The ligand, KMP was able to quench the intrinsic fluorescence of these three proteins efficiently through static quenching mode. The binding constant (Kb) for the interactions of KMP with these three proteins were found in the following order HSA-KMP > BHb-KMP > HEWL-KMP. Different non-covalent forces such as hydrogen bonding and hydrophobic forces played a major role in the binding of KMP with HSA and HEWL, whereas hydrogen bonding and van der Waals forces contribute to the complexation of BHb with KMP. KMP was able to alter the micro-environment near the Trp fluorophore of the proteins. KMP altered the secondary structural component of all three proteins. The putative binding sites and the residues surrounding the KMP molecule within the respective protein matrix were determined through molecular docking and molecular dynamics (MD) simulation studies. The conformational flexibility of the ligand KMP and the three individual proteins were also evident from the MD simulation studies. Meat Standards Australia (MSA) sensory protocols have been effectively utilised in beef for international consumers however, to date sheepmeat testing is largely limited to Australian consumers. This study measured the sensory responses (liking of odour, tenderness, juiciness, liking of flavour, and overall liking) of 2160 untrained American, Australian and Chinese consumers to grilled longissimus lumborum (LL) and semimembranosus (SM) muscles from 164 lambs and 168 yearlings. Across countries there was no difference in juiciness or overall liking sensory scores. American consumers scored tenderness, flavour and odour slightly higher than Australian consumers, and Chinese consumer scores were lowest. Consistently for all countries, sensory scores were greatest in the LL muscle, in lambs compared to yearlings particularly for the LL, and Merino sired and female lambs. These results indicate that cultural background has minimal impact on sensory perceptions of sheepmeat, and provides valuable information for future eating quality prediction models. Such divisions are viewed as a threat to the advancement of science because they tend to polarise researchers and tear apart scientific communities. The objective of this article is to highlight that a piece of information is not either intelligence or evidence, and that a forensic scientist is not either an investigator or an evaluator, but that these notions must all be applied in conjunction to successfully understand a criminal problem or solve a case. To capture the scope, strength and contribution of forensic science, this paper proposes a progressive but non-linear continuous model that could serve as a guide for forensic reasoning and processes. In this approach, hypothetico-deductive reasoning, iterative thinking and the notion of entropy are used to frame the continuum, situate forensic scientists' operating contexts and decision points. Situations and examples drawn from experience and practice are used to illustrate the approach. The authors argue that forensic science, as a discipline, should not be defined according to the context it serves (i.e. an investigation, a court decision or an intelligence process), but as a general, scientific and holistic trace-focused practice that contributes to a broad range of goals in various contexts. Since forensic science does not work in isolation, the approach also provides a useful basis as to how forensic scientists should contribute to collective and collaborative problem-solving to improve justice and security. INTRODUCTION Plastination allows anatomical samples to be preserved in excellent condition for an indefinite period, free of formalin, and in a format that allows biosafe manipulation by students, academics, and researchers. As with other tissue preservation techniques, it is important to establish the level of conservation of deoxyribonucleic acid (DNA) for use in future applications. The object of the present work was to extract and evaluate DNA from plastinated tissues. METHODS We used samples of liver from Canis lupus familiaris and skeletal muscle from Rattus norvegicus, Sprague-Dawley strain, extracted from specimens plastinated with silicone at room temperature. The tissue samples were deplastinated by incubation in 5% sodium methoxide dissolved in methanol for 24 or 48 h. The samples were divided into two equal parts and DNA was extracted using two different protocols. After extraction, the DNA was quantified by fluorometry and its integrity was assessed by electrophoresis in a 1% agarose gel. RESULTS AND DISCUSSION A high yield of DNA was obtained from the deplastinated samples and the DNA was intact.

  • OBJECTIVE AND BACKGROUND Developmental endothelial locus-1 (Del-1), lymphocyte function-associated antigen-1 (LFA-1), and interleukin 17 (IL-17) play critical roles in transendothelial migration of neutrophils in periodontal diseases. The aim of this study was to evaluate salivary Del-1, IL-17, and LFA-1 protein levels in patients with gingivitis (G), chronic periodontitis (CP), and generalized aggressive periodontitis (GAP). METHODS A total of 180 systemically healthy, non-smoking patients (45 periodontally healthy (H) and 45 G, 50 CP, and 40 GAP) individuals (between March 2014 and February 2016) were included in this study according to Armitage's (1999) classification. Clinical periodontal parameters, including clinical attachment level, probing depth, plaque index, and gingival index, were recorded. Del-1, IL-17, and LFA-1 protein expression levels were measured in unstimulated saliva samples collected from patients by using enzyme-linked immunosorbent assays. Kruskal-Wallis and Mann-Whitney U tests were Del-1, LFA-1, and IL-17 before and after periodontal therapy are required to understand the exact roles of these cytokines during the periodontal healing period. © 2020 John Wiley & Sons A/S. BACKGROUND The dual process theory suggests that both conscious and non-conscious processes influence the adoption of physical activity. However, the role of regulatory focus and time preference in this process was not yet examined. DESIGN A cross-sectional online survey among 143 nurses during August-November 2017. METHODS Validated measures were used to estimate physical activity habit strength (a non-conscious process) and intention, planning and behaviour control (conscious processes), physical activity level, regulatory focus and time preference. Multivariable ordered logit and logistic models were specified to examine determinants of both processes. RESULTS Promotion focus was positively associated with having a strong habit of physical activity among nurses with moderate-to-low activity levels, but it was negatively associated with habit for active nurses. As for time preference, higher delay discounting was negatively associated with nurses' conscious intention to adopt physical activity and with their action planning. CONCLUSION Promotion focus and time preference are associated with both conscious and non-conscious processes of physical activity adoption among nurses and should be considered in future health promotion interventions targeted to this population. IMPACT Promotion focus and time preference have a significant role in this dual process. OBJECTIVE To determine the effect of treatment with an occlusal splint (OS), manual therapy (MT), counselling (CS) and the combination of an occlusal splint and counselling (OSCS) on pain and anxiety in patients with TMD. MATERIALS AND METHODS A randomised clinical trial was conducted with 89 patients diagnosed with TMD through RDC/TMD (Research Diagnostic Criteria for Temporomandibular Disorders) and divided into four groups of treatment OSCS (n = 25); OS (n = 24); MT (n = 21); and CS (n = 19). Participants were assessed before and after 1 month of therapy for pain, anxiety and TMD diagnosis. Pain was measured by a visual analogue scale. To assess anxiety, Hospital Anxiety and Depression Scale (HADS), **** Anxiety Inventory (BAI) and the State-Trait Anxiety Inventory (STAI-S and T) were used. The data were analysed using SPSS (Statistical Package for Social Science) 22.0. RESULTS The four groups obtained a significant reduction (P  less then  0.001) in the pain after 1 month of treatment. Treatment in all groups promoted a significant reduction in anxiety symptoms 1 month after completion, HADS (P  less then  0.001), BAI (P  less then  0.001), STAI-T (P = 0.006). Thus, no group was superior to the other in reducing the studied variables. CONCLUSION The therapies used were effective in reducing pain and anxiety in patients diagnosed with TMD. In this study, the reversed lipid-based nanoparticle (RLBN) technique was used to encapsulate a water-soluble drug to facilitate the preparation of SLM. METHODS The model drug used was atomoxetine hydrochloride (ATX), and a three-step method was used to prepare ATX-RLBN. Taste-masking microsphere (ATX-RLBN-SLM) was prepared by the spray chilling method. The drug release mechanism was studied by high-performance liquid chromatography and scanning electron microscopy. Moreover, in vitro taste evaluation method was established and ATX bioavailability was investigated employing pharmacokinetic studies. KEY FINDINGS The obtained ATX-RLBN-SLM had smooth spherical particles with a size of about 80 μm. The drug encapsulation and loading efficiencies were 98.28% ± 0.59% and 0.89% ± 0.04%, respectively. In vitro drug release studies showed that nearly 96% drug was retained in the microspheres within 10 min at pH 6.8 and a complete release was triggered by lipase, accompanied by variation in the morphology. Taste assessment revealed that ATX-RLBN-SLM could efficiently mask the bitter taste and improved the bioavailability of ATX. CONCLUSIONS Atomoxetine hydrochloride-reversed lipid-based nanoparticle-solid lipid microsphere exhibited excellent taste-masking effect with negligible leakage in the oral cavity environment and thorough collapse upon lipase stimulation, simultaneously enhancing the bioavailability of ATX.   • In this paper we examined the photocatalytic efficiency of a laser-synthesized colloidal solution of ZnO nanoparticles synthesized by laser ablation in water. The average size of the obtained colloidal ZnO nanoparticles is about 47 nm. As revealed by electron microscopy, other nanostructures were also present in the colloidal solution, especially nanosheets. A photocatalytic degradation of UV-irradiated Methylene Blue and Rhodamine B solutions of different concentration in the presence of different ZnO catalyst mass concentrations was studied in order to examine their influence on photodegradation rates. ZnO nanoparticles have shown high photocatalytic efficiency, which is limited due to different effects related to UV light transmittivity through the colloidal solution. Therefore, increasing catalyst concentration is effective way to increase photocatalytic efficiency up to some value where photodegradation rate saturation occurs. The photodegradation rate increases as the dye concentration decreases. Adenine-fed rats developed renal dysfunction, glomerular and tubulointerstitial damage, hypertension, placental abnormalities, and reduced fetal weights.     In this paper we examined the photocatalytic efficiency of a laser-synthesized colloidal solution of ZnO nanoparticles synthesized by laser ablation in water. The average size of the obtained colloidal ZnO nanoparticles is about 47 nm. As revealed by electron microscopy, other nanostructures were also present in the colloidal solution, especially nanosheets. A photocatalytic degradation of UV-irradiated Methylene Blue and Rhodamine B solutions of different concentration in the presence of different ZnO catalyst mass concentrations was studied in order to examine their influence on photodegradation rates.     Glaucoma, a worldwide eye disease, may cause irreversible vision damage. If not treated properly at an early stage, glaucoma eventually deteriorates into blindness. Various glaucoma screening methods, e.g. Ultrasound Biomicroscopy (UBM), Optical Coherence Tomography (OCT), and Heidelberg Retinal Scanner (HRT), are available. However, retinal fundus image photography examination, because of its low cost, is one of the most common solutions used to diagnose glaucoma. Clinically, the cup-to-disk ratio is an important indicator in glaucoma diagnosis. Therefore, precise fundus image segmentation to calculate the cup-to-disk ratio is the basis for screening glaucoma.

    In this paper, we propose a deep neural network that uses anatomical knowledge to guide the segmentation of fundus images, which accurately segments the optic cup and the optic disc in a fundus image to accurately calculate the cup-to-disk ratio. Optic disc and optic cup segmentation are typical small target segmentation problems in biomedical images. We propose to use an attention-based cascade network to effectively accelerate the convergence of small target segmentation during training and accurately reserve detailed contours of small targets.

    Our method, which was validated in the MICCAI REFUGE fundus image segmentation competition, achieves 93.31% dice score in optic disc segmentation and 88.04% dice score in optic cup segmentation. Moreover, we win a high CDR evaluation score, which is useful for glaucoma screening.

  • What affects the efficacy of alendronate for prevention of glucocorticoid-induced (GI) fractures remains unclear. We aimed to explore the factors affecting alendronate's efficacy, and further identify subgroup effects of alendronate in preventing GI fractures.

    We searched 3 databases. Random-effects meta-analysis was conducted to synthesize risk ratio (RR) and 95% confidence interval (CI) for each endpoint. Meta-regression analysis was used to explore sources of heterogeneity, and subgroup analysis was used to address heterogeneity and evaluate subgroup effects. We detected publication bias using funnel plots and Egger tests.

    We included 13 papers from 12 unique studies involving 46431 participants. Glucocorticoid (GC) dosage (P = .053) and proportion of previous vertebral fracture (PVF) (P = .047) were probably 2 sources of heterogeneity in meta-analysis for vertebral fractures, while GC duration (P = .020) was probably 1 for nonvertebral fractures. Alendronate reduced vertebral fractures in the high dosage subgroup (RR 0.61, 95% CI 0.44-0.86), but didn't in the low dosage subgroup (RR 1.56, 95% CI 0.20-12.02). Alendronate reduced vertebral fractures (RR 0.53, 95% CI 0.40-0.68) in the subgroup of PVF proportion <5%, but didn't (RR 0.76, 95% CI 0.42-1.37) in the subgroup of this proportion ≥5%. Alendronate reduced nonvertebral and hip fractures, whether in primary or in secondary prevention subgroup.

    The findings in our study support that alendronate is used for the primary and secondary prevention of GI fractures, but do not support that alendronate is recommended as a first-line agent for patients receiving a low dose of GCs or patients with PVF.

The aim was to investigate the distribution characteristics of ABO and RhD blood groups in Chongqing, and to provide scientific-effective data for a more reasonable blood collection program. We retrospectively analyzed 795 698 blood donors who had donated blood from 2014 to 2019 at the Chongqing Blood Center. The data on ABO and RhD blood groups were extracted based on blood management system. We used percentages to describe the extraction of data on blood donors by gender, age, and nationality. The data on the distribution of ABO (A, B, AB and O) and RhD (RhD-Positive and RhD-Negative) blood groups were reported also in percentages. Of those, 427 516 (53.73%) were males and 368 182 (46.27%) were females. Among all the blood donors, 321 916 (40.46%) were under the age of 25, followed by 26-35 years age group (22.65%), 36-45 years age group (18.95%), 46-55 years age group (16.98%) and 56-60 years age group (0.96%). There were 755439 (94.94%) of the blood donors who were of the Han nationality. The distribution of blood groups O, A, B and AB were 35.54%, 31.90%, 24.14% and 8.42%, respectively. The distributions of RhD-Negative group was found in 4362 (0.55%) blood donors. The distribution characteristics of the ABO and RhD blood group should be considered when improving blood collection program in Chongqing.

Logistic regression was used to establish a model to predict the vitreous volume.A total of 280 emmetropic eyes and 290 eyes with pathological myopia were included. Males represented 60.7% and 65.5% of the individuals. The mean axial lengths of those two groups were 23.1 ± 0.8 mm (95%CI 22.7-23.4 mm) and 28.3 ± 2.2 mm (95%CI 27.5-29.2 mm), respectively (P  less then  .001). The regression model in the pathological myopic group for calculating the vitreous volume according to the axial length was Vitreous volume = 546.27 × axial length - 6977.12. The regression model in the emmetropic group for calculating the vitreous volume according to the axial length was Vitreous volume = 458.35 × axial length - 6331.14 (R = 0.360, P = .001).Elongation of the axial length is involved in eyeball enlargement in pathological myopic eyes. Measurement of the axial length could be recommended for the estimation of the vitreous volume during vitrectomy if vitreous cavity filling is needed.
    What affects the efficacy of alendronate for prevention of glucocorticoid-induced (GI) fractures remains unclear. We aimed to explore the factors affecting alendronate's efficacy, and further identify subgroup effects of alendronate in preventing GI fractures. We searched 3 databases. Random-effects meta-analysis was conducted to synthesize risk ratio (RR) and 95% confidence interval (CI) for each endpoint. Meta-regression analysis was used to explore sources of heterogeneity, and subgroup analysis was used to address heterogeneity and evaluate subgroup effects. We detected publication bias using funnel plots and Egger tests. We included 13 papers from 12 unique studies involving 46431 participants. Glucocorticoid (GC) dosage (P = .053) and proportion of previous vertebral fracture (PVF) (P = .047) were probably 2 sources of heterogeneity in meta-analysis for vertebral fractures, while GC duration (P = .020) was probably 1 for nonvertebral fractures. Alendronate reduced vertebral fractures in the high dosage subgroup (RR 0.61, 95% CI 0.44-0.86), but didn't in the low dosage subgroup (RR 1.56, 95% CI 0.20-12.02). Alendronate reduced vertebral fractures (RR 0.53, 95% CI 0.40-0.68) in the subgroup of PVF proportion <5%, but didn't (RR 0.76, 95% CI 0.42-1.37) in the subgroup of this proportion ≥5%. Alendronate reduced nonvertebral and hip fractures, whether in primary or in secondary prevention subgroup. The findings in our study support that alendronate is used for the primary and secondary prevention of GI fractures, but do not support that alendronate is recommended as a first-line agent for patients receiving a low dose of GCs or patients with PVF. The aim was to investigate the distribution characteristics of ABO and RhD blood groups in Chongqing, and to provide scientific-effective data for a more reasonable blood collection program. We retrospectively analyzed 795 698 blood donors who had donated blood from 2014 to 2019 at the Chongqing Blood Center. The data on ABO and RhD blood groups were extracted based on blood management system. We used percentages to describe the extraction of data on blood donors by gender, age, and nationality. The data on the distribution of ABO (A, B, AB and O) and RhD (RhD-Positive and RhD-Negative) blood groups were reported also in percentages. Of those, 427 516 (53.73%) were males and 368 182 (46.27%) were females. Among all the blood donors, 321 916 (40.46%) were under the age of 25, followed by 26-35 years age group (22.65%), 36-45 years age group (18.95%), 46-55 years age group (16.98%) and 56-60 years age group (0.96%). There were 755439 (94.94%) of the blood donors who were of the Han nationality. The distribution of blood groups O, A, B and AB were 35.54%, 31.90%, 24.14% and 8.42%, respectively. The distributions of RhD-Negative group was found in 4362 (0.55%) blood donors. The distribution characteristics of the ABO and RhD blood group should be considered when improving blood collection program in Chongqing. Copyright © Wang et al.Effect of atorvastatin combined with routine therapy on the expression of hypoxia inducible factor (HIF-1) and vascular endothelial growth factor (VEGF) in rats with acute myocardial infarction (AMI) and its therapeutic effect were investigated. The rat models of acute myocardial infarction were established and divided into routine therapy, study, model, single drug and control group according to the treatment plan, with 10 cases in each group. Enzyme-linked immunosorbent assay (ELISA) was used to detect the concentration of HIF-1 and VEGF in serum of rats before treatment (T0), and 3 days (T1), 5 days (T2) and 7 days (T3) after treatment, and the cardiac function was measured at the same time. The concentrations of HIF-1 and VEGF in serum of the study group and the routine therapy group after treatment were significantly higher than those before treatment. The concentrations of HIF-1 and VEGF in serum of model group at T0 were significantly lower than those at T3 (P less then 0.05). After treatment, the concentrations of HIF-1 and VEGF in serum of the study group were significantly higher than those of the routine therapy, the model and the control group (P less then 0.05). One week after administration, there were significant differences in the left ventricular function among the five groups (P less then 0.001). The left ventricular function in the study group was better than that in the routine therapy, model and control group. The levels of HIF-1 and VEGF in the serum of rats with myocardial infarction were negatively correlated with LVIDs and LVIDd, and positively correlated with LVEF% and LVFS%. In conclusion, atorvastatin combined with routine therapy can better reduce serum HIF-1 and VEGF levels and improve the left ventricular function in rats than routine therapy. The percentages of CD4+CD25+FOXP3+ Tregs in CD4+ T lymphocytes from 15 children with asthma and 15 healthy controls were measured by flow cytometry, and FOXP3 mRNA expression in the CD4+CD25+ Tregs was measured by reverse transcriptase-quantitative PCR. In addition, the forced expiratory volume in one second (FEV1) was measured to determine lung function. The methylation statuses of 16 CpG sites in two regions of the FOXP3 gene's exon and intron were analysed with bisulfite-specific PCR and pyrophosphate sequencing. The differences in methylation levels between the asthma and control groups were compared. The percentage of CD4+CD25+FOXP3+ Tregs in CD4+ T lymphocytes and FOXP3 mRNA expression were significantly lower in children with asthma than in control children (P less then 0.05). The FOXP3 mRNA levels in children with asthma were positively correlated with FEV1 (P less then 0.001; r=0.895). The methylation levels in 12 of the 16 studied CpG loci of the FOXP3 gene, and of the 6th CpG locus in the exon regions, were significantly higher in the asthma group compared with the control group (P less then 0.05). In the present study, we aimed to evaluate the in vitro effects of vasoconstrictive retraction agents on primary human gingival fibroblasts (HGFs). A total of six commercial adrenergic solutions (0.05 and 0.01% HCl-epinephrine, two based on 0.05% HCl-tetrahydrozoline, 0.05% HCl-oxymetazoline, and 10% HCl-phenylephrine) and three experimental gel formulations (EG-1, EG-2, and EG-3) were used to treat primary HGFs. The biological effect of the retraction treatment on the expression of collagen types I and III was detected by performing immunocytochemical analysis. The generation of reactive oxygen species triggered by the retraction agents were evaluated by using the dichlorofluorescein (DCF) fluorescent probe. The effect of retraction agents on the expression of fibronectin was visualized by confocal laser scanning microscopy. According to the results, experimental retraction gels did not limit the expression of collagen types I and III. EG-3 even induced the synthesis of both types of collagen. The DCF assay indicated oxidative stress similar to the control cells for most of the selected retraction agents. Experimental gels did not cause degradation of the cellular shape and morphology of the primary HGFs. The proposed experimental retraction gels in the present study demonstrated higher biocompatibility with primary HGFs, suggesting their use as clinical vasoconstrictive agents for the application of gingival retraction with minimal damage to periodontal tissues. AQP4 is a water channel protein and mediates water homeostasis in a variety of brain disorders. In the current study, a rat TBI model was established, and the features of brain edema following TBI were assessed using multimodal MRI. The results of the multimodal MRI were useful, reliable and were used to evaluate the extent and the type of brain edema following TBI. Brain edema was also successfully alleviated using an intracerebral injection of AQP4 small interfering (si)RNA. The expression of AQP4 and its role in brain edema were also examined in the present study. The AQP4 siRNA was demonstrated to downregulate AQP4 expression following TBI and reduced brain edema at the early stages of TBI (6 and 12 h).

  • The common bed bug, Cimex lectularius L., resurged as an important urban pest in the last 20 yr. Yet, there are no commercial repellent products labeled for bed bugs available in the United States. We evaluated the repellency of two catnip oils from newly developed cultivars, CR3 and CR9, and compared each to 10 and 25% N,N-diethyl-meta-toluamide (DEET). CR3 contains 63.4% E,Z-nepatalactone and 27.5% Z,E-nepatalactone, and CR9 contains 94.9% Z,E-nepatalactone. Arena studies showed that CR3 and CR9 oils were more effective than DEET within a 24-h period. At 10% concentration, both CR3 and CR9 oils exhibited a repellency of over 94% during the first 8 h. At 25% concentration, the repellency of CR3 and CR9 oils increased to 100%, and repellency of DEET was 92% during the first 24 h. Repellency of 25% CR3 and CR9 oils became lower than 25% DEET after being aged for 3 d. After 28-d aging, repellency of 25% CR3, CR9, and DEET reduced to 25, 64, and 92%, respectively. Soiled socks were placed above repellent treated bands to determine if the repellent can protect soiled socks from being infested. The 20% CR3 oil prevented 100% of bed bugs from infesting soiled socks showing that it was more effective than DEET. These results indicate that catnip oils from CR3 and CR9 cultivars are more repellent than DEET over a 24-h period following application, but their longevity is shorter than DEET after 72 h.Following the deregistration of lindane, several neonicotinoid insecticides have been registered as seed treatments for controlling wireworms in cereal crops. Unlike lindane, which did both, neonicotinoids provide crop protection but do not reduce wireworm populations. Hence populations of various economic species are growing in key wheat production areas of the United States and Canada, and there is a need for novel seed treatments that match the efficacy of lindane. https://www.selleckchem.com/products/glafenine.html Herein we evaluated broflanilide, a novel GABA-gated Cl- channel allosteric modulator that differs from the cyclodienes (e.g., lindane) in that it binds to a unique site in the GABA receptor. As such, broflanilide confers a new mode of insecticidal action (Group 30 MOA), and is the first meta-diamide insecticide developed. When evaluated in field trials over 7 yr at various rates, broflanilide at 5.0 g [AI]/100 kg wheat seed was as effective at protecting wheat stand from wireworm (Agriotes obscurus) injury as the current industry standard thiamethoxam at 20-30 g [AI]/100 kg seed. In addition, broflanilide at 5.0 g reduced neonate wireworms (produced from eggs laid in established wheat during the growing season) and resident wireworms (in the field at time of planting) by 73.1 and 81.1%, respectively, which is comparable to that reported for the previous industry standard lindane (75.3 and 57.6%, respectively). These studies show that broflanilide at 5.0 g [AI] will provide consistent wheat stand protection (equal to thiamethoxam at 20-30 g [AI]), and A. obscurus wireworm population reduction (equal to lindane at 59 g [AI]), and will do so at far lower dosages per hectare.
    In the past two decades, scientists in different laboratories have assayed gene expression from millions of samples. These experiments can be combined into compendia and analyzed collectively to extract novel biological patterns. Technical variability, or "batch effects," may result from combining samples collected and processed at different times and in different settings. Such variability may distort our ability to extract true underlying biological patterns. As more integrative analysis methods arise and data collections get bigger, we must determine how technical variability affects our ability to detect desired patterns when many experiments are combined.

    We sought to determine the extent to which an underlying signal was masked by technical variability by simulating compendia comprising data aggregated across multiple experiments.

    We developed a generative multi-layer neural network to simulate compendia of gene expression experiments from large-scale microbial and human datasets. We compared simulated compendia before and after introducing varying numbers of sources of undesired variability.

    The signal from a baseline compendium was obscured when the number of added sources of variability was small. Applying statistical correction methods rescued the underlying signal in these cases. However, as the number of sources of variability increased, it became easier to detect the original signal even without correction. In fact, statistical correction reduced our power to detect the underlying signal.

Platelets express receptors that are important for platelet adhesion, aggregation, participation in inflammatory responses, and for triggering degranulation and enhancing thrombin generation. They carry a cargo of granules bearing enzymes, adhesion molecules, growth factors and cytokines, and have the ability to generate reactive oxygen species. The platelet is at the frontline of a host of cellular responses to invading pathogens, injury, and infection. Perhaps because of this intrinsic responsibility of a platelet to rapidly respond to thrombotic, pathological and immunological factors as part of their infantry role; platelets are susceptible to targeted attack by the adaptive immune system. Such attacks are often transitory but result in aberrant platelet activation as well as significant loss of platelet numbers and platelet function, paradoxically leading to elevated risks of both thrombosis and bleeding. Here, we discuss the main molecular events underlying immune-based platelet disorders with specific focus on events occurring at the platelet surface leading to activation and clearance.

Arena studies showed that CR3 and CR9 oils were more effective than DEET within a 24-h period. At 10% concentration, both CR3 and CR9 oils exhibited a repellency of over 94% during the first 8 h. At 25% concentration, the repellency of CR3 and CR9 oils increased to 100%, and repellency of DEET was 92% during the first 24 h. Repellency of 25% CR3 and CR9 oils became lower than 25% DEET after being aged for 3 d. After 28-d aging, repellency of 25% CR3, CR9, and DEET reduced to 25, 64, and 92%, respectively. Soiled socks were placed above repellent treated bands to determine if the repellent can protect soiled socks from being infested. The 20% CR3 oil prevented 100% of bed bugs from infesting soiled socks showing that it was more effective than DEET. When evaluated in field trials over 7 yr at various rates, broflanilide at 5.0 g [AI]/100 kg wheat seed was as effective at protecting wheat stand from wireworm (Agriotes obscurus) injury as the current industry standard thiamethoxam at 20-30 g [AI]/100 kg seed. We developed a generative multi-layer neural network to simulate compendia of gene expression experiments from large-scale microbial and human datasets. We compared simulated compendia before and after introducing varying numbers of sources of undesired variability. The signal from a baseline compendium was obscured when the number of added sources of variability was small. Applying statistical correction methods rescued the underlying signal in these cases. However, as the number of sources of variability increased, it became easier to detect the original signal even without correction.     The ImPACT® Quick Test is a brief iPad-based battery of neurocognitive tests that has been standardized on a sample of children, adolescents, and adults (ages 12-70). This study provides information on the prevalence of ImPACT® Quick Test scores falling below specific percentiles in the normative sample to aid in clinical interpretation and reduce the risk of over-interpreting, or misinterpreting, a single low score.

    Participants were 683 individuals ranging in age from 12 to 70, who were assessed individually. The ImPACT® Quick Test includes five subtests, contributing to three factor scores motor speed, memory, and attention tracker. The prevalence of low factor scores, stratified by age and sex, were calculated using multivariate base rates.

    In the total sample, obtaining 1 or more scores below the 25th percentile was common (base rate, BR=47.2%), but obtaining 2 or more scores in this range was uncommon (BR=15.3%). Similarly, obtaining 1 or more scores below the 16th percentile was common (BR=31.4%), but obtaining two or more scores in this range was uncommon (BR=6.9%). There were small differences in BRs between sexes and the number of low scores was fairly similar across the age groups.

    Postoperative hypercortisolemia mandates further therapy in patients with Cushing's disease (CD). Delayed remission (DR) is defined as not achieving postoperative immediate remission (IR), but having spontaneous remission during long-term follow-up.

    We aimed to develop and validate machine learning (ML) models for predicting DR in non-IR patients with CD.

    We enrolled 201 CD patients, and randomly divided them into training and test datasets. We then used the recursive feature elimination (RFE) algorithm to select features and applied 5 ML algorithms to construct DR prediction models. We used permutation importance and local interpretable model-agnostic explanation (LIME) algorithms to determine the importance of the selected features and interpret the ML models.

    Eighty-eight (43.8%) of the 201 CD patients met the criteria for DR. Overall, patients who were younger, had a low body mass index, a Knosp grade of III-IV, and a tumor not found by pathological examination tended to achieve a lower rate of DR. After RFE feature selection, the Adaboost model, which comprised 18 features, had the greatest discriminatory ability, and its predictive ability was significantly better than using Knosp grading and postoperative immediate morning serum cortisol (PoC). The results obtained from permutation importance and LIME algorithms showed that preoperative 24-hour urine free cortisol, PoC, and age were the most important features, and showed the reliability and clinical practicability of the Adaboost model in DC prediction.

  • Oxidized LDLs (oxLDLs) and oxysterols play a key role in endothelial dysfunction and the development of atherosclerosis. The loss of vascular endothelium function negatively impacts vasomotion, cell growth, adhesiveness and barrier functions. While for some of these disturbances, a reasonable explanation can be provided from a mechanistic standpoint, for many others, the molecular mediators that are involved are unknown. Caveolae, specific plasma membrane domains, have recently emerged as targets and mediators of oxLDL-induced endothelial dysfunction. Caveolae and their associated protein caveolin-1 (Cav-1) are involved in oxLDLs/LDLs transcytosis, mainly through the scavenger receptor class B type 1 (SR-B1 or SCARB1). In contrast, oxLDLs endocytosis is mediated by the lectin-like oxidized LDL receptor 1 (LOX-1), whose activity depends on an intact caveolae system. In addition, LOX-1 regulates the expression of Cav-1 and vice versa. On the other hand, oxLDLs may affect cholesterol plasma membrane content/distribution thus influencing caveolae architecture, Cav-1 localization and the associated signalling. Overall, the evidence indicate that caveolae have both active and passive roles in oxLDL-induced endothelial cell dysfunction. First, as mediators of lipid uptake and transfer in the subendothelial space and, later, as targets of changes in composition/dynamics of plasma membrane lipids resulting from increased levels of circulating oxLDLs. The introduction of CBD as the medicine Epidiolex in the United States (in 2018) and as Epidyolex in the European Union (in 2019) as the first cannabis-derived therapeutic for the treatment of seizures was underpinned by preclinical research performed at the University of Reading. As professional anthropologists and geneticists aim to accurately communicate their science and its implications for understanding human differences to the public, they must contend with these substantive claims and metapolitical contexts.Protein glycosylation is crucial for the central nervous system and brain functions, including processes that are defective in Alzheimer disease (AD) such as neurogenesis, synaptic function, and memory formation. Still, the roles of glycans in the development of AD are relatively unexplored. Glycomics studies of cerebrospinal fluid (CSF) have previously shown altered glycosylation pattern in patients with different stages of cognitive impairment, including AD, compared to healthy controls. As a consequence, we hypothesized that the glycan profile is altered in the brain of patients with AD and analyzed the asparagine-linked (N-linked) glycan profile in hippocampus and cortex in AD and control brain. Glycans were enzymatically liberated from brain glycoproteins and analyzed by liquid chromatography-tandem mass spectrometry (LC-MS/MS). Eleven glycans showed significantly different levels in hippocampus compared to cortex in both control and AD brain. Two glycans in cortex and four in hippocampus showed different levels in AD compared to control brain. All glycans that differed between controls and AD brain had similar structures with one sialic acid, at least one fucose and a confirmed or potential bisecting N-acetylglucosamine (GlcNAc). The glycans that were altered in AD brain differed from those that were altered in AD CSF. One glycan found to be present in significantly lower levels in both hippocampus and cortex in AD compared to control contained a structurally and functionally interesting epitope that we assign as a terminal galactose decorated with fucose and sialic acid. Altogether, these studies suggest that protein glycosylation is an important component in the development of AD and warrants further studies.

Beyond these substantive aims, on a "metapolitical" level, they serve to reframe concepts and standards for political and scientific discussion of race, challenge structures of academic legitimacy and expertise, and build a cadre of ideological foot soldiers armed with an argumentative toolkit. As professional anthropologists and geneticists aim to accurately communicate their science and its implications for understanding human differences to the public, they must contend with these substantive claims and metapolitical contexts.Protein glycosylation is crucial for the central nervous system and brain functions, including processes that are defective in Alzheimer disease (AD) such as neurogenesis, synaptic function, and memory formation. Still, the roles of glycans in the development of AD are relatively unexplored. Glycomics studies of cerebrospinal fluid (CSF) have previously shown altered glycosylation pattern in patients with different stages of cognitive impairment, including AD, compared to healthy controls. As a consequence, we hypothesized that the glycan profile is altered in the brain of patients with AD and analyzed the asparagine-linked (N-linked) glycan profile in hippocampus and cortex in AD and control brain. Glycans were enzymatically liberated from brain glycoproteins and analyzed by liquid chromatography-tandem mass spectrometry (LC-MS/MS). Eleven glycans showed significantly different levels in hippocampus compared to cortex in both control and AD brain. Two glycans in cortex and four in hippocampus showed different levels in AD compared to control brain. All glycans that differed between controls and AD brain had similar structures with one sialic acid, at least one fucose and a confirmed or potential bisecting N-acetylglucosamine (GlcNAc). The glycans that were altered in AD brain differed from those that were altered in AD CSF. One glycan found to be present in significantly lower levels in both hippocampus and cortex in AD compared to control contained a structurally and functionally interesting epitope that we assign as a terminal galactose decorated with fucose and sialic acid. Altogether, these studies suggest that protein glycosylation is an important component in the development of AD and warrants further studies.
