  • Objective the aim of the present study was to compare the effectiveness of three approaches [the seasonal auto-regressive integrated moving average (SARIMA) model, the nonlinear autoregressive exogenous (NARX) artificial neural networks and Wood's model] to the prediction of milk yield during lactation. Methods the dataset comprised monthly test-day records from 965 Polish Holstein-Friesian Black-and-White primiparous cows. The milk yields from cows in their first lactation (from 5 to 305 days in milk) were used. Each lactation was divided into ten lactation stages of approximately 30 days. Two age groups and four calving seasons were distinguished. The records collected between 2009 and 2015 were used for model fitting and those from 2016 for the verification of predictive performance. Results no significant differences between the predicted and the real values were found. The predictions generated by SARIMA were slightly more accurate, although they did not differ significantly from those produced by the NARX and Wood's models. SARIMA had a slightly better performance, especially in the initial periods, whereas the NARX and Wood's models in the later ones. Conclusion the use of SARIMA was more time-consuming than that of NARX and Wood's model. The year was divided into three seasons cool/dry (from November to February), hot/dry (from March to June), and wet (from July to October), based on the maximal temperature and rainfall in Pursat, to analyze the relationship between ambient conditions and the reproductive performance of cows. Body condition scores (BCS) and feeding schemes were also analyzed in these seasons. Results The number of cows receiving AI was significantly higher in the cool/dry season than the wet season. The number of calving and calving rate were significantly higher in cows receiving AI in the cool/dry season compared with the hot/dry and wet seasons. The cows showed higher BCSs in the cool/dry season compared to the hot/dry and wet seasons probably due to the seasonal changes in the feeding schemes These cows grazed on wild grasses in the cool/dry season but fed with a limited amount of grasses and straw in the hot/dry and wet seasons. Conclusion The present study suggests that the low number of cows receiving AI, low number of calving and low calving rate could be mainly due to poor body condition as a result of the poor feeding schemes during the hot/dry and wet seasons. The ImC animals had higher flight speeds (P=0.022) and tended to have higher cortisol levels (P= 0.059) than the NoC animals. Animals fed ZH and RH did not differ in cortisol levels, respiratory rate, rectal temperature, temperature measured by IRT, or temperament behaviour. Methods A double 3 × 3 Latin square design was used with three PAP treatments (control, powdered and liquid PAP) and two adaptation protocols (adapted, unadapted; applied to the square). Adapted animals were transitioned for 2 weeks from an all-forage to an 80% concentrate diet, while unadapted animals were switched abruptly. Results Interactions between sampling time and adaptation were observed; 12 h after feeding, the adapted group had lower ruminal pH and greater total short chain fatty acid concentrations than the unadapted group, while the opposite was observed after 24 h. Acetatepropionate ratio, molar proportion of butyrate and ammonia nitrogen concentration were generally greater in adapted than unadapted cattle up to 36 h after feeding. Adaptation promoted 3.5 times the number of Entodinium protozoa but copy numbers of S. bovis and Fibrobacter succinogens genes in rumen fluid were not affected. However, neither liquid nor powdered forms of PAP altered rumen acidosis variables in adapted or unadapted animals. Conclusion Adaptation of cattle to highly fermentable carbohydrate diets promoted a more stable ruminal environment, but PAP was not effective in this study in which no animal experienced acute or sub-acute rumen acidosis.

  • One can roughly judge the adaptability of bioleaching of silicon in EMR using Paenibacillus mucilaginosus if the main form of silicate minerals in EMR is determined.In this work, a miniaturized flow-through leaching test is presented for rapid screening of potential chemical extractants to explore the bioaccessibility of lead (Pb) in contaminated shooting range soils in Valkeala, Finland. The method combines the versatility of microcolumn-based extraction methods with on-line inductively coupled plasma optical emission spectrometry (ICP OES) analysis for expedient assessment of the magnitude of the bioaccessible pools and the leaching kinetics of lead from polluted soils under variable physicochemical scenarios. Acids and salt solutions were studied as potential extractants. To elucidate the potential exposure of PPCPs in drinking water, this study assessed the occurrences and treatment efficiencies of PPCPs in the drinking water of Taiwan. Raw and finished water samples collected from five main drinking water treatment plants (DWTPs) in February, June, and November 2018 were analyzed. Furthermore, laboratory-scale water treatment processes were conducted to evaluate the treatment efficiencies of these chemicals. Most of the water samples from the DWTPs had a low concentration ( less then 30 ng/L) of PPCPs. Only samples from a DWTP was observed to have higher concentration of ibuprofen (55.6 ng/L), benzophenone (92.5 ng/L), caffeine (390.5 ng/L), and diethyltoluamide (DEET) (434.9 ng/L) in raw water than others. The results of laboratory simulations indicated that the pre-chlorination process was the key step responsible for the removal of PPCPs in conventional water treatment processes, which can remove most of the hormone treatment products, parabens, oxybenzone, and acetaminophen in water sources. However, the filtration process with anthracite as a medium could remove some of the parabens (approximately 11.9%-41.2%), hormones (approximately 18.2%-44.8%), suntan lotions (37.5%-68.8%), and naproxen (30.1%) from Milli-Q water. The removal efficiencies of the aforementioned chemicals were marginally lower in raw water. However, analgesics, caffeine, and DEET cannot be removed effectively through conventional drinking water treatment.Dichloroacetonitrile (DCAN) is one of the most toxic and common nitrogenous disinfection by-products in water treatment. It is necessary to understand how this compound can be removed. In this study, the effectiveness of vacuum ultraviolet (VUV) at 185 nm was evaluated to destroy DCAN. When water is exposed to VUV, hydroxyl radicals (HO•), hydrogen atoms (H•), and hydrated electrons (eaq-) are generated. The individual contributions of these reactive species to DCAN degradation were distinguished using multiple scavengers. The results showed that eaq- was the most important species for DCAN degradation. The second-order rate constant for eaq- reacting with DCAN was calculated to be 3.16 × 1010 M-1s-1 using a quantitative structure-activity relationship (QSAR) method adopted from previous study, and determined to be 3.76 (±0.02) × 1010 M-1s-1 by competition kinetics. Although dissolved oxygen (DO) at 8 mg/L consumed 86% eaq-, the rest of eaq- still led to 93% removal of DCAN within 20 min. Chloride was the major inorganic product of DCAN degradation, while nitrate and nitrite were minor products. Quantum chemical calculation and mass balance calculation under an oxygen free condition further suggested that cleavage of C-Cl bonds was the major pathway by eaq- attack. This study demonstrated the significant role of eaq- in micropollutant destruction during VUV treatment.Systemic neurotoxic insecticides are widely used to control aphid pests worldwide and their potential non-target effects on aphid predators are often unknown. Behavioral responses linked to biological control services are crucial when assessing the compatibility of chemicals with biocontrol organisms. This is particularly relevant for insecticides at low and sublethal concentrations. We studied the acute toxicity and the sublethal effect on the voracity of the generalist predator Harmonia axyridis (Coleoptera Coccinellidae) caused by the exposure to three systemic insecticides routinely used against aphids. The tested insecticide concentrations were the Lethal Concentration 50% (LC50), 20% (LC20) and 1% (LC1) estimated for the target pest Aphis gossypii (Hemiptera Aphididae) in a companion study. The survival and the voracity differed among the tested chemicals and concentrations, but only thiamethoxam at LC50 caused a significant predator mortality, and individuals that survived showed a reduced predation rate. The predators showed a density independent functional response after the exposure to most of the insecticide-concentration combinations, while an inverse density dependence of the prey consumption rate was observed for coccinellids exposed to sulfoxaflor and thiamethoxam at their lowest tested concentration. The estimated parameters, i.e., the attack rate and the prey handling time, were affected at higher concentrations by both imidacloprid and sulfoxaflor. These findings stress the importance of carefully evaluating side effects of insecticides at very low concentrations on beneficial arthropods in the risk assessment schemes for sustainable pest control programmes.Biodegradable mulch films are an alternative to polyethylene films used in agriculture for weed control, improving crop productivity. This change could minimize the residue production and costs related to the final disposal. Nevertheless, the environmental safety of these biodegradable products is scarcely investigated. In this work, samples of poly(butylene adipate-co-terephthalate)-PBAT mulch films, with and without UV stabilizer additives, were prepared. Aqueous extracts of soil samples, where mulch films were disposed, were investigated using bioassays with Lactuca sativa, Allium cepa, and cell culture HepG2/C3A. As PBAT is expected to suffer photodegradation and biodegradation, soil samples mixed with films before and after these processes were evaluated. Soil aqueous extracts promoted root grown (mainly hypocotyl) of L. sativa, probably due to presence of nutrients. So, to evaluate toxicity potential, in this case it was necessary to use aqueous extract prepared with soil instead of ultrapure water as the control.

  • It was found that the magnetization transfer rate and chemical shift anisotropy for different functional groups present in patiromer play a large role when optimizing parameters for spectral acquisition. External stressors alter the mitochondrial dynamics of RGCs, leading to ROS buildup, energy shortages, decreased biogenesis and increased mitophagy, and ultimately axon degeneration and ganglion cell death. New therapeutic alternatives targeting these newly understood pathophysiological changes in the mitochondria and directly addressing the genetic mutations involved in LHON are being developed.BACKGROUND The inverse relationship between body mass index (BMI) and natriuretic peptide levels complicates the diagnosis of heart failure (HF) in obese patients. METHODS We performed a comprehensive echocardiographic evaluation within 24 h from hospital admission in patients with HF, including lung B-lines and IVC diameter, and studied their relationship with BMI and outcome. RESULTS 216 patients (median age 81 (77-86) years) were enrolled. Median number of B-lines was 31 (IQR 26-38), median IVC diameter was 23 (22-25) mm and median BNP 991 (727-1601) pg/mL. BMI was inversely correlated with B-lines (r = - 0.50, p  less then  0.001), but not with IVC diameter (r = - 0.04, p = 0.58). Compared to overweight patients (BMI 25-29.9 kg/m2; n = 100) or with a normal BMI (BMI  less then  25 kg/m2; n = 59), obese patients (BMI ≥ 30 kg/m2; n = 57) had lower B-lines [28 (24-33) vs 30 (26-35), and vs 38 (32-42), respectively; p  less then  0.001] but similar IVC diameter. During the first 60 days of follow-up, there were 53 primary events 29 patients died and 24 had a HF-related hospitalisation. B-lines and IVC diameter were independently associated with an increased risk. However, B-lines were less likely to predict outcome in the subgroup of patients with a BMI ≥ 30 kg/m2. CONCLUSIONS Assessment of IVC diameter or B-lines in patients admitted with AHF identifies those at greater risk of death or HF readmission. Non-fatal strokes were absolutely infrequent during follow-up (0 vs. 1.1%). Severe non-fatal bleedings were observed only among CKD patients (1.4 vs. 0%; p = 0.021). OBJECTIVES To characterize ICI-mediated myocarditis and re-introduction to immunotherapy. METHODS During 2019, 849 patients were treated with ICI at Tel Aviv Sourasky Medical Center for the diagnosis of lung adenocarcinoma, gastric adenocarcinoma, urothelial carcinoma, and hepatocellular carcinoma. Overall, seven (0.8%) patients were diagnosed with ICI-mediated myocarditis, according to the European Society of Cardiology guidelines of myocarditis 2013. We retrospectively evaluated their presentation, severity, and clinical outcomes. RESULTS Among the seven patients, only one had a history of cardiac disease. The majority were diagnosed with lung adenocarcinoma and treated with anti-programmed death-1 antibody. All patients were treated with single-agent ICI. Most patients presented with cardiac symptoms, elevated troponin and typical cardiac magnetic resonance; however, only three had reduced ejection fraction. Overall, three patients were chosen for re-introduction with concomitant low dose steroids and weekly troponin follow-up. Two patients diagnosed with grade I and II renewed therapy successfully with no recurrence of symptoms and improvement in disease burden. The one patient diagnosed with grade III developed worsening of cardiac symptoms after the 1st cycle and, therefore, therapy was interrupted permanently. CONCLUSIONS ICI-mediated myocarditis is potentially fatal and leads to permanent interruption of life-saving cancer therapy. The current data suggest that re-introduction may be considered in low-grade patients; however, a better definition of the diagnosis and grading is needed.

  • BACKGROUND Fetal programming during in utero life defines the set point of physiological and metabolic responses that lead into adulthood; events happening in "the first 1,000 days" (from conception to 2-years of age), play a role in the development of non-communicable diseases (NCDs). The infant gut microbiome is a highly dynamic organ, which is sensitive to maternal and environmental factors and is one of the elements driving intergenerational NCDs' transmission. The A.MA.MI (Alimentazione MAmma e bambino nei primi MIlle giorni) project aims at investigating the correlation between several factors, from conception to the first year of life, and infant gut microbiome composition. We described the study design of the A.MA.MI study and presented some preliminary results. METHODS A.MA.MI is a longitudinal, prospective, observational study conducted on a group of mother-infant pairs (n = 60) attending the Neonatal Unit, Fondazione IRCCS Policlinico San Matteo, Pavia (Italy). RESULTS We assessed the Endurance Shuttle - Nine Hole Peg Test (ESNHPT), - Box and Block Test (ESBBT) and - Walk Test (ESWT) in 61 patients with SMA types 2-4, 25 healthy controls (HC) and 15 disease controls (DC). Convergent validity, discriminative validity and test-retest reliability were investigated. Additionally, we compiled the Endurance Shuttle Combined Score (ESTCS) by selecting the most relevant endurance test of each individual. 54, 70 and 73% of patients with SMA demonstrated increased fatigability on the ESNHPT, ESBBT and the ESWT. Endurance response in SMA was characterized by a decrease in muscle strength, an increase in muscle fatigue and an increase in motor adaptions, thereby confirming convergent validity. Patients with SMA showed increased drop-out rates and a shorter endurance time compared to HC and DC demonstrating good discriminative validity. The virulent Mtb strain, Rv caused double strand breaks (DSBs) whereas the non-virulent Ra strain triggered single stranded DNA generation. Accumulation of DSBs mediated through Rv activates ATM-Chk2 pathway of DNA damage response (DDR) signaling, resulting in altered cell cycle. Instead of the classical ATM-Chk2 DDR, Mtb gains survival advantage through ATM-Akt signaling cascade. Notably, in vivo infection with Mtb led to sustained DSBs and ATM activation during chronic phase of tuberculosis. Furthermore, the association of patient f0 stability and therapist coregulation with patients' perceived rationale plausibility was analyzed. N = 197 therapy videos-used to deduct f0-from a multicenter randomized controlled trial evaluating therapist-guided exposure on CBT outcome were analyzed post hoc. Plausibility of the exposure rationale was assessed by patients after its development. This trial-specific rating aggregates plausibility ratings for every manual component in the development of the exposure rationale and showed good internal consistency (Cronbach's alpha = .85). Stability in f0 and its coregulation were calculated using cross-lagged Actor-Partner Interdependence Models (APIMs), and APIM dyad estimates were associated with plausibility using linear regression analyses. Analyses indicated a relative stability in emotional arousal within both patients and therapists. Therapists' f0 had a significant effect on patients in that with therapist covariation, patients' f0 departed from their equilibrium level, while patients' f0 had no effect on therapists. Study 1A (n = 707) provides exploratory factor analytic evidence supporting the separability of power and color evasion. Study 1B (n = 710) provides confirmatory evidence of this factor structure and evidence of discriminant validity. In Study 1B, 3 latent profiles based on power and color evasion were identified acknowledgers (low color evasion, low power evasion), evaders (high color evasion, average power evasion), and deniers (average color evasion, average power evasion), which differed on relevant variables (e.g., modern racism, support for affirmative-action). However, efforts to address this question have been hampered by cross-study inconsistencies in how item valence is assessed. Thus, the goal of this study was to establish reference ratings for the valence of RMET items. In Study 1, we recorded valence ratings for each RMET item with a large sample of raters (n = 164). We illustrated how valence categories are essentially arbitrary and largely influenced by sample size. In addition, valence ratings were continuously distributed, further questioning the validity of imposing categorical distinctions. In Study 2, we used an archival dataset to demonstrate how the different categorization schemes resulted in conflicting conclusions about the association between item valence and RMET performance.

The Event History Calendar is a potential tool for research and care practices to comprehend the needs of adolescents, with reminder of key personal events, culturally and socially specific. The comprehension of retrospective data referring to activities, behaviors, experiences and transitions of life, in certain periods of time, enables dialogue and new understandings about the history of adolescents. FINAL CONSIDERATIONS the Event History Calendar provides nursing professionals with an expansion of their practice in educational, relational, and communicative dimensions, as well as to instruct care planning and management.OBJECTIVES to present the nurses' experience with technological tools to support the early identification of sepsis. METHODS experience report before and after the implementation of artificial intelligence algorithms in the clinical practice of a philanthropic hospital, in the first half of 2018. RESULTS describe the motivation for the creation and use of the algorithm; the role of the nurse in the development and implementation of this technology and its effects on the nursing work process. FINAL CONSIDERATIONS technological innovations need to contribute to the improvement of professional practices in health. Thus, nurses must recognize their role in all stages of this process, in order to guarantee safe, effective and patient-centered care. In the case presented, the participation of the nurses in the technology incorporation process enables a rapid decision-making in the early identification of sepsis.This study was designed to investigate the expression of RBM8A protein in patients with gastric cancer (GC) and to explore its correlation with clinical pathological features as well as prognosis. One hundred pairs of gastric carcinoma tissues and adjacent tissues from patients undergoing gastrectomy for GC were included in this study. The protein expression level of RBM8A was determined by immunohistochemistry using tissue microarrays. We also detected the mRNA expression level of RBM8A in 16 pairs of gastric carcinoma tissues and adjacent tissues. Meanwhile, we predicted the potential correlation between RBM8A and tumor stages as well as survival condition in patents with GC based on The Cancer Genome Atlas (TCGA) database. The correlation of RBM8A with the clinical pathological features and prognosis of the 100 patients with GC was also elucidated. The expression level of RBM8A was significantly higher in gastric carcinoma tissues compared to the adjacent tissues. The protein level of RBM8A was correlated with tumor size (P=0.031), depth of invasion (P less then 0.001), lymph node metastasis (P less then 0.001), TNM stage ( less then 0.001), and distant metastasis (P=0.001). Patients with increased RBM8A expression (P less then 0.0018, 95%CI=0.322-0.871), higher TNM stage (P less then 0.001, 95%CI=4.990-11.283), and lymph node metastasis (P less then 0.001, 95%CI=2.873-4.002) had a lower overall survival. Quantitative polymerase chain reaction (qRT-PCR) was utilized to detect the expression levels of ZEB1-AS1, microRNA-216a-5p (miR-216a-5p), and bone morphogenetic protein 7 (BMP7). Western blot assay was used to evaluate the protein levels of BMP7, epithelial-to-mesenchymal transition (EMT)-related proteins, and fibrosis markers. The proinflammatory cytokine interleukin 1 beta (IL-1β) is abundantly expressed in OA and plays a crucial role in cartilage remodeling, although its role in the activity of chondrocytes in cartilage and subchondral remodeling remains unclear. We demonstrate that in p53 deleted colon cancer cells, LQ1 causes nucleolar stress, impairs ribosomal RNA processing, leading to the accumulation of pre-ribosomal RNAs, arrests cells in the G2/M phase and induces early apoptosis. Serum lipid abnormalities and renal ectopic lipid accumulation have been associated with the development of kidney diseases, in particular diabetic nephropathy. Chronic hyperinsulinemia, often seen in type 2 diabetes, plays a crucial role in blood and liver lipid metabolism abnormalities, thus resulting in increased non-esterified fatty acids (NEFA). Excessive lipid accumulation alters cellular homeostasis and activates lipogenic and glycogenic cell-signaling pathways. Recent evidences indicate that both quantity and quality of lipids are involved in renal damage associated to lipotoxicity by activating inflammation, oxidative stress, mitochondrial dysfunction, and cell-death. The pathological effects of lipotoxicity have been observed in renal cells, thus promoting podocyte injury, tubular damage, mesangial proliferation, endothelial activation, and formation of macrophage-derived foam cells. Therefore, this review examines the recent preclinical and clinical research about the potentially harmful effects of lipids in the kidney, metabolic markers associated with these mechanisms, major signaling pathways affected, the causes of excessive lipid accumulation, and the types of lipids involved, as well as offers a comprehensive update of therapeutic strategies targeting lipotoxicity.

  • In colon cancer, the prognostic value of macrophages is controversial, and it is still unknown how hypoxia modulates macrophage-cancer cell crosstalk. To unravel this, co-cultures of human primary macrophages and colon cancer cells were performed at 20% and 1% O2, followed by characterization of both cellular components. Different colon cancer patient cohorts were analyzed for hypoxia and immune markers, and their association with patient overall survival was established. A positive correlation between HIF1A and CD68 in colon cancer patients was identified but, unexpectedly, in cases with higher macrophage infiltration, HIF1A expression was associated with a better prognosis, in contrast to breast, gastric, and lung cancers. The intervention included drawing pictures related to three topics (1) war-related stressors, (2) coping resources, and (3) integration of the stressful image and the resources drawing. The major aims of the study were (1) to examine whether significant changes occurred in MHP distress levels after the intervention; (2) to explore the narratives of the three drawing and their compositional characteristics; and (3) to determine which of selected formats of the integrated drawing and compositional transformations of the stressful image are associated with greater distress reduction. Results indicate that MHP distress levels significantly decreased after the intervention. This stress-reducing effect was also reflected in differences between the compositional elements of the 'stress drawing' and the 'integrated drawing,' which includes elements of resources. Reduced distress accompanied compositional transformations of the stressful image. The first group underwent AAAs (n = 26), the second group chose a relaxation technique (n = 28), and the last one was a control group (n = 39). This shows that new therapeutic agents targeting several factors at the same time are needed.   • With the increasing necessity of animal models in biomedical research, there is a vital need to harmonise findings across species by establishing similarities and differences in rodent and primate neuroanatomy. Using connectivity fingerprint matching, we compared cortico-striatal circuits across humans, non-human primates, and **** using resting-state fMRI data in all species. Our results suggest that the connectivity patterns for the nucleus accumbens and cortico-striatal motor circuits (posterior/lateral putamen) were conserved across species, making them reliable targets for cross-species comparisons. However, a large number of human and macaque striatal voxels were not matched to any mouse cortico-striatal circuit (mouse->human 85% unassigned; mouse->macaque 69% unassigned; macaque->human; 31% unassigned). These unassigned voxels were localised to the caudate nucleus and anterior putamen, overlapping with executive function and social/language regions of the striatum and connected to prefrontal-projecting cerebellar lobules and anterior prefrontal cortex, forming circuits that seem to be unique for non-human primates and humans. © 2020, Balsters et al.In hypoxic stress conditions, glycolysis enzymes assemble into singular cytoplasmic granules called glycolytic (G) bodies. METHODS We analyzed data for 63,299 adolescents aged 13 to 17 years in the National Immunization Survey-Teen, 2015-2017. We compared HPV vaccination initiation coverage estimates (≥1 dose) in the DRA region with coverage estimates in areas in the 8 Delta states outside the DRA region and non-Delta states. We examined correlates of HPV vaccination coverage initiation and reasons parents did not intend to vaccinate adolescents. RESULTS Vaccination rates in the DRA region (n = 2,317; 54.3%) and in Delta areas outside the DRA region (n = 6,028; 56.2%) were similar, but these rates were significantly lower than rates in non-Delta states (n = 54,954; 61.4%). Inside the DRA region, reasons for parents' vaccine hesitancy or refusal were similar to those expressed by parents in the Delta areas outside the DRA region. Some parents believed that the vaccine was not necessary or had concerns about vaccine safety. CONCLUSION HPV vaccination coverage in the DRA region is similar to coverage in other Delta counties and parishes, but it is significantly lower than in non-Delta states. Widely adopted individual precautionary measures, coupled with early government actions, might slow transmission early in the outbreak.Context Few cases of neuromyelitis optica spectrum disorder (NMOSD) with an onset older than 75 years old have been reported.Finding Herein, we report an 81-year-old Chinese male initially suspected of acute stroke but was ultimately diagnosed with NMOSD.Conclusion Even in the elderly, a diagnosis of NMOSD should be considered for patients with myelitis, especially those with longitudinally extensive spinal cord involvement. Testing for aquaporin 4 antibody in this scenario is recommended for further confirmation.
