  • Elastography is a new promising ultrasonographic technique which is used to differentiate benign and malignant breast lesions based on the stiffness of the lesion.

    The aim of this study was to identify the ultrasound features which are associated with malignancy in subcentimetre thyroid nodules.

    This retrospective study included 454 thyroid nodules <10 mm in size in 413 patients from 2012 to 2016, which were subjected to fine needle aspiration cytology. Each nodule was classified according to the ultrasound patterns of the 2015 American Thyroid Association guidelines and the high suspicion ultrasound features (solid, hypo-echogenicity, irregular margins, microcalcifications, taller-than-wide, interrupted rim calcifications, and extrathyroidal extension) were identified for evaluation of their diagnostic performance.

    Of the American Thyroid Association high suspicion ultrasound features, univariate analysis showed that hypo-echogenicity (sensitivity 81.6% (95% CI 65.7-92.3%), specificity 50.0% (95% CI 43.4-56.6%)), irregular margins (sensitivity 34.2% (95% CI 19.6-51.4%), specificity 92.2% (95% CI 88.0-95.3%)), microcalcifications (sensitivity 23.7% (95% CI 11.4 significantly associated with malignancy in subcentimetre thyroid nodules in our study. Hence, subcentimetre nodules which are hypoechoic with irregular margins may warrant further follow-up.
    Cerebral blood flow is increasingly monitored in preterm infants. Doppler ultrasound of the carotid artery is a widely available method but is operator dependent. Our aim was to design and produce a realistic flow phantom model of the carotid artery of preterm infants.

    Data from cerebral blood flow measurements using Doppler ultrasound of the right common carotid artery from 21 premature newborn infants were used to produce a Doppler flow phantom model with three different vessel diameters. Vessel diameter, continuous and pulsatile flow volume measurements were performed by two blinded observers (with more than eight and 20 years of experience).

    Vessel diameter measurements using the phantom were underestimated by 7%. Continuous flow volume measurements were overestimated by 7% by both observers (observer 1 mean difference 1.5 ± 1.96 SD -3.3 to 6.3 ml/min versus observer 2, 1.9 ± 1.96 SD -3.6 to 7.4 ml/min). Pulsatile flow measurements were overestimated by 12.6% by observer 1 (2.7 ± 1.96 SD -0.6 to 5.9materials, this could be explored using a wall-less model. The flow phantom could be utilised for research and training clinicians in measuring cerebral blood flow using the carotid artery in this vulnerable group of infants.

The emotional labour on sonographers is increased as they struggle to provide patient-centred care, given the limits put on their communication during the ultrasound examination. The aim of this study was to explore Australian sonographers' views on communicating adverse findings, including their level of autonomy in communicating with patients and how this influences their sense of professional identity.

    Following a national survey, seven purposively selected participants, who were qualified to perform obstetric ultrasound examinations, completed follow-up interviews. The interviews were thematically analysed with iterative comparison to the survey results. Three case Sanmiao pill (SMP), a Chinese traditional formula, had been used to treat gouty arthritis (GA). However, the active compounds and underlying mechanism remained unclear. Hence, network pharmacology and molecular docking were utilized to explore bioactive compounds and potential mechanism of action of SMP in treating GA. In the study, the compounds of SMP, corresponding targets, and GA-related targets were mined from various pharmacological databases. Then, herb-compound-target, compound-target, PPI, and target-pathway networks were constructed. Ultimately, molecular docking was carried out to verify the predicted results. The results indicated that 47 active compounds, 338 targets, and 144 disease targets were collected. Network analysis implied that Phellodendron chinense Schneid. played a vital role in the whole formula. Moreover, 7 compounds (quercetin, kaempferol, wogonin, rutaecarpine, baicalein, beta-sitosterol, and stigmasterol) and 4 targets (NFKB1, RELA, MAPK1, and TNF) might be the kernel compounds and targets of SMP against GA. According to GOBP and KEGG pathway enrichment analysis and target-pathway network, SMP might exert a therapeutic role in GA by regulating numerous biological processes and pathways, including lipopolysaccharide-mediated signaling pathway, positive regulation of transcription, Toll-like receptor signaling pathway, JAK-STAT signaling pathway, NOD-like receptor signaling pathway, and MAPK signaling pathway. The results of molecular docking showcased that 11 pairs of compound with targets had tight binding strength. Thereinto, 4 compounds of MAPK1 and 5 compounds of NFKB1 possessed a better combination, suggesting that MAPK1 and NFKB1 might be considered as therapeutic targets in treatment of GA.   • CDK4/6 inhibitors are considered a breakthrough in cancer therapy. Currently approved for breast cancer treatment, CDK4/6 inhibitors are extensively tested in other cancer subtypes. Frequently observed side effects include hematological abnormalities such as reduced numbers of neutrophils, erythroid cells and platelets that are associated with anemia, bleedings and a higher risk of infections. To understand whether the adverse effects within the hematopoietic system are related to CDK4 or CDK6 we generated transgenic **** that lack either CDK4 or CDK6 in adult hematopoiesis. Anemia and perturbed erythroid differentiation are associated with the absence of CDK6 but did not manifest in CDK4-deficient ****. Total CDK6 knockout **** accumulate the most dormant fraction of hematopoietic stem cells due to an impaired exit of the quiescent state. We recapitulated this finding by deleting CDK6 in adult hematopoiesis. In addition, unlike total CDK6 knockout, all stem cell fractions were affected and increased in numbers. The deletion of CDK6 was also accompanied by neutropenia which is frequently seen in patients receiving CDK4/6 inhibitors. This was not the case in the absence of CDK4; CDK4 deficiency resulted in elevated numbers of myeloid progenitors without translating into numeric changes of differentiated myeloid cells. Cell death was associated with cytoplasmic sequestration of heat shock protein 70 and was rescued by induction of HbF synthesis. Importantly, we document that in bone marrow of SCD patients similar cell loss occurs during the final stages of terminal differentiation. Our study provides evidence for ineffective erythropoiesis in SCD and highlights an anti-apoptotic role for HbF during the terminal stages of erythroid differentiation. These findings imply that the beneficial effect on anemia of increased HbF levels is not only due to the increased life span of red cells but also a consequence of decreased ineffective erythropoiesis.In situ follicular neoplasia (ISFN) is the earliest morphologically identifiable precursor of follicular lymphoma (FL). Although it is genetically less complex than FL and has low risk for progression, ISFN already harbors secondary genetic alterations, in addition to the defining t(14;18)(q32;q21) translocation. FL, in turn, frequently progresses to diffuse large B-cell lymphoma (DLBCL) or high-grade B-cell lymphoma (HGBL). By BCL2 staining of available reactive lymphoid tissue obtained at any time point in patients with aggressive B-cell lymphoma (BCL), we identified 10 paired cases of ISFN and DLBCL/HGBL, including 6 de novo tumors and 4 transformed from FL as intermediate step, and investigated their clonal evolution using microdissection and next generation sequencing. A clonal relationship between ISFN and aggressive BCL was established by immunoglobulin and/or BCL2 rearrangements and/or the demonstration of shared somatic mutations for all 10 cases. Targeted sequencing revealed CREBBP, KMT2D, EZH2, TNFRSF14 and BCL2 as the genes most frequently mutated already in ISFN. Based on the distribution of private and shared mutations, two patterns of clonal evolution were evident. In most cases, the aggressive lymphoma, ISFN and, when present, FL revealed divergent evolution from a common progenitor, whereas linear evolution with sequential accumulation of mutations was less frequent.     The COVID-19 pandemic is changing the landscape of healthcare delivery in many countries, with a new shift toward remote patient monitoring (RPM).

    The goal of this perspective is to highlight the existing and future role of wearable and RPM optical technologies in an increasingly at-home healthcare and research environment.

    First, the specific changes occurring during the COVID-19 pandemic in healthcare delivery, regulations, and technological innovations related to RPM technologies are reviewed. Then, a review of the current state and potential future impact of optical physiological monitoring in portable and wearable formats is outlined.

    New efforts from academia, industry, and regulatory agencies are advancing and encouraging at-home, portable, and wearable physiological monitors as a growing part of healthcare delivery. It is hoped that these shifts will assist with disease diagnosis, treatment, management, recovery, and rehabilitation with minimal in-person contact. Some of these trends are likely to persist for years to come. Optical technologies already account for a large portion of RPM platforms, with a good potential for future growth.

The goal of this perspective is to highlight the existing and future role of wearable and RPM optical technologies in an increasingly at-home healthcare and research environment. First, the specific changes occurring during the COVID-19 pandemic in healthcare delivery, regulations, and technological innovations related to RPM technologies are reviewed. Then, a review of the current state and potential future impact of optical physiological monitoring in portable and wearable formats is outlined. New efforts from academia, industry, and regulatory agencies are advancing and encouraging at-home, portable, and wearable physiological monitors as a growing part of healthcare delivery. It is hoped that these shifts will assist with disease diagnosis, treatment, management, recovery, and rehabilitation with minimal in-person contact. Some of these trends are likely to persist for years to come. Optical technologies already account for a large portion of RPM platforms, with a good potential for future growth.   • In forensic facial approximation, facial soft tissue thickness (FSTT) measurements play a major role. These values are affected by many factors such as ethnicity, age and sex, in addition to measurement errors. We hypothesize that an additional source of error is the lack of consideration of facial type in the assessment of FSTT norms. The purpose of this study was to 1- evaluate the presence of significant effects of vertical facial type within the FSTT measurements in adults and 2- assess the correlations between FSTT and hard and soft tissue cephalometric measurements. The sample consisted of the lateral cephalometric radiographs of 222 adult individuals (87 males; 135 females, 23.49±6.24 years of age) with normal occlusion and balanced profiles. Hard and soft tissue cephalometric measurements were taken, in addition to FSST at 10 facial landmarks. The sample was categorized into 3 vertical pattern groups based on the MP/SN angle hypodivergent, normodivergent and hyperdivergent. Statistical analyses included MANOVA test and Pearson moment product for associations among variables. Statistically significant effect of vertical divergence on FSTT values was limited to the levels of Stomion, Labiomentale and Pogonion and FSTT measurements were associated with measurements related to the lower face (Lm and Pog) Moderate to high correlations between mandibular length and ramus length and FSTT values related to the lower face (LL, Lm and Pog mainly) emphasize further the important role of the underlying skeleton.Pluronics are a class of amphiphilic tri-block copolymers with wide pharmaceutical applicability. The attractive inherent antitumor properties of Pluronic polymers in combination with cell targeting and stimuli-responsive ligands greatly improved antitumoral therapeutic effects of tested drugs. In spite of that, the extraordinary complexity of biological challenges in the delivery of micellar drug payload makes their therapeutic potential still not exploited to the fullest. In this review paper we attempt to present the latest developments in the field of Pluronic based nanovehicles and their application in anticancer therapy with an overview of the chemistry involved in the preparation of these nanovehicles.The lateral mobility serves as a key function of the cell membrane and can be regulated by the cholesterol. Here supported lipid bilayers were used to quantitatively analyse the influence of cholesterol on the fluidity of lipid bilayer in the environments with and without an electric field. We observed that with the increase of cholesterol proportion (0-30 mol%), the diffusion coefficient of DOPC lipid bilayer gradually decreased from 1.30 ± 0.15 μm2 s-1 to 0.28 ± 0.13 μm2 s-1, where as that of DPPC lipid bilayer increased slightly from ∼0 to 0.45 ± 0.18 μm2 s-1. We then showed that cholesterol also regulated the movement of charged lipids in the bilayer in the electric field. The migration rate of charged lipids in the DOPC bilayers slowed down as the cholesterol increased. The clarification of the dose-effect relationship of cholesterol for the bidirectional regulating effect on the mobility of lipid bilayer will extend applications of supported lipid bilayers as the model of cell membrane.The development of targeted delivery systems can improve the selectivity of cancer drugs. Additionally, a system that promotes the controlled delivery of the drug triggered by an external stimulus in the exact target tissue is highly desirable. Regarding the light stimulus, the NIR window (650-950 nm) is the most suitable due to its higher capacity of penetration in human tissues and less harmful effects on normal cells. In this work, new red-light-responsive nanoparticles for doxorubicin delivery were developed. The nanoparticles were based on cleavable di-block copolymers of poly(ethylene glycol) (PEG) and poly(lactic acid) (PLA) linked by a red-light sensitive segment (1,2-bis(2-hydroxyethylthio)ethylene, BHETE). The PEG-BHETE-PLA copolymers were synthesized under mild conditions and exhibited a narrow polydispersity. The nanoparticles presented a size between 53 and 133 nm, with a doxorubicin loading capacity between 1.2 and 4.4 wt%. Release study of the encapsulated doxorubicin confirms the light-triggered nanoparticle disassembly process. In vitro cytotoxicity tests in MCF7 cell line, for the light-triggered nanoparticles, showed a decrease in cancer cells' viability higher than 25% compared to non-irradiated cells. Due to the promising results obtained with the light-sensitive PEG-BHETE-PLA nanoparticles, these materials have great potential to be used in drug delivery systems for cancer therapy.Two plant-associated bacterial strains were isolated from Beijing, China. The two strains possessed almost identical 16S rRNA gene sequences. The respiratory quinone was ubiquinone-8 and the major polar lipids were phosphatidylethanolamine, phosphatidylglycerol and diphospatidylglycerol. The major fatty acids of strain 502T were C10 03-OH (26.2%), C160 (12.9%), C170 cyclo (14.5%) and summed feature 3 (21.4%). Furthermore, both strains showed the potential of plant growth promotion. Based on these results, the two isolates could be considered to represent a novel species of the genus Variovorax, for which the name Variovorax beijingensis sp. nov., is proposed, with 502T (= DSM 106862T = CGMCC 1.16560T) as the type strain.

Interestingly, however, it was found that the addition of the thiophene ring in (E)-1-(1H-pyrrol-2-yl)-3-(thiophen-2-yl)prop-2-en-1-one increased both the melting point and solubility of the sample compared with (E)-3-phenyl-1-(1H-pyrrol-2-yl)prop-2-en-1-one.

  • Number concentration is an important index to measure atmospheric particle pollution. However, tailored methods for data preprocessing and characteristic and source analyses of particle number concentrations (PNC) are rare and interpreting the data is time-consuming and inefficient. In this method-oriented study, we develop and investigate some techniques via flexible conditions, C++ optimized algorithms, and parallel computing in R (an open source software for statistics and graphics) to tackle these challenges. The data preprocessing methods include deletions of variables and observations, outlier removal, and interpolation for missing values (NA). They do better in cleaning data and keeping samples and generate no new outliers after interpolation, compared with previous methods. Besides, automatic division of PNC pollution events based on relative values suites PNC properties and highlights the pollution characteristics related to sources and mechanisms. Additionally, basic functions of k-means clustering, Principal Component Analysis (PCA), Factor Analysis (FA), Positive Matrix Factorization (PMF), and a newly-introduced model NMF (Non-negative Matrix Factorization) were tested and compared in analyzing PNC sources. Only PMF and NMF can identify coal heating and produce more explicable results, meanwhile NMF apportions more distinctly and runs 11-28 times faster than PMF. Traffic is interannually stable in non-heating periods and always dominant. Coal heating's contribution has decreased by 40%-86% in recent 5 heating periods, reflecting the effectiveness of coal burning control.
    Artificial light at night (ALAN) may be an anthropogenic stressor for mental health disturbing humans' natural day-night cycle. However, the few existing studies used satellite-based measures of radiances for outdoor ALAN exposure assessments, which were possibly confounded by traffic-related air pollutants.

    To assess 1) whether living in areas with increased exposure to outdoor ALAN is associated with depressive symptoms; and 2) to assess the potential confounding effects of air pollution.

    We used cross-sectional data from people (N = 10,482) aged 18-65 years in the Netherlands. Depressive symptoms were assessed with the Patient Health Questionnaire (PHQ-9). Satellite-measured annual ALAN were taken from the Visible Infrared Imaging Radiometer Suite. ALAN exposures were assessed at people's home address within 100 and 600 m buffers. We used generalized (geo)additive models to quantify associations between PHQ-9 scores and quintiles of ALAN adjusting for several potential confounders including PM
    and ith depressive symptoms. Future evaluations of health effects from ALAN should consider potential confounding by traffic-related exposures (i.e., NO

Plastic items have a great floatability, being this one of the main reasons for their proportional dominance among beach litter, especially in the wrack line. Hundreds of measurements were made of different items using Raman spectroscopy, an effective technique to determine the type of plastic. Cellulose acetate represented the most abundant material (79%) linked to cigarette butts, followed by polypropylene, 9% and polyethylene, 8%. The remaining were composed by polystyrene, polyethylene terephthalate and polyvinyl chloride. Smoke related items were the most common in the study area, followed by film and hard pieces, cotton bud sticks, fishing strings, food wrappers, caps/lids and single-use plastics. Beach litter can be related to beachgoers but some categories can be attributed to fishing activities or wastewater discharges. Plastics cause a multitude of damages to the coastal ecosystem, due to their toxic components and adsorbed substances and their facility of fragmentation and dispersion. Principal objective is the compositional characterization of plastic beach litter items and their spatial variability. It is therefore important to avoid unnecessary consumption, promote recycling and use other materials that are less harmful to the environment.Pollutants from navigation sector are key contributors to emission inventories of most coastal cities with heavy port activities. The use of high fuel sulfur content (FSC) bunker oil by ocean going vessels (OGVs) has been identified as a major source of sulfur dioxide (SO2). Government authorities all over the world, including Hong Kong government, have implemented air pollution control regulations to cap FSC of fuel used by OGVs to 0.5%, from the existing 3.5%, to reduce SO2 emissions. However, the lack of efficient screening tools to identify non-compliant OGVs has prevented effective enforcement. This study developed and evaluated an unmanned aerial vehicle (UAV)-borne lightweight (750 g) microsensor system (MSS), which is capable of measuring ship plume SO2, NO2, NO, CO2, CO, and particulate matter in real-time. Extensive experiments were conducted on the sensor system to evaluate its performance during laboratory and field operations.   • Using well-characterized Arabidopsis npq mutants, we showed for the first time the quantitative link between SIF, photosynthetic efficiency, NPQ components and leaf reflectance. We discuss the functional potential and limitations of SIF and reflectance measurements for estimating photosynthetic efficiency and NPQ in the field.Sildenafil (SIL) has potential as an interesting gastroprotective drug. However, the pathways of its protective effect still needs to be clarified, and its use as a potential gastroprotective agent validated. This study aims to evaluate the effects of SIL via modulation of oxidative stress in a NSAID-induced gastric lesion model. Male Swiss **** were divided into six groups control (CON, water), nonsteroidal anti-inflammatory drug (NSAID, water), proton pump inhibitor (PPI, 30 mg/kg of lansoprazole), SIL 5 (5 mg/kg), SIL 25 (25 mg/kg) and SIL 50 (50 mg/kg). The animals were treated by gavage (a single dose) after 24 hours of fasting, and gastric lesions were performed after 30 minutes, with indomethacin (40 mg/kg, by gavage). After 6h, the animals were killed and the stomach was removed to evaluate reactive oxygen species (ROS) production, oxidation of macromolecules, quantification of antioxidant enzymes, DNA fragmentation, apoptosis and macroscopic and histologic analysis of gastric lesions. SIL exerts a dose-dependent gastroprotective effect against NSAID-induced mucosal injury, also reducing cytoplasmic levels of ROS and consequent oxidative damage to macromolecules. In addition, SIL increases nitric oxide bioavailability, antioxidant enzymes and gastric cellular viability, as well as restoring important factors involved in gastroprotection.     The proposed CNN (denoted as Incept-net) followed the general framework of encoder-decoder network, with an assumption that local image information was sufficient for modeling the nonlinear physical process of multi-energy CT. Incept-net was implemented with a customized loss function, including an in-house-designed image-gradient-correlation (IGC) regularizer to improve edge preservation. The network consisted of two types of customized multibranch modules exploiting multiscale feature representation to improve the robustness over local image noise and artifacts. Women with obesity of >35 years-of-age (n = 100) and ≤35 years (n = 44) participated in a behavioral weight loss treatment and were assessed on psychological factors and physical activity and eating behaviors over 6 months. At baseline, the younger group demonstrated significantly higher scores on physical activity and depression, and significantly lower scores on fruit/vegetable intake. Improvements in Tension (i.e., anxiety), Overall Negative Mood, and dimensions of EE from baseline-Month 3 were significantly greater in the younger group. Similar significant improvements in physical activity and the intake of sweets and fruits/vegetables over 6 months were observed in both groups. Incorporating aggregated data, change in physical activity was significantly related to mood improvements.     The dog displayed intermittent, non-weight bearing lameness of the right forelimb for approximately 18 months before presenting at the veterinary medical center. Medial subluxation of the right elbow joint was detected by palpation. The Campbell test was consistent with an increased range of motion during supination. At ultrasonographic examination, medial luxation of the triceps brachii tendon was noted, whereas collateral ligaments appeared normal. No skeletal deformities were found on radiographs of the right forelimb. The luxation of the triceps brachii tendon was surgically corrected with antirotational suture, a stopper pin, medial retinaculum release, and imbrication of the lateral retinaculum.

    The right triceps brachii tendon and elbow joint were successfully reduced. Gait returned to normal by 55 days postoperatively. No implant failure or recurrence were observed 3.5 years after surgery.

This review investigated the effect of these changes on global all-cause and cause-specific mortality in polytrauma patients admitted to the intensive care unit (ICU). Moreover, changes in trauma mechanism over time and differences between continents were analyzed. A systematic review of literature on all-cause mortality in polytrauma patients admitted to ICU was conducted. All-cause and cause-specific mortality rates were extracted as well as trauma mechanism of each patient. Poisson regression analysis was used to model time trends in all-cause and cause-specific mortality. Thirty studies, which reported mortality rates for 82,272 patients, were included and showed a decrease of 1.8% (95% CI 1.6-2.0%) in all-cause mortality per year since 1966. The relative contribution of brain injury-related death has increased over the years, whereas the relative contribution of death due to multiple organ dysfunction syndrome (MODS), acute respiratory distress syndrome, and sepsis decreased. MODS was the most common cause of death in North America, and brain-related death was the most common in Asia, South America, and Europe. However, results of validated and standardized in vivo efficacy tests allowing the comparison of different products are lacking.

    Efficacy of six skin protection products against sodium lauryl sulphate-induced irritant contact dermatitis was investigated in a double blind, randomized and controlled repetitive irritation study in healthy volunteers using skin physiological methods. Transepidermal water loss (TEWL) results were used to calculate an efficacy index of protection by mathematical/statistical means according to a protocol developed in a German Statutory Accident Insurance (DGUV) multicenter study (research project FP 275).

In contrast, the influence of the topology of the central π-electron system on the relevant properties such as the band gap or the fluorescence behavior has not yet been addressed. In this work, pyrene is compared with its structural isomer azupyrene, which has a π-electron system with non-alternant topology. Using photoelectron spectroscopy, near edge X-ray absorption fine structure spectroscopy, and other methods, it is shown that the electronic band gap of azupyrene is by 0.72 eV smaller than that of pyrene. The difference of the optical band gaps is even larger with 1.09 eV, as determined by ultraviolet-visible absorption spectroscopy. The non-alternant nature of azupyrene is also associated with a more localized charge distribution.

In this context, bicatenary biobased lipidic-cyclodextrins of low DS were easily synthesized from a fatty ester epoxide by means of alternative methods (ball-milling conditions, use of enzymes). The ring opening reaction of methyl oleate epoxide needs ball-milling and is highly specific of cyclodextrins in solventless conditions. L-CDs are thus composed of complex mixtures that were deciphered by an extensive structural analysis using mainly mass spectrometry and NMR spectroscopy. In addition, as part of their potential use as vectors of active drugs, these products were submitted to an integrity study on in vitro model of the blood-brain-barrier (BBB) and the intestinal epithelium. No toxicity has been observed, suggesting that applications for the vectorization of active ingredients can be expected.Hepatocellular carcinoma (HCC) is a complex biological process and is often diagnosed at advanced stages with no effective treatment options. With advances in tumor biology and molecular genetic profiling, several different signaling pathways and molecular mechanisms have been identified as responsible for initiating and promoting HCC. Targeting these critical pathways, which include the receptor tyrosine kinase pathways, the Ras mitogen-activated protein kinase (Ras/Raf/MAPK), the phosphatidylinositol 3-kinase (PI3K)/AKT/mammalian target of rapamycin (mTOR), the Wnt/β-catenin signaling pathway, the ubiquitin/proteasome degradation and the hedgehog signaling pathway has led to the identification of novel therapeutics for HCC treatment. The WNK2-methylated human glioblastoma cell line A172 WT (wild type) was compared to transfected clones A172 EV (empty vector), and A172 WNK2 (WNK2 overexpression) for the evaluation of autophagy using an inhibitor (bafilomycin A1-baf A1) and an inducer (everolimus) of autophagic flux. Western blot and immunofluorescence approaches were used to monitor autophagic markers, LC3A/B and SQSTM1/p62. A172 WNK2 cells presented a significant decrease in LC3B and p62 protein levels, and in LC3A/B ratio when compared with control cells, after treatment with baf A1 + everolimus, suggesting that WNK2 overexpression inhibits the autophagic flux in gliomas. The mTOR pathway was also evaluated under the same conditions, and the observed results suggest that the inhibition of autophagy mediated by WNK2 occurs through a mTOR-independent pathway. A targeted metabolomics analysis was performed using an AbsoluteIDQ® p180 kit, flow injection analysis, and liquid chromatography-tandem mass spectrometry. The highest variable importance in projection (VIP) scores for the progression to diabetes of the amino acids and phospholipids were associated with isoleucine and LPC acyl C281, respectively. Using logistic regression analysis, we found that high baseline isoleucine concentration was associated with a higher incidence of diabetes, while high LPC acyl 281 was associated with a lower incidence. Isoleucine and LPC acyl 281 were independently associated with incident diabetes in a model that also included conventional risk factors for diabetes (baseline fasting plasma glucose (FPG), age, sex, and body mass index (BMI)). In addition, isoleucine and LPC acyl 281 were independently associated with serum HbA1c 5 years later in a robust regression model that also included baseline FPG, age, sex, and BMI. Isoleucine, LPC acyl 281, age, and FPG were significantly associated with HbA1c at this time. In conclusion, these results provide evidence that isoleucine and LPC acyl C281 have respective positive and negative independent associations with incident diabetes.

Targeting these critical pathways, which include the receptor tyrosine kinase pathways, the Ras mitogen-activated protein kinase (Ras/Raf/MAPK), the phosphatidylinositol 3-kinase (PI3K)/AKT/mammalian target of rapamycin (mTOR), the Wnt/β-catenin signaling pathway, the ubiquitin/proteasome degradation and the hedgehog signaling pathway has led to the identification of novel therapeutics for HCC treatment. In this review, we elaborated on our current understanding of the signaling pathways involved in the development and initiation of HCC and anticipate the potential targets for therapeutic drug development.Autophagy is a cell-survival pathway with dual role in tumorigenesis, promoting either tumor survival or tumor death. WNK2 gene, a member of the WNK (with no lysine (K)) subfamily, acts as a tumor suppressor gene in gliomas, regulating cell migration and invasion; however, its role in autophagy process is poorly explored. The WNK2-methylated human glioblastoma cell line A172 WT (wild type) was compared to transfected clones A172 EV (empty vector), and A172 WNK2 (WNK2 overexpression) for the evaluation of autophagy using an inhibitor (bafilomycin A1-baf A1) and an inducer (everolimus) of autophagic flux. Western blot and immunofluorescence approaches were used to monitor autophagic markers, LC3A/B and SQSTM1/p62. A172 WNK2 cells presented a significant decrease in LC3B and p62 protein levels, and in LC3A/B ratio when compared with control cells, after treatment with baf A1 + everolimus, suggesting that WNK2 overexpression inhibits the autophagic flux in gliomas. The mTOR pathway was also evaluated under the same conditions, and the observed results suggest that the inhibition of autophagy mediated by WNK2 occurs through a mTOR-independent pathway. Using a water-assisted FCCVD process with optimized conditions, a transition from multi- to double-walled CNTs was observed with a decrease in diameters from 19-23 nm to 10-15 nm in tandem with an elevated Raman IG/ID ratio up to 10.23, and corroborated from the decomposition peak shifts in thermogravimetric data. To characterize the mechanical and electrical improvements, the FCCVD-CNT/bismaleimide (BMI) composites with different water concentrations were manufactured, revealing high electrical conductivity of 1720 S/cm along the bundle alignment (collection) direction, and the nano-indentation tests showed an axial reduced modulus at 65 GPa. A consistent value of the anisotropic ratio at ~3 was observed comparing the longitudinal and transverse properties. The continuous capability of the presented method while maintaining high quality is expected to result in an improved DWCNT mass production process and potentially enhance the structural and electrical applications of CNT nanocomposites.Branched-chain amino acids (BCAAs) and lysophosphatidylcholines (LPCs) have been reported to be associated with diabetes. The purpose of the present study was to investigate the relative contributions of BCAAs and LPCs to the progression of prediabetes to diabetes using a targeted metabolomic approach. This study was part of a health survey of employees of the Electricity Generating Authority of Thailand (n = 79; nine females and 70 males). A targeted metabolomics analysis was performed using an AbsoluteIDQ® p180 kit, flow injection analysis, and liquid chromatography-tandem mass spectrometry. The highest variable importance in projection (VIP) scores for the progression to diabetes of the amino acids and phospholipids were associated with isoleucine and LPC acyl C281, respectively. Using logistic regression analysis, we found that high baseline isoleucine concentration was associated with a higher incidence of diabetes, while high LPC acyl 281 was associated with a lower incidence. Isoleucine and LPC acyl 281 were independently associated with incident diabetes in a model that also included conventional risk factors for diabetes (baseline fasting plasma glucose (FPG), age, sex, and body mass index (BMI)). In addition, isoleucine and LPC acyl 281 were independently associated with serum HbA1c 5 years later in a robust regression model that also included baseline FPG, age, sex, and BMI. Isoleucine, LPC acyl 281, age, and FPG were significantly associated with HbA1c at this time. In conclusion, these results provide evidence that isoleucine and LPC acyl C281 have respective positive and negative independent associations with incident diabetes.
