Skin being the largest, superficial, easily accessible and assessable organ of the body, may be a promising target for gene therapy research in the recent future.The aim of the study was to evaluate the relationship between tumor mutational burden (TMB) and immune infiltration in ovarian cancer. We extracted somatic mutational data and gene expression profiles of ovarian cancer from The Cancer Genome Atlas (TCGA). The samples were separated into low and high TMB groups. Correlations between TMB and cancer prognosis were analyzed and immune cell infiltration in the high and low TMB subgroups was calculated using the CIBERSORT package software. High TMB was significantly related to an improved survival rate. We identified 4 TMB-related core genes that were significantly associated with prognosis. Furthermore, mutations in the 4 genes were associated with immune cell infiltration. We also found a high proportion of naive B cells and activated NK cells in the high TMB group, while increased proportions of memory B cells and plasma cells were found in the low TMB group. Overall, our study indicated that patients with a higher TMB level experienced a favorable survival outcome and this may influence immune infiltration in ovarian cancer. Furthermore, the 4 TMB-related core genes were highly correlated with prognosis and the level of immune cell infiltration.

The expression of miR-10a was detected by qRT-PCR. Target mRNA candidates were detected by bioinformatic analysis. Proliferative and migration capability of cSCC cell were examined by MTT assay, wound healing assay, and invasion assay, respectively. miR-10a expression was monitored in cSCC patients to elucidate the relationship between miR-10a expression and outcomes of cSCC.

In our study, we found that expression of miR-10a was significantly down-regulated in cSCC cell in vitro and in vivo. Moreover, our results revealed that SDC-1 was a likely target of miR-10a in regulating biologic function of cSCC cell. Additionally, miR-10a expression level was inversely correlated with cSCC cell differentiation and tumor progression.

These findings in this study indicate the importance of miR-10a in cSCC cell hallmarks and its use as a novel target for cSCC treatment.

It may form multiple tumors. When SP is combined with carcinoid, adenoma, or other tumors (although rare), diagnosis is more complicated. SP mixed with carcinoid tumor is rare. At present, only 4 cases have been reported in English literature. Here, we report a case of sclerosing pneumocytoma combined not only with carcinoid, but also with clear cell adenoma of the lung. The patient was a 52-year-old female and CT found a nodule in the middle lobe of the right lung. SP was not excluded by intraoperative frozen section diagnosis. The above 3 components formed a 1.4 cm nodule. The related literature is reviewed to strengthen the understanding of SP, and aid clinicopathological diagnosis.

By surgery, we completely removed the mass. Unlike previous literature reports, we found that the surface of the mass was not encapsulated, and subsequent pathologic reports confirmed that it was indeed a plexiform schwannoma. As of now, the patient did not have adverse reactions or postoperative recurrence.

We recommend a pathologic diagnosis of isolated tissues after they are completely removed during surgery. Furthermore, if the same ear has been operated on before, it may increase the difficulty of this operation.

Cholangiocarcinoma (CHOL) is one of the most fatal malignancies worldwide. PBRM1 is a tumor suppressor gene in diverse cancers. It regulates cell cycle, genomic stability, centromeric cohesion, and apoptosis. However, its relevance to remodel tumor cell immune response of PBRM1 in CHOL remains unclear.

PBRM1 mutation and expression of CHOL patients were analyzed by the TCGA database using R packages and cBioPortal site. The correlation between PBRM1 and tumor cell immune infiltrates among CHOL patients was investigated by TIMER2.0. Correlation analysis between PBRM1 and gene markers of tumor-infiltrating immune cells in CHOL was analyzed by GEPIA. Pathway enrichment analysis and protein-protein interaction network of PBRM1 mutation and expression was investigated using STRING and Cytoscape.

Among CHOL patients, PBRM1 has a high mutation probability and significant differential expression. Mutations and differential expression of PBRM1 both have a significant effect on the infiltration of cancer associated fibroblasts (CAF) in CHOL patients.
