The representative target therapeutic agent, erlotinib (EGFR tyrosine kinase inhibitor; EGFR TKI), is effective in inactivating EGFR in lung cancer patients. However, approximately 50-60% of patients are resistant to EGFR TKI. These populations are associated with the EGFR mutation. To overcome resistance to EGFR TKI, we discovered a JAK1 inhibitor, CJ14939. We investigated the efficacy of CJ14939 in human NSCLC cell lines in vitro and in vivo. Our results showed that CJ14939 induced the inhibition of cell growth. Moreover, we demonstrated that combination treatment with erlotinib and CJ14939 induced cell death in vitro and inhibited tumor growth in vivo. Moreover, co-treatment with bortezomib, which is a newly identified anticancer drug for osteosarcoma, and kuanoniamine C suppressed GRP78 protein expression, which is essential for the stimulation of bortezomib-induced cell death. MAIN BODY This review summarizes current knowledge of NY-ESO-1 as efficient biomarker and target of immunotherapy. It also addresses limitations and challenges preventing a robust immune response to NY-ESO-1 expressing cancers, and describes pre-clinical and clinical observations relevant to NY-ESO-1 immunity, holding potential therapeutic relevance for cancer treatment. CONCLUSION NY-ESO-1 induces strong immune responses in cancer patients but has limited objective clinical responses to NY-ESO-1 expressing tumors due to effect of competitive negative signaling from immune-checkpoints and immune-suppressive tumor microenvironment. Medical records were retrospectively reviewed, and patients were assessed using postal questionnaire after a mean follow-up of 8.3 years [3-14]. Clinical assessment was performed using Constant score, Rowe score, and American Shoulder and Elbow Surgeons score. RESULTS The overall recurrence rate was 22%. The Kaplan-Meier failure-free survival estimates. were 80% at 5 years and 70% at 10 years. Nearly half (54.5%) of recurrences occurred at 2 years postoperative. Compared with normal shoulder, there were statistical differences in all 3 scores. Failure rate was significantly affected by age at the time of surgery with 86% of recurrence cases observed in patients aged 30 years or younger. Nevertheless, Younger age at the time of surgery (P = 0.007) as well age at the time of initial instability (P = 0.03) was found to correlate negatively with early recurrence within 2 years of surgery. Among those with recurrent instability, recurrence rate was found to be higher if there had been more than 5 instability episodes preoperatively (P = 0.01). Return to the preinjury sport and occupational level was possible in 41 and 78%, respectively. CONCLUSION Failure-free survival rates dropped dramatically over time. Alternative reconstruction techniques should be considered in those aged ≤30 years due to the high recurrence rate.

The synagogue in Córdoba has become an irreplaceable Jewish tourist destination in Spain due to its origins, construction, conservation, and recognition by international organizations. This article analyzes the loyalty of tourism in synagogues using variables such as satisfaction, loyalty and the expected and perceived value of the destination and the historical monument using a structural research equations methodology. No previous studies on tourism at synagogues using structural equations have been made. The data was obtained by means of a survey given to 350 tourists visiting this cultural heritage site. The anaerobic acetogenic consortium in mineral medium containing CO 2 / HCO 3 - as the inorganic carbon source used suspended humin as the energy source under mesophilic dark conditions. Retardation of acetogenesis under the CO 2 -deficient conditions demonstrated that humin did not function as the organic carbon source but as electron donor in the CO 2 -reducing acetogenesis. The consortium with humin also achieved anaerobic dechlorination with limited methanogenic activity. Total electron-donating capacity of humin was estimated at about 87 µeeq/g-humin. The metagenomic sequencing of 16S rRNA genes showed the predominance of Firmicutes (71.8 ± 2.5%) in the consortium, and Lachnospiraceae and Ruminococcaceae were considered as the CO 2 -reducing acetogens in the consortium. Thus, microbial fixation of CO 2 using humin introduces new insight to the holistic approach for sustainable treatment of contaminants in environment.

  • BACKGROUND This study aimed to investigate the biomechanical effect on the Takeuchi classification of lateral hinge fracture (LHF) after an opening wedge high tibial osteotomy (HTO). METHODS We performed an FE simulation for type I, type II, and type III in accordance with the Takeuchi classification. The stresses on the bone and plate, wedge micromotion, and forces on ligaments were evaluated to investigate stress-shielding effect, plate stability, and biomechanical change, respectively, in three different types of LHF HTO and with the HTO without LHF model (non-LHF) models. RESULTS The greatest stress-shielding effect and wedge micromotion were observed in type II LHF (distal portion fracture). The type II and type III (lateral plateau fracture) models exhibited a reduction in ACL force and an increase in PCL force compared with the HTO without LHF model. METHODS Incisor Plus Blade (IPB; Smith & Nephew, Andover, MA) and Double Serrated Plus Blade (DSPB; BJKMC, Shanghai, China) were selected for resection performance and quality test. For resection performance testing, the resection torque, which is the minimum torque required to cut off silicone blocks with the same cross-sectional area, was measured to evaluate the resection performance of shaver blades when the other factors remain the same. For quality testing, tensile and torsion tests of the shavers' joint pa the torsional strength of the joint part, and the corrosion resistance of IPB and DSPB may show comparable properties, whereas the tensile strength of the shavers' joint part showed some level of difference.H3K27ac is well recognized as a marker for active enhancers and a great indicator of enhancer activity. However, its functional impact on transcription has not been characterized. By substituting lysine 27 in histone variant H3.3 with arginine in mouse embryonic stem cells, we diminish the vast majority of H3K27ac at enhancers. However, the transcriptome is largely undisturbed in these mutant cells, likely because the other enhancer features remain largely unchanged, including chromatin accessibility, H3K4me1, and histone acetylation at other lysine residues. Autophagy flow was detected by RFP-GFP-LC3 double fluorescence molecule. The expression of TFEB, DRAM, MAPLC3, and MITF were analyzed by Western blot, and the expression of genes NITF, Bcl2, and TYR in rat cartilage tissues were detected by RT-PCR. RESULTS The number of autophagosomes was increasing in the auto group compared with the inhibitor-auto group and normal group. There was a significant difference of LC3 levels in the auto group and inhibitor-auto group compared with the normal control. The expression of TFEB, DRAM, MAPLC3, and MITF proteins by Western blot analysis were significantly increased in the auto group and decreased in the inhibitor-auto group. The expression of NITF, Bcl2, and TYR by RT-PCR determination were higher in the auto group and inhibitor-auto group than the normal group. CONCLUSIONS Autophagy can inhibit apoptosis, promote chondrocyte growth and bone regeneration, and restore knee joint injury of rats. We investigated the risk of long-term sick leave, disability pension, and premature death in individuals with AHP compared to the general population. METHODS In a nationwide cohort study from 1992 to 2017, records of 333 persons (total person-years = 6728) with a confirmed AHP diagnosis were linked to several national compulsory registries (reference population = 5,819,937). We conducted survival analyses to assess additional risk. RESULTS Persons with AHP had higher risks of accessing long-term sick leave (adjusted hazard ratio (aHR) 1.5, 95% confidence interval (CI) 1.3, 1.7) and disability pension (aHR 1.9, CI 1.5, 2.4). The risk was highest in persons who had been hospitalised for acute attacks, while no additional risk was observed in asymptomatic AHP gene mutation carriers. The median age when accessing disability pension was 45 years, 21 years younger than the general population. AHP was associated with increased risk of mortality due to hepatocellular carcinoma (adjusted mortality rate ratio (aMRR) 84.4, CI 37.8, 188.2), but no overall increased risk of premature death was observed. CONCLUSIONS Persons with symptomatic AHP were at increased risk of accessing long-term sick leave and disability pension but not of premature death.

  • In this study, a novel Al°-CNTs-Cu2O composite, capable of activating O2 to generate H2O2 and further to reactive oxygen species (ROSs) at a wide pH range, was synthetized, characterized and applied for the degradation of sulfamerazine. In the activation of O2 by Al°-CNTs-Cu2O composite, H2O2 was generated from the reaction of O2 with Al°-CNTs, which could be catalytically decomposed into O2- and OH by Cu2O, the formed Cu(II) could be rapidly reduced to Cu2O by Al°-CNTs in composite, which made Al°-CNTs-Cu2O composite reusable and decreased the leaching of copper ions into solution. The removal efficiency of SMR and TOC was 73.91 % and 56.80 %, respectively at initial pH = 5.8, T = 20 °C, O2 flow rate = 100 mL/min, Al°-CNTs-Cu2O dosage = 2 g/L, SMR = 50 mg/L, and reaction time = 60 min. The removal efficiency of SMR kept almost unchanged and the concentration of copper ions in solution was below 0.5 mg/L. The Al°-CNTs-Cu2O/O2 process could be used as a novel catalyst for the degradation of refractory organic contaminants in water and wastewater by Fenton-like process at a wide pH range through the in situ generation of H2O2. Coal fly ash (CFA) is a valuable industrial solid waste, but conventional methods used for its disposal can lead to serious and long-lasting environmental issues. The study of technologies for CFA recycling has been of major concern, while the harm caused by CFA is only partially understood, limiting its reuse. In this review, the basic physico-chemical properties of CFA are introduced, followed by a systematic summary and discussion of the leachability of CFA via different leaching methods and the chemical speciation of some typical metal elements in CFA, which is related to its harmful effects. The specific harm that CFA causes to humans, wild animals, and plants and the study status of magnetic property of CFA are presented. Because of the pervasive concerns of many people, the utilisation of CFA in the USA and Europe and an economic and environmental analysis of its disposal is provided and discussed. Finally, some possible directions for future research involving CFA are proposed. China has applied the ultra-low emission technology in coal-fired power plants to control traditional air pollutants and to reduce Hg emissions synergically. In this study, we applied field experiment, model calculation, and literature review to evaluate the Hg control effect of ultra-low emission technology and the potential cross-media effect comprehensively. The dominant ultra-low emission technology significantly improves the atmospheric Hg removal efficiency from 75% to 87%. Such improvement mainly comes from the effect of dust removal devices. Based on the calculated distribution characteristic of Hg content of wastes, we find out that the improvement of Hg control effect of air pollution control devices significantly increase the Hg content of fly ash, which rises from 0.16 mg/kg to 0.33 mg/kg. However, the Hg content of gypsum decreases from 0.75 mg/kg to 0.51 mg/kg. Whether or not to carry out ultra-low emission retrofits, Hg contents of wastes from coal-fired power plants are overall lower than the limit of 25 mg/kg which is intended to be set as the limit for Hg-containing wastes. However, the embodied more than two hundreds of tons Hg in these wastes still require policies to guide the disposal of these wastes. V.Nanoscale zerovalent iron (nZVI) and sulfidated nanoscale zerovalent iron (S-nZVI) have been increasingly studied for heavy metal removal in the subsurface. However, a comprehensive comparison of the effectiveness of the technologies and the stability of derived metal-adsorbed composites is lacking. In this study, we evaluated the colloidal stability and transport of nZVI, S-nZVI and S-nZVI modified with nanosized silica (FeSSi). Furthermore, we monitored the metal immobilization performance of the three nanoparticles (NPs) under anoxic conditions in synthetic groundwater for 30 days.     In this study, a novel Al°-CNTs-Cu2O composite, capable of activating O2 to generate H2O2 and further to reactive oxygen species (ROSs) at a wide pH range, was synthetized, characterized and applied for the degradation of sulfamerazine. In the activation of O2 by Al°-CNTs-Cu2O composite, H2O2 was generated from the reaction of O2 with Al°-CNTs, which could be catalytically decomposed into O2- and OH by Cu2O, the formed Cu(II) could be rapidly reduced to Cu2O by Al°-CNTs in composite, which made Al°-CNTs-Cu2O composite reusable and decreased the leaching of copper ions into solution. The removal efficiency of SMR and TOC was 73.91 % and 56.80 %, respectively at initial pH = 5.8, T = 20 °C, O2 flow rate = 100 mL/min, Al°-CNTs-Cu2O dosage = 2 g/L, SMR = 50 mg/L, and reaction time = 60 min. The removal efficiency of SMR kept almost unchanged and the concentration of copper ions in solution was below 0.5 mg/L. The Al°-CNTs-Cu2O/O2 process could be used as a novel catalyst for the degradation of refractory organic contaminants in water and wastewater by Fenton-like process at a wide pH range through the in situ generation of H2O2. Coal fly ash (CFA) is a valuable industrial solid waste, but conventional methods used for its disposal can lead to serious and long-lasting environmental issues. The study of technologies for CFA recycling has been of major concern, while the harm caused by CFA is only partially understood, limiting its reuse. In this review, the basic physico-chemical properties of CFA are introduced, followed by a systematic summary and discussion of the leachability of CFA via different leaching methods and the chemical speciation of some typical metal elements in CFA, which is related to its harmful effects. The specific harm that CFA causes to humans, wild animals, and plants and the study status of magnetic property of CFA are presented. Because of the pervasive concerns of many people, the utilisation of CFA in the USA and Europe and an economic and environmental analysis of its disposal is provided and discussed. Finally, some possible directions for future research involving CFA are proposed. China has applied the ultra-low emission technology in coal-fired power plants to control traditional air pollutants and to reduce Hg emissions synergically. In this study, we applied field experiment, model calculation, and literature review to evaluate the Hg control effect of ultra-low emission technology and the potential cross-media effect comprehensively. The dominant ultra-low emission technology significantly improves the atmospheric Hg removal efficiency from 75% to 87%. Such improvement mainly comes from the effect of dust removal devices. Based on the calculated distribution characteristic of Hg content of wastes, we find out that the improvement of Hg control effect of air pollution control devices significantly increase the Hg content of fly ash, which rises from 0.16 mg/kg to 0.33 mg/kg. However, the Hg content of gypsum decreases from 0.75 mg/kg to 0.51 mg/kg. Whether or not to carry out ultra-low emission retrofits, Hg contents of wastes from coal-fired power plants are overall lower than the limit of 25 mg/kg which is intended to be set as the limit for Hg-containing wastes. However, the embodied more than two hundreds of tons Hg in these wastes still require policies to guide the disposal of these wastes. V.Nanoscale zerovalent iron (nZVI) and sulfidated nanoscale zerovalent iron (S-nZVI) have been increasingly studied for heavy metal removal in the subsurface. However, a comprehensive comparison of the effectiveness of the technologies and the stability of derived metal-adsorbed composites is lacking.   • Objective  In the present study, we present the results with at least 10 years of follow-up of the cervical disc prosthesis implanted in a single level. Methods  Retrospective study of patients undergoing single-level total cervical disc replacement (TCDR). Clinical results included the neck disability index (NDI) and the visual analogue scale (VAS) in the preoperative period, one year postoperatively, and a minimum of 10 years of follow-up. The radiographic parameters included cervical mobility, segmental lordosis, C2-C7 angle, heterotopic ossification (HO), facet and joint degeneration (FJD) and adjacent segment disease (ASD). Results  We identified 22 patients, 16 women and 6 men with mean age of 39.7 years old (26-51 years old), of which fifteen completed a minimum follow-up of 10 years. There was a statistically significant improvement of NDI and VAS ( p   10 years). At the end of 10 years, HO was observed in 59% of the cases. The mobility of the implant was preserved in 80% of the patients. Radiological evidence of ASD was recorded in 6 patients (40%). There was no correlation between the clinical parameters evaluated and the presence of ASD or the different classes of HO. Conclusion  Clinical improvement in all evaluated parameters, which persists over time. Most implants maintained mobility, as has already been demonstrated in other studies with shorter follow-ups. In a significant percentage of cases, ASD was observed, questioning the concept of motion preserving technology. However, we did not have any surgical intervention for this reason, since there was no correlation with worse clinical results.Objective  The aim of the present study is to identify the incidence, predisposing factors and prognostic impact of blood loss in patients with neuromuscular scoliosis submitted to corrective surgery. https://www.selleckchem.com/products/Trichostatin-A.html Methods  Retrospective cohort study, including pediatric patients diagnosed with neuromuscular scoliosis undergoing instrumentation and posterior vertebral fusion in a university hospital. Patient characteristics were collected from the hospital information system. Results  A total of 39 patients were included in the study. The intraoperative blood losses were 962 ml, representing a loss of 35.63% of the blood volume. In 20 cases, there was a massive blood loss (> 30%) and only 7 patients did not need a transfusion. The group of patients with massive blood loss had a slightly higher age (13.75 versus 13.53 years old), a lower body mass index (BMI) percentile (25 versus 50), and for each decrease of 0.38 in the BMI, intraoperative blood losses increased 1% ( p  less then  0.05). The value of preoperative albumin had influence on the percentage of blood loss, and for each decrease of 0.4 of albumin, blood loss increased 1% ( p  less then  0.05). Conclusions  The factors that most contributed to the differences in blood loss were age, BMI, and preoperative albumin value. There was no significant association between Cobb angle, number of fusion levels and duration of surgery. Method  In the adaptation process, the original NCCPC-PV was translated, ****-translated, its versions were discussed by a committee of experts, and the resulting tool was tested in 20 health professionals and 20 caregivers of CCIC patients regarding its semantic clarity. Results  Data from the present study and its participants were analyzed and their results were described. Thus, "Lista de Verificação de Dor em Crianças Não Comunicantes - Versão Pós-operatória" (Br-NCCPC-PV) was obtained as the final version in Brazilian Portuguese. Conclusion  After the present study, the Br-NCCPC-PV was considered adequate for use in the Brazilian population.Objective  To elaborate a protocol for the harvest, transport, and preservation of human osteochondral tissue for use in tissue banks (TBs). Methods  Osteochondral fragments measuring 2 cm 3 of 5 corpse donors aged between 15 and 45 years old were analyzed. The samples were stored in cell preservation medium containing human albumin, Iscove's and vancomycin preserved at 4°C. The concentration of proteoglycans in the extracellular medium was quantified by the use of Safranin-O, while tissue structural analysis was assessed by histological study with hematoxylin-eosin stained slides. The images obtained were analyzed according to the histological scores of Mankin and the score proposed by the OsteoArthritis Research Society International. The samples were analyzed with 0, 15, 30 and 45 days of preservation. Results  The osteochondral fragments studied showed a progressive decrease in proteoglycan concentration with increased preservation time. After 30 days of preservation, structural changes were identified with discontinuity of the cartilage surface layer. According to the results obtained by the Mankin score, there was a statistically significant difference between 15 and 30 days of tissue preservation. Conclusion  The protocol described defined knee transport immersed in Lactated Ringer at a controlled temperature of 10° C until its arrival at the TB. After processing, the preservation solution was composed of Iscove's serum-free cell culture medium supplemented with 10% human albumin and 100 μg/ml vancomycin. The tissue was preserved at a temperature of 4°C until the moment of transplantation characterizing the fresh preservation.Objective  To compare antisepsis techniques using chlorhexidine-based soap associated with ethyl alcohol and alcohol-based chlorhexidine or chlorhexidine-based soap associated with alcohol-based chlorhexidine alone in surgical orthopedic procedures. Methods  This is a primary, randomized, analytical and single-center clinical trial consisting of 170 patients, who were divided into 2 groups. The combinations chlorhexidine-based soap + alcohol-based chlorhexidine (CSAC) and chlorhexidine-based soap + 70% ethyl alcohol + alcohol-based chlorhexidine (CSAAC) were tested in each group. The cultures were grown in mannitol and eosin methylene blue (EMB) after collection before skin preparation (time point 0), after skin preparation (time point 1) and at the end of the surgical procedure (time point 2). Results  There was no statistically significant difference regarding bacterial growth in mannitol and EMB between the groups at any time point. Moreover, there was no statistical difference between groups and time points regarding the type of bacterial growth in culture media.

  • Rurality was associated with significantly higher in-hospital mortality rates (1.44% vs 1.89%, OR 1.32, P less then .001). This association strengthened after adjusting for medical comorbidities and hospital characteristics. Further, disparities between urban and rural mortality were found to be growing, with the gap almost doubling between 2012 and 2016. Conclusions Rural and urban uninsured patients differed significantly, specifically in terms of race and median income. Among the uninsured, rurality was associated with higher in-hospital mortality, and the gap between urban and rural in-hospital mortality was widening. Our findings suggest the rural uninsured are a vulnerable population in need of informed, tailored policies to reduce these disparities.Purpose People living in rural areas experience greater health disparities than their nonrural counterparts, but little is known about the association between rural status and quality of life (QOL) in non-Hodgkin's lymphoma (NHL) survivors. We compared self-reported quality of life and impact of cancer in rural and nonrural NHL survivors. Methods This study is a secondary analysis of 566 NHL cancer survivors recruited from cancer registries at 2 large academic medical centers in 1 state. Standardized measures collected information on demographics and clinical characteristics, quality of life (QOL; SF-36), and the Impact of Cancer (IOCv2). Rural residence was determined by Rural-Urban Commuting Area (RUCA) codes designated as nonmetropolitan. Multiple linear regression analysis, adjusted for demographic and clinical covariates, was used to evaluate the relationship between rural residence and QOL and impact of cancer. Findings Among the 566 participants (83% response rate), rural residence was independently associated with lower SF-36 physical component summary scores and the physical function subscale (all P less then .05). Rural residence was also associated with higher IOCv2 positive impact scores and the subscales of altruism/empathy and meaning of cancer scores in the adjusted models (all P less then .05). Methods MRI quality recommendations for spatial integrity, image blurring, fat saturation, and null banding in RT were developed based on the literature. Guaranteed (maximum) and typical B0 field homogeneity specifications for various diameter spherical volumes (DSVs) were provided by GE, Philips, Siemens, and Canon. For each system, the DSV that conforms to each MRI quality recommendation and anatomical RT application was estimated based on the manufacturer specifications. The unopposed serine protease activity due to reduced LEKTI-1 inhibition in NS leads to premature degradation of corneodesmosomes, causing decreased stratum corneum cohesion and thinning.Blepharismins are photodynamic hypericin-like dianthrones produced as a variable pigment blend in Blepharisma ciliates and mostly studied in the Afro-Asiatic Blepharisma japonicum. The present work describes the bioactivity of pigments from the Brazilian Blepharisma sinuosum. Comparative analyses showed that the pigments from both species can trigger photo-induced modifications in phospholipids, but different redox properties and biological activities were assigned for each pigment blend. Stronger activities were detected for B. sinuosum pigments, with the lethal concentration LC50 10 × lower than B. japonicum pigments in light-irradiated tests against Bacillus cereus and less than half for treatments on the human HeLa tumor cells. HPLC showed B. sinuosum producing a simpler pigment blend, mostly with the blepharismin-C (~ 70%) and blepharismin-E (~ 30%) types. Each blepharismin engaged a specific dose-response profile on sensitive cells. The blepharismin-B and blepharismin-C were the most toxic pigments, showing LC50 ~ 2.5-3.0 µm and ~ 100 µm on B. cereus and HeLa cells, respectively, after illumination. Similarity clustering analysis compiling the bioactivity data revealed two groups of blepharismins the most active, B and C, and the less active, A, D and E. The B. sinuosum pigment blend includes one representative of each clade. Functional and medical implications are discussed.

Our findings suggest the rural uninsured are a vulnerable population in need of informed, tailored policies to reduce these disparities.Purpose People living in rural areas experience greater health disparities than their nonrural counterparts, but little is known about the association between rural status and quality of life (QOL) in non-Hodgkin's lymphoma (NHL) survivors. We compared self-reported quality of life and impact of cancer in rural and nonrural NHL survivors. Methods This study is a secondary analysis of 566 NHL cancer survivors recruited from cancer registries at 2 large academic medical centers in 1 state. Standardized measures collected information on demographics and clinical characteristics, quality of life (QOL; SF-36), and the Impact of Cancer (IOCv2). Rural residence was determined by Rural-Urban Commuting Area (RUCA) codes designated as nonmetropolitan. Multiple linear regression analysis, adjusted for demographic and clinical covariates, was used to evaluate the relationship between rural residence and QOL and impact of cancer. Findings Among the 566 participants (83% response rate), rural residence was independently associated with lower SF-36 physical component summary scores and the physical function subscale (all P less then .05). Rural residence was also associated with higher IOCv2 positive impact scores and the subscales of altruism/empathy and meaning of cancer scores in the adjusted models (all P less then .05). Stronger activities were detected for B. sinuosum pigments, with the lethal concentration LC50 10 × lower than B. japonicum pigments in light-irradiated tests against Bacillus cereus and less than half for treatments on the human HeLa tumor cells. HPLC showed B. sinuosum producing a simpler pigment blend, mostly with the blepharismin-C (~ 70%) and blepharismin-E (~ 30%) types. Each blepharismin engaged a specific dose-response profile on sensitive cells. The blepharismin-B and blepharismin-C were the most toxic pigments, showing LC50 ~ 2.5-3.0 µm and ~ 100 µm on B. cereus and HeLa cells, respectively, after illumination. Similarity clustering analysis compiling the bioactivity data revealed two groups of blepharismins the most active, B and C, and the less active, A, D and E. The B. sinuosum pigment blend includes one representative of each clade. Functional and medical implications are discussed.

The administration of PV significantly improved running endurance and glycogen accumulation in the liver and muscle of PV supplemented rats compared to sedentary and exercised control groups. In addition, PV supplementation elicited lower fatigue-related serum biomarkers, for instance, ammonia, inorganic phosphate, and lactate. PV administered rats exhibited higher lactate dehydrogenase activity and glutathione peroxidase activity, and lower creatine kinase activity and malondialdehyde levels. Furthermore, phenolic compounds in PV were identified using HPLC analysis. The phenolic acids analyzed in PV were protocatechuic acid, syringic acid, chlorogenic acid, and its derivates. These results indicate that the administration of PV with antioxidative property contributes to the improvement of fatigue recovery in exhausted rats. Non-coding RNAs operate mainly in mammals as epigenetic modifiers and enhancers of proteome diversity. The prominent evolutionary expansion of the gene number of lncRNAs in mammals, particularly in primates and humans, and their preferential neuronal expression does represent a driving force that enhanced the layering of synaptic control mechanisms. In the last few years, remarkable alterations of the expression of lncRNAs have been reported in psychiatric conditions such as schizophrenia, autism, and depression, suggesting unprecedented mechanistic insights into disruption of fine synaptic tuning underlying severe behavioral manifestations of psychosis. In this review, we integrate literature data from rodent pathological models and human evidence that proposes the biology of lncRNAs as a promising field of neuropsychiatric investigation.Elevated levels of high-density lipoprotein cholesterol (HDL-C) have been associated with a decreased risk of coronary heart disease (CHD). The sociodemographic data of 7543 participants, comprising 3472 men and 4071 women aged between 30 and 70 years, were retrieved from the Taiwan Biobank. The participants were grouped into three exercise categories-no exercise, aerobic exercise, and badminton exercise. The HDL-C levels were compared using an analysis of variance (ANOVA). Multivariate linear regression models were used to determine the associations between HDL and exercise.   • Scope The development of novel compounds that trigger non-apoptotic cell death may represent alternative therapeutic strategies for esophageal squamous cell carcinoma (ESCC) treatment. Cellular senescence suppresses tumorigenesis by halting the proliferation of tumor cells, implying the induction of senescence as a promising anticancer strategy, especially when combined with senolytic agents that specially kill senescent cells. This study is designed to screen novel anti-ESCC compounds from a natural product resource and identify its mechanism-of-action. Methods and results Identified are the significant anti-cancer effect and underlying mechanism of SFN, an isothiocyanate derived from cruciferous vegetables, through RNA sequencing, western blot, and immunofluorescent assays. It is found that SFN inhibits proliferation of ESCC cells through inducing senescence. Mechanistically, SFN induces reactive oxygen species (ROS) via disrupting the balance between glutathione and oxidized glutathione, leading to DNA damage. In addition, ROS deregulates autophagy and promotes lysosome abnormal biogenesis through regulating mTOR/TFE3 axis. Finally, the inhibited autophagic flux facilitates exosome production, resulting in exosome-mediated paracrine senescence. Conclusions This study suggests the important roles of autophagy and exosome-mediated paracrine senescence in cancer therapy and highlights SFN as a potent anti-ESCC drug candidate.We have read with great interest the article published by Ramadan et al(1). We investigated the effect of a colonic-delivery formulation of butyrate on the fecal microbiota of patients with inflammatory bowel diseases (IBDs). Methods In this double-blind, placebo-controlled, pilot study, 49 IBD patients (n = 19 Crohn's disease, CD and n = 30 ulcerative colitis, UC) were randomized to oral administration of microencapsulated-sodium-butyrate (BLM) or placebo for 2 months, in addition to conventional therapy. Eighteen healthy volunteers (HVs) were recruited to provide a healthy microbiota model of the local people. Fecal microbiota from stool samples was assessed by 16S sequencing. Clinical disease activity and quality of life (QoL) were evaluated before and after treatment. Key results At baseline, HVs showed a different microbiota composition compared with IBD patients. The proposals include using national inpatient death data to define a standardized denominator to calculate comparable donation rates among OPO's. Based on these objective metrics, OPO's not performing at a pre-specified threshold will be required to rapidly improve performance to avoid decertification. We sought to determine whether rapid OPO improvement was possible based on objective donation metrics, and whether leadership change was associated with rapid improvement. We evaluated UNOS and CDC data from 2011-2018, and measured donation rates using CDC data on inpatient deaths from causes consistent with donation, based on the location of deaths. During the two four-year cycles, we found that an OPO's ranking relative to other OPO's were fairly static, with more than 90% of the OPO's at risk of flagging at the end of each four-year cycle (2014, 2018) being in the bottom 75% of OPO's in the preceding three years. Leadership changes were only associated with an improvement in OPO rankings during the 2011-2014 cycle. These data demonstrate that rapid improvements in OPO performance are uncommon, and while leadership changes increase the odds of rapid improvement, they do not guarantee improvement.Background Raoiella indica Hirst (Acari Tenuipalpidae) is the most critical coconut and banana pest recently introduced in Brazil. Once mite pests are introduced, it is essential to understand their dynamics in important crops under open-field climatic conditions to implement strategies for their management and determine the periods when species populations may increase in the field. Modelling tools have been used to determine the potential distribution of species and implications for the management of invasive species. Thus, our aim in this study was to determine the seasonal variation in R. indica and the influence of the monthly climate using CLIMEX modelling. We adjusted the CLIMEX model for R. indica based on distribution data, additional biological characteristics, and fluctuations in the R. indica population in a commercial coconut plantation. Results The model for the current climate shows a good match between the ecoclimatic index and the global distribution of R. indica. The model results demonstrate that most states of Brazil and several regions worldwide include areas with highly suitable climatic conditions for R. indica. We observed variations in the density of R. indica in commercial coconut crops, with the highest incidence occurring during the first months of the year. Conclusion Our results showed different alterations in seasonal suitability for R. indica that may provide information for the implementation of methods for time management, such as strategies for sampling and control during periods with a high degree of suitability for R. indica. We sought to determine whether rapid OPO improvement was possible based on objective donation metrics, and whether leadership change was associated with rapid improvement. We evaluated UNOS and CDC data from 2011-2018, and measured donation rates using CDC data on inpatient deaths from causes consistent with donation, based on the location of deaths. During the two four-year cycles, we found that an OPO's ranking relative to other OPO's were fairly static, with more than 90% of the OPO's at risk of flagging at the end of each four-year cycle (2014, 2018) being in the bottom 75% of OPO's in the preceding three years. Leadership changes were only associated with an improvement in OPO rankings during the 2011-2014 cycle. Modelling tools have been used to determine the potential distribution of species and implications for the management of invasive species. Thus, our aim in this study was to determine the seasonal variation in R. indica and the influence of the monthly climate using CLIMEX modelling. We adjusted the CLIMEX model for R. indica based on distribution data, additional biological characteristics, and fluctuations in the R. indica population in a commercial coconut plantation. Results The model for the current climate shows a good match between the ecoclimatic index and the global distribution of R. indica. The model results demonstrate that most states of Brazil and several regions worldwide include areas with highly suitable climatic conditions for R. indica. We observed variations in the density of R. indica in commercial coconut crops, with the highest incidence occurring during the first months of the year. Conclusion Our results showed different alterations in seasonal suitability for R. indica that may provide information for the implementation of methods for time management, such as strategies for sampling and control during periods with a high degree of suitability for R. indica. Results show that after nearly one year of exposure, the microbial biomass was not affected by UV-B incidence, and this effect has not yet been fully understood for tropical ecosystems. Modeled using an exponential deceleration equation, the removal of UV-B radiation decelerated the plant litter decomposition rate for the control conditions by 21% compared to litter exposed to full sun. Interestingly, shaded litter exhibited similar mass loss compared to litter exposed to full sun. Furthermore, differences in the decay constant among radiation treatments due to the UV-B effect were independent of lignin loss. Overall, our study suggests that UV-B radiation contributed to plant litter decomposition through carbon losses but had no discernible effect on nitrogen, lignin, or cellulose loss specifically. V.Plastic mulching is suspected to be a significant source of microplastics in terrestrial environments owing to its intensive application and improper disposal. However, there has been a comparative lack of studies examining this hypothesis. In this study, the occurrence of macroplastics in agricultural soils was investigated by analysing 384 soil samples collected from 19 provinces across China. Additionally, the abundance of microplastics was investigated in potential hotspots that have carried out plastic mulching for over 30 years. Macroplastic concentrations in the soil samples ranged from 0.1 to 324.5 kg/ha, with an average of 83.6 kg/ha; the concentrations were higher in western China than in eastern China. A highly significant linear correlation (R2 = 0.61) was found between the consumption of mulching film and the plastic residue in soils, indicating plastic film mulching may be a major source of macroplastics. The abundances of microplastic particles increased over time in the locations where plastic mulching was continuously employed, with concentrations of 80.3 ± 49.3, 308 ± 138.1, and 1075.6 ± 346.8 pieces/kg soil in fields with 5, 15, and 24 y of continuous mulching, respectively. Fourier transform infrared analyses revealed that the composition of the microplastics matched that of the mulching films, suggesting the microplastic particles originated from the mulching films. The Mediterranean basin is a hotspot of mercury (Hg) contamination owing to intense anthropogenic emissions, volcanic activity and oligotrophic conditions. Little work has been done to assess the sources of Hg exposure for seabirds and, particularly, the physiological consequences of Hg bioaccumulation. In this study, we (i) describe the individual and temporal variation in blood concentration of total Hg (THg) over three breeding seasons, (ii) identify the factors that affect the THg exposure and (iii) determine the individual- and population-level connections between THg and blood-based markers of oxidative status in chicks of Scopoli's shearwaters (Calonectris diomedea) breeding on the island of Linosa in the southern Mediterranean. We carried out the work on chicks near fledging because they are fed with prey captured near the colony, thus their Hg levels reflect local contamination. The concentration of THg in erythrocytes varied from 0.23 to 4.29 μg g-1 dw. Chicks that were fed upon higher trophic level prey (i.e., higher δ15N values) had higher THg levels. Individual variation in THg concentrations was not explained by parental identity, sex nor δ13C values. There was significant variation in THg among chicks born from the same mother in different years. We found significant correlations between THg and markers of oxidative status; however, these correlations were no longer significant when we took into account the annual variation in mean values of all metrics. Males with higher values of body condition index had higher blood THg, while THg and body condition index were not correlated in females. Our data indicate that THg levels were moderate to high if compared to other seabirds. However, there is little evidence for harmful short-term detrimental effects owing to THg exposure. Thus, high-resolution analysis of dust storms and comparison of dust sources in different regions of northern China are important to clarify the formation mechanism of East Asian dust storms and predict or even prevent such storms. Here, we analyzed spatiotemporal trends in dust storms that occurred in three main dust source regions during 1960-2007 Taklimakan Desert (western region [WR]), Badain Jaran and Tengger Deserts (middle region [MR]), and Otindag Sandy Land (eastern region [ER]). We analyzed daily dust storm frequency (DSF) at the 10-day scale (first [FTDM], middle [MTDM], and last [LTDM] 10 days of a month), and investigated the association of dust storm occurrences with meteorological factors. The 10-day DSF was greatest in the FTDM (accounting for 77.14% of monthly occurrences) in the WR, MTDM (45.85%) in the MR, and LTDM (72.12%) in the ER, showing a clear trend of movement from the WR to the ER. Temporal analysis of DSF revealed trend changes over time at annual and 10-day scales, with mutation points at 1985 and 2000.

Descriptive statistics was used to present data as frequencies, proportions and percentages. Chi-square test and logistic regression analysis were used to evaluate relationships between variables. Two hundred and nine students from the two optometry training institutions in Ghana; Kwame Nkrumah University of Science and Technology (49.8%) and University of Cape Coast (51.2%) responded to the questionnaire. The mean (SD) age of students was 23.6 (1.9) years (males = 65.6%). On seeking admission into the university, optometry (65.6%) and human biology/medicine (28.2%) were the leading first choice programmes among participants. Participants largely aspire to be in clinical practice (64.6%) or Academia/Research (28.2%). The major factors which influenced career choices were interest in career field (64.1%) and potential good income (38.3%). Females were twice more likely to practice optometry and pursue an interest in paediatric optometry than males. Institution of study (p = .028) and information on career opportunities (p = .018) were significant predictors of students' decision to pursue a career in academia/research. Optometry students in Ghana largely aspire to be in clinical practice, a finding which is useful for optometry training institutions and relevant stakeholders in developing the optometry programme and projecting its future in Ghana.Background Retesting for HIV is critical to identifying newly-infected persons and reinforcing prevention efforts among at-risk adults. Incentives can increase one-time HIV testing, but their role in promoting retesting is unknown. We sought to test feasibility and acceptability of incentive strategies, including commitment contracts, to promote HIV retesting among at-risk adults in rural Uganda. Methods At-risk HIV-negative adults were enrolled in a pilot trial assessing feasibility and acceptability of incentive strategies to promote HIV retesting three months after enrollment. Participants were randomized (113) to 1) no incentive; 2) standard cash incentive (~US$4); and 3) commitment contract participants could voluntarily make a low- or high-value deposit that would be returned with added interest (totaling ~US$4 including the deposit) upon retesting or lost if participants failed to retest. Contracts sought to promote retesting by leveraging loss aversion and addressing present bias via pre-commitment. Outcomes included acceptability of trial enrollment, contract feasibility (proportion of participants making deposits), and HIV retesting uptake. Results Of 130 HIV-negative eligible adults, 123 (95%) enrolled and were randomized 74 (60%) to commitment contracts, 25 (20%) to standard incentives, and 24 (20%) to no incentive. Of contract participants, 69 (93%) made deposits. Overall, 93 (76%) participants retested for HIV uptake was highest in the standard incentive group (22/25 [88%]) and lowest in high-value contract (26/36 [72%]) and no incentive (17/24 [71%]) groups.
