  • The purpose of this study was to improve the repulsion ability of sulfonated poly(ether ether ketone) (SPEEK) membrane for the vanadium ions crossover. For this purpose graphene oxide (GO) nanosheet and titanium dioxide (TiO₂) nanoparticles were employed into the polymer matrix to prepare SPEEK/GO/TiO₂ hybrid membrane via solution-casting method for vanadium redox flow battery (VRFB). The morphology, permeability of vanadium ions and device performance of asprepared membrane were investigated and discussed. It was observed that with the barrier block effect by the filler, the VRFB single cell with the optimized SPEEK/GO/TiO₂ hybrid membrane exhibited high coulombic efficiency (~99%), excellent energy efficiency (~85%) and vigorous cyclability (~97.2% capacity retention after 100 cycles). Moreover, the VRFB cell with this blend membrane showed lower vanadium ions permeability than Nafion 212 or pure SPEEK membranes. However, the Vth, p decreases as the Ge mole fraction of the Si1-xGex S/D becomes greater than 0.5 due to the higher valence band energy (Ev) of the NS channels. On the other hand, the Vth of n-type FETs (NFETs) (Vth, n) consistently increases as the C mole fraction of the Si1-xCx S/D increases due to monotonously retarded phosphorus dopants diffusing from the Si1-xCx S/D into the NS channels. On the other hand, the Vth, p and Vth, n consistently decreases and increases, respectively, as Si/Si0.7Ge0.3 intermixing becomes severer because both Ev and conduction band energies (Ec) of the NS channels become higher. In addition, the Vth, p variations are more sensitive to the Si/Si0.7Ge0.3 intermixing than the Vth, n variations because the Ge mole fraction in NS channels affects the Ev remarkably rather than the Ec. The device using a TiO₂/SiN gate-insulator shows better gate leakage current characteristics than the device with only TiO₂ gate-insulator because of the high quality SiN gate-insulator. Therefore, the device using a dual gate-insulator can overcome disadvantages of a device using only TiO₂ gate-insulator. To better predict the power consumption and the switching speed, we simulated the specific on-resistance (Ron, sp) according to the gate-to-drain distance (LGD) using the two-dimensional ATLAS simulator.   • AIM Peyronie's disease (PD) or plastic induration of the penis, require complete evaluation of plaques in order to decide the best therapeutic option for patient. The purpose of this study is to compare the findings of three-dimensional ultrasound (3D US) and two-dimensional ultrasound (2D US) in patients with PD. MATERIALS AND METHODS Twenty patients with PD aged 30 to 72 years were included in study. The examination was performed with a 12 MHz linear probe, using 2D US and 3D US. Localization and size of plaques were determined and time needed for imagine acquisition was determined in every case. RESULTS 3D ultrasound permits the visualization of the entire plaque in the coronal plane of plaque with its precise measurements. No statistical difference in plaque dimensions and its surface area assessment using 3D US and 2D US was found (127.72 mm² vs. 128.74 mm², p>0.05). The possibility to perform detailed analysis of the acquired images using generated digital cube reduced the average duration of the acquisition to 69.8 seconds (median 64 seconds) for 3D US vs. 151.25 seconds (median 145.5 seconds) for 2D US (p less then 0.05). A supplementary plaque was detected using 3D US. CONCLUSIONS 3D US seems to be a valuable complement of 2D US for patients with PD. The acquisition time is significantly reduced using 3D US comparing to 2D US and thus it is more comfortable for the patient.AIMS To examine the diagnostic accuracy of plain radiography, abdominal ultrasonography (US), and their combination in pediatric patients with suspected gastrointestinal (GI) tract obstruction. MATERIAL AND METHODS A cohort of 48 patients (age, 0-14 years, 27 boys) with clinical manifestations of GI tract obstruction underwent plain radiography and abdominal US examination. The final diagnoses were based on intraoperative findings, rectal biopsies (in Hirschsprung's disease), or adequate follow-ups. RESULTS The GI tract obstruction was diagnosed in 40 patients. The sensitivity, specificity, positive predictive value and negative predictive value of plain radiography in diagnosing GI tract obstruction were 87.5%, 75.0%, 94.6%, and 54.6%, respectively. The corresponding values were 95%, 100%, 100%, and 80%, respectively when US was used alone; and 97.5%, 100%, 100% and 88.9%, respectively when radiography and US were used together. Except for two patients (one with Hirschsprung's disease and the other with massive peritonitis), US detected the underlying causes of obstruction correctly in all patients. CONCLUSIONS US is a highly sensitive and specific modality in diagnosing pediatric GI tract obstructions, as well as their causes. The combination of plain radiography and US further increase the diagnostic sensitivity and negative predictive value.AIM Τo examine the inter- and intra-muscular differences in the anatomical cross-sectional area (CSA) of the quadricep muscles, using extended - field of view (EFOV) ultrasonography (US). MATERIAL AND METHODS Panoramic transverse US images of the thigh were acquired from 10 young participants at five different locations across the thigh, in two sessions, spaced a week apart. The CSA of the vastus medialis (VM), rectus femoris (RF), vastus intermedius (VI), vastus lateralis (VL) and tensor vastus intermedius (TVI) was quantified. RESULTS The intraclass correlation coefficients ranged from 0.75 to 0.97 and the standard error of measurement ranged from 0.78% to 6.61%, indicating high test-retest reliability. Cortical developmental regionalization, i.e., the landscape of cortical heterogeneity in development, reflects the organization of underlying microstructures, which are closely related to the functional principles of the cortex. Therefore, prospecting early cortical developmental regionalization can provide neurobiologically meaningful units for precise region localization, which will advance our understanding on brain development in this critical period. However, due to the absence of dedicated computational tools and large-scale datasets, our knowledge on early cortical developmental regionalization still remains intact. To fill both the methodological and knowledge gaps, we propose to explore the cortical developmental regionalization using a novel method based on nonnegative matrix factorization (NMF), due to its ability in analyzing complex high-dimensional data by representing data using several bases in a data-driven way. Specifically, a novel multi-view NMF (MV-NMF) method is proposed, in which multiple distinct and complementary cortical properties (i.e., multiple views) are jointly considered to provide comprehensive observation of cortical regionalization process. To ensure the sparsity of the discovered regions, an orthogonal constraint defined in Stiefel manifold is imposed in our MV-NMF method. Meanwhile, a graph-induced constraint is also included to improve the compactness of the discovered regions. Capitalizing on an unprecedentedly large dataset with 1,560 longitudinal MRI scans from 887 infants, we delineate the first neurobiologically meaningful representation of early cortical regionalization, providing a valuable reference for brain development studies.Glioblastoma (GBM) is the most common and aggressive form of malignant glioma in adults with a median overall survival (OS) time of 16-18 months and a median age of diagnosis at 64 years old. Recent work has suggested that depression and psychosocial distress are associated with worse outcomes in patients with GBM. We therefore hypothesized that the targeted neutralization of psychosocial distress with selective serotonin reuptake inhibitor (SSRI) antidepressant treatment would be associated with a longer OS among patients with GBM. To address this hypothesis, we retrospectively studied the association between adjuvant SSRI usage and OS in GBM patients treated by Northwestern Medicine-affiliated providers. The medical records of 497 GBM patients were analyzed after extraction from the Northwestern Medicine Enterprise Data Warehouse. Data were retrospectively studied using a multivariable Cox model with SSRI use defined as a time-dependent variable for estimating the association with OS. Of the 497 patients, 315 individuals died, while 182 were censored due to the loss of follow-up or were alive at the end of our study.

    The purpose of this study was to improve the interpretability of the Non-arthritic Hip Score (NAHS) by determining the minimal clinically important difference (MCID), patient acceptable symptomatic state (PASS), and substantial clinical benefit (SCB) after hip arthroscopy for femoroacetabular impingement. The secondary aim was to identify variables associated with achievement of the thresholds.

    Patients who underwent hip arthroscopy for femoroacetabular impingement and completed postoperative questionnaires between August 2019 and March 2020 were included. Patients were excluded if they underwent previous ipsilateral hip surgery, underwent gluteus medius repair, or had a previous hip condition. The MCID, PASS, and SCB thresholds were calculated for the NAHS at minimum 1-, 2-, and 5-year follow-up. Distribution- and anchor-based methods with receiver operating characteristic analysis were used to determine the thresholds. Multivariate logistic regression was performed to determine predictors of achieving tlue ranged from 91.9 to 94.4. The preoperative NAHS was found to be positively associated with achievement of the PASS and inversely related to achievement of the MCID.

Melanocortins and their receptors (MCRs) exert multiple beneficial effects in pulmonary inflammation. We tested the idea that treatment with the synthetic α-melanocyte-stimulating hormone analogue [Nle4,D-Phe7]-α-MSH (NDP-MSH) during ex vivo lung perfusion (EVLP) could exert positive influences in lungs exposed to different injuries. Rats were assigned to one of the following protocols (N = 10 each) 1) ischemia/reperfusion (IR) or 2) cardiac death (CD) followed by ex vivo perfusion. NDP-MSH treatment was performed in five rats of each protocol before lung procurement and during EVLP. Pulmonary function and perfusate concentration of gases, electrolytes, metabolites, nitric-oxide, mediators, and cells were assessed throughout EVLP. ATP content and specific MCR expression were investigated in perfused lungs and in biopsies collected from rats in resting conditions (Native, N = 5). NDP-MSH reduced the release of inflammatory mediators in perfusates of both the IR and the CD groups. Treatment was likewise associated with a lesser amount of leukocytes (IR p = 0.034; CD p = 0.002) and reduced lactate production (IR p = 0.010; CD p = 0.008). In lungs exposed to IR injury, the NDP-MSH group showed increased ATP content (p = 0.040) compared to controls. In CD lungs, a significant improvement of vascular (p = 0.002) and airway (Ppeak p less then 0.001, compliance p less then 0.050, pO2 p less then 0.001) parameters was observed. Finally, the expression of MC1R and MC5R was detected in both native and ex vivo-perfused lungs. Using this method, the original intensity and phase information of the object to be observed can be correctly restored. Therefore, we propose a novel method to calculate the degree of intensity and phase modulation by calculating the direct and alternating current components obtained from the output of the sum and difference of the two photodetectors. The degree of spatial frequency modulation is corrected according to the electrical signal frequency to detect transparent or unstained cells. We first performed laser scanning of an object. Then, signals were detected using two photodetectors placed in the far-field, separated by the optical axis as the boundary. The output signals of the photodetectors were processed and the intensity and phase were unambiguously separated, thus allowing the visualization of the phase information of the transparent bodies and unstained cells. Spatial frequency correction was performed to correct the modulation. Our method successfully separated the information related to the intensity and optical path difference (OPD). In future work, by accurately correcting the intensity and OPD, it will be possible to separate the absorption rate from the ratio of the irradiation light intensity to the observed intensity and to separate the OPD into the refractive index and the thickness information. This method allows the accurate determination of these parameters in a noninvasive manner.
    Although group studies provide some support for the material-specific model of memory function, there are considerable individual variations in memory function in people with temporal lobe epilepsy, even in those with the same underlying pathology. In this proof-of-concept study, we examined the sensitivity and specificity of a single measure of an individual's relative strength for the encoding of verbal or visual learning.

    Six hundred ninety-two patients with left hemisphere language dominance and unilateral hippocampal sclerosis completed verbal and visual encoding tasks with similar test structures as part of their presurgical evaluation. Three hundred one patients had right hippocampal sclerosis (RHS), and 391 patients had left hippocampal sclerosis (LHS). A memory specialization index (MSI) was calculated by subtracting the Visual Learning z-score from the Verbal Learning z-score. A positive value on the MSI indicates a relative strength in verbal learning. A negative score indicates a relative strevisual and verbal encoding may evolve with age and duration of epilepsy, and clinicians should be aware of these factors when interpreting the lateralizing significance of test scores, particularly in a presurgical setting.

We applied permutation distribution clustering to time series of successive shift lengths, between-shift rest periods, and shift starting times to identify clusters of similar working hour patterns over time. We examined associations of clusters spanning on average 23 months with SA during the following 23 months.

    We identified eight distinct working hour patterns in shift work (i) regular morning (M)/evening (E) work, weekends off; (ii) irregular M work; (iii) irregular M/E/night (N) work; (iv) regular M work, weekends off; (v) irregular, interrupted M/E/N work; (vi) variable M work, weekends off; (vii) quickly rotating M/E work, non-standard weeks; and (viii) slowly rotating M/E work, non-standard weeks. The associations of these eight working-hour clusters with risk of future SA varied. The cluster of irregular, interrupted M/E/N work was the strongest predictor of increased SA (days per year) with an incidence rate ratio of 1.77 (95% confidence interval 1.74-1.80) compared to regular M/E work, weekends off.

    Structural brain maturation and sleep are complex processes that exhibit significant changes over adolescence and are linked to many physical and mental health outcomes. We investigated whether sleep-gray matter relationships are developmentally-invariant (i.e., stable across age) or developmentally-specific (i.e., only present during discrete time windows) from late childhood through young adulthood.

    We constructed the Neuroimaging and Pediatric Sleep Databank from 8 research studies conducted at the University of Pittsburgh (2009- 2020). Participants completed a T1-weighted structural MRI scan (sMRI) and 5-7 days of wrist actigraphy to assess naturalistic sleep. The final analytic sample consisted of 225 participants without current psychiatric diagnoses (9-25 years). We extracted cortical thickness and subcortical volumes from sMRI. Sleep patterns (duration, timing, continuity, regularity) were estimated from wrist actigraphy. Using regularized regression, we examined cross-sectional associations between sMRI measures and sleep patterns, as well as the effects of age, sex, and their interaction with sMRI measures on sleep.

As a proof-of-concept, copper determination in fuel bioethanol was performed, as such ions catalyse oxidation processes during storage and transport. Square-wave anodic-stripping voltammetry (SWASV) of copper was achieved after sample dilution in 0.1 mol L-1 HCl as supporting electrolyte (resulting in 3070% v/v ethanolwater). The linear responses were in the range between 10 and 300 μg L-1 (R = 0.999), inter-day precision was lower than 8% (n = 10, for 20 μg L-1) and limits of detection (LOD) and quantification (LOQ) using 180 s as deposition time were 0.097 μg L-1 and 0.323 μg L-1, respectively. Recovery values between 95 and 103% for the analysis of bioethanol spiked with known amounts of copper were obtained. These results show great promise of the application of 3D-printed sensors for the quality control of biofuels. The Viennese university teacher immediately lost his employment and teaching authority and was forced to emigrate.While Gottlieb's exceptional scientific position in oral pathology is well documented, the complex implications of his deprivation of rights and forced emigration in the Third Reich have so far received little attention. Against this background, the present contribution poses the question of the concrete effects of this drastic event on Gottlieb's life and work.In order to clarify this question, Gottlieb's career status, his scientific success up to 1938, the concrete background of his forced emigration, as well as the further course of his life and career in the USA (his immigration country) are scrutinized. In addition, the paper analyzes the extent to which Gottlieb was able to build on his professional career after 1945 and posthumously. The work is based on a thorough analysis of Gottlieb's academic career using archival sources and a re-analysis of the relevant research literature.The study concludes that Gottlieb suffered a severe setback after his emigration. Several reasons played a role. PATIENTS AND METHODS The study incorporated 40 patients with chronic HCV-related HCC and 40 healthy controls. Patients were radiologically assessed for hepatic focal lesions and portal vein thrombosis. Response to HCC treatment and overall survival (OS) outcomes were determined. The expression of Tim-3, PD-1, and CCR5 among CD19+, CD4+, and CD8+ lymphocytes was assessed by flow cytometry. RESULTS Higher frequencies of CD4+ and CD8+ cells expressing each of Tim-3 and PD-1 and PD-1+CD19+ cells were observed in the HCV-related HCC patients in comparison with controls. The highest expression of Tim-3 and PD-1 was by the CD8+ cells. Strong relations were detected among PD-1+CD19+, PD-1+CD4+ and PD-1+CD8+ cells. Elevated levels of PD-1+ lymphocytes were significantly associated with poor treatment response and shorter OS. CONCLUSION Modulation of the expression of immune checkpoints as Tim-3 and PD-1, and of CCR5 on T cells is somehow related to HCC. CD8+ T cells expressing PD-1 were the most relevant to HCC prognosis (OS and treatment response) and could represent a promising target for immune therapy against HCC. Future studies need to focus on exploring PD-1+ B cells and Tim-3+CD4+ cells, which seem to play a significant role in the pathogenesis of HCC.Tumor-related leukocytosis (TRL) is correlated with poor survival in various types of cancers, but the microenvironment of TRL-associated human tumors has not been fully elucidated. LShigh tumors also expressed higher levels of CXCR2 chemokines, CSF2, and CSF3. In the pan-cancer cohort (n = 9984), LShigh tumors also exhibited poor survival, signatures of a suppressive immune microenvironment, and higher expression of CXCR2 chemokines. Our data provide evidence for a suppressive immune microenvironment in patients with TRL and suggest promising targets, such as the CXCR2 axis, for its therapeutic intervention.

Graphical abstract.We show that fused deposition modelling (FDM) 3D-printed electrodes can be used for quality control of fuel bioethanol. 3D-printing using carbon black/polylactic acid (CB-PLA) filaments resulted in conductive and biodegradable electrodes for biofuel analysis. As a proof-of-concept, copper determination in fuel bioethanol was performed, as such ions catalyse oxidation processes during storage and transport. Square-wave anodic-stripping voltammetry (SWASV) of copper was achieved after sample dilution in 0.1 mol L-1 HCl as supporting electrolyte (resulting in 3070% v/v ethanolwater). The linear responses were in the range between 10 and 300 μg L-1 (R = 0.999), inter-day precision was lower than 8% (n = 10, for 20 μg L-1) and limits of detection (LOD) and quantification (LOQ) using 180 s as deposition time were 0.097 μg L-1 and 0.323 μg L-1, respectively. Recovery values between 95 and 103% for the analysis of bioethanol spiked with known amounts of copper were obtained. The Viennese university teacher immediately lost his employment and teaching authority and was forced to emigrate.While Gottlieb's exceptional scientific position in oral pathology is well documented, the complex implications of his deprivation of rights and forced emigration in the Third Reich have so far received little attention. Against this background, the present contribution poses the question of the concrete effects of this drastic event on Gottlieb's life and work.In order to clarify this question, Gottlieb's career status, his scientific success up to 1938, the concrete background of his forced emigration, as well as the further course of his life and career in the USA (his immigration country) are scrutinized. In addition, the paper analyzes the extent to which Gottlieb was able to build on his professional career after 1945 and posthumously. The work is based on a thorough analysis of Gottlieb's academic career using archival sources and a re-analysis of the relevant research literature.The study concludes that Gottlieb suffered a severe setback after his emigration. Several reasons played a role. In particular, cultural and age-related adjustment problems, difficult local conditions, and scarce financial resources hampered the seamless continuation of Gottlieb's career in the USA. Patients were radiologically assessed for hepatic focal lesions and portal vein thrombosis. Response to HCC treatment and overall survival (OS) outcomes were determined. The expression of Tim-3, PD-1, and CCR5 among CD19+, CD4+, and CD8+ lymphocytes was assessed by flow cytometry. RESULTS Higher frequencies of CD4+ and CD8+ cells expressing each of Tim-3 and PD-1 and PD-1+CD19+ cells were observed in the HCV-related HCC patients in comparison with controls. The highest expression of Tim-3 and PD-1 was by the CD8+ cells. Strong relations were detected among PD-1+CD19+, PD-1+CD4+ and PD-1+CD8+ cells. Elevated levels of PD-1+ lymphocytes were significantly associated with poor treatment response and shorter OS. CONCLUSION Modulation of the expression of immune checkpoints as Tim-3 and PD-1, and of CCR5 on T cells is somehow related to HCC. CD8+ T cells expressing PD-1 were the most relevant to HCC prognosis (OS and treatment response) and could represent a promising target for immune therapy against HCC.   • PURPOSE We expect physicians to be lifelong learners. Learning from clinical practice is an important potential source for that learning. To support physicians in this process, a better understanding of how they learn in clinical practice is necessary. This study investigates how physicians recognize and use informal feedback from interactions with patients in outpatient settings as learning cues to adjust their communication behaviours in daily practice. METHODS To understand physicians' use of informal feedback, we combined non-participant observations with semi-structured interviews. We enrolled 10 respiratory physicians and observed 100 physician-patient interactions at two teaching hospitals in the Netherlands. Data collection and analysis were performed iteratively according to the principles of constructivist grounded theory. RESULTS Following stages of open, axial and selective coding, we were able to conceptualize how physicians use cues to reflect on and adjust their communication. METHODS The interaction of previously identified factors that could induce tolerance, granulocyte-macrophage colony stimulating factor (GM-CSF) and thymic stromal lymphopoietin (TSLP), with the periodontitis network was confirmed in silico. The effect of the cytokines on DC migration was explored in vitro using time-lapse imaging. Finally, regulatory T-cell enrichment in the dermis and periodontal tissue in response to alginate hydrogels delivering TSLP and GM-CSF was examined in vivo in **** using immunohistochemistry and live-animal imaging. RESULTS The GM-CSF and TSLP interactome connects to the periodontitis network. GM-CSF enhances DC migration in vitro. An intradermal injection of an alginate hydrogel releasing GM-CSF enhanced DC numbers and the addition of TSLP enriched FOXP3+ regulatory T-cells locally. Injection of a hydrogel with GM-CSF and TSLP into the periodontal tissue in **** increased DC and FOXP3+ cell numbers in the tissue, FOXP3+ cells in the lymph node, and IL-10 in the tissue. By comparing their stomatal dynamics with that of the same genotypes grown in a glasshouse, we were able to test whether differences between these growing conditions interacted with genotypic differences in affecting stomatal dynamics and responses to soil water deficit. • We found that, despite higher stomatal density and smaller size, in the field stomatal dynamics were **** slower than in the glasshouse. Overall, differences among genotypes and their response to soil water deficit were **** less pronounced in the field compared to the glasshouse. • These results indicate that stomatal dynamics are regulated by both genotype-specific and environmental factors. OBJECTIVES To evaluate clinical features and primary efficacy (composite of stroke, systemic embolic events, myocardial infarction and cardiovascular mortality during study period) and safety endpoints (composite of major and clinically relevant nonmajor bleeding during on-treatment period) in patients awaiting ECV of AF with a transesophageal echocardiography (TEE)-guided vs a non-TEE-guided strategy. METHODS In this prospective, randomized, open-label, blinded endpoint study, 2199 patients were randomized to edoxaban 60 mg once-daily (30 mg for creatinine clearance 15-50 mL/min, weight ≤60 kg and/or concomitant use of P-glycoprotein inhibitor) or enoxaparin-warfarin. Primary efficacy endpoint and safety endpoint were reported. Associates of TEE use, efficacy endpoint and safety endpoint were explored using multivariable logistic regression. RESULTS In total, 589 patients from the edoxaban stratum and 594 from the enoxaparin-warfarin stratum were allocated to the TEE-guided strategy. Primary efficacy was similar regardless of TEE approach (P = .575). There were no significant differences in bleeding rates, regardless of TEE approach (P = .677). Independent predictors of TEE use were as follows history of ischaemic stroke/ transient ischaemic attack, hypertension and valvular heart disease. Mean CHA2 DS2 VASc and HAS-BLED score were independent predictors of the efficacy endpoint whilst mean age was an independent predictor of the safety endpoint. CONCLUSIONS Thromboembolic and bleeding events were not different between patients undergoing TEE-guided strategy and in those undergoing an optimized conventional anticoagulation approach for ECV of AF. © 2020 Stichting European Society for Clinical Investigation Journal Foundation.

To better understand the mechanistic effect cromolyn has in slowing down or halting the neuroinflammatory response associated with AD progression, we tested the effect of cromolyn to dampen the inflammatory response in the human HMC3 microglia cell line. The direct effect of cromolyn on HMC3 microglia is on cytokines and chemokines production following their activation by the inflammatory cytokine TNF-α. Cromolyn and a new fluorinated analog dramatically reduced the secretion of a wide spectrum of inflammatory mediators, which included cytokines such as IL-1β, IL-6, IL-8 and IFN-γ, and chemokines such as CXCL10, CCL2, CCL3 and CCL4. These results bolster our understanding of how our cromolyn platform modulates toxic microglia behavior as a dynamic future treatment option for neurodegenerative disorders.Extracellular vesicles (EVs) derived from tumor cells have the potential to provide a ****-needed source of non-invasive molecular biomarkers for liquid biopsies. However, current methods for EV isolation have limited specificity towards tumor-derived EVs that limit their clinical use. Here, we present an approach called immunomagnetic sequential ultrafiltration (iSUF) that consists of sequential stages of purification and enrichment of EVs in approximately 2 h. In iSUF, EVs present in different volumes of biofluids (0.5-100 mL) can be significantly enriched (up to 1000 times), with up to 99% removal of contaminating proteins (e.g., albumin). The EV recovery rate by iSUF for cell culture media (CCM), serum, and urine corresponded to 98.0% ± 3.6%, 96.0% ± 2.0% and 94.0% ± 1.9%, respectively (p > 0.05). The final step of iSUF enables the separation of tumor-specific EVs by incorporating immunomagnetic beads to target EV subpopulations. The direct effect of cromolyn on HMC3 microglia is on cytokines and chemokines production following their activation by the inflammatory cytokine TNF-α. Cromolyn and a new fluorinated analog dramatically reduced the secretion of a wide spectrum of inflammatory mediators, which included cytokines such as IL-1β, IL-6, IL-8 and IFN-γ, and chemokines such as CXCL10, CCL2, CCL3 and CCL4. Here, we present an approach called immunomagnetic sequential ultrafiltration (iSUF) that consists of sequential stages of purification and enrichment of EVs in approximately 2 h. In iSUF, EVs present in different volumes of biofluids (0.5-100 mL) can be significantly enriched (up to 1000 times), with up to 99% removal of contaminating proteins (e.g., albumin). The EV recovery rate by iSUF for cell culture media (CCM), serum, and urine corresponded to 98.0% ± 3.6%, 96.0% ± 2.0% and 94.0% ± 1.9%, respectively (p > 0.05).     The objective of this study is to determine whether modification of opioid prescribing presets in the EHR could change prescribing patterns for an entire hospital system.

    We performed a quasi-experimental retrospective pre-post analysis of a 24-month period before and after modifications to our EHR's opioid prescription presets to match Centers for Disease Control and Prevention guidelines. We included all opioid prescriptions prescribed at our institution for nonchronic pain. Our modifications to the EHR include (1) making duration of treatment for an opioid prescription mandatory, (2) adding a quick button for 3 days' duration while removing others, and (3) setting the default quantity of all oral opioid formulations to 10 tablets. We examined the quantity in tablets, duration in days, and proportion of prescriptions greateand quantity of opioid prescriptions could reduce the risk of dependence and overdose.

We developed an internet-based decision aid (DA) to help patients with CF make better informed decisions regarding treatments and advance care planning. We built the DA around two major treatment decisions whether to have a lung transplant and whether to agree to invasive mechanical ventilation (intubation).

    This study aims to conduct usability testing of the InformedChoices CF DA among key stakeholder groups.

    We performed a patient needs assessment using think-aloud usability testing with patients with CF, their surrogates, and CF clinicians. Think-aloud participants provided feedback while navigating the DA, and after viewing, they answered surveys. Transcripts from the think-aloud sessions and survey results were categorized into common, generalizable themes and optimizations for improving content, comprehension, and navigation. We assessed the ease of use of the DA (System Usability Scale) and also assessed ing the DA's future success. In this study, we examined the structure, diversity and co-occurrence networks of fungal community in rhizosphere and non-rhizosphere soils of planted and natural Picea asperata forests using high-throughput sequencing technique and bioinformatic methods. The results showed that Inocybaceae and Sebacinaceae were dominant family in soils of planted and natural forests, respectively. At the genus level, Inocybe was dominant one in soils of planted and natural forests. There were significant differences in β-diversity of fungal communities between rhizosphere and non-rhizosphere soils in both planted and natural forests. There were no significant correlations between environmental variables and the relative abundance and α-diversity of fungal communities. Herb layer coverage, soil water content, total organic carbon concentration, and plant species richness played important roles in explaining the variations of β-diversity of fungal communities. Results of the network analysis showed that the negative correlations were dominant among soil fungal communities in natural forest, suggesting that the competition of different groups in natural forest. The results showed that 1) The reduction of nitrogen fertilization decreased nematode number by 15.3%-68.5%. 2) Protorhabditis was the dominant genus (19.6%-50.4%) across all treatments. The reduction of nitrogen fertilizer application increased the abundance of fungivores, herbivores, and predators-omnivores, while that of bacterivores decreased first and then increased. Combined application of organic fertilizer decreased the abundance of bacterivores and fungivores, while increased that of herbivores and predators-omnivores. 3) N240 and F225 increased the Shannon diversity (H) of nematode community by 48.1% and 58.5%, respectively. We identified and prioritized GI network centers in Fuzhou downtown area using the MSPA and the landscape connectivity evaluation. The least cost path method and gravity model were used to construct the potential corridors at multiple levels. The density analysis and blind area analysis were used to extract and prioritize the GI nodes and to obtain the optimized GI network. The results showed that the first-level GI network centers were mainly distributed in the north and south of Fuzhou downtown, while those in the central region were small and scattered. The comprehensive resistance of landscape was low in the periphery but high in the middle, with poor integral connectivity. The GI corridor system with existing corridors and potential corridors was employed to enhance the connectivity among network centers. Furthermore, the GI nodes were extracted to provide a "transfer station" for material circulation and energy flow, which could partly solve the problems including excessive substrate resistance and the long connection corridor in some areas. The spatial prioritization of GI elements could make the construction of GI network more scientific and also provide reference for the future planning period and construction timing of GI network in Fuzhou.The extreme precipitation characteristics of Ansai District in 40 years was analyzed from the perspective of precipitation intensity and precipitation frequency with 11 extreme precipitation indices, based on daily precipitation data of Ansai Meteorological Station from 1980 to 2019. The trend of extreme precipitation in the future was predicted. The results showed that the extreme precipitation indicators generally showed a downward trend during 1980-2019. The downtrend of heavy precipitation days and simple daily precipitation intensity reached significant level, with their climate tendency slopes being -0.65 d·(10 a)-1, -0.32 mm·d-1·(10 a)-1, respectively. Except for consecutive wet days and extremely wet day precipitation, the other extreme precipitation indices had mutation points. The spatial prioritization of GI elements could make the construction of GI network more scientific and also provide reference for the future planning period and construction timing of GI network in Fuzhou.The extreme precipitation characteristics of Ansai District in 40 years was analyzed from the perspective of precipitation intensity and precipitation frequency with 11 extreme precipitation indices, based on daily precipitation data of Ansai Meteorological Station from 1980 to 2019. The trend of extreme precipitation in the future was predicted. The results showed that the extreme precipitation indicators generally showed a downward trend during 1980-2019. The downtrend of heavy precipitation days and simple daily precipitation intensity reached significant level, with their climate tendency slopes being -0.65 d·(10 a)-1, -0.32 mm·d-1·(10 a)-1, respectively. Except for consecutive wet days and extremely wet day precipitation, the other extreme precipitation indices had mutation points. After the mutation, most of them had a downward trend, with significant decreases of annual precipitation, moderate precipitation days, heavy precipitation days, very heavy precipitation days, very wet day precipitation, simple daily precipitation intensity.

  • Historical theories of metastasis have been informed by the seed and soil hypothesis, the Halsteadian paradigm proposing an orderly spread from local to distant sites, and the presumption that cancer is an inherently systemic process even in the earliest cases. The more contemporary spectrum theory now suggests that the propensity for distant spread exists along a continuum of metastatic virulence. Tumors with limited metastatic potential represent one subset along this spectrum that could potentially be cured with local ablative therapy. Integrating clinical and molecular features to biologically inform the classification of not only oligometastatic or oligoprogressive disease but also the entire metastatic spectrum holds great promise to improve prognostication and inform clinical decision making. To this end, the inclusion of molecular correlative studies and biospecimen collection on prospective protocols is imperative.Pain signals the presence of potential harm, captures attention, and can inhibit performance on concurrent tasks. What is less well known, however, is whether such attentional capture also occurs in a wider social context, such as when observing people in pain. In order to explore this possibility, we adopted a novel social-cue detection methodology the bodies-in-the-crowd task. Two experiments are reported that consider whether nonverbal cues of pain, happiness and anger as expressed through body postures would capture and hold attention. Both experiments recruited 40 (20 male, 20 female) pain-free individuals. To fill this void, we studied placebo effects in healthy and chronic pain participants self-identified as either African-American/Black (AA/Black) or White. We enrolled 372 study participants, 186 with a diagnosis of temporomandibular disorder (TMD) and 186 race, sex, and age matched healthy participants to participate in a placebo experiment. Using a well-established paradigm of classical conditioning with verbal suggestions, each individual pain sensitivity was measured to calibrate the temperatures for high- and low-pain stimuli in the conditioning protocol. These two temperatures were then paired with a red and green screen, respectively, and participants were told that the analgesic intervention would activate during the green screens to reduce pain. Participants then rated the painfulness of each stimulus on a Visual Analog Scale ranging from 0-100. Racial influences were tested on conditioning strength, reinforced expectations and placebo hypoalgesia. We found that White participants reported greater conditioning effects, reinforced relief expectations, and placebo effects when compared to their AA/Black counterparts. Racial effects on placebo were observed in TMD although negligible, short-lasting, and mediated by conditioning strength. Secondary analyses on the effect of experimenter-participant race and sex concordance indicated that same experimenter-participant race induced greater placebo hypoalgesia in TMDs while different sex induced greater placebo hypoalgesia in healthy participants. This is the largest study to analyze racial effects on placebo hypoalgesia and has implications for both clinical research and treatment outcomes.We investigated the contribution of nucleus locus ceruleus (LC) to the development of pain-associated affective behavior. **** of both sexes were subjected to sciatic nerve cuffing, a model of peripheral nerve injury, and monitored for 45 days. While the thermal and mechanical thresholds were equally decreased in both males and females, only the male **** developed anxiodepressive-like behavior, which was complemented by suppressed hippocampal neurogenesis. Furthermore, the LC activity was lower in males when compared to females subjected to sciatic cuffing. Next, we used a chemogenetic approach to modulate the activity of LC projections to the dentate gyrus of the hippocampus in females without cuffs and in males with sciatic cuffs. Sustained inhibition of the LC projections to the dentate gyrus for 15 days induced anxiodepressive-like behavior and reduced the hippocampal neurogenesis in females. Activation of the LC projections to the dentate gyrus for 15 days prevented the development of anxiodepressive-like behavior and increased the hippocampal neurogenesis in males with cuffs. To inform how best to deliver care, we undertook two narrative reviews examining existing evidence on a) models of care for fibromyalgia and b) patients' experiences, preferences and unmet needs regarding their healthcare. Seven databases were systematically searched. Quantitative data was narratively synthesised and qualitative data thematically analysed. No evidence-based model of care covering the patient journey through the entire healthcare system was identified. Limited evidence suggests no clear benefit for ongoing care in secondary care settings. Patients with fibromyalgia report difficult interactions with the healthcare system that might equally be expressed by those with other longterm conditions, such as inconsistent and poorly coordinated care. However, they also face unique problems; fibromyalgia was often not viewed as a real condition, resulting in difficult encounters with healthcare staff, in particular not feeling believed or listened to.
