  • Aberrant activation of the WNT/β-catenin signaling pathway is implicated in various types of cancers. Inhibitors targeting the Wnt signaling pathway are intensively studied in the current cancer research field, the outcomes of which remain to be determined. In this study, we have attempted to discover novel potent WNT/β-catenin pathway inhibitors through tankyrase 1/2 structure-based virtual screening. After screening more than 13.4 million compounds through molecular docking, we experimentally verified one compound, LZZ-02, as the most potent inhibitor out of 11 structurally representative top hits. LiCl-induced HEK293 cells containing TOPFlash reporter showed that LZZ-02 inhibited the transcriptional activity of β-catenin with an IC50 of 10 ± 1.2 μM. Mechanistically, LZZ-02 degrades the expression of β-catenin by stabilizing axin 2, thereby diminishing downstream proteins levels, including c-****and cyclin D1. LZZ-02 also inhibits the growth of colonic carcinoma cell harboring constitutively active β-catenin. More importantly, LZZ-02 effectively shrinks tumor xenograft derived from colonic cell lines. Our study successfully identified a novel tankyrase 1/2 inhibitor and shed light on a novel strategy for developing inhibitors targeting the WNT/β-catenin signaling axis.Acinetobacter baumannii is one of the most common causes of nosocomial infections in intensive care units. Its ability to acquire diverse mechanisms of resistance limits the therapeutic choices for its treatment. This especially concerns colistin, which has been reused recently as a last-resort drug against A. baumannii. Here, we explored the impact of gaining colistin resistance on the susceptibility of A. baumannii to other antibiotics and linked colistin resistance acquisition to a gene mutation in A. baumannii. The susceptibility of 95 A. baumannii isolates revealed that 89 isolates were multi-drug resistance (MDR), and nine isolates were resistant to colistin. Subsequently, three isolates, i.e., MS48, MS50, and MS64, exhibited different resistance patterns when colistin resistance was induced and gained resistance to almost all tested antibiotics. Upon TEM examination, morphological alterations were reported for all induced isolates and a colistin-resistant clinical isolate (MS34Col-R) compared to the parental sensitive strains. Finally, genetic alterations in PmrB and LpxACD were assessed, and a point mutation in LpxD was identified in the MS64Col-R and MS34Col-R mutants, corresponding to Lys117Glu substitution in the lipid-binding domain. Our findings shed light on the implications of using colistin in the treatment of A. baumannii, especially at sub-minimum inhibitory concentrations concentrations, since cross-resistance to other classes of antibiotics may emerge, beside the rapid acquisition of resistance against colistin itself due to distinct genetic events.A correlation between autophagy and autolysis has been proposed in order to acceleratethe acquisition of wine organoleptic properties during sparkling wine elaboration. Diabetic retinopathy (DR) is the most serious complication of the eye caused by diabetes. The aim of this research was to assess the prevalence of diabetic retinopathy in type 1 and type 2 diabetes mellitus patients in north-east Poland. Materials and Methods The eye fundus was assessed on the basis of two-field 50 degrees color fundus photographs that showed the optic nerve and macula in the center after the pupil was dilated with 1% tropicamide. Results The experimental group included 315 (26%) patients with type 1 diabetes mellitus (DM1) and 894 (74%) patients with DM2. DM1 patients were diagnosed with DR in 32.58% of cases, with non-proliferative diabetic retinopathy (NPDR) in 24.44% of cases, proliferative diabetic retinopathy (PDR) in 1.59% of cases, diabetic macular edema (DME) in 5.40% of cases, and PDR with DME in 0.95% of cases. DR was found in DM2 patients in 23.04% of cases, NPDR in 17.11% of cases, PDR in 1.01% of cases, DME in 4.81% of cases, and PDR with DME in 0.11% of cases. Conclusions The presented study is the first Polish study on the prevalence of diabetic retinopathy presenting a large group of patients, and its results could be extrapolated to the whole country. Diabetic retinopathy was found in 25.48% of patients in the whole experimental group. The above results place Poland within the European average, indicating the quality of diabetic care offered in Poland, based on the number of observed complications.BACKGROUND In patients with neuromuscular disorder, only little data of myalgia frequency and characterization exists. To date, only a weak correlation between pain intensity and pressure pain threshold has been found, and it remains enigmatic whether high pain intensity levels are equivalent to high pain sensitivity levels in neuromuscular disorders. METHODS 30 sequential patients with suspected neuromuscular disorder and myalgia were analyzed with regard to myalgia characteristics and clinical findings, including symptoms of depression and anxiety and pain- threshold. RESULTS A neuromuscular disorder was diagnosed in 14/30 patients. Muscular pain fasciculation syndrome (MPFS) without evidence for myopathy or myositis was diagnosed in 10/30 patients and 6/30 patients were diagnosed with pure myalgia without evidence for a neuromuscular disorder (e.g., myopathy, myositis, MPFS, polymyalgia rheumatica). Highest median pain scores were found in patients with pure myalgia and polymyalgia rheumatica. Pressure pain threshold measurement showed a significant difference between patients and controls in the biceps brachii muscle.

The analysis of our results showed that this plant oil rich in terpenoids and phenolic antioxidants could be a quite good in vitro additive with high potential for the world of medically assisted reproduction.Simultaneous testing of multiple genetic variants for association is widely recognized as a valuable complementary approach to single-marker tests. As such, principal component regression (PCR) has been found to have competitive power. We focus on exploring a robust test for an unknown genetic mode of all SNPs, an unknown Hardy-Weinberg equilibrium (HWE) in a population, and a large number of all SNPs. First, we propose a new global test by means of the use of codominant codes for all markers and PCR. The new global test is built on an empirical Bayes-type score statistic for testing marginal associations with each single marker. The new global test gains power by robustly exploiting the Hardy-Weinberg equilibrium in the control population and effectively using linkage disequilibrium among test markers. The new global test reduces to PCR when the genotype for each marker is coded as the number of minor alleles. This connection lends insight into the power of the new global test relative to PCR and some other popular multimarker test methods. Second, we propose a robust test method based on the new global test and the ordinary PCR test built on a prospective score statistic for testing marginal associations with each single marker when the genotype for each marker is coded as the number of minor alleles by taking the minimum p value of these two tests. Finally, through extensive simulation studies and analysis of the association between pancreatic cancer and some genes of interest, we show that the proposed robust test method has desirable power and can often identify association signals that may be missed by existing methods.This study identified 57 basic leucine zipper (bZIP) genes from the pineapple genome, and the analysis of these bZIP genes was focused on the evolution and divergence after multiple duplication events in relation to the pineapple genome fusion. According to bioinformatics analysis of a phylogenetic tree, the bZIP gene family was divided into 11 subgroups in pineapple, Arabidopsis, and rice; gene structure and conserved motif analyses showed that bZIP genes within the same subgroup shared similar intron-exon organizations and motif composition. Further synteny analysis showed 17 segmental duplication events with 27 bZIP genes. The study also analyzed the pineapple gene expression of bZIP genes in different tissues, organs, and developmental stages, as well as in abiotic stress responses. The RNA-sequencing data showed that AcobZIP57 was upregulated in all tissues, including vegetative and reproductive tissues. AcobZIP28 and AcobZIP43 together with the other 25 bZIP genes did not show high expression levels in any tissue. Six bZIP genes were exposed to abiotic stress, and the relative expression levels were detected by quantitative real-time PCR. A significant response was observed for AcobZIP24 against all kinds of abiotic stresses at 24 and 48 h in pineapple root tissues. Our study provides a perspective for the evolutionary history and general biological involvement of the bZIP gene family of pineapple, which laid the foundation for future functional characterization of the bZIP genes in pineapple.Background Sepsis is a life-threatening condition and a systemic inflammatory response syndrome (SIRS) driven by infection. Methods Expression of miR-103 was measured using quantitative real-time PCR. A receiver operating characteristics curve was plotted to evaluate the diagnostic vale of miR-103. Serum and cell supernatant levels of proinflammatory cytokines were analyzed using ELISA. The interaction between miR-103 and Toll-like receptors 4 (TLR4) was analyzed using luciferase reporter assay. The effect of miR-103 on inflammation was examined in LPS-treated monocytes. Results Serum expression of miR-103 was decreased in noninfectious SIRS and sepsis patients compared with healthy controls, and the lowest expression value was observed in sepsis patients (all P less then 0.05). Serum levels of miR-103 have considerable diagnostic accuracy in distinguishing sepsis patients from SIRS patients and healthy controls. A negative correlation was found between miR-103 and inflammatory responses in sepsis patients. TLR4 was demonstrated to be a direct target of miR-103 and was negatively regulated by miR-103 in monocytes. The promoted inflammatory responses by LPS in monocytes were reversed by the overexpression of miR-103. This paper presents a detailed methodology followed in the development of a stochastic groundwater model under uncertain recharge. The methodology is comprised of two steps (1) The development of a groundwater flow model and (2) the development of a stochastic solution- stochastic groundwater flow model considering recharge as uncertain parameter.•The groundwater flow model was developed and simulated using MODFLOW 2000 software and calibrated using Parameter Estimation Program (PEST). Validated model compared well with calibrated model.

This may be achieved through reductions in the allocation of energy for various purposes (e.g. thermoregulation, locomotion, etc.). We studied whether such trade-offs also include suppression of the innate immune response, by subjecting captive male Svalbard ptarmigan (Lagopus muta hyperborea) to bacterial lipopolysaccharide (LPS) during exposure to either mild temperature (0°C) or cold snaps (acute exposure to -20°C), in constant winter darkness when birds were in energy-conserving mode, and in constant daylight in spring. The innate immune response was mostly unaffected by temperature. However, energy expenditure was below baseline when birds were immune challenged in winter, but significantly above baseline in spring. This suggests that the energetic component of the innate immune response was reduced in winter, possibly contributing to energy conservation. Immunological parameters decreased (agglutination, lysis, bacteriostatic capacity) or did not change (haptoglobin/PIT54) after the challenge, and behavioural modifications (anorexia, mass loss) were lengthy (9 days). While we did not study the mechanisms explaining these weak, or slow, responses, it is tempting to speculate they may reflect the consequences of having evolved in an environment where pathogen transmission rate is presumably low for most of the year. C57BL/6J **** in obese (body mass gain >45%), lean (body mass gain less then 15%), and control (body mass gain less then 1%) groups were exposed to high (23±1°C) or low (4±1°C) ambient temperature for 28 days. Our data showed that low temperature exposure attenuated DIO, but enhanced brown adipose tissue (BAT) thermogenesis. Low temperature exposure also resulted in increased norepinephrine (NE) concentrations in the hypothalamus, decreased TRP melastatin 8 (TRPM8) expression in the small intestine, and altered composition and diversity of gut microbiota.

We studied whether such trade-offs also include suppression of the innate immune response, by subjecting captive male Svalbard ptarmigan (Lagopus muta hyperborea) to bacterial lipopolysaccharide (LPS) during exposure to either mild temperature (0°C) or cold snaps (acute exposure to -20°C), in constant winter darkness when birds were in energy-conserving mode, and in constant daylight in spring. The innate immune response was mostly unaffected by temperature. However, energy expenditure was below baseline when birds were immune challenged in winter, but significantly above baseline in spring. This suggests that the energetic component of the innate immune response was reduced in winter, possibly contributing to energy conservation. Immunological parameters decreased (agglutination, lysis, bacteriostatic capacity) or did not change (haptoglobin/PIT54) after the challenge, and behavioural modifications (anorexia, mass loss) were lengthy (9 days). While we did not study the mechanisms explaining these weak, or slow, responses, it is tempting to speculate they may reflect the consequences of having evolved in an environment where pathogen transmission rate is presumably low for most of the year. In both cases the effect of α-tocopherol appears similar. Spectral moments reveal that the main chain melting transition of POPE-d31 and PDPE-d31 is broadened beyond detection. A spectral component attributed to the formation of inverted hexagonal HII phase in coexistence with lamellar Lα phase by POPE-d31 (20 %) and PDPE-d31 (18 %) is resolved following the addition of α-toc. Order parameters in the remaining Lα phase are increased slightly more for POPE-d31 (7%) than PDPE-d31 (4%). Preferential interaction with polyunsaturated phospholipid is not apparent in these results. The propensity for α-toc to form phase structure with negative curvature that is more tightly packed at the membrane surface, nevertheless, may restrict the contact of free radicals with lipid chains on phosphatidylethanolamine molecules that accumulate polyunsaturated fatty acids.Endothelial progenitor cells (EPCs) are important to tissue repair and regeneration especially after ischemic injury, and very heterogeneous in phenotypes and biological features. Limb blood flow, function, muscle structure, and capillary density were evaluated with laser Doppler perfusion imaging, treadmill test, and immunohistochemistry, respectively, at day 3, 7, 14 and 21 post ischemia. Reactive oxygen species production in circulating and BM mononuclear cells and EPCs populations were significantly increased in BM and blood in **** with acute and chronic ischemia. We present a population dynamical model of a three-species system involving antagonism between species sharing a mutualist partner species with two explicit life stages. We find that, regardless of whether the antagonism is predatory or non-consumptive, persistence of the shared mutualist is possible only under a restrictive set of conditions. As the rate of antagonism between the species sharing the mutualist increases, indirect rather than direct interactions increasingly determine species' densities and sometimes result in complex, oscillatory dynamics for all species. Surprisingly, persistence of the mutualistic system is particularly dependent upon the degree to which each of the two mutualistic interactions is specialized. Primary outcome was the rate of complete muscularis mucosae (MM) resection, defined by the proportion of MM under the tumor more than 80% of the tumor's horizontal dimension. Secondary outcomes were the rates of negative lateral and vertical margins, fragmentation of resected specimens, conversion to hot-snare mucosal resection, intraprocedural bleeding, delayed bleeding, and perforation.

  • There is growing interest in the mechanisms for natural sensory learning in pro-social contexts. Studies using a maternal model of social behavior in the mouse have provided new insight into the auditory processing of behaviorally relevant pup vocalizations, which are used as communication signals to elicit pup retrieval behavior by adult females. Whether neural and behavioral plasticity in response to these vocalizations reflect auditory associative learning linking the sounds to pups, versus simply a change in maternal responsiveness to evolved vocal signals, remains an open question. Here, we describe a T-maze paradigm to track auditory learning as we pair an initially neutral, non-ethological stimulus with delivery of a pup for retrieval, which is intrinsically reinforcing for rodents. Training is rapid and completely appetitive. Over a period of 7 × 50-minute daily training sessions, animals increasingly use the sound to guide their arm choice for pup retrieval, with an increase in performance from chance to an average of ~80% on day 7. This pairing method establishes a newly-formed sensory association using a natural maternal behavioral response, and lays a solid foundation for studies into the neurochemical and circuit mechanisms that mediate auditory associative learning in natural social contexts.Background Infection may be an acute precipitant of cardiovascular events. However, the relationships between different types of infection and cardiovascular events are less known. Our objective was to determine if exposure to infections of different organ systems in different time periods increases risk of myocardial infarction or venous thromboembolism. Method We used case-crossover analysis with conditional logistic regression to estimate odds ratios (OR) for the association for each infection type during 3 case periods (30, 60, and 90 days prior to index event) compared with control periods (exactly 1 year before). Results This study had a total number of index admissions of 338,021 individuals, of which 82,986 were female; the mean age for individuals with myocardial infarction and venous thromboembolism was 68.48 years and 62.33 years, respectively. With every infection type, there was an increased likelihood of venous thromboembolism. The greatest association was for skin infections, with an OR of 5.39 (95% confidence interval, 4.08- 7.12) within the 7-day window. In a total of 35 fish examined, five (14.3%) had myxozoan plasmodia. Taxonomic analysis was performed integrating multiple characters, including morphometrical, biological traits, ssrDNA sequence data and host ecological characters. Molecular phylogenetic analysis revealed that the obtained ssrDNA sequence did not match any existing sequences in GenBank but showed M. iquitoensis n. sp. to be a close species of M. figueirae. Nonetheless, the ssrDNA sequences of those species show large genetic divergence. This is the first description and phylogenetic study of a myxozoan parasitizing fish of the genus Otocinclus from South America, as well the first report of these parasites infecting a fish belonging to the Loricariidae family from Amazon basin. Considering the endangered status of the host, the high degree of host-specificity of freshwater histozoic myxobolids, the low occurrence shown by the new myxozoan, and the fact that this is the only host known for this myxozoan, the conservation status of the new species of myxozoan is likely to be connected to the future survival of its host.Objective To report detailed knowledge about the clinical manifestations, ciliary phenotypes, genetic spectrum as well as phenotype/genotype correlation in primary ciliary dyskinesia (PCD) in Chinese children. A high prevalence of congenital abnormalities (30.2%, 13/43), observed in patients who underwent comprehensive examination for comorbidities, included thoracic deformity (11.6%, 5/43), congenital heart disease (9.3%, 4/43) and sensorineural deafness (2.3%, 1/43). For 24 children aged >6 years, the mean predicted values of forced expiratory volume in 1 second (FEV1) were 87.2%. Bronchiectasis evident on high-resolution computed tomography was reported in 38.1% of patients (16/42). Biallelic mutations (81 total; 57 novel) were identified in 13 genes:DNAAF3, DNAAF1, DNAH5, DNAH11, CCDC39, CCDC40, CCDC114, CCDC103, HYDIN, CCNO, DNAI1, OFD1 and SPAG1. Overall, ciliary ultrastructural and beat pattern correlated well with the genotype. However, variable phenotypes were also observed in CCDC39 and DNAH5 mutant cilia. Conclusions This large PCD cohort in China broadens the clinical, ciliary phenotypes and genetic characteristics children with PCD. Our findings are roughly consistent with previous studies besides some peculiarities such as high prevalence of associated abnormalities.
    There is growing interest in the mechanisms for natural sensory learning in pro-social contexts. Studies using a maternal model of social behavior in the mouse have provided new insight into the auditory processing of behaviorally relevant pup vocalizations, which are used as communication signals to elicit pup retrieval behavior by adult females. Whether neural and behavioral plasticity in response to these vocalizations reflect auditory associative learning linking the sounds to pups, versus simply a change in maternal responsiveness to evolved vocal signals, remains an open question. Here, we describe a T-maze paradigm to track auditory learning as we pair an initially neutral, non-ethological stimulus with delivery of a pup for retrieval, which is intrinsically reinforcing for rodents. Training is rapid and completely appetitive. Over a period of 7 × 50-minute daily training sessions, animals increasingly use the sound to guide their arm choice for pup retrieval, with an increase in performance from chance to an average of ~80% on day 7. This pairing method establishes a newly-formed sensory association using a natural maternal behavioral response, and lays a solid foundation for studies into the neurochemical and circuit mechanisms that mediate auditory associative learning in natural social contexts.Background Infection may be an acute precipitant of cardiovascular events. However, the relationships between different types of infection and cardiovascular events are less known. Our objective was to determine if exposure to infections of different organ systems in different time periods increases risk of myocardial infarction or venous thromboembolism. Method We used case-crossover analysis with conditional logistic regression to estimate odds ratios (OR) for the association for each infection type during 3 case periods (30, 60, and 90 days prior to index event) compared with control periods (exactly 1 year before). Results This study had a total number of index admissions of 338,021 individuals, of which 82,986 were female; the mean age for individuals with myocardial infarction and venous thromboembolism was 68.48 years and 62.33 years, respectively. With every infection type, there was an increased likelihood of venous thromboembolism. The greatest association was for skin infections, with an OR of 5.39 (95% confidence interval, 4.08- 7.12) within the 7-day window. Finding an experimentally measurable and theoretically computable descriptor for evaluating surface-adsorbate interactions is a significant step toward achieving this goal. By training a machine learning neural network with a large data set of first-principles calculations, we achieve quick and accurate predictions of molecular adsorption energy and transferred charge. Using a serial concentration, an ultrahigh concentration fold of approximately 1100 could be achieved. Our device offers numerous advantages, such as high-processing throughput, high concentration fold, simple channel design, and low-cost fabrication. An unsymmetrical small molecule of a solubilizing donor(Dsol)-acceptor(A)-dye donor(Ddye) type connected with twisted conjugation system is designed for broadband detection (ranging from 250 nm to 700 nm). This molecule has high solubility, thereby facilitating the formation of uniformly dispersed nanoparticles in an insulating polymer matrix, which is deposited on top of OPT semiconductors by a simple solution process. The broadband photodetection shown by the organic photosensitizer is realized with improved mobility by close to an order of magnitude and high on/off current ratio (~105) of the organic semiconductor. Furthermore, p-type charge transport behavior in the channel of the OPT is enhanced through the intrinsic electron-accepting ability of the organic photosensitizer, caused by the unique molecular configuration. Here we report the synthesis of a zinc-layered hydroxide salt (LHS) and its use as a novel nanocontainer for this purpose, storing the corrosion inhibitor molybdate in the interlayer region of the LHS. Layered zinc hydroxide molybdate (ZHM) was obtained by anion-exchange reactions using layered zinc hydroxide acetate (ZHA) as a precursor, obtained by alkaline coprecipitation. The release behavior of molybdate from the ZHM nanocontainers in aqueous NaCl solution (0.05 mol/L) was evaluated using UV-vis absorption spectroscopy. The molybdate release from the ZHM nanocontainers was realized by the anion-exchange mechanism, where chloride anions replaced intercalated molybdate anions.   • To improve the neural network detection accuracy of the electric power bushings in infrared images, a modified algorithm based on the You Only Look Once version 2 (YOLOv2) network is proposed to achieve better recognition results. Specifically, YOLOv2 corresponds to a convolutional neural network (CNN), although its rotation invariance is poor, and some bounding boxes (BBs) exhibit certain deviations. To solve this problem, the standard Hough transform and image rotation are utilized to determine the optimal recognition angle for target detection, such that an optimal recognition effect of YOLOv2 on inclined objects (for example, bushing) is achieved. With respect to the problem that the BB is biased, the shape feature of the bushing is extracted by the Gap statistic algorithm, based on K-means clustering; thereafter, the sliding window (SW) is utilized to determine the optimal recognition area. Experimental verification indicates that the proposed rotating image method can improve the recognition effect, and the SW can further modify the BB. The accuracy of target detection increases to 97.33%, and the recall increases to 95%.Aberrant angiogenesis is a pathological feature of a number of diseases and arises from the uncoordinated expression of angiogenic factors as response to different cellular stresses. Age-related macular degeneration (AMD), a leading cause of vision loss, can result from pathological angiogenesis. As a mutation in the mitochondrial ferritin (FTMT) gene has been associated with AMD, its possible role in modulating angiogenic factors and angiogenesis was investigated. FTMT is an iron-sequestering protein primarily expressed in metabolically active cells and tissues with high oxygen demand, including retina. In this study, we utilized the human retinal pigment epithelial cell line ARPE-19, both as undifferentiated and differentiated cells. The effects of proinflammatory cytokines, FTMT knockdown, and transient and stable overexpression of FTMT were investigated on expression of pro-angiogenic vascular endothelial growth factor (VEGF) and anti-angiogenic pigment epithelial-derived factor (PEDF). Proinflammatory cytokines induced FTMT and VEGF expression, while NF-κB inhibition significantly reduced FTMT expression. VEGF protein and mRNA expression were significantly increased in FTMT-silenced ARPE-19 cells. Using an in vitro angiogenesis assay with endothelial cells, we showed that conditioned media from FTMT-overexpressing cells had significant antiangiogenic effects. Collectively, our findings indicate that increased levels of FTMT inhibit angiogenesis, possibly by reducing levels of VEGF and increasing PEDF expression. The cellular models developed can be used to investigate if increased FTMT may be protective in angiogenic diseases, such as AMD.An adaptive beamformer is sensitive to model mismatch, especially when the desired signal exists in the training samples. Focusing on the problem, this paper proposed a novel adaptive beamformer based on the interference-plus-noise covariance (INC) matrix reconstruction method, which is robust with gain-phase errors for uniform or sparse linear array. In this beamformer, the INC matrix is reconstructed by the estimated steering vector (SV) and the corresponding individual powers of the interference signals, as well as noise power. Firstly, a gain-phase errors model of the sensors is deduced based on the first-order Taylor series expansion. Secondly, sensor gain-phase errors, the directions of the interferences, and the desired signal can be accurately estimated by using an alternating descent method. Thirdly, the interferences and noise powers are estimated by solving a quadratic optimization problem. To reduce the computational complexity, we derive the closed-form solutions of the second and third steps with compressive sensing and total least squares methods. Simulation results and measured data demonstrate that the performance of the proposed beamformer is always close to the optimum, and outperforms other tested methods in the case of gain-phase errors.Industrial food animal production uses huge amounts of antibiotics worldwide. This study evaluates whether bacterial resistance in the red kite (Milvus milvus) differs between two wintering areas selected based on patent differences in farming practices-particularly in the industrial production of food animals (primarily ***** and poultry) vs. scarce and declining sheep herding. The results support the hypothesis that intensification in food animal production is associated with increased bacterial multidrug resistance in wildlife.     To improve the neural network detection accuracy of the electric power bushings in infrared images, a modified algorithm based on the You Only Look Once version 2 (YOLOv2) network is proposed to achieve better recognition results. Specifically, YOLOv2 corresponds to a convolutional neural network (CNN), although its rotation invariance is poor, and some bounding boxes (BBs) exhibit certain deviations. To solve this problem, the standard Hough transform and image rotation are utilized to determine the optimal recognition angle for target detection, such that an optimal recognition effect of YOLOv2 on inclined objects (for example, bushing) is achieved. With respect to the problem that the BB is biased, the shape feature of the bushing is extracted by the Gap statistic algorithm, based on K-means clustering; thereafter, the sliding window (SW) is utilized to determine the optimal recognition area. Experimental verification indicates that the proposed rotating image method can improve the recognition effect, and the SW can further modify the BB. The accuracy of target detection increases to 97.33%, and the recall increases to 95%.Aberrant angiogenesis is a pathological feature of a number of diseases and arises from the uncoordinated expression of angiogenic factors as response to different cellular stresses. Age-related macular degeneration (AMD), a leading cause of vision loss, can result from pathological angiogenesis. As a mutation in the mitochondrial ferritin (FTMT) gene has been associated with AMD, its possible role in modulating angiogenic factors and angiogenesis was investigated. FTMT is an iron-sequestering protein primarily expressed in metabolically active cells and tissues with high oxygen demand, including retina. In this study, we utilized the human retinal pigment epithelial cell line ARPE-19, both as undifferentiated and differentiated cells. The effects of proinflammatory cytokines, FTMT knockdown, and transient and stable overexpression of FTMT were investigated on expression of pro-angiogenic vascular endothelial growth factor (VEGF) and anti-angiogenic pigment epithelial-derived factor (PEDF). Proinflammatory cytokines induced FTMT and VEGF expression, while NF-κB inhibition significantly reduced FTMT expression. VEGF protein and mRNA expression were significantly increased in FTMT-silenced ARPE-19 cells. Using an in vitro angiogenesis assay with endothelial cells, we showed that conditioned media from FTMT-overexpressing cells had significant antiangiogenic effects. Collectively, our findings indicate that increased levels of FTMT inhibit angiogenesis, possibly by reducing levels of VEGF and increasing PEDF expression. The cellular models developed can be used to investigate if increased FTMT may be protective in angiogenic diseases, such as AMD.An adaptive beamformer is sensitive to model mismatch, especially when the desired signal exists in the training samples. Focusing on the problem, this paper proposed a novel adaptive beamformer based on the interference-plus-noise covariance (INC) matrix reconstruction method, which is robust with gain-phase errors for uniform or sparse linear array. In this beamformer, the INC matrix is reconstructed by the estimated steering vector (SV) and the corresponding individual powers of the interference signals, as well as noise power. Firstly, a gain-phase errors model of the sensors is deduced based on the first-order Taylor series expansion. Secondly, sensor gain-phase errors, the directions of the interferences, and the desired signal can be accurately estimated by using an alternating descent method. Thirdly, the interferences and noise powers are estimated by solving a quadratic optimization problem. To reduce the computational complexity, we derive the closed-form solutions of the second and third steps with compressive sensing and total least squares methods. Simulation results and measured data demonstrate that the performance of the proposed beamformer is always close to the optimum, and outperforms other tested methods in the case of gain-phase errors.Industrial food animal production uses huge amounts of antibiotics worldwide. Vultures and other scavengers can be directly exposed to active antibiotics ingested while feeding on livestock carcasses. This study evaluates whether bacterial resistance in the red kite (Milvus milvus) differs between two wintering areas selected based on patent differences in farming practices-particularly in the industrial production of food animals (primarily swine and poultry) vs. scarce and declining sheep herding. The results support the hypothesis that intensification in food animal production is associated with increased bacterial multidrug resistance in wildlife. Resistance was positively correlated with time elapsed since the beginning of the commercial application of each antibiotic in human and veterinary medicine, with clear differences depending on farming intensification between areas.   • Thanks to the development of high-throughput sequencing technologies, target enrichment sequencing of nuclear ultraconserved DNA elements (UCEs) now allows routinely inferring phylogenetic relationships from thousands of genomic markers. Recently, it has been shown that mitochondrial DNA (mtDNA) is frequently sequenced alongside the targeted loci in such capture experiments. Despite its broad evolutionary interest, mtDNA is rarely assembled and used in conjunction with nuclear markers in capture-based studies. https://www.selleckchem.com/products/paeoniflorin.html Here, we developed MitoFinder, a user-friendly bioinformatic pipeline, to efficiently assemble and annotate mitogenomic data from hundreds of UCE libraries. We compared the efficiency of four different assemblers (IDBA-UD, MEGAHIT, MetaSPAdes, and Trinity) for assembling both UCE and mtDNA loci. Using MitoFinder, we show that metagenomic assemblers, in particular MetaSPAdes, are well suited to assemble both UCEs and mtDNA. Mitogenomic signal was successfully extracted from all 501 UCE libraries allowing confirming species identification using CO1 barcoding. Moreover, our automated procedure retrieved 296 cases in which the mitochondrial genome was assembled in a single contig, thus increasing the number of available ant mitogenomes by an order of magnitude. By leveraging the power of metagenomic assemblers, MitoFinder provides an efficient tool to extract complementary mitogenomic data from UCE libraries, allowing testing for potential mito-nuclear discordance. AIM The present study examined the prevalence of PIU and its associated risk factors including socio-demographic variables, internet use behaviors, and other psychological variables including loneliness, self-esteem, and psychological distress. METHODS A cross-sectional study was conducted among Bangladeshi students (N = 605). Measures included the Internet Addiction Test (IAT), Rosenberg's Self-Esteem Scale, the UCLA Loneliness Scale, and the General Health Questionnaire. RESULTS Using a cut-off score of ≤60 (out of 100) on the IAT, the prevalence of PIU was 16.5% (n = 100). Regression analysis showed that sleep disturbance (more or less than 6-7 hours sleeping time), not using internet for academic purposes, and online chatting were risk factors for PIU. Loneliness and psychological distress were positively correlated with PIU, whereas self-esteem was negatively correlated. To extend the findings obtained using tissue from epilepsy patients, we evaluated GABAR expression and modulation by allopregnanolone and Ro15-4513 in cultured rat hippocampal neurons exposed to high extracellular potassium (HK) to increase neuronal activity. It leads to considerable economic losses as a result of low productivity of infected animals and the long debilitating illness in humans. Despite its impact on human and animal health, little attention has been paid on Brucella infections in domestic animals. It is in this light that the prevalence of Brucella antibodies was determined in domestic animals with the overarching goal of improving our knowledge on brucellosis in southern Cameroon. During cross-sectional studies conducted from December 2016 to August 2018 in five sites of southern Cameroon, blood samples were collected in cattle, sheep, goat, pig and dog. Plasma was obtained from each blood sample and Brucella antibodies were detected using the Rose Bengal test and the enzyme-linked immunosorbent assay (ELISA). Endothelial-stromal interactions were observed across the whole cornea transversing DM during early postnatal development (P3), while these interactions became restricted to the periphery in the mature murine cornea (P30). In adult human corneas, endothelial extensions through the DM were observed in the peripheral cornea. The pattern of FRAP in both mature **** and human central corneas demonstrated endothelial-endothelial cell communication.

ACZ-loaded transfersomes were formulated by the ethanol injection method, using phosphatidylcholine (PC) and different edge activators, including Tween 80, Span 60, and Cremophor RH 40. The effects of the ratio of lipid to surfactant and type of surfactant on % drug released after 8 h (Q8h) and entrapment efficiency (EE%) were investigated by using Design-Expert software. The optimized formula was formulated as TGS, using poloxamers as gelling agents. In vitro and in vivo characterization of ACZ-loaded TGS was performed. According to optimization study, F8 had the highest desirability value and was chosen as the optimized formula for preparing TGS. F8 appeared as spherical elastic nanovesicles with Q8h of 93.01 ± 3.76% and EE% of 84.44 ± 2.82. Compared to a free drug, TGS exhibited more prolonged drug release of 71.28 ± 0.46% after 8 h, higher ex vivo permeation of 66.82 ± 1.11% after 8 h and a significant lowering of intraocular pressure (IOP) for 24 h. A total of 30 randomized controlled trials (RCT), comprising 43 trials and 326 participants in total, were included in this review. The meta-analysis showed an overall significant effect in Time-To-Exhaustion (TTE) and Time-Trial (TT) performance, when ingesting carbohydrates and proteins (CHO-PRO) compared to CHO-only (p = 0.03 and p = 0.0007, respectively). A subgroup analysis demonstrated a significant effect in TTE by ingesting CHO-PRO compared to CHO, when supplements were provided during and/or following an exercise bout. CHO-PRO significantly improved TTE compared to CHO-only, when a long-term recovery (i.e., ≥8 h) was implemented (p = 0.001). To do so, samples of chitosan and pullulan impregnated with CN-NL/GA and CN-NL were analysed through Short Wave Infrared (SWIR) and Visible-Near Infrared (VisNIR) hyperspectral cameras. Two different chemometric tools for qualitative and quantitative analysis have been applied, principal component analysis (PCA) and partial least square regression (PLSR) models. Promising results were obtained in the VisNIR range, which made it possible for us to visualize the CN-NL/GA compound on chitosan and CN-NL on pullulan substrates. Additionally, the PLSR model results had determination coefficient ( R C 2 ) for calibration and cross-validation ( R C V 2 )   values of 0.983 and 0.857, respectively. Minimum values of root-mean-square error for calibration (RMSEC) and cross-validation (RMSECV) of CN-NL/GA were 0.333 and 0.993 g, respectively. A subgroup analysis demonstrated a significant effect in TTE by ingesting CHO-PRO compared to CHO, when supplements were provided during and/or following an exercise bout. In a univariate analysis, dogs with pulmonary metastases and high pain at the time of irradiation had short overall survival times; use of high radiation doses and chemotherapy were associated with improved survival.   • BACKGROUND The treatment and management of long-term health conditions is the greatest challenge facing health systems around the world today. Innovative approaches to patient care in the community such as Anticipatory Care Planning (ACP), which seek to help with the provision of high-quality comprehensive care to older adults at risk of functional decline, require evaluation. This study will evaluate one approach that will include primary care as the setting for ACP. METHODS/DESIGN This study will help to determine the feasibility for a definitive randomised trial to evaluate the implementation and outcomes of an ACP intervention. The intervention will be delivered by specially trained registered nurses in a primary care setting with older adults identified as at risk of functional decline. The intervention will comprise (a) information collection via patient assessment; (b) facilitated informed dialogue between the patient, family carer, general practitioner and other healthcare practitioners; and, (c) documentation of the agreed support plan and follow-up review dates. Through a structured consultation with patients and their family carers, the nurses will complete a mutually agreed personalised support plan. DISCUSSION This study will determine the feasibility for a full trial protocol to evaluate the implementation and outcomes of an (ACP) intervention in primary care to assist older adults aged 70 years of age or older and assessed as being at risk of functional decline. The study will be implemented in two jurisdictions on the island of Ireland which employ different health systems but which face similar health challenges. This study will allow us to examine important issues, such as the impact of two different healthcare systems on the health of older people and the influence of different legislative interpretations on undertaking cross jurisdictional research in Ireland. METHODS We comprehensively integrated gene expression data acquired from 374 HCC and 50 normal tissues in The Cancer Genome Atlas (TCGA). Differentially expressed genes (DEGs) analysis and univariate Cox regression analysis were performed to identify DEGs that related to overall survival. An immune prognostic model was constructed using the Lasso and multivariate Cox regression analyses. Furthermore, Cox regression analysis was applied to identify independent prognostic factors in HCC. The correlation analysis between immune-related signature and immune cells infiltration were also investigated. IPS has been extensively studied in severe mental illness and physical disabilities, but there have been no formal randomised controlled trials (RCTs) in alcohol and drug dependence. The Individual Placement and Support for Alcohol and Drug Dependence (IPS-AD) study should determine whether IPS for patients with alcohol use disorder (AUD), opioid use disorder (OUD) and other drug use disorder is effective. DESIGN/METHODS The IPS-AD study is a seven-site, pragmatic, two-arm, parallel-group, superiority RCT. IPS-AD includes a realist process evaluation. Eligible patients (adult, unemployed or economically inactive for at least 6 months and wishing to obtain open job market employment and enrolled in ongoing community treatment-as-usual (Tponding National Insurance contributions and tax paid); (2) time from randomisation to first competitive employment; (3) number of competitive job appointments; (4) job tenure (length of longest held competitive employment); (5) sustained employment (tenure in a single appointment for at least 13 weeks); and (6) job search self-efficacy. A primary cost-benefit analysis and a secondary cost-effectiveness analysis will be done using the primary outcome and secondary vocational outcomes, respectively and will include addiction treatment and social and health outcomes and their associated reference costs. The process evaluation will address IPS implementation and delivery. DISCUSSION The IPS-AD study is the first large-scale, multi-site, definitive, superiority RCT of IPS for people with alcohol and drug dependence. Acupuncture is an effective tool for pain management. However, electroacupuncture specifically for the relief of postoperative pain after nasal endoscopic surgery has not yet been studied in a randomized controlled trial. METHODS/DESIGN This randomized sham-controlled patient- and assessor-blind pilot trial has been designed to evaluate the efficacy and safety of electroacupuncture in managing postoperative pain following nasal endoscopic surgery to treat sinusitis due to nasal polyps. Altogether, 30 participants will be randomly allocated to an electroacupuncture or non-invasive sham control in a 11 ratio. Treatment will occur within 2 h before the operation, immediately after the operation upon arrival in the recovery ward, and once daily for 3 days. The primary outcome is the pain numerical rating scale, which will be analyzed using the area under the curve. The secondary outcome measures include heart rate and blood pressure after the operation, sleep quality during the hospital stay (actigraph), quality of recovery, and the 36-item short form health survey.

IPS has been extensively studied in severe mental illness and physical disabilities, but there have been no formal randomised controlled trials (RCTs) in alcohol and drug dependence. The Individual Placement and Support for Alcohol and Drug Dependence (IPS-AD) study should determine whether IPS for patients with alcohol use disorder (AUD), opioid use disorder (OUD) and other drug use disorder is effective. DESIGN/METHODS The IPS-AD study is a seven-site, pragmatic, two-arm, parallel-group, superiority RCT. IPS-AD includes a realist process evaluation. Eligible patients (adult, unemployed or economically inactive for at least 6 months and wishing to obtain open job market employment and enrolled in ongoing community treatment-as-usual (Tponding National Insurance contributions and tax paid); (2) time from randomisation to first competitive employment; (3) number of competitive job appointments; (4) job tenure (length of longest held competitive employment); (5) sustained employment (tenure in a single appointment for at least 13 weeks); and (6) job search self-efficacy. A primary cost-benefit analysis and a secondary cost-effectiveness analysis will be done using the primary outcome and secondary vocational outcomes, respectively and will include addiction treatment and social and health outcomes and their associated reference costs. The process evaluation will address IPS implementation and delivery. DISCUSSION The IPS-AD study is the first large-scale, multi-site, definitive, superiority RCT of IPS for people with alcohol and drug dependence. Registered on 1 February 2018.BACKGROUND Postoperative pain is common after nasal endoscopic surgery. It interferes with the quality of sleep and delays postoperative recovery. Acupuncture is an effective tool for pain management. However, electroacupuncture specifically for the relief of postoperative pain after nasal endoscopic surgery has not yet been studied in a randomized controlled trial. METHODS/DESIGN This randomized sham-controlled patient- and assessor-blind pilot trial has been designed to evaluate the efficacy and safety of electroacupuncture in managing postoperative pain following nasal endoscopic surgery to treat sinusitis due to nasal polyps. Altogether, 30 participants will be randomly allocated to an electroacupuncture or non-invasive sham control in a 11 ratio. Treatment will occur within 2 h before the operation, immediately after the operation upon arrival in the recovery ward, and once daily for 3 days. The primary outcome is the pain numerical rating scale, which will be analyzed using the area under the curve. The secondary outcome measures include heart rate and blood pressure after the operation, sleep quality during the hospital stay (actigraph), quality of recovery, and the 36-item short form health survey.
