  • Many studies have verified the safety of combined radiotherapy and immune checkpoint blockades (ICBs) without the specific radiation dose or sequencing of combination. We aimed to evaluate the expression and response of PD-1, TIM-3, LAG-3 after neoadjuvant radiotherapy (NRT) and explore the possibility and optimal schedule of combining immunotherapy with radiotherapy in treating rectal cancer. However, TIM-3 in ICs and tumor cells (TCs) were both decreased (80% vs. 50%, P = 0.011, 90% vs. 0%, P = 0.000, respectively). The LAG-3 expression was higher in patients treated with short-course RT than long-course RT (22.5% vs. 8.0%, P = 0.0440 in ICs; 0% vs. 70%, P eight weeks after long-course RT (P = 0.045). The expressions of PD-1, ICs TIM-3, ICs LAG-3, CD3, and CD8 were associated with the disease-free survival (DFS) in univariate analysis (P = 0.036, 0.008, 0.018, 0.025, and 0.004, respectively). Adjusted by the relevant variables, PD-1 (HR 0.274; 95% CI 0.089-0.840; P = 0.024) and ICs TIM-3 (HR 0.425; 95% CI 0.203-0.890; P = 0.023) were independent prognostic factors of DFS in rectal cancer patients following NRT. In conclusion, we have identified that PD-1 and ICs LAG-3 presented a trend towards increased expression after NRT, supporting the ICBs and NRT combination as a potential treatment option for local advanced rectal cancer patients. Initially, the expression of miR-204 in human NSCLC tissues and cells was determined by RT-qPCR, which demonstrated that miR-204 was downregulated in NSCLC tissues and cells. Next, Krüppel-like factor 7 (KLF7) was predicted and validated to be a target of miR-204 using dual-luciferase reporter gene assay. NSCLC A549 cells were treated with ****-derived exosomes, after which the migration and invasion of A549 cells were detected and expression of EMT-related proteins (E-cadherin, N-cadherin, and Vimentin), KLF7, p-AKT/AKT, and HIF-1α were measured. The data was subjected to logistic regression analyses and revealed that the subjects carrying AA genotype of rs3787016 in POLR2E had a significant 1.85-1.98-fold increased risk of GC when compared with those carrying GG genotype (adjusted OR=1.979, 95% CI=1.198-3.267; p=0.008) or those carrying AG/GG genotypes (adjusted OR=1.847, 95% CI=1.222-2.793; p=0.004). For the GC patients, the AA genotype of rs3787016 was significantly correlated with poorly differentiated GC (p=0.018), advanced TNM stage (p=0.023), higher depth of invasion (p=0.022), positive lymph node metastasis (p=0.01), and worse overall survival (OS; p=0.004). Multivariate analysis confirmed that the POLR2E rs3787016 polymorphism is an independent prognostic factor for GC (HR=1.668, 95% CI=1.058-2.631; p=0.028). Our cumulative results thus suggest that the presence of POLR2E rs3787016 polymorphism could serve as a genetic factor that affects the susceptibility to and the prognosis of GC.Protein phosphatase 1H (PPM1H) is the metal-dependent protein phosphatase, however, its role in tumorigenesis and tumor progression remains controversial. Non-small cell lung cancer (NSCLC) is the most common histological type of lung cancer but the expression and clinical significance of PPM1H in NSCLC is unknown. In our study, we detected the mRNA of PPM1H in 25 pairs of NSCLC tissues and their corresponding adjacent tissues with qRT-PCR. Moreover, we investigated PPM1H expression in 474 NSCLC tissues and divided them into subgroups with low and high PPM1H. We further evaluated its correlation with the clinicopathological factors. The correlation between PPM1H and other biomarkers involved in tumor progression including chromosome segregation 1-like protein (CSE1L), p53, and Ki67 was also estimated. In addition, the prognostic significance of PPM1H was investigated by univariate and multivariate analyses. The mRNA levels of PPM1H in NSCLCs were significantly higher than those in tumor-adjacent tissues. Patients with low and high PPM1H expression accounted for 54.64% (259/474) and 45.36% (215/474) respectively in all the NSCLCs. PPM1H expression (P = 0.012), patients' sex (P = 0.009), tumor size (P less then 0.001), histological grade (P = 0.026), T stage (P = 0.002), N stage (P less then 0.001), M stage (P = 0.011), and TNM stage (P less then 0.001) were all associated with the poor prognosis. With multivariate analysis, PPM1H was determined as an independent prognostic factor of NSCLC (HR = 1.42, 95% CI = 1.14-1.75, P = 0.001). Moreover, high PPM1H was significantly with high Ki67 (P = 0.022), indicating the oncogenic role of PPM1H. PPM1H is an independent prognostic factor indicating an unfavorable prognosis of NSCLC. Our results indicated that PPM1H was an important supplement of NSCLC molecular profile and detecting PPM1H may help recognize the high-risk patients for further treatment.

  • Physical activity is known to be beneficial for bone; however, some athletes who train intensely are at risk of bone stress injury (BSI). Incidence in adolescent athlete populations is between 3.9 and 19% with recurrence rates as high as 21%. Participation in physical training can be highly skeletally demanding, particularly during periods of rapid growth in adolescence, and when competition and training demands are heaviest. Sports involving running and jumping are associated with a higher incidence of BSI and some athletes appear to be more susceptible than others. Maintaining a very lean physique in aesthetic sports (gymnastics, figure skating and ballet) or a prolonged negative energy balance in extreme endurance events (long distance running and triathlon) may compound the risk of BSI with repetitive mechanical loading of bone, due to the additional negative effects of hormonal disturbances. The following review presents a summary of the epidemiology of BSI in the adolescent athlete, risk factors for BSI (physical and behavioural characteristics, energy balance and hormone disruption, growth velocity, sport-specific risk, training load, etc.), prevention and management strategies.The robustness of precast reinforced concrete (RC) frames is relatively poor, while the precast RC frames are strengthened to mitigate progressive collapse, avoiding "strong beams and weak columns" and the anchorage failure of strengthening materials under large deformation condition are the key problems. Aiming to discuss these problems, this paper carried out an experimental research of strengthening on three half-scale assembled monolithic frame subassemblages to mitigate progressive collapse. One specimen was strengthened by implanting carbon fiber rope (CFR) with polymer into concrete, one specimen was strengthened by binding CFR with special knot, and the last one was not strengthened. The failure mode, collapse failure mechanism and strengthening effect of subassemblages were discussed. Analytical models of load capacity increment contributed by CFR and construction suggestions of precast RC frame to mitigate progressive collapse were proposed. The results indicated that none of the strengthened specimens had anchorage failure. The two strengthening methods significantly increased the load capacity of the subassemblages in the catenary action (CA) stage with little effect on the flexural action (FA) stage and compressive arch action (CAA) stage.A laser-irradiated surface is the paradigm of a self-organizing system, as coherent, aligned, chaotic, and complex patterns emerge at the microscale and even the nanoscale. A spectacular manifestation of dissipative structures consists of different types of randomly and periodically distributed nanostructures that arise from a homogeneous metal surface. The spectroscopy shows strong spatial variations in the density of states (DOS), and the superconducting map constructed from spectroscopy discloses a localized superconducting phase, as small as a single unit cell. The comparison of the spectra taken at 4.2 K and 22 K (below vs. close to the bulk superconducting transition temperature) from the exact same area confirms the superconducting behavior. Nanoscale superconducting states have been found near Pr dopants, which can be identified using dI/dV conductance maps at +300 mV. There is no correlation of the local superconductivity to the surface reconstruction domain and surface defects, which reflects its intrinsic bulk behavior. We, therefore, suggest that the local strain of Pr dopants is competing with defects induced local magnetic moments; this competition is responsible for the local superconducting states observed in this Fe-based filamentary superconductor.To use implantable biomedical devices such as electrocardiograms and neurostimulators in the human body, it is necessary to package them with biocompatible materials that protect the internal electronic circuits from the body's internal electrolytes and moisture without causing foreign body reactions. Herein, we describe a hydrogel surface-modified polyurethane copolymer film with concurrent water permeation resistance and biocompatibility properties for application to an implantable biomedical device. To achieve this, hydrophobic polyurethane copolymers comprising hydrogenated poly(ethylene-co-butylene) (HPEB) and aliphatic poly(carbonate) (PC) were synthesized and their hydrophobicity degree and mechanical properties were adjusted by controlling the copolymer composition ratio. When 10 wt% PC was introduced, the polyurethane copolymer exhibited hydrophobicity and water permeation resistance similar to those of HPEB; however, with improved mechanical properties. Subsequently, a hydrophilic poly(vinyl pyrrolidone) (PVP) hydrogel layer was formed on the surface of the polyurethane copolymer film by Fenton reaction using an initiator and crosslinking agent and the effect of the initiator and crosslinking agent immobilization time, PVP concentration and crosslinking agent concentration on the hydrogel properties were investigated. Finally, MTT assay showed that the hydrogel surface-modified polyurethane copolymer film displays excellent biocompatibility.

High mobility group box 1 (HMGB1) is a highly conserved nuclear protein with a nuclear and extracellular role that functions as an extracellular signaling molecule under specific conditions. Several studies have shown that HMGB1 is a critical regulator of autophagy. Here, we describe a novel emission ratiometric flow cytometry method based on F66, a 3-hydroxiflavon derivative, and demonstrate that 6-ketocholestanol, cholesterol and 7-dehydrocholesterol, saturated stearic acid (SA) and ω-6 γ-linolenic acid (GLA) increase, while ω-3 α-linolenic acid (ALA) decreases the DP. These changes do not correlate with alterations in cell viability or membrane fluidity. Pretreatment with ALA counteracts, while SA or GLA enhances cholesterol-induced DP elevations. Furthermore, ALA (but not SA or GLA) increases endo-lysosomal escape of penetratin, a cell-penetrating peptide.     To evaluate the long term visual, refractive, and corneal tomographic outcomes of epithelium-off accelerated corneal collagen cross-linking (ACXL) in the management of pediatric keratoconus (KC).

    This retrospective study included patients under 18 years old with progressive KC who underwent ACXL between 2012 and 2019 at Dicle University Hospital. Complete ophthalmic examination was performed including uncorrected distance visual acuity (UDVA), best spectacle-corrected distance visual acuity (CDVA), manifest refraction, and corneal tomography. Evaluations were performed preoperatively and at 6 months intervals postoperatively.

    Forty-nine eyes of 49 patients were included in the study. The mean age of patients at the time of ACXL was 14.2 ± 1.8 (range 9.5-17.3) years. Mean follow up was 4.61 ± 1.90 (range 2.0-8.1) years. The mean LogMAR UDVA improved from 0.94 ± 0.41 to 0.81 ± 0.43, 0.69 ± 0.41, and 0.67 ± 0.33 after 1, 3, and 5 years respectively (
     = 0.001). The mean LogMAR CDVA improved from 0.58 ± 0.36 to 0.46 ± 0.31, 0.34 ± 0.23, and 0.39 ± 0.27 after 1, 3, and 5 years respectively (

    A 84-year old woman with worsening of her herpes simplex keratitis, diagnosed, and treated 2 days before. The slit lamp showed deep paracentral infiltrate and hypopyon. A corneal sample was collected for culture prior to initiation of empiric antibiotic therapy with vancomycin and ceftazidime fortified, oral acyclovir, and cyclopentolate. The strain was identified as
    and topical antibiotic therapy was adjusted to ciprofloxacin and ceftazidime. After 2 weeks, the epithelial defect and the infiltrate were resolved and prednisolone was added to the regimen. As the corneal oedema and neovascularization decreased, acyclovir, and prednisolone were slowly tapered. About 4 months later, the visual outcome was 20/50 and the ophthalmic examination showed a clear cornea with a paracentral leucoma.

    The Supported Employment Demonstration (SED), a multiyear (2016-2022), randomized controlled trial funded by the Social Security Administration, recruited a nontraditional sample of benefits applicants with self-reported or documented mental health conditions who were denied disability benefits and who expressed a desire for employment. This study describes the characteristics of the SED sample at baseline.

    The authors analyzed baseline data from the 2,960 eligible enrollees, including responses to the Composite International Diagnostic Interview, the 12-item Short-Form Health Survey (SF-12), and the Work Disability Functional Assessment Battery (WD-FAB).

    A majority of SED enrollees self-identified as female (57%), White (56%), and non-Hispanic (87%). Many were 35 years or older (58%), reported at least a high school education (81%), lived with relatives (69%), had never married (55%), were unemployed (81%), and were poor. Median monthly household income was $1,200. Anxiety disorders (71%), personality disorders (65%), and mood disorders (61%) were prevalent. Enrollees reported a mean±SD of 2.5±1.3 mental health conditions and 3.5±2.1 general medical conditions. Health-related quality of life was low, relative to national norms mean scores for the sample were 32.6±12.5 on the SF-12 mental component summary and 38.3±13.0 on the physical component summary. Mean scores on the WD-FAB subdomains were more than a SD below norms.

  • Compared with general anesthesia, monitored anesthesia led to less adverse anesthetic effects.     We aimed to assess sleep quality of Tunisian medical students during home confinement due to the COVID-19 pandemic, and to analyze the relationship between sleep quality and sociodemographic, clinical, confinement-related and psychological variables.

    A correlational cross-sectional study was conducted from April 11th to May 3rd 2020. Medical students who have been in home confinement and who accepted to participate in an online survey were targeted. Sociodemographic data, clinical variables, and data related to home confinement were collected. Participants also completed Pittsburgh Sleep Quality Index, Depression, Anxiety and Stress Scale and **** Hopelessness Scale.

    Results showed a high prevalence of poor sleepers among medical students (72.5%) with poor subjective sleep quality, increased sleep latency, sleep disturbances and daytime dysfunction. Multiple regression analysis revealed that family history of suicide attempts, tobacco use, perception of home confinement and reduced physical activity dursleep parameters need to be assessed in this particular population and adequate measures aiming to promote quality of sleep need to be enhanced, given the crucial regenerative, homeostatic and psychological roles of sleep.

Hence, a novel feature of DES in MOF synthesis is uncovered its role as a structure-directing agent, triggering the transformation between two different MOF structures.By using extensive all-atom molecular dynamics simulations of an atactic linear polymer chain, we provide microscopic insights into poly(N-isopropylacrylamide) (PNIPAM) coil-to-globule transition addressing the roles played by both temperature and pressure. In this study, we systemically investigated the formation process of VO conductive filaments in β-Ga2O3 memory. There were at least three kinds of VOs and conductive filaments with different low resistance states (LRSs) in β-Ga2O3 memory, suggesting their potential for multilevel storage application. Interestingly, these conductive filaments preferred to be formed along the [010] direction and with a single VO cluster rather than a mixed VO cluster due to the lower single VO cluster formation energy and ellipsoid charge distribution. The lowest migration and activation barriers for different kinds of VOs in +2 charge states (V2+Os) were discrepant and lower than the neutral charge states. Meanwhile, the forward migration energy of VO was different from the reversed migration path, so that the conductive filament formation and rupture were not an inverse process in the experiment. The detailed mechanisms were revealed by the charge density and migration process of these VOs. These results not only revealed the function of the VO conductive filaments in β-Ga2O3 memory but also predicted the potential of β-Ga2O3 memory for multilevel storage application.

It was unequivocally demonstrated that, upon heating in reline, HKUST-1 converts to the non-porous chloride-incorporating material. Hence, a novel feature of DES in MOF synthesis is uncovered its role as a structure-directing agent, triggering the transformation between two different MOF structures.By using extensive all-atom molecular dynamics simulations of an atactic linear polymer chain, we provide microscopic insights into poly(N-isopropylacrylamide) (PNIPAM) coil-to-globule transition addressing the roles played by both temperature and pressure. We detect a coil-to-globule transition up to large pressures, showing a reentrant behavior of the critical temperature with increasing pressure in agreement with experimental observations. Furthermore, again confirming the experimental findings, we report the existence at high pressures of a new kind of globular state. It is characterized by a more structured hydration shell that is closer to PNIPAM hydrophobic domains, as compared to the globular state observed at atmospheric pressure. In this study, we systemically investigated the formation process of VO conductive filaments in β-Ga2O3 memory. There were at least three kinds of VOs and conductive filaments with different low resistance states (LRSs) in β-Ga2O3 memory, suggesting their potential for multilevel storage application. Interestingly, these conductive filaments preferred to be formed along the [010] direction and with a single VO cluster rather than a mixed VO cluster due to the lower single VO cluster formation energy and ellipsoid charge distribution. The lowest migration and activation barriers for different kinds of VOs in +2 charge states (V2+Os) were discrepant and lower than the neutral charge states. Meanwhile, the forward migration energy of VO was different from the reversed migration path, so that the conductive filament formation and rupture were not an inverse process in the experiment. The detailed mechanisms were revealed by the charge density and migration process of these VOs. These results not only revealed the function of the VO conductive filaments in β-Ga2O3 memory but also predicted the potential of β-Ga2O3 memory for multilevel storage application.

While many species rely on behavioural thermoregulation to avoid thermal extremes, others build structures (nests) that confer a shelter against climate variability and extremes. However, the microclimate inside nests remains unknown for most insects. We investigated the thermal environment inside the nest of a temperate winter-developing insect species, the pine processionary moth (PPM), Thaumetopoea pityocampa. https://www.selleckchem.com/products/lw-6.html Gregarious larvae collectively build a silken nest at the beginning of the cold season. We tested the hypothesis that it provides a warmer microenvironment to larvae. First, we monitored temperature inside different types of nests varying in the number of larvae inside. At noon, when global radiation was maximal, nest temperature exceeded air temperature by up to 11.2-16.5 °C depending on nest type. Second, we developed a biophysical model to predict temperature inside PPM nests based on heat transfer equations and to explain this important temperature excess. We conclude that some pest insects already live in warm microclimates by building their own sheltering nest. Considering that temperature has a strong influence on insect voltinism (i.e., number of generations per year), climate change may affect the population growth of insects, with potential consequences for food production. The southern armyworm, Spodoptera eridania, is a multivoltine species native to the American tropics that causes severe damage to several crops. In this context, this study evaluated the impacts of climate change on the voltinism of S. eridania in southern Brazil. Current and future daily temperature data were combined with non-linear and degree-day models to estimate the voltinism of this pest. Under current climate conditions, the voltinism of S. eridania ranged from 2.9 to 9.2 generations, with fewer cohorts in colder regions and more in warmer ones. A higher number of generations was predicted for the future climate scenarios evaluated, reaching up to 12.1 annual generations in certain regions by 2070. Most of the variation in voltinism was explained by location (87.7%) and by the interaction between location and mathematical model (3.0%). The degree-day model estimated an increase in the number of generations in the entire study area, while the non-linear model predicted a decrease in voltinism in the warmer regions under future climate change scenarios. Given these differences between the predictions provided by degree-day and non-linear models, the selection of the best method to be used in climate change studies should be carried out carefully, considering how species respond to temperature. A considerable increase in the number of generations of S. eridania was projected for most of the study area under the climate change scenarios evaluated, suggesting a possible rise in pest incidence levels in the coming decades.High ambient temperature has potential influence on oxidative stress, or systemic inflammation affecting poultry production and immune status of chickens. Therefore, we designed an experiment to analyze the effects of 35 ± 2 °C HS on salmonella infection in chickens through regulation of the immune responses. 40 broiler chickens were randomly divided into 4 groups control group, heat stress (HS) group, salmonella typhimurium (ST) group and model group (heat stress + salmonella typhimurium, HS + ST). Birds in HS and model group were treated with 35 ± 2 °C heat stress 6 h a day and for 14 continuous days. Then, ST and model group birds were orally administrated with 1 mL ST inoculum (109 cfu/mL). Chickens were sacrificed at the 4th day after ST administration and ileum tissues were measured. We observed that heat stress decreased ileum TNF-α and IL-1β protein expressions. Concomitantly heat stress decreased NLRP3 and Caspase-1 protein levels. The protein expressions of p-NF-κB-p65 and p-IκB-α in ileum. Heat stress also inhibited IFN-α, p-IRF3 and p-TBK1, showing a deficiency in the HS + ST group birds. Together, the present data suggested that heat stress suppressed intestinal immune activity in chickens infected by salmonella typhimurium, as observed by the decrease of immune cytokines levels, which regulated by NF-κB-NLRP3 signaling pathway.The present study was aimed to assess the effect of temperatures on egg incubation, growth, standard metabolic rate (SMR), and thermal tolerance of a near threatened Himalayan hill stream chocolate mahseer (Neolissochilus hexagonolepis). Experiment I (for validation of the CTmax method) was carried out by incubating eggs at 17 °C and 23 °C. The CTmax was estimated in response to different warming rates (1-18°C h-1), acclimation temperatures (17 and 23°C), and the age of fishes (8, 15, 35 dph). The results suggested that a warming rate of 18°C h-1 could be used for the thermal tolerance study of yolk-sac larvae (8 dph) and 35 dph larvae, but for free-swimming larvae (15 dph) up to 3°C h-1 is suitable. Experiment II (for growth, SMR and thermal tolerance) was carried by acclimatizing 15 dph larvae in five temperatures (15, 19, 23, 27, and 31 °C) for 60 days.

  • Over the last 200 years, conversion of non-cultivated land for agriculture has substantially reduced global soil organic carbon (SOC) stocks in upper soil layers. Nevertheless, practices such as no- or reduced tillage, application of organic soil amendments, and maintenance of continuous cover can increase SOC in agricultural fields. While these management practices have been well studied, the effects on SOC of cropping systems that incorporate irrigation are poorly understood. Given the large, and expanding, agricultural landbase under irrigation across the globe, this is a critical knowledge gap for climate change mitigation. We undertook a systematic literature review and subsequent meta-analysis of data from studies that examined changes in SOC on irrigated agricultural sites through time. We investigated changes in SOC by climate (aridity), soil texture, and irrigation method with the following objectives (i) to examine the impact of irrigated agriculture on SOC storage; and (ii) to identify the conditions under which irrigated agriculture is most likely to enhance SOC. Overall, irrigated agriculture increased SOC stocks by 5.9%, with little effect of study length (2-47 years). However, changes in SOC varied by climate and soil depth, with the greatest increase in SOC observed on irrigated semi-arid sites at the 0-10 cm depth (14.8%). Additionally, SOC increased in irrigated fine- and medium-textured soils but not coarse-textured soils. Furthermore, while there was no overall change to SOC in flood/furrow irrigated sites, SOC tended to increase in sprinkler irrigated sites, and decrease in drip irrigated sites, especially at depths below 10 cm. This work sheds light on the nuances of SOC change across irrigated agricultural systems, highlights the importance of studying SOC storage in deeper soils, and will help guide future research on the impacts of irrigated agriculture on SOC.Annual variation in phenology can have profound effects on the behavior of animals. We investigated the influence of spring phenology on the behavior of migratory, barren-ground caribou (Rangifer tarandus), a species that has evolved to cope with short Arctic summers. Specifically, we examined the effect of spring snow melt and vegetation growth on the current and potential future space-use patterns of the Porcupine Caribou Herd (PCH), which exhibits large, inter-annual shifts in their calving and post-calving distributions across the U.S.-Canadian border. We quantified PCH selection for snow melt and vegetation phenology using machine learning models, determined how selection resulted in annual shifts in space-use, and then projected future distributions based on climatmaintaining sufficient suitable habitat to allow for behavioral plasticity.Anthropogenic nitrogen (N) input is known to alter plant and microbial α-diversity, but how N enrichment influences β-diversity of plant and microbial communities remains poorly understood. Using a long-term multilevel N addition experiment in a temperate steppe, we show that plant, soil bacterial and fungal communities exhibited different responses in their β-diversity to N input. Plant β-diversity decreased linearly as N addition increased, as a result of increased directional environmental filtering, where soil environmental properties largely explained variation in plant β-diversity. Soil bacterial β-diversity first increased then decreased with increasing N input, which was best explained by corresponding changes in soil environmental heterogeneity. Soil fungal β-diversity, however, remained largely unchanged across the N gradient, with plant β-diversity, soil environmental properties, and heterogeneity together explaining an insignificant fraction of variation in fungal β-diversity, reflecting the importance of stochastic community assembly. Here, we characterized rhizosphere fungal communities of 53 native and invasive plants spanning approximately 1800 km in China, as well as eight pairs of phylogenetically related native versus invasive plants in a greenhouse experiment. The results of both field survey and greenhouse experiment showed that rhizosphere fungal composition was primarily predicted by plant phylogeny (e.g. family and species), and plant geographic origin (native vs. invasive) and abiotic factors had **** smaller effects. We detected no differences in the number and relative abundance of total and family/species-specific OTUs (i.e. overall, pathogens and mutualists) associated with these native and invasive plants on average, suggesting novel co-evolution between native soil fungi and these invasive plants.
