Differentiation therapies with all-trans retinoic acid (ATRA) have been successful in treating acute promyelocytic leukemia, a rare subtype of acute myeloid leukemia (AML). However, their efficacy is limited in the case of other AML subtypes. Here, we show that the combination of ATRA with salt-inducible kinase (SIK) inhibition significantly enhances ATRA-mediated AML differentiation. SIK inhibition augmented the ability of ATRA to induce growth inhibition and G1 cell cycle arrest of AML cells. Moreover, combining ATRA and SIK inhibition synergistically activated the Akt signaling pathway but not the MAPK pathway. Pharmacological blockade of Akt activity suppressed the combination-induced differentiation, indicating an essential role for Akt in the action of the combination treatment. Taken together, our study reveals a novel role for SIK in the regulation of ATRA-mediated AML differentiation, implicating the combination of ATRA and SIK inhibition as a promising approach for future differentiation therapy.Hereditary red blood cell (RBC) membranopathies are characterized by mutations in genes encoding skeletal proteins that alter the membrane complex structure. The recent availability of targeted next generation sequencing (t-NGS) and its combination with RBC deformability measured with a laser-assisted optical rotational ektacytometer (LoRRca) has demonstrated to be the most powerful contribution to lower the percentage of hereditary hemolytic anemia undiagnosed cases. In order to know the kind and frequency of RBC membrane mutations in our geographical area (Catalonia) and to better understand their pathophysiology, 42 unrelated, non-transfusion-dependent (NTD) patients with hereditary hemolytic anemia have been studied by combining t-NGS and LoRRca. The osmoscan module of LoRRca provides three rheological profiles that reflect the maximal deformability (EImax), osmotic fragility (Omin), and hydration state (Ohyper) of RBCs and contribute to a better understanding of the contribution RBC rheology to the severity of anemia. From the 42 patients studied, 37 were suspected to be a RBC membrane defect due to phenotypic characteristics and abnormal RBC morphology and, from these, in 31 patients (83.8% of cases) the mutation was identified by t-NGS. No definite diagnosis was achieved in 11 patients (26.2% of cases), including 6 out of 37 cases, with suspected membranopathy, and 5 with unclassifiable HHA. In all these undiagnosed patients, the existence of hemoglobinopathy and/or enzymopathy was ruled out by conventional methods.Due to overcrowding and subsequent unavoidable close contact, poor ventilation, and decreased hygiene standards and healthcare services as compared with in the community setting, the prison environment is highly conducive to the transmission of infections, including COVID-19. Physical distancing measures may be difficult to implement without introducing interventions that may violate human rights. Given that Indigenous women represent over 41% of federally incarcerated women, this is a cause for concern. Indigenous women are also more likely to have higher rates of numerous chronic conditions, including respiratory illnesses, substantially increasing vulnerability to COVID-19 complications. This can be exacerbated in prisons as access to healthcare may be limited. Outbreaks within the prison setting can not only overwhelm an already over-stretched healthcare system but also spread to the community and disproportionately impact marginalized communities and populations. In this commentary, we explore the public health and human rights implications of COVID-19 in prisons while calling particular attention to the unique needs and circumstances of incarcerated Indigenous women based on international best practice-based guidance to preventive and responsive measures to COVID-19.
Prostate cancer is a major cause of cancer-related death in males worldwide and, in addition to impairing prostate function, also causes testicular adaptations. In this study, we aim to investigate the preventive effect of exercise training on PCa-induced testicular dysfunction.
As a model, we used fifty Wistar Unilever male rats, randomly divided in four experimental groups. Prostate cancer was chemically and hormonally induced in two groups of animals (PCa groups). One control group and one PCa group was submitted to moderate intensity treadmill exercise training. Fifty weeks after the start of the training the animals were sacrificed and sperm, prostate, testis and serum were collected and analyzed. Sperm concentration and morphology, and testosterone serum levels were determined. In addition, histological analyses of the testes were performed, and testis proteomes and metabolomes were characterized.
We found that prostate cancer negatively affected testicular function, manifested as an arrest of spermatogenesis. Oxidative stress-induced DNA damage, arising from reduced testis blood flow, may also contribute to apoptosis of germ cells and consequential spermatogenic impairment. Decreased utilization of the glycolytic pathway, increased metabolism of ketone bodies and the accumulation of branched chain amino acids were also evident in the PCa animals. Conversely, we found that the treadmill training regimen activated DNA repair mechanisms and counteracted several metabolic alterations caused by PCa without impact on oxidative stress.
These findings confirm a negative impact of prostate cancer on testis function and suggest a beneficial role for exercise training in the prevention of prostate cancer-induced testis dysfunction.

Deubiquitination, the inverse process of ubiquitination, is catalyzed by deubiquitinases (DUBs) that remove ubiquitin from target proteins and subsequently prevent their degradation by proteasomes. Previously, deubiquitination has been found to be involved in hepatocellular carcinoma (HCC) progression. As yet, however, little is known about the exact role of deubiquitination in the development and/or progression of this type of cancer.
HCC tissues and tissue microarrays were used to detect expression of the DUB ubiquitin-specific protease 2a (USP2a). The critical role of USP2a in HCC development and progression was assessed in both in vitro cell and in vivo animal models. LC-MS/MS analyses were performed to identify potential targets of USP2a in HCC cells, after which regulation of target protein stability and ubiquitin status by USP2a were investigated.

Primary sources include data collected from interviews and surveys of Indigenous peoples during expeditions to the Nyda settlement, the Nydinskaya tundra, the Tazovsky settlement, the Tazovskaya tundra, the Nakhodka tundra, the Gyda and Gydansky settlements, the Yavai-Salinskaya tundra, the Seyakha settlement, the Seyakhinskaya and Tambeyskaya tundras located along the southern coast of the Ob Bay, the northeast coast of the Yamal Peninsula, the Tazovsky and Gydansky Peninsulas, and the Shuryshkarsky district. Data were collected during the summers and winters of 2014-2020.The utilization of a glass-adhesive-steel joint in modern machine and vehicle production is constantly growing. Despite the technological regime during the production process, various defects in the adhesive joints may occur. One of the major problems is lack of adhesive between joined materials. Due to the widespread use of non-transparent layers increasing adhesion, it is frequently not possible to conduct simple visual inspections. Hence, it is important to develop a non-destructive adhesive path continuity examination procedure. In that process, the different physical properties of the joint materials must be taken into account. Therefore, in this paper various testing methods were used, including ultrasound, thermographic and electromagnetic methods operating in the microwave and terahertz frequency range. Different physical phenomena of the methods allowed for examination of the joint in a wide context. In order to verify the information brought by each method, the results were transferred into common coordinate space, compared and finally integrated. Various opinion pooling strategies were conducted to fuse data, which allowed us to emphasize convergent and complementary information on adhesive continuity. The obtained results are promising and constitute the basis for further work on an integrated system for automatic evaluation of a wide range of possible defects in glass-adhesive-steel joints.Natural pathogen pressure is an important factor that shapes the host immune defense mechanism. The transcriptome of two-spotted field crickets was assembled into 233,172 UniGenes, which yielded approximately 163.58 million reads. On the other hand, there were 43,055 genes in common that were shared among all the biological samples. Gene Ontology analysis successfully annotated 492 immune-related genes, which comprised mainly Pattern Recognition Receptors (62 genes), Signal modulators (57 genes), Signal transduction (214 genes), Effectors (36 genes), and another immune-related 123 genes. In summary, the identified wide range of immune-related genes from G. bimaculatus indicates the existence of a sophisticated and specialized broad spectrum immune mechanism against invading pathogens, which provides, for the first time, insights into the molecular mechanism of disease resistance among two-spotted field crickets.In order to enable timely actions to prevent major losses of crops caused by lack of nutrients and, hence, increase the potential yield throughout the growing season while at the same time prevent excess fertilization with detrimental environmental consequences, early, non-invasive, and on-site detection of nutrient deficiency is required. Therefore, this metalloporphyrin derived from biomass represents an interesting alternative as a potential therapeutic drug for the treatment of CL through PDT, especially for patients with intolerance to the chemotherapeutic drugs currently available.Cutaneous angiosarcoma is a rare neoplasm. One important predictor of recurrence is the resection margin; however, identifying the tissue area containing malignant cells is difficult. Indocyanine green fluorescence angiography (ICG-FA) has been used to identify superficial malignancies, including malignant tumors in the liver and sentinel lymph node metastasis of breast cancer. ICG-FA is also used to identify and define the resection margin of cutaneous angiosarcomas. However, there are currently only a few reports on the application of ICG-FA for detecting cutaneous angiosarcomas. We report a case of cutaneous angiosarcoma in the scalp in which one lesion, located in the parietal scalp, showed high fluorescence intensity corresponding exactly with the lesion was defined by physical examination, whereas the malignant area of the second lesion, located in the occipital scalp, was revealed more accurately by ICG-FA than by physical examination. Further, the second lesion was the first case diagnosed as angiosarcoma by the limited-area biopsy for a high-intensity area of ICG-FA. By determining where ICG is located within cutaneous angiosarcomas and quantitating the ICG intensity level corresponding to the malignant area, ICG-FA could be a promising tool for identifying cutaneous angiosarcomas.

Corresponding low sensitivities (0% to 21%) and kappa-values (<0.50) in detecting aortic sac changes were found. The risk of missing true growth between three and 12 months were lower (6%, 5%, 6%, and 6%, respectively), and matching sensitivities 33%, 33%, 17%, and 17%, respectively.

Indocyanine green fluorescence angiography (ICG-FA) has been used to identify superficial malignancies, including malignant tumors in the liver and sentinel lymph node metastasis of breast cancer. ICG-FA is also used to identify and define the resection margin of cutaneous angiosarcomas. However, there are currently only a few reports on the application of ICG-FA for detecting cutaneous angiosarcomas. We report a case of cutaneous angiosarcoma in the scalp in which one lesion, located in the parietal scalp, showed high fluorescence intensity corresponding exactly with the lesion was defined by physical examination, whereas the malignant area of the second lesion, located in the occipital scalp, was revealed more accurately by ICG-FA than by physical examination. Further, the second lesion was the first case diagnosed as angiosarcoma by the limited-area biopsy for a high-intensity area of ICG-FA. By determining where ICG is located within cutaneous angiosarcomas and quantitating the ICG intensity level corresponding to the malignant area, ICG-FA could be a promising tool for identifying cutaneous angiosarcomas. Perampanel is a highly selective and non-competitive α-amino-3-hydroxy-5 -methyl-4-isoxazole propionate (AMPA) receptor (AMPAR) antagonist, which has been licensed as an orally administered antiepileptic drug in more than 55 countries. Recently, perampanel was found to exert neuroprotective effects in hemorrhagic and ischemic stroke models.

We found that perampanel significantly preserved morphological changes, attenuated lactate dehydrogenase (LDH) release and inhibited caspase-3 activation after TNI. The TNI-induced necroptosis, as evidenced by flow cytometry, was markedly reduced by perampanel treatment. The results of western blot showed that perampanel decreased the expression and phosphorylation of the necroptotic factors, receptor protein interacting kinase 1 (RIPK1) and RIPK3. In addition, treatment with perampanel increased the phosphorylation of Akt and GSK3β in a time-dependent manner up to 24 h after TNI. Treatment with the Akt inhibitor LY294002 partially reversed the protective effects of perampanel.
Our present data suggest that necroptosis plays a key role in the pathogenesis of neuronal death after TNI, and that perampanel might have therapeutic potential for patients with traumatic brain injury (TBI).

Protein levels of beta-catenin, MMP-2 and MMP-9 were evaluated by Western blotting.
In this study, we found that GPR4 and TCF7 had the capacity to control cell division by altering cell cycle distribution, anchorage-independent growth, and directional cell motility of ovarian cancer cell, A2780. Also, we showed that knockdown of GPR4 and TCF7 in ovarian cancer cells A2780 induced significantly decreased cell growth and decreased invasion, as well as increased apoptosis. Down-regulation of TCF7 resulted in the decreased MMP-2 and MMP-9 level.
The results implicate that GPR4 behaves like an oncogene and may function through WNT pathway molecule, TCF7. Downregulation of GPR4 and TCF7 essentially inhibited cell growth and invasion and enhanced apoptosis of ovarian cancer cells, which may lay the foundation of ovarian cancer treatment.

For decades, Praziquantel has been the undisputed drug of choice for all schistosome infections, but rising concerns due to the unelucidated mechanism of action of the drug and unavoidable reports of emerging drug resistant strains has necessitated the need for alternative treatment drug. Moreover, current apprehension has been reinforced by total dependence on the drug for treatment hence, the search for novel and effective anti-schistosomal drugs. Uses This study made use of bioinformatic tools to determine the structural binding of the Universal G4LZI3 stress protein (USP) in complex with ten polyphenol compounds, thereby highlighting the effectiveness of these recently identified 'lead' molecules in the design of novel therapeutics targeted against schistosomiasis. Upregulation of the G4LZI3 USP throughout the schistosome multifaceted developmental cycle sparks interest in its potential role as a druggable target. The integration of in silico tools provides an atomistic perspective into the binding of potential inhibitors to target proteins.

Testing under what conditions a product satisfies the desired properties is a fundamental problem in manufacturing industry. If the condition and the property are respectively regarded as the input and the output of a black-box function, this task can be interpreted as the problem called level set estimation (LSE) the problem of identifying input regions such that the function value is above (or below) a threshold. Although various methods for LSE problems have been developed, many issues remain to be solved for their practical use. As one of such issues, we consider the case where the input conditions cannot be controlled precisely-LSE problems under input uncertainty. We introduce a basic framework for handling input uncertainty in LSE problems and then propose efficient methods with proper theoretical guarantees. The proposed methods and theories can be generally applied to a variety of challenges related to LSE under input uncertainty such as cost-dependent input uncertainties and unknown input uncertainties. We apply the proposed methods to artificial and real data to demonstrate their applicability and effectiveness.The ability to encode and manipulate data structures with distributed neural representations could qualitatively enhance the capabilities of traditional neural networks by supporting rule-based symbolic reasoning, a central property of cognition. Here we show how this may be accomplished within the framework of Vector Symbolic Architectures (VSAs) (Plate, 1991; Gayler, 1998; Kanerva, 1996), whereby data structures are encoded by combining high-dimensional vectors with operations that together form an algebra on the space of distributed representations. In particular, we propose an efficient solution to a hard combinatorial search problem that arises when decoding elements of a VSA data structure the factorization of products of multiple codevectors. Our proposed algorithm, called a resonator network, is a new type of recurrent neural network that interleaves VSA multiplication operations and pattern completion. We show in two examples-parsing of a tree-like data structure and parsing of a visual scene-how the factorization problem arises and how the resonator network can solve it. More broadly, resonator networks open the possibility of applying VSAs to myriad artificial intelligence problems in real-world domains. The companion article in this issue (Kent, Frady, Sommer, & Olshausen, 2020) presents a rigorous analysis and evaluation of the performance of resonator networks, showing it outperforms alternative approaches.Pruning is an effective way to slim and speed up convolutional neural networks. Generally previous work directly pruned neural networks in the original feature space without considering the correlation of neurons. We argue that such a way of pruning still keeps some redundancy in the pruned networks. In this letter, we proposed to prune in the intermediate space in which the correlation of neurons is eliminated. To achieve this goal, the input and output of a convolutional layer are first mapped to an intermediate space by orthogonal transformation. Then neurons are evaluated and pruned in the intermediate space. Extensive experiments have shown that our redundancy-aware pruning method surpasses state-of-the-art pruning methods on both efficiency and accuracy. Notably, using our redundancy-aware pruning method, ResNet models with three times the speed-up could achieve competitive performance with fewer floating point operations per second even compared to DenseNet.We develop theoretical foundations of resonator networks, a new type of recurrent neural network introduced in Frady, Kent, Olshausen, and Sommer (2020), a companion article in this issue, to solve a high-dimensional vector factorization problem arising in Vector Symbolic Architectures. Given a composite vector formed by the Hadamard product between a discrete set of high-dimensional vectors, a resonator network can efficiently decompose the composite into these factors. We compare the performance of resonator networks against optimization-based methods, including Alternating Least Squares and several gradient-based algorithms, showing that resonator networks are superior in several important ways. This advantage is achieved by leveraging a combination of nonlinear dynamics and searching in superposition, by which estimates of the correct solution are formed from a weighted superposition of all possible solutions. While the alternative methods also search in superposition, the dynamics of resonator networks allow them to strike a more effective balance between exploring the solution space and exploiting local information to drive the network toward probable solutions. Resonator networks are not guaranteed to converge, but within a particular regime they almost always do. In exchange for relaxing the guarantee of global convergence, resonator networks are dramatically more effective at finding factorizations than all alternative approaches considered.Working memory is essential it serves to guide intelligent behavior of humans and nonhuman primates when task-relevant stimuli are no longer present to the senses. Thus far, neural network models of working memory have been unable to offer an integrative account of how such control mechanisms can be acquired in a biologically plausible manner. Here, we present WorkMATe, a neural network architecture that models cognitive control over working memory content and learns the appropriate control operations needed to solve complex working memory tasks. Key components of the model include a gated memory circuit that is controlled by internal actions, encoding sensory information through untrained connections, and a neural circuit that matches sensory inputs to memory content. Testing under what conditions a product satisfies the desired properties is a fundamental problem in manufacturing industry. If the condition and the property are respectively regarded as the input and the output of a black-box function, this task can be interpreted as the problem called level set estimation (LSE) the problem of identifying input regions such that the function value is above (or below) a threshold. Although various methods for LSE problems have been developed, many issues remain to be solved for their practical use. As one of such issues, we consider the case where the input conditions cannot be controlled precisely-LSE problems under input uncertainty. We introduce a basic framework for handling input uncertainty in LSE problems and then propose efficient methods with proper theoretical guarantees. The proposed methods and theories can be generally applied to a variety of challenges related to LSE under input uncertainty such as cost-dependent input uncertainties and unknown input uncertainties. We apply the proposed methods to artificial and real data to demonstrate their applicability and effectiveness.The ability to encode and manipulate data structures with distributed neural representations could qualitatively enhance the capabilities of traditional neural networks by supporting rule-based symbolic reasoning, a central property of cognition. Here we show how this may be accomplished within the framework of Vector Symbolic Architectures (VSAs) (Plate, 1991; Gayler, 1998; Kanerva, 1996), whereby data structures are encoded by combining high-dimensional vectors with operations that together form an algebra on the space of distributed representations. In particular, we propose an efficient solution to a hard combinatorial search problem that arises when decoding elements of a VSA data structure the factorization of products of multiple codevectors. Our proposed algorithm, called a resonator network, is a new type of recurrent neural network that interleaves VSA multiplication operations and pattern completion. In this letter, we proposed to prune in the intermediate space in which the correlation of neurons is eliminated. To achieve this goal, the input and output of a convolutional layer are first mapped to an intermediate space by orthogonal transformation. Then neurons are evaluated and pruned in the intermediate space. Extensive experiments have shown that our redundancy-aware pruning method surpasses state-of-the-art pruning methods on both efficiency and accuracy. Notably, using our redundancy-aware pruning method, ResNet models with three times the speed-up could achieve competitive performance with fewer floating point operations per second even compared to DenseNet.We develop theoretical foundations of resonator networks, a new type of recurrent neural network introduced in Frady, Kent, Olshausen, and Sommer (2020), a companion article in this issue, to solve a high-dimensional vector factorization problem arising in Vector Symbolic Architectures. Given a composite vector formed by the Hadamard product between a discrete set of high-dimensional vectors, a resonator network can efficiently decompose the composite into these factors. We compare the performance of resonator networks against optimization-based methods, including Alternating Least Squares and several gradient-based algorithms, showing that resonator networks are superior in several important ways. This advantage is achieved by leveraging a combination of nonlinear dynamics and searching in superposition, by which estimates of the correct solution are formed from a weighted superposition of all possible solutions. While the alternative methods also search in superposition, the dynamics of resonator networks allow them to strike a more effective balance between exploring the solution space and exploiting local information to drive the network toward probable solutions. Resonator networks are not guaranteed to converge, but within a particular regime they almost always do. Chimeric antigen receptor (CAR)-T cell therapy has drawn **** attention due to its recent clinical success in B-cell malignancies. In general, the CAR-T cell discovery process consists of CAR identification, T-cell activation, transduction, and expansion, as well as assessment of CAR-T cytotoxicity. The current evaluation methods for the CAR-T discovery process can be time-consuming, low-throughput and requires the preparation of multiple sacrificial samples in order to produce kinetic data. In this study, we employed the use of a plate-based image cytometer to monitor anti-CAIX (carbonic anhydrase IX) G36 CAR-T generation and assess its cytotoxic potency of direct and selective killing against CAIX+ SKRC-59 human renal cell carcinoma cells. The transduction efficiency and cytotoxicity results were analyzed using image cytometry and compared directly to flow cytometry and Chromium 51 (51 Cr) release assays, showing that image cytometry was comparable against these conventional methods. Image cytometry method streamlines the assays required during the CAR-T cell discovery process by analyzing a plate of T cells from CAR-T generation to in vitro functional assays with minimum disruption. The proposed method can reduce assay time and uses less cell samples by imaging and analyze the same plate over time without the need to sacrifice any cells. Discontinuation or non-publication of trials may hinder scientific progress and violates the commitment made to research participants. We sought to identify the prevalence of discontinuation and non-publication of heart failure (HF) clinical trials.
We conducted a cross-sectional search of ClinicalTrials.gov to identify all completed and discontinued HF clinical trials. We limited our search to only include trials that were completed by 31 December 2017. Trials were investigated to identify reasons for discontinuation. Informative termination was defined as trial termination due to safety or efficacy concerns. Data pertaining to the trial phase, funding, intervention, enrolment, and trial completion date were extracted for each trial. A total of 572 trials were included. Of these, 21% (n=118) were discontinued before completion. Patient accrual was the most frequently cited reason (n=42; 36%) for trial discontinuation, followed by informative termination (n=16; 14%) and funding (n=14; 12%). Overall, 24780 patients were enrolled in trials that were terminated. Of trials that were completed and not terminated, nearly one-third (n=131/454; 29%) were not published. Seventy-nine (24%) trials were published within 12months, 192 (59%) within 24months, and 252 (78%) trials within 36months.

Morphological and chemical characterization of the prepared materials were performed using scanning electron microscopy (SEM), energy-dispersive spectroscopy (EDS), and X-ray photoelectron spectroscopy (XPS). We aimed to study the prevalence of human papillomavirus (HPV) in conjunctival intraepithelial neoplasia and carcinoma. Furthermore, we aimed to explore whether geographical differences or different detection modalities are associated with the conflicting information regarding HPV and the development of the disease.
We searched the MEDLINE, EMBASE and Scopus databases for studies reporting on HPV and conjunctival intraepithelial neoplasia or carcinoma. The pooled prevalence proportions, odds ratio (OR) and corresponding 95% confidence intervals were calculated assuming a random-effects model. Subgroup analyses and meta-regression explored possible sources of heterogeneity.
A total of 39 studies were included in the systematic review. To analyze the radiographic and clinical outcomes of deltoid ligament repair, we studied 34 supination external rotation type IV ankle fracture patients with deltoid ruptures who required syndesmosis fixation.
We retrospectively evaluated 34 cases of ruptured deltoid ligaments with associated supination external rotation type IV ankle fractures with a widening of the syndesmosis between 2011 and 2017. All of the ankle fractures were treated with a similar surgical protocol (plate and screw fixation). Syndesmotic fixations were performed on all patients. The deltoid ligaments were surgically repaired in 19 patients and in 15 patients, they were not. The preoperative, immediate postoperative, and final follow-up radiographic outcomes (medial clear space, overlap space, and clear space) and clinical scores (Foot and Ankle Outcome Scores and the Foot Function Index) were compared. Patients were followed for an average of 13.6months.

The immune evasion of tumor cells may be the main cause of the difficulties of γδT cell-based treatment. In the present study, we explored whether and how B7-H3 regulates the resistance of CC cells to the cytotoxicity of Vγ9Vδ2 (Vδ2) T cells. We observed that B7-H3 overexpression promoted, while B7-H3 knockdown inhibited, CC cell resistance to the killing effect of Vδ2 T cells in vitro and in vivo. Mechanistically, we showed that B7-H3-mediated CC cell resistance to the cytotoxicity of Vδ2 T cells involved a molecular pathway comprising STAT3 activation and decreased ULBP2 expression. ULBP2 blockade or knockdown abolished the B7-H3 silencing-induced increase in the cytotoxicity of Vδ2 T cells to CC cells. Furthermore, cryptotanshinone, a STAT3 phosphorylation inhibitor, reversed the B7-H3 overexpression-induced decrease in ULBP2 expression and attenuated the killing effect of Vδ2 T cells on CC cells. Moreover, there was a negative correlation between the expression of B7-H3 and ULBP2 in the tumor tissues of CC patients. The arabinan utilization operon of L. crispatus DSM29598 encodes enzymes for degradation of arabinan, α-galactosidases, β-galactosidases, and enzymes and for utilization of arabinose including phosphoketolase. The two putative extracellular arabinanases, AbnA and AbnB, are homologous to family GH43 endo-arabinanases. In Lactobacillaceae, homologs of these enzymes were identified exclusively in vertebrate-adapted species of the genus Lactobacillus. L. crispatus grew with arabinan from sugar beet pectin as sole carbon source, indicating extracellular arabinanase activity, and produced lactate and acetate, indicating metabolism via the phosphoketolase pathway. The two arabinanases AbnA and AbnB were heterologously expressed and purified by affinity chromatography. AbnA hydrolyzed linear and branched arabinan, while AbnB hydrolyzed only linear arabinan. The optimum pH for AbnA and AbnB was 6 and 7.5, respectively; 40 °C was the optimum temperature for both enzymes. The application of arabinan degrading L. crispatus as probiotic or as synbiotic with pectins may improve the production of short-chain fatty acids from pectin to benefit host health. We observed that B7-H3 overexpression promoted, while B7-H3 knockdown inhibited, CC cell resistance to the killing effect of Vδ2 T cells in vitro and in vivo. Mechanistically, we showed that B7-H3-mediated CC cell resistance to the cytotoxicity of Vδ2 T cells involved a molecular pathway comprising STAT3 activation and decreased ULBP2 expression. ULBP2 blockade or knockdown abolished the B7-H3 silencing-induced increase in the cytotoxicity of Vδ2 T cells to CC cells. Furthermore, cryptotanshinone, a STAT3 phosphorylation inhibitor, reversed the B7-H3 overexpression-induced decrease in ULBP2 expression and attenuated the killing effect of Vδ2 T cells on CC cells. Moreover, there was a negative correlation between the expression of B7-H3 and ULBP2 in the tumor tissues of CC patients. Our results suggest that the B7-H3-mediated STAT3/ULBP2 axis may be a potential candidate target for improving the efficiency of γδT cell-based immunotherapy in CC.Extracellular glycosyl hydrolases are uncommon in lactobacilli and include amylases and fructosidases mediating starch and fructan utilization, respectively. This study is aimed at identifying the function of an arabinan utilization operon in Lactobacillus crispatus DSM29598 and at characterizing two putative extracellular arabinanases that are located on that operon. The arabinan utilization operon of L. crispatus DSM29598 encodes enzymes for degradation of arabinan, α-galactosidases, β-galactosidases, and enzymes and for utilization of arabinose including phosphoketolase. The two putative extracellular arabinanases, AbnA and AbnB, are homologous to family GH43 endo-arabinanases. In Lactobacillaceae, homologs of these enzymes were identified exclusively in vertebrate-adapted species of the genus Lactobacillus. L. crispatus grew with arabinan from sugar beet pectin as sole carbon source, indicating extracellular arabinanase activity, and produced lactate and acetate, indicating metabolism via the phosphoketolase pathway. The two arabinanases AbnA and AbnB were heterologously expressed and purified by affinity chromatography. AbnA hydrolyzed linear and branched arabinan, while AbnB hydrolyzed only linear arabinan. The optimum pH for AbnA and AbnB was 6 and 7.5, respectively; 40 °C was the optimum temperature for both enzymes. The application of arabinan degrading L. crispatus as probiotic or as synbiotic with pectins may improve the production of short-chain fatty acids from pectin to benefit host health. In comparison to control, we have identified 867 and 9,053 differentially expressed genes in the hormone-treated female brain and ovary, respectively. In the brain, most of the identified genes were significantly enriched in 18 KEGG pathways. In the ovarian tissue, the identified genes were significantly involved in 9 pathways. In the hormone-treated carp, genes were involved in calcium signalling pathway, cAMP signalling pathway, insulin secretion, and oxidative phosphorylation pathway, which showed obvious associations with ovarian maturation. Indeed, compared with the other selected Countries (France, Germany, UK, and Spain), the Italian infection cases resulted to be lower, in the same analysed period. This work is devoted to find a possible justification of the unexpected situation found in Italy. A comparison of the imposed restrictions in the considered Countries allows to highlight that some policies result more effective to limice this pandemic period.Parabens are antimicrobial preservatives used in a wide range of consumer products such as personal care products, cosmetics, pharmaceuticals, and food. Consequently, the general population is ubiquitously exposed to these substances via dermal absorption, ingestion, and inhalation. Parabens promote estrogenic activity and are hence under assessment as endocrine disrupting substances. Urine samples from 3- to 17-year-old children and adolescents (N = 516) living in Germany were analysed for concentrations of nine parabens in the population representative German Environmental Survey for Children and Adolescents 2014-2017 (GerES V). Detection rates and urinary concentrations of the parabens decreased with increasing length of the alkyl chain. Methyl paraben was quantified in 97% of the samples with a geometric mean (GM) concentration of 7.724 μg/L (6.714 μg/gcreatinine), ethyl paraben was quantified in 69% (GM 0.943 μg/L and 0.825 μg/gcrea), and n-propyl paraben in 31% (GM 0.563 μg/L and 0.493 μg/gcrea). Concentrations of iso-propyl paraben, butyl paraben, iso-butyl paraben, and benzyl paraben were below the limit of quantification in most samples. Pentyl paraben and heptyl paraben were not detected in any of the samples. Paraben concentrations in urine were found to be associated with frequent usage of leave-on personal care products and cosmetics. Students (n = 357) were surveyed for asthma symptoms and measured for fractional exhaled nitric oxide (FeNO). Asthma was not associated with the overall bacterial richness but associated with specific phylogenetic classes. Taxa richness and abundance in Clostridia, including Ruminococcus, Blautia, Clostridium and Subdoligranulum, were positively associated with asthma (p less then 0.05), and these taxa were mainly derived from the human gut. Taxa richness in Alphaproteobacteria and Actinobacteria were marginally protectively associated with asthma, and these taxa were mainly derived from the outdoor environment. Bacterial richness and abundance were not associated with FeNO levels. Building age was associated with overall bacterial community variation in air and floor dust (p less then 0.05), but not associated with the asthma-related microorganisms. Our data shows that taxa from different phylogenetic classes and derived habitats have different health effects, indicating the importance of incorporating phylogenetic and ecological concepts in revealing patterns in the microbiome asthma association analysis.

A pot experiment was conducted to study the impact of low- and high-dose zinc amendments on plant growth and biomass yield, with Calcic Chernozem as a growing medium and barley (Hordeum vulgare L.) as a model plant. The distribution of zinc in various plant parts was also investigated. Zn (II) was added in powder as bulk ZnO and in solution as ZnO nanoparticles and ZnSO4 in two dosages (3 and 30 mmol kg-1 soil) prior to planting. The plants were harvested after 10 days of growth. The three sets of data were taken under identical experimental conditions. The application of zinc in aqueous solution and in particulate form (having particle sizes in the range of 500 nm) at concentration of 3 and 30 mmol Zn kg-1 to the soil resulted in decreased growth (root length, shoot length) and biomass yield; the only exception was the addition of 30 mmol Zn kg-1 in the form of bulk ZnO, which had a positive effect on the root growth. The dry weight reduction (sprout biomass) was lowest in plants grown in soil treated with dissolved zinc. There were no statistically significant changes in the content of chlorophyll a, chlorophyll b, and total chlorophyll, although flame atomic absorption spectrometry (FAAS) analysis indicated that plants bioaccumulated the zinc applied. This implies that the transport of zinc into the above-ground plant parts is controlled by the presence of effective mechanical and physiological barriers in roots. Crop performance under zinc stress in relation to biomass production and the growth of roots and shoots is also partly a reflection of the effects of soil properties. Eight residues were important for ligand binding. Mutations of three residues impaired Gs-cAMP signaling without changing the binding properties. Investigation on constitutive activities of all the mutants in the cAMP pathway revealed that six residues were involved in constraining the receptor in inactive states and five residues were important for receptor activation in the absence of an agonist.

Eight residues were important for ligand binding. Mutations of three residues impaired Gs-cAMP signaling without changing the binding properties.
