Thirty-one children were eligible for inclusion, but five were excluded from the analysis (three were treated with a lower than recommended dose, and two were lost to follow-up). Of the 26 children included in the analysis, the treatment completion rate was 100%. Adverse drug reactions (ADRs) were reported in 38.5% (10/26) of the patients. The most common ADRs were gastrointestinal symptoms (19.2%,5/26), and all ADRs were rated as Grade 1. The 3HP regimen has a high completion rate, and it seems well tolerated in our study population. However, further randomized controlled clinical trial with larger sample size are warranted.

    This is a detailed MRI analysis of a prospective, monocenter study, including 49 consecutive patients (37 female, 12 male; median age 50) with symptoms due to endogenous hyperinsulinemic hypoglycemia (EHH) and mostly negative prior-imaging. All patients received a 3-T MRI and a 68Ga-DOTA-exendin-4-PET/CT. Pooled accuracy, sensitivity, specificity and inter-reader agreement were calculated.     CD155 is an immune checkpoint protein. Its overexpression is an indicator of poor prognosis in some types of cancer. However, the significance of CD155 expression in patients with triple-negative breast cancer, and the relationship between CD155 and programmed death-ligand 1 (PD-L1) expression, have not yet been analyzed in detail.

    Using immunohistochemical staining and tissue microarrays, we analyzed the expression profiles of CD155 and PD-L1 in 61 patients with triple-negative breast cancer. Relapse-free survival and overall survival rates were compared according to CD155 expression. The correlation between CD155 expression and clinicopathological factors, including PD-L1 expression (using SP142 and 73-10 assays), was also examined.

    CD155 expression was noted in 25 patients (41.0%) in this cohort. CD155 expression did not correlate with pathological stage, histological grade, Ki-67 labeling index, or stromal tumor-infiltrating lymphocytes. Only PD-L1 expression in tumor cells by SP142 assay significantly correlated with CD155 expression (p = 0.035); however, PD-L1 expression in tumor cells by 73-10 assay did not show a correlation (p = 0.115). Using the 73-10 assay, 59% of patients showed CD155 and/or PD-L1 expression in tumor cells. Moreover, using the SP142 assay, 63.3% of patients showed CD155 and/or PD-L1 expression in immune cells. CD155 expression did not correlate with either relapse-free survival or overall survival (p = 0.485 and 0.843, respectively).

    During the early months of the COVID-19 pandemic, mortality associated with the disease declined in the United States. The standard of care for pharmacological interventions evolved during this period as new and repurposed treatments were used alone and in combination. Though these medications have been studied individually, data are limited regarding the relative impact of different medication combinations. The objectives of this study were to evaluate the association of COVID-19-related mortality and observed medication combinations and to determine whether changes in medication-related practice patterns and measured patient characteristics, alone, explain the decline in mortality seen early in the COVID-19 pandemic.

    A retrospective cohort study was conducted at a multi-hospital healthcare system exploring the association of mortality and combinations of remdesivir, corticosteroids, anticoagulants, tocilizumab, and hydroxychloroquine. Multivariable logistic regression was used to identify predictors of mortality for both the overall population and the population stratified by intensive care and non-intensive care unit admissions.

It involves the material recovery and segregation stage (MRSS), organic extraction (pulping), thermophilic anaerobic digestion (AD), composting, effluent treatment plant (ETP), and biogas genset stages producing 11.90% recyclables, 33% refused derived fuel (RDF), 5% compost of total waste received, 70 m3/day recyclable water and 0.435 MWh/day electricity. The biogas and methane yield were 0.535 and 0.350 m3/kg VSadded (avg.), respectively, with 40% VS removal (avg total solids (TS) 10%). Less than 3% (inert) of total waste received was subjected to landfill disposal. The MBT plant's revenue generation is 995 US$ per day/148 tons ($ 6.72/ton) waste processed. The gross OPEX is 24 US$/ton making the net OPEX of 17 US$/ton (minus revenue), which could be considered as the excellent OPEX for MSW based MBT plants as per global benchmarks. Further, local usage of RDF can significantly reduce the OPEX to 14 US$/ton, as almost 16% of the OPEX goes towards RDF disposal to cement companies located at a distance of 200-500 km from the MBT plant site. As per LCA study, the total GHG emissions have been calculated to be -25.68 tons CO2 eq./100 tons MSW. The negative emissions result from the export of electricity, compost, and RDF as well as recycling of paper and plastic products. In addition to maximizing the supply of basin demands during the operation period, this model aims to reduce the salinity of the inflow to Shadegan Wetland. Due to the importance of the wetland as a seasonal habitat for birds and the importance of protecting its ecosystem, the development of a quantitative-qualitative optimization model for optimal use of available water resources is the aim of this study. First, based on current conditions, the prepared model is developed as a reference scenario for a future 30-year period(2021-2050). To achieve the best system efficiency in terms of quality and quantity, the optimization is performed by means of the NSGA-II algorithm. The results indicate that the optimizer model performs appropriately in supplying various demands and also decreasing the salinity of the inflow to Shadegan Wetland compared to the reference scenario so that in addition to supplying the demands with more than92% reliability in the whole system, it is expected that the salinity of the river at the entrance to Shadegan Wetland to be reduced by about50%., especially in low water months. The coupling model proposed in this research is applicable for other study areas with quantitative-qualitative operation approach and is able to detect critical points of rivers in terms of quantity and quality.   • The proposed method is tested on a self-collected dataset. The results demonstrate that proposed method achieves state-of-the-art performance in terms of DCs = 0.92±0.03 (condyles) and 0.90±0.04 (glenoid fossae), and MSDs =0.20±0.19 mm (condyles) and 0.19±0.08 mm (glenoid fossae).

    Spinal cord injury (SCI) has an immediate and devastating impact on the control over various movements and sensations. However, no effective therapies for SCI currently exist.

    To identify and analyze SCI subtypes, we obtained the expression profile data of the 1,057 genes (889 intersection genes) in GSE45550 using weighted gene co-expression network analysis (WGCNA), and 14 co-expression gene modules were identified. Next, we filtered out the network degree top 10 (degree >80) genes, considered the final key SCI genes. A multifactor regulatory network (105 interaction pairs), consisting of messenger RNAs (mRNAs), long non-coding RNAs (lncRNAs), and transcription factors (TFs) was constructed. This network was involved in the co-expression of key genes. We selected the top 10 regulatory factors (degree >4) as core regulators in the multifactor regulatory network.

    The results of functional enrichment analysis of the target gene expressing the core regulatory factor [1,059] showed that these target genes were enriched in pathways for human cytomegalovirus infection, chronic myeloid leukemia, and pancreatic cancer. Further, we used the key genes in the co-expression network to categorize the SCI samples in GSE45550. The expression levels of the top 6 genes (

    A retrospective analysis was conducted of 182 patients with T1-2 cancer with anterior vocal commissure (AVC) involvement. The local control (LC), disease-free survival (DFS) and overall survival (OS) rates at 5-year follow-up and the influencing factors were analyzed.

To identify and analyze SCI subtypes, we obtained the expression profile data of the 1,057 genes (889 intersection genes) in GSE45550 using weighted gene co-expression network analysis (WGCNA), and 14 co-expression gene modules were identified. Next, we filtered out the network degree top 10 (degree >80) genes, considered the final key SCI genes. A multifactor regulatory network (105 interaction pairs), consisting of messenger RNAs (mRNAs), long non-coding RNAs (lncRNAs), and transcription factors (TFs) was constructed. This network was involved in the co-expression of key genes. We selected the top 10 regulatory factors (degree >4) as core regulators in the multifactor regulatory network. Available control measures are not always successful, therapeutic options are limited, and there is no vaccine available against human leishmaniasis, although several candidate antigens have been evaluated over the last decades. Plenty of studies have aimed to evaluate the immune response development and a diverse range of host immune factors have been described to be associated with protection or disease progression in leishmaniasis; however, to date, no comprehensive biomarker(s) have been identified as surrogate marker of protection or exacerbation, and lack of enough information remains a barrier for vaccine development. Most of the current understanding of the role of different markers of immune response in leishmaniasis has been collected from experimental animal models. Although the data generated from the animal models are crucial, it might not always be extrapolated to humans. Here, we briefly review the events during Leishmania invasion of host cells and the immune responses induced against Leishmania in animal models and humans and their potential role as a biomarker of protection against human leishmaniasis.The aim of the study was to evaluate the pathogenic potential of Bacteroides pyogenes, rarely identified in clinical laboratories anaerobic bacteria. To increase the knowledge about this poorly understood anaerobic microorganism, the study also includes cases of infections described so far in the literature. Only the use of 16S rRNA sequencing and mass spectrometry technique allowed the identification of B. pyogenes from clinical specimens. We reported 13 severe human infections caused by B. pyogenes. Bacteria were cultured from the wound after biting by animals, chronic infections within the oral cavity, from patients with histologically or radiological proven osteomyelitis, surgical site infection, and from urine sample collected after a urological procedure. Most (9/13) of the patients required hospitalization. Almost 70% of them needed urgent admission via the emergency room. Two inpatients due to a life-threatening condition were admitted to the intensive care unit. Almost 50% of isolates were resistant to penicillin. Collaborating with student peers, four UFCWC faculty members have facilitated forms of peer support developed within service-user movements, while openly identifying experiences of reclaiming their own lives from the impacts of adversity, intense mental distress, and traumatizing responses of others to their distress. In the wake of the current pervasive health, economic, and social justice crises, professionals have a collective opportunity to recognize the human experience and rights of those suffering mental distress. Overall increased spontaneous firing patterns and altered coherence between the thalamic medial geniculate body (MGB) and auditory cortices is observed in animal models of tinnitus. It is likely that the functional connectivity between the MGB and primary and secondary auditory cortices is reduced in humans. Conversely, there are indications for increased connectivity between the MGB and several areas in the cingulate cortex and posterior cerebellar regions, as well as variability in connectivity between the MGB and frontal areas regarding laterality and orientation in the inferior, medial and superior frontal gyrus. We suggest that these changes affect adaptive sensory gating of temporal and spectral sound features along the auditory pathway, reflecting dysfunction in an extensive thalamo-cortical network implicated in predictive temporal adaptation to the auditory environment. The FTIR spectra of drug-loaded U and FU showed the presence of new carboxylic and aliphatic groups of OX and FA.   • Individuals with social anxiety disorder (SAD) experience significant interpersonal impairment. However, little is known about the physiological processes that are associated with interpersonal dysfunction in the disorder. In the present study we examined heart-rate (HR) synchrony in SAD during "getting-acquainted" interactions between opposite-sex partners. Participants included 118 individuals who formed 59 dyads 30 dyads that included one individual with SAD and one non-socially anxious (NSA) individual (SAD dyads) and 29 dyads that included two NSA individuals (control dyads). Dyads were randomly assigned to either a closeness-generating conversation or a small talk conversation. For closeness-generating conversations, we found that social anxiety was positively associated with HR synchrony in control dyads but negatively associated with HR synchrony in SAD dyads. These results remained when controlling for depressive symptoms and participants' movement. Our findings suggest that in more intimate social contexts, SAD may impair the ability to create HR synchrony between interlocuters and this can have negative relational consequences. Our findings are further discussed in the context of cognitive behavioral and interpersonal models of SAD, and clinical and research implications are delineated.Ammonia is an important environmental pollutant and can induce serious damages to the organs of aquatic animals, especially the intestine which is mostly exposed to external environment. Results showed that ammonia significantly changed the structure of intestines by decreasing the thickness of intestinal wall, shortening the length of intestinal villus, extending lamina proprias, and inducing inflammatory cells appearance when the turtles were exposed to ammonia (1.418 mg NH3 L-1) for 30 d. In addition, the downregulation of epithelial tight junction genes indicated that ammonia increased selective paracellular permeability. Simultaneously, the upregulation of cytokines suggested that ammonia induced intestinal immune and inflammatory responses. Furthermore, ammonia altered the dominant bacterial composition, and decreased the abundance of beneficial intestinal bacteria in the host. Our results demonstrated that ammonia impaired the intestinal health and changed the composition of residential microbiota in T. s. elegans. This study provides a new insight to evaluate the toxic effects of ammonia on aquatic turtles and helps to build a framework for the effective conservation of turtles.
    Colorectal cancer (CRC) is a kind of malignant tumor of digestive system severely affecting human health. The occurrence of CRC is a polygenic and multi-step complex process involving genetic and epigenetic alterations. Bulk mucosa and laser-captured microdissected intestinal epithelial cell (IEC) gene expression were analyzed by gene set enrichment and gene set variation analyses to identify significant pathways and cells, respectively, altered in CC.     A retrospective review was conducted of all patients undergoing immediate postmastectomy bilateral breast reconstruction for unilateral breast cancer between January 2008 and January 2019 at a single institution. Data were collected on patient demographics, cancer and adjuvant/neoadjuvant treatments, tumor, reconstruction, hospital stay, and complications. Complications were compared between the cancerous and the noncancerous breasts.

    One hundred sixty patients met the inclusion criteria of this study. An increasing number of women are choosing to undergo contralateral prophylactic mastectomy with immediate bilateral breast reconstruction. Operating on the contralateral noncancer side is not without its own set of risks. We sought to compare complication rates between the cancerous and contralateral prophylactic breasts. A retrospective review was conducted of all patients undergoing immediate postmastectomy bilateral breast reconstruction for unilateral breast cancer between January 2008 and January 2019 at a single institution. Data were collected on patient demographics, cancer and adjuvant/neoadjuvant treatments, tumor, reconstruction, hospital stay, and complications. Complications were compared between the cancerous and the noncancerous breasts. One hundred sixty patients met the inclusion criteria of this study. Of these 160 patients, 33 (20.6%) had complications (major and minor) only to the cancerous breast, 7 (4.4%) had complications only to the noncancerous breast, and 7 (4.4%) had bilaterac side when stratifying by specific complications. Although contralateral prophylactic mastectomy with immediate bilateral reconstruction is not without added risk when compared with a unilateral procedure, this study shows that the incidence of complications to the noncancerous breast is less than that to the cancerous breast. This information can be used to help counsel patients with unilateral breast cancer on their treatment options and associated risks.     It is hard to differentiate transient symptoms associated with infarction (TSI) from transient ischemic stroke (TIA) without MRI in the early onset. However, they have distinct clinical outcomes and respond differently to therapeutics. Therefore, we aimed to develop a risk prediction model based on the clinical features to identify TSI.

    We enrolled 230 consecutive patients with transient neurologic deficit in the Department of Neurology, Tongji University Affiliated Tenth People's Hospital from March 2014 to October 2019. All the patients were assigned into TIA group (DWI-negative) or TSI group (DWI-positive) based on MRI conducted within five days of onset. We summarized the clinical characteristics of TSI by univariate and multivariate analyses. And then, we developed and validated a nomogram to identify TSI by the logistic regression equation.

    Of the 230 patients, 41.3% were diagnosed with TSI. According to the multivariate analysis, four independent risk factors, including smoking history, low-density lipoprotein cholesterol, brain natriuretic peptide precursor, and ABCD3 score, were incorporated into a nomogram. We developed a predictive model named ABCD3-SLOPE. The calibration curve showed good agreement between nomogram prediction and observation. The concordance index (C-index) of the nomogram for TSI prediction was 0.77 (95% confidence interval, 0.70-0.83), and it was well-calibrated.

GO and KEGG analysis showed that proteins coded by the corresponding differentially expressed genes were mainly enriched in the mitochondrial oxidative respiratory chain, and the expression of three proteins (COX17, COX5B, ATP5J) was upregulated. The three proteins with higher scores were verified by Western blot. The apoptosis rate of the gastrin knockdown cancer cell was significantly increased; meanwhile, gastrin knockdown leads to increase of membrane potential and decrease of intracellular ROS production.   • Discovering new anticancer agents and analyzing their activities is a vital part of drug development, but it requires a huge amount of time and resources, leading to the increasing demands for more-effective techniques. Herein, a novel and simple cell-based electrochemical biosensor, referred to as a cytosensor, was proposed to investigate the electrochemical behavior of human skin malignant melanoma (SK-MEL28) cells and the anticancer effect of saponin on cell viability. To enhance both electrocatalytic properties and biocompatibility, gold nanoparticles were electrochemically deposited onto a conductive substrate, and poly-L-lysine was further added to the electrode surface. Electric signals from SK-MEL28 cells on the electrodes were obtained from cyclic voltammetry and differential pulse voltammetry. The cathodic peak current was proportional to the cell viability and showed a detection range of 2,880-40,000 cells per device with an excellent linear cell number-intensity relationship (R2= 0.9952). Furthermore, the anticancer effect of saponin on SK-MEL28 cells was clearly established at concentrations higher than 20 μM, which was highly consistent with conventional assays. Our results revealed that the productivity variables above- and belowground (i.e., net primary productivity, aboveground net primary productivity, belowground net primary productivity, total biomass, aboveground biomass, root biomass, gross ecosystem productivity, and net ecosystem productivity) were decreased across all ecosystems. However, drought did not significantly affect litter mass across all ecosystems, and the responses of above- and belowground productivity to drought were non-uniform. Furthermore, the responses of these productivity variables to drought were more pronounced with drought intensity and duration, and consistent across ecosystem types and background climates. Drought significantly decreased soil moisture, soil C concentrations, soil CN ratios, and microbial biomass C, whereas it enhanced soil pH values and fungibacteria ratios. Moreover, the negative effects of drought on above- and belowground productivity variables were correlated mostly with the response of soil pH to drought among all soil factors. Concentrations of NEOs (arithmetic means in fall and spring, respectively) in greenhouse were the highest (2.52×102 and 4.59×102 ng g-1), followed by in orchard (35.1 and 1.31×102 ng g-1), park (50.4 and 1.02×102 ng g-1), residential area (20.2 and 1.38×102 ng g-1) and farm (25.5 and 84.2 ng g-1). The contribution of individual NEO varied in soils of different land use types. Both IMI and THX were largely used in greenhouse, while IMI was the main NEO in the other four lands. The NEO levels in soils planted with different crops varied greatly. Extremely high levels of NEOs (>103 ng g-1) were observed in soils planted with watermelon, tomato and peach in greenhouse. The ubiquitous presence of NEOs in soils deserves more attention, particularly in greenhouse.In this study, the impact of long term exposure of elevated ozone (+20 ppb above ambient) on photosynthetic traits and anti-oxidative defense system of Leucaena leucocephala, a tree of great economic importance, was studied in a Free Air Ozone Concentration Enrichment (O3-FACE) facility at different time intervals (6, 12, 18, and 24 months). Results showed that net photosynthesis, photosynthetic pigments and lipid peroxidation were significantly reduced after 6, 12 and 24 months of exposure to elevated ozone (eO3) whereas stomatal conductance and transpiration rate were significantly decreased after 12 months of exposure to eO3. Antioxidant enzymatic activities (catalase, ascorbate peroxidase and glutathione reductase) were significantly increased after 12 months of exposure to eO3. Ascorbate was increased significantly after 6 and 12 months of exposure to eO3 while reduced glutathione content declined significantly after 6 and 24 months of exposure to eO3. The study showed that there were several negative long lasting physiological and biochemical responses in Leucaena.

The ubiquitous presence of NEOs in soils deserves more attention, particularly in greenhouse.In this study, the impact of long term exposure of elevated ozone (+20 ppb above ambient) on photosynthetic traits and anti-oxidative defense system of Leucaena leucocephala, a tree of great economic importance, was studied in a Free Air Ozone Concentration Enrichment (O3-FACE) facility at different time intervals (6, 12, 18, and 24 months). Results showed that net photosynthesis, photosynthetic pigments and lipid peroxidation were significantly reduced after 6, 12 and 24 months of exposure to elevated ozone (eO3) whereas stomatal conductance and transpiration rate were significantly decreased after 12 months of exposure to eO3. Antioxidant enzymatic activities (catalase, ascorbate peroxidase and glutathione reductase) were significantly increased after 12 months of exposure to eO3. Ascorbate was increased significantly after 6 and 12 months of exposure to eO3 while reduced glutathione content declined significantly after 6 and 24 months of exposure to eO3. The study showed that there were several negative long lasting physiological and biochemical responses in Leucaena.
