataset), nuclear segmentation in microscope images (DSB 2018) and neuron structure segmentation (ISBI 2012). The dice global scores of liver and tumor segmentation (LiTS 2017) reach 0.949 and 0.799. The dice coefficients of lung segmentation and nuclear segmentation are 0.9797 and 0.9081 respectively, and the information theoretic score for the last one is 0.9703. The segmentation results are all more accurate than U-Net with fewer parameters, and the problem of slow convergence speed of U-Net on DBS 2018 is solved.Acute respiratory distress syndrome (ARDS) is a life-threatening lung injury with global prevalence and high mortality. Chest x-rays (CXR) are critical in the early diagnosis and treatment of ARDS. However, imaging findings may not result in proper identification of ARDS due to a number of reasons, including nonspecific appearance of radiological features, ambiguity in a patient's case due to the pathological stage of the disease, and poor inter-rater reliability from interpretations of CXRs by multiple clinical experts. This study demonstrates the potential capability of methodologies in artificial intelligence, machine learning, and image processing to overcome these challenges and quantitatively assess CXRs for presence of ARDS. We propose and describe Directionality Measure, a novel feature engineering technique used to capture the "cloud-like" appearance of diffuse alveolar damage as a mathematical concept. This study also examines the effectiveness of using an off-the-shelf, pretrained deep learning modg the proposed methodologies to complement current clinical analysis for detection of ARDS from CXRs.The p38α MAP Kinase has been an important target of drug design for treatment of inflammatory diseases and cancers. This work applies multiple replica Gaussian accelerated molecular dynamics (MR-GaMD) simulations and the molecular mechanics generalized Born surface area (MM-GBSA) method to probe the binding mechanism of inhibitors L51, R24 and 1AU to p38α. Dynamics analyses show that inhibitor bindings exert significant effect on conformational changes of the active helix α2 and the conserved DFG loop. The rank of binding free energies calculated with MM-GBSA not only agrees well with that determined by the experimental IC50 values but also suggests that mutual compensation between the enthalpy and entropy changes can improve binding of inhibitors to p38α. The analyses of free energy landscapes indicate that the L51, R24 and 1AU bound p38α display a DFG-out conformation. With the regulation of perfluorooctanesulfonate (PFOS), 62 chlorinated polyfluoroalkyl ether sulfonate (62 Cl-PFESA) has been used as a potential PFOS alternative in electroplating. In this study, the uptake, translocation and phytotoxicity of PFOS and 62 Cl-PFESA in mung bean (Vigna radiata (Linn.) Wilczek.) were investigated. The uptake kinetics of PFOS and 62 Cl-PFESA fit the Michaelis-Menten equation well, suggesting that the uptake is a carrier-mediated process. The root concentration factor (RCF) of 62 Cl-PFESA (34.55 mL g-1 dw) was 1.27 times that of PFOS (27.11 mL g-1 dw), and the translocation factor (TF) of 62 Cl-PFESA (0.177) was 1.07 times that of PFOS (0.165). Exposure to 62 Cl-PFESA and PFOS both resulted in the inhibition of mung bean seedling development. Treatment with 62 Cl-PFESA and PFOS led to the concentration-dependent elevation of malondialdehyde (MDA), carbonyl groups, and phosphorylated histone H2AX (γ-H2AX) levels in mung bean roots. The MDA and carbonyl group contents induced by 62 Cl-PFESA were 1.10-1.35 and 1.03-1.14 times, respectively, those of PFOS. The hydroxyl free radical (·OH) levels in mung bean roots after exposure to PFOS and 62 Cl-PFESA were elevated significantly, and the ·OH levels induced by 62 Cl-PFESA were higher than those induced by PFOS. Hydroxyl free radical levels were positively correlated with the MDA and carbonyl group contents in mung bean roots (p less then 0.05). The dynamic changes in some antioxidative enzyme activities in mung bean seedlings were determined, including peroxidase (POD), superoxide dismutase (SOD), and catalase (CAT). The results demonstrated the phytotoxicities of 62 Cl-PFESA and PFOS to mung bean in the early developmental stage. 62 Cl-PFESA is more harmful to mung beans than PFOS. U-Net is well-liked in medical image segmentation, but it doesn't fully explore useful features of the channel and capitalize on the contextual information. Therefore, we present an improved U-Net with residual connections, adding a plug-and-play, very portable channel attention (CA) block and a hybrid dilated attention convolutional (HDAC) layer to perform medical image segmentation for different tasks accurately and effectively, and call it HDA-ResUNet, in which we fully utilize advantages of U-Net, attention mechanism and dilated convolution. In contrast to the simple copy splicing of U-Net in the skip connection, the channel attention block is inserted into the extracted feature map of the encoding path before decoding operation. Since this block is lightweight, we can apply it to multiple layers in the backbone network to optimize the channel effect of this layer's coding operation. In addition, the convolutional layer at the bottom of the "U"-shaped network is replaced by a hybrid dilated attention convolutional (HDAC) layer to fuse information from different sizes of receptive fields. The proposed HDA-ResUNet is evaluated on four datasets liver and tumor segmentation (LiTS 2017), lung segmentation (Lung dataset), nuclear segmentation in microscope images (DSB 2018) and neuron structure segmentation (ISBI 2012). The dice global scores of liver and tumor segmentation (LiTS 2017) reach 0.949 and 0.799. The dice coefficients of lung segmentation and nuclear segmentation are 0.9797 and 0.9081 respectively, and the information theoretic score for the last one is 0.9703. The segmentation results are all more accurate than U-Net with fewer parameters, and the problem of slow convergence speed of U-Net on DBS 2018 is solved.Acute respiratory distress syndrome (ARDS) is a life-threatening lung injury with global prevalence and high mortality. Dynamics analyses show that inhibitor bindings exert significant effect on conformational changes of the active helix α2 and the conserved DFG loop. In vitro, pretreatment of **** human lung fibroblasts with GRg1 prior to exposure to cigarette smoke extract (CSE) reversed the cell ultrastructure disorder, decreased the expression of IL-6 and TNF-α, and significantly attenuated transdifferentiation of **** cells by suppressing α-SMA and collagen I expression. Additionally, GRg1 suppressed the TGF-β1/Smad3 signaling pathway in CSE-stimulated **** cells, whereas Smad3 over-expression abolished the anti-transdifferentiation effect of GRg1. In this study, we extracted and characterized FAs from dried Lucilia sericata larvae and investigated the molecular basis by which FAs promote cutaneous wound healing. We first confirmed that FA sodium salts (FASSs) stimulated proliferation, migration, and tube formation of cultured human umbilical vein endothelial cells (HUVECs) in a dose-dependent manner. We then showed that FASSs promoted endothelial-to-mesenchymal transition (EndMT), which plays an important role in stabilizing the neovasculature during angiogenesis. Mechanistically, FASSs up-regulated the expression of angiogenesis-related growth factors, platelet-derived growth factor (PDGF), transforming growth factor-β1 (TGF-β1), and vascular endothelial growth factor A (VEGFA), and activated angiogenesis-related signaling pathways, AKT, ERK, and TGF-β/Smad3. In a rat acute cutaneous-wound model, FAs promoted wound healing. Following treatment, we further found that expression of anti-apoptosis-related factors (c-****and Bcl-2) was up-regulated and expression of apoptosis-related factors (p53 and Bad) was down-regulated. Our findings suggest that FAs can promote cutaneous wound healing by inducing angiogenesis, partly by activating AKT, ERK, and TGF-β/Smad3 signaling. The relationships among statin therapy, LDL-cholesterol levels, and ICH risk remain controversial. Here, we conduct a systematic review and meta-analysis of dose-dependent statin therapy and ICH risk. Eligible articles were identified by searching MEDLINE from inception up to December 1, 2018. Reference lists of previous meta-analyses were manually searched to retrieve all relevant publications. Statin doses were allocated into one of two groups according to the observed reduction of LDL cholesterol doses that lowered LDL-cholesterol levels ≥35% were regarded as high-dose statin therapy, whereas those that lowered LDL-cholesterol levels less then 35% were regarded as low-dose statin therapy. We retrieved 33 studies involving 203,305 subjects. The pooled analysis indicated that high-dose statin treatment significantly increased the risk of ICH [relative risk (RR), 1.35; 95% confidence interval (CI), 1.08-1.68] and reduced the risk of all stroke (RR, 0.85; 95% CI, 0.78-0.92), ischemic stroke (RR, 0.79; 95% CI, 0.72-0.87), and all-cause mortality (RR, 0.94; 95% CI, 0.90-0.98). The analyses did not detect any association between low-dose statin treatment and ICH (RR, 1.05; 95% CI, 0.88-1.25). Low-dose statin therapy significantly reduced the incidence of all stroke (RR, 0.84; 95% CI, 0.79-0.89), ischemic stroke (RR, 0.81; 95% CI, 0.76-0.86), and all-cause mortality (RR, 0.94; 95% CI, 0.92-0.97). Our data indicate that low-dose statin therapy is a safe and effective ICH treatment, whereas high-dose statin therapy is associated with increased ICH risk. Fenofibrate, a common drug used to treat severe hypertriglyceridemia and mixed dyslipidemia, reversed cellular metabolism and mitochondrial dysfunction in gastric cancer cells through PPARα. Our results show that fenofibrate altered glucose and lipid metabolism, inhibited gastric cancer cell proliferation, and promoted apoptosis in gastric cancer cells. We further show that fenofibrate induced mitochondrial reprogramming via CPT1 and the fatty acid oxidation pathway, as well as by activating the AMPK pathway and inhibiting the HK2 pathway. Additionally, fenofibrate inhibited subcutaneous gastric cancer cell tumor growth without obvious toxicity in ****. Collectively, our results indicate that fenofibrate exhibits anti-tumor activity in vitro and in vivo via the mitochondria and metabolic reprogramming, demonstrating that mitochondrial regulation and the normalization of cancer cell metabolism are novel therapeutic strategies for cancer.

Increasing data support the use of molecular data to refine the diagnostic approach to persistent monocytosis. The absence of a TET2, SRSF2, or ASXL1 mutation has ≥ 90% negative predictive value for a diagnosis of CMML. These data may also reliably differentiate chronic myelomonocytic leukemia, the malignancy that is most associated with mature monocytosis, from several other diseases that can be associated with typically a lesser degree of monocytosis. These include acute myelomonocytic leukemia, acute myeloid leukemia with monocytic differentiation, myelodysplastic syndromes, and myeloproliferative neoplasms driven by BCR-ABL1, PDGFRA, PDGFRB, or FGFR1 rearrangements or PCM1-JAK2 fusions among other rarer aberrations. The combination of monocyte partitioning with molecular data in patients with persistentmyelomonocytic leukemia, acute myeloid leukemia with monocytic differentiation, myelodysplastic syndromes, and myeloproliferative neoplasms driven by BCR-ABL1, PDGFRA, PDGFRB, or FGFR1 rearrangements or PCM1-JAK2 fusions among other rarer aberrations. The combination of monocyte partitioning with molecular data in patients with persistent monocytosis may increase the predictive power for the ultimate development of CMM but has not been prospectively validated. After reasonably excluding a secondary or reactive monocytosis, there should be a concern for and investigation of malignant monocytosis, which includes hematopathologic review of blood and marrow tissues, flow cytometric analysis, and cytogenetic and molecular studies to arrive at an appropriate diagnosis.
Coronavirus Disease 2019 (COVID-19) and cardiovascular (CV) disease have a close relationship that emerged from the earliest reports. The aim of this review is to show the possible associations between COVID-19 and heart failure (HF) with preserved ejection fraction (HFpEF).

We evaluate these predictions using an ethnobiological knowledge assessment tool developed for an off-grid pastoralist population known as Choyeros, from Baja California Sur, Mexico. Our results indicate that while individuals acquire knowledge of most dangerous items and edible resources by early adulthood, knowledge of plants and animals relevant to the age and sex divided labor domains and ecologies (e.g., women's house gardens, men's herding activities in the wilderness) continues to develop into middle adulthood but to different degrees and at different rates for men and women. As the demands of offspring on parents accumulate with age, reproductive-aged adults continue to develop their knowledge to meet their children's needs. After controlling for vision, our analysis indicates that many post-reproductive adults show the greatest ethnobiological knowledge. The thermal hyperalgesia was evaluated by hot plate test, while mechanical allodynia was evaluated by von Frey filaments. After CCI, animals showed a reduction in the nociceptive threshold (p less then 0.001) when compared to the sham group. In von Frey test, the CCI + saline + PBM 50 J/cm2 group showed an increase in nociceptive threshold (p less then 0.001) in all measurement moments in comparison with groups CCI + SALINE + PBM 0 J/cm2, CCI + WAY100635 + PBM 50 J/cm2, and CCI + WAY100635 + PBM 0 J/cm2.
In this review, we provide a comprehensive and contemporary understanding of malignant monocytosis and provide a framework by which the appropriate diagnosis with malignant monocytosis can be rendered. Increasing data support the use of molecular data to refine the diagnostic approach to persistent monocytosis. The absence of a TET2, SRSF2, or ASXL1 mutation has ≥ 90% negative predictive value for a diagnosis of CMML. These data may also reliably differentiate chronic myelomonocytic leukemia, the malignancy that is most associated with mature monocytosis, from several other diseases that can be associated with typically a lesser degree of monocytosis. These include acute myelomonocytic leukemia, acute myeloid leukemia with monocytic differentiation, myelodysplastic syndromes, and myeloproliferative neoplasms driven by BCR-ABL1, PDGFRA, PDGFRB, or FGFR1 rearrangements or PCM1-JAK2 fusions among other rarer aberrations. We evaluate these predictions using an ethnobiological knowledge assessment tool developed for an off-grid pastoralist population known as Choyeros, from Baja California Sur, Mexico. Our results indicate that while individuals acquire knowledge of most dangerous items and edible resources by early adulthood, knowledge of plants and animals relevant to the age and sex divided labor domains and ecologies (e.g., women's house gardens, men's herding activities in the wilderness) continues to develop into middle adulthood but to different degrees and at different rates for men and women. As the demands of offspring on parents accumulate with age, reproductive-aged adults continue to develop their knowledge to meet their children's needs. After controlling for vision, our analysis indicates that many post-reproductive adults show the greatest ethnobiological knowledge.
