  • The activation process of G protein-coupled receptors (GPCRs) has been extensively studied, both experimentally and computationally. In particular, Molecular Dynamics (MD) simulations have proven useful in exploring GPCR conformational space. The typical behaviour of class A GPCRs, when subjected to unbiased MD simulations from their crystallized inactive state, is to fluctuate between inactive and intermediate(s) conformations, even with bound agonist. Fully active conformation(s) are rarely stabilized unless a G protein is also bound. In order to thoroughly examine the conformational landscape of A2aR activation, we performed unbiased microsecond-length MD simulations in quadruplicate, starting from the inactive conformation either in apo or with bound agonists endogenous adenosine or synthetic NECA, embedded in two homogeneous phospholipid membranes 1,2-dioleoyl-sn-glycerol-3-phosphoglycerol (DOPG) or 1,2-dioleoyl-sn-glycerol-3-phosphocholine (DOPC). In DOPC with bound adenosine or NECA, we observe transition to an intermediate receptor conformation consistent with the known adenosine-bound crystal state. In apo state in DOPG, two different intermediate conformations are obtained. One is similar to that observed with bound adenosine in DOPC, while the other is closer to the active state but not yet fully active. Exclusively, in DOPG with bound adenosine or NECA, we reproducibly identify receptor conformations with fully active features, which are able to dock Gs protein. These different receptor conformations can be attributed to the action/absence of agonist and phospholipid-mediated allosteric effects on the intracellular side of the receptor.Gene duplication and diversification drive the emergence of novel functions during evolution. Domain-swap experiments identified the α/β domain of Ku80c as the major determinant for its specialized function, while its C-terminal part is required for excision of only a small subset of IESs located in IES-dense regions. We conclude that Ku80c has acquired the ability to license Pgm-dependent DNA cleavage, securing precise DNA elimination during programmed rearrangements. Founded in 2005, the Duke Chemistry Outreach (DCO) employs a pedagogical approach to outreach that aims to teach its audience a new scientific concept, while instilling a pure enjoyment of science. DCO has performed 583 events reaching over 70,000 participants throughout 2,270 hours, with the majority of events in Durham, the surrounding North Carolinian communities, and across 8 other states. The flexibility and diversity of this outreach program creates a framework amendable for others to adopt in both secondary and higher education settings.In 1997, during the 41st session of the Regional Committee for the Eastern Mediterranean, the 21 countries in the World Health Organization (WHO) Eastern Mediterranean Region* (EMR) passed a resolution to eliminate† measles (1). In 2015, this goal was included as a priority in the Eastern Mediterranean Vaccine Action Plan 2016-2020 (EMVAP) (2), endorsed at the 62nd session of the Regional Committee (3). To achieve this goal, the WHO Regional Office for the Eastern Mediterranean developed a four-pronged strategy 1) achieve ≥95% vaccination coverage with the first dose of measles-containing vaccine (MCV1) among children in every district of each country through routine immunization services; 2) achieve ≥95% vaccination coverage with a second MCV dose (MCV2) in every district of each country either through implementation of a routine 2-dose vaccination schedule or through supplementary immunization activities§ (SIAs); 3) conduct high-quality, case-based surveillance in all countries; and 4) provide optimal measles clinical case management, including dietary supplementation with vitamin A (4).

  • There was no significant difference in the lesion area measurements (SD=1.09, P = 0.08). The lesion with the largest difference in area measurements resulted from focal areas of atrophy, misdiagnosed as polypoidal lesions on ICGA, and a low-lying serous retinal pigment epithelial detachment (PED), erroneously identified as part of the BVN by ICGA graders. SS-OCTA imaging correctly diagnosed the focal areas of atrophy and the serous PED. Conclusions SS-OCTA imaging was comparable to ICGA for the diagnosis of treatment-naïve PCV. However, SS-OCTA might be better than ICGA in correctly identifying both polypoidal lesions and BVNs in treatment-naïve PCV.Posterior fossa A (PFA) ependymomas are lethal malignancies of the hindbrain in infants and toddlers. Lacking highly recurrent somatic mutations, PFA ependymomas are proposed to be epigenetically driven tumors for which model systems are lacking. Here we demonstrate that PFA ependymomas are maintained under hypoxia, associated with restricted availability of specific metabolites to diminish histone methylation, and increase histone demethylation and acetylation at histone 3 lysine 27 (H3K27). PFA ependymomas initiate from a cell lineage in the first trimester of human development that resides in restricted oxygen. Unlike other ependymomas, transient exposure of PFA cells to ambient oxygen induces irreversible cellular toxicity. PFA tumors exhibit a low basal level of H3K27me3, and, paradoxically, inhibition of H3K27 methylation specifically disrupts PFA tumor growth. Targeting metabolism and/or the epigenome presents a unique opportunity for rational therapy for infants with PFA ependymoma.The United States (US) spends more on healthcare than any other country with little evidence of better, or even comparable, outcomes. We reflect on the US and the COVID-19 pandemic and focus on cultural, economic and structural barriers that threaten both current and future responses to infectious diseases emergencies. These include the US healthcare delivery model, the defunding of public health, a scarcity of infectious diseases physicians, the market failure of vaccines and anti-infectives and the concept of American exceptionalism. In addition, signaling pathways in breast cancer have been extensively studied as therapeutic targets due to their roles in tumor growth and metastasis. However, it is challenging to put signaling pathways and gene expression profiles together to characterize biological mechanisms of breast cancer subtypes since many signaling events result from post-translational modifications, rather than gene expression differences. We designed a logic-based computational framework to explain the differences in gene expression profiles among breast cancer subtypes using Pathway Logic and transcriptional network information. Pathway Logic is a rewriting-logic-based formal system for modeling biological pathways including post-translational modifications. Our method demonstrated its utility by constructing subtype-specific path from key receptors (TNFR, TGFBR1 and EGFR) to key transcription factor (TF) regulators (RELA, ATF2, SMAD3 and ELK1) and identifying potential association between pathways via TFs in basal-specific paths, which could provide a novel insight on aggressive breast cancer subtypes. To improve the performance of a drug property prediction model, it is important to extract complex molecular dynamics from limited data. Recent machine learning or deep learning based models have employed the atom- and bond-type information, as well as the structural information to predict drug properties. However, many of these methods can be used only for the graph representations. Message passing neural networks (MPNNs) (Gilmer et al., 2017) is a framework used to learn both local and global features from irregularly formed data, and is invariant to permutations. This network performs an iterative message passing (MP) operation on each object and its neighbors, and obtain the final output from all messages regardless of their order. In this study, we applied the MP-based attention network (Nikolentzos et al., 2019) originally developed for text learning to perform chemical classification tasks. Before training, we tokenized the characters, and obtained embeddings of each molecular sequence. We conducted various experiments to maximize the predictivity of the model. We trained and evaluated our model using various chemical classification benchmark tasks. Our results are comparable to previous state-of-the-art and baseline models or outperform. To the best of our knowledge, this is the first attempt to learn chemical strings using an MP-based algorithm. We will extend our work to more complex tasks such as regression or generation tasks in the future.Background Some studies, mainly from high-income countries (HICs), report that women receive less care (investigations and treatments) for cardiovascular disease than do men and might have a higher risk of death. We recorded information on participants' sociodemographic characteristics, risk factors, medication use, cardiac investigations, and interventions.

All rights reserved.Purpose Late gadolinium enhancement (LGE) of the left atrium is susceptible to artifacts arising from the right pulmonary veins, caused by inflowing blood tagged by the navigator restore pulse. The purpose of this study was to evaluate a new method to reduce the inflow artifact using an adaptive flip-angle restore pulse. Methods A low-restore angle reduces the inflow artifact but may lead to a poor navigator SNR. The proposed approach aims to determine the patient-specific restore angle, which optimizes the trade-off between inflow artifacts and navigator SNR. Three-dimensional LGE with adaptive navigator restore (3D LGEA ) was implemented by incrementing the flip angle of the restore pulse from a starting value of 0°, based on the navigator normalized cross-correlation. Magnetic resonance imaging experiments were performed on a 1.5T scanner. The value of 3D LGEA was compared with 3D LGE with a constant 180° restore pulse (3D LGE180 ) in 22 patients with heart diseases. The values of 3D LGEA and 3D LGE180 were compared in terms of pulmonary vein blood signal relative to reference blood in the descending aorta (PVrel ) and visual scoring to determine level of motion artifacts using a 4-point scale (1 = severe artifacts; 4 = no artifacts). Results The value of PVrel was significantly lower for 3D LGEA than for 3D LGE180 (1.16 ± 0.23 vs. 1.59 ± 0.29, P less then .001). Furthermore, visual scoring of the motion artifacts yielded no difference (P = .78). Conclusion Adaptively adjusting the navigator restore flip angle based on the navigator normalized cross-correlation reduces the 3D LGE inflow artifact without affecting image quality or the scan time.Background This study evaluated the acceptability of meat (sensorial and visual analyses) of 40 Nellore heifers (finished in feedlots; for 73 days) fed with different diets supplemented with essential oils (EOs) and an active principle blend. The five diets tested were CON diet without essential oil and active principle blend (eugenol, thymol, and vanillin), ROS diet supplemented with rosemary EO, BLE supplemented with blend, BCL diet with clove EO+ blend and ****diet with rosemary, clove EOs + blend. The acceptance of diet and ageing time were evaluated by consumer. Results Diet affected consumer acceptability and visual analysis (meat color). The diets with EO and the blend showed better sensory acceptance by the consumers; meat aged during seven days obtained the highest scores than meat aged for one day. Meat from heifers that received both EOs + blend (BCL, BRC) obtained the highest scores in the visual evaluation. Conclusion Thus, the use of natural compounds in ruminant diets improves sensory characteristics of meat without damage visual acceptability and may be an alternative to the conventional additive market. We investigated if treatment with zoledronate could prevent bone loss and if the timing of the zoledronate infusion influenced the outcome. 2-year randomized, open label, interventional study including 61 patients with osteopenia, discontinuing denosumab after 4.6 ± 1.6 years. We administrated zoledronate six (6 M group, n = 20) or nine months (9 M group, n = 20) after the last denosumab injection or when bone turnover had increased (OBS group, n = 21). We monitored the patients with DXA and bone turnover markers. Our primary endpoints were change in lumbar spine BMD (LSBMD) six months after zoledronate and the proportion of patients who failed to maintain BMD. The study is ongoing (clinicaltrials.gov NCT03087851). We included 61 participants and 59 patients completed follow-up 12 months after zoledronate. Six months after zoledronate, LSBMD had decreased significantly by 2.1 ± 0.9% (mean ± SEM), 4.3 ± 1.1% and 3.0 ± 1. Conclusions Self-harm content is a fraught issue at the centre of current debates around risks and opportunities for child and adolescent mental health in the digital age. Whilst the importance of supporting young people's online safety is clear, moves to eradicate self-harm content must be undertaken with caution so as not to cause unintentional harm. Our research highlights a need to think beyond a model of contagion, instead attending to other mechanisms of harm and benefit. The purpose of this study was to evaluate a new method to reduce the inflow artifact using an adaptive flip-angle restore pulse. Methods A low-restore angle reduces the inflow artifact but may lead to a poor navigator SNR. The proposed approach aims to determine the patient-specific restore angle, which optimizes the trade-off between inflow artifacts and navigator SNR. Three-dimensional LGE with adaptive navigator restore (3D LGEA ) was implemented by incrementing the flip angle of the restore pulse from a starting value of 0°, based on the navigator normalized cross-correlation. Magnetic resonance imaging experiments were performed on a 1.5T scanner. The value of 3D LGEA was compared with 3D LGE with a constant 180° restore pulse (3D LGE180 ) in 22 patients with heart diseases. The values of 3D LGEA and 3D LGE180 were compared in terms of pulmonary vein blood signal relative to reference blood in the descending aorta (PVrel ) and visual scoring to determine level of motion artifacts using a 4-point scale (1 = severe artifacts; 4 = no artifacts). Results The value of PVrel was significantly lower for 3D LGEA than for 3D LGE180 (1.16 ± 0.23 vs. 1.59 ± 0.29, P less then .001). The five diets tested were CON diet without essential oil and active principle blend (eugenol, thymol, and vanillin), ROS diet supplemented with rosemary EO, BLE supplemented with blend, BCL diet with clove EO+ blend and BRC diet with rosemary, clove EOs + blend. The acceptance of diet and ageing time were evaluated by consumer. Results Diet affected consumer acceptability and visual analysis (meat color). The diets with EO and the blend showed better sensory acceptance by the consumers; meat aged during seven days obtained the highest scores than meat aged for one day. Meat from heifers that received both EOs + blend (BCL, BRC) obtained the highest scores in the visual evaluation. Conclusion Thus, the use of natural compounds in ruminant diets improves sensory characteristics of meat without damage visual acceptability and may be an alternative to the conventional additive market. We investigated if treatment with zoledronate could prevent bone loss and if the timing of the zoledronate infusion influenced the outcome. 2-year randomized, open label, interventional study including 61 patients with osteopenia, discontinuing denosumab after 4.6 ± 1.6 years. We administrated zoledronate six (6 M group, n = 20) or nine months (9 M group, n = 20) after the last denosumab injection or when bone turnover had increased (OBS group, n = 21). We monitored the patients with DXA and bone turnover markers. Our primary endpoints were change in lumbar spine BMD (LSBMD) six months after zoledronate and the proportion of patients who failed to maintain BMD. The study is ongoing (clinicaltrials.gov NCT03087851). We included 61 participants and 59 patients completed follow-up 12 months after zoledronate. Six months after zoledronate, LSBMD had decreased significantly by 2.1 ± 0.9% (mean ± SEM), 4.3 ± 1.1% and 3.0 ± 1. Subsequently, the classifier was applied to N = 26 TW. Finally, we compared brain volumes of CG-men, women, and TW-pre/post treatment cross-sex hormone treatment (CHT) in a univariate analysis controlling for sexual orientation, age, and total brain volume. The application of our biological sex classifier to the transgender sample resulted in a significantly lower true positive rate (TPR-male = 56.0%). The TPR did not differ between CG-individuals with (TPR-male = 86.9%) and without depression (TPR-male = 88.5%). The univariate analysis of the transgender application-sample revealed that TW-pre/post treatment show brain-structural differences from CG-women and CG-men in the putamen and insula, as well as the whole-brain analysis. In order to rapidly discover lead compounds for clinical use, we initiated a program of combined structure-assisted drug design, virtual drug screening and high-throughput screening to identify new drug leads that target the COVID-19 virus main protease (Mpro). Mpro is a key CoV enzyme, which plays a pivotal role in mediating viral replication and transcription, making it an attractive drug target for this virus5,6. Here, we identified a mechanism-based inhibitor, N3, by computer-aided drug design and subsequently determined the crystal structure of COVID-19 virus Mpro in complex with this compound. Next, through a combination of structure-based virtual and high-throughput screening, we assayed over 10,000 compounds including approved drugs, drug candidates in clinical trials, and other pharmacologically active compounds as inhibitors of Mpro. Six of these compounds inhibited Mpro with IC50 values ranging from 0.67 to 21.4 μM. Ebselen also exhibited promising antiviral activity in cell-based assays. Mental health variables were assessed via the Insomnia Severity Index (ISI), the Symptom Check List-revised (SCL-90-R), and the Patient Health Questionnaire-4 (PHQ-4), which included a 2-item anxiety scale and a 2-item depression scale (PHQ-2). RESULTS Compared with nonmedical health workers (n = 1,255), medical health workers (n = 927) had a higher prevalence of insomnia (38.4 vs. 30.5%, p less then 0.01), anxiety (13.0 vs. 8.5%, p less then 0.01), depression (12.2 vs. 9.5%; p less then 0.04), somatization (1.6 vs. 0.4%; p less then 0.01), and obsessive-compulsive symptoms (5.3 vs. 2.2%; p less then 0.01). They also had higher total scores of ISI, GAD-2, PHQ-2, and SCL-90-R obsessive-compulsive symptoms (p ≤ 0.01). Among medical health workers, having organic disease was an independent factor for insomnia, anxiety, depression, somatization, and obsessive-compulsive symptoms (p less then 0.05 or 0.01). Living in rural areas, being female, and being at risk of contact with COVID-19 patients were the most common risk factors for insomnia, anxiety, obsessive-compulsive symptoms, and depression (p less then 0.01 or 0.05). Among nonmedical health workers, having organic disease was a risk factor for insomnia, depression, and obsessive-compulsive symptoms (p less then 0.01 or 0.05). MATERIALS AND METHODS Peak oxygen uptake (V∙O2peak) and exercise-limiting factors were evaluated by a treadmill cardiopulmonary exercise test (CPET) 6-60 months after BLTx. Physical activity was measured with accelerometers, and results were compared with Norwegian normative data and the World Health Organization's (WHO) recommendations for physical activity. RESULTS In 54 included BLTx recipients (mean age 50 ± 15 years, 50% females), V∙O2peak (mL × kg-1 × min-1) was 21.8 ± 7.7 for men and 22.4 ± 6.2 for women, corresponding to 57 ± 17 and 70 ± 12% of predicted, respectively. Three patients (6%) met criteria for normal V∙O2peak.

METHODS A total of 70 patients were randomized into an observation group (35 cases) and a control group (34 cases, 1 case dropped off). Internal medical basic treatment and breathing training were given in both groups. Besides, Xingnao Kaiqiao acupuncture was applied at Neiguan (PC 6), Shuigou (GV 26), Sanyinjiao (SP 6), Jiquan (HT 1), Chize (LU 5) and Weizhong (BL 40) in the control group, the needles were sustained for 30 min. On the basis of treatment in the control group, EA was applied at Tianding (LI 17), Fengchi (GB 20) and Jiaji (EX-B 2, C3-C5 ) in the observation group, with continuous wave, 10-20 Hz, 30 min each time. The treatment was given once a day, 6 times a week for 4 weeks in both groups. Before and after treatment, the diaphragmatic motility was measured by SonoSite ultrasound system (M-Turbo) in the two groups. RESULTS Compared before treatment, the diaphragmatic motility after treatment was increased in both groups (P less then 0.01), and the improvement in the observation group was superior to the control group (P less then 0.01). CONCLUSION Electroacupuncture can promote the recovery of diaphragmatic function in patients with post-stroke tracheotomy tube.OBJECTIVE To compare the clinical therapeutic effect of midnight-noon ebb-flow acupuncture combined with rehabilitation therapy and simple rehabilitation therapy in severe craniocerebral trauma patients with vegetative state. METHODS A total of 100 patients were randomized into an observation group and a control group, 50 cases in each one. Basic treatment of medication, hyperbaric oxygen therapy and specialized nursing were given in both groups. In the control group, rehabilitation therapy was adopted for 30 min each time, once a day. On the basis of the control group, midnight-noon ebb-flow acupuncture was applied in the observation group, the needles were sustained for 30 min, once a day, 5 times a week. The treatment was for 30 days in both groups. Before treatment and after 10, 20, 30 days of treatment, scores of Glasgow coma scale (GCS) and coma recovery scale-revised (CRS-R) were observed, and the conscious rate after treatment was calculated in both groups. RESULTS Compared before treatment, the GCS and CRS-R scores after 10, 20, 30 days of treatment were increased in both groups (P less then 0.01), and the scores in the observation group were superior to those in the control group (P less then 0.01). After treatment, the conscious rate was 20.0% (10/50) in the observation group, which was superior to 12.0% (6/50) in the control group (P less then 0.01). CONCLUSION Midnight-noon ebb-flow acupuncture combined with rehabilitation therapy can effectively treat the severe craniocerebral trauma patients with vegetative state, improve the consciousness level, and have superior therapeutic effect compared with simple rehabilitation therapy.OBJECTIVE To compare the therapeutic effect of transcutaneous electrical acupoint stimulation (TEAS) combined with conventional western medicine and western medicine alone on early enteral nutrition tolerance in patients with sepsis. METHODS Forty-nine patients with sepsis were randomly divided into an observation group (24 cases) and a control group (25 cases). The control group was treated with conventional western medicine, including limited fluid resuscitation, anti-infection and maintenance of hemodynamics, and enteral nutrition was provided when the hemodynamics became stable and the dosage of vasoactive drugs was reduced. The observation group, on the basis of the treatment of the control group, was treated with TEAS at Tianshu (ST 25), Shangjuxu (ST 37), Jiexi (ST 41), Diji (SP 8), Zusanli (ST 36), Zhongwan (CV 12) and Daheng (SP 15) when the enteral nutrition was provided (dilatational wave, 2 Hz/10 Hz, twice a day, 30 min each time), and the TEAS stopped when enteral nutrition was normal. CONCLUSION Conducting research using EHRs has provided important learning about the potential of this type of data and the promise they hold for future research. IMPLICATIONS FOR PRACTICE Organisations willing for existing data to be used in research might consider embedding pathways for collecting data that are easy for potential researchers to navigate.

Besides, Xingnao Kaiqiao acupuncture was applied at Neiguan (PC 6), Shuigou (GV 26), Sanyinjiao (SP 6), Jiquan (HT 1), Chize (LU 5) and Weizhong (BL 40) in the control group, the needles were sustained for 30 min. Cohorts were grouped by time interval between ultrasound and delivery 0-7 days versus 8-14 days. The detection rate (DR) and false positive rate (FPR) for detection of birthweight (BW) ≥ 4500 g were compared between groups with subgroup analysis for diabetic women. Area under the receiver operator curve (AUC) was calculated to analyze all possible SEFW cutoffs within our cohort. RESULTS A total of 330 patients met inclusion criteria with 250 (75.8 %) having SEFW within 7 days and 80 (24.2 %) with SEFW 8-14 days prior to delivery. The rate of macrosomia was 15.1 % (N = 51). The DR for macrosomia was significantly higher when SEFW was performed within 7 days of delivery compared to 8-14 days among non-diabetic (73.0 % vs 7.1 %; p less then 0.001) and diabetic women (76.5 % vs 16.7 %; p = 0.02). There was no significant change in FPR in either group. The AUC for detection of macrosomia was significantly higher when SEFW was performed within 7 days versus 8-14 days (0.89 vs 0.63; p less then 0.01). This was associated with reduced cardiac glucose concentrations and decreased calcium/calmodulin-dependent protein kinase (CaMKII) activation with subsequent less phosphorylation of the ryanodine receptor (RyR). The PDDs values of the gafchromic films in front of the disks were 107%, 105%, 104%, and 94% for the Al/Pb, Al/Steel, Al/Cu, and PMMA/Cu/PMMA disks respectively. CONCLUSIONS The dose values above and under the shielding disks were acceptable for the four studied shielding types. Demonstrated it is possible to use any of them clinically, while the best shielding disk was the Al/Pb since it has minimum thickness and a small backscatter enhancement. Further flow cytometry analysis indicated that hyalinocytes were the main hemocytes subpopulation responsible for ingesting foreign fluorescent beads, and this ingestion process mainly depends on the endocytic way of macropinocytosis.   • We found indirect effects of the constructs in the extended TPB model on consumption to be different across food items. Practical implications to improve consumption of different food items were proposed accordingly.For digital impression-making of two-piece oral implants, scan bodies are used to transfer the exact intraoral implant position to the dental laboratory. In this in vitro investigation, the accuracy of digitizing a one-piece ceramic oral implant without a scan body (OC) was compared to that of a standard two-piece titanium implant with a scan body (TT) and a preparation of a natural single tooth (ST). Furthermore, incomplete scans of OC simulating clinical compromising situations (OC1-4) were redesigned using a virtual reconstruction tool (RT) and superimposed to OC. OC and TT oral implants and one ST were inserted into a mandible typodont model and digitized (N = 13) using two different intraoral scanners. The resulting virtual datasets were superimposed onto a three-dimensional (3D) laser scanner-based reference. Test and reference groups were aligned using an inspection software according to a best-fit algorithm, and circumferential as well as marginal discrepancies were measured. For the statistical evaluation, multivariate analyses of variance with post-hoc Tukey tests and students t-tests to compare both scanners were performed. A total of 182 datasets were analyzed. For circumferential deviations, no significant differences were found between ST, TT, and OC (p > 0.964), but increased deviations for OC1-4 (p 0.979). Except for marginal deviation of OC (p less then 0.001), the outcome was not affected by the scanner. Within the limitations of this study, digitization of OC is as accurate as that of TT, but less than that of ST. In the case of known geometries, post-processing of compromised scans with a virtual reconstruction results in accurate data.The aim of this paper is to analyze the primary stress factors female professors at online universities are exposed to. The prevalence of musculoskeletal discomfort in any part of the body in the past year among the respondents was 93.0%, with the highest prevalence of 66.3% and 51.8% in the hands/wrists (right and left), followed by the prevalence of 50.6% and 45.8% in the shoulders (right and left) and the lower **** (48.2%), respectively. The results also revealed that during the 3 dough processing operations, the workers' wrist movements in specific operations were close to the recommended limits suggested in previous studies, especially the ulnar deviation and palm flexion of the right wrist during dough kneading and the radial deviation of the left wrist during dough rolling and rounding.   • Rheumatoid arthritis (RA) is an autoimmune disease. Wasp venom (WV), which is considered as a traditional folk medicine in Jingpo nationality in Yunnan, China, relieves rheumatoid arthritis. The current study aimed to investigate the effect of wasp venom ameliorating rheumatoid arthritis symptoms in experimental rats. We established a model of type II collagen- (CII-) induced arthritis (CIA) in SD rats and examined the inhibition of inflammation and autoimmune response. The antiarthritic effects of WV were evaluated through the paw swelling, and histopathological score and histopathology changes of the affected paw were assessed. The anti-inflammation effects were assayed by the level of IL-6, TNF-α, IL-1β, and the number of inflammatory cells in peripheral blood. The alteration of the T cell subset ratio in the spleen of rats was detected by flow cytometry, and at the same time, the viscera index and immune serum globulin levels were evaluated. The results suggested that various doses of WV (0.125, 0.25, and 0.5 mg/kg) significantly alleviated paw swelling and arthritis score in CIA rats with the untreated control (P less then 0.05). WV (0.25 and 0.5 mg/kg) relieved synovial tissue lesions of ankle joints and histopathology scores of synoviocyte hyperplasia and inflammatory cell infiltration with vehicle group (P less then 0.05). Regarding immunological regulation, 0.5 mg/kg WV lowered the immune serum globulin levels (P less then 0.05), and we further found that WV (0.5 mg/kg) suppressed the immune response of Th cells, while enhancing the functions of Tc cells and Treg cells in spleen cells markedly (P less then 0.05). The immunosuppressive action of WV displayed was analogous to its inhibitory effect on IL-1β, TNF-α, IL-8, IL-6, COX-2, and PGE2 levels in rat serum. In conclusion, these findings demonstrated that WV exhibited antiarthritic activity, which might be associated with their inhibitory effects on immunoregulation and anti-inflammatory action. Berberine induced apoptosis of MGC803 and increased the apoptotic rate with higher doses. Berberine induced the accumulation of fatty acid of MGC803 and suppressed the protein expression of FABPs and PPARα. The FABP inhibitor BMS309403 recapitulated the effects of berberine on MGC803 cells. Methods Randomized controlled trials comparing EA with other treatments that met the eligibility criteria published in databases were included. The differences were observed and quantified through the risk ratio (RR) for dichotomous outcomes and the standardized mean difference (SMD) for continuous outcomes. Then, their 95% confidence intervals (CI) were recorded. Results Twenty-three studies involving 1985 participants were included. META-analysis results showed that EA was better than manual acupuncture for PFP (RR 1.16, 95% CI 1.11 to 1.22, for responding rate; SMD 2.26, 95% CI 0.15 to 4.37, for facial nerve function) and current promoted recovery (RR 1.21, 95% CI 1.15 to 1.27, for responding rate; SMD 2.87, 95% CI 1.16 to 4.58, for facial nerve function). When combined with other treatments, EA improved their effectiveness (RR 1.19, 95% CI 1.12 to 1.28, responding rate; SMD 1.85, 95% CI 0.67 to 3.03, facial nerve function). Conclusion Patients with PFP received EA (used separately or combined with other treatments) resulting in a better prognosis. However, the quality of evidence was very low-to-moderate. Considering the poor quality of evidence, we are not very confident in the results. We look forward to more research and update results in the future and improve the evidence quality. The aim of this study was to investigate the effect of forced and voluntary training before and after induction of experimental autoimmune encephalomyelitis (EAE) on accumulation of neural cell adhesion molecule (NCAM) and polysialic acid (PSA) in neuromuscular junction denervation in plantaris and soleus muscles in C57BL/6 female ****. A total of 40 female C57BL/6 ****, 10-week-old, were randomly divided into four groups, including induced control groups without EAE induction, induced EAE without training, and forced and voluntary training groups. Myelin oligodendrocyte glycoprotein peptide 35-55 (300 μg in saline; MOG 35-55; KJ Ross-Petersen ApS, Denmark) was injected subcutaneously at the base of the tail of each mouse. Clinical assessment of EAE was performed daily using a 15-point scoring system following immunization. Training groups performed ctable both in pre and post-soleus and plantaris. However, voluntary activity resulted in more expression levels of NCAM and PSA than that of compulsory. In conclusion, since it has been suggested that depletion of NCAM is one of the factors associated with or possibly responsible for MS progression, these findings show exercise MS progression may be reduced by increasing expression of exercise-related adhesion molecule such as NCAM and PSA (a glycan modification of the NCAM).
