Low levels of BST integrity were observed during baseline. Immediately after viewing a 13-min training video, 2 participants implemented BST at mastery (i.e., 100% integrity). The remaining 2 participants required brief supplemental performance feedback to reach criterion performance. The effects of the training generalized to training 2 novel skills and maintained in follow-up probes conducted 1 to 4 weeks posttraining. A social validity measure indicated high levels of satisfaction with the video-based training and high acceptability of BST. Implications of these findings for organizational training practices and directions for future research are discussed.Bacillus coagulans, a thermophilic facultative anaerobe, is a favorable chassis strain for the biosynthesis of desired products. In this study, B. coagulans was converted into an efficient malic acid producer by metabolic engineering and promoter engineering. Promoter mapping revealed that the endogenous promoter Pldh was a tandem promoter. Accordingly, a promoter library was developed, covering a wide range of relative transcription efficiencies with small increments. A reductive tricarboxylic acid pathway was established in B. coagulans by introducing the genes encoding pyruvate carboxylase (pyc), malate dehydrogenase (mdh), and phosphoenolpyruvate carboxykinase (pckA). Five promoters of various strengths within the library were screened to fine-tune the expression of pyc to improve the biosynthesis of malic acid. In addition, genes involved in the competitive metabolic pathways were deleted to focus the substrate and energy flux toward malic acid. Dual-phase fed-batch fermentation was performed to increase the biomass of the strain, further improving the titer of malic acid to 25.5 g/L, with a conversion rate of 0.3 g/g glucose. Our study is a pioneer research using promoter engineering and genetically modified B. coagulans for the biosynthesis of malic acid, providing an effective approach for the industrialized production of desired products using B. coagulans.Identifying interspecific associations is very important for understanding the community assembly process. However, most methods provide only an average association and assume that the association strength does not vary along the environmental gradient or with time. The scale effects are generally ignored. We integrated the idea of wavelet and network topological analysis to provide a novel way to detect nonrandom species associations across scales and along gradients using continuous or presence-absence ecological data. We first used a simulated species distribution data set to illustrate how the wavelet correlation analysis builds an association matrix and demonstrates its statistical robustness. Then, we applied the wavelet correlation network to a presence-absence data set of soil invertebrates. We found that the associations of invertebrates varied along an altitudinal gradient. We conclude by discussing several possible extensions of this method, such as predicting community assembly, utility in the temporal dimension, and the shifting effects of highly connected species within a community. The combination of the multiscale decomposition of wavelet and network topology analysis has great potential for fostering an understanding of the assembly and succession of communities, as well as predicting their responses to future climate change across spatial or temporal scales.

To assess the malignancy risk in patients with secukinumab-treated psoriasis, psoriatic arthritis (PsA) and ankylosing spondylitis (AS).

This integrated safety analysis from both the secukinumab clinical trial programme and postmarketing safety surveillance data included any patient receiving at least one approved dose of secukinumab with a maximum of 5years of follow-up. Safety analyses evaluated the rate of malignancy using exposure-adjusted incidence rates [EAIR; incidence rates per 100 patient treatment-years (PTY)]. Standardized incidence ratios (SIRs) were reported using the Surveillance, Epidemiology, and End Results Program (SEER) database as a reference population. Crude incidence of malignancy was also reported using postmarketing surveillance data.

Safety data from 49 clinical trials with secukinumab-treated patients were included 10685 patients with psoriasis, 2523 with PsA and 1311 with AS. Across indications over a 5-year period, the EAIR of malignancy was 0·85 per 100 PTY [95% confidence interval (CI) 0·74-0·98] in secukinumab-treated patients, corresponding to 204 patients per 23908 PTY. Overall, the observed vs. expected number of malignancies from secukinumab clinical trial data were comparable, as indicated by an SIR of 0·99 (95% CI 0·82-1·19) across indications. The estimated crude cumulative incidence reporting rate per 100 PTY for malignancy was 0·27 in the postmarketing surveillance data across indications with a cumulative exposure of 285811 PTY.
