These data identify platelet XPR1 as an integral regulator of platelet polyphosphate metabolism and reveal a fundamental role for phosphate homeostasis in thrombosis.Activated B-cell (ABC)-diffuse large B-cell lymphomas (DLBCLs) are clinically aggressive and phenotypically complex malignancies, whose transformation mechanisms remain unclear. Partially differentiated antigen-secreting cells (plasmablasts) have long been regarded as cells-of-origin for these tumors, despite lack of definitive experimental evidence. Recent DLBCL reclassification based on mutational landscapes identified MCD/C5 tumors as specific ABC-DLBCLs with unfavorable clinical outcome, activating mutations in the signaling adaptors MYD88 and CD79B, and immune evasion through mutation of antigen-presenting genes. MCD/C5s manifest prominent extranodal dissemination and similarities with primary extranodal lymphomas (PENLs). In this regard, recent studies on TBL1XR1, a gene recurrently mutated in MCD/C5s and PENLs, suggest that aberrant memory B cells (MBs), and not plasmablasts, are the true cells-of-origin for these tumors. Moreover, transcriptional and phenotypic profiling suggests that MCD/C5s, as a class, represent bona fide MB tumors. Based on emerging findings we propose herein a generalized stepwise model for MCD/C5 and PENLs pathogenesis, whereby acquisition of founder mutations in activated B cells favors the development of aberrant MBs prone to avoid plasmacytic differentiation on recall and undergo systemic dissemination. Cyclic reactivation of these MBs through persistent antigen exposure favors their clonal expansion and accumulation of mutations, which further facilitate their activation. As a result, MB-like clonal precursors become trapped in an oscillatory state of semipermanent activation and phenotypic sway that facilitates ulterior transformation and accounts for the extranodal clinical presentation and biology of these tumors. In addition, we discuss diagnostic and therapeutic implications of a MB cell-of-origin for these lymphomas.International attention on the environmental impacts of China's Belt and Road Initiative (BRI) is increasing, but little is known internationally about the large corpus of Chinese BRI environmental research. We present the first systematic review of the Chinese and English-language BRI environmental research, supported with text mining and sentiment analysis. We found that the research is dominated by Chinese authors writing about BRI routes within China in Chinese, even though concerns around BRI are largely about impacts and benefits within host countries, and the volume of publications in English is recently catching up. Different disciplines and methods are well-represented across languages, apart from specific types of Chinese social science papers. The sentiments of academic research are largely neutral and less polarised than media discourse. We recommend that scientists and practitioners should pay more attention to BRI environmental impacts in developing countries and proactively engage local voices.In this work, we demonstrate a discrete dispersion scan scheme using a low number of flat windows to vary the dispersion of laser pulses in discrete steps. Monte Carlo simulations indicate that the pulse duration can be retrieved accurately with less than 10 dispersion steps, which we verify experimentally by measuring few-cycle pulses and material dispersion curves at 3 and 10 µm wavelength. This minimal measuring scheme using only five optical components without the need for linear positioners and interferometric alignment can be readily implemented in many wavelength ranges and situations.We present an experimental characterization of the amplification of sub-nanosecond duration laser pulses at a wavelength of 1538 nm in short custom-made ErYb phosphate glass fibers with different core diameters. The fibers vary in their diameter from 100 µm (highly multi-mode) down to 12 µm (single-mode). The peak power, energy per pulse, and spectral shape of the amplified signal are presented. With our input pulses, the measurements show that the large core diameter fibers do not increase the amplification of the 1538 nm signal. We believe this is due to the high re-absorption of the Er3+ ions in the phosphate fiber. The optimal fiber geometry was found to have a core diameter of 20 µm with a length of 14 cm. The maximum peak power is 8.25 kW, corresponding to a net gain of 10.9 dB, with a pulse duration of 0.7 ns and a repetition rate of 40 kHz.A high-energy, tunable, long-wave mid-infrared optical parametric oscillator (OPO) based on the BaGa4Se7 crystal was demonstrated in this Letter with 1064 nm laser pumping. The mid-infrared OPO was designed as a double-pass single resonant oscillator (DP-SRO) to reduce the threshold and improve the outputs. Further optimization on the cavity length was theoretically and experimentally studied. With a short cavity length of 30 mm, the output energy of over 1 mJ/pulse at 11 µm was obtained with the pump energy of 39.5 mJ/pulse. https://www.selleckchem.com/products/gdc-0068.html In addition, a wide tuning range of 8-14 µm was experimentally achieved by rotating the BaGa4Se7 crystal.We simulate and experimentally demonstrate a feedback-based probabilistic constellation shaping (FB-PCS) of a 10 Gbaud 16-ary quadrature amplitude modulation (16-QAM) signal. Our approach is to adaptively modify the distribution of transmitted symbols based on errors at the receiver, and assumptions about the channel model are not required. Specifically, the error feedback enables solving an optimization problem to find the distribution that maximizes the mutual information between the input and output of the channel without knowledge of the channel itself. A known training sequence with uniform distribution is transmitted, and the errors at each constellation point are counted at the receiver. This information is relayed to the transmitter, which then updates the data constellation with a new probability distribution such that constellation points with more errors are used less frequently. We examine four different system scenarios in simulation and one scenario in experiment. In simulation, we find that FB-PCS (a) reduces the number of errors when compared to uniform shaping for the four scenarios, and (b) reduces symbol error rate (SER) by approximately an order of magnitude or has similar SER compared to conventional Maxwell-Boltzmann (M-B) shaping.
