  • Oxaliplatin (OXA) resistance is the major obstacle to the anticancer effects of chemotherapy in colorectal cancer (CRC) patients. MicroRNAs (miRNAs) play an important role in the chemoresistance of various tumors. Our objective is to clarify the underlying mechanism of miRNAs in chemoresistance and provide a potential strategy to improve the response of CRC patients to chemotherapeutics. Methods MiRNA microarray and Real-time PCR were performed to compare changes in miRNA expression between oxaliplatin-resistant and the parental cells. ONIVYDE is a novel liposome formulation, entrapping CPT-11 in the aqueous core of vesicles using a modified gradient loading method. Due to toxicity concerns, it is essential to explore a rapid and reliable method to effectively isolate and quantify the non-liposomal, namely, free CPT-11and total CPT-11 in plasma. This study focuses on separation of non-liposomal CPT-11, evaluation of the pharmacokinetics of free CPT-11 and total CPT-11 and bio-distribution after intravenous administration of CPT-11 liposome. Free CPT-11 in plasma was separated by solid-phase extraction (SPE). Pharmacokinetic studies of β-lactamase in rats were performed to study the contribution of rOat1/3 to the inhibition of tazobactam on β-lactamase. The AUC (from 2.93 ± 0.58 to 6.52 ± 1.44 mg·min/ml) and the plasma clearance (CLP ) (from 2.41 ± 1.20 to 0.961 ± 0.212 ml/min/kg) of tazobactam were both altered after the intravenous coadministration of piperacillin and tazobactam in the bacteremia rats. The renal clearance (CLR ) of tazobactam decreased from 1.30 ± 0.50 to 0.361 ± 0.043 ml/min/kg. In summary, there was a beneficial interaction between piperacillin and tazobactam mediated by rOat1 and rOat3. Piperacillin enhances the inhibitory effect of tazobactam on β-lactamase through the inhibition of rOat1 and rOat3 in rats. Discriminatory dissolution media were used to obtain reliable dissolution results. Liposomal delivery is a well-established approach to increase the therapeutic index of hydrophobic drugs. In this study, a PEG-modified liposome was developed for efficiently encapsulating CBZ, thus enhancing its specific tumor inhibition effect and reducing the systemic toxicity. It was found that the loading efficiency of CBZ into the liposome could be improved with the increase of lipophilic materials, as it could be over 80% under the weight ratio of 201 (total lipid CBZ). The diameter of CBZ loaded liposome (CBZ@Lipo) was ∼100 nm. And the liposome suspending in aqueous medium was stable at 4 °C for at least one month, according to the change of its size distribution. The killing ability of CBZ@Lipo to cancer cells was significantly lower comparing to that of CBZ solution, which could be attributed to the slow release of CBZ from the liposomes. However, CBZ@Lipo could induce an obvious apoptosis of the cancer cells at low concentration.

  • STUDY OBJECTIVES Disrupted nighttime sleep (DNS) is a core narcolepsy symptom of unconsolidated sleep resulting from hypocretin neuron loss. In this study, we define a DNS objective measure and evaluate its diagnostic utility for pediatric narcolepsy type 1 (NT1). METHODS This was a retrospective, multi-site, cross-sectional study of polysomnograms (PSGs) in 316 patients, ages 6-18 years [n=150 NT1, n=22 narcolepsy type 2, n=27 idiopathic hypersomnia, and n=117 subjectively sleepy subjects]. We assessed sleep continuity PSG measures for a) their associations with subjective and objective daytime sleepiness, daytime sleep onset REM periods (SOREMPs), self-reported disrupted nocturnal sleep and CSF hypocretin levels, and b) their predictive value for NT1 diagnosis. We then combined the best performing DNS measure with nocturnal SOREMP (nSOREMP) to assess the added value to the logistic regression model and the predictive accuracy for NT1 compared to nSOREMP alone. RESULTS The Wake/N1 Index [the number of transitions from any sleep stage to wake or NREM stage 1 normalized by total sleep time] was associated with objective daytime sleepiness, daytime SOREMPs, self-reported disrupted sleep, and CSF hypocretin levels (p's less then 0.003) and held highest AUC for NT1 diagnosis. When combined with nSOREMP, the DNS index had greater accuracy for diagnosing NT1 [AUC=0.91(0.02)] than nSOREMP alone [AUC=0.84, (0.02), LR test p less then 0.0001). CONCLUSION The Wake/N1 Index is an objective DNS measure that can quantify DNS severity in pediatric NT1. The Wake/N1 Index in combination with or without nSOREMP is a useful sleep biomarker that improves recognition of pediatric NT1 using only the nocturnal PSG. © Sleep Research Society 2020. Thus, in order to illuminate mechanisms underlying these physiological processes, it is crucial to understand how cells deform and respond to external mechanical stimuli. During recent decades, the mechanical properties of cells have been studied extensively using diverse measurement techniques. A number of experimental studies have shown that cells are far from linear elastic materials. Cells exhibit a wide variety of nonlinear elastic and inelastic properties. Such complicated properties of cells are known to emerge from unique mechanical characteristics of cellular components. In this review, we introduce major cellular components that largely govern cell mechanical properties and provide brief explanations of several experimental techniques used for rheological measurements of cell mechanics. Then, we discuss the representative nonlinear elastic and inelastic properties of cells. Finally, continuum and discrete computational models of cell mechanics, which model both nonlinear elastic and inelastic properties of cells, will be described. However, these drugs also induce immune-related adverse events, among which interstitial lung disease (ILD) is potentially fatal. The underlying mechanism of ILD induction by ICIs is largely unknown. With the use of flow cytometry, we determined the expression levels of the immune-checkpoint proteins PD-1, TIM-3, TIGIT, LAG-3, and PD-L1 in T cells of bronchoalveolar lavage fluid (BALF) from patients with ICI-related ILD and compared them with those for patients with sarcoidosis or with ILD related to connective tissue disease or cytotoxic drug use. The proportions of CD8+ T cells positive for both PD-1 and TIM-3 or for TIGIT in BALF were significantly higher for ICI-related ILD patients than for those with other types of ILD. A prominent increase in the proportion of PD-1+PD-L1+ cells among CD8+ T cells was also apparent in BALF of a patient with a fatal case of ICI-related ILD, and the proportion of such cells was positively correlated with the grade of ICI-related ILD. Our data reveal the immune-checkpoint profiles of T cells in ICI-related ILD and may provide mechanistic insight into the development of this adverse event. © The Japanese Society for Immunology. 2020. While many recommendations exist to reduce the risk of VTE, the actual VTE prophylaxis practices of aesthetic plastic surgeons remain unknown. OBJECTIVES The primary aim of this study was to elucidate plastic surgeons' experiences with VTE, preferred VTE prophylaxis practices and areas in which VTE prevention may be improved. METHODS Members of The American Society for Aesthetic Plastic Surgery (ASAPS) were queried via a 55-question electronic survey regarding their experience with VTE as well as their VTE prophylaxis practices. Anonymous responses were collected and analyzed by the Mayo Clinic Survey Research Center. RESULTS The survey was sent to 1729 ASAPS members, of whom 286 responded. Fifty percent, 38%, and 6% of respondents reported having had a patient develop a DVT, PE, or death secondary to VTE. Procedures performed on the **** or trunk were associated with the highest rate of VTE.

While many recommendations exist to reduce the risk of VTE, the actual VTE prophylaxis practices of aesthetic plastic surgeons remain unknown. OBJECTIVES The primary aim of this study was to elucidate plastic surgeons' experiences with VTE, preferred VTE prophylaxis practices and areas in which VTE prevention may be improved. METHODS Members of The American Society for Aesthetic Plastic Surgery (ASAPS) were queried via a 55-question electronic survey regarding their experience with VTE as well as their VTE prophylaxis practices. Anonymous responses were collected and analyzed by the Mayo Clinic Survey Research Center. RESULTS The survey was sent to 1729 ASAPS members, of whom 286 responded. Fifty percent, 38%, and 6% of respondents reported having had a patient develop a DVT, PE, or death secondary to VTE. Procedures performed on the back or trunk were associated with the highest rate of VTE.

  • Pancreatic adenocarcinoma is one of the most lethal cancers in both men and women, with a median five-year survival of around 5%. Therefore, pancreatic adenocarcinoma represents an unmet medical need. Neoplastic diseases, such as pancreatic adenocarcinoma, often are associated with microbiome dysbiosis, termed oncobiosis. In pancreatic adenocarcinoma, the oral, duodenal, ductal, and fecal microbiome become dysbiotic. Furthermore, the pancreas frequently becomes colonized (by Helicobacter pylori and Malassezia, among others). The oncobiomes from long- and short-term survivors of pancreatic adenocarcinoma are different and transplantation of the microbiome from long-term survivors into animal models of pancreatic adenocarcinoma prolongs survival. The oncobiome in pancreatic adenocarcinoma modulates the inflammatory processes that drive carcinogenesis. In this review, we point out that bacterial metabolites (short chain fatty acids, secondary bile acids, polyamines, indole-derivatives, etc.) also have a role in the microbiome-driven pathogenesis of pancreatic adenocarcinoma. Finally, we show that bacterial metabolism and the bacterial metabolome is largely dysregulated in pancreatic adenocarcinoma. The pathogenic role of additional metabolites and metabolic pathways will be identified in the near future, widening the scope of this therapeutically and diagnostically exploitable pathogenic pathway in pancreatic adenocarcinoma.In recent years, Zirconia-reinforced Lithium Silicate ceramic (ZLS), combining lithium-silicate and zirconia features, has shown to have excellent mechanical and aesthetic characteristics. Thus, the aim of this study was to compare the fracture strength of ZLS single crowns cemented with two different cementation techniques. Twenty crowns were realised and cemented on teeth replicas achieved from an extracted premolar human tooth. The samples were divided into two groups of 10 specimens each, Glass-ionomeric cement (GIC) group and Self-Adhesive Resin Cement (ARC) group. The mechanical test was performed using a universal testing machine. The specimens were then evaluated with a scanning electron microscope (SEM) to identify for all crowns and related abutments the pattern of fracture after the breaking point. The data obtained were statistically analysed. The mean fracture toughness values and standard deviations (±SD) were 2227 ± 382 N and 3712 ± 319 N respectively for GIC and ARC groups. In fact, t-test showed a statistically significant difference between the two groups (p less then 0.001). Moreover, the SEM results demonstrated portions of abutments still attached to the crown fragments in the ARC group, whilst these were not present in the GIC group.   • Poyang Lake, the largest freshwater lake of China, provides critical ecological functions for water circulation and biodiversity conservation as a dynamic wetland system. However, recent climate change and human activities exerted strong pressures on this ecosystem. In this paper, we applied object-based image analysis (OBIA) and Radom Forests (RF) classifier to ten Landsat images to examine the land cover composition and its change during 1987-2017 low water season at Poyang Lake. NDVI time series (2000-2017) derived from MODIS imagery was used to document the changes of vegetation growth status. To investigate the potential driving mechanism of the inundation patterns, we differentiated the spatial-temporal changes of vegetation coverage and NDVI accumulation on eight elevation bands. Major result indicates that the vegetation area increased by 15.5% of the lake area during 1987-2017. A **** faster-increasing rate (58.0 km2 year-1) can be observed during 2001-2009 as compared to that of the overall study period (18.4 km2 year-1). Analysis of NDVI accumulation showed that 42.1% of the lake's area displayed a significant increasing trend during 2000-2017. Spatially, the increase of vegetation area and NDVI accumulation mainly took place in the 11-12 m elevation band in the lower lake center. Early dry season and prolonged exposure period after the operation of Three Gorges Dam (TGD) was the major reason for the spatio-temporal evolution of the wetland vegetation in Poyang Lake. The Lake's water level started to fall below 12 m before 9th November might cause a boost of vegetation growth in the low lake center, and in turn, triggering xerophilization for the vegetation in the highlands and a shift in foraging patterns of waterbirds due to phenology variations. The findings of this study provide a clear reference for sustaining the inter-annual stability of the ecosystem by controlling the depth of water in the lake. V.Optimizing long-term best management practices (BMPs) is of vital importance for water quality management, especially for nonpoint source (NPS) pollution. The relative incident irradiance distribution across the exposed area was examined by spectroradiometry and nitrate-nitrite uniformity assay. The two methods showed a good correlation and high distribution factors (>0.89 and >0.94 for 96- and 24-well plates, respectively). In addition, the potential of the new system for determining disinfection efficacy of E. coli and MS2 coliphage by UV-LEDs emitting at central wavelengths of 265 nm and 285 nm was demonstrated. Both the colorimetric and fluorescent titration studies are performed with different metal ions. The results have suggested that the probe L is very selective and sensitive towards Zn2+ ions with significant changes in color. The pendant armed macrocyclic "tet a" probe has exhibited 1.28× 105 M-1 binding constant and virtuous selectivity for Zn2+ ion than other common metal ions. The detection limit of the probe towards Zn2+ ion is 0.027 nM. The selective sensing of Zn2+ ion is efficiently reversible with EDTA, which is demonstrated for five cycles without losing sensitivity. The time-resolved single-photon counting (TCSPC) studies have determined the average lifetime value for the probe L and L+ Zn2+ ion of 1.29 and 2.96 ns, respectively. The theoretical DFT studies have well supported the experimental outcomes. The practical application of the probe in visualizing intracellular Zn2+ ion distribution in live Artemia salina has proved the low cytotoxicity and cell membrane permeability of probe, which makes it capable of sensing Zn2+ ion in HeLa cells. Fear conditioning and extinction serve as a dominant model for the development and maintenance of pathological anxiety, particularly for phasic fear to specific stimuli or situations. The validity of this model would be supported by differences in the physiological or subjective fear response between patients with fear-related disorders and healthy controls, whereas the model's validity would be questioned by a lack of such differences. We derived pupillometry, skin conductance response and startle electromyography as well as unconditioned stimulus expectancy in a two-day fear acquisition, immediate extinction and recall task and compared an unmedicated group of patients (n = 73) with phobias or panic disorder and a group of patients with posttraumatic stress disorder (PTSD, n = 21) to a group of carefully screened healthy controls (n = 35). Bayesian statistics showed no convincing evidence for a difference in physiological and subjective responses between the groups during fear acquisition, extinction learning or recall.

  • Schizophrenia participants showed higher accuracy when targets appeared in the same location as a directional cue (i.e., congruency) for nonsocial, but not social, cues. Contrary to expectations, reduced social attention was not significantly correlated with clinically rated social motivation deficits or decreased social functioning in the schizophrenia group. CONCLUSION These findings provide evidence for social attention deficits in schizophrenia, but without a clear mapping of its influence on social motivation. Data were analyzed using linear mixed models to evaluate change in psychological distress and minority stress across time points, and the influence of exposure to the marriage equality debate, sexual identity, and gender identity on psychological distress and minority stress. Reported symptoms of psychological distress and minority stress significantly decreased following the postal survey period. Greater exposure to the marriage equality campaign was associated with greater psychological distress and perceived stigma but not internalized stigma. Sexual and gender identity subgroups significantly differed on outcome variable means. This study documents the longitudinal effects on a minority group of a public vote and the enactment of legislation regarding their human rights. Each couple engaged in 2 video-recorded discussions so that each person took a turn as discloser and responder. Trained coders rated disclosures for specificity (i.e., whether or not the discloser used specific examples). Following each discussion, couples rated perceived partner responsiveness. Hypotheses were tested with multilevel modeling. Findings suggest that individuals perceived their partners' reactions as less responsive when they disclosed specific, partner-inclusive vulnerabilities and their partners had lower mindfulness. When partners had higher mindfulness, individuals perceived their partners as similarly responsive when disclosing partner-inclusive and -exclusive vulnerabilities, regardless of specificity.     Schizophrenia participants showed higher accuracy when targets appeared in the same location as a directional cue (i.e., congruency) for nonsocial, but not social, cues.   • Although van der Waals heterostructures composed of graphene and hexagonal boron nitride (h-BN) have attracted wide interest, it is still challenging to prepare them with high quality and controllability. Since contamination induced by transfer cannot be avoided in the case of growth on a metal catalyst, the non-catalytic growth of graphene and h-BN is highly desired. However, unlike graphene, few studies have reported the non-catalytic growth of h-BN, and the lack of controllability in terms of crystal orientation and nucleation density, and size of h-BN has hindered the practical applications of the heterostructures. In this work, we demonstrate the heteroepitaxial growth of aligned monolayer h-BN single-crystals on exfoliated graphite by chemical vapour deposition (CVD) without a metal catalyst. Triangular shaped domains were aligned with each other, which suggests the epitaxy between h-BN and the underlying graphite. Characterisation by Raman spectroscopy, Auger electron spectroscopy, and X-ray photoelectron spectroscopy also confirmed that the h-BN/graphite samples were of high quality. A growth kinetics study over different temperatures indicated an increase in the growth rate at high temperature. Control of nucleation density was realised by changing the hydrogen pressure during CVD or by the heating temperature in air before CVD. Under the optimised growth conditions, the edge length of h-BN single-crystals grew to ∼1 μm, which is the largest size to date for non-catalytic growth. These results will help to obtain structure-controlled, large-area, and impurity-free heterostructures based on h-BN and graphene.Despite extensive studies on the distinctive properties of water confined in a nanospace, the underlying mechanism and significance of the lengthscale involved in the confinement effects are still subjects of controversy. The dielectric constant and the refractive index in particular are key parameters in modeling and understanding nanoconfined water, yet experimental evidence is lacking. We report the measurement of the refractive indices of water in 10-100 nm spaces by exploiting the confinement of water and localized surface plasmons in a physicochemically well-defined nanocavity. The results revealed significantly low values and the scaling behavior of the out-of-plane refractive index n⊥ of confined water. They are attributed to the polarization suppression at the interfaces and the long-range correlation in electronic polarization facilitated by the strengthened H-bonding network. Using the refractive index as a sensing probe, we also observed anomalous stability of water structures over a wide range of temperature. Our measurement results provide essential feedback information for benchmarking water models and molecular interactions under nanoconfinement. This study also opens up a new methodology of using plasmon resonance in characterizing nanoconfined molecules and chemical reactions, and thus gives us fundamental insight into confinement effects.Amyotrophic lateral sclerosis is a progressive neurodegenerative disease characterized by a loss of function of motor neurons. The etiology of this disorder is still largely unknown. Gene-environment interaction arises as a possible key factor in the development of amyotrophic lateral sclerosis. We assessed the levels of trace metals, copper (Cu), iron (Fe), and manganese (Mn), of 9 amyotrophic lateral sclerosis cases and 40 controls by measuring their content in cerebrospinal fluid. The following trace element species were quantified using ion chromatography-inductively coupled plasma mass spectrometry univalent copper (Cu-I), divalent Cu (Cu-II), divalent Fe (Fe-II), trivalent Fe (Fe-III), divalent Mn (Mn-II), trivalent Mn (Mn-III), and also unidentified Mn species (Mn-unknown) were present in some samples. When computing the relative risks for amyotrophic lateral sclerosis through an unconditional logistic regression model, we observed a weak and imprecise positive association for iron (Fe III, adjusted odds ratio 1.48, 95% CI 0.46-4.76) and manganese (total-Mn and Mn-II; adjusted odds ratio 1.11, 95% CI 0.74-1.67, and 1.13, 95% CI 0.79-1.61, respectively). Increased risk for copper was found both in the crude analysis (odds ratio 1.14, 95% CI 0.99-1.31) and in multivariable analysis after adjusting for sex, age, and year of storage (1.09, 95% CI 0.90-1.32). Our results suggest a possible positive association between Cu and genetic amyotrophic lateral sclerosis, while they give little indication of involvement of Fe and Mn in disease, though some correlations found also for these elements deserve further investigation.Outfitted with abundant hydrogen bonding and coordination active groups, carboxymethyl chitosan (CMC) possesses a class of naturally occurring ligands for coordination with metal ions, establishing its excellent potential for various fields. Herein, by incorporating the naturally derived CMC into a thermally reconfigurable agarose (Agar) gel medium, a novel type of metal-biopolymer coordinated double network hydrogel (DN gel) was successfully fabricated via the strong coordination interactions. The interpenetrated CMC was confirmed to retain its excellent chelating abilities within the bulk gel matrix, which resulted in a series of metal-coordinated DN gels through spontaneous self-associative complexation with metal ions such as Cu2+, Zn2+, Ni2+, Co2+, Fe3+, and Cr3+. Moreover, these two types of physical cross-links are functionally independent and reversible, which enables the programming of the hydrogel with multi-functionality, including pH-regulated shape memory behavior, multi-staged self-healing properties and durable antibacterial activities.   • BACKGROUND Great progress has recently been made in immunotherapy of solid carcinomas. The advent of immunotherapy with checkpoint inhibitors has brought not only a completely different mechanism of action but also different types of adverse reactions. These adverse reactions are similar to those of autoimmune diseases and are referred to as immune-related adverse events. Most adverse reactions are due to lower grade toxic effects. Neurotoxicity is a less frequent. On the other hand, with the spread of immunotherapy, it can be assumed that adverse events with a lower incidence may occur in a relatively higher frequency due to borader usage of immunotherapeutics and thus, physicians outside of cancer centres may be presented with these events. The main therapeutic intervention is immunosuppressive therapy with corticoids. However, in some cases, or in case of delayed intervention, they may be fatal. PURPOSE Therefore, it is of great importance to increase the physicians and patients knowledge of the possible complications of this promising treatment modality. It is essential to apply proper management and help patients engage positively to prevent these situations occurring. The side effects of these treatments are similar in nature to those of autoimmune diseases. Recently, increasing evidence has indicated that some adverse effects of immunotherapy are associated with the beneficial effect of this treatment. PURPOSE The aim of this review was to summarize current knowledge of the association between the adverse effects of checkpoint inhibitors and the outcomes of patients treated with this therapy. CONCLUSION The association between the effect of immunotherapy and the occurrence of adverse reactions has been identified in a number of studies. It has been best documented in patients with malignant melanoma, non-small cell lung cancer, and renal cell carcinoma. Here, we show that based on current data multimodal distributions cannot be unambiguously excluded. Furthermore, the transferability of cluster results to the liquid can be neither justified nor refuted on the basis of currently available experimental ultraviolet photoelectron spectra.The efficacy of a sunscreen tends to be associated with its sun protection factor (SPF) value, a figure determined in a test that relies on the independence of the SPF value to both UV radiation dose and irradiance. We probe these assumptions when photoinduced product degradation is present, and we estimate that the theoretical limit for their validity is when the sunfilter active molecule relaxation time is faster than ∼10 ns. While such threshold relaxation time should be compatible with the expected ultrafast relaxation mechanisms of sunfilter molecules (picoseconds), recent research on sunfilter photodynamics has identified the existence of **** longer-lived molecular states. Here, we have developed novel SCS models, nonkeratinized (lipid only) and keratinized, to study the effect of keratin hydrolysates on the dehydration rate. We have confirmed that the lipid organizational structure of the SCS was similar to that of the human SC using X-ray scattering. We have revealed that keratin hydrolysates play a significant role in the dehydration rate, accelerating the rate for the short term.
    BACKGROUND Great progress has recently been made in immunotherapy of solid carcinomas. The advent of immunotherapy with checkpoint inhibitors has brought not only a completely different mechanism of action but also different types of adverse reactions. These adverse reactions are similar to those of autoimmune diseases and are referred to as immune-related adverse events. Most adverse reactions are due to lower grade toxic effects. Neurotoxicity is a less frequent. On the other hand, with the spread of immunotherapy, it can be assumed that adverse events with a lower incidence may occur in a relatively higher frequency due to borader usage of immunotherapeutics and thus, physicians outside of cancer centres may be presented with these events. The main therapeutic intervention is immunosuppressive therapy with corticoids. However, in some cases, or in case of delayed intervention, they may be fatal. PURPOSE Therefore, it is of great importance to increase the physicians and patients knowledge of the possible complications of this promising treatment modality. It is essential to apply proper management and help patients engage positively to prevent these situations occurring. Monoclonal antibodies against CTLA-4 (cytotoxic T-lymphocyte antigen-4) and PD-1 (programmed cell death-1) or PD-L1 (programmed cell death-1 ligand) are the most commonly used agents. The side effects of these treatments are similar in nature to those of autoimmune diseases. Recently, increasing evidence has indicated that some adverse effects of immunotherapy are associated with the beneficial effect of this treatment. PURPOSE The aim of this review was to summarize current knowledge of the association between the adverse effects of checkpoint inhibitors and the outcomes of patients treated with this therapy. CONCLUSION The association between the effect of immunotherapy and the occurrence of adverse reactions has been identified in a number of studies. It has been best documented in patients with malignant melanoma, non-small cell lung cancer, and renal cell carcinoma.   • Salt intake was significantly associated with annual change in eGFR of -0.11 (95% confidence interval (0.20 - - 0.02) mL/min/1.73m2 per gram of salt, whereas protein intake was not (-0.00001 (-0.01 - 0.01) mL/min/1.73m2 per gram of protein. The effect of salt intake on eGFR slope was significantly mediated by plasma copeptin (crude analysis 77% mediation, and, adjusted analysis 45% mediation), but not by systolic blood pressure. Thus, higher salt, but not higher protein intake may be detrimental in ADPKD. The substantial mediation by plasma copeptin suggests that this effect is primarily a consequence of a salt-induced rise in vasopressin.The popularity of laparoscopy to perform radical hysterectomy has massively increased over the two last decades. However, oncologic outcomes (overall and disease-free survivals) have been found to be better in patients managed by laparotomy compared to laparoscopy, challenging this surgical route. Compared to laparotomy, vaginal access reduces post-operative morbidity, while avoiding potential cancer spread associated with laparoscopy. We describe the procedure of Schauta-Amreich radical vaginal hysterectomy with bilateral salpingo-oophorectomy assisted laparoscopically and associated with a pelvic sentinel lymph node procedure in a 56-year-old woman with a FIGO IB-2 cervical epidermoid carcinoma. A sentinel lymph node (SLN) procedure was first performed by laparoscopy. Radical hysterectomy was prepared by laparoscopy by dividing the infundibulopelvic, round and broad ligaments. The procedure was continued by the vaginal route using the Schuchardt incision. We describe each step of the procedure and provide a video. Histology showed a margin-free resection in both the vagina and parametrium with negative SLNs. This description of the Schauta-Amreich radical vaginal hysterectomy technique with a video file could support the teaching of a procedure which may gain in popularity.The mitochondrial F-ATP synthase is responsible for coupling the transmembrane proton gradient, generated through the inner membrane by the electron transport chain, to the synthesis of ATP. This enzyme shares a basic architecture with the prokaryotic and chloroplast ones, since it is composed of a catalytic head (F1), located in the mitochondrial matrix, a membrane-bound part (FO), together with a central and a peripheral stalk. In this review we compare the structural and functional properties of F-ATP synthase in plant mitochondria with those of yeast and mammals. We also present the physiological impact of the alteration of F-ATP synthase in plants, with a special regard to its involvement in cytoplasmic male sterility. Furthermore, we show the involvement of this enzyme in plant stress responses. Finally, we discuss the role of F-ATP synthase in shaping the curvature of the mitochondrial inner membrane and in permeability transition pore formation.The incorporation of mitochondria in the eukaryotic cell is one of the most enigmatic events in the course of evolution. This important organelle was thought to be only the powerhouse of the cell, but was later learnt to perform many other indispensable functions in the cell. Two major contributions of mitochondria in spermatogenesis concern energy production and apoptosis. Apart from this, mitochondria also participate in a number of other processes affecting spermatogenesis and fertility. Mitochondria in sperm are arranged in the periphery of the tail microtubules to serve to energy demand for motility. Apart from this, the role of mitochondria in germ cell proliferation, mitotic regulation, and the elimination of germ cells by apoptosis are now well recognized. Eventually, mutations in the mitochondrial genome have been reported in male infertility, particularly in sluggish sperm (asthenozoospermia); however, heteroplasmy in the mtDNA and a complex interplay between the nucleus and mitochondria affect their penetrance. In this article, we have provided an update on the role of mitochondria in various events of spermatogenesis and male fertility and on the correlation of mitochondrial DNA mutations with male infertility.Recently, functional liposomes modified with versatile polymer and cell-based- biomimetic nanoparticles have emerged as the most advanced lipid-polymer hybrid nanocarriers (LPNs) for drug delivery. This review highlights the advances of these two LPNs in the delivery of active ingredients and fractions from Chinese medicine with promising therapeutic, chemopreventive, or chemosensitive effects. To understand their complete potency, the relationship between the nanoparticle characteristics and their in vitro and in vivo performance characteristics, such as improved physical stability, mucosal penetration, biological and stimuli-responsive feature, and transport across various biological barriers have been discussed. Polymer-modified liposomes and cell-based biomimetic nanoparticles are beneficial for improving absorption, modulating release, targeting and overcoming multidrug resistance, and reducing side effects. The associated challenges, current limitations, and opportunities in this field are also discussed.Objectives Achromobacter xylosoxidans is an emerging pathogen in cystic fibrosis (CF). Relatively little is known about its clinical impact and optimal management. In the present study, the in vitro bactericidal activities of meropenem, either alone or in combination with colistin, levofloxacin, or chloramphenicol, were assessed using A. xylosoxidans strains isolated from CF patients. The synergistic interactions of these combinations were also investigated. Methods Minimal inhibitory concentrations were determined by microbroth dilution. Bactericidal and synergistic effects of the tested antibiotic combinations were assessed by using the time-kill curve technique.   • The striatal dopamine transporter-specific binding ratios were not correlated with the QSM values of the substantia nigra but were inversely correlated with those of the striatum (putamen, r = -0.478, P = 0.009; caudate nucleus, r = -0.462, P = 0.011). The QSM values of the putamen were positively correlated with motor parkinsonism scores (Movement Disorder Society Unified Parkinson's Disease Rating Scale, r = 0.505, P = 0.003), and those of the caudate nucleus were negatively correlated with cognitive impairment scores (Montreal Cognitive Assessment, r = -0.525, P  less then  0.001). CONCLUSIONS This study showed that striatal iron accumulations were correlated with dopaminergic deficits and neurophysiological signs in patients with PD, highlighting the potential of QSM as an auxiliary biomarker for parkinsonism and cognitive dysfunction. © 2020 International Parkinson and Movement Disorder Society. © 2020 International Parkinson and Movement Disorder Society.OBJECTIVE This study aimed to evaluate whether a master and PhD thesis in Brazilian graduate programs in dentistry can be based on a systematic review (SR). METHODS In this cross-sectional study, we surveyed the coordinators of Brazilian graduate programs in dentistry using a self-administered questionnaire including questions about whether an SR meets requirements for approval of a master or PhD thesis, instructions regarding how to use an SR as the method for a master or PhD thesis, and reasons and barriers that SRs were not recognized as acceptable for a master or PhD thesis. The responses were analyzed descriptively, and tables and graphs were created to summarize the data. RESULTS Among 101 graduate programs invited to participate in the study, 38 (37.6%) returned the questionnaire. Over three-quarters of surveyed programs (n = 30, 78.1%) answered that SRs, in whole or in part, meet the criteria for a master or PhD thesis. Likewise, most programs answered that a master or PhD thesis could be exclusively based on an SR (n = 28, 73.7%). The same number of programs pointed out that one possible reason for not recognizing SRs as a master or PhD thesis is either the lack of SR expertise among committee members (n = 28, 73.7%) or the lack of adequate training of candidates in SR methodology (n = 26, 68.4%). CONCLUSIONS SRs are well accepted as a master or PhD thesis by Brazilian graduate programs in dentistry. DESIGN Single center cohort study. SETTING Renmin Hospital of Wuhan University, Wuhan, Hubei province, China. POPULATION OR SAMPLE We studied 13 COVID-19 infected pregnant women diagnosed between January 31 and March 9, 2020. https://www.selleckchem.com/products/r428.html METHODS We collected clinical data, vaginal secretions and stool specimens, breast milk from COVID-19 infected women during different stages of pregnancy and neonatal throat, anal swabs. MAIN OUTCOME(S) AND MEASURE(S) We assessed viral presence in different biosamples. RESULTS Of the 13 women with COVID-19, 5 were in their first trimester, 3 in their second trimester, and 5 in their third trimester. Of the 5 women during their third trimester who gave birth, all delivered live newborns. Among these 5 deliveries, the primary adverse perinatal outcomes included premature delivery (n = 2) and neonatal pneumonia (n = 2). One of 9 stool samples was positive; and all 13 vaginal secretion samples, and 5 throat swabs and 4 anal swabs collected from newborns were negative for the novel coronavirus. However, 1 of 3 samples of breast milk was positive by viral nucleic acid testing. CONCLUSIONS In this case series of 13 pregnant women with COVID-19, we observed negative viral test results in vaginal secretion specimens, suggesting that a vaginal delivery may be a safe delivery option. All rights reserved.OBJECTIVE Acetyl zingerone (AZ), a derivative of the phytochemical zingerone from Zingiber officinale (******), is a novel compound that is purported to have anti-aging properties. The objective of this clinical study was to assess the role of acetyl zingerone in its ability to improve the appearance of facial skin wrinkles, redness, pigmentation, and photoaging was assessed. METHODS Thirty-one healthy participants (age 44 ± 7 years) were randomized in blinded fashion to apply either 1% AZ or placebo, consisting of the vehicle base cream, to the full face twice daily for 8 weeks with a total of 3 visits. Signs of photoaging, including wrinkles, dyspigmentation, and redness were assessed with facial image analysis photography and software. RESULTS There was a significant decrease in average wrinkle severity (p=0.019; Mean=-25.7% change), total wrinkle volume (p=0.003; Mean=-30.1 % change), pigment intensity (p=0.021; Mean=-25.6% change), and redness intensity (p=0.035; Mean=-20.7% change) in the AZ group by 8 weeks compared to the placebo. No significant itching, burning, or stinging was noted by study participants. There was also no significant difference between both groups in the clinical assessment of scaling, erythema, hypopigmentation, or hyperpigmentation. CONCLUSIONS AND RELEVANCE Topical AZ improves photodamage and decreases the appearance of wrinkles, dyspigmentation, and redness intensity when compared to placebo (vehicle) formulation. METHODS In this cross-sectional study, we surveyed the coordinators of Brazilian graduate programs in dentistry using a self-administered questionnaire including questions about whether an SR meets requirements for approval of a master or PhD thesis, instructions regarding how to use an SR as the method for a master or PhD thesis, and reasons and barriers that SRs were not recognized as acceptable for a master or PhD thesis. The responses were analyzed descriptively, and tables and graphs were created to summarize the data. RESULTS Among 101 graduate programs invited to participate in the study, 38 (37.6%) returned the questionnaire. Over three-quarters of surveyed programs (n = 30, 78.1%) answered that SRs, in whole or in part, meet the criteria for a master or PhD thesis. Likewise, most programs answered that a master or PhD thesis could be exclusively based on an SR (n = 28, 73.7%). The same number of programs pointed out that one possible reason for not recognizing SRs as a master or PhD thesis is either the lack of SR expertise among committee members (n = 28, 73.7%) or the lack of adequate training of candidates in SR methodology (n = 26, 68.4%). CONCLUSIONS SRs are well accepted as a master or PhD thesis by Brazilian graduate programs in dentistry. Educational initiatives should be encouraged to promote understanding of SR methodology and increase acceptance among graduate programs worldwide. © 2020 Chinese Cochrane Center, West China Hospital of Sichuan University and John Wiley & Sons Australia, Ltd.OBJECTIVE To assess whether vaginal secretions and breast milk of COVID-19 patients contain SARS-CoV-2 virus. DESIGN Single center cohort study. SETTING Renmin Hospital of Wuhan University, Wuhan, Hubei province, China. POPULATION OR SAMPLE We studied 13 COVID-19 infected pregnant women diagnosed between January 31 and March 9, 2020. https://www.selleckchem.com/products/r428.html METHODS We collected clinical data, vaginal secretions and stool specimens, breast milk from COVID-19 infected women during different stages of pregnancy and neonatal throat, anal swabs. MAIN OUTCOME(S) AND MEASURE(S) We assessed viral presence in different biosamples. RESULTS Of the 13 women with COVID-19, 5 were in their first trimester, 3 in their second trimester, and 5 in their third trimester. Of the 5 women during their third trimester who gave birth, all delivered live newborns. Among these 5 deliveries, the primary adverse perinatal outcomes included premature delivery (n = 2) and neonatal pneumonia (n = 2). One of 9 stool samples was positive; and all 13 vaginal secretion samples, and 5 throat swabs and 4 anal swabs collected from newborns were negative for the novel coronavirus. However, 1 of 3 samples of breast milk was positive by viral nucleic acid testing. CONCLUSIONS In this case series of 13 pregnant women with COVID-19, we observed negative viral test results in vaginal secretion specimens, suggesting that a vaginal delivery may be a safe delivery option. All rights reserved.OBJECTIVE Acetyl zingerone (AZ), a derivative of the phytochemical zingerone from Zingiber officinale (ginger), is a novel compound that is purported to have anti-aging properties. The objective of this clinical study was to assess the role of acetyl zingerone in its ability to improve the appearance of facial skin wrinkles, redness, pigmentation, and photoaging was assessed. METHODS Thirty-one healthy participants (age 44 ± 7 years) were randomized in blinded fashion to apply either 1% AZ or placebo, consisting of the vehicle base cream, to the full face twice daily for 8 weeks with a total of 3 visits. Signs of photoaging, including wrinkles, dyspigmentation, and redness were assessed with facial image analysis photography and software. RESULTS There was a significant decrease in average wrinkle severity (p=0.019; Mean=-25.7% change), total wrinkle volume (p=0.003; Mean=-30.1 % change), pigment intensity (p=0.021; Mean=-25.6% change), and redness intensity (p=0.035; Mean=-20.7% change) in the AZ group by 8 weeks compared to the placebo. No significant itching, burning, or stinging was noted by study participants. There was also no significant difference between both groups in the clinical assessment of scaling, erythema, hypopigmentation, or hyperpigmentation. CONCLUSIONS AND RELEVANCE Topical AZ improves photodamage and decreases the appearance of wrinkles, dyspigmentation, and redness intensity when compared to placebo (vehicle) formulation. Acetyl zingerone is well tolerated with daily use. This article is protected by copyright.
