Laccase is one of the best known biocatalysts which degrade wide varieties of complex molecules that are both non-cyclic and cyclic in structure. The study focused on enzyme kinetics of a purified laccase from Trametes hirsuta L. fungus and its application on biotransformation of a carcinogenic molecule 1,4-dioxane.
Laccase was purified from white-rot fungus T. hirsuta L. which showed specific activity of 978.34 U/mg after the purification fold of 54.08. The stable laccase activity (up to 16h) is shown at 4-6 pH and 20-40°C temperature range. The purified enzyme exhibited significant stability for 10 metal ions up to 10mM concentration, except for Fe
and Hg
. The Cu
ion induced laccase activity up to 142% higher than the control at 10mM concentration. The laccase enzyme kinetic parameters K
was 20 ± 5µM and 400 ± 60µM, whereas K
was 198.29 ± 0.18/s and 80.20 ± 1.59/s for 2,2'-azino-bis (3-ethylbenzothiazoline-6-sulphonic acid) (ABTS) and guaiacol respectively. The cyclic ether 1,4-dioxane (100ppm) was completely degraded in presence of purified laccase within 2h of incubation and it was confirmed by HPLC and GC analysis. The oxidation reaction was accelerated by 25, 22, 6 and 19% in presence of 1mM syringaldehyde, vanillin, ABTS and guaiacol mediators respectively.

In this study, fungal laccase (a natural biocatalyst) based degradation of synthetic chemical 1,4-dioxane was reported for the first time. This method has added advantages over the multiple methods reported earlier being a natural remedy.Critical literatures, and public discourses, on public health policies and practices often present fixated concerns with paternalism. In this paper, rather than focus on the question of whether and why intended instances of paternalistic policy might be justified, we look to the wider, real-world socio-political contexts against which normative evaluations of public health must take place. We explain how evaluative critiques of public health policy and practice must be sensitive to the nuance and complexity of policy contexts. This includes sensitivity to the 'imperfect' reach and application of policy, leading to collateral effects including collateral paternalism. We argue that theoretical critiques must temper their demandingness to real-world applicability, allowing for the detail of social and policy contexts, including harm reduction apparent knock-down objections of paternalism cannot hold if they are limited to an abstract or artificially-isolated evaluation of the reach of a public health intervention.A room temperature benzene and formaldehyde gas sensor system with an ionogel as sensing material is presented. The sensing layer is fabricated employing poly(N-isopropylacrylamide) polymerized in the presence of 1-ethyl-3-methylimidazolium ethyl sulfate ionic liquid onto gold interdigitated electrodes. When the ionogel is exposed to increasing formaldehyde concentrations employing N2 as a carrier gas, a more stable response is observed in comparison to the bare ionic liquid, but no difference in sensitivity occurs. On the other hand, when air is used as carrier gas the sensitivity of the system towards formaldehyde is decreased by one order of magnitude. At room temperature, the proposed sensor exhibited in air higher sensitivities to benzene, at concentrations ranging between 4 and 20 ppm resulting, in a limit of detection of 47 ppb, which is below the standard permitted concentrations. The selectivity of the IL towards HCHO and C6H6 is demonstrated by the absence of response when another IL is employed. Humidity from the ambient air slightly affects the resistance of the system proving the protective role of the polymeric matrix. Furthermore, the gas sensor system showed fast response/recovery times considering the thickness of the material, suggesting that ionogel materials can be used as novel and highly efficient volatile organic compounds sensors operating at room temperature.Graphical abstract.
Sepsis is a complication in acute-on-chronic liver failure (ACLF) patients associated with high rates of mortality and morbidity. Early diagnosis of sepsis in ACLF patients can improve prognosis. This study aimed to explore potential effective biomarkers for the early diagnosis of sepsis in ACLF patients.
Ninety-four ACLF patients with sepsis were enrolled from 10 hospitals across China from January 2015 to June 2016 as well as 49 ACLF patients without infection from Xiangya Hospital. The first-day admission data and SOFA score and CLIF-SOFA score were collected. The differences of indicators between groups were compared with Kruskal-Wallis test. The receiver-operating characteristic (ROC) curve was analyzed to evaluate the diagnostic efficiency of the selected factors.
Soluble triggering receptor expressed on myeloid cell-1 (sTREM-1) and presepsin were significantly higher in ACLF-sepsis patients compared with ACLF patients with no infection (P < 0.001). sTREM-1 and presepsin presented higher diagnostic value in sepsis for ACLF patients compared with other biomarkers [white blood cells (WBC), procalcitonin (PCT) and C-reactive protein (CRP)]. Combining sTREM-1 or presepsin with the CLIF-SOFA score increased the diagnostic efficiency (AUC = 0.876 or AUC = 0.913, respectively).
sTREM-1 and presepsin are potential biomarkers for the early diagnosis of sepsis in ACLF patients. The combination of presepsin and the CLIF-SOFA score is a promising method for diagnosing sepsis in ACLF patients.
