  • Non-linear mixed effects modeling was carried out to characterize dacomitinib pharmacokinetics in the presence and absence of the concomitant use of a PPI, rabeprazole. Several absorption models, some more empirical, and some more physiologically based, were tested transit compartment, first-order absorption with and without lag time, and variations of combined zero- and first-order absorption kinetics models. RESULTS The presence of a PPI was a significant covariate affecting the extent (F) and rate (ka) of dacomitinib absorption, as previously reported in the dedicated clinical study. In the era of personalized medicine, the prospective centralized collection of high-quality biological material, expert pathological revision, and association to well-controlled clinical data are important or even essential added values to clinical trials. Here, we present the academic experience of the MITO (Multicenter Italian Trial in Ovarian Cancer) group, including gynecologists, pathologists, oncologists, biostatisticians, and translational researchers, whose effort is dedicated to the care and basic/translational research of gynecologic cancer. In our ten years of experience, we have been able to collect and process, for translational analyses, formalin-fixed, paraffin-embedded blocks from more than one thousand ovarian cancer patients. Standard operating procedures for collection, shipping, and processing were developed and made available to MITO researchers through the coordinating center's web-based platform. Clinical data were collected through dedicated electronic case report forms hosted in a web-based electronic platform and stored in a central database at the trial's coordinating center, which performed all the analyses related to the proposed translational researches. During this time, we improved our strategies of block management from retrospective to prospective collection, up to the design of a prospective collection with a quality check for sample eligibility before patients' accrual. The final aim of our work is to share our experience by suggesting a guideline for the process of centralized collection, revision processing, and storing of formalin-fixed, paraffin-embedded blocks for translational purposes.Maize plants are often infected with fungal pathogens of the genus Fusarium. HRM analysis thus offers a cost-efficient method suitable for the diagnosis of multiple fungal pathogens.A joint initiative between community and public health stakeholders in a low-income Canadian city was created to describe the developmental profiles of children aged 2-5 years. A two-phase, cross-sectional design was used. Children's development was assessed using an online screening procedure. Those at risk of delays were invited for a school-readiness face-to-face brief assessment. Descriptive and exploratory analyses were conducted. In Phase 1, 223 families were screened; 100 children were at risk of delays (45%); 13% were at risk in ≥3 developmental domains; 26% were at risk in the fine motor domain. Risk of delay was associated with parental concerns, accessing more healthcare professionals, and using fewer public health/community programs. Lower incomes, and not attending day care showed trends towards an increased risk of delay. In Phase 2, 49 children were assessed; 69% were at risk of school-readiness delays; 22% had potential motor delays; 37% were at risk in the social domain. This study found a higher proportion of children at risk of delay than typically reported. Creating community partnerships could help identify all children needing developmental and school-readiness support. More research is needed to ensure these community-based partnerships are integrated into health/community programs responding to children's needs and parental concerns.The primary means for electronic position fixing currently in use in majority of contemporary merchant ships are shipborne GPS (Global Positioning System) receivers or DGPS (Differential GPS) and IALA (International Association of Lighthouse Authorities) radio beacon receivers. More advanced GNSS (Global Navigation Satellite System) receivers able to process signals from GPS, Russian GLONASS, Chinese Beidou, European Galileo, Indian IRNSS, Japan QZSS, and satellite-based augmentation systems (SBAS), are still relatively rare in maritime domain. However, it is expected that such combined or multi-system receivers will soon become more common in maritime transport and integrated with gyro, inertial, radar, laser, and optical sensors, and they will become indispensable onboard maritime autonomous surface ships (MASS). To be prepared for a malfunction of any position sensors, their state-of-the-art integrity monitoring should be developed and standardized, taking into account the specificity of MASS and e-navigation safety.

  • Results indicated that response inhibition performance was significantly worse during faster speeds of play, and that the role of arousal in this relationship was independent of any motor priming affect. The implications of these findings for gambling legislation and gambling harm-minimisation approaches are discussed.Differences in guiding catheters (GCs) manipulations and selections among different access sites are currently unclear. We examined the differences in the routes of GCs for the left coronary artery (LCA) among the right radial, left radial, and femoral approaches. We used a combined angiography-computed tomography (CT) system that enabled to perform CT scans during percutaneous coronary intervention (PCI). We enrolled 88 patients who underwent CT scans during LCA PCI or percutaneous transluminal septal myocardial ablation. To evaluate the route of GCs, we analyzed the positions of the catheter's contact point on the contralateral aortic wall to the LCA ostium, which were expressed by the angle formed by the vertical line and the diagonal line from the GC shaft to the center of the aorta. The procedures were performed via the right radial in 47 cases, left radial in 20, and femoral approach in 21. The positions of the catheter's contact point were significantly different depending on the approaches (interquartile range - 3.7 [- 14.3 to 7.8], - 46.5 [- 76.9 to - 9.3], and - 30.7 [- 39.4 to - 22.4] degrees, respectively; p less then 0.001). Multivariate analysis demonstrated that access sites and LCA ostium locations had significant impacts on the positions of the catheter's contact point. The routes of LCA GCs were different among the right radial, left radial, and femoral approaches.Dendritic spines are tiny membrane specialization forming the postsynaptic part of most excitatory synapses. In this paper, I discuss the ecology and physiology of gut bacteria associated with insect herbivores and how they may shape interactions between insects and their various host plants. I first establish how microbial associations vary between insects with different feeding styles, and how the insect host physiology and ecology can shape stable or transient relationships with gut bacteria. Then, I describe how these relationships factor in with plant nutrition and plant defenses. Within this framework, I suggest that many of the interactions between plants, insects, and the gut microbiome are context-dependent and shaped by the type of defense and the isolates present in the environment. Relationships between insects and plants are not pairwise, but instead highly multipartite, and the interweaving of complex microbial interactions is needed to fully explore the context-dependent aspects of the gut microbiome in many of these systems. I conclude the review by suggesting studies that would help reduce the unsureness of microbial interactions with less-defined herbivore systems and identify how each could provide a path to more robust roles and traits.Selenium (Se) is an essential trace element for both humans and animals. Se deficiency leads to myocardial injury, reproductive disorder, increased exudation, inflammatory injury, and other diseases. The present study analyzed the relationships of Se deficiency, inflammation, and smooth muscle contraction in the small intestine, which is the main tissue that digests and absorbs Se. The model was established by feeding the animals diets with different concentrations of Se. The results showed that the dietary Se content was positively correlated with the blood Se concentration and the intestinal Se concentration. ROS and MPO activity increased with the lack of Se. TNF-α, IL-1β, and IL-6 expression was increased at both the mRNA and protein levels with Se deficiency. The pathways tested showed that the IκBα, NF-κB p65, p38, ERK, and JNK phosphorylation levels were significantly increased with the lack of Se. Moreover, the contractility analysis confirmed that contraction of the intestinal smooth muscle was attenuated by Se deficiency, as shown by the MedLab data acquisition system.

This paper aims to assess and quantify the healing performance rates (HR) of half-warm recycled asphalt (HWRA) mixtures containing electric arc furnace (EAF) **** and total recycled asphalt pavement (RAP) rates. To this end, a novel assisted thermomechanical healing treatment (i.e., a recompaction-based technique and microwave heating energy) was put forward to promote the potential healing effect of this treatment on the mechanical properties of the asphalt mixtures. In order to do this, three microwave heating temperatures (25 °C, 60 °C, and 80 °C) and three mechanical recompaction levels (0, 25, and 50 gyrations) were selected. After that, the healing performance rates (%, HR) of the asphalt mixtures were calculated by repeated indirect tensile strength (ITS) and indirect tensile stiffness modulus (ITSM). The results indicated that the 8% EAF **** mixture was found to provide significant microwave heating energy savings by up to 69% compared with the benchmark 100% RAP mixture, and, at the same time, it experienced a remarkable stiffness recovery response of 140% of the initial mechanical properties. These findings encourage greater confidence in promoting this innovative thermomechanical-based healing treatment for in-situ surface course asphalt mixtures of road pavements.Kidney transplantation is a well-established therapy for patients with end-stage renal disease. Heme-oxygenase (HO) is the rate-limiting enzyme in heme catabolism, converting heme to iron, carbon monoxide, and biliverdin. Heme-oxygenase overexpression may be observed in all phases of transplant processes, including brain death, recipient management, and acute and chronic rejection. HO induction has been proved to provide a significant reduction of inflammatory response and a reduction of ischemia and reperfusion injury in organ transplantation, as well as providing a reduction of incidence of acute rejection. In this review, we will summarize data on HO and kidney transplantation, suggesting possible clinical applications in the near future to improve the long-term outcomes.Fiber optic sensors represent one of the most promising technologies for the monitoring of various engineering structures. A major challenge in the field is to analyze and predict the strain transfer to the fiber core reliably. Many authors developed analytical models of a coated optical fiber, assuming null strain at the ends of the bonding length. Gastric cancer (GC) is the fourth commonest cause of cancer worldwide and is the second leading cause of cancer-related death annually. Although the number of new diagnoses has declined over recent decades, prognosis remains poor, with only 15% surviving to five years. Geographical differences in clinicopathological features are also apparent, with epidemiological and genetic studies revealing GC to be a highly heterogeneous disease with phenotypic diversity as a result of etiological factors. The molecular heterogeneity associated with GC dictates that a single 'one size fits all' approach to management is unlikely to be successful. Wnt pathway dysregulation has been observed in approximately 50% of GC tumors and may offer a novel therapeutic target for patients who would otherwise have a poor outcome. This mini review will highlight some recent discoveries involving Wnt signaling in GC.Previously, we showed the preventive effects of Lactobacillus plantarum ZS2058 (ZS2058) on Salmonella infection in murine models. In this work, we found that eugenol has a selective antibacterial effect, which inhibited Salmonella more than probiotics ZS2058 in vitro. It suggested a synergistic effect of them beyond their individual anti-Salmonella activity. We verified the conjecture in murine models. The results showed that the combination of ZS2058 and eugenol (CLPZE) significantly increased (p = 0.026) the survival rate of Salmonella-infected **** from 60% to 80% and the effect of CLPZE on preventing Salmonella-infection was 2-fold that of ZS2058 alone and 6-fold that of eugenol alone. CLPZE had a synergistic effect on inhibiting ST growth (the coefficient drug interaction ((CDI) = 0.829), reducing its invasiveness (CDI = 0.373) and downregulating virulence genes' expression in vitro. CLPZE helped the host form a healthier gut ecosystem. CLPZE also elicited a stronger and earlier immune response to systemic infection.

This paper aims to assess and quantify the healing performance rates (HR) of half-warm recycled asphalt (HWRA) mixtures containing electric arc furnace (EAF) slag and total recycled asphalt pavement (RAP) rates. To this end, a novel assisted thermomechanical healing treatment (i.e., a recompaction-based technique and microwave heating energy) was put forward to promote the potential healing effect of this treatment on the mechanical properties of the asphalt mixtures. In order to do this, three microwave heating temperatures (25 °C, 60 °C, and 80 °C) and three mechanical recompaction levels (0, 25, and 50 gyrations) were selected. After that, the healing performance rates (%, HR) of the asphalt mixtures were calculated by repeated indirect tensile strength (ITS) and indirect tensile stiffness modulus (ITSM). The results indicated that the 8% EAF slag mixture was found to provide significant microwave heating energy savings by up to 69% compared with the benchmark 100% RAP mixture, and, at the same time, it experienced a remarkable stiffness recovery response of 140% of the initial mechanical properties. Heme-oxygenase overexpression may be observed in all phases of transplant processes, including brain death, recipient management, and acute and chronic rejection. HO induction has been proved to provide a significant reduction of inflammatory response and a reduction of ischemia and reperfusion injury in organ transplantation, as well as providing a reduction of incidence of acute rejection. In this review, we will summarize data on HO and kidney transplantation, suggesting possible clinical applications in the near future to improve the long-term outcomes.Fiber optic sensors represent one of the most promising technologies for the monitoring of various engineering structures.   • BACKGROUND After accounting for known risk factors for CKD progression in children, clinical outcomes among children with CKD still vary substantially. Biomarkers of tubular injury (such as KIM-1), repair (such as YKL-40), or inflammation (such as MCP-1, suPAR, TNF receptor-1 [TNFR-1], and TNFR-2) may identify children with CKD at risk for GFR decline. METHODS We investigated whether plasma KIM-1, YKL-40, MCP-1, suPAR, TNFR-1, and TNFR-2 are associated with GFR decline in children with CKD and in subgroups defined by glomerular versus nonglomerular cause of CKD. We studied participants of the prospective CKiD Cohort Study which enrolled children with an eGFR of 30-90 ml/min per 1.73 m2 and then assessed eGFR annually. Biomarkers were measured in plasma collected 5 months after study enrollment. The primary endpoint was CKD progression, defined as a composite of a 50% decline in eGFR or incident ESKD. RESULTS Of the 651 children evaluated (median age 11 years; median baseline eGFR of 53 ml/min per 1.73 m2), 195 (30%) had a glomerular cause of CKD. Over a median follow-up of 5.7 years, 223 children (34%) experienced CKD progression to the composite endpoint. After multivariable adjustment, children with a plasma KIM-1, TNFR-1, or TNFR-2 concentration in the highest quartile were at significantly higher risk of CKD progression compared with children with a concentration for the respective biomarker in the lowest quartile (a 4-fold higher risk for KIM-1 and TNFR-1 and a 2-fold higher risk for TNFR-2). Plasma MCP-1, suPAR, and YKL-40 were not independently associated with progression. When stratified by glomerular versus nonglomerular etiology of CKD, effect estimates did not differ significantly. CONCLUSIONS Higher plasma KIM-1, TNFR-1, and TNFR-2 are independently associated with CKD progression in children. Even though detailed information on the genetic aberrations in cell signaling and cell cycle checkpoint control is available, no effective targeted treatment has been developed. Despite the advanced molecular defects, glioblastoma cells may have remnants of normal growth inhibitory pathways, such as the bone morphogenetic protein (BMP) signaling pathway. We have evaluated the growth inhibitory effect of BMP4 across a broad spectrum of patient samples, using a panel of 40 human glioblastoma initiating cell (GIC) cultures. A wide range of responsiveness was observed. BMP4 sensitivity was positively correlated with a proneural mRNA expression profile, high SOX2 activity, and BMP4-dependent up-regulation of genes associated with inhibition of the MAPK pathway, as demonstrated by gene set enrichment analysis (GSEA). BMP4 response in sensitive cells was mediated by the canonical BMP receptor pathway involving SMAD1/5/9 phosphorylation and SMAD4 expression. SOX2 was consistently down-regulated in BMP4-treated cells. Forced expression of SOX2 attenuated the BMP4 sensitivity including a reduced up-regulation of MAPK inhibitory genes, implying a functional relationship between SOX2 down-regulation and sensitivity. The results show an extensive heterogeneity in BMP4 responsiveness among GICs, and identify a BMP4 sensitive subgroup, in which SOX2 is a mediator of the response. Implications Development of agonists targeting the BMP signaling pathway in glioblastoma remains an attractive avenue towards a better treatment - our study may help find biomarkers that predict the outcome of such treatment and help stratification of patients. In all of these incurable patients, de novo lethal cancer evolves capacities for both metastasis and resistance. Therefore, cancers in different patients appear to follow the same eco-evolutionary path that independently manifests in affected patients. This convergent outcome that always includes the ability to metastasize and exhibit resistance demands an explanation beyond the slow and steady accrual of stochastic mutations. The common denominator may be that cancer starts as a speciation event when a unicellular protist breaks away from its multicellular host and initiates a cancer clade within the patient. As the cancer cells speciate and diversify further, some evolve the capacity to evolve evolvability. Evolvability becomes a heritable trait that influences the available variation of other phenotypes that can then be acted upon by natural selection. Although previous reports suggest the contribution of tissue factor and podoplanin, the mechanistic linkage between VTE and bladder cancer cell derived molecules is unknown. Therefore, we compared distinct procoagulant pathways in four different cell lines. In vitro findings were further confirmed by microfluidic experiments mimicking the pathophysiology of tumor blood vessels and in tissue samples of bladder cancer patients by transcriptome analysis and immunohistology. In vitro and microfluidic experiments identified bladder cancer derived vascular endothelial growth factor A (VEGF-A) as highly procoagulant because it promoted the release of von Willebrand factor (VWF) from endothelial cells and thus platelet aggregation. In tissue sections from bladder cancer patients, we found that VWF mediated blood vessel occlusions were associated with a poor outcome.

  • Anabolic steroids (AS) are synthetic testosterone-derivatives developed by the pharmaceutical industry to mimic testosterone biological effects. So far, AS have been implicated in the treatment of pathological conditions, such as hypogonadism, anemia, and cachexia. Since their discovery, though, AS have been illicitly used by elite and recreational athletes, bodybuilders and weightlifters in order to enhance athletic and aesthetic performance. This practice is characterized by cycles of administration and withdrawal, the combination of different AS compounds, and administration of doses 50 - 1000 times higher than those recommended for therapeutic purposes. AS excess has been correlated to cardiovascular detrimental effects, including cardiac hypertrophy, arrhythmias, and hypertension. Particularly, acute myocardial infarction (AMI) has been extensively reported by clinical and post-mortem studies. Atherosclerosis, hypercoagulability state, increased thrombogenesis and vasospasm have arisen as potential causes of myocardial ischemia in AS users. Additionally, several experimental reports have demonstrated that AS can increase the susceptibility to cardiac ischemia/reperfusion injury, whereas the cardioprotection elicited by physical exercise and ischemic postconditioning is blunted. Altogether, these factors can contribute to increased AMI morbidity and mortality during AS excess, particularly when AS are combined with other compounds, such as thyroid hormones, growth hormones, insulin, and diuretics.NF-κB is a transcription factor that activates super enhancers (SEs) and typical enhancers (TEs) and triggers threshold and graded gene expression, respectively. However, the mechanisms by which NF-κB selectively participates in these enhancers remain unclear. Here we show using mouse primary B lymphocytes that SE activity simultaneously associates with chromatin opening and enriched NF-κB binding, resulting in a higher fold change and threshold expression upon B cell receptor (BCR) activation. The higher fold change results from longer DNA, whereas the threshold response is explained by synergy in DNA-NF-κB binding and is supported by the coexistence of PU.1 and NF-κB in a SE before cell stimulation. So far, AS have been implicated in the treatment of pathological conditions, such as hypogonadism, anemia, and cachexia. Since their discovery, though, AS have been illicitly used by elite and recreational athletes, bodybuilders and weightlifters in order to enhance athletic and aesthetic performance. This practice is characterized by cycles of administration and withdrawal, the combination of different AS compounds, and administration of doses 50 - 1000 times higher than those recommended for therapeutic purposes. AS excess has been correlated to cardiovascular detrimental effects, including cardiac hypertrophy, arrhythmias, and hypertension. Particularly, acute myocardial infarction (AMI) has been extensively reported by clinical and post-mortem studies. Atherosclerosis, hypercoagulability state, increased thrombogenesis and vasospasm have arisen as potential causes of myocardial ischemia in AS users. Additionally, several experimental reports have demonstrated that AS can increase the susceptibility to cardiac ischemia/reperfusion injury, whereas the cardioprotection elicited by physical exercise and ischemic postconditioning is blunted. Altogether, these factors can contribute to increased AMI morbidity and mortality during AS excess, particularly when AS are combined with other compounds, such as thyroid hormones, growth hormones, insulin, and diuretics.NF-κB is a transcription factor that activates super enhancers (SEs) and typical enhancers (TEs) and triggers threshold and graded gene expression, respectively. However, the mechanisms by which NF-κB selectively participates in these enhancers remain unclear. Here we show using mouse primary B lymphocytes that SE activity simultaneously associates with chromatin opening and enriched NF-κB binding, resulting in a higher fold change and threshold expression upon B cell receptor (BCR) activation. The higher fold change results from longer DNA, whereas the threshold response is explained by synergy in DNA-NF-κB binding and is supported by the coexistence of PU.1 and NF-κB in a SE before cell stimulation. This model indicates that the pre-existing NF-κB functions as a seed and triggers its processive binding upon BCR activation. Conclusion We detected marked small nerve fiber damage in both lower and upper limbs in SLE patients using CSP. We also utilized kinase translocation reporters (KTRs) to identify single cell effects of mitochondrial state on signaling cascades, PI3K/Akt/mTOR and Ras/Raf/MEK/ERK, commonly activated in TNBC. Furthermore, we determined effects of fission and fusion states on metastasis, bone destruction, and signaling in mouse models of breast cancer. Results Enforcing mitochondrial fission through chemical or genetic approaches inhibited migration, invasion, and metastasis in TNBC. Breast cancer cells with predominantly fissioned mitochondria exhibited reduced activation of Akt and ERK both in vitro and in mouse models of breast cancer. Treatment with leflunomide, a potent activator of mitochondrial fusion proteins, overcame inhibitory effects of fission on migration, signaling, and metastasis. Mining existing datasets for breast cancer revealed that increased expression of genes associated with mitochondrial fission correlated with improved survival in human breast cancer. Conclusions In TNBC, mitochondrial fission inhibits cellular processes and signaling pathways associated with cancer progression and metastasis. We aimed to determine which aspects of the development, content, and use of health outcome descriptors were valuable to guideline developers. Methods We developed 24 health outcome descriptors related to breast cancer screening and diagnosis for the European Commission Breast Guideline Development Group (GDG). Eighteen GDG members provided feedback in written format or in interviews. We then evaluated the process and conducted two health utility rating surveys. Results Feedback from GDG members revealed that health outcome descriptors are probably useful for developing recommendations and improving transparency of guideline methods. Time commitment, methodology training, and need for multidisciplinary expertise throughout development were considered important determinants of the process. Comparison of the two health utility surveys showed a decrease in standard deviation in the second survey across 21 (88%) of the outcomes.   • The recent availability of small and low-cost sensor carrying unmanned aerial systems (UAS, commonly known as drones) coupled with advances in image processing software (i.e., structure from motion photogrammetry) has made drone-collected imagery a potentially valuable tool for rangeland inventory and monitoring. Drone-imagery methods can observe larger extents to estimate indicators at landscape scales with higher confidence than traditional field sampling. They also have the potential to replace field methods in some instances and enable the development of indicators not measurable from the ground. **** research has already demonstrated that several quantitative rangeland indicators can be estimated from high-resolution imagery. Developing a suite of monitoring methods that are useful for supporting management decisions (e.g., repeatable, cost-effective, and validated against field methods) will require additional exploration to develop best practices for image acquisition and analytical workflows that can replace most field methods in large monitoring programs, they could be a valuable enhancement for pressing local management needs.There is considerable evidence that animals are able to discriminate between quantities. Despite the fact that quantitative skills have been extensively studied in adult individuals, research on their development in early life is restricted to a limited number of species. We, therefore, investigated whether 2-month-old puppies could spontaneously discriminate between different quantities of food items. We therefore used the transgenic approach of overexpressing gibberellin 2-oxidase 6 (OsGA2ox6), a rice GA deactivation gene, to investigate its functional effect in miniaturizing Phalaenopsis and to create a stable miniaturization platform to facilitate a supply for the potential demands of the miniature flower market. RESULTS A commercial moth orchid, Phalaenopsis Sogo Yukidian 'SPM313', was transformed with the plasmid vector UbiOsGA2ox6 and successfully overexpressed the OsGA2ox6 gene in planta. The transgenic lines displayed darker-green, shorter, and wider leaves, thicker roots and **** shorter flower spikes (10 cm vs 33 cm) than the nontransgenic line with a normal flower size and ******** ability and are therefore an ideal miniaturized form of Phalaenopsis orchids. CONCLUSIONS We demonstrated that the ectopic expression of OsGA2ox6 can miniaturize Phalaenopsis Sogo Yukidian 'SPM313' while preserving its ******** ability, providing an alternative, useful method for miniaturizing Phalaenopsis species. This miniaturization by a transgenic approach can be further expanded by using GA2ox genes from different plant species or different gene variants, thereby expanding the technical platform for miniaturizing Phalaenopsis species to meet the potential demands of the miniature Phalaenopsis flower market.The MIR gene is not an Oryza sativa orphan gene, but an Oryza genus-specific gene that evolved before AA lineage speciation by a complex origination process. Rice (Oryza sativa L.) is a model species and an economically relevant crop. The Oryza genus comprises 25 species, with genomic data available for several Oryza species, making it a model for genetics and evolution. The Mitochondrial Iron-Regulated (MIR) gene was previously implicated in the O. sativa Fe deficiency response, and was considered an orphan gene present only in rice. Here we show that MIR is also found in other Oryza species that belong to the Oryza sativa complex, which have AA genome type and constitute the primary gene pool for O. sativa breeding. Our data suggest that MIR originated in a stepwise process, in which sequences derived from an exon fragment of the raffinose synthase gene were pseudogenized into non-coding, which in turn originated the MIR gene de novo. All species with a putative functional MIR gene conserve their regulation by Fe deficiency, with the exception of Oryza barthii. In O. barthii, the MIR coding sequence was translocated to a different chromosomal position and separated from its regulatory region, leading to a lack of Fe deficiency responsiveness. Moreover, the MIR co-expression subnetwork cluster in O. sativa is responsive to Fe deficiency, evidencing the importance of the newly originated gene in Fe uptake. This work establishes that MIR is not an orphan gene as previously proposed, but a de novo originated gene within the genus Oryza. We also showed that MIR is undergoing genomic changes in one species (O. barthii), with an impact on Fe deficiency response.We investigated the prevalence and characteristics of defective mismatch repair (dMMR) in colorectal cancer (CRC) patients who would potentially benefit from anti-programmed cell death protein 1 (PD-1) immunotherapy. Medical records were obtained and reviewed for 1147 patients who underwent surgical resection of stage I-IV CRC, in whom universal screening for Lynch syndrome using immunohistochemistry for MMR proteins had been undertaken. The molecular characteristics of dMMR CRCs were also investigated. Defective MMR accounted for 5.2% of stage I-IV CRC patients, including 12 (1.0% of all CRC patients) who had stage IV disease or recurrence after curative resection (n = 6 each). These 12 patients included patients with LS (n = 3) and Lynch-like syndrome (n = 1). Defective MMR tumors were predominantly located in the right-sided colon (P  less then  0.01). Approximately 1% of stage I-IV CRC patients could potentially benefit from anti-PD-1 immunotherapy, while one-third would require genetic counseling and/or MMR gene testing.

The transgenic lines displayed darker-green, shorter, and wider leaves, thicker roots and much shorter flower spikes (10 cm vs 33 cm) than the nontransgenic line with a normal flower size and blooming ability and are therefore an ideal miniaturized form of Phalaenopsis orchids. CONCLUSIONS We demonstrated that the ectopic expression of OsGA2ox6 can miniaturize Phalaenopsis Sogo Yukidian 'SPM313' while preserving its blooming ability, providing an alternative, useful method for miniaturizing Phalaenopsis species. This miniaturization by a transgenic approach can be further expanded by using GA2ox genes from different plant species or different gene variants, thereby expanding the technical platform for miniaturizing Phalaenopsis species to meet the potential demands of the miniature Phalaenopsis flower market.The MIR gene is not an Oryza sativa orphan gene, but an Oryza genus-specific gene that evolved before AA lineage speciation by a complex origination process. Rice (Oryza sativa L.) is a model species and an economically relevant crop. The Oryza genus comprises 25 species, with genomic data available for several Oryza species, making it a model for genetics and evolution. The Mitochondrial Iron-Regulated (MIR) gene was previously implicated in the O. sativa Fe deficiency response, and was considered an orphan gene present only in rice. Here we show that MIR is also found in other Oryza species that belong to the Oryza sativa complex, which have AA genome type and constitute the primary gene pool for O. sativa breeding. Our data suggest that MIR originated in a stepwise process, in which sequences derived from an exon fragment of the raffinose synthase gene were pseudogenized into non-coding, which in turn originated the MIR gene de novo. All species with a putative functional MIR gene conserve their regulation by Fe deficiency, with the exception of Oryza barthii. In O. barthii, the MIR coding sequence was translocated to a different chromosomal position and separated from its regulatory region, leading to a lack of Fe deficiency responsiveness. Moreover, the MIR co-expression subnetwork cluster in O. sativa is responsive to Fe deficiency, evidencing the importance of the newly originated gene in Fe uptake. This work establishes that MIR is not an orphan gene as previously proposed, but a de novo originated gene within the genus Oryza. We also showed that MIR is undergoing genomic changes in one species (O. barthii), with an impact on Fe deficiency response.We investigated the prevalence and characteristics of defective mismatch repair (dMMR) in colorectal cancer (CRC) patients who would potentially benefit from anti-programmed cell death protein 1 (PD-1) immunotherapy. Medical records were obtained and reviewed for 1147 patients who underwent surgical resection of stage I-IV CRC, in whom universal screening for Lynch syndrome using immunohistochemistry for MMR proteins had been undertaken. The molecular characteristics of dMMR CRCs were also investigated. Defective MMR accounted for 5.2% of stage I-IV CRC patients, including 12 (1.0% of all CRC patients) who had stage IV disease or recurrence after curative resection (n = 6 each). These 12 patients included patients with LS (n = 3) and Lynch-like syndrome (n = 1). Defective MMR tumors were predominantly located in the right-sided colon (P  less then  0.01). Approximately 1% of stage I-IV CRC patients could potentially benefit from anti-PD-1 immunotherapy, while one-third would require genetic counseling and/or MMR gene testing.

We found three main areas where misuse of self-assessments may have resulted in inaccurate measures of learning outcomes measures of "confidence" or "comfort", pre-post self-assessments, and the use of ambiguous learning objectives. To prevent future misuse and to build towards more valid and reliable data for program evaluations, we present the following recommendations measure competence instead of confidence or comfort; use pre-test self-assessments for instructional purposes only (and not for data); ask participants to do the post-intervention self-assessments first followed by retrospective pre-intervention self-assessments afterwards; and use observable, clear, specific learning objectives in the educational intervention that can then be used to create the self-assessment statements.
    International health experiences (IHEs) are popular among medical learners and provide a valuable learning experience. IHE participants have demonstrated an increased intention to care for underserved populations in the future, but what is its actual impact on practice? This study evaluates the effect of postgraduate IHE participation on the future careers of clinicians regarding their work among underserved populations.

    We conducted a systematic review and meta-analysis of peer-reviewed articles comparing the populations served by physicians who had participated in an IHE with those of physicians who had not participated in an IHE.

    764 titles were scanned, 28 articles were reviewed, with an eventual 3 studies of fair-good or good quality identified. These addressed physicians' service to domestic underserved populations, and also addressed future service in a low- or middle-income country (LMIC). Meta-analysis demonstrated a statistically-significant increase in service by IHE graduates to domestic underserved populations (OR = 2.12; CI = 95%; P = 0.03). The certainty of the evidence was low due to limitations in study design (non-randomised studies) and inconsistency in effects.

    Participation in an IHE may cause an increase in care for domestic underserved populations in future clinical practice, though further research from high quality randomised trials is needed to increase the certainty of the effect. Further study is needed to establish whether there is a similar effect with increased future service in a LMIC setting.

    Clinical placements are essential for applied learning experiences in health professions education. Unfortunately, there is little consensus on how best to prepare learners for the transition between academic and clinical learning. We explored learners' perceptions of hospital-based orientation and resulting preparedness for clinical placement.

    Sixty-three learners participated in a total of 18 semi-structured focus groups, during their clinical placements. Data were analyzed thematically.

    Previous literature has explored the underrepresentation of women in surgery. However, this research has often been quantitative or limited by considering only the perspectives and experiences of women at more advanced career stages. Here, we use a qualitative methodology and a sample of women and men across the career continuum to identify the role that gender plays in the decision to pursue a surgical career.

    We audio-recorded and transcribed semi-structured interviews conducted with 12 women and 12 men ranging in their level of medical training from medical students to residents to staff surgeons. We used Braun and Clarke's six-step approach to thematic analysis to analyze the data, maintaining trustworthiness and credibility by employing strategies including reflexivity and participant input.

    Our findings suggested that the characteristics of surgery and early exposure to the profession served as important factors in participants' decisions to pursue a surgical career. Although not explicitly mentioned by participants, each of these areas may implicitly be gendered.

  • Interface engineering multi-component core-shell nanostructures with highly efficient and reversible faradaic reactions for energy conversion storage devices is still a challenge. Here, Co3O4 nanowires@NiO ultrafine nanowires on Ni foam were well fabricated via coating the NiO ultrafine nanowires on porous Co3O4 nanowire arrays. The combination of structural and compositional advantages endows the Co3O4@NiO core-shell composites with excellent electrochemical performance, such as a favorable specific capacity of 0.71 mA h cm-2 at 3 mA cm-2, excellent rate capability and 85% retention rate up to 10,000 cycles. Rechargeable alkaline batteries with the Co3O4@NiO core-shell composites and AC as cathode and anode, respectively, had a high specific capacity of 0.51 mA h cm-2 and stable cycling ability (81% retention after 5000 cycles). The hierarchical core-shell heterostructure electrode exhibits excellent electrochemical performance, making it a very promising material for next-generation energy storage device applications.A novel coordination tailoring top-down delamination strategy involving the controllable partial disassembly of water-labile 3D MOFs has been explored to fabricate ultrathin 2D MOF nanosheets, and it exhibits many advantages such as green, convenience and high efficiency. Accordingly, 2D ultrathin MOF nanosheets with a two-fold interpenetrated architecture have been rapidly obtained from a 3D pillar-layered MOF, and they show distinguished fluorescence responses to different solvents.Motivated by the discovery of neutral noble gas hydrides, herein, we have explored the possibility of the existence of a novel class of neutral noble gas compounds, HNgBO, HNgOB, HNgAlO and HNgOAl (Ng = Xe and Rn), through the insertion of a Ng atom into the hydroxides of icosagens and their isomers, namely, HBO, HOB, HAlO and HOAl. Second-order Møller-Plesset perturbation theory (MP2), density functional theory (DFT), and coupled-cluster theory (CCSD(T))-based methods have been employed to investigate the structures, stabilities, energetics, harmonic vibrational frequencies, and charge distribution of the predicted molecules. The HXeBO, HXeOAl, HRnBO, HRnAlO and HRnOAl molecules are found to be thermodynamically stable with respect to all plausible 2-body and 3-body dissociation channels except the 2-body dissociation pathway, leading to the formation of global minimum products (Ng + HBO), (Ng + HOAl) and (Ng + HAlO). However, the very large activation energy barrier heights provide enough kinetic stability conclusion and also suggest that the predicted metastable HNgBO and HNgOAl molecules should essentially exist in the form of [HNg]+[BO]- and [HNg]+[OAl]-, respectively. All the calculated results reported in this work indicate that it might be possible to prepare and characterize the predicted molecules via suitable experimental technique(s) under cryogenic conditions.The evolution of retained species during the whole methanol-to-olefins process was revealed with the aid of GC-MS, thermogravimetric analysis (TG) and density functional theory (DFT) calculations. The stronger preference for 〈210〉 vacancy-vacancy alignments can explain why long range ordering is identified by distinct C-type like superlattice peaks in neutron diffraction patterns for Nd2Ce2O7 whereas they appear to be almost invisible in La2Ce2O7.

We observe more collective chains of migrating oxygen in Nd2Ce2O7- a manifestation of a stronger preference for a dynamic local oxygen vacancy order - than in La2Ce2O7. The stronger preference for 〈210〉 vacancy-vacancy alignments can explain why long range ordering is identified by distinct C-type like superlattice peaks in neutron diffraction patterns for Nd2Ce2O7 whereas they appear to be almost invisible in La2Ce2O7.

  • Hepatocellular carcinoma (HCC) is a frequent and increasing cause of cancer-related deaths worldwide. Reversing this trend is complicated by the varied aetiological factors leading to liver cirrhosis resulting in molecular genetic and clinical heterogeneity, combined with frequent presentation at advanced stage. Large-scale genomic studies have identified alterations in key signalling pathways for HCC development and progression, but these findings have not yet directly influenced patient management in the clinical setting. RESULTS Most patients (59%) received all of their mental health care in medical settings and from unaffiliated providers, including a large portion (16%) with serious mental illness. Despite the availability of integrated care in the medical setting and use of unaffiliated mental health providers, rates of mental health inpatient admissions were high among all patients in this setting (including those with serious and with nonserious mental illness), considerably higher than for patients treated in a mental health clinic within the system. Rates of medical and substance abuse inpatient admissions were also **** higher for patients treated in the medical setting and by unaffiliated providers, compared with those treated in the system's mental health clinics. CONCLUSIONS Findings suggest that when mental health services are available in medical and mental health settings within the same system, either patients with more severe physical illnesses are more likely to receive their mental health care in medical settings and from unaffiliated providers and thus have more hospitalizations of all types or affiliated mental health settings more effectively address clinical needs and thus reduce hospitalizations.OBJECTIVE The Community Informant Detection Tool (CIDT) is a vignette- and picture-based method of proactive case detection to promote help seeking for persons with depression, psychosis, alcohol use disorder, and epilepsy. The authors evaluated the effectiveness of the CIDT to increase help-seeking behavior in rural Nepal, where a district mental health care plan was being implemented. METHODS Twenty-four health facilities were randomly assigned to one of two methods for training their all-female cadre of community health volunteers standard training or standard training that included the CIDT. The authors compared the number of patients with depression, psychosis, alcohol use disorder, and epilepsy who were registered in the routine health information system prior to and 6 months after the training. RESULTS At health facilities where volunteers received CIDT training, 309 patients were registered as having depression, psychosis, alcohol use disorder, or epilepsy, compared with 182 patients at facilities where volunteers received standard training. The median number of patients registered was 47% greater at facilities where CIDT training was included (24 patients) than at facilities with standard training (16 patients) (p=0.04, r=0.42). The difference in the number of registered patients remained significant when the analysis factored in the population catchment (N=18 patients [CIDT] versus N=14 [standard] per 10,000 population; p=0.05, r=0.40). CONCLUSIONS The median number of patients registered as having a mental illness was 47% greater at primary care facilities in which community health volunteers used the CIDT than at facilities where volunteers received standard training. Challenges for inpatient psychiatry include risky close contact among staff and patients, space constraints, and structural barriers in care delivery. Nuanced considerations of five contingency planning strategies in response to COVID-19 are described, including COVID-19-specific precautions, visitor restrictions, physician workforce considerations, operational adjustments, and group therapy changes. Organized leadership and clear communication are identified as early priorities in pandemic response to minimize misinformation and address immediate challenges.OBJECTIVE The purpose of this study was to evaluate a 16-week, reverse-integrated care (bringing primary care interventions/services into the psychiatric setting) behavioral and educational group intervention for individuals with serious mental illness and diabetes. METHODS The primary outcome was change in glycated hemoglobin (HbA1c). Secondary outcomes included body mass index (BMI), blood pressure, lipid levels, physical activity, diabetes knowledge, and self-care. RESULTS Thirty-five participants attended at least one group and were included in a modified intent-to-treat analysis. From baseline to week 16, HbA1c improved, from 7.5±1.6 to 7.1±1.4, p=0.01, and BMI improved, from 33.3±3.8 to 32.9±4.1, p less then 0.001, as did measures of diabetes knowledge and self-care. One-year follow-up in a subset of participants showed no evidence of rebound in HbA1c. CONCLUSIONS This 16-week behavioral and educational group intervention resulted in improvements in glycemic control, BMI, diabetes knowledge, and self-care.

We believe these findings can inform the development of policy and support the implementation of SRHR related guidelines to support staff working with young people with ID.Background The latest advancements in DNA sequencing technologies have facilitated the resolution of the phylogeny of insects, yet parts of the tree of Holometabola remain unresolved. The phylogeny of Neuropterida has been extensively studied, but no strong consensus exists concerning the phylogenetic relationships within the order Neuroptera. Here, we assembled a novel transcriptomic dataset to address previously unresolved issues in the phylogeny of Neuropterida and to infer divergence times within the group. We tested the robustness of our phylogenetic estimates by comparing summary coalescent and concatenation-based phylogenetic approaches and by employing different quartet-based measures of phylogenomic incongruence, combined with data permutations. Results Our results suggest that the order Raphidioptera is sister to Neuroptera + Megaloptera. Coniopterygidae is inferred as sister to all remaining neuropteran families suggesting that larval cryptonephry could be a ground plan feature of Neuroptera. A claw insights into the timing of diversification of Neuropterida. Despite the vast amount of analyzed molecular data, we found that certain nodes in the tree of Neuroptera are not robustly resolved. Therefore, we emphasize the importance of integrating the results of morphological analyses with those of sequence-based phylogenomics. We also suggest that comparative analyses of genomic meta-characters should be incorporated into future phylogenomic studies of Neuropterida.Background Immunotherapy has paved the way for new therapeutic opportunities in cancer but has failed to show any efficacy in Pancreatic Adenocarcinoma (PDAC), and its therapeutic role remains unclear. The objective of this study is to examine the impact of immunotherapy in combination with chemotherapy, RT, and chemoradiation on the overall survival (OS) of PDAC patients who received definitive surgery of the tumor using the National Cancer Database (NCDB). Methods Patients with PDAC who received definitive surgery of the pancreatic tumor and were diagnosed between 2004 and 2016 from the NCDB were identified. Cox proportional hazard analysis was used to assess the survival difference between patients who received chemotherapy plus immunotherapy and chemoradiation therapy plus immunotherapy and their counterparts who only receive these treatments without immunotherapy. The multivariable analysis was adjusted for age of diagnosis, race, sex, place of living, income, education, treatment facility type, insurancnterparts who only received chemoradiation without immunotherapy (HR 0.852 CI 0.757-0.958; P less then 0.008). Conclusions In this study, the addition of immunotherapy to chemoradiation therapy was associated with significantly improved OS in PDAC patients who received definitive surgery. Methods This retrospective, single-center, single-surgeon case-control study included patients who underwent primary THA between 2008 and 2016. We sent postal questionnaires to 38 patients with Crowe type III and IV dysplasia. Among the questionnaire respondents, 23 patients, excluding those with a follow-up period of 50. Conclusion The postoperative VAS score was higher in Crowe type III and IV dysplasia than in Crowe type I dysplasia, but no significant differences were detected in the postoperative satisfaction, JOA score, and SF-36 score. These findings may help explain the effects of THA preoperatively to patients with Crowe type III and IV dysplasia. Presence of bone marrow edema (using T2W sequences with fat suppression), diffusion restriction in Diffusion Weighted Imaging (DWI) or Diffusion Weighted Imaging with Background Suppression (DWIBS), and dynamic contrast enhancement were evaluated. Each SI joint was divided into 4 quadrants upper iliac, lower iliac, upper associated with IBD; it is often asymptomatic and clinically underdetected, with an unrelated progression with respect to the underlying IBD. MRE offers the possibility to study SI joints in young patients with IBD who undergo MRE for the investigation of their intestinal condition. Furthermore, we observed that gadolinium enhancement does not improve diagnostic specificity in sacroiliiitis detection.
