LPP amplitudes were significantly smaller in the social exclusion group than those in the social inclusion group for painful stimuli. Our results indicate that social exclusion does not affect empathic responses during the early emotional sharing stage. However, it down-regulates empathic responses at the late cognitive controlled stage, and this modulation is attenuated gradually. Pedunculopontine nucleus (PPN) region DBS has been used for medication-refractory FoG, with mixed findings. FoG, as a paroxysmal phenomenon, provides an ideal framework for the possibility of closed-loop DBS (CL-DBS). Methods In this clinical trial (NCT02318927), five subjects with medication-refractory FoG underwent bilateral GPi DBS implantation to address levodopa-responsive PD symptoms with open-loop stimulation. https://www.selleckchem.com/products/ik-930.html Additionally, PPN DBS leads were implanted for CL-DBS to treat FoG. The primary outcome of the study was a 40% improvement in medication-refractory FoG in 60% of subjects at 6 months when "on" PPN CL-DBS. Secondary outcomes included device feasibility to gauge the recruitment potential of this four-lead DBS approach for a potentially larger clinical trial. Safety was judged based on adverse events and explantation rate. Findings The feasibility of this approach was demonstrated as we recruited five subjects with both "on" and "off" medication freezing. The safety for this population of patients receiving four DBS leads was suboptimal and associated with a high explantation rate of 40%. The primary clinical outcome in three of the five subjects was achieved at 6 months. However, the group analysis of the primary clinical outcome did not reveal any benefit. Interpretation This study of a human PPN CL-DBS trial in medication-refractory FoG showed feasibility in recruitment, suboptimal safety, and a heterogeneous clinical effect in FoG outcomes.The present study examines the comorbidity between specific learning disorders (SLD) and attention deficit and hyperactivity disorder (ADHD) by comparing the neuropsychological profiles of children with and without this comorbidity. Ninety-seven schoolchildren from 8 to 14 years old were tested a clinical sample of 49 children with ADHD (n = 18), SLD (n = 18) or SLD in comorbidity with ADHD (n = 13), and 48 typically-developing (TD) children matched for age and intelligence. Participants were administered tasks and questionnaires to confirm their initial diagnosis, and a battery of executive function (EF) tasks testing inhibition, shifting, and verbal and visuospatial updating. Using one-way ANOVAs, our results showed that all children in the clinical samples exhibited impairments on EF measures (inhibition and shifting tasks) when compared with TD children. A more specific pattern only emerged for the updating tasks. Only children with SLD had significant impairment in verbal updating, whereas children with ADHD, and those with SLD in comorbidity with ADHD, had the worst performance in visuospatial updating. The clinical and educational implications of these findings are discussed.Embodied theories of grounded semantics postulate that, when word meaning is first acquired, a link is established between symbol (word form) and corresponding semantic information present in modality-specific-including primary-sensorimotor cortices of the brain. Direct experimental evidence documenting the emergence of such a link (i.e., showing that presentation of a previously unknown, meaningless word sound induces, after learning, category-specific reactivation of relevant primary sensory or motor brain areas), however, is still missing. Here, we present new neuroimaging results that provide such evidence. We taught participants aspects of the referential meaning of previously unknown, senseless novel spoken words (such as "Shruba" or "Flipe") by associating them with either a familiar action or a familiar object. After training, we used functional magnetic resonance imaging to analyze the participants' brain responses to the new speech items. We found that hearing the newly learnt object-related word sounds selectively triggered activity in the primary visual cortex, as well as secondary and higher visual areas.These results for the first time directly document the formation of a link between the novel, previously meaningless spoken items and corresponding semantic information in primary sensory areas in a category-specific manner, providing experimental support for perceptual accounts of word-meaning acquisition in the brain.It has been suggested that the thalamus acts as a blackboard, on which the computations of different cortical modules are composed, coordinated, and integrated. This article asks what blackboard role the thalamus might play, and whether that role is consistent with the neuroanatomy of the thalamus. It does so in a context of Bayesian belief updating, expressed as a Free Energy Principle. We suggest that the thalamus-as-a-blackboard offers important questions for research in spatial cognition. Several prominent features of the thalamus-including its lack of olfactory relay function, its lack of internal excitatory connections, its regular and conserved shape, its inhibitory interneurons, triadic synapses, and diffuse cortical connectivity-are consistent with a blackboard role.Different thalamic nuclei may play different blackboard roles (1) the Pulvinar, through its reciprocal connections to posterior cortical regions, coordinates perceptual inference about "what is where" from multi-sense-data. (2) The Mediodorsal (MD) nucleus, through its connections to the prefrontal cortex, and the other thalamic nuclei linked to the motor cortex, uses the same generative model for planning and learning novel spatial movements. (3) The paraventricular nucleus may compute risk-reward trade-offs. We also propose that as any new movement is practiced a few times, cortico-thalamocortical (CTC) links entrain the corresponding cortico-cortical links, through a process akin to supervised learning. Subsequently, the movement becomes a fast unconscious habit, not requiring the MD nucleus or other thalamic nuclei, and bypassing the thalamic bottleneck.
