Using an established PNS model, we tested whether crocin could have a rapid and persistent antidepressant-like effect in PNS ****. Growth hormone secretagogue receptor (GHSR) and phosphoinositide 3-kinase (PI3K) inhibitors were used to test their effects in antidepressant-like effect of crocin. Hippocampal GHSR-PI3K signaling was examined both in PNS **** treated with a single dose of crocin and in combination of GHSR inhibitor. PNS **** showed depression-like behaviors at juvenile and adulthood, and crocin induced an instant and persistent antidepressant-like response in PNS **** in a dose-dependent manner. Moreover, crocin increased the expression of hippocampal synaptic plasticity-associated proteins through the restoration of GHSR-PI3K signaling. Inhibitions of both GHSR and PI3K abolished the effect of crocin in alleviating depressive-like behaviors. More importantly, GHSR inhibitor JMV2959 blocked the enhanced expression of hippocampal plasticity-related proteins induced by crocin. The present study demonstrated that crocin induced a fast-onset and prolonged antidepressant effect in PNS **** and suggested that GHSR-PI3K signaling may play a key role in crocin's effect at least partially by a restoration of hippocampal synaptic plasticity-associated proteins.Metal-nitrogen-carbon (MNC) nanocomposites have been hailed as promising and efficient electrocatalysts toward oxygen reduction reaction (ORR), due to the formation of MNx coordination moieties. This limited accessibility compromises the electrocatalytic performance. Herein, we describe a wet-impregnation procedure by facile thermal refluxing, whereby palladium is atomically dispersed and enriched onto the surface of hollow, nitrogen-doped carbon cages (HNC) forming Pd-N coordination bonds. The obtained Pd-HNC nanocomposites exhibit an ORR activity in alkaline media markedly higher than that of metallic Pd nanoparticles, and the best sample even outperforms commercial Pt/C and relevant Pd-based catalysts reported in the literature. The results suggest that atomic dispersion and surface enrichment of palladium in a carbon matrix may serve as an effective strategy in the fabrication of high-performance ORR electrocatalysts.A flexible electronic-nose (E-nose) was constructed by assembling graphene oxide (GO) using different types of metal ions (Mx+) with different ratio of GO to Mx+. Owing to the cross-linked networks, the Mx+-induced assembly of graphene films resulted in different porous structures. A chemi-resistive sensor array was constructed by coating the GO-M hybrid films on PET substrate patterned with 8 interdigited electrodes, followed by in situ reduction of GO to rGO with hydrazine vapor. Each of the sensing elements on the sensor array showed a cross-reactive response toward different types of gases at room temperature. Compared to bare rGO, incorporation of metal species into rGO significantly improved sensitivity owing to the additional interaction between metal species and gas analyte. Principle component analysis (PCA) showed that four types of exhaled breath (EB) biomarkers including acetone, isoprene, ammonia, and hydrothion in sub-ppm concentrations can be discriminated well. To overcome the interference from humidity in EB, a protocol to collect and analyze EB gases was established and further validated by simulated EB samples. Data from mouse, zebrafish, Drosophila, and rat show that disruption of LPHN3 results in hyperactivity, and in the Sprague-Dawley Lphn3 knockout rat, exhibit deficits in learning and memory and changes in dopamine (DA) markers in the neostriatum. To determine the effects of Lphn3 deletion on DA neurotransmission, we compared the concentration, duration, and frequency of DA transients in KO and wild-type rats using fast-scan cyclic voltammetry in brain slices. Lphn3 KO rats showed higher release of DA, and the duration and interevent time were markedly decreased compared with wild-type rats. The data demonstrate that LPHN3 plays a heretofore unrecognized role in DA signaling and may represent a new target for small molecule regulation of DA neurotransmission with translational implications.
