  • In the present work, different natural compounds from coffee by-product extracts (coffee silverskin and spent coffee) rich in polyphenols, was investigated against beauvericin (BEA) induced-cytotoxicity on SH-SY5Y cells. Spent coffee arise as waste products through the production of instant coffee and coffee brewing; while the silverskin is a tegument which is removed and eliminated with toasting coffee grains. First of all, polyphenol extraction methods, measurement of total polyphenols content and its identification were carried out. Afterwards evaluating in vitro effects with MTT assay on SH-SY5Y cells of coffee by-product extracts and mycotoxins at different concentrations and exposure times was performed. TPC in silverskin coffee by-product extracts was >10 times higher than in spent coffee by-product extracts. Chlorogenic acid was the majority polyphenol detected. Later, the erythroid colony-forming capacity of GDF-15-treated hematopoietic stem cells (HSCs) was evaluated by CFC assay. Any effect of elevated levels of GDF-15 on erythroid maturation was ultimately examined by expression analysis of erythroid-associated transcription factors and flow cytometry analysis of CD235a expression. GDF-15 mRNA expression increased during erythroid differentiation and also in β-thalassemia and MDS patients which was directly correlated with erythropoiesis severity. Treating the cells with high GDF-15 concentration (50 ng/ml) resulted in an approximate 30% decline in the capacity of erythroid colony formation of HSCs and CD235a positive cells. Additionally, erythroid-specific transcription factors showed significant down-regulation in the early stages of erythroid differentiation.     Western consumers interest in Eastern fermented foods has been growing, due to their nutritional and healthy properties. In this study, new sweet misos and salty misos were produced using grass pea (Lathyrus sativus L.) - traditional Portuguese legume from local producers - to promote its consumption and preservation. The evolution of the new misos was evaluated in comparison to traditional miso (made from soybean), through analysis of the chemical composition, colour, texture and linear viscoelastic behaviour.

    Throughout the fermentation process, the ascorbic acid and phenolic compounds content - with important nutritional value - increased in all misos, mainly in misos produced using grass pea, besides, grass pea sweet miso presented the fastest evolution and darkest colour. The texture parameters (firmness and adhesiveness) of misos decreased over time grass pea sweet miso showed the highest firmness reduction (51.63 N to 6.52 N) and soybean sweet miso the highest adhesiveness reduction (27.76 N to 3.11 N). Viscoelastic moduli also decreased, reflecting a reduction in the degree of internal structuring for all misos. However, grass pea misos presented more structured internal systems with faster maturation kinetics than soybean misos, for which stabilization started earlier.

    In the present work we propose the use of accelerating energies (microwaves and ultrasounds) to the maceration process of sherry vinegar with citrus fruit peels (orange and lemon). For the application of microwaves, an experimental design has been developed in order to optimize the maceration conditions. To evaluate the effect of these energies on the maceration, the volatile and polyphenolic content of the samples has been analyzed, as well as their sensory characteristics.

    Orange peel provided a higher number of volatile and polyphenolic compounds to the vinegar, while lemon's yielded a greater amount of some of them. The multivariate analysis showed that the samples macerated using microwaves were the most similar to the samples macerated in the traditional way. This aspect was corroborated by the sensory analysis, which was more noticeable when orange peel was used in the macerations.

    Therefore, it seems that the use of microwaves to accelerate maceration is a good alternative to the traditional method of making sherry vinegars macerated with citrus peel, since it cuts down the maceration time from 3 days to just a few minutes. © 2020 Society of Chemical Industry.

    Waxy maize (Zea mays L. sinensis Kulesh) is a good material for brewing. Waxy maize wine, a kind of Chinese rice wine, is strongly affected by a fermentation starter named Qu. In this study, an innovative mixed Qu, consisting of two yeasts and three molds, was produced and the raw-starch brewing method was applied in winemaking. Three other waxy maize wines fermented by three kinds of commercial Qu were also analyzed for comparison.

    Due to superb growth and fermentation characteristics, Saccharomyces cerevisiae CICC1009 and Pichia anomala CICC1851 were chosen to produce yeast Qu. The addition amount of yeast Qu was determined to be 30 g kg
    . In terms of chemical properties, mixed Qu was more suitable for making maize wine by the raw-starch brewing method than the three kinds of commercial Qu with which it was compared. The most influential components for the overall aroma profile in maize wines fermented by mixed Qu and Mifeng Qu were ethyl butyrate and β-damascenone, respectively, while in maize wines fermented by Angel Qu and Like Qu the most influential component was ethyl octanoate. Obvious differences were found among four maize wines regarding bitterness, umami, richness, saltiness, and sourness by the electronic tongue. The olfactory characteristics of maize wine fermented by Mifeng Qu were quite different from the other three according to the electronic nose.

  • The phenomenon of increasing accidents caused by reduced vigilance does exist. In the future, the high accuracy of vigilance estimation will play a significant role in public transportation safety. We propose a multimodal regression network that consists of multichannel deep autoencoders with subnetwork neurons (MCDAEsn). After we define two thresholds of ``0.35'' and ``0.70'' from the percentage of eye closure, the output values are in the continuous range of 0-0.35, 0.36-0.70, and 0.71-1 representing the awake state, the tired state, and the drowsy state, respectively. To verify the efficiency of our strategy, we first applied the proposed approach to a single modality. Then, for the multimodality, since the complementary information between forehead electrooculography and electroencephalography features, we found the performance of the proposed approach using features fusion significantly improved, demonstrating the effectiveness and efficiency of our method.Fuzzy-rough cognitive networks (FRCNs) are recurrent neural networks (RNNs) intended for structured classification purposes in which the problem is described by an explicit set of features. The advantage of this granular neural system relies on its transparency and simplicity while being competitive to state-of-the-art classifiers. Despite their relative empirical success in terms of prediction rates, there are limited studies on FRCNs' dynamic properties and how their building blocks contribute to the algorithm's performance. In this article, we theoretically study these issues and conclude that boundary and negative neurons always converge to a unique fixed-point attractor. Moreover, we demonstrate that negative neurons have no impact on the algorithm's performance and that the ranking of positive neurons is invariant. Moved by our theoretical findings, we propose two simpler fuzzy-rough classifiers that overcome the detected issues and maintain the competitive prediction rates of this classifier. Toward the end, we present a case study concerned with image classification, in which a convolutional neural network is coupled with one of the simpler models derived from the theoretical analysis of the FRCN model. The numerical simulations suggest that once the features have been extracted, our granular neural system performs as well as other RNNs.Recent advances in high-throughput single-cell technologies provide new opportunities for computational modeling of gene regulatory networks (GRNs) with an unprecedented amount of gene expression data. Current studies on the Boolean network (BN) modeling of GRNs mostly depend on bulk time-series data and focus on the synchronous update scheme due to its computational simplicity and tractability. However, such synchrony is a strong and rarely biologically realistic assumption. In this study, we adopt the asynchronous update scheme instead and propose a novel framework called SgpNet to infer asynchronous BNs from single-cell data by formulating it into a multiobjective optimization problem. SgpNet aims to find BNs that can match the asynchronous state transition graph (STG) extracted from single-cell data and retain the sparsity of GRNs. To search the huge solution space efficiently, we encode each Boolean function as a tree in genetic programming and evolve all functions of a network simultaneously via cooperative coevolution. In the future, the high accuracy of vigilance estimation will play a significant role in public transportation safety. We propose a multimodal regression network that consists of multichannel deep autoencoders with subnetwork neurons (MCDAEsn). After we define two thresholds of ``0.35'' and ``0.70'' from the percentage of eye closure, the output values are in the continuous range of 0-0.35, 0.36-0.70, and 0.71-1 representing the awake state, the tired state, and the drowsy state, respectively. To verify the efficiency of our strategy, we first applied the proposed approach to a single modality. Then, for the multimodality, since the complementary information between forehead electrooculography and electroencephalography features, we found the performance of the proposed approach using features fusion significantly improved, demonstrating the effectiveness and efficiency of our method.Fuzzy-rough cognitive networks (FRCNs) are recurrent neural networks (RNNs) intended for structured classification purposes in which the problem is described by an explicit set of features. The advantage of this granular neural system relies on its transparency and simplicity while being competitive to state-of-the-art classifiers. Despite their relative empirical success in terms of prediction rates, there are limited studies on FRCNs' dynamic properties and how their building blocks contribute to the algorithm's performance. In this article, we theoretically study these issues and conclude that boundary and negative neurons always converge to a unique fixed-point attractor. Moreover, we demonstrate that negative neurons have no impact on the algorithm's performance and that the ranking of positive neurons is invariant. Moved by our theoretical findings, we propose two simpler fuzzy-rough classifiers that overcome the detected issues and maintain the competitive prediction rates of this classifier. Toward the end, we present a case study concerned with image classification, in which a convolutional neural network is coupled with one of the simpler models derived from the theoretical analysis of the FRCN model.     Polyphenols may interact with protein via covalent bonds and non-covalent interactions, improving the structures and functional properties of the protein. The cross-linking between the polyphenol and protein is susceptible to salt (sodium chloride, NaCl) concentrations. Our study investigated the combined effects of quercetin (Q) and NaCl concentrations on wheat gliadin (G) structure and physicochemical properties.

    Q and NaCl addition resulted in a more compact protein microstructure. The improved foaming and emulsifying properties indicated that the modified G might be potent as a novel surface-active agent. Differential scanning calorimetry analysis indicated that Q protected the thermal stability from destruction at 50 and 200 mmol L
    NaCl concentrations, with narrower protein denaturation peaks. Fourier transform infrared and the Raman spectral analyses showed the secondary structural and microenvironmental changes of G. NaCl addition imparted a rearrangement of hydrogen bonds in the polypeptide chain and the disorder of protein structure, whereas Q enhanced the transition from β-sheets and random coils to α-helices and β-turns, forming a more ordered structure. The concentrations of these two medicines in ***** plasma were determined using high-performance liquid chromatographic-tandem mass spectrometry, and the data were analysed using a noncompartmental model with the WinNonlin software. After intravenous administration, both substances were absorbed rapidly and reached their effective therapeutic concentration quickly. CA was eliminated more slowly compared with AMX. Moreover, the distribution volume of AMX was larger than that of CA, suggesting that AMX could penetrate tissues better. After oral administration of the granular formulation, no significant difference was observed in the mean elimination half-life value between AMX and CA. The mean maximal plasma concentrations of AMX and CA, reached after 1.14 and 1.32 hr, were 2.58 and 1.91 μg/m, respectively. The mean oral bioavailability of AMX and CA was 23.6% and 26.4%, respectively. After oral administration, the T>MIC50 for three common respiratory pathogens was over 6.12 hr.     Four-dimensional cone-beam computed tomography (4D CBCT) imaging has been suggested as a solution to account for interfraction motion variability of moving targets like lung and liver during radiotherapy (RT) of moving targets. However, due to severe sparse view sampling artifacts, current 4D CBCT data lack sufficient image quality for accurate motion quantification. In the present paper, we introduce a deep learning-based framework for boosting the image quality of 4D CBCT image data that can be combined with any CBCT reconstruction approach and clinical 4D CBCT workflow.

    Boosting is achieved by learning the relationship between so-called sparse view pseudo-time-average CBCT images obtained by a projection selection scheme introduced to mimic phase image sparse view artifact characteristics and corresponding time-average CBCT images obtained by full view reconstruction. The employed convolutional neural network architecture is the residual dense network (RDN). The underlying hypothesis is that the RDN learns the appearance of the streaking artifacts that is typical for 4D CBCT phase images - and removes them without influencing the anatomical image information.

    After identifying 2018's 20 best-selling US video games and selecting for each a YouTube video of all cut scenes and bridging game play, we coded each video for the presence of five types of tobacco imagery. We also recorded the length of time tobacco content was visible during the video.

    Seven of the top 20 video games contained tobacco imagery, which comprised between 7 seconds and 38 minutes of game play. All types of tobacco imagery were accounted for visible tobacco paraphernalia, tobacco products used to further game play, background characters using tobacco products, and main characters (playable and nonplayble) using tobacco products. Visible tobacco paraphernalia was the most common type of tobacco imageink between exposure to tobacco marketing in media such as films and subsequent smoking behavior, policies to minimize tobacco imagery in video games that exceed the scant protections offered by the Master Settlement Agreement could help reduce smoking prevalence among youth.

    Previous research has found that male users of androgens are diagnosed approximately twice as often with infertility. We therefore set out to investigate the fertility in men using androgens.

    The study included 545 males who tested positive for androgens in an anti-doping test program in Danish fitness centers during the period from January 3, 2006, to March 1, 2018. The confirmed androgen users were matched by birth year with 5450 male controls. We followed this cohort from 10 years prior to testing positive until the end of follow-up in May 2018.

    During the 10-year period prior to testing positive, the group of androgen users experienced a 26% lower fertility rate than the controls (rate ratio [RR] 0.74; 95% CI, 0.60-0.90; P = 0.0028). However, in the years following the doping sanction, they made a significant catch-up, and at completed follow-up the total fertility rate was only 7% lower than expected (RR 0.93, 95% CI, 0.84-1.03). The prevalence of assisted reproduction was 5.69% in the group of androgen users and 5.28% in the control group (P = 0.69).

    Androgen use was associated with a temporary decline in fertility and most androgen users achieved parenthood without any help from the health care system. CNSs exist in many eukaryotes and are assumed to be involved in protein expression control. Version 1 of dbCNS, introduced here, includes a powerful and precise CNS identification pipeline for multiple vertebrate genomes. Mutations in CNSs may induce morphological changes and cause genetic diseases. For this reason, many vertebrate CNSs have been identified, with special reference to primate genomes. We integrated ∼6.9 million CNSs from many vertebrate genomes into dbCNS, which allows users to extract CNSs near genes of interest using keyword searches. In addition to CNSs, dbCNS contains published genome sequences of 161 species. With purposeful taxonomic sampling of genomes, users can employ CNSs as queries to reconstruct CNS alignments and phylogenetic trees, to evaluate CNS modifications, acquisitions, and losses, and to roughly identify species with CNSs having accelerated substitution rates. dbCNS also produces links to dbSNP for searching pathogenic single-nucleotide polymorphisms in human CNSs.     Telehealth (TH) practices among pediatric infectious disease specialists prior to the coronavirus disease 2019 (COVID-19) pandemic are largely unknown.

    In 2019, the Pediatric Infectious Diseases Society (PIDS) Telehealth Working Group surveyed PIDS members to collect data on the use of TH modalities, adoption barriers, interest, extent of curbside consultations (CC), and reimbursement.

    Of 1,213 PIDS members, 161 (13.3%) completed the survey, and the responses of 154 (12.7%) from the US were included in our report. Medical school (63.6%) and hospital (44.8%) were the commonest work settings with 16.9% practicing in both of them. The most common TH modalities used were synchronous provider-patient virtual visits (20.8%) and synchronous provider-provider consultations (13.6%). TH services included outpatient consultations (48.1%), vaccine recommendations (43.5%), inpatient consultations (39.6%) and travel advice (39.6%). The prevalence of assisted reproduction was 5.69% in the group of androgen users and 5.28% in the control group (P = 0.69). CNSs exist in many eukaryotes and are assumed to be involved in protein expression control. Version 1 of dbCNS, introduced here, includes a powerful and precise CNS identification pipeline for multiple vertebrate genomes. Mutations in CNSs may induce morphological changes and cause genetic diseases. For this reason, many vertebrate CNSs have been identified, with special reference to primate genomes. We integrated ∼6.9 million CNSs from many vertebrate genomes into dbCNS, which allows users to extract CNSs near genes of interest using keyword searches. In addition to CNSs, dbCNS contains published genome sequences of 161 species. With purposeful taxonomic sampling of genomes, users can employ CNSs as queries to reconstruct CNS alignments and phylogenetic trees, to evaluate CNS modifications, acquisitions, and losses, and to roughly identify species with CNSs having accelerated substitution rates. dbCNS also produces links to dbSNP for searching pathogenic single-nucleotide polymorphisms in human CNSs. Thus, dbCNS connects morphological changes with genetic diseases. A test analysis using 38 gnathostome genomes was accomplished within 30 s. dbCNS results can evaluate CNSs identified by other stand-alone programs using genome-scale data. Telehealth (TH) practices among pediatric infectious disease specialists prior to the coronavirus disease 2019 (COVID-19) pandemic are largely unknown. In 2019, the Pediatric Infectious Diseases Society (PIDS) Telehealth Working Group surveyed PIDS members to collect data on the use of TH modalities, adoption barriers, interest, extent of curbside consultations (CC), and reimbursement. Of 1,213 PIDS members, 161 (13.3%) completed the survey, and the responses of 154 (12.7%) from the US were included in our report. Medical school (63.6%) and hospital (44.8%) were the commonest work settings with 16.9% practicing in both of them. The most common TH modalities used were synchronous provider-patient virtual visits (20.8%) and synchronous provider-provider consultations (13.6%). TH services included outpatient consultations (48.1%), vaccine recommendations (43.5%), inpatient consultations (39.6%) and travel advice (39.6%). The VIRTUS project has provided the impetus to objectively demonstrate the anatomical coverage provided by side plates. METHOD CT scans of 120 male UK Armed Forces personnel were analysed to ascertain the vertical distance between the anterior axillary fold and iliac crest, and horizontal distance between anterior and posterior borders of the liver, delineating the boundaries of essential medical coverage from the side aspects. The percentage of shot-lines intersected by the existing Enhanced Combat Body Armour (ECBA) plates as well as an optimised plate based on the maximum potential dimensions of essential coverage was determined in the Coverage of Armour Tool. RESULTS ECBA plates were 101 mm shorter and 4 mm narrower than a plate with dimensions providing essential medical coverage for the 50th percentile subject (157×315 mm). Coverage increased by 35% when using two ECBA plates as side coverage in addition to using the front and rear OSPREY plates in the VIRTUS vest. Two side plates with dimensions providing essential medical coverage for the 50th percentile increased anatomical coverage by a further 16%. CONCLUSIONS This analysis has provided strong evidence that ECBA plates are already optimised for side protection, despite not being originally designed for this purpose. They are correctly positioned within the VIRTUS soft body armour vest and the width of the ECBA plate is only 3% less than what would be optimum size for the 50th percentile. The activities of these P450s were determined by the formation of metabolites from probe drugs for each P450 using UPLC-QTOFMS. DILI was induced at mild to severe levels in a dose-dependent manner in 200, 300, and 400 mg/kg APAP treated groups at child and adult ages, but not at the infant age. Significantly decreased expression at mRNA and protein levels as well as enzymatic activities of CYP2E1, 3A11, 1A2, and 2C29 was found at child and adult ages. Adult male **** were more susceptible to ALIL than female **** with more decreased expression of P450s. These results implicate that altered levels of P450s in severely injured livers caused by drugs may affect the therapeutic efficacy of drugs, which are metabolized by P450s, more particularly for males.   • Targeted therapies in the treatment of head and neck squamous cell carcinoma (HNSCC) are subject to extensive research. Different mutations of genes belonging to the fibroblast growth factor (FGF) family have been detected in HNSCC. In this study, we examined the expression of FGF1 and FGF2 after treatment with small-molecule tyrosine kinase inhibitors (TKIs) and an inhibitor of mechanistic target of rapamycin (mTOR) in vitro using human papillomavirus (HPV)-positive and -negative SCC lines.

    Cells of two human HPV-negative cell lines (UMSCC-11A/-14C) and one HPV-positive cell line (CERV196) were incubated with 20 μmol/l of erlotinib, gefitinib, nilotinib, dasatinib, or everolimus for 24-96 h.     Since pathways involving LRRC17 are related to the survival of patients with various cancers, we analyzed LRRC17 as a prognostic gene in serous ovarian cancer.

    Data were collected from Gene Expression Omnibus (GSE9891, GSE13876, and GSE26712) and The Cancer Genome Atlas (TCGA). We performed survival analyses using C statistics, area under the curve, survival plot with optimal cutoff level, and cox proportional regression. Zebrafish embryos were used as an in vivo model.

Multivariate regression analysis showed that LRRC17 expression, age, and stage were independently related with survival.   • Two different deep learning networks are used for human action recognition and contact detection, which in combination, are expected to lead to the enhancement of human safety and an increase in the level of cobot perception about human intentions. The results show a promising path for future AI-driven solutions in safe and productive human-robot collaboration (HRC) in industrial automation.Trichogramma spp. is an important egg parasitoid wasp for biocontrol of agriculture and forestry insect pests. Trehalose serves as an energy source or stress protectant for insects. To study the potential role of trehalose in cold resistance on an egg parasitoid, cDNA for trehalose-6-phosphate synthase (TPS) and soluble trehalase (TRE) from Trichogramma dendrolimi were cloned and characterized. Gene expressions and enzyme activities of TdTPS and TdTRE were determined in larvae, prepupae, pupae, and adults at sustained low temperatures, 13 °C and 16 °C. TdTPS and TdTRE expressions had similar patterns with higher levels in prepupae at 13 °C and 16 °C. TdTPS enzyme activities increased with a decrease of temperature, and TdTRE activity in prepupae decreased sharply at these two low temperatures. In vitro reared T. dendrolimi could complete entire development above 13 °C, and the development period was prolonged without cold injury. Results indicated trehalose might regulate growth and the metabolic process of cold tolerance. Moreover, 13 °C is the cold tolerance threshold temperature and the prepupal stage is a critical developmental period for in vitro reared T. dendrolimi. These findings identify a low cost, prolonged development rearing method, and the cold tolerance for T. dendrolimi, which will facilitate improved mass rearing methods for biocontrol.There is a lack of prediction markers for early diabetic nephropathy (DN) in patients with type 2 diabetes mellitus (T2DM). Sensitivity, specificity, and other estimates of accuracy were calculated using the bivariate random effects model. The hierarchical summary receiver operating characteristic curve hsROC) was used to pool data and evaluate the area under curve (AUC). The sources of heterogeneity were explored by sensitivity analysis. Publication bias was assessed using Deek's test. The meta-analysis enrolled 14 studies involving 598 healthy individuals, 765 T2DM patients with normoalbuminuria, 549 T2DM patients with microalbuminuria, and 551 T2DM patients with macroalbuminuria, in total for both biomarkers. The AUC of uKIM-1 and YKL-40 for T2DM patients with normoalbuminuria, was 0.85 (95%CI; 0.82-0.88) and 0.91 (95%CI; 0.88-0.93), respectively. The results of this meta-analysis suggest that both uKIM-1 and YKL-40 can be considered as valuable biomarkers for the early detection of DN in T2DM patients with the latter showing slightly better performance than the former.Malaria control relies heavily on the small number of existing antimalarial drugs.   • Objective We performed a meta-analysis to quantify the overall incidence and risk of proteinuria associated with five newly approved VEGFR-TKIs (regorafenib, vandetanib, cabozantinib, lenvatinib, axitinib) in cancer patients.Methods Pubmed, Embase, ASCO abstracts, and ESMO abstracts were searched to identify relevant studies. Overall incidence rates, relative risk (RR), and 95% confidence intervals (CI) were estimated using random or fixed effects models according to the heterogeneity of included studies.Results A total of 9,446 patients from 20 RCTs were included for the meta-analysis. The use of newly approved VEGFR-TKIs was associated with an increased risk of all-grade (RR 2.35, 95% CI 1.69-3.27, P less then  0.001) and high-grade (RR 3.70, 95% CI 2.09-6.54, P less then  0.001) proteinuria. On subgroup analysis, lenvatinib, axitinib, and vandetanib significantly increased the risk of all-grade proteinuria, and lenvatinib was associated with an increased risk of high-grade proteinuria. Analyses of previously published recognition data elucidated this finding by showing that confidence is not affected by cue validity for items recognized as "old" regardless of accuracy (i.e., hits and false alarms). However, confidence is affected by cue validity for items judged "new" regardless of accuracy (i.e., correct rejections and misses). Both children and adults demonstrated a polysemy over ambiguity learning advantage on each task after exposure, showing better learning of novel words with multiple related meanings than novel words with unrelated meanings. Stimuli in the polysemy condition were designed and then normed to guard against learners relying on a simple definition to distinguish the multiple target meanings for each word from foils. We retested available participants after a week-long delay without providing additional exposure and found that adults' performance remained strong in the polysemy condition in 1 task, and children's performance remained strong in the polysemy condition in both tasks.
