METHODS This study included 6343 men and women aged ≥50 years from the Korean National Health and Nutrition Examination Survey. Osteoarthritis was defined as Kellgren-Lawrence grade ≥2 in the knee on radiographic images with knee pain. Mental health and health-related QoL were assessed according to three dimensions (depressive mood, psychological distress and suicidal ideation) and five domains (impaired mobility, impaired self-care, impaired usual activities, pain/discomfort and anxiety/depression). The odds ratios and 95% confidence intervals (CI) for mental health and health-related QoL were calculated using multiple logistic regression analyses. RESULTS Compared with the control group, the odds ratios for depressive mood, psychological distress and suicidal ideation in the osteoarthritis group were 2.80 (95% CI 1.31-3.31), 1.92 (95% CI 1.21-3.05) and 1.97 (95% CI 1.31-2.94) in men, and 1.51 (95% CI 1.16-1.95), 1.36 (95% CI 1.07-1.72) and 1.92 (95% CI 1.49-2.46) in women after adjusting for potential confounding variables. Similarly, the odds ratios for impaired mobility, impaired self-care, impaired usual activities, pain/discomfort and anxiety/depression in the osteoarthritis group were 12.37 (95% CI 8.17-18.72), 3.02 (95% CI 1.84-4.94), 7.33 (95% CI 4.89-10.98), 8.92 (95% CI 5.92-13.45) and 2.56 (95% CI 1.52-4.29) in men, and 8.29 (95% CI 6.41-10.73), 2.53 (95% CI 1.74-3.69), 4.40 (95% CI 3.33-5.80), 4.79 (95% CI 3.72-6.17) and 1.85 (1.44-2.37) in women after adjusting for the same covariables. CONCLUSIONS Knee osteoarthritis was significantly associated with deteriorated mental health and health-related QoL in middle-aged and older men and women. Most of the treatments currently used for Parkinson's disease - the second most prevalent - are only symptomatic. Therefore, it is urgent to develop drugs that are able to act simultaneously on different targets, being able to stop neuronal death and promote the recovery of neuronal populations already affected. In this work, we studied the activity of a series of hybrid molecules, which combine the structure of both coumarin and an alkynylamine group inspired on rasagiline, as MAO inhibitors, antioxidants and neuroprotective agents. Half of the studied hybrids turned out to be selective monoamine oxidase B (hMAO-B) inhibitors in the low micro/nanomolar range, demonstrating that positions 3 (compounds 1-3) and 7 (compounds 8 and 10) of the coumarin scaffold are the most suitable for the incorporation of the alkynylamine chain. All the studied compounds proved to be capable of neutralizing free radicals (DPPH). Finally, the 4-(but-2-yn-1-ylamino)coumarin (5) showed neuroprotective effects on glial cells and the 4-methyl-7-(pent-2-yn-1-ylamino)coumarin (8) inhibited intraneuronal ROS production as well. © 2020 Wiley-VCH Verlag GmbH & Co. METHODS Data from 366 consecutive PICC insertions by five trained angiography-specialized radiographers were prospectively collected over a 12-month period. For each PICC insertion, patient demographics, including past medical history of cystic fibrosis (CF), number of punctures, vein used, final tip position, contrast administration and screening time were recorded. Institutional review board approval was obtained. RESULTS The overall PICC insertion success rate was 100%. Fifty-five (15%) had a known medical history of CF. Three hundred and thirty-one (90%) PICC insertions required a single puncture and 32 (9%) required two punctures. The remaining three insertions required three punctures. Since we previously found that GATA6 promotes the stemness in HCT-116 and HT-29 human colorectal cancer (CRC) cells, we aimed to identify the downstream mediator(s) of the stemness-stimulating effect of GATA6 herein. LRH-1 was found as a direct target of GATA6 and its upregulation promoted the stemness in both HCT-116 and HT-29 cells. Subsequently, hypoxia-inducible factor-1α (HIF-1α) was identified as a direct target of LRH-1 and its expression level and activity were significantly elevated in the LRH-1-overexpressing clones established from the aforementioned two CRC lines. Accordingly, the expression levels of several HIF-1α targets were also markedly increased, resulting in a stronger glycolysis associated with dramatic elevations of the lactate levels in these cells. Strikingly, higher mitochondrial activities were also found in these clones which might be attributed to the increase of PGC-1α stimulated by the lactate uptaken through the upregulated MCT-1.
