Here, we review relevant aspects of the TOS, EMS, and NSF epidemic outbreaks of chemically-induced systemic fibrosing disorders that provide strong support to the hypothesis that SSc is caused by a toxic or biological agent that following its internalization by endothelial cells induces in genetically predisposed individuals a series of molecular alterations that result in the development of SSc clinical and pathological alterations.
We aimed to determine the real-world prevalence and investigate risk factors for rheumatoid arthritis (RA)-related lung disease on chest computed tomography (CT) imaging. We also investigated the impact of RA-related lung disease on mortality.
We studied chest CT imaging abnormalities among RA patients. We determined the presence and type of abnormalities using the chest CT imaging radiologic report. RA-related lung disease was defined as interstitial lung disease (ILD), bronchiectasis, or pleural disease. We examined whether demographics and RA characteristics were associated with RA-related lung disease using logistic regression. RA-related lung disease and mortality was described using survival curves and Cox regression.
We analyzed 190 patients who had chest CT imaging performed for clinical indications. Mean age was 64.2 years (SD 11.8), 80.0% were female, and 75.3% were seropositive. RA-related lung disease was detected in 54 patients (28.4%); 30 (15.8%) had ILD, 27 (14.2%) had bronchiectasis, and 18 (9.5%) had pleural disease. RA-related lung disease was reported in both seropositive and seronegative RA (28.7% vs. https://www.selleckchem.com/products/crt-0105446.html 27.7%, p=1.00). Male sex (OR 2.62, 95%CI 1.17-5.88) and current methotrexate use (OR 2.73, 95%CI 1.27-5.61vs. not current) were associated with RA-related lung disease. Twenty-four (44.4%) patients with RA-related lung disease died during mean 7.0 years of follow-up. RA-related lung disease had HR of 5.35 (95%CI 0.72-39.9) for mortality compared to normal chest CT.
In this real-world study, RA-related lung disease was commonly detected on chest CT imaging regardless of RA serostatus. RA-related lung disease had high mortality, emphasizing the importance in close monitoring of these patients.

Osteoarthritis Research Society International (OARSI) Expert Consensus Guidelines recommend topical non-steroidal anti-inflammatory drugs as first-line medications for osteoarthritis (OA) knee pain, but several voluminous daily applications are required to achieve efficacy. There is a need to develop new and improved topical analgesics with a faster onset, longer duration of action, and the requirement to apply less gel. This trial investigated the safety and efficacy of a new 3.06% diclofenac gel (AMZ001) in subjects with knee OA.
In total, 444 subjects (AMZ001 twice daily (BID) [n=121], AMZ001 once daily (QD)+placebo QD [n=121], placebo BID [n=121], or Voltaren 1% 4-times daily [n=81]) were enrolled. All except Voltaren 1% (single-blinded) were applied topically in a double-blind manner for a total of 4-weeks. The primary endpoint was the change from baseline to week 4 in the WOMAC pain sub-score in the target knee. Secondary and exploratory endpoints included additional efficacy measures (WOMAC total snificant improvements vs placebo. AMZ001 was generally well tolerated.Priming effect (PE) is increasingly recognized as an important mechanism in the microbial degradation of dissolved organic matter (DOM) from freshwater to the ocean. However, potential PE during the mixing of DOM from different sources and the effects on different DOM constituents are still largely unknown. This study examined the PE after adding DOM from typical natural and anthropogenic sources (rainwater, fresh plant, leaf litter, and wastewater) into pre-aged river DOM, using dissolved organic carbon (DOC) measurement, absorption spectroscopy, and fluorescence excitation-emission matrices-parallel factor analysis (EEMs-PARAFAC). The plant-derived DOM had a low humic content and was dominated by benzoic acid-like and tyrosine-like fluorescent components (C4 and C5), which showed a high DOC bioavailability of 80%. DOC in rainwater and wastewater also had high bioavailabilities (45%-50%), while DOM in the leaf litter leachate showed high aromaticity, average molecular weight, and humic content but low DOC bioavailability (12%). There was generally limited PE ( less then 5% of the initial values) on the degradation of DOC and chromophoric DOM (CDOM) for most samples. Two humic-like components (C2 and C3) showed little PE, while the humic-like C1 and C6, tyrosine-like C5, and tryptophan-like C7 showed variable PE after adding rainwater, wastewater, and plant leachate. Overall, the results revealed that the DOM from typical natural and anthropogenic sources had different composition and bioavailability, and their inputs to aquatic environments would result in variable PE on the bulk DOC and different DOM components.The electro-assisted autohydrogenotrophic reduction of perchlorate (ClO4-) was investigated in a dual-chamber biofilm-electrode reactor (BER), in which the microbial community was inoculated from natural sediments. To avoid the effect of extreme pH and direct electron transfer on perchlorate reduction, a novel cathode configuration was designed. The pH of the cathode compartment was successfully controlled in the range of 7.2-8.4 during whole experiment. The effective biological autohydrogenotrophic reduction of perchlorate was achieved using hydrogen generated in-situ on the electrode surface, and the removal rate of 10 mg L-1 perchlorate reached 98.16% at HRT of 48 h. The highest perchlorate removal flux reached to 1498.420 mg m-2·d-1 with a 0.410 kW·h g-perchlorate-1 energy consumption. The microbial community evolution in the BER was determined by high-throughput sequencing and the results indicated that the Firmicutes and Bacteroidetes were dominant at phylum level when perchlorate concentration was 10 mg L-1 or lower.
