The synthesis of several biomolecules is often regulated by a quorum sensing (QS) mechanism, a chemical cell-to-cell communication based on the production and diffusion of small molecules named "autoinducers", most of which belonging to the N-acyl homoserine lactones' (AHLs) family. There are few reports about this mechanism in extremophiles, in particular thermophiles. Here, we report the identification of a signal molecule, the N-butanoyl-homoserine lactone (C4-HSL), from the milieu of A. amylolyticus. Moreover, investigations performed by supplementing a known QS inhibitor, trans-cinnamaldehyde, or exogenous C4-HSL in the growth medium of A. amylolyticus suggested the involvement of QS signaling in the modulation of extracellular α-amylase activity. The data showed that the presence of the QS inhibitor trans-cinnamaldehyde in the medium decreased amylolytic activity, which, conversely, was increased by the effect of exogenous C4-HSL. Overall, these results represent the first evidence of the production of AHLs in thermophilic microorganisms, which could be responsible for a communication system regulating thermostable α-amylase activity.The COVID-19 lockdown was imposed in a context of notable inequalities in the distribution of the social determinants of health. It is possible that the housing conditions in which children and their families experienced the confinement, and the adoption of healthy behaviors, may have followed unequal patterns. The aim was to describe social inequalities in housing conditions and in health-related behaviors among children during the lockdown in Spain. This cross-sectional study was based on data from an online survey collecting information on the child population (3-12 years) living in Spain (n = 10,765). The outcome variables used were several housing conditions and health-related behaviors. The socioeconomic variables used were financial difficulties and parents' educational level. Using a single-case, concurrent multiple baseline design, autism spectrum disorder symptomology (i.e., higher-order restrictive and repetitive behaviors and interests; higher-order RRBIs) was targeted in toddlers (21-35 months) waiting for a diagnostic appointment. Caregivers were coached via telehealth to mediate early intervention to decrease interfering, inflexible higher-order RRBIs during play using four evidence-based applied behavior analytic strategies modeling, prompting, differential reinforcement of appropriate behaviors, and response interruption and redirection. Six mother-child dyads were recruited from pediatrician offices and early intervention service districts in the United States. All families were considered under-served, under-resourced, or living in rural locations. A visual analysis of the data combined with Tau-U revealed a strong basic effect between the intervention package and parent strategy use and child flexible and inflexible behavior. Findings were consistent across participants with one exception demonstrating a moderate effect for flexible behaviors yet a strong effect for inflexible behaviors. Standardized mean difference was beyond zero for all participants. Implications for science and practice include support for early intervention of higher-order RRBIs for young children with and at risk for ASD.A non-contact heartbeat/respiratory rate monitoring system was designed using narrow beam millimeter wave radar. Equipped with a special low sidelobe and small-sized antenna lens at the front end of the receiving and transmitting antennas in the 120 GHz band of frequency-modulated continuous-wave (FMCW) system, this sensor system realizes the narrow beam control of radar, reduces the interference caused by the reflection of other objects in the measurement background, improves the signal-to-clutter ratio (SCR) of the intermediate frequency signal (IF), and reduces the complexity of the subsequent signal processing. In order to solve the problem that the accuracy of heart rate is easy to be interfered with by respiratory harmonics, an adaptive notch filter was applied to filter respiratory harmonics. Meanwhile, the heart rate obtained by fast Fourier transform (FFT) was modified by using the ratio of adjacent elements, which helped to improve the accuracy of heart rate detection. The experimental results show that when the monitoring system is 1 m away from the human body, the probability of respiratory rate detection error within ±2 times for eight volunteers can reach 90.48%, and the detection accuracy of the heart rate can reach 90.54%. Finally, short-term heart rate measurement was realized by means of improved empirical mode decomposition and fast independent component analysis algorithm.

  • Hidradenoma papilliferum is a rare benign neoplasm arising from apocrine glands. It occurs commonly on the anogenital region of middle-aged women. It usually presents as a slow growing, solitary asymptomatic, skin colored or red nodule less than 1 cm in diameter.

    The case is a 38-year-old, white woman who presented with a painful nodule occurring within a month in the himenal region of the posterior vaginal introitus. The nodule was excisied and the histology revealed a hidradenoma papilliferum. The diagnosis and treatment of hidradenoma papilliferum is possible with surgical removal and histopathological evaluation of nodules.

    When an adult woman presents with a noduler lesion in the anogenital area, sexually transmitted diseases and other benign and malignant vulvar lesions, as well as malignant transformation is very rare but,should be kept in mind; however because it has been reported and long-term clinical follow-up is suggested.

    We report a unique case of synchronous sacrococcygeal chordoma in association with rectal invasive adenocarcinoma. Retrorectal tumors are a rare disease caused by a variety of pathologies. To our knowledge, no prior cases of such a coincidental finding of both cancers have been reported in the literature.

    This is the case of a 74-year-old white middle eastern man, with known hypertension under treatment, who presented with complaints of progressive lower **** pain associated with urinary incontinence over the past 12months. Magnetic resonance imaging (MRI) of the pelvis showed a large midline, well-defined, oval-shaped lesion replacing the sacrococcygeal portion of the spine, with extension to the presacral region. https://www.selleckchem.com/products/az20.html Computed tomography (CT)-guided Tru-Cut biopsy revealed features suggestive of chordoma. At surgery, we performed excision of the entire mass en bloc, sacrectomy with rectus abdominis myocutaneous flap reconstruction and end sigmoid colostomy. Surgical histopathology proved it to be sacral dedifferentiated chordoma and rectal invasive adenocarcinoma. Overall, the patient recovered well postoperatively, was discharged home with functional stoma and on permanent Foley catheter use.

    To the best of our knowledge, this is the only reported case of such a presentation, and sheds light on the approach and management. We hope that reporting such a case will add value to the medical literature.

It causes chronic animal welfare issues and economic losses. Eradication of scab has proved impossible in many sheep-rearing areas and recent reports of resistance to macrocyclic lactones, a key class of parasiticide, highlight the importance of improving approaches to scab management. To allow this, the current study aimed to develop a stochastic spatial metapopulation model for sheep scab transmission which can be adapted for use in any geographical region, exhibited here using data for Great Britain. The model uses agricultural survey and sheep movement data to geo-reference farms and capture realistic movement patterns. Reported data on sheep scab outbreaks from 1973 to 1991 were used for model fitting with Sequential Monte Carlo Approximate Bayesian Computation methods. The outbreak incidence predicted by the model was from the same statistical distribution as the reported outbreak data ([Formula see text] = 115.3, p = 1) and the spatial location of sheep scab outbreaks predicted was positively correlated with the observed outbreak data by county ([Formula see text] = 0.55, p  less then  0.001), confirming that the model developed is able to accurately capture the number of farms infected in a year, the seasonality of scab incidence and the spatial patterns seen in the data. This model gives insight into the transmission dynamics of sheep scab and will allow the exploration of more effective control strategies.
    Non-small cell lung carcinoma (NSCLC) is one of the most common human cancers, comprising approximately 80-85% of all lung carcinomas. An estimated incidence of NSCLC is approximately 2 million new cases per year worldwide.

    In recent decade, the treatment of NSCLC has made breakthrough progress owing to a large number of targeted therapies which were approved for clinical use. Epidemiology, genetic susceptibility, and molecular profiles in patients are likely to play an important factor in response rates and survival benefits to these targeted treatments and thus warrant further investigation on ethnic differences in NSCLC. In this study, a total number of 1500 Chinese patient samples,1000 formalin fixed paraffin-embedded (FFPE) and 500 blood samples, from patients with NSCLC were analyzed by targeted sequencing to explore mutational landscape in ethnic groups associated with China.

    Overall, the data presented here provide a comprehensive analysis of NSCLC mutational landscape in Chinese patients and findings are discussed in the context of similar studies on different ethnic groups.

The family Echiniscidae is one such lineage; it is one of the most diverse groups of limno-terrestrial tardigrades and can be characterized as having achieved great evolutionary success. In this contribution, using populations representing several species that originated from the Indomalayan region, we reconstructed phylogenetic affinities within Nebularmis, a recently erected genus within the Echiniscus lineage. Nebularmis auratus sp. nov. and Nebularmis burmensis sp. nov. are described from the Eastern Yoma Mountains and the Shan Hills (Myanmar), Nebularmis bhutanensis sp. nov. is described from the Eastern Himalayas (Bhutan), and Nebularmis indicus sp. nov. is described from the foothills of the Western Ghats (Goa, India). Moreover, males are reported in populations of the last two species. All known members of the genus can be phenotypically differentiated based on minute details of their dorsal sculpture and claws.

    Postpartum hemorrhage is a leading cause of pregnancy-related morbidity and mortality; however, there is limited ability to identify women at risk of this obstetrical complication.

    This study aimed to develop and validate a prediction model for postpartum hemorrhage based on antenatal and intrapartum risk factors.

    This was a retrospective cohort study of women who delivered between April 2016 and March 2019 at a single safety net hospital. The prevalence of postpartum hemorrhage, defined as blood loss of ≥1000 mL at the time of delivery, was determined, and characteristics were compared between women with and without postpartum hemorrhage. Women were randomly assigned to a prediction or a validation cohort. The selection of predictors to be included in the model was based on known antenatal and intrapartum risk factors for postpartum hemorrhage. A multivariable logistic regression with a backward stepwise approach was used to create a prediction model. Area under the receiver operating characteristic cu74.2%, a positive predictive value of 18.6%, a negative predictive value of 98.8%, a false-negative rate of 13.1%, and a false-positive rate of 25.9%.

    The model performed reasonably well in identifying women at risk of postpartum hemorrhage. Further studies are necessary to evaluate the model in clinical practice and its effect on decreasing the prevalence of postpartum hemorrhage and associated maternal morbidity.

    Although mass vaccination against COVID-19 may prove to be the most efficacious end to this deadly pandemic, there remain concern and indecision among the public toward vaccination. Because pregnant and reproductive-aged women account for a large proportion of the population with particular concerns regarding vaccination against COVID-19, this survey aimed at investigating their current attitudes and beliefs within our own institution.

    This study aimed to understand vaccine acceptability among pregnant, nonpregnant, and breastfeeding respondents and elucidate factors associated with COVID-19 vaccine acceptance.

    We administered an anonymous online survey to all women (including patients, providers, and staff) at our institution assessing rates of acceptance of COVID-19 vaccination. Respondents were contacted in 1 of 3 ways by email, advertisement flyers, and distribution of quick response codes at virtual town halls regarding the COVID-19 vaccine. Based on their responses, respondents were divided into 3spondents the second most likely (55.2%). Pregnant respondents had the lowest rate of vaccine acceptance (44.3%; P<.001). Prevalence ratios revealed all non-White races except for non-Hispanic Asian respondents, and Spanish-speaking respondents were less likely to accept vaccination (Table 3). Working in healthcare was not found to be associated with vaccine acceptance among our cohort.

A biphasic viscoelastic model using an ellipsoidal fiber distribution was created with three distinct layers of cartilage (superficial zone, middle zone, and deep zone) along with a layer of subchondral bone. For stress-relaxation in unconfined compression, our results for compressive stress, radial stress, and effective fluid pressure were compared with established biphasic analytical solutions. Our model also shows the development of fluid pressure gradients at the cartilage-bone interface during loading. Fluid pressure gradients that develop at the cartilage-bone interface show consistently higher pressures in cartilage following the initial loading to 10% stain, followed by convergence of the pressures in cartilage and bone during the 400 s relaxation period. These results provide additional evidence that fluid is transported between cartilage and bone during loading and improves upon estimates of the magnitude of this effect through incorporating a realistic distribution and estimate of the collagen ultrastructure. Understanding fluid transport between cartilage and bone may be key to new insights about the mechanical and biological environment of both tissues in health and disease.

This study aimed to develop and validate a prediction model for postpartum hemorrhage based on antenatal and intrapartum risk factors. This was a retrospective cohort study of women who delivered between April 2016 and March 2019 at a single safety net hospital. The prevalence of postpartum hemorrhage, defined as blood loss of ≥1000 mL at the time of delivery, was determined, and characteristics were compared between women with and without postpartum hemorrhage. Women were randomly assigned to a prediction or a validation cohort. The selection of predictors to be included in the model was based on known antenatal and intrapartum risk factors for postpartum hemorrhage. A multivariable logistic regression with a backward stepwise approach was used to create a prediction model. Area under the receiver operating characteristic cu74.2%, a positive predictive value of 18.6%, a negative predictive value of 98.8%, a false-negative rate of 13.1%, and a false-positive rate of 25.9%. The model performed reasonably well in identifying women at risk of postpartum hemorrhage. We administered an anonymous online survey to all women (including patients, providers, and staff) at our institution assessing rates of acceptance of COVID-19 vaccination. We determine the optimal number and duration of intensity measurements through a cycle of the retarder for these two types of setups as a function of instrument and noise parameters. We show that this number mainly depends on the type of noise that corrupts the measurements. We also show that with these setups, the starting angle of the retarder need not be known precisely and can be autocalibrated, which facilitates synchronization of the rotating retarder with the camera. We investigate the precision and feasibility domain of this autocalibration and show the RRDOFP setup has more attractive properties compared with RRFP setup. These results are important to optimize and facilitate the operation of polarization imagers based on a rotating retarder.The discovery of single structure Ce3+ doped garnet transparent ceramics (TCs) with a broad full width at half maximum (FWHM) is essential to realize a high CRI for high-power white light emitting diodes (LEDs) and laser diodes (LDs). In this work, by utilizing the ion substitution engineering strategy, pure phase Gd3Sc2Al3O12Ce3+ (GSAGCe) TC with a broad FWHM of 132.4 nm and a high CRI value of 80.7 was fabricated through the vacuum sintering technique for the first time. The optimized in-line transmittance of TCs was 58.4% @ 800 nm. Notably, the GSAGCe TCs exhibited a remarkable red shift from 546 nm to 582 nm, with a high internal quantum efficiency (IQE) of 46.91%. The degraded thermal stability in CeGSAG TCs was observed compared with that of CeYAG TC, owing to the narrowed band gap of GSAG. Additionally, remote excitation white LEDs/LDs were constructed by combining GSAGCe TCs with blue LED chips or laser sources. A tunable color hue from yellow to shinning white was achieved in white LEDs, whereas the acquired CRI and CCT of the white LDs were 69.5 and 7766 K, respectively. This work provides a new perspective to develop TCs with high CRI for their real applications in high-power white LEDs/LDs.We report the spatiotemporal mode-locked multimode fiber laser operating at 1.55 µm based on semiconductor saturable absorber mirrors with the mode-locking threshold as low as 104 mW. The lack or limited therapeutic armamentarium against some viral infections and increasing drug resistance issues make the search for new antiviral agents urgent. In recent years, a growing literature highlighted the use of triazolopyrimidine (TZP) heterocycles in the development of antiviral agents, with numerous compounds that showed potent antiviral activities against different RNA and DNA viruses. TZP core represents a privileged scaffold for achieving biologically active molecules, thanks to i) the synthetic feasibility that allows to variously functionalize TZPs in the different positions of the nucleus, ii) the ability of TZP core to establish multiple interactions with the molecular target, and iii) its favorable pharmacokinetic properties. In the present review, after mentioning selected examples of TZP-based compounds with varied biological activities, we will focus on those antivirals that appeared in the literature in the last 10 years. Approaches used for their identification, the hit-to-lead studies, and the emerged structure-activity relationship will be described. A mention of the synthetic methodologies to prepare TZP nuclei will also be given. In addition, their mechanism of action, the binding mode within the biological target, and pharmacokinetic properties will be analyzed, highlighting the strengths and weaknesses of compounds based on the TZP scaffold, which is increasingly used in medicinal chemistry.

In recent years, a growing literature highlighted the use of triazolopyrimidine (TZP) heterocycles in the development of antiviral agents, with numerous compounds that showed potent antiviral activities against different RNA and DNA viruses. TZP core represents a privileged scaffold for achieving biologically active molecules, thanks to i) the synthetic feasibility that allows to variously functionalize TZPs in the different positions of the nucleus, ii) the ability of TZP core to establish multiple interactions with the molecular target, and iii) its favorable pharmacokinetic properties. In the present review, after mentioning selected examples of TZP-based compounds with varied biological activities, we will focus on those antivirals that appeared in the literature in the last 10 years. Approaches used for their identification, the hit-to-lead studies, and the emerged structure-activity relationship will be described. A mention of the synthetic methodologies to prepare TZP nuclei will also be given.   • The study examined the infradian rhythms in total motor activity and the periods of "freerunning" circadian rhythm in male C57BL/6 **** and European greenfinch Chloris chloris exposed to constant light. In birds and ****, the motor activity was characterized with inphase 4-day-long rhythm. The periods of free-running circadian rhythms in birds and **** were 23 and 25 h, respectively, and they were modulated with 4-day-long periodicity. The maximal deviations in the period of free-running circadian rhythm from the individual mean levels coincided with the acrophase of 4-day-long rhythm of motor activity in both species.We analyzed functional status of blood leukocytes in diabetes mellitus and after addition of glucose in vitro. To this end, generation of ROS and reactive halogen species by monocytes and neutrophils from patients with diabetes mellitus and healthy donors was assayed using lucigenin- and luminol-dependent chemiluminescence after stimulation with phorbol 12-myristate 13-acetate or opsonized zymosan in vitro. Formation of neutrophil extracellular traps was evaluated in the blood after addition of glucose. In comparison with donors, leukocytes from patients with diabetes mellitus were primed and this effect can be modeled by addition of glucose to the blood in vitro. Addition of glucose to donor blood also triggered the formation of neutrophil extracellular traps.Comparative evaluation of blood content of VEGF, sVEGFR1, and sVEGFR2 in 104 primary gastric cancer patients and 65 healthy persons was performed and associations of these markers with the principal clinical and morphological characteristics of gastric cancer were analyzed. The median levels of VEGF and sVEGFR1 in gastric cancer patients significantly surpassed the control by 1.5 (p less then 0.001) and 1.2 times (p less then 0.01), respectively. On the contrary, sVEGFR2 level in patients was below the control (p less then 0.001). We also demonstrated that the opioid (opicor) can improve cardiac contractility during the reperfusion period.The protocol of optogenetic ChR2-mediated activation of astrocytes was used in a model of artificial neurogenic niche, neurospheres implanted into ex vivo organotypic cultures of mouse hippocampus. The electrophysiological characteristics of the hippocampus and expression of molecules involved in the mechanisms of activation of astrocytes and microglia (GFAP, CD38, C3/C3b, Cx43, CD11b, and CD18) were evaluated. Photoactivation of astrocytes led to activation of neurogenesis and changes in the expression of molecules (Cx43 and CD38) that determine bioavailability of NAD+ to ensure proliferative activity of cells in the neurogenic niche. Implantation of neurospheres into organotypic slices of the hippocampus caused an increase in C3/C3b expression and suppression of the synaptic plasticity of hippocampal neurons.

Propagation of independent excitation waves in the right and left atria increases heterogeneity of depolarization and leads to the formation of atrial arrhythmias.We studied the expression of small heat shock proteins HSP10 and HSP27 in left ventricular cardiomyocytes in animals with arterial hypertension, insulin-dependent diabetes mellitus, and their combination. The experiment was performed on 38-week-old male Wistar-Kyoto and 38-57-week-old SHR (spontaneously hypertensive) rats. Insulin-dependent diabetes mellitus was modeled by single parenteral injection of streptozotocin (65 mg/kg). Expression of HSP10 and HSP27 in left ventricular cardiomyocytes was evaluated by immunohistochemical assay. It was found that the content of HSP10 in the left ventricular cardiomyocytes decreased in comparison with the control in case of isolated diabetes mellitus and, on the contrary, increased in case of arterial hypertension combined with diabetes mellitus. The intensity of HSP27 expression decreased in case of 38-week arterial hypertension and a combination of arterial hypertension with diabetes mellitus. However, in case of 57-week arterial hypertension we observed an increase in the content of HSP27 in cardiomyocytes.We studied the relationship between body weight and behavioral characteristics (anxiety, social status, aggressive and exploratory behavior, and sociability) of socially housed adult male cynomolgus macaques. Of the studied parameters, only anxiety showed pronounced relationship with body weight. We also demonstrated that the opioid (opicor) can improve cardiac contractility during the reperfusion period.The protocol of optogenetic ChR2-mediated activation of astrocytes was used in a model of artificial neurogenic niche, neurospheres implanted into ex vivo organotypic cultures of mouse hippocampus. The electrophysiological characteristics of the hippocampus and expression of molecules involved in the mechanisms of activation of astrocytes and microglia (GFAP, CD38, C3/C3b, Cx43, CD11b, and CD18) were evaluated. Photoactivation of astrocytes led to activation of neurogenesis and changes in the expression of molecules (Cx43 and CD38) that determine bioavailability of NAD+ to ensure proliferative activity of cells in the neurogenic niche. Implantation of neurospheres into organotypic slices of the hippocampus caused an increase in C3/C3b expression and suppression of the synaptic plasticity of hippocampal neurons.

  • Liver transplantation (LT) for children with urea cycle disorders (UCDs) is capable of correcting the enzymatic defect and preventing progressive neurologic injury. We describe the characteristics and outcomes of pediatric LT recipients with UCDs. We identified all pediatric ( less then 18 years) LT candidates with UCDs in the United Network for Organ Sharing (UNOS) database (February 2002 to September 2020). Multivariable Cox and logistic regression were used to determine risk factors for graft loss and cognitive delay, respectively. Of 424 patients, 1.9% (8/424) experienced waitlist mortality and 95.0% underwent LT (403/424). The most frequently encountered UCDs in our cohort were ornithine transcarbamylase deficiency (46.2%), citrullinemia (20.3%), and argininosuccinic aciduria (ASA; 12.9%). While skinfold callipers showed poor absolute reliability (mean differences (Δ) [95% CI] = 0.54% [0.22, 0.87], standard error of measurement (SEM) = 0.63%), ultrasound and the 3DPS showed excellent absolute (Δ = 0.17% [-0.25, 0.58], SEM = 0.78%; and Δ = -0.01% [-0.43, 0.40], SEM = 0.67%, respectively) and relative reliability (ICC2,1 = 0.988 [0.979, 0.993]; and ICC2,1 = 0.983 [0.968, 0.991], respectively). Compared to ADP (n = 43), skinfold callipers underestimated %BF (Δ = -4.53 [-7.72, -1.34]; p = 0.003), while ultrasound (Δ = -0.32 [-3.51, 2.87]; p = 0.99) and the 3DPS (Δ = 1.06 [-2.12. 4.26]; p = 0.77) were not significantly different. In the present study, the involvement of TIP30 in combating gefitinib resistance in NSCLC was determined for the first time in vitro and in vivo. Gain and loss functional studies showed that overexpression of TIP30 effectively sensitized cells to gefitinib in vitro, whereas TIP30 inhibition promoted gefitinib cell resistance. Moreover, TIP30 negatively regulated the activation of the p-AKT and p-MEK signaling pathways in PC9/GR. Importantly, PC9/GR harbored high levels of nuclear EGFR, and overexpression of TIP30 restored irregular EGFR trafficking and degradation from early endosomes to the late endosomes, decreasing the nuclear accumulation of EGFR, which may partly or totally inhibit EGFR-mediated induction of c-****transcription. Xenographic tumors induced by overexpression of TIP30 by PC9/GR cells in nude **** were suppressed compared with their original counterparts.   • Frictional sliding between patterned surfaces is of fundamental and practical importance in the haptic engineering of soft materials. In emerging applications such as remote surgery and soft robotics, thin fluid films between solid surfaces lead to a multiphysics coupling between solid deformation and fluid dissipation. These viral histones form 'forced' heterodimers, and a heterotetramer of four such heterodimers assembles DNA to form structures virtually identical to canonical eukaryotic nucleosomes.Hendra virus (HeV) and Nipah virus (NiV) are henipaviruses (HNVs) causing respiratory illness and severe encephalitis in humans, with fatality rates of 50-100%. There are no licensed therapeutics or vaccines to protect humans. HeV and NiV use a receptor-binding glycoprotein (G) and a fusion glycoprotein (F) to enter host cells. HNV F and G are the main targets of the humoral immune response, and the presence of neutralizing antibodies is a correlate of protection against NiV and HeV in experimentally infected animals. We describe here two cross-reactive F-specific antibodies, 1F5 and 12B2, that neutralize NiV and HeV through inhibition of membrane fusion. Cryo-electron microscopy structures reveal that 1F5 and 12B2 recognize distinct prefusion-specific, conserved quaternary epitopes and lock F in its prefusion conformation. We provide proof-of-concept for using antibody cocktails for neutralizing NiV and HeV and define a roadmap for developing effective countermeasures against these highly pathogenic viruses.Alveolata comprises diverse taxa of single-celled eukaryotes, many of which are renowned for their ability to live inside animal cells. Both the initial cellular events that gave rise to this intracellular lifestyle and the role of host immune modulation in coral-dinoflagellate endosymbiosis are poorly understood. Here, we use a comparative approach in the cnidarian endosymbiosis model Aiptasia, which re-establishes endosymbiosis with free-living dinoflagellates every generation. We find that uptake of microalgae is largely indiscriminate, but non-symbiotic microalgae are expelled by vomocytosis, while symbionts induce host cell innate immune suppression and form a lysosomal-associated membrane protein 1-positive niche. We demonstrate that exogenous immune stimulation results in symbiont expulsion and, conversely, inhibition of canonical Toll-like receptor signalling enhances infection of host animals. Our findings indicate that symbiosis establishment is dictated by local innate immune suppression, to circumvent expulsion and promote niche formation. This work provides insight into the evolution of the cellular immune response and key steps involved in mediating endosymbiotic interactions.Many wastewater treatment plants around the world suffer from the operational problem of foaming. This is characterized by a persistent stable foam that forms on the aeration basin, which reduces effluent quality. The foam is often stabilized by a highly hydrophobic group of Actinobacteria known as the Mycolata1. Gordonia amarae is one of the most frequently reported foaming members1. With no currently reliable method for treating foams, phage biocontrol has been suggested as an attractive treatment strategy2. Phages isolated from related foaming bacteria can destabilize foams at the laboratory scale3,4; however, no phage has been isolated that lyses G. amarae. Here, we assemble the complete genomes of G. amarae and a previously undescribed species, Gordonia pseudoamarae, to examine mechanisms that encourage stable foam production. We show that both of these species are recalcitrant to phage infection via a number of antiviral mechanisms including restriction, CRISPR-Cas and bacteriophage exclusion. Instead, we isolate and cocultivate an environmental ultrasmall epiparasitic bacterium from the phylum Saccharibacteria that lyses G. amarae and G. pseudoamarae and several other Mycolata commonly associated with wastewater foams. The application of this parasitic bacterium, 'Candidatus Mycosynbacter amalyticus', may represent a promising strategy for the biocontrol of bacteria responsible for stabilizing wastewater foams.Next-generation sequencing (NGS) technologies are now established in clinical laboratories as a primary testing modality in genomic medicine. With no currently reliable method for treating foams, phage biocontrol has been suggested as an attractive treatment strategy2. Phages isolated from related foaming bacteria can destabilize foams at the laboratory scale3,4; however, no phage has been isolated that lyses G. amarae. Here, we assemble the complete genomes of G. amarae and a previously undescribed species, Gordonia pseudoamarae, to examine mechanisms that encourage stable foam production. We show that both of these species are recalcitrant to phage infection via a number of antiviral mechanisms including restriction, CRISPR-Cas and bacteriophage exclusion. Instead, we isolate and cocultivate an environmental ultrasmall epiparasitic bacterium from the phylum Saccharibacteria that lyses G. amarae and G. pseudoamarae and several other Mycolata commonly associated with wastewater foams. The application of this parasitic bacterium, 'Candidatus Mycosynbacter amalyticus', may represent a promising strategy for the biocontrol of bacteria responsible for stabilizing wastewater foams.Next-generation sequencing (NGS) technologies are now established in clinical laboratories as a primary testing modality in genomic medicine. Fungi, such as mushrooms, produce a relatively large amount of BG, the structure and molecular weight of which varies depending on the species of fungi. This study was conducted to develop a detection probe for quantifying or detecting BG from fungi using BG-binding proteins. The binding properties of a new β-glucan recognition protein (BGRP) against various BGs were compared. With reference to the amino acid sequences of BGRP in insects, an artificial BGRP (supBGRP) was designed with higher production efficiency using gene recombination technology. SupBGRP was produced in Escherichia coli with high efficiency, and its reactivity with BG from fungi was the highest among the BG-binding proteins examined. SupBGRP exhibited high reactivity with 1,6-branched BG and will be useful for the quantification and detection of fungal BG.Isaria cicadae is an entomopathogenic fungus possessing several therapeutic properties and has a potential role in traditional Chinese medicine. The present study was designed to describe the taxonomic details of a new isolate of I. cicadae collected from the Northern Himalayas of India and to study its vegetative and reproductive growth responses under in vitro conditions. Proximate composition, biochemical profiling, and radical scavenging activities were studied to establish the bioactivity of the isolate. Micromorphological characteristics of conidia and conidiophore formation were studied using scanning electron microscopy. The optimum temperature and pH for mycelial growth was 25°C and 7.0, respectively. Pinhead initiation was observed at day 10 after inoculation, but the fully developed, branched, and coral to club-shaped fruiting bodies could be observed after 30 days of inoculation. Proximate analysis indicated that carbohydrates are the major constituents (50.2%) of the fruit bodies, along with a lower quantity of protein (4.46%), crude fat (6.4%), and crude fiber (1.55%). Vitamin D content of I. cicadae was 3,605.84 IU/g. Radical scavenging activity based on the DPPT (1,1-diphenyl-2-picrylhydrazyl) assay was 21.2%. ABTS [2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid] and potassium ferricyanide reducing activity were quite high, at around 93% and 99.3%, respectively. The findings of this study provide insight into the biochemical constituents of I. cicadae and its cultivation practices for further exploitation of this mushroom at a larger scale.Microwave-assisted extraction (MAE) is a highly selective method that reduces the time and number of solvents used, has high efficiency, and is environmentally friendly. This study aimed to evaluate the antioxidant, anticancer, and antibacterial activities of Iraqi Terfezia claveryi desert truffle extract and silver nanoparticles by MAE using water and hexane as extraction solvents. Chemical compositions were identified by gas chromatography-mass spectrometry. Silver nanoparticles were characterized by field emission scanning electron microscopy. Antioxidant activity was assessed with the 2,2-diphenyl-1-picrylhydrazyl method. Antibacterial activity was evaluated against four types of pathogenic bacteria, and anticancer activity was also assessed. The T. claveryi hexane extract showed approximately 18 bioactive compounds, with an antioxidant half-maximal inhibitory concentration of 6.896 μg/mL. This extract also had significant antibacterial action against Escherichia coli and Pseudomonas aeruginosa. A cytotoxicity test of the hexane extract showed moderate toxicity against hepatocellular carcinoma mouse (HCAM) liver cancer cells. Acridine orange/ ethidium bromide staining showed apoptosis and DNA damage in HCAM cells treated with the hexane extract. The results of this study suggest that components of the T. claveryi wild desert truffle could have important nutrition functions that might support the immune system in fighting liver diseases when taken in daily regular doses.In this study, acetone and water extracts obtained from edible mushrooms, Rhizopogon luteolus Fr. and Rh. roseolus (Corda) Th. Total antioxidant capacity (TAC) and total oxidative stress (TOS) levels were examined on human peripheral lymphocyte culture treated with the respective extracts. Levels of genetic damage and cytotoxic effects of the respective extracts on lymphocytes were also tested. In general, when TAC levels of the extracts on cells were examined, a concentration-dependent increase was observed; a negative correlation was found between TOS data and concentration. Finally, the inhibition modes of extracts on α-amylase and α-glucosidase were detected by kinetic assay. The results showed that TPC, TFC, and the content of phenolic compounds in the extracts were different. EAF contained the highest contents of both TPC and TFC and exhibited strongest inhibitory effects on α-amylase and α-glucosidase, with half maximal inhibitory concentration (IC50) values of 0.
