  • The authors propose "Barocrinology", a novel terminology in medical literature, to comprehensively describe the rapidly expanding field of obesity medicine. This new term highlights the need to appreciate the role of endocrine physiology in the evolution of obesity, insights into its complications and changes in the hormonal milieu following weight loss therapies, including bariatric surgery. This term would not only reduce the stigma associated with obesity in affected individuals but also give them a better appreciation of its biological origin rather than self-criticism. This paper also undermines the importance of a multi-disciplinary approach needed in this field for achieving practical and sustainable goals, individualized to each person.This communication describes a framework to help plan and implement glucose lowering strategies in hospitalized patients. Termed the SECURE model, it utilizes six clinical characteristics to decide intensity of insulin therapy. The SECURE rubric is more comprehensive than the conventional gluco-centric approach promoted by routinely used algorithms. SECURE promotes good clinical sense and pragmatic care in the critical care setting.Management of endogenous Cushing syndrome is based on its aetiology. Dynamic post contrast MRI is currently the most common investigation implied to diagnose pituitary adenoma; however, it comes with the drawback of low specificity and high false positive results. Inferior petrosal sinus sampling (IPSS) is an established invasive procedure performed to differentiate central versus peripheral source of ACTH which, in turn, results in hypercortesolaemia. This is a series of 14 patients who underwent IPSS at the Department of Radiology, Aga Khan University Hospital, Karachi, from January 2006 to December 2018. The case series emphasises the role of IPSS in the management of ACTH-dependent Cushing syndrome and combined efficacy of Dynamic post-contrast MRI and the procedure under focus.This case report is of a 31-year-old female who presented with splinted and faulty metalloceramic prosthesis on maxillary anterior teeth, which was responsible for localised periodontal disease and poor aesthetics. At multiple sites the ceramic had chipped off and the prosthesis colour did not match with the adjacent healthy teeth. A multidisciplinary approach of endodontics, periodontics and prosthodontics was employed for this case. The treatment plan consisted of removing the faulty prosthesis, endodontic retreatment of inadequately root-treated teeth and endodontics in the other coronally prepared abutment teeth. Crown lengthening surgery was performed for the correction of periodontal pockets and high smile line and maintenance of an adequate biological width. After periodontal healing, all-ceramic individual crowns were placed on the maxillary anterior teeth along with a three-unit all-ceramic bridge for the replacement of the left upper maxillary first premolar. It is, therefore, advisable to consider placing individual crowns rather than multiunit splinted prosthesis whenever possible.Neural Tube Defects (NTDs) are serious congenital abnormalities and most of them are incompatible with life. The extremely debilitating quality of life, if one survives, calls for actions to prevent such sufferings. Experts agree on the role of Folic Acid in primary prevention of NTDs, yet, despite best efforts, the use of Folic Acid has reduced NTDs by only 50%. These cases too can be prevented by employing secondary preventive measures. These involve timely interruption of pregnancy -- a decision which, in addition to a medical judgment, is based on ethics, social, cultural and Muslim religious value systems in Pakistan. Indeed, it is a complex issue but empathic understanding and strong co-ordination, once established between different disciplines, can help parents to decide and opt for necessary secondary prevention by interruption of malformed foetus within the given time frame mandated by medical and religious authorities.Lipomas in the submandibular region are very uncommon. Large submandibular neck mass (greater than 10 cm) with a rapid growth rate, may raise concern about possible malignancy of salivary glands. Failure to distinguish salivary gland tumour and liposarcoma from a lipoma may represent a medico-legal pitfall. It is very important for the surgeon to rule out liposarcomas when dealing with giant lipomas in such regions. We report a case of a 65-year-old male who presented in the OPD with complains of pain and a 15x12 cm, giant submandibular solitary lipoma of anterior neck which had rapidly increased in size. The diagnosis of lipoma was confirmed on physical examination, radiological investigations and Fine Needle Aspiration Cytology (FNAC). The Patient underwent surgical excision. The surgery produced excellent cosmetic results and no functional impairment. This study illustrates the literature regarding aetiology, epidemiology, followed by diagnostic and treatment modalities of submandibular lipomas.Neuroendocrine tumours (NETs) of the liver are generally metastatic lesions from broncho-pulmonary or gastrointestinal primary lesions. Liver is an unusual primary site for a NET with only 150 reported cases in literature. We encountered two cases of primary hepatic NET (PHNET) at the Shaukat Khanum Memorial Cancer Hospital, Lahore. Both the patients had abdominal pain and hepatomegaly. Imaging revealed a large mass in the right lobe of the liver in both the cases. The tru-cut biopsy showed neuroendocrine tumour. Extensive workup to look for primary lesion elsewhere in the body turned out to be negative. One patient received neo-adjuvant chemotherapy along with right main portal vein embolisation. Once adequate future liver remnant was achieved, the patient underwent right hepatic trisectionectomy. In the other patient, anticipated future liver remnant was sufficient and underwent surgery without portal vein embolization. No immediate postoperative complication was observed, and both the patients were followed for more than one year.

  • Eight new compounds (Entanutilins O-V; 1-8), including four limonoids, two steroids, one triterpenoid and one lignan were isolated from the stem barks of Entandrophragma utile. Their structures were determined by detailed spectroscopic analyses (HRESIMS and 1D/2D-NMR). Bioactivity screening indicated that compounds 1, 6 and 7 exhibited effective in reversing resistance in MCF-7/DOX cells at a nontoxic concentration of 30 μM with 18.18-, 7.43- and 7.94-fold enhancing effect, respectively, meanwhile, compounds 5 and 6 showed moderate NO inhibitory activities in LPS-activated RAW 264.7 macrophages. We have shown previously that alpha-santalol, a major component of sandalwood oil inhibits growth of cultured prostate cancer cells in vitro by causing apoptosis, but the mechanism of cell death is not fully elucidated. The present study was undertaken to investigate the role of PI3K/Akt/survivin pathway in alpha-santalol-induced apoptosis employing cultured LNCaP and PC-3 human prostate cancer cells. Treatment of prostate cancer cells with alpha-santalol (20, 40 μM) resulted in the down regulation of survivin and p-AKT (s-473) expression and statistically significant reduction in total survivin levels as evidenced by survivin ELISA assay. Furthermore, inhibition of PI3K-Akt pathway by pharmacological inhibitor, LY294002 enhanced the apoptotic cell death induced by alpha-santalol as determined by cell viability, cellular morphology, active caspase-3 activity and expression of cleaved PARP, cleaved caspase-3 levels. In conclusion, the present study provides novel insight into the molecular circuitry of alpha-santalol-induced cell death and reveals that alpha-santalol targets Akt/Survivin pathway to induce cell death and that the cell death is increased in the presence of a known inhibitor of the pathway. Five new alkaloids (1-5), including three new aporphine alkaloids and two new phenanthrene alkaloids, together with 10 known compounds (6-15) were obtained from the roots of Stephania tetrandra. Their structures were elucidated by spectroscopic methods, single-crystal X-ray diffraction, and electronic circular dichroism analyses. Compounds 7-10, and 13 showed antioxidant activities with malondialdehyde (MDA) inhibitory rates of 62.50 ± 1.91 to 98.44 ± 0.34% at the concentration of 10 μM. Eight new flavonoids daemoflavans A-H, including two dimeric proanthocyanidins (1 and 2), four flavans (3-6), two 2-arylbenzofurans (7 and 8), along with nine known compounds (9-17), were isolated from the fruit of Daemonorops draco. Their structures, including the absolute configurations, were elucidated by extensive spectroscopic data, ECD analysis, and X-ray crystal diffraction. Besides, the X-ray crystal data of a known compound dracoflavan B1 (9) was firstly reported. Recent studies have demonstrated that programmed necrosis (necroptosis) is a delayed component of ischemic neuronal injury and our previous study has shown that pannexin 1 channel is involved in cerebral ischemic injury and cellular inflammatory response. Here, we examined whether the pannexin 1 channel inhibitor, 10panx, could reduce focal ischemic brain injury in rats by inhibiting cellular necroptosis and the associated inflammation. Male Sprague-Dawley rats were randomly divided into sham-operated, MCAO (transient middle cerebral artery occlusion) group, and 10panx-treated groups. We investigated the effect of 10panx by assessing infarct volume and neurological deficit. Further, we determined the potential mechanism using immunofluorescent staining, Western blotting, enzyme-linked immunosorbent assay (ELISA) and TUNEL assay. We demonstrated that 10panx reduced infarct volume and alleviated neurological deficit in the MCAO injury model. 10panx ameliorated post-ischemic neuronal death, but it did not reduce the TUNEL positive neurons and expression of cleaved-caspase3. In contrast, expression of necroptosis related protein receptor-interacting protein 3 (RIP3) was significantly decreased. Furthermore, 10panx reduced the release of high mobility group box 1 (HMGB1) from neurons and inhibited microglial activation and secretion of pro-inflammatory factors. Immunent co-labeling of RIP3 with HMGB1 showed that RIP3 protein was closely related with the release of HMGB1 from nucleus to cytoplasm. Our data suggested that 10panx treatment may ameliorate MCAO injury by reducing RIP3-mediated necroptosis, HMGB1 release and associated inflammatory response. RIP3 may play an important role in the release of HMGB1 and inflammation after stroke. AIMS SGLT2 inhibitors have been proposed as an adjunct to insulin therapy for glycemic control in type 1 diabetes (T1D) patients. However, concern has been raised due to an increase in renin-angiotensin-system (RAS) activity reported in a clinical trial in which an SGLT2 inhibitor was added while insulin dose was reduced in T1D patients. We previously reported that insulin inhibits intrarenal angiotensinogen (Agt) gene transcription and RAS activation. We hypothesized that insulin, rather than SGLT2 inhibition might regulate the intrarenal RAS. METHODS We compared RAS activity in non-diabetic wild type ****, Akita **** (T1D model) and Akita **** treated with insulin or the SGLT2 inhibitor canagliflozin. RESULTS Treatment of Akita **** with insulin or canagliflozin produced similar reductions in blood glucose, whereas insulin, but not canagliflozin, reduced elevated systolic blood pressure. Akita **** exhibited increased renal Agt mRNA/protein expression, which was attenuated by insulin, but not by canagliflozin.

  • Therapeutic efficiency of amphiphilic methotrexate-camptothecin (MTX-CPT) prodrug compared to free drug mixture (MTX/CPT) has been investigated using all-atom molecular dynamics simulation and first principles density functional theory calculations. This comparison revealed that MTX-CPT prodrug tends to form spherical self-assembled nanoparticle (NP), while free MTX/CPT mixture forms rod-shape NP. These observations are attributed to a structural defect in the MTX-CPT prodrug and solvation free energies of MTX, CPT and MTX-CPT molecules. The results provided evidence that noncovalent interactions (NCIs) among the pharmaceutical drugs play a very important role in anticancer agents aggregation process, leading to enhanced stability of the self-assembled NPs. It is found that the stability of MTX-CPT self-assembled NP is greater than the MTX/CPT NP due to the synergistic effect of hydrogen bonding between monomers and solvent (water). Moreover, the noncatalyzed as well as catalyzed hydrolysis reactions of MTX-CPT prodrug are theoretically studied at the PCM(water)//M06-2X/6-31G(d,p) computational level to shed additional light on the role of acidic condition in tumor tissues. We found that the ester hydrolysis in mild acidic solutions is a concerted reaction. In an agreement between theory and experiment, we also confirmed that the activation energies of the catalyzed-hydrolysis steps are **** lower than the activation energies of the corresponding steps in the noncatalyzed reaction. BACKGROUND Despite the negative impact of marital violence on female nurses' health, few studies have investigated its physical and mental aspects. METHOD A descriptive correlational design was used to investigate the relationship between marital violence and the physical and mental health of female Jordanian nurses. One hundred ninety-eight female Jordanian nurses from five governmental hospitals were recruited. CONCLUSION Jordanian female nurses suffered hazardous physical and mental health consequences of violence that adversely affect their entire life.     Therapeutic efficiency of amphiphilic methotrexate-camptothecin (MTX-CPT) prodrug compared to free drug mixture (MTX/CPT) has been investigated using all-atom molecular dynamics simulation and first principles density functional theory calculations. This comparison revealed that MTX-CPT prodrug tends to form spherical self-assembled nanoparticle (NP), while free MTX/CPT mixture forms rod-shape NP. These observations are attributed to a structural defect in the MTX-CPT prodrug and solvation free energies of MTX, CPT and MTX-CPT molecules. The results provided evidence that noncovalent interactions (NCIs) among the pharmaceutical drugs play a very important role in anticancer agents aggregation process, leading to enhanced stability of the self-assembled NPs. It is found that the stability of MTX-CPT self-assembled NP is greater than the MTX/CPT NP due to the synergistic effect of hydrogen bonding between monomers and solvent (water). Moreover, the noncatalyzed as well as catalyzed hydrolysis reactions of MTX-CPT prodrug are theoretically studied at the PCM(water)//M06-2X/6-31G(d,p) computational level to shed additional light on the role of acidic condition in tumor tissues. We found that the ester hydrolysis in mild acidic solutions is a concerted reaction. In an agreement between theory and experiment, we also confirmed that the activation energies of the catalyzed-hydrolysis steps are much lower than the activation energies of the corresponding steps in the noncatalyzed reaction. This study investigated the role of the feeding regime on cellular (lysosomal membrane impairment), oxidative (superoxides and nitric oxides generation, as well as lipid peroxidation) and genotoxic (nuclear abnormalities) biomarkers measured in hemocytes of mussels Mytilus galloprovincialis treated with diclofenac (DCF). Specifically, unfed mussels, or mussels fed ad libitum with algal species Tisochrysis lutea or Tetraselmis suecica (Tiso/DCF- and Tetra/DCF- treated mussels, respectively) were exposed to DCF (20 μgL-1) for 4 days. The results showed that biomarkers' responsiveness against DCF, were more pronounced in unfed and Tetra/DCF-, rather than Tiso/DCF- treated mussel hemocytes, thus revealing food deprivation, changes in mussel feeding/filtration rate and digestion processes, as potent factors of mussels' immune efficiency and response against DCF. Those findings could provide valuable data for the optimization of mussels' feeding regime during laboratory studies, in order to assess reliably the effects of emerging contaminants on non-target sentinel organisms, such as mussels. Wild fish belonging to four species belonging to different trophic groups were captured at three distances from fish farm facilities long distance (>5 Km), medium distance (1.5 Km) and close to sea-cages. Flesh, brain, liver and gonads were sampled for fatty acid analysis. Fish aggregated near sea-cages showed accumulation of fatty acids of vegetable origin in the studied tissues, due to surplus feed consumption or via predation of fish that consumed the feed. Results of the toxicity tests showed that the microalgae sensitivity was not correlated. The integration of chemical data did not allow to discriminate toxicity effects but contributed to highlight that D. tertiolecta was the most sensitive microalgae (no cell wall) followed by P. tricornutum and S. costatum. Further analysis, including lines of evidence and weight of evidence approaches to calculate risk quotients of elutriate samples, confirmed these results. The effects of increasing or decreasing extreme temperatures on bivalves depend on their physiological and biochemical capacity to respond to changes in ambient temperature. We tested the response of the clam Anomalocardia flexuosa to simulated marine heat waves and cold spells, under summer and winter experimental conditions. We sought information about physiological and biochemical parameters, as well as survival rates during two bioassays of 43 days each. The winter cold spell simulations showed that extreme temperatures acted as a physiological and biochemical stimulus, linked to an increase in metabolic rates, and consequently higher maintenance costs, as acclimatory strategies. On the other hand, the summer heat wave extreme temperatures exceeded the individuals' thermal tolerance limits, resulting in an inability to acclimate and a high mortality. These experiments suggest that A. flexuosa can be considered as a sensitive indicator of heat wave events. In April and August 2015, a massive fire occurred in the Chernobyl Exclusion zone. The fire spread to neighboring areas due to the prevailing strong winds. In this paper, we analyze the peat fires as a unique source of radioactive contamination. After an expedition directly to the peat fire site (the evacuation zone of the Bryansk region), we collected a number of aerosol samples. We came to the conclusion that peat fires cannot be the reason for radioactive particle transport in the Bryansk region as well as in the Chernobyl evacuation zone. During the peat fire, radioactive contamination was not transferred by aerosols beyond 500 m. The 137Cs concentration in the aerosol filters varied between 0.55 and 0.64 Bq/m3, and that at the same distance from the fire seat and peat edge was 4.4∙10-3 Bq/m3; the activity values in the peat bog and in the nearest inhabited locality did not exceed the background values. Strontium-90 was not found in aerosol samples. The soil-to-air transport rate of 137Cs was 2.7∙10-6. After the Chernobyl accident, the majority of the 137Cs was incorporated into the structure of clay minerals, and these did not change during the peat fire. The mobility of 137Cs in the flight peat material particles was established. To confirm the territory status of the evacuation zone, we also collected some food samples. The purpose of our study was to conduct a comparative analysis of the morphological states of the dentition of individuals living in regions exposed to radiation to determine the groups at risk for the main classes of dental anomalies. We believe our results will support the development of a differentiated system for dental rehabilitation and follow-up of individuals exposed to radiation. After an expedition directly to the peat fire site (the evacuation zone of the Bryansk region), we collected a number of aerosol samples. We came to the conclusion that peat fires cannot be the reason for radioactive particle transport in the Bryansk region as well as in the Chernobyl evacuation zone. During the peat fire, radioactive contamination was not transferred by aerosols beyond 500 m. The 137Cs concentration in the aerosol filters varied between 0.55 and 0.64 Bq/m3, and that at the same distance from the fire seat and peat edge was 4.4∙10-3 Bq/m3; the activity values in the peat bog and in the nearest inhabited locality did not exceed the background values. Strontium-90 was not found in aerosol samples. The soil-to-air transport rate of 137Cs was 2.7∙10-6. After the Chernobyl accident, the majority of the 137Cs was incorporated into the structure of clay minerals, and these did not change during the peat fire. The mobility of 137Cs in the flight peat material particles was established. To confirm the territory status of the evacuation zone, we also collected some food samples. Berries and mushrooms consumed at the assumed rate for dose estimation would result in doses that exceed the public dose limit by approximately a factor of five. The 1986 accident at the Chernobyl Nuclear Power Plant led to large-scale changes in the environmental situation. The purpose of our study was to conduct a comparative analysis of the morphological states of the dentition of individuals living in regions exposed to radiation to determine the groups at risk for the main classes of dental anomalies. We believe our results will support the development of a differentiated system for dental rehabilitation and follow-up of individuals exposed to radiation. The prevalence rate of dental anomalies was studied in 1,889 patients of both sexes divided by age in accordance with dentition formation stages and by regions of residence in accordance with the 137Cs soil-contamination level. A statistically significant decrease was observed in the number of patients with normal dentition for their age among those who had been exposed to prenatal radiation. Specifically, unfed mussels, or mussels fed ad libitum with algal species Tisochrysis lutea or Tetraselmis suecica (Tiso/DCF- and Tetra/DCF- treated mussels, respectively) were exposed to DCF (20 μgL-1) for 4 days. We sought information about physiological and biochemical parameters, as well as survival rates during two bioassays of 43 days each. The winter cold spell simulations showed that extreme temperatures acted as a physiological and biochemical stimulus, linked to an increase in metabolic rates, and consequently higher maintenance costs, as acclimatory strategies. On the other hand, the summer heat wave extreme temperatures exceeded the individuals' thermal tolerance limits, resulting in an inability to acclimate and a high mortality.   • This manuscript reviews the state-of-the-art regarding human biological monitoring (HBM) of mycotoxins in plasma serum and blood samples. After a comprehensive and systematic literature review, with a focus on the last five years, several aspects were analyzed and summarized a) the biomarkers analyzed and their encountered levels, b) the analytical methodologies developed and c) the relationship between biomarker levels and some illnesses. In the literature reviewed, aflatoxin B1-lysine (AFB1-lys) and ochratoxin A (OTA) in plasma and serum were the most widely studied mycotoxin biomarkers for HBM. Regarding analytical methodologies, a clear increase in the development of methods for the simultaneous determination of multiple mycotoxins has been observed. For this purpose, the use of liquid chromatography (LC) methodologies, especially when coupled with tandem mass spectrometry (MS/MS) or high resolution mass spectrometry (HRMS), has grown. A high percentage of the samples analyzed for OTA or aflatoxin B1 (mostly as AFB1-lys) in the reviewed papers were positive, demonstrating human exposure to mycotoxins. This review confirms the importance of mycotoxin human biomonitoring and highlights the important challenges that should be faced, such as the inclusion of other mycotoxins in HBM programs, the need to increase knowledge of mycotoxin metabolism and toxicokinetics, and the need for reference materials and new methodologies for treating samples. In addition, guidelines are required for analytical method validation, as well as equations to establish the relationship between human fluid levels and mycotoxin intake.Prostaglandin A2-AcMe (1) and Prostaglandin A2 (2) were isolated from the octocoral Plexaura homomalla and three semisynthetic derivatives (3-5) were then obtained using a reduction protocol. All compounds were identified through one- and two-dimensional (1D and 2D) nuclear magnetic resonance (NMR) experiments. Additionally, evaluation of in vitro cytotoxic activity against the breast (MDA-MB-213) and lung (A549) cancer cell lines, in combination with enzymatic activity and molecular docking studies with the enzymes p38α-kinase, Src-kinase, and topoisomerase IIα, were carried out for compounds 1-5 in order to explore their potential as inhibitors of cancer-related molecular targets. Results showed that prostaglandin A2 (2) was the most potent compound with an IC50 of 16.46 and 25.20 μg/mL against MDA-MB-213 and A549 cell lines, respectively. In addition, this compound also inhibited p38α-kinase in 49% and Src-kinase in 59% at 2.5 μM, whereas topoisomerase IIα was inhibited in 64% at 10 μM. Enzymatic activity was found to be consistent with molecular docking simulations, since compound 2 also showed the lowest docking scores against the topoisomerase IIα and Src-kinase (-8.7 and -8.9 kcal/mol, respectively). For the longitudinal validation, we recruited 563 diabetic patients and collected 1363 urine samples with the clinical data during a follow-up of 6 years. Comparative urinary proteomics identified four proteins Apolipoprotein A-I (APOA1), Beta-2-microglobulin (B2M), E-cadherin (CDH1) and Lithostathine-1-alpha (REG1A), which differentiated with high statistical strength (p less then 0.05) between DN patients and the other groups. Label-free mass spectrometric quantification of the candidates confirmed the discriminatory value of E-cadherin and Lithostathine-1-alpha (p less then 0.05). Immunological validation highlighted E-cadherin as the only marker able to differentiate significantly between the different DN stages with an area under the curve (AUC) of 0.85 (95%-CI [0.72, 0.97]).   • Similar to other studies, our study showed that gcgr-/- zebrafish exhibited decreased ureagenesis and impaired cholesterol metabolism. More interestingly, we found that the glycerophospholipid metabolism was disrupted, the arachidonic acid metabolism was up-regulated, and the tryptophan metabolism pathway was down-regulated in gcgr-/- zebrafish. Based on the omics data, we further validated our findings by revealing that gcgr-/- zebrafish exhibited dampened melatonin diel rhythmicity and increased locomotor activity. These global omics data provide us a better understanding about the role of GCGR in regulating metabolic network and new insight into GCGR physiological functions.
    Colon cancer (CC) currently ranks as the third most common human cancer worldwide with an increasing incidence and a poor prognosis. Recently, circular RNAs have been reported to regulate the progression of diverse human cancers. However, the role of circRNA hsa_circ_0020095 in CC remains largely unclear.

    Expression levels of the related circRNAs, microRNAs and mRNA in CC tissues and cells were determined. The impacts of circ_0020095 or miR-487a-3p on CC cells were examined at the indicated times after transfection. Meanwhile, a luciferase-reporter experiment was employed to validate the interplay between miR-487a-3p and circ_002009695 or SOX9. Moreover, the
    tumor growth assay was applied to further evaluate the effects of circ_0020095 knockdown on CC progression.

    We demonstrated that circ_0020095 was highly expressed in CC tissues and cells. The proliferation, migration, invasion, and cisplatin resistance of CC were suppressed by silencing circ_0020095

    or by ectopic expression of miR-487a-3p
    . Mechanistically, circ_0020095 could directly bind to miR-487a-3p and subsequently act as a miR-487a-3p sponge to modulate the activity by targeting the 3'-UTR of SOX9. Interestingly, overexpression of circ_0020095 dramatically reversed the suppressive effects of miR-487a-3p mimics on CC cells.

    Similar to other studies, our study showed that gcgr-/- zebrafish exhibited decreased ureagenesis and impaired cholesterol metabolism. More interestingly, we found that the glycerophospholipid metabolism was disrupted, the arachidonic acid metabolism was up-regulated, and the tryptophan metabolism pathway was down-regulated in gcgr-/- zebrafish. Based on the omics data, we further validated our findings by revealing that gcgr-/- zebrafish exhibited dampened melatonin diel rhythmicity and increased locomotor activity. Colon cancer (CC) currently ranks as the third most common human cancer worldwide with an increasing incidence and a poor prognosis. Recently, circular RNAs have been reported to regulate the progression of diverse human cancers. However, the role of circRNA hsa_circ_0020095 in CC remains largely unclear. Expression levels of the related circRNAs, microRNAs and mRNA in CC tissues and cells were determined. The impacts of circ_0020095 or miR-487a-3p on CC cells were examined at the indicated times after transfection. Meanwhile, a luciferase-reporter experiment was employed to validate the interplay between miR-487a-3p and circ_002009695 or SOX9. Moreover, the tumor growth assay was applied to further evaluate the effects of circ_0020095 knockdown on CC progression. We demonstrated that circ_0020095 was highly expressed in CC tissues and cells. The proliferation, migration, invasion, and cisplatin resistance of CC were suppressed by silencing circ_0020095 and or by ectopic expression of miR-487a-3p . Mechanistically, circ_0020095 could directly bind to miR-487a-3p and subsequently act as a miR-487a-3p sponge to modulate the activity by targeting the 3'-UTR of SOX9. Interestingly, overexpression of circ_0020095 dramatically reversed the suppressive effects of miR-487a-3p mimics on CC cells. METHODS A total of 164 caregivers of children diagnosed with ASD were asked which autism services they were currently using and completed the Beach Center Family Quality of Life Scale and the Nisonger Child Behaviour Rating Form. RESULTS Findings revealed that service usage type significantly predicted families' satisfaction with their emotional well-being, physical/material well-being, and disability-related support. Specifically, families using a combination of mental health services and ADL therapies reported greater satisfaction in these FQOL domains. Electrochemical behavior of the DPMC-CuNPs and DPMC-NiNPs was analyzed by cyclic voltammetry method. DNA binding studies of the synthesized compounds with CT-DNA were examined by four different techniques such as UV-Visible and emission spectroscopy, cyclic voltametry and viscometric measurments. Thermal denaturation and sono-chemical denaturation studies of DNA with the DPMC, DPMC-CuNPs and DPMC-NiNPs results also suggest the synthesized compounds have good DNA binding ability. Various antioxidant scavenging studies results shows that DPMC and its copper and nickel nanoparticles have significant antioxidant activity. Antimicrobial studies of the DPMC and its MNPs were studied by Agar-Agar well diffusion method. Anticancer studies of the DPMC and its MNPs show that the DPMC-CuNPs and DPMC-NiNPs have significant anticancer activity with least toxicity than the standard drug cis-platin. The above was confirmed by the absence of ANS fluorescence in the presence of a fluorescein isothiocyanate (FITC)-labeled N-domain. Further, the labeling of the N-domain with FITC was hindered by the presence of ANS, i.e., ANS was bound to the ATP binding site. Importantly, ANS displayed a higher affinity than ATP. In addition, ANS binding led to quenching the N-domain intrinsic fluorescence displaying a FRET pattern, which suggested the existence of a Trp-ANS FRET couple.   • Induction of intrinsic liver regeneration is an unmet need that can be achieved by temporally activating key hepatocyte regenerative pathways. Here, we establish an efficient, safe, non-integrative method to transiently express hepatocyte-growth-factor (HGF) and epidermal-growth-factor (EGF) in hepatocytes via nucleoside-modified, lipid-nanoparticle-encapsulated mRNA (mRNA-LNP) delivery in ****. We confirm specific hepatotropism of mRNA-LNP via intravenous injection of firefly luciferase encoding mRNA-LNP, with protein expression lasting about 3 days. In the liver, virtually all hepatocytes are transfected along with a subpopulation of endothelial and Kupffer cells. In homeostasis, HGF mRNA-LNP efficiently induce hepatocyte proliferation. Here we assessed in detail its function by performing RNA-sequencing profiling coupled with bioinformatic prediction to identify HELLS targets and transcriptional cooperators. We demonstrated that HELLS, together with the transcription factor YY1, contributes to an appropriate cytokinesis via the transcriptional regulation of genes involved in cleavage furrow regulation. Binding target promoters, HELLS primes YY1 recruitment and transcriptional activation of cytoskeleton genes including the small GTPases RhoA and RhoU and their effector kinase Pak2. Single or multiple knockdowns of these genes reveal that RhoA and RhoU mediate HELLS effects on cell proliferation and cell division of ALK-ALCLs.

We recently identified the DNA-helicase HELLS as central for proficient ALK-ALCL proliferation and progression. Here we assessed in detail its function by performing RNA-sequencing profiling coupled with bioinformatic prediction to identify HELLS targets and transcriptional cooperators. We demonstrated that HELLS, together with the transcription factor YY1, contributes to an appropriate cytokinesis via the transcriptional regulation of genes involved in cleavage furrow regulation. Binding target promoters, HELLS primes YY1 recruitment and transcriptional activation of cytoskeleton genes including the small GTPases RhoA and RhoU and their effector kinase Pak2. Single or multiple knockdowns of these genes reveal that RhoA and RhoU mediate HELLS effects on cell proliferation and cell division of ALK-ALCLs.
