Citrus canker caused by Xanthomonas citri subsp. citri (Xcc) is one of the most devastating diseases in citrus. Meiwa kumquat (Fortunella crassifolia) has shown a durable resistance against Xcc. Here, we aimed to characterize the mechanisms responsible for such a durable resistance by characterizing the transcriptional and physiological responses of Meiwa kumquat to Xcc. Inoculation of Meiwa kumquat with Xcc promoted immune responses such as upregulation of PR genes, accumulation of salicylic acid, hypersensitive response (HR)-like cell death and early leaf abscission. Hypertrophy and hyperplasia symptoms, which are known to be caused by Xcc-induction of the canker susceptibility gene LOB1 through the transcription activator-like effector (TALE) PthA4, always appear prior to the development of cell death. Mutation of pthA4 in Xcc abolished the induction of LOB1, canker symptoms, cell death, and leaf abscission and reduced the expression of PR genes in inoculated kumquat leaves without reducing Xcc titers in planta. Transcriptome analysis demonstrated that PthA4 promotes plant biotic and abiotic stress responses and the biosynthesis of abscisic acid. Transcriptional induction of LOB1 homologs in Meiwa kumquat by Xcc pthA4 mutant strains carrying a repertoire of designer TALEs promoted the elicitation of HR-like phenotype and leaf abscission, suggesting that kumquat response to Xcc is associated with upregulation of LOB1. Our study suggests a novel mechanism of plant resistance to Xanthomonas via elicitation of immune responses by upregulation of a host susceptibility gene.

A prospective cohort study of obstetric patients admitted to MBBM Foundation and Buzzi Hospital (Lombardy, Northern Italy) from March 16th to May 22nd, 2020. There were 1,177 women screened by the questionnaire, which yielded a positive result in 130 (11.0%) cases. SARS-CoV-2 RT-PCR was performed in 865 (73.5%) patients, identifying 51 (5.9%) infections. During the first period, there were 29 infected mothers, 4 (13.8%) oflow risk of SARS-CoV-2 infection in the context of a targeted and a universal viral testing approach. In 15.7% of infected women, correct classification as a suspected case of infection was due to investigation of olfactory and taste disorders. Extension of the assessed time-frame to 30 days may be worth considering to increase the questionnaire's performance.
Cytopenias affect the outcomes of highly active anti-retroviral therapy that results in higher morbidity, mortality, and impaired quality of life. The purpose of this study was to assess the prevalence of cytopenia and its associated factors among HIV infected adults on highly active antiretroviral therapy at Mehal Meda Hospital, North Shewa Zone, Ethiopia.
A cross-sectional health facility based study was conducted among 499 consecutively selected adult HIV infected patients taking HAART for at least six months from January to April 2018. The study participant's socio-demographic and clinical information was collected using a pre-tested questionnaire and reviewing of medical records by trained clinical nurses. Complete blood count and CD4 T cell count were determined by Sysmex KX-21 N and BD FACS count respectively. Bivariate and multivariate analysis was performed to identify the independently associated factors of cytopenia and prevalence ratios and their 95% confidence intervals were estimated using Prevalence increased as the CD4 count decreases. Therefore, these warrant the need for monitoring hematological parameters of HIV infected patients on HAART to reduce morbidity and mortality.The rising public health threat of antimicrobial resistance, the influence of food service companies, as well as the overall lack of positive image of using medical products in intensive farming are major drivers curbing antimicrobial use. In the future, government policies may affect practices of antimicrobial use in beef production in feedlots, a prominent current user of antimicrobials in animal agriculture, but also the agricultural industry generating the highest cash receipt in the U.S. Our objective was to estimate the cost effect from the following policies in feedlots 1) using antimicrobials for disease prevention, control, and treatment; 2) using antimicrobials only for treatment of disease; and 3) not using antimicrobials for any reason. However, the effect of the OTU protease activity in the context of the full-length L protein (approximately 4000 amino acids) is only poorly characterized, and recombinant CCHFV with the C40A mutation could not be rescued. Here, we employed transcriptionally active virus-like particles (tc-VLPs) to investigate the interaction between the L-embedded OTU protease and the IFN system. Our data show a cis requirement of the OTU protease for optimal CCHFV polymerase activity in human HuH-7 cells.

Probiotics and proanthocyanidin-rich cinnamon extract (PRCE) have numerous potential health benefits, but they are very sensitive to degradation in various environmental conditions. Additionally, the combination of these two materials into a single structure could possibly enhance their therapeutic properties. Thus, the aim of this study was to produce and evaluate the solid lipid microparticles covered by electrostatic interactions of polymers in which Lactobacillus paracasei (BGP1) and Bifidobacterium animalis subsp. lactis (****) were either encapsulated alone or co-encapsulated with PRCE. https://www.selleckchem.com/screening-libraries.html Through turbidimetric titration and zeta potential measurement, the optimum coacervates were obtained at a pH of 4.2 with the proteinpolysaccharide mixing ratio of 61. In China, there are few studies on the authenticity of fish products in Henan province of China. In this study, 179 fish samples were collected from supermarkets in Zhengzhou city and Xinxiang city in Henan province, China. COI gene sequences were obtained with PCR technology by designing specific primers and universal primers. COI gene sequences of all samples were obtained to identify species, which is used to investigate species substitution and mislabeling of the fish sold in the two regional markets. The molecular identification results showed that 28.49% (51/179) fish samples were not consistent with the labels. Substitution of high-price fish by low-price fish was prevalent. For example, halibut (Pleuronectiformes) and cod (Gadus) are replaced by striped catfish (Pangasianodon hypophthalmus), and some merchants label bighead carp (Hypophthalmichthys nobilis) as cod (Gadus), there are also accidental labeling errors (such as labels for greenfin horse-faced filefish (Thamnaconus septentrionalis) have been identified as grass carp (Ctenopharyngodon idella) and Wuchang bream (Megalobrama amblycephala) etc. Most of the samples labeled correctly are the fish of low economic value and the fresh fish. This study shows that almost all the commercial fish can be identified by COI DNA barcoding by newly designed primers. However, the effects and mechanisms of Monascus purpureus-fermented common buckwheat (HQ) on non-alcoholic fatty liver disease (NAFLD) and dyslipidemia are unclear. Here, oral supplementation of HQ significantly inhibited the abnormal growth of body weight and epididymal white adipose tissue (eWAT), prevented the hypertrophy of epididymal adipocytes, ameliorated some biochemical parameters of serum and liver related to lipid metabolism in **** fed a high-fat and high-cholesterol diet (HFD). Histological analysis also showed that the excessive accumulation of lipid droplets in the livers induced by HFD-feeding was greatly alleviated by HQ supplementation. Metagenomic analysis revealed that HQ supplementation made significant structural changes in the intestinal microflora of **** fed with HFD. The Spearman's correlation analysis revealed that physiological index, serum and liver lipid profiles were positively correlated with Bacteroidales S24-7, Streptococcus, Allobaculum, and Clostridiales XIII, but negatively associated with Lactobacillus, Ruminococcaceae_NK4A214 group, Ruminiclostridium, and Alistipes. UPLC-QTOF/MS-based liver metabolomics demonstrated that HQ intervention had significant regulatory effects on the metabolic pathways of primary bile acid biosynthesis, pyrimidine metabolism, ether lipid metabolism, glutathione metabolism, glycine, serine and threonine metabolism, and amino sugar and nucleotide sugar metabolism, etc. Additionally, HQ intervention regulated the mRNA levels of hepatic genes involved in hepatic lipid metabolism and bile acid homeostasis. Its infusion is traditionally ingested for the treatment and prevention of hepatic disorders. Here we report that an inulin type fructan is present in the B. trimera infusion and contributes to the hepatoprotective effect of the species. In vitro, inulin at 300 μg/mL, was able to scavenger 97% of the DPPH radicals. In vivo experiments showed that it protected the liver against CCl4-induced injuries. The administration of inulin at low dose of 1 mg/kg significantly reduced the blood levels of ALT, AST and ALP, reduced the lipid peroxidation and increased the catalase activity and the levels of reduced glutathione in the liver of CCl4-treated ****. Moreover, the administration of inulin at 100 mg/kg increased GSH levels in the liver of Naïve ****. No signs of toxicity were observed. Thus, inulin present in B. trimera infusion protects the liver from the oxidative stress caused by CCl4 administration and can corroborate with the hepatoprotective effects presented by the species infusion.

Solid organ transplant (SOT) recipients may be more vulnerable to coronavirus disease 2019 (COVID-19). Data on the clinical course of COVID-19 in immunosuppressed patients are limited, and the optimal management strategy for these patients is yet unclear.
We present 53 SOT recipients (31 kidney transplant recipients, 8 liver transplant recipients, 5 heart transplant recipients, 5 lung transplant recipients, 3 liver-kidney transplant recipients, and 1 kidney-after-heart transplant recipient), transplanted at a Swedish high-volume transplant center and each diagnosed with COVID-19 between February 21, 2020 and June 22, 2020. Demographic, clinical, and treatment data were extracted from the electronic patient files.
Patients reported fever (61%), cough (43%), diarrhea (31%), and upper respiratory symptoms (29%). The median age was 56 years, and 57% were male. According to severity, 55% had mild, 13% had moderate, 19% had severe, and 13% had critical disease. Thirty-seven patients (70%) were hospitalized, w of patients could be managed on an outpatient basis, while keeping a low threshold for admission. Mild and moderate disease forms seem to have a good outcome.
This study aimed to investigate the incidence and risk factors of endophthalmitis after transconjunctival pars plana vitrectomy (PPV) without intraoperative subconjunctival antibiotics.

Consecutive cases of transconjunctival 25-gauge PPV without intraoperative subconjunctival antibiotics performed by three retina surgeons at a single surgical site at the Dean McGee Eye Institute from 2012 to 2018 were reviewed.
Of 4,263 cases of PPV without intraoperative subconjunctival antibiotics, five cases (0.117%, 5/4,263) of post-PPV endophthalmitis were identified. Of these five cases, four cases (80%, 4/5) received combined cataract extraction or secondary intraocular lens implantation at the time of PPV. Data on the clinical course of COVID-19 in immunosuppressed patients are limited, and the optimal management strategy for these patients is yet unclear. We present 53 SOT recipients (31 kidney transplant recipients, 8 liver transplant recipients, 5 heart transplant recipients, 5 lung transplant recipients, 3 liver-kidney transplant recipients, and 1 kidney-after-heart transplant recipient), transplanted at a Swedish high-volume transplant center and each diagnosed with COVID-19 between February 21, 2020 and June 22, 2020. Demographic, clinical, and treatment data were extracted from the electronic patient files. Patients reported fever (61%), cough (43%), diarrhea (31%), and upper respiratory symptoms (29%). A promising TTM practice for treatment of pressure ulcers was selected to conduct a clinical trial. This study aimed to evaluate the efficacy of the TTM practice for the treatment of pressure ulcers using honey or a Thai Herbal Oil preparation (THO) based on the TTM wound diagnosis comparing with the standard practice. Methods The study design was an open-label randomized controlled trial. Sixty-six participants, with pressure ulcers at least stage II-IV or unstageable, were allocated to two groups via minimization. A TTM practice group received honey or THO depending on the TTM diagnosis via the Thai Traditional Medicine Pressure Ulcer Assessment Tool (TTM-PUAT). A standard practice group received advanced dressings, including hydrogel, alginate, silver-impregnated, or hydrocolloid dressings. The primary outcome was the Pressure Ulcer Scale for Healing (PUSH). Results Both TTM practice and standard practice showed a significant reduction in PUSH scores after treatments. However, there was no significant difference in PUSH score reduction between the groups. The mean PUSH score reduction over the 6-week period was 2.58 ± 3.38 (95% CI 1.34-3.82) in the TTM practice group and 3.24 ± 3.49 (95% CI 1.91-4.57) in the standard practice group (p = 0.284). The TTM practice and standard practice accelerated pressure ulcer healing without statistically significant difference between the practices, during 6 weeks in a home-based care setting. Aims To evaluate whether spironolactone added to an ongoing antipsychotic treatment improves cognitive functioning in schizophrenia. Methods The add-on spironolactone for the treatment of schizophrenia trial (SPIRO-TREAT) is a multicenter randomized, placebo-controlled trial with three arms (spironolactone 100 mg, spironolactone 200 mg and placebo). Schizophrenia patients are treated for three weeks and then followed-up for additional nine weeks. As primary outcome, we investigate changes in working memory before and at the end of the intervention phase. We will randomize 90 patients. Eighty-one patients are intended to reach the primary endpoint measure at the end of the three-week intervention period. Secondary endpoints include other measures of cognition, psychopathology, safety measures and biological measures. Conclusions SPIRO-TREAT is the first study evaluating the efficacy of the mineralocorticoid receptor antagonist spironolactone to improve cognitive impairments in schizophrenia patients targeting the NRG1-ERBB4 signaling pathway. We previously reported elevated levels of phosphorylated JNK (pJNK), indicative of JNK hyperactivation, in the CA1 hippocampus of chronically epileptic rats. We also showed that pharmacological inhibition of JNK activity reduced seizure frequency in a dose-dependent fashion (Tai TY et al., Neuroscience, 2017). Building on these observations, the objectives of this current study were to investigate the timeline of JNK activation during epileptogenesis, and to identify the JNK isoform(s) that undergo hyperactivation in the chronic epilepsy stage. As Covid-19 forces an oil surplus and lowered production, there are opportunities to enact such changes - particularly by redirecting oil industry labor toward the growing problem of well decommissioning.Canada's Extractive Sector Transparency Measures Act (ESTMA) is the culmination of a series of proposals and consultations with government, industry and civil society organizations to address conflict over Canadian extractive industry. Created in the context of a global call for extractive industry accountability, as well as increasing scrutiny of Canadian mining activities for alleged human rights and environmental abuses, the ESTMA aims to deter corruption via financial reporting requirements for Canadian extractive firms operating in Canada and abroad. By mandating that firms publicly disclose payments to various levels of government, however, the ESTMA is constructed atop global corruption discourse that identifies host states in the Global South as the source of social pathologies that facilitate corruption, largely excluding a critical analysis of extractive firms in the Global North. Drawing on interviews, document analysis of material related to the ESTMA and case studies of extractive firm financial reporting, this paper argues that under the ESTMA's financial reporting processes, corporate risk management trumps meaningful social regulation. While the Act does mandate disclosures useful to the advocacy community, limited oversight, a lack of standardized reporting and excluded activities under the Act mean that the ESTMA offers limited leverage to substantively address the human and ecological cost of Canada's extractive industry. As has resulted from transparency policies more broadly, however, the ESTMA provides firms a means to counter broader critique and, in complying with audit culture, promotes investment security.This special section Beyond Transparency Rethinking the Government of Extraction examines the relationship between international transparency discourse in the extractive sector, and the persistent association of unaccountable government, socioeconomic injustice and ongoing environmental hazards associated with extractive firms and their operations. Our critical analyses of transparency- situate the discourse and practice within the overall turn-of-millennium regulatory capture of states in the global North - including Canada, the US and the UK - by oil and mining industry interests. Contributors probe how transparency regimes have been applied to oil and extractive sector 'host states' in the global South, in particular Nigeria, while the rent-seeking practices that these regimes seek to expose are rarely tied to corporate malfeasance in the North. We employ this introduction to consider global transparency discourse and regulatory regimes in the light of the full cost of extraction. In order to achieve this ambition, there is a need for new and more mineral extraction which is necessary for the technology for this low-carbon transition. The research identifies the key questions that need to be resolved under each element of justice and the unfortunate limited timeframes for action. Critical justice areas include taxation, environmental impact assessments, waste management, social license to operate, and cross-border actions. Resolving these issues will directly address societal issues of inequality and ensure a just transition to a low-carbon economy. Already there is a global race for critical minerals, and justice needs a stronger role in its development based on evidence to-date.The aim of this paper is to present a brief historical analysis of the Australian mining industry and the development of its social licence to operate. Commencing with the discovery of coal in the 18th century, to gold and copper and base metals in the 19th century, to the world class iron ore, mineral sands and diamond mines of the 20th century to the current day, the paper will attempt to determine how and why an industry, formerly well-respected by the public, is considered to be a pariah in the eyes of many sections of society. The theory of social licence and its use in the Australian minerals industry is briefly described and a working definition of "an honest, transparent engagement resulting in a beneficial outcome to all parties before, during and after mining" is adopted throughout the paper. Case studies illustrating examples where mining operations were, and continue to be sustained for decades, provide clear evidence of having a social licence to operate. On the other hand, examples are provided illustrating where poor environmental management practices, tailings dam failures, disputes with landowners, and/or government intervention have resulted in the loss of the social licence and early, unplanned closure.
As Covid-19 forces an oil surplus and lowered production, there are opportunities to enact such changes - particularly by redirecting oil industry labor toward the growing problem of well decommissioning.Canada's Extractive Sector Transparency Measures Act (ESTMA) is the culmination of a series of proposals and consultations with government, industry and civil society organizations to address conflict over Canadian extractive industry. Created in the context of a global call for extractive industry accountability, as well as increasing scrutiny of Canadian mining activities for alleged human rights and environmental abuses, the ESTMA aims to deter corruption via financial reporting requirements for Canadian extractive firms operating in Canada and abroad. By mandating that firms publicly disclose payments to various levels of government, however, the ESTMA is constructed atop global corruption discourse that identifies host states in the Global South as the source of social pathologies that facilitate corruption, largely excluding a critical analysis of extractive firms in the Global North. Drawing on interviews, document analysis of material related to the ESTMA and case studies of extractive firm financial reporting, this paper argues that under the ESTMA's financial reporting processes, corporate risk management trumps meaningful social regulation. While the Act does mandate disclosures useful to the advocacy community, limited oversight, a lack of standardized reporting and excluded activities under the Act mean that the ESTMA offers limited leverage to substantively address the human and ecological cost of Canada's extractive industry. As has resulted from transparency policies more broadly, however, the ESTMA provides firms a means to counter broader critique and, in complying with audit culture, promotes investment security.This special section Beyond Transparency Rethinking the Government of Extraction examines the relationship between international transparency discourse in the extractive sector, and the persistent association of unaccountable government, socioeconomic injustice and ongoing environmental hazards associated with extractive firms and their operations. Our critical analyses of transparency- situate the discourse and practice within the overall turn-of-millennium regulatory capture of states in the global North - including Canada, the US and the UK - by oil and mining industry interests. Contributors probe how transparency regimes have been applied to oil and extractive sector 'host states' in the global South, in particular Nigeria, while the rent-seeking practices that these regimes seek to expose are rarely tied to corporate malfeasance in the North. We employ this introduction to consider global transparency discourse and regulatory regimes in the light of the full cost of extraction. Since the turn of the millennium, we argue, attention to extraction's full costs have been largely overshadowed in policy discourse via global transparency regimes, notably the Extractive Industries Transparency Initiative.Since the 1970s, environmentalists have warned that overconsumption, especially of minerals and fossil fuels, will lead to resource depletion. But there are compelling reasons to question the assertion that we are running out. On the one hand, new technologies and discoveries have increased the supply of petroleum and natural gas. On the other, concerns about global climate change and the competitiveness of renewable energy are turning coal into a sunset industry and threaten to transform oil reserves into stranded assets. In contrast to fossil fuels, which are consumed in the process of generating energy, virtually all of the metals excavated in the past remain available even after they have been put to use. Even though the average size and degree of mineralization of recently-discovered ore bodies is on the decline, there has never been as much copper, silver, and gold available for human consumption as there is today. Whether incorporated into digital technology or infrastructure, the majority of metals remain available for recycling, which is generally less expensive, uses less energy, and has fewer environmental impacts than extracting minerals from the earth. The threat posed by climate change from continued use of fossil fuels, and the impacts of environmental degradation caused by resource extraction, demand greater attention than the misleading specter of peak oil or running out of metals.It is a fundamental objective to transition towards a low-carbon economy worldwide which is supported by an international legal agreement - the 2015 Paris Agreement. In order to achieve this ambition, there is a need for new and more mineral extraction which is necessary for the technology for this low-carbon transition. Mid-frequency active sonar (MFAS), used for antisubmarine warfare (ASW), has been associated with multiple beaked whale (BW) mass stranding events. Multinational naval ASW exercises have used MFAS offshore of the Mariana Archipelago semi-annually since 2006. We report BW and MFAS acoustic activity near the islands of Saipan and Tinian from March 2010 to November 2014. Signals from Cuvier's (Ziphius cavirostris) and Blainville's beaked whales (Mesoplodon densirostris), and a third unidentified BW species, were detected throughout the recording period. Both recorders documented MFAS on 21 August 2011 before two Cuvier's beaked whales stranded on 22-23 August 2011. We compared the history of known naval operations and BW strandings from the Mariana Archipelago to consider potential threats to BW populations. Eight BW stranding events between June 2006 and January 2019 each included one to three animals. Half of these strandings occurred during or within 6 days after naval activities, and this co-occurrence is highly significant. We highlight strandings of individual BWs can be associated with ASW, and emphasize the value of ongoing passive acoustic monitoring, especially for beaked whales that are difficult to visually detect at sea. We strongly recommend more visual monitoring efforts, at sea and along coastlines, for stranded cetaceans before, during and after naval exercises.In disease-suppressive soils, microbiota protect plants from root infections. Bacterial members of this microbiota have been shown to produce specific molecules that mediate this phenotype. To date, however, studies have focused on individual suppressive soils and the degree of natural variability of soil suppressiveness remains unclear. Here, we screened a large collection of field soils for suppressiveness to Fusarium culmorum using wheat (Triticum aestivum) as a model host plant. A high variation of disease suppressiveness was observed, with 14% showing a clear suppressive phenotype. The microbiological basis of suppressiveness to F. culmorum was confirmed by gamma sterilization and soil transplantation. Amplicon sequencing revealed diverse bacterial taxonomic compositions and no specific taxa were found exclusively enriched in all suppressive soils. Nonetheless, co-occurrence network analysis revealed that two suppressive soils shared an overrepresented bacterial guild dominated by various Acidobacteria. In addition, our study revealed that volatile emission may contribute to suppression, but not for all suppressive soils. Our study raises new questions regarding the possible mechanistic variability of disease-suppressive phenotypes across physico-chemically different soils. Accordingly, we anticipate that larger-scale soil profiling, along with functional studies, will enable a deeper understanding of disease-suppressive microbiomes.Adopting a new diet is a significant evolutionary change, and can profoundly affect an animal's physiology, biochemistry, ecology and genome. To study this evolutionary transition, we investigated the physiology and genomics of digestion of a derived herbivorous fish, Cebidichthys violaceus. We sequenced and assembled its genome (N50 = 6.7 Mb) and digestive transcriptome, and revealed the molecular changes related to digestive enzymes (carbohydrases, proteases and lipases), finding abundant evidence of molecular adaptation. Specifically, two gene families experienced expansion in copy number and adaptive amino acid substitutions amylase and carboxyl ester lipase (cel), which are involved in the digestion of carbohydrates and lipids, respectively. Both show elevated levels of gene expression and increased enzyme activity. Because carbohydrates are abundant in the prickleback's diet and lipids are rare, these findings suggest that such dietary specialization involves both exploiting abundant resources and scavenging rare ones, especially essential nutrients, like essential fatty acids.Gliding animals traverse cluttered aerial environments when performing ecologically relevant behaviours. However, it is unknown how gliders execute collision-free flight over varying distances to reach their intended target. We quantified complete glide trajectories amid obstacles in a naturally behaving population of gliding lizards inhabiting a rainforest reserve. In this cluttered habitat, the lizards used glide paths with fewer obstacles than alternatives of similar distance. Their takeoff direction oriented them away from obstacles in their path and they subsequently made mid-air turns with accelerations of up to 0.5 g to reorient towards the target tree. These manoeuvres agreed well with a vision-based steering model which maximized their bearing angle with the obstacle while minimizing it with the target tree. Nonetheless, negotiating obstacles reduced mid-glide shallowing rates, implying greater loss of altitude. Finally, the lizards initiated a pitch-up landing manoeuvre consistent with a visual trigger model, suggesting that the landing decision was based on the optical size and speed of the target. They subsequently followed a controlled-collision approach towards the target, ending with variable impact speeds. Overall, the visually guided path planning strategy that enabled collision-free gliding required continuous changes in the gliding kinematics such that the lizards never attained theoretically ideal steady-state glide dynamics.Rapid and unprecedented ecological change threatens the functioning and stability of ecosystems. On coral reefs, global climate change and local stressors are reducing and reorganizing habitat-forming corals and associated species, with largely unknown implications for critical ecosystem functions such as herbivory. The minimum bactericidal concentration of this recombinant protein was found to be 4.54 μM against A. hydrophila and Staphylococcus aureus. Interestingly, rLrNKEF-B showed relative percent survival of 72.6 % in A. hydrophila challenged L. rohita, and the survival was found to be associated with a high level of expression of different cytokines, anti-oxidant genes and perforin in the rLrNKEF-B treated L. rohita. An indirect ELISA assay for estimation of NKEF was developed in L. rohita, and the concentrations of NKEF-B increased with time periods post A. hydrophila challenge viz., 0 h (42.56 ng/mL), 12 h (174 ng/mL) and 48 h (370 ng/mL) in rohu serum. Our results suggest a crucial role of LrNKEF-B in innate immunity against biotic stress and oxidative damage and also having antibacterial activity.In Alzheimer's disease (AD), a decline in function of neural progenitor cells (NPCs) results in a reduced capacity for neural regeneration. It has been shown that plasma oxidized low-density lipoprotein (ox-LDL) levels are positively correlated with severity in patients with AD. However, the direct effects of ox-LDL on NPCs are unknown. Thus, we examined the effects of ox-LDL on the proliferation and differentiation of mouse NPCs into neural cells. Mouse induced pluripotent stem (iPS) cell-derived embryoid bodies were stimulated with Noggin and SB431542 for 4 days. Mouse NPCs were then collected using anti-polysialic acid-neural cell adhesion molecule antibodies in a magnetic separator. The proliferation of mouse NPCs was examined using the MTT assay. The differentiation of mouse NPCs into neural cells was examined by the expression of NeuN (a neuronal-specific nuclear protein) using immunofluorescence staining and Western blot analysis. Treatment with ox-LDL did not affect the proliferation of mouse NPCs. While treatment with all-trans retinoic acid (ATRA), epidermal growth factor (EGF), and basic fibroblast growth factor (FGF) significantly induced NeuN expression in the differentiated NPCs (P less then 0.01), the addition of ox-LDL significantly inhibited the NeuN expression (P less then 0.05). Pretreatment with SC-79 (an Akt activator) significantly reversed the inhibitory effect of ox-LDL on NeuN expression (P less then 0.05). Treatment with ox-LDL significantly inhibited Akt phosphorylation (P less then 0.05) and CREB phosphorylation induced by ATRA, EGF, and basic FGF (P less then 0.05). In this study, we aimed to investigate the anti-allergic activities of PF. The pathogenesis of several NDs including Alzheimer's (AD), Parkinson's (PD), and Huntington's (HD) diseases are associated with oxidative stress (OS). In order to maintain the normal functions of neurons, lower levels of reactive oxygen species (ROS) and reactive nitrogen species (RNS) are important, since their increased levels can cause neuronal cell death. It has been found that OS-mediated neurodegeneration involves a number of events including mitochondrial dysfunction, Ca2+ overload, and excitotoxicity. A growing number of studies are suggesting the benefit of using polyphenols for the treatment of neurodegenerative disorders. Indeed, in order to treat most of the NDs, synthetic drugs are extensively used which are found to exert side effects in the course of the treatment. There is mounting evidence that researchers have identified several naturally-occurring chemical compounds in plants, which are used for the management of NDs. Overall, polyphenolic phytochemicals are safer in nature and have negligible side effects. In this article, we have focused on the potential efficacy of polyphenols such as epigallocatechin-3-gallate, curcumin, resveratrol, quercetin and methylated polyphenols berberine against the most common neurodegenerative disorders.The present study investigated the accumulation and depuration effects of hexavalent chromium (Cr6+) in ten tissues (gills, intestines, liver, kidney, blood, heart, bladder, spleen, skin and muscle) of the bighead carp (Aristichthys nobilis). Fish were exposed to graded levels of waterborne Cr6+ (0.01, 0.1, 1 and 5 mg/L) for 4, 7 and 14 days, and subsequently transferred to Cr6+-free water for 14 days. After 14-day exposure, a dose-dependent increase of Cr6+ has been observed in most tissues. While after 14-day depuration, Cr6+ contents were significantly decreased in various tissues except in kidney and spleen where Cr6+ contents significantly increased at the group of 5 mg/L. Considering that Cr6+ highly accumulated in gills, intestines, liver and kidney, the oxidative damage of Cr6+ on the four tissues were further investigated and found that the antioxidant response to Cr6+ were organ-specific. The results in this study indicated that a 14-day period is effective for accumulation and depuration of Cr6+ in bighead carp and there was no health risk of fish muscle consumption.

The present study indicates that treatment with ox-LDL inhibits the differentiation of mouse NPCs into neural cells by inhibiting Akt and CREB activation.Paeoniflorin (PF), a monoterpene glycoside isolated from the aqueous extract of the Chinese herb Radix Paeoniae Alba, has been used for treating various inflammatory diseases. In this study, we aimed to investigate the anti-allergic activities of PF. The anti-anaphylactic activity of PF was investigated using mast cell (MC) degranulation assay as well as Ca2+ influx in vitro and skin swelling and extravasation assays in vivo. It has been found that OS-mediated neurodegeneration involves a number of events including mitochondrial dysfunction, Ca2+ overload, and excitotoxicity. A growing number of studies are suggesting the benefit of using polyphenols for the treatment of neurodegenerative disorders. Indeed, in order to treat most of the NDs, synthetic drugs are extensively used which are found to exert side effects in the course of the treatment. There is mounting evidence that researchers have identified several naturally-occurring chemical compounds in plants, which are used for the management of NDs. Overall, polyphenolic phytochemicals are safer in nature and have negligible side effects. In this article, we have focused on the potential efficacy of polyphenols such as epigallocatechin-3-gallate, curcumin, resveratrol, quercetin and methylated polyphenols berberine against the most common neurodegenerative disorders.The present study investigated the accumulation and depuration effects of hexavalent chromium (Cr6+) in ten tissues (gills, intestines, liver, kidney, blood, heart, bladder, spleen, skin and muscle) of the bighead carp (Aristichthys nobilis). Fish were exposed to graded levels of waterborne Cr6+ (0.01, 0.1, 1 and 5 mg/L) for 4, 7 and 14 days, and subsequently transferred to Cr6+-free water for 14 days. After 14-day exposure, a dose-dependent increase of Cr6+ has been observed in most tissues. While after 14-day depuration, Cr6+ contents were significantly decreased in various tissues except in kidney and spleen where Cr6+ contents significantly increased at the group of 5 mg/L. Considering that Cr6+ highly accumulated in gills, intestines, liver and kidney, the oxidative damage of Cr6+ on the four tissues were further investigated and found that the antioxidant response to Cr6+ were organ-specific. Human movement, rehabilitation, and allied sciences have embraced their ambitions within the cycle of "RehabMove" congresses over the past 30years. This combination of disciplines and collaborations in the Netherlands has tried to provide answers to questions in the fields of rehabilitation and adapted sports, while simultaneously generating new questions and challenges. These research questions help us to further deepen our understanding of (impaired) human movement and functioning, with and without supportive technologies, and stress the importance of continued multidisciplinary (inter)national collaboration.
This position stand provides answers that were conceived by the authors in a creative process underlining the preparation of the 6th RehabMove Congress.
The take-home message of the RehabMove2018 Congress is a plea for continued multidisciplinary research in the fields of rehabilitation and adapted sports. This should be aimed at more individualized notions of human functioning, practice, and traand the quality there-off.
With this, we anticipate to support the development of rehabilitation sciences and technology and to stimulate the use of rehabilitation notions in general health care. We also hope to help ensure a stronger embodiment of preventive and lifestyle medicine in rehabilitation practice. Indeed, general health care and rehabilitation practice require a healthy and active lifestyle management and research agenda in the context of primary, secondary, and tertiary prevention. IMPLICATIONS FOR REHABILITATION Continued multidisciplinary (international) collaboration will stimulate the development of rehabilitation and human movement sciences. To map spasticity-related goals using the International Classification of Functioning, Disability and Health (ICF) linking rules, and review goal syntax to direct future goal setting.
1633 treatment goals, set during the TOWER study, were linked to the ICF framework and EQ-5D domains. Goals were mapped independently by two investigators with expertise in ICF linking rules.
In total, 1630 (99.8%) goals could be mapped to the ICF (inter-rater agreement on the main ICF category 96.1%). Most goals (80.2%) were mapped to a single ICF category and were related to activities/participation (54.9%). 170 (10.4%) goals were related to general tasks and activities, such as positioning, stretching, and strengthening. In total, 1072 goals (65.6%) mapped to the EQ-5D domains (inter-rater agreement 90.8%). Analysis of the goal syntax highlighted the need to include a verb in patient-centered goals to direct active behavior.

Action Observation Treatment is a novel rehabilitation approach exploiting a neurophysiological mechanism that allows one to recruit the neural structures sub-serving action execution during the mere observation of those same actions. Action Observation Treatment is effective in the rehabilitation of several neurological diseases. In this pilot study, we used Action Observation Treatment in a telerehabilitation setting in children with Cerebral Palsy.
Ten children with Cerebral Palsy, aged 5-12years, entered the study. They followed the Action Observation Treatment rehabilitation program at home with remote supervision by a child neurologist located at the hospital. Outcome measures were the scores at the Melbourne Assessment of Unilateral Upper Limb Function Scale and the Assisting Hand Assessment.
Scores obtained after treatment and at a two months' follow-up significantly differed from baseline and overlapped those obtained in randomized controlled studies carried out in a conventional setting.
Action Observation Treatment is therefore a promising approach that can be used on a large scale in a telerehabilitation setting. IMPLICATIONS FOR REHABILITATION Tele-rehabilitation has the potential to enhance early intervention service provision for children with Cerebral Palsy. The aim of this study was to explore the everyday life experiences and perspectives on challenges related to pregnancy, childbirth and parenthood when living with neuromuscular disorders (NMDs) and identify important elements to take into consideration for parents and their health professionals.
In all, 27 people (17 with a NMD and 10 partners without a NMD) were included in this study. The participants were all parents or expecting parents. Both mothers and fathers and ambulant and non-ambulant participants were included to broaden the perspective. Three types of semi-structured interviews were applied. The analysis was guided by the Interpretive Description methodology and Antonovsky's Sense of Coherence theory.
The findings illuminated insights to ongoing challenges in the everyday lives of parents with NMD.

Post-chemotherapy-induced cognitive dysfunction remains one of the challenges in cancer survivors. Cytokine-induced neurotoxicity manifests in subjects at any time after doxorubicin (DOX) chemotherapy. We examined the effect of bioactive Cordyceps militaris mycelia extract (CM) on the energy status, oxidative stress, and acetylcholinesterase activity in the brain of DOX treated rats. The CM (150 and 300 mg/kg b.w.) and DL-α lipoic acid (LA, 100 mg/kg b.w) were administered orally once daily for 5 days to male Wistar rats prior to the DOX administration (18 mg/kg as 3 doses of 6 mg/kg, i.p. b.w.) and continued for 6 more days. Cellular antioxidant status, Krebs cycle dehydrogenases, electron transport chain complexes (ETC) (I, III, and IV), adenosine triphosphate (ATP) level, advanced oxidation of protein products (AOPP), and acetylcholinesterase (AchE) activities were determined in the brain homogenate. The DOX alone treated group of animals showed significant decrease (p less then 0.05) of brain antioxidant levels, Krebs cycle dehydrogenases activities, ETC complex activities, and decreased ATP level, while lipid peroxidation and AOPP levels were elevated. CM at 300 mg/kg b.w. or LA at 100 mg/kg b.w. elevated antioxidant status, Krebs cycle dehydrogenases, and complex activities and thus alleviated the toxicity. CM also inhibited the AchE activity in brain. The experimental results thus reveal that CM possessed excellent capacity to attenuate oxidative stress, upregulate respiratory chain complex activity and ATP levels, as well as inhibition of AchE activity.To improve the production of mycelial biomass and intracellular polysaccharide (IPS), the culture medium composition and culture condition were respectively optimized by response surface methodology and one-factor-at-a-time methodology. The impact of fermentation process on the transcription of genes involved in IPS synthesis was evaluated using quantitative reverse transcription PCR (qRT-PCR). The yield of biomass and IPS were increased by 150% and 177% under the optimized fermentation process, respectively. Moreover, the optimization of fermentation process significantly increased the transcription of genes involved in IPS synthesis, especially pgm-1, ugp, and pgi. This is the first exploration of gene transcription profile for IPS synthesis of Auricularia auricula-judae under submerged fermentation. Samples were analyzed for antioxidant potential and the chemical composition of the major bioactive chemical components. Chromatographic procedures were used to analyze phenolic compounds, free amino acids, vitamins, and fatty acids. T. versicolor was analyzed for moisture (87.21 ± 1.08 g/100 g fw), protein (11.07 ± 0.85 g/100 g dw), and fat (1.35 ± 0.09 g/100 g dw) content. Phenolic compounds in T. versicolor were found as p-hydroxy benzoic (113.16 ± 0.22 μg/g dw), protocatechuic (10.07 ± 0.54 μg/g dw), vanillic (5.21 ± 0.10 μg/g dw), and homogentisic acids (1.24 ± 0.15 μg/g dw). In the studied mushroom, essential and nonessential amino acids were determined as leucine (72.41 ± 0.16 mg/100 g dw), isoleucine (60.07 ± 0.10 mg/100 g dw), methionine (53.51 ± 0.09 mg/100 g dw), tyrosine (33.37 ± 0.27 mg/100 g dw), glutamine (15.48 ± 0.19 mg/100 g dw), and asparagine (10.90 ± 0.13 mg/100 g dw). Vitamin content of T. versicolor was found as nicotinic acid (26.52 ± 0.10 mg/100 g dw) and nicotinamide (12.18 ± 0.05 mg/100 g dw). Fatty acids in T. versicolor were linoleic acid (182n6c), oleic acid (181n9c), palmitic acid (C160), stearic acid (C180), and linolenic acid (183n3). In conclusion, the results of the present study support the potential use of T. versicolor as a promising source of bioactive products for pharmaceutical and also food industries.This study was designed to understand the effect of extraction temperature, i.e., room temperature (GLRT), 50°C (GL50), 100°C (hot water; GL100), and 200°C (GL200) on antioxidant and biological activity of G. lucidum. The % yield obtained was 5.3%, 7.6%, 10.7%, and 13.2% at various extraction temperatures; room temperature, 50°C, 100°C and 200°C, respectively. Similarly, phenolic content (51.6, 57.9, 82.9, and 93.1 mg/g extract) and flavonoid content (18.8, 23.2, 34.3, and 36.3 mg/g extract) were observed to be increased with rise in extraction temperature. However, extraction temperature resulted in loss of antioxidant activities above 100°C as evident by chemical assays such as DPPH, FRAP, ABTS, and TRP conducted on extracts. In contrast, three bioactive compounds, i.e., adenine (3.26, 3.48, 2.16, and 1.45 mg/g extract), uracil (3.99, 3.21, 2.51, and 1.47 mg/g extract), and adenosine (5.92, 5.62, 2.22 and 0.7 mg/g extract), quantified by high performance thin layer chromatography showed decrease in their content with increasing extraction temperature.

In the studied mushroom, essential and nonessential amino acids were determined as leucine (72.41 ± 0.16 mg/100 g dw), isoleucine (60.07 ± 0.10 mg/100 g dw), methionine (53.51 ± 0.09 mg/100 g dw), tyrosine (33.37 ± 0.27 mg/100 g dw), glutamine (15.48 ± 0.19 mg/100 g dw), and asparagine (10.90 ± 0.13 mg/100 g dw). Vitamin content of T. versicolor was found as nicotinic acid (26.52 ± 0.10 mg/100 g dw) and nicotinamide (12.18 ± 0.05 mg/100 g dw). Fatty acids in T. versicolor were linoleic acid (182n6c), oleic acid (181n9c), palmitic acid (C160), stearic acid (C180), and linolenic acid (183n3). The aforementioned SBH's antimicrobial results and characterization study of its bacterial diversity are hoped to carve the pathway towards extending its probiotic ability into our everyday lives.Haze pollution has a serious impact on China's economic development and people's livelihood. We used data on PM2.5 concentration, industrial energy consumption structure, economic development and transportation in Beijing-Tianjin-Hebei and surrounding cities from 2000 to 2017, and analyzed the spatial effect of industrial energy consumption structure and traffic factors on haze pollution by using spatial autoregressive model (SAR) and spatial error model (SEM). The results indicated that (1) The global spatial correlation analysis showed that haze pollution had a significant positive spatial correlation, and the local spatial correlation analysis showed that the high-high clusters of PM2.5 were located in the south and middle of the region; (2) The change of industrial energy consumption structure was highly correlated with haze pollution, namely, the increase of industrial energy consumption led to the deterioration of environmental quality; (3) The change of economic development was highly correlated with haze pollution. There was no clear EKC relationship between haze pollution and economic development in Beijing-Tianjin-Hebei region and surrounding cities. MSM was tested in human keratinocytes, exhibiting a good cytotoxicity profile and anti-inflammatory activity by the reduction in IL-8 and TNF-α under LPS stimuli. A clinical study was performed to check product safety and efficacy against nail brittleness and alterations such as Beau's lines and onychorrhexis. A reduction in both alterations and in surface roughness without alteration of nail structure was observed, with a good level of patient acceptance and satisfaction.
Post-laminectomy syndrome is a common cause of dissatisfaction after endoscopic interlaminar approach. Our aim was to evaluate the efficacy and safety of our two newly designed instruments for laminotomy, a dural protector attached to the scope and a knot pusher for water-tight suturing of the incidental dural tears.
This was a multicenter evaluation. Efficacy was quantified as the pre-to-postoperative improvement in pain (visual analog scale), disability (Oswestry Disability Index), patient satisfaction (modified MacNab score), and length of hospital stay. Safety was quantified by the incidence and location of dural tears, rate of revision, and radiological outcomes. Outcomes were evaluated between the control (before instrument development) and experimental (after instrument development) groups.
There was a significant improvement in leg pain in the experimental group (
= 0.03), with greater patient satisfaction in the control group (
< 0.01). There was no incidence of dural tears in the area of the traversing and exiting nerve roots in the experimental group. Water-tightness of sutures was confirmed radiologically.

To investigate the potential effect of intracoronary administration of the glycoprotein IIb/IIIa inhibitor tirofiban on the microvascular obstruction (MVO) assessed by cardiac magnetic resonance (CMR) imaging compared to the intravenous route in patients with ST-segment-elevation myocardial infarction undergoing primary percutaneous coronary intervention (PCI). Two hundred eight patients were randomized into two groups (tirofiban i.v. and tirofiban i.c.). CMR was completed within 3-7 days after ST-segment-elevation myocardial infarction. One hundred thirty-two patients had a follow-up CMR at 6 months after discharge. The primary end point was the CMR measurements including myocardium strain, myocardial perfusion index, final infarct size, prevalence and extent of MVO, and the change of left ventricular end-diastolic volume (LVEDV) at six months follow-up. These roles are dictated by a number of complex factors including expression levels of other C/EBP family members and their stoichiometry relative to the isoform in question, binding site affinity, post-translational modifications, co-factor expression, and even hormone levels and lactogenic status. Here we summarize the historical work up to the latest findings in the field on C/EBPß in the mammary gland and in breast cancer. With the current emphasis on improving immunotherapy in breast cancer the role of specific C/EBPß isoforms in regulating specific chemokine and cytokine expression and the immune microenvironment will be of increasing interest.PURPOSE To evaluate the effect of vitamin D on severity of restless legs syndrome in patients with idiopathic restless legs syndrome (RLS). METHODS Patients with idiopathic RLS completed questionnaires including the International Restless Legs Severity Scale (IRLSS) and were evaluated for vitamin D deficiency. Patients with deficiency of vitamin D were treated with 50,000 units per week for 2 months. At the end of the 2 months, vitamin D levels were re-measured and disease severity was re-evaluated in patients who reached adequate vitamin D level. Subgroups of IRLSS questionnaire were also analyzed. RESULTS Of 35 patients enrolled, 21 (60%) had vitamin D deficiency and received vitamin D therapy. In 2 patients, vitamin D levels did not rise to sufficient levels with supplementation and these 2 patients were excluded from analysis. The remaining 19 patients showed vitamin D levels increased from 13.2 ± 4.0 to 42.8 ± 9.6 ng/mL while IRLSS improved from 24.9 ± 5.1 to 21.1 ± 2.9 points (p less then 0.001). Therefore, six candidate DNA barcode sequences (ITS, ITS2, psbA-trnH, matK, rbcL, and trnL-F) were tested for their capacity to identify Stephania species. The barcodes were analyzed either as a single region or in combination by tree-based [neighbor-joining (NJ) and Bayesian inference (BI)], distance-based (PWG-distance), and sequence similarity-based (TaxonDNA) methods. Amplification and sequencing success rates were 100% for all six candidate barcodes. A comparison of six barcode regions showed that ITS exhibited the highest number of variable and informative sites (182/179), followed by psbA-trnH (173/162). DNA barcoding gap assessment showed that interspecific distances of the six barcodes were greater than intraspecific distances. The identification results showed that species discrimination rates of combination barcodes were higher than those of single-region barcodes. Based on best match and best close match methods, the ITS+psbA-trnH combination exhibited the highest discrimination power (93.93%). Further, all Stephania species could be resolved in the phylogenetic trees based on ITS+psbA-trnH (NJ, BI). This study demonstrates that DNA barcoding is an efficient method to identify Stephania species and recommends that the ITS+psbA-trnH combination is the best DNA barcode for the identification of Stephania species.
