We previously identified ten linear immunodominant sites on the SARS-CoV-2 spike protein of which four are located within the RBD. Therefore, we designed two linkerimmunodominant site (LIS) vaccine candidates which are composed of four immunodominant sites within the RBD (RBD-ID) or all the 10 immunodominant sites within the whole spike (S-ID). They were administered by subcutaneous injection and were tested for immunogenicity and in vivo protective efficacy in a hamster model for COVID-19. We showed that the S-ID vaccine induced significantly better neutralizing antibody response than RBD-ID and alum control. As expected, hamsters vaccinated by S-ID had significantly less body weight loss, lung viral load, and histopathological changes of pneumonia. The synthesized compounds were identified by spectral studies such as IR, NMR and HRMS. Dielectric properties of the target compounds were experimentally determined by dielectric spectroscopy in the frequency range of 20 Hz - 1 MHz. The real part of the dielectric constant, dielectric loss tangent and conductivity of the samples were investigated as a function of applied frequency. Dielectric measurements showed Ata7 has the maximum dielectric constant at 1 kHz, while Ata1 has a negative dielectric constant value. When the result is evaluated with theoretical calculations, grain boundaries play an effective role in the experimental observed dielectric constant. Additionally, in this research the pyrrole-3-one derivatives (Ata1-9) were theoretically optimized and over these structures, NMR with GIAO (gauge-independent atomic orbital), UV with TD (time dependent), frontier orbitals (HOMO and LUMO), NLO (nonlinear optical properties) and MEP (molecular electrostatic potential) analysis were carried out.

We found co-occurrences of clusters of elevated concentrations of air pollutants such as PM2.5, high poverty indexes, and high TB incidence in Lima. These findings suggest an interplay of socio-economic and environmental in driving TB incidence.Interprofessional (IP) practice and education are important when seeking to respond to the growing demand for primary and preventive care services. Multiple professions with synergistic expertise are needed to effectively provide health promotion, disease prevention, and patient education and to help patients with multiple comorbidities, chronic health conditions, and care coordination. A recent study by Schor et al. titled, "Multidisciplinary work promotes preventive medicine and health education in primary care a cross-sectional survey," compares the implementation of preventive services in three primary care models. Higher rates of health services, patient education, and health outcomes were documented in two different models of care involving persons in multiple professions when compared with independent solo physicians' practices. In this commentary, we focus on the value of IP team-based care, continuing professional development, and the impact of the team on practice performance and health outcomes. Key components of effective IP teams include using consistent terminology to describe the team composition and function, team structures with purposeful selection of professions to address gaps in care, leadership support, and IP continuing professional development and education.OBJECTIVES Circulating tumour cells (CTCs) from liquid biopsies provide an exceptional opportunity to obtain real-time tumour information and are under current investigation in several cancers, including cutaneous melanoma, but face significant drawbacks in terms of non-standardised methodology, low viable cell numbers and accuracy of CTC identification. In this pilot study, we report that chemosensitivity assays using liquid biopsy-derived metastatic melanoma (MM) CTCs, from 7 patients with stage IIIC, BRAF wild-type metastatic melanomas, localized exclusively to the pelvic region, un-eligible for immunotherapy and treated with melphalan hypoxic pelvic perfusion (HPP), is both feasible and useful in predicting response to therapy. Viable MM CTCs (> 5 cells/ml for all 7 blood samples), enriched by transient culture, were characterised in flow cytometry-based Annexin V-PE assays for chemosensitivity to several drugs. RESULTS Using melphalan as a standard, chemosensitivity cut-off values of > 60% cell death, were predictive of patient RECIST 1.1 response to melphalan HPP therapy, associated with calculated 100% sensitivity, 66.67% specificity, 33.33% positive predictive, 100% negative predictive, and 71.43% accuracy values. We propose that the methodology in this study is both feasible and has potential value in predicting response to therapy, setting the stage for a larger study.   • Acha grains and sandpaper leaf are commonly used by locals for diabetes and hypertension management, respectively. However, this study sought to produce biscuits from acha-sandpaper leaf composite flour and evaluate their ameliorative potentials in streptozotocin (STZ)/Nω-nitro-l-arginine methylester hydrochloride (l-NAME)-induced hypertensive-diabetic rats. Group I (non-diabetic/normotensive), group II (negative control), group III (positive control) rats were placed on standard drugs, and groups IV-VII rats were placed on formulated biscuits supplemented with 0%, 1.25%, 2.5%, and 3.75% sandpaper leaf flour. The fasting blood glucose (FBG) and blood pressure (BP) of the experimental rats were measured. In addition, the activities of α-amylase, α-glucosidase, and angiotensin-1-converting enzyme (ACE) as well as antioxidant status were evaluated in all the groups and compared. A 9-year-old male castrated mixed dog presented with an episodic history of nonweight-bearing thoracic limb lameness. Additional clinical signs included an atrophied thoracic limb with cool paw pads and painful axillary region. Magnetic resonance imaging, computed tomography, ultrasound, and exploratory surgery confirmed a chronic thrombus of the right brachial artery. No underlying cause for the thrombus was identified. The dog has been successfully managed on long-term rivaroxaban and clopidogrel. Follow-up ultrasound of the thrombus suggested early remodeling.
    Acha grains and sandpaper leaf are commonly used by locals for diabetes and hypertension management, respectively. However, this study sought to produce biscuits from acha-sandpaper leaf composite flour and evaluate their ameliorative potentials in streptozotocin (STZ)/Nω-nitro-l-arginine methylester hydrochloride (l-NAME)-induced hypertensive-diabetic rats. Group I (non-diabetic/normotensive), group II (negative control), group III (positive control) rats were placed on standard drugs, and groups IV-VII rats were placed on formulated biscuits supplemented with 0%, 1.25%, 2.5%, and 3.75% sandpaper leaf flour. The fasting blood glucose (FBG) and blood pressure (BP) of the experimental rats were measured.     The purposes of this study are to outline trends in the prevalence of disability among Australian adults and to analyse the relationship between obesity, and physical activity with disability.

    A retrospective longitudinal research design.

    The study utilized the most recent 14 waves (wave 6 through 19) of the nationally-representative Household, Income and Labour Dynamics in Australia (HILDA) survey (2006-2019). The Generalized Estimating Equation (GEE) with the logistic link function model was employed to estimate the relationships between obesity and physical activity with disability. The final study sample consisted of 189,519 person-year observations from 26,208 participants.

    The pooled prevalence of disability in adults is 28%. The prevalence of disability among older adults (65 and above years) is more than 50%, irrespective of gender. Further, it identifies obesity and physical activity as risk and protective factors of disability for adults, respectively. The odds of acquisition of a disability was 1.33 times (Odds Ratios [OR] 1.33, 95% Confidence Interval [CI] 1.28-1.39) higher among obese adults than healthy weight counterparts. However, adults undertaking recommended level of physical activity (more than thrice a week to everyday) per week have 17% (OR 0.83, 95% CI 0.81-0.85) lower chances of disability acquisition.

    Our purpose was to establish a noninvasive quantitative method for assessing intracranial pressure (ICP) levels in patients with traumatic brain injury (TBI) through investigating the Hounsfield unit (HU) features of computed tomography (CT) images.

    In this retrospective study, 47 patients with a closed TBI were recruited. Hounsfield unit features from the last cranial CT and the initial ICP value were collected. Three models were established to predict intracranial hypertension with Hounsfield unit (HU model), midline shift (MLS model), and clinical expertise (CE model) features.

    The HU model had the highest ability to predict intracranial hypertension. In 34 patients with unilateral injury, the HU model displayed the highest performance. In three classifications of intracranial hypertension (ICP ≤ 22, 23-29, and ≥30 mmHg), the HU model achieved the highest F1 score.

    This radiological feature-based noninvasive quantitative approach showed better performance compared with conventional methods, such as the degree of midline shift and clinical expertise. The results show its potential in clinical practice and further research.

Group 1 was the fruit pulp (5g) fermented in 45mL of MRS-B medium for 24h. Group 2 was the fruit pulp wrapped in banana leaves for 3days before fermented in MRS-B for 24h. Group 3 was the fruit pulp wrapped in banana leaves for 3days before fermentation in MRS-B for 48h. Fermentation broth of each condition was taken and then filtered using millipore (0.2µm). As many as 50µL of filtrates was tested for its inhibitory activity against

The aims of this study were to analyze the antibiotic use and factors affected to the quality of antibiotic use in PHCC in Banjarbaru City, South Kalimantan, Indonesia.

Benefits of WALANT include a bloodless field, improved recovery, on-table testing, as well as cost and time savings. Whilst more clinical trials are underway to fully elucidate safety-profile and outcomes, there is a lack of consensus and clarity over contraindications to WALANT. A survey of trainees identified that only one-in-five were aware of the appropriate reversal agent in the event of inadequate perfusion.     This study examined cross-sectional associations among device-measured sedentary and physical activity behavior with perceived sleep quality in adults with MS.

    Adults with MS (N=290) completed the Pittsburgh Sleep Quality Index (PSQI) and wore an accelerometer for seven days providing a measure of time spent in sedentary behavior, light physical activity (LPA), and moderate-to-vigorous physical activity (MVPA) using MS-specific cut-points. We conducted multiple linear regression analysis to identify the independent contributions of variables for explaining PSQI scores.

    The overall model accounted for 2% of the variance in global PSQI scores, and MVPA was significantly and independently associated with global PSQI scores (β=─0.123; p=0.045; partial r=─0.118) when accounting for average wear time, sedentary behavior, and time spent in LPA. There were no other significant associations with PSQI global score.

    Our results suggest that time spent in MVPA may be associated with better sleep quality in adults with MS, but adults with MS do not spend sufficient time in physical activity. Researchers should evaluate these relationships in longitudinal study designs and behavior change interventions, as physical activity may provide a unique opportunity to improve sleep quality outcomes.

Benefits of WALANT include a bloodless field, improved recovery, on-table testing, as well as cost and time savings. Whilst more clinical trials are underway to fully elucidate safety-profile and outcomes, there is a lack of consensus and clarity over contraindications to WALANT. A survey of trainees identified that only one-in-five were aware of the appropriate reversal agent in the event of inadequate perfusion. We feel that a WALANT checklist should be developed and implemented for use immediately prior to administration of local anaesthetic with adrenaline to an extremity, building on the successes of the World Health Organisation (WHO) and the Royal College of Anaesthetists checklists. Significant effort has been spent on identifying these variations and their impact on transmission, virulence and pathogenicity of SARS-CoV-2. Binding of the SARS-CoV-2 spike protein to the angiotensin converting enzyme 2 (ACE2) promotes cellular entry. Therefore, human ACE2 variations could also influence susceptibility or resistance to the virus. A deeper understanding of the evolution and genetic variations in SARS-CoV-2 as well as ACE2 could contribute to the development of effective treatment and preventive measures. Here, we review the literature on SARS-CoV-2 and ACE2 variations and their role in COVID-19. There is a fourfold higher prevalence of sleep problems in multiple sclerosis (MS) than the general population. This study examined cross-sectional associations among device-measured sedentary and physical activity behavior with perceived sleep quality in adults with MS. Adults with MS (N=290) completed the Pittsburgh Sleep Quality Index (PSQI) and wore an accelerometer for seven days providing a measure of time spent in sedentary behavior, light physical activity (LPA), and moderate-to-vigorous physical activity (MVPA) using MS-specific cut-points. We conducted multiple linear regression analysis to identify the independent contributions of variables for explaining PSQI scores. The overall model accounted for 2% of the variance in global PSQI scores, and MVPA was significantly and independently associated with global PSQI scores (β=─0.123; p=0.045; partial r=─0.118) when accounting for average wear time, sedentary behavior, and time spent in LPA. There were no other significant associations with PSQI global score. Our results suggest that time spent in MVPA may be associated with better sleep quality in adults with MS, but adults with MS do not spend sufficient time in physical activity.   • Glioblastoma (GBM) is the most lethal and frequent type of brain tumor, leading patients to death in approximately 14 months after diagnosis. GBM treatment consists in surgical removal followed by radio and chemotherapy. However, tumors commonly relapse and the treatment promotes only a slight increase in patient survival. Thus, uncovering the cellular mechanisms involved in GBM resistance is of utmost interest, and the use of cell lines has been shown to be an extremely important tool. In this work, the exploration of RNAseq data from different GBM cell lines revealed different expression signatures, distinctly correlated with the behavior of GBM cell lines regarding proliferation indexes and radio-resistance. U87MG and U138MG cells, which presented expressively reduced proliferation and increased radio-resistance, showed a particular expression signature encompassing enrichment in many extracellular matrix (ECM) and receptor genes. Contrasting, U251MG and T98G cells, that presented higher proliferation and gs suggest the clinical relevance of ECM-receptor genes signature found here for radiotherapy decision and as biomarkers of glioma prognosis.
    In certain malignancies, patients with oligometastatic disease benefit from radical ablative or surgical treatment. The SABR-COMET trial demonstrated a survival benefit for oligometastatic patients randomized to local stereotactic ablative radiation (SABR) compared to patients receiving standard care (SC) alone. Our aim was to determine the cost-effectiveness of SABR.

    A decision model based on partitioned survival simulations estimated costs and quality-adjusted life years (QALY) associated with both strategies in a United States setting from a health care perspective. Analyses were performed over the trial duration of six years as well as a long-term horizon of 16 years. Model input parameters were based on the SABR-COMET trial data as well as best available and most recent data provided in the published literature. An annual discount of 3% for costs was implemented in the analysis. All costs were adjusted to 2019 US Dollars according to the United States Consumer Price Index. SABR costs were reported wiancers and represents an intermediate- and long-term cost-effective treatment strategy.

We report the case of a 24-year-old patient with chemotherapy-refractory metastatic IMT harboring ROS1 kinase fusion, who experienced a significant clinical and pathological response to crizotinib. This clinical case highlights the need to assess all patients with unresectable IMTs for chromosomal abnormalities and gene mutations and address them to targeted agents as well as clinical trials.

Our study aims to investigate whether there is a link between pretreatment hemoglobin (Hb) levels and survival to immunotherapy in patients with AGC and MGC.

    We retrospectively reviewed patients with AGC or ****treated at the oncology department of the Chinese PLA general hospital receiving PD-1 inhibitor. The Propensity Score Matching (PSM) (11) was performed to balance potential baseline confounding factors. Progression-free survival (PFS) and overall survival (OS) was analyzed among different Hb level (normal Hb group and decreased Hb group). Objective response rate (ORR), disease control rate (DCR) were also analyzed. Univariate analysis and multivariate analysis were performed further to validate the prognostic value of Hb level.

    We included 137 patients with AGC and ****who received PD-1 inhplus anti-angiogenic therapy with respect to PFS (10.3m vs 2.8m, HR 95% CI 0.37(0.15, 0.95), P=0.031) and OS(15m vs 5.7m, HR 95% CI 0.21 (0.08, 0.58), P=0.001).

To date, the proteome profile of **** from buffalo origin is not available. In the present study, we have profiled in-depth proteome of in vitro cultured buffalo **** (BuMECs) during active proliferation using high throughput tandem mass spectrometry (MS). MS analysis identified a total of 8330, 5970, 5289, 4818 proteins in four sub-cellular fractions (SCFs) that included cytosolic (SCF-I), membranous and membranous organelle's (SCF-II), nuclear (SCF-III), and cytoskeletal (SCF-IV). However, 792 proteins were identified in the conditioned media, which represented the secretome. Altogether, combined analysis of all the five fractions (SCFs- I to IV, and secretome) revealed a total of 12,609 non-redundant proteins. The KEGG analysis suggested that these proteins were associated with 325 molecular pathways. Some of the highly enriched molecular pathways observed were metabolic, MAPK, PI3-AKT, insulin, estrogen, and cGMP-PKG signalling pathway. In this paper, GS of a Ti-6Al-4V alloy is generated by pre-torsion and characterized by electron backscatter diffraction. Then the VHCF behavior of the GS specimen is studied. The fractography and synchrotron radiation X-ray microtomography presented detailed characteristics of the internal crack initiation region in VHCF of the titanium alloy with GS. The results indicated that, in contrast to the low- and high-cycle regimes, the VHCF strength is reduced for the specimens with GS. Thus, the GS induced by pre-torsion cannot enhance the VHCF strength of the titanium alloy. This implies that VHCF test (property) is an important consideration for the microstructural designed materials. We measured hypoglycemia-induced changes in endothelial parameters, oxidative stress markers and inflammation at baseline and after a 24-hour period in type 2 diabetic (T2DM) subjects versus age-matched controls. Case-control study 10 T2DM and 8 control subjects. Blood glucose was reduced from 5 (90 mg/dl) to hypoglycemic levels of 2.8 mmol/L (50 mg/dl) for 1 hour by incremental hyperinsulinemic clamps using baseline and 24 hour samples. Measures of endothelial parameters, oxidative stress and inflammation at baseline and at 24-hours post hypoglycemia were performed proteomic (Somalogic) analysis for inflammatory markers complemented by C-reactive protein (hsCRP) measurement, and proteomic markers and urinary isoprostanes for oxidative measures, together with endothelial function. Between baseline and 24 -hours after hypoglycemia, 15 of 140 inflammatory proteins differed in T2DM whilst only 1 of 140 differed in controls; all returned to baseline at 24-hours. However, elevated hsCRP levels were seen at 24-hours in T2DM (2.4 mg/L (1.2-5.4) vs. 3.9 mg/L (1.8-6.1), Baseline vs 24-hours, P  less then  0.05). In patients with T2DM, between baseline and 24-hour after hypoglycemia, only one of 15 oxidative stress proteins differed and this was not seen in controls. In this paper, GS of a Ti-6Al-4V alloy is generated by pre-torsion and characterized by electron backscatter diffraction. Then the VHCF behavior of the GS specimen is studied. The fractography and synchrotron radiation X-ray microtomography presented detailed characteristics of the internal crack initiation region in VHCF of the titanium alloy with GS. The results indicated that, in contrast to the low- and high-cycle regimes, the VHCF strength is reduced for the specimens with GS. Thus, the GS induced by pre-torsion cannot enhance the VHCF strength of the titanium alloy. The graphical abstract is available in Supplementary information.Intensive diabetes control has been associated with increased mortality in type 2 diabetes (T2DM); this has been suggested to be due to increased hypoglycemia. We measured hypoglycemia-induced changes in endothelial parameters, oxidative stress markers and inflammation at baseline and after a 24-hour period in type 2 diabetic (T2DM) subjects versus age-matched controls. Case-control study 10 T2DM and 8 control subjects. Blood glucose was reduced from 5 (90 mg/dl) to hypoglycemic levels of 2.8 mmol/L (50 mg/dl) for 1 hour by incremental hyperinsulinemic clamps using baseline and 24 hour samples. Measures of endothelial parameters, oxidative stress and inflammation at baseline and at 24-hours post hypoglycemia were performed proteomic (Somalogic) analysis for inflammatory markers complemented by C-reactive protein (hsCRP) measurement, and proteomic markers and urinary isoprostanes for oxidative measures, together with endothelial function. Between baseline and 24 -hours after hypoglycemia, 15 of 140 inflammatory proteins differed in T2DM whilst only 1 of 140 differed in controls; all returned to baseline at 24-hours. However, elevated hsCRP levels were seen at 24-hours in T2DM (2.4 mg/L (1.2-5.4) vs. 3.9 mg/L (1.8-6.1), Baseline vs 24-hours, P  less then  0.05). In patients with T2DM, between baseline and 24-hour after hypoglycemia, only one of 15 oxidative stress proteins differed and this was not seen in controls. An increase (P = 0.016) from baseline (73.4 ng/mL) to 24 hours after hypoglycemia (91.7 ng/mL) was seen for urinary isoprostanes. Hypoglycemia resulted in inflammatory and oxidative stress markers being elevated in T2DM subjects but not controls 24-hours after the event.
