  • In patients with schizophrenia, clinical symptoms and cognitive impairment are its core features, both of which have a significant impact on the prognosis and functional outcome. Empathy, as an important social cognition, has been found to be associated with the clinical symptoms in schizophrenia, but the conclusions on this issue are inconsistent. Therefore, this study will continue to explore it through a large sample of inpatients with chronic schizophrenia in the Chinese Han population.

    We obtained the sociodemographic characteristics of 987 inpatients, measured their clinical symptoms using the Positive and Negative Syndrome Scale (PANSS), and assessed their self-reported empathy using the Interpersonal Reactivity Index (IRI). The factor score for negative symptoms (FSNS) of PANSS was additionally calculated.

    Correlation and linear regression analysis showed that patients' PANSS scores were widely correlated with their IRI scores. In particular, the negative symptoms of patients were significantly correlated to IRI total score (r=-0.131, p<.001) and subscales such as Perspective Taking (PT) (r=-0.233, p<.001). FSNS had close relationships with empathy as well. There are also many significant associations between other dimensions, such as general psychopathology and Perspective Taking (PT) or Fantasy (FS) (all p<.05).

    Our results indicated that clinical symptoms, especially negative symptoms, were closely related to their current empathy in patients with schizophrenia, suggesting that the severity of clinical symptoms may be a powerful factor in predicting social cognition such as empathy of schizophrenia.

    To evaluate whether the extent of COVID-19 pneumonia on CT scans using quantitative CT imaging obtained early in the illness can predict its future severity.

    We conducted a retrospective single-center study on confirmed COVID-19 patients between January 18, 2020 and March 5, 2020. A quantitative AI algorithm was used to evaluate each patient's CT scan to determine the proportion of the lungs with pneumonia (VR) and the rate of change (RAR) in VR from scan to scan. Patients were classified as being in the severe or non-severe group based on their final symptoms. Penalized B-splines regression modeling was used to examine the relationship between mean VR and days from onset of symptoms in the two groups, with 95% and 99% confidence intervals.

    Median VR max was 18.6% (IQR 9.1-32.7%) in 21 patients in the severe group, significantly higher (P<0.0001) than in the 53 patients in non-severe group (1.8% (IQR 0.4-5.7%)). RAR was increasing with a median RAR of 2.1% (IQR 0.4-5.5%) in severe and 0.4% (IQR 0.1-0.9%) in non-severe group, which was significantly different (P<0.0001). Penalized B-spline analyses showed positive relationships between VR and days from onset of symptom. The 95% confidence limits of the predicted means for the two groups diverged 5days after the onset of initial symptoms with a threshold of 11.9%.

    Education about patient safety has historically been limited in health professionals curricula and largely inaccessible to the general public. We developed a massive open online course (MOOC), The Science of Safety in Healthcare, to present the foundational patient safety principles to a broad global audience of health professionals, learners, and patients interested in patient safety.

    To describe the Science of Safety in Healthcare MOOC, its effects on patient safety competence, and the satisfaction of course participants.

    The five-week video-based course was delivered in 2013 and 2014, and was organized in five modules 1) overview of science of safety and safety culture, 2) enabling and contextual factors that influence patient safety and quality, 3) methods to improve safety and quality, 4) translating evidence intro practice and leading change, and 5) summary and opportunities for capacity building. Each module had three or four segments. Participants were introduced to key concepts, and tools and s4 (95% CI 0.61, 0.68), and Culture 0.72 (95% CI 0.68, 0.75). About 8% of participants in each cohort earned a certificate of completion. At 6-months post-course, the majority of the participants agreed or agreed strongly that the course content was useful (93%) and that the course positively influenced their clinical practice (69%) and communication (84%).

    The MOOC course allowed educators to reach a large, diverse audience. The course was well-received and participants reported a significant increase in patient safety competence. As with most MOOCs, rates of completion were low.

    Related teaching and learning strategies were deployed in the course using the Community of Inquiry framework. All students who completed the course were invited to complete an online survey that included a validated Online Student Engagement questionnaire (OSE). Pearson's correlations were used to determine the association between perceived learning satisfaction and learning engagement. A logistic regression model was used to explore the associations of gender, age, working experience and perceived learning satisfaction with higher learning engagement.

  • Nb2 exhibited the highest melting temperature (Tm); thus, Nb2 is a promising GFP nanobody candidate for use in applications requiring harsh testing conditions. We also compared the binding sites of available GFP nanobodies and showed that some of them can simultaneously bind with GFP and assemble into multifunctional complexes to manipulate GFP-tagged target proteins. Our results provide atomic-scale binding information for Nb2-sfGFP, which is important for the further development of GFP-nanobody based fusion protein manipulation techniques.Nano/microplastics (NPs/MPs), a tiny particle of plastic pollution, are known as one of the most important environmental threats to marine ecosystems. Wastewater treatment plants can act as entrance routes for NPs/MPs to the aquatic environment as they breakdown of larger fragments of the plastic component during the treatment process; therefore, it is necessary to remove NPs/MPs during the wastewater treatment process. In this study, understanding the effect of water shear force on the fragmentation of larger size MPs into smaller MPs and NPs and their removal by air flotation and nano-ferrofluid (i.e., magnetite and cobalt ferrite particle as a coagulant) and membrane processes were investigated as a proof-of-concept study. It is found that a two-blade mechanical impeller could fragment MPs from 75, 150 and 300 μm into mean size NPs/MPs of 0.74, 1.14 and 1.88 μm, respectively. Results showed that the maximum removal efficiency of polyethylene, polyvinyl chloride and polyester was 85, 82 and 69%, respectively, in the air flotation process. Increasing the dose of behentrimonium chloride surfactant from 2 to 10 mg/L improved the efficiency of the air flotation process for NPs/MPs removal. It is also found that the removal efficiency of NPs/MPs by the air flotation system depends on solution pH, size, and types of NPs/MPs. This study also found a less significant removal efficiency of NPs/MPs by both types of ferrofluid used in this study with an average removal of 43% for magnetite and 55% for cobalt ferrite. All three plastics tested had similar removal efficiency by the nano-ferrofluid particles, meaning that this removal technique does not rely on the plastic component type. Among all the process tested, both ultrafiltration and microfiltration membrane processes were highly effective, removing more than 90% of NPs/MPs fragment particles. Overall, this study has confirmed the effectiveness of using air flotation and the membrane process to remove NPs/MPs from wastewater.Semiconducting membranes integrated with nanomaterials have placed themselves in new emerging researches tremendously for seawater desalination, oil-water separation, disinfection, removal of inorganic as well as organic pollutants. Howbeit, only nanoparticles unified membranes show quite a lot lags in their performance, although some of these particles associated with the demerits of high cost. In contrast, graphitic carbon nitride incorporated membranes offered improved aforementioned properties corresponding to absolute essential qualities such as cost-effective, environmentally friendly, easy-to-operate, green manufacturing, anti-fouling, and low energy consumption. Moreover, their high mechanical strength, high stability against harsh environment and long-term utilization without flux reduction are strong plus. Even though there are some undeniable downsides of these membranes in real world applications as bulk synthesis, consistent dispersion of graphitic carbon nitride, low photocatalytic efficiency etc. Accordingly, in the present article, these frailties of the membranes having graphitic carbon nitride as a filler and their respective synthesis procedures and properties are discussed. A comprehensive analysis over the application of semiconducting graphitic carbon nitride incorporated membranes with and without special surface modification; and exploration of the future challenges and difficulties associated to these membranes are also reviewed. Consequently, the current article provides brief overview about graphitic carbon nitride integrated composite membranes as well as their applications, and it finished up with new thoughts of further improvements/modifications to overcome their shortcomings in actual environmental conditions.Efforts are being made to develop fast, cost-effective and sensitive sensor to detect water contamination by toxic heavy metal ions. The oxygenated functional groups decorated graphene quantum dots (GQDs) effectively enhances the aqueous solubility and considered as a more desirable and simple sensing material with high sensitivity. Here, photoluminescence (PL) property of GQDs has been employed to devise an optical nanosensor for the detection of toxic heavy metal ions in aqueous media. Hydrothermal method was employed to synthesize highly fluorescent and water soluble GQDs. The fluorescence intensity reduces with the increase in toxic heavy metal ions concentration. Annual quarters were defined as L-1 (November-January), L-2 (February-April), H-1 (April-July), and H-2 (July-October). The total nitrogen removal rate was stable at 0.08-0.11 kg-N/m3/d, even during temperature fluctuations. Removal efficiencies of biochemical oxygen demand and total nitrogen were consistently high at 95-99% and 69-81%, respectively. The anammox activity and abundance of anammox bacteria were highest in granule L-1 and lowest in granule H-2. The optimal temperature for anammox activity shifted from 35 °C in granules H-1 and H-2 to 30 °C in granules L-1 and L-2, while the latter maintained a moderate activity compared to the former at low temperature.

  • This study demonstrates that Beishan granite has a good reducing capacity, which is suitable for the immobilization of redox-sensitive radionuclides. However, potential oxidation of spent fuel by Fe3+ in the granite should also been taken into account. Organic amendments affect the behavior of radiocesium in soil-plant systems in a complex way; they can inhibit radiocesium fixation by clay minerals by blocking selective sorption sites, whereas K supplied to the soil solution by amendments can reduce Cs uptake by plant roots. Here, we investigated the influence of inorganic and organic amendments on the transferability of radiocesium from soil to grass seedling in a humus-rich Andosol with high exchangeable K content. Soil samples were spiked with a137Cs tracer, treated with N, N-P-K, compost (cattle manure using rice straw), or no amendment (control soil), and subjected to repeated two-week wetting and air-drying treatments for one year in an artificial climate chamber. Small-scale cultivations of orchard grass were performed four times during the experimental period to assess temporal changes of availability of 137Cs in the soils. The 137Cs transfer factor (TF), defined as the 137Cs concentration in the plant divided by that in the soil, decreased with time in the control soil. The soil treated with compost showed higher TFs than the control soil in each cultivation and a slower attenuation of 137Cs transferability. By comparing the extractability of 137Cs, NH4+, and K+ with the observed TFs, we show that K released from the compost was not effective in reducing root uptake of 137Cs, but enhanced 137Cs desorption from the soil under K-rich conditions. This result suggests that organic amendment is ineffective in reducing root uptake of radiocesium under high exchangeable K concentrations, and may instead enhance the long-term availability of radiocesium in soils. This article summarizes and discusses mass balance calculations of the activities of Fukushima-derived 137Cs released to the atmosphere and ocean prior to 2018 as well as the 137Cs inventories on land and in the ocean, biota, and sediment. We propose that the consensus value of the total amount of 137Cs released to the atmosphere was 15-21 PBq; atmospheric deposition of 137Cs on land was 3-6 PBq; atmospheric deposition of 137Cs on the North Pacific was 12-15 PBq; and direct discharge of 137Cs to the ocean was 3-6 PBq. We also evaluated the movement of 137Cs from one domain to another for several years after the accident. We calculated that the amount of 137Cs transported by rivers might be 40 TBq. The annual deposition of 137Cs due to resuspension at Okuma during the period 2014-2018 was 4-10 TBq year-1. The 137Cs discharged to the ocean was 0.73-1.0 TBq year-1 in 2016-2018. The integrated amount of FNPP1-derived 137Cs that entered the Sea of Japan from the Pacific Ocean from 2011 until 2017 was 270 ± 20 TBq, 6.4% of the estimated amount of FNPP1-derived 137Cs in Subtropical Mode Water in the North Pacific. The integrated amount of FNPP1-derived 137Cs that returned to the North Pacific Ocean through the Tsugaru Strait from the Sea of Japan was 110 ± 10 TBq. Decontamination efforts removed 134 TBq of 137Cs from surface soil prior to February 2019, an amount that corresponded to 4% of the137Cs deposited on land in Japan. Wine dating methods based on anthropogenic 14C and 137Cs, as well as on the cosmogenic 14C were studied with the aim to improve the accuracy and precision of the dating results. While the 14C dating method has proved to be useful for dating young and old wines, the 137Cs has been effective for dating of wines originating around the 137Cs bomb-peak observed in 1963. However, potential oxidation of spent fuel by Fe3+ in the granite should also been taken into account. Organic amendments affect the behavior of radiocesium in soil-plant systems in a complex way; they can inhibit radiocesium fixation by clay minerals by blocking selective sorption sites, whereas K supplied to the soil solution by amendments can reduce Cs uptake by plant roots. Here, we investigated the influence of inorganic and organic amendments on the transferability of radiocesium from soil to grass seedling in a humus-rich Andosol with high exchangeable K content. Soil samples were spiked with a137Cs tracer, treated with N, N-P-K, compost (cattle manure using rice straw), or no amendment (control soil), and subjected to repeated two-week wetting and air-drying treatments for one year in an artificial climate chamber. Small-scale cultivations of orchard grass were performed four times during the experimental period to assess temporal changes of availability of 137Cs in the soils. The 137Cs transfer factor (TF), defined as the 137Cs concentration in the plant divided by that in the soil, decreased with time in the control soil. The soil treated with compost showed higher TFs than the control soil in each cultivation and a slower attenuation of 137Cs transferability. By comparing the extractability of 137Cs, NH4+, and K+ with the observed TFs, we show that K released from the compost was not effective in reducing root uptake of 137Cs, but enhanced 137Cs desorption from the soil under K-rich conditions. This result suggests that organic amendment is ineffective in reducing root uptake of radiocesium under high exchangeable K concentrations, and may instead enhance the long-term availability of radiocesium in soils. This article summarizes and discusses mass balance calculations of the activities of Fukushima-derived 137Cs released to the atmosphere and ocean prior to 2018 as well as the 137Cs inventories on land and in the ocean, biota, and sediment. We propose that the consensus value of the total amount of 137Cs released to the atmosphere was 15-21 PBq; atmospheric deposition of 137Cs on land was 3-6 PBq; atmospheric deposition of 137Cs on the North Pacific was 12-15 PBq; and direct discharge of 137Cs to the ocean was 3-6 PBq. We also evaluated the movement of 137Cs from one domain to another for several years after the accident. We calculated that the amount of 137Cs transported by rivers might be 40 TBq. The annual deposition of 137Cs due to resuspension at Okuma during the period 2014-2018 was 4-10 TBq year-1. The 137Cs discharged to the ocean was 0.73-1.0 TBq year-1 in 2016-2018. The integrated amount of FNPP1-derived 137Cs that entered the Sea of Japan from the Pacific Ocean from 2011 until 2017 was 270 ± 20 TBq, 6.4% of the estimated amount of FNPP1-derived 137Cs in Subtropical Mode Water in the North Pacific. The integrated amount of FNPP1-derived 137Cs that returned to the North Pacific Ocean through the Tsugaru Strait from the Sea of Japan was 110 ± 10 TBq. Decontamination efforts removed 134 TBq of 137Cs from surface soil prior to February 2019, an amount that corresponded to 4% of the137Cs deposited on land in Japan.   • To evaluate risk factors and describe the association between maternal pre-pregnancy body-mass-index (BMI) and neonatal brachial plexus palsy (BPP) in vaginal deliveries with and without shoulder dystocia.

    This is a retrospective cohort study of singleton, non-anomalous, term vaginal deliveries in California (2007-2011). Deliveries were classified as with or without shoulder dystocia. Our primary outcome was BPP and the independent variable of interest was maternal pre-pregnancy BMI, which was categorized as underweight (<18.5 kg/m

This work examined antiglycation activity compounds isolated from the antidiabetic extract of T. alnifolia stem bark via in vitro and computational [molecular dynamics (MD)] approach. Phytochemical investigation of ethyl acetate fraction and the application of spectroscopic methods led to the isolation and elucidation of 3 compounds quercetin (1), kaempferol (2), and gallic acid (3). Compounds 1, 2 and 3 were then screened for antioxidant and antiglycation activities. Results show that the ethanol extract of T. alnifolia demonstrated good antioxidant activity compared to the standard gallic acid. https://www.selleckchem.com/products/myci361.html There was a significant reduction in fasting blood glucose level progressively in diabetic rats, for 21 days compared to diabetic control.     Until autumn 2018 the GPs in Bergen Municipality did not attend emergency patients outside the emergency primary care centre. The ambulance staff handled emergencies on their own or were assisted by an anaesthesiologist from the helicopter emergency medical service (HEMS). The aim of this study was to investigate procedures performed by the HEMS anaesthesiologist and to assess the level of skills needed to perform these procedures.

    This study was a retrospective assessment of data from the period 2011 to 2013 on all emergency missions in which patients were dealt with by HEMS, using a rapid-response car in Bergen Municipality. All emergency missions were sorted into three categories No intervention, Basic or Advanced intervention. This list was made by a research group with anaesthesiologists working for Bergen HEMS and GPs with OOH experience. The list is based on curriculum found in acute medicine courses.

    HEMS responded to 716 (2.3%) out of a total of 31,696 emergencies in Bergen Municipality during call GP could have adequately treated many of the patients in this study in terms of practical skills.Objective The current study summarizes the results of a 2020 survey that solicited information regarding backgrounds, beliefs, practices, and incomes of clinical neuropsychologists who practice in Canada. Methods Clinical neuropsychologists who practice in Canada were invited to participate in an online survey that was available from 1/17/20 to 4/02/20. Available survey findings were obtained from 111 respondents, which reflects a response rate of 51.3% of the 216 doctoral-level Canadian neuropsychologists identified in at least one major North American or international professional organization membership list (AACN, INS, NAN, or SCN). Results Most of the current respondents were White/Caucasian women who identified as adult providers and worked full-time in urban institutional settings. Four Canadian provinces (Alberta, British Columbia, Ontario, Quebec) accounted for more than 91% of the current respondent sample. Incomes and career satisfactions were largely encouraging, though some important variations were noted by province, work setting, and professional identity.

Rubingh's regular solution theory was used to determine the interaction behavior of the two components (Cremophor RH 40 and Phospholipon 80H) and of the mixed micelles and synergistic behavior was confirmed. The mixed miceller system was formulated using solvent evaporation method and a 32 factorial design was used for the optimization of selected independent variables. Miceller systems were further characterized in terms of morphology, particle size, zeta potential, percent entrapment efficiency, and drug loading. Fourier transform infrared and differential scanning calorimetry measurements confirmed the entrapment of EFZ in the micelles. The optimized formulation presented desirable qualities viz., nanometric size (17.27 ± 0.079), high entrapment efficiency, and good colloidal stability. The prepared optimized micelles can be potential carriers for EFZ in ARV therapies.Virtual reality perspective-taking (VRPT) experiences effectively increase both empathy and prosocial behaviors toward related social targets (e.g., cutting down a tree in virtual reality increases concern for the environment). This project tests the prediction that empathy is analogous to a muscle that increases with practice and can transfer to unrelated contexts instead of being a mental state that increases only for a specific context or target. This study examines the extent to which VRPT experiences can train empathic skills that are applied to unrelated social targets and contexts. Two thirds of the participants engaged in VRPT experiences either showing what it is like to become homeless or how ocean acidification affects the marine environment. A third of the participants were in the control condition and did not complete a VRPT task. Results replicate previous findings showing that VRPT tasks increase related context empathy and prosocial behaviors; however, the results on VRPTs effect on empathy and prosocial behaviors for unrelated contexts were mixed. The VRPT ocean acidification task was more effective at inducing empathy for the homeless, an unrelated social target, than the control condition, but the empathy-transfer effect did not occur from the homeless context to the ocean context. Replicating previous work, participants who experienced what it is like to become homeless signed a petition supporting the homeless at significantly higher rates than participants in the control condition. The mixed miceller system was formulated using solvent evaporation method and a 32 factorial design was used for the optimization of selected independent variables. Miceller systems were further characterized in terms of morphology, particle size, zeta potential, percent entrapment efficiency, and drug loading. Fourier transform infrared and differential scanning calorimetry measurements confirmed the entrapment of EFZ in the micelles. The optimized formulation presented desirable qualities viz., nanometric size (17.27 ± 0.079), high entrapment efficiency, and good colloidal stability. This project tests the prediction that empathy is analogous to a muscle that increases with practice and can transfer to unrelated contexts instead of being a mental state that increases only for a specific context or target. This study examines the extent to which VRPT experiences can train empathic skills that are applied to unrelated social targets and contexts. Two thirds of the participants engaged in VRPT experiences either showing what it is like to become homeless or how ocean acidification affects the marine environment. A third of the participants were in the control condition and did not complete a VRPT task. Results replicate previous findings showing that VRPT tasks increase related context empathy and prosocial behaviors; however, the results on VRPTs effect on empathy and prosocial behaviors for unrelated contexts were mixed. The VRPT ocean acidification task was more effective at inducing empathy for the homeless, an unrelated social target, than the control condition, but the empathy-transfer effect did not occur from the homeless context to the ocean context. Replicating previous work, participants who experienced what it is like to become homeless signed a petition supporting the homeless at significantly higher rates than participants in the control condition.   • Memantine, a low- to moderate-affinity uncompetitive N-methyl-D-aspartate receptor antagonist, has been shown to improve cognitive functions in animal models of Alzheimer's disease (AD).     Biodegradation of antibiotics is a promising method for the large-scale removal of antibiotic residues in the environment. However, the enzyme that is involved in the biodegradation process is the key information to be revealed.

    In this study, the beta-lactamase from Ochrobactrum tritici that mediates the biodegradation of penicillin V was identified and characterized. When searching the proteins of Ochrobactrum tritici, the β-lactamase (OtLac) was identified. OtLac consists of 347 amino acids, and predicted isoelectric point is 7.0. It is a class C β-lactamase according to BLAST analysis. The coding gene of OtLac was amplified from the genomic DNA of Ochrobactrum tritici. The OtLac was overexpressed in E. coli BL21 (DE3) and purified with Ni
    column affinity chromatography. The biodegradation ability of penicillin V by OtLac was identified in an in vitro study and analyzed by HPLC. The optimal temperature for OtLac is 32℃ and the optimal pH is 7.0. Steady-state kinetics showed that OtLac was highly active against penicillin V with a Km value of 17.86μM and a kcat value of 25.28 s

This research focused on the effectiveness of two levels of active office interventions to reduce SB in office workers. Participants were 78 nonacademic university employees divided into a control (CON) group and an intervention (INT) group. At the organizational level, it was found that the organizational health culture, the physical and social environment, and the organizational health behavior were dramatically changed. At the individual level, compared with the CON group, the INT group was significantly higher in the METs rate; light-intensity physical activity (LPA); and moderate-to-vigorous-intensity physical activity, and was lower in SB (CON, 397.30 ± 39.33 minutes vs. INT, 389.09 ± 37.59 minutes), all p less then .05.   • Systemic sclerosis (SSc) is a chronic disease characterized by autoimmunity, vasculopathy, and visceral and cutaneous fibrosis. Vitamin D has several functions in the immunological system, and different studies have suggested a potential role in triggering autoimmune diseases. Patients with SSc may present with low serum levels of vitamin D, but the association between hypovitaminosis D and disease onset or any clinical manifestation is still obscure. Our goal was to verify the causal relationship between hypovitaminosis D and SSc onset or any particular clinical manifestation in the literature.

    Forty articles met the eligibility criteria and the main results of each study are described. Additionally, in some reports serum levels of vitamin D were inversely correlated with the severity of SSc. Oral supplementation did not seem to affect serum levels of vitamin D.     Resilience research to date has been criticised for its consideration of resilience as a personal trait instead of a process, and for identifying individual factors related to resilience with no consideration of the ecological context. The overall aim of the current study was to explore the multi-level process through which adults recovering from EDs develop resilience, from the perspectives of clients and clinicians. The objective of this research was to outline the stages involved in the process of developing resilience, which might help to inform families and services in how best to support adults with EDs during their recovery.

    Thirty participants (15 clients; 15 clinicians) took part in semi-structured interviews, and responded to questions relating to factors associated with resilience. Using an inductive approach, data were analysed using reflexive thematic analysis.

    The overarching theme which described the process of developing resilience was 'Bouncing **** to being me', which involved three stl-being of the person recovering. One way to achieve this is to focus on resilience, which was identified as a fundamental aspect of eating disorder recovery in previous research. This study conceptualises resilience as a dynamic process that is influenced not only at a personal level but also through the environment in which the person lives. This study gathered data from adults with eating disorders and their treating clinicians, to devise a framework for resilience development for adults recovering from eating disorders. The paper discussed ways in which these findings and the framework identified can be easily implemented in clinical practice to facilitate a better understanding of eating disorder resilience and to enhance recovery outcomes.
    Osteoarthritis (OA) is defined as a degenerative disease. Pivotal roles of long non-coding RNA (lncRNAs) in OA are widely elucidated. Herein, we intend to explore the function and molecular mechanism of lncRNA KCNQ1OT1 in CHON-001 cells.

    Relative expression of KCNQ1OT1, miR-126-5p and TRPS1 was determined by quantitative real-time polymerase chain reaction (qRT-PCR). Cell viability was examined by MTT assay. The migratory ability of chondrocytes was assessed by transwell assay. Western blot was used to determine relative protein expression of collagen II, MMP13 and TRPS1. Dual-luciferase reporter (DLR) assay was applied to test the target of lncRNA KCNQ1OT1 or miR-126-5p.

    Relative expression of KCNQ1OT1 and TRPS1 was reduced, whereas miR-126-5p was augmented in cartilage tissues of post-traumatic OA patients compared to those of subjects without post-traumatic OA. Increased KCNQ1OT1 or decreased miR-126-5p enhanced cell viability and migration, and repressed extracellular matrix (ECM) degradation in CHON-001 cells. MiR-126-5p was the downstream target of KCNQ1OT1, and it could directly target TRPS1. There was an inverse correlation between KCNQ1OT1 and miR-126-5p or between miR-126-5p and TRPS1. Meantime, there was a positive correlation between KCNQ1OT1 and TRPS1. The promoting impacts of KCNQ1OT1 on cell viability and migration as well as the suppressive impact of KCNQ1OT1 on ECM degradation were partially abolished by miR-126-5p overexpression or TRPS1 knockdown in CHON-001 cells.

While reasonably successful in achieving physical recovery (i.e. weight restoration), both empirical and anecdotal accounts highlight a lack of attention to the psychological recovery of the adolescent within manualised FBT. As such, there is a need for innovation to explore treatment variations and alternatives for the proportion of adolescents with AN who do not respond favourably to this first-line treatment. This paper introduces a new treatment framework to the field for clinical consideration and empirical assessment. Adolescent and Parent Treatment (APT) for adolescent AN draws from both family-based and individual treatment models, applying a developmental lens. APT attends to physical and psychological recovery simultaneously and from the start of treatment, with capacity to tailor individual psychological modules to the adolescent formulation. While clearly in its infancy, APT provides an exciting new avenue for exploration within the field, as we seek new avenues to support young people and their families to effectively combat this deadly illness.

This study gathered data from adults with eating disorders and their treating clinicians, to devise a framework for resilience development for adults recovering from eating disorders. The paper discussed ways in which these findings and the framework identified can be easily implemented in clinical practice to facilitate a better understanding of eating disorder resilience and to enhance recovery outcomes. Osteoarthritis (OA) is defined as a degenerative disease. Pivotal roles of long non-coding RNA (lncRNAs) in OA are widely elucidated. Herein, we intend to explore the function and molecular mechanism of lncRNA KCNQ1OT1 in CHON-001 cells. Relative expression of KCNQ1OT1, miR-126-5p and TRPS1 was determined by quantitative real-time polymerase chain reaction (qRT-PCR). Cell viability was examined by MTT assay. The migratory ability of chondrocytes was assessed by transwell assay. Western blot was used to determine relative protein expression of collagen II, MMP13 and TRPS1. Dual-luciferase reporter (DLR) assay was applied to test the target of lncRNA KCNQ1OT1 or miR-126-5p. Relative expression of KCNQ1OT1 and TRPS1 was reduced, whereas miR-126-5p was augmented in cartilage tissues of post-traumatic OA patients compared to those of subjects without post-traumatic OA. Increased KCNQ1OT1 or decreased miR-126-5p enhanced cell viability and migration, and repressed extracellular matrix (ECM) degradation in CHON-001 cells. MiR-126-5p was the downstream target of KCNQ1OT1, and it could directly target TRPS1. There was an inverse correlation between KCNQ1OT1 and miR-126-5p or between miR-126-5p and TRPS1. Meantime, there was a positive correlation between KCNQ1OT1 and TRPS1. The promoting impacts of KCNQ1OT1 on cell viability and migration as well as the suppressive impact of KCNQ1OT1 on ECM degradation were partially abolished by miR-126-5p overexpression or TRPS1 knockdown in CHON-001 cells. Overexpression of KCNQ1OT1 attenuates the development of OA by sponging miR-126-5p to target TRPS1. Our findings may provide a possible therapeutic strategy for human OA in clinic. While reasonably successful in achieving physical recovery (i.e. weight restoration), both empirical and anecdotal accounts highlight a lack of attention to the psychological recovery of the adolescent within manualised FBT. As such, there is a need for innovation to explore treatment variations and alternatives for the proportion of adolescents with AN who do not respond favourably to this first-line treatment. This paper introduces a new treatment framework to the field for clinical consideration and empirical assessment. Adolescent and Parent Treatment (APT) for adolescent AN draws from both family-based and individual treatment models, applying a developmental lens. APT attends to physical and psychological recovery simultaneously and from the start of treatment, with capacity to tailor individual psychological modules to the adolescent formulation. While clearly in its infancy, APT provides an exciting new avenue for exploration within the field, as we seek new avenues to support young people and their families to effectively combat this deadly illness.

  • Co-infection with ancillary pathogens is a significant modulator of morbidity and mortality in infectious diseases. There have been limited reports of co-infections accompanying SARS-CoV-2 infections, albeit lacking India specific study. The present study has made an effort toward elucidating the prevalence, diversity and characterization of co-infecting respiratory pathogens in the nasopharyngeal tract of SARS-CoV-2 positive patients. Two complementary metagenomics based sequencing approaches, Respiratory Virus Oligo Panel (RVOP) and Holo-seq, were utilized for unbiased detection of co-infecting viruses and bacteria. The limited SARS-CoV-2 clade diversity along with differential clinical phenotype seems to be partially explained by the observed spectrum of co-infections. We found a total of 43 bacteria and 29 viruses amongst the patients, with 18 viruses commonly captured by both the approaches. In addition to SARS-CoV-2, Human Mastadenovirus, known to cause respiratory distress, was present in a majority of the samples. We also found significant differences of bacterial reads based on clinical phenotype. Of all the bacterial species identified, ∼60% have been known to be involved in respiratory distress. Among the co-pathogens present in our sample cohort, anaerobic bacteria accounted for a preponderance of bacterial diversity with possible role in respiratory distress. Clostridium botulinum, Bacillus cereus and Halomonas sp. are anaerobes found abundantly across the samples. Our findings highlight the significance of metagenomics based diagnosis and detection of SARS-CoV-2 and other respiratory co-infections in the current pandemic to enable efficient treatment administration and better clinical management. To our knowledge this is the first study from India with a focus on the role of co-infections in SARS-CoV-2 clinical sub-phenotype.Cell surface hydrophobicity (CSH) and adhesion are very important phenotypical traits for probiotics that confer them a competitive advantage for the resilience in the human gastrointestinal tract. This study was aimed to understand the effects over time of a 50 MPa hyperbaric treatment on the surface properties of Lactobacillus acidophilus 08 including CSH, autoaggregation, and in vitro adhesion (mucin layer and Caco-2 cells). Moreover, a link between the hurdle applied and the expression of genes involved in the general stress response (groEL and clpP) and adhesion processes (efTu and slpA) was evaluated. High pressure homogenization (HPH) at 50 MPa significantly increased the CSH percentage (H%), autoaggregation and in vitro adhesion on mucin of L. acidophilus 08 cells compared with the untreated cells. Moreover, the hyperbaric hurdle induced an upregulation of the stress response genes groEL and ef-TU together with a down regulation of the clpP and S-layer slpA genes. Looking at the protein profile, HPH-treatment showed an increase in the number or intensity of protein bands at high and low molecular weights.The level of water table and temperature are two environmental variables shaping soil bacterial communities, particularly in peatland ecosystems. However, discerning the specific impact of these two factors on bacterial communities in natural ecosystems is challenging. To address this issue, we collected pore water samples across different months (August and November in 2017 and May 2018) with a gradient of water table changes and temperatures at the Dajiuhu peatland, Central China. The samples were analyzed with 16S rRNA high-throughput sequencing and Biolog EcoMicroplates. Bacterial communities varied in the relative abundances of dominant taxa and harbored exclusive indicator operational taxonomic units across the different months. Despite these differences, bacterial communities showed high similarities in carbon utilization, with preferences for esters (pyruvic acid methyl ester, Tween 40, Tween 80, and D-galactonic acid γ-lactone), amino acids (L-arginine and L-threonine), and amines (phenylethylamine a fluctuation of the water table.In this study, the mitogenome of Hannaella oryzae was sequenced by next-generation sequencing (NGS) and successfully assembled. The H. oryzae mitogenome comprised circular DNA molecules with a total size of 26,444 bp. We found that the mitogenome of H. oryzae partially deleted the tRNA gene transferring cysteine. Comparative mitogenomic analyses showed that intronic regions were the main factors contributing to the size variations of mitogenomes in Tremellales. Introns of the cox1 gene in Tremellales species were found to have undergone intron loss/gain events, and introns of the H. oryzae cox1 gene may have different origins. Gene arrangement analysis revealed that H. oryzae contained a unique gene order different from other Tremellales species. Phylogenetic analysis based on a combined mitochondrial gene set resulted in identical and well-supported topologies, wherein H. oryzae was closely related to Tremella fuciformis. This study represents the first report of mitogenome for the Hannaella genus, which will allow further study of the population genetics, taxonomy, and evolutionary biology of this important phylloplane yeast and other related species.Recent advances in 3D printing have led to a rise in the use of 3D printed materials in prosthetics and external medical devices. This abundant use of 3D printed devices in the medical fields warrants studies to assess the ability of different microorganisms to attach and colonize to such surfaces. In this study, we describe methods to determine bacterial biofouling and biofilm formation on 3D printed materials. We explored the biofilm-forming ability of multiple opportunistic pathogens commonly found on the human body including Escherichia coli, Pseudomonas aeruginosa, and Staphylococcus aureus to colonize eight commonly used polylactic acid (PLA) polymers.
