  • Humans are strongly affected by social exclusion, a multifaceted and complex phenomenon of social life. #link# However, individuals tend to respond differently depending on a multitude of individual and contextual factors. Firstly, with a view to increasing the ecological validity and experimental control of an exclusion manipulation in the laboratory setting, we made use of immersive virtual environment technology (IVET; an Oculus Rift Virtual Reality headset) to create a new exclusion paradigm. Secondly, given that a recent meta-analytic report on reflexive responses (i.e., affect and physiology) to manipulations of exclusion in the laboratory setting cites inconsistencies across findings (Blackhart et al., 2009), we focused on the form of exclusion manipulated to illustrate how this factor may help to explain the divergences in responses. We thus investigated how explicit and implicit forms of social exclusion may have a differential impact on self-reported affect, as well as on electrodermal and cardiovascular activity. Results from this laboratory study conducted with a varied sample of the local population made salient the affordances of IVET as a tool in exclusion research. They also helped to reconcile the conflicting findings in the literature relating to differences in the level of negative affect generated and shed light on physiological arousal in the wake of being excluded in different ways.Sleep is a fundamental property conserved across species. The homeostatic induction of sleep indicates the presence of a mechanism that is progressively activated by the awake state and that induces sleep. Several lines of evidence support that such function, namely, sleep need, lies in the neuronal assemblies rather than specific brain regions and circuits. However, the molecular mechanism underlying the dynamics of sleep need is still unclear. This review aims to summarize recent studies mainly in rodents indicating that protein phosphorylation, especially at the synapses, could be the molecular entity associated with sleep need. Genetic studies in rodents have identified a set of kinases that promote sleep. The activity of sleep-promoting kinases appears to be elevated during the awake phase and in sleep deprivation. Furthermore, the proteomic analysis demonstrated that the phosphorylation status of synaptic protein is controlled by the sleep-wake cycle. Therefore, a plausible scenario may be that the awake-dependent activation of kinases modifies the phosphorylation status of synaptic proteins to promote sleep. The review was conducted according to PRISMA-statement through searches in the scientific database PsychINFO, PsycARTICLES, MEDLINE, and PUBMED. Studies included in this review had at least one bilingual and monolingual group, participants aged between 5 and 17 years, and at least one executive function measure. Studies on second language learners, multilingual people, and the clinical population were excluded. Fifty-three studies were included in the systematic review. Evidence supporting the bilingual effect seems to appear when assessing inhibition and cognitive flexibility, but to disappear when working memory is considered. The inconsistent results of the studies do not allow drawing definite conclusions on the bilingual effect. Further studies are needed; they should consider the role of some modulators (e.g., language history and context, methodological differences) on the observed results.Solving problems that are perceptually dissimilar but require similar solutions is a key skill in everyday life. In adults, this ability, termed analogical transfer, draws on memories of relevant past experiences that partially overlap with the present task at hand. Thanks to this support from long-term memory, analogical transfer allows remarkable behavioral flexibility beyond immediate situations. However, little is known about the interaction between long-term memory and analogical transfer in development as, to date, they have been studied separately. Here, for the first time, effects of age and memory on analogical transfer were investigated in 2-4.5-olds in a simple tool-use setup. Children attempted to solve a puzzle box after training the correct solution on a different looking box, either right before the test or 24 h earlier. We found that children (N = 105) could transfer the solution regardless of the delay and a perceptual conflict introduced in the tool set. For children who failed to transfer (N = 54) and repeated the test without a perceptual conflict, the odds of success did not improve. Our findings suggest that training promoted the detection of functional similarities between boxes and, thereby, flexible transfer both in the short and the long term.
    Studies on autistic strengths are often focused on what they reveal about autistic intelligence and, in some cases, exceptional and atypical reasoning abilities. An emerging research trend has demonstrated how interests and strengths often evident in autism can be harnessed in interventions to promote the well-being, adaptive, academic and professional success of autistic people. However, abilities in certain domains may be accompanied by major limitations in others, as well as psychiatric and behavioral issues, which may challenge their inclusion in support programs.

    To provide an in-depth, pragmatic, real-life example of the psychological and psychiatric management of interests and strengths in an autistic adolescent.

    An autistic teenager, C.A., with above-average calendar calculation and musical abilities, received psychiatric, neuropsychological, and language standardized and clinical assessments, combined with a measurement of his musical and calendar calculation abilities. C.A. and his parents then received psychiatric and psychological support over a 14-month period, targeting their perceptions of C.

  • Sensors based on fluorogenic RNA aptamers have emerged in recent years. These sensors have been used for in vitro and intracellular detection of a broad range of biological and medical targets. However, the potential application of fluorogenic RNA-based sensors for point-of-care testing is still little studied. Here, we report a paper substrate-based portable fluorogenic RNA sensor system. Target detection can be simply performed by rehydration of RNA sensor-embedded filter papers. This affordable sensor system can be used for the selective, sensitive, and rapid detection of different target analytes, such as antibiotics and cellular signaling molecules. We believe that these paper-based fluorogenic RNA sensors show great potential for point-of-care testing of a wide range of targets from small molecules, nucleic acids, proteins, to various pathogens.A rapid analytical procedure is proposed for determining two antimicrobial onion organosulfur compounds, propyl disulfide (PDS) and propyl propane thiosulfonate (PTSO), in animal feed. The use of PTSO as a natural ingredient in animal feed is allowed due to its antimicrobial activity against pathogenic organisms. Two analytical methodologies using gas chromatography coupled to mass spectrometry (GC-MS) are compared. After the extraction of the compounds from animal feed with acetonitrile, dispersive solid phase extraction (DSPE) as a cleaning stage with C18, or dispersive liquid-liquid microextraction (DLLME), using 100 μL of CHCl3, was tried. Both the methods were validated using a pig feed sample and the best results were achieved by DLLME. This technique provided cleaner extracts, five-times greater linear ranges and lower detection limits than simple cleaning due to the enrichment factor achieved. The relative standard deviation decreased from 22% with DSPE to 13% with DLLME. The usefulness of the DLLME-GC-MS methodology was tested by analysing 10 different samples of chicken, calf, hen, *** and fish feed. The concentrations of PDS were in the 0.1-1.7 μg g-1 range and those of PTSO were between 0.09 and 2.1 μg g-1.A constant magnetic field (CMF) was used to arrange the orientation of graphene oxide (GO) which was modified on a self-made screen-printed electrode. We evaluated the efficiency of this method for potential analytical application towards the sensing of hydrogen peroxide (H2O2). Mesoporous silica (MS)-encapsulated horseradish peroxidase (HRP) was immobilized on the electrode with vertically arranged GO to construct an H2O2 sensor (denoted as CMF/GO/HRP@MS). The linear range of the response of the CMF/GO/HRP@MS sensor to H2O2 was 0.1-235 μM, and the detection limit was as low as 0.01 μM. #link# The results demonstrated that the vertical arrangement of GO resulting from the CMF on the electrode surface could increase the electron transfer rate. The excellent selectivity and anti-interference ability of this sensor to H2O2 in physiological samples may be attributed to the synergistic effect of mesoporous silica, GO and constant magnetic field.Bovine antibodies, such as immunoglobulin G (IgG), cannot pass the placental barrier and as such are not transferred from the mother to the foetus, in utero. Instead a calf must absorb antibodies following ingestion of colostrum postpartum. Thus, continuous early monitoring of IgG absorption in a calf, within the first 6 to 12 hours of life, is imperative to allow faster treatment and prevent FPT. Importantly, it was observed that the magnitude of the effect was shown to vary with the length of the linking chain. To investigate the origins of these solvent effects, temperature dependent kinetic studies were performed to obtain activation parameters at high and low mole fractions of ionic liquid. The observed activation parameters showed the rate constant enhancement was due to interaction of the ionic liquid with the starting materials, consistent with previous results. Significantly, however, these data also showed that the balance of enthalpic and entropic effects varied dramatically with the length of the alkyl chain between the cationic centres.Herein, gold@platinum (Au@Pt) bimetallic nanoparticles with high catalytic ability were in situ decorated onto a molybdenum disulfide (MoS2) surface to obtain nanocomposites (MoS2-Au@Pt) with high peroxidase-mimicking activity, which were used to construct a colorimetric sensor for cysteine (Cys) detection. Interestingly, this sensor can efficiently distinguish Cys from homocysteine (Hcy), glutathione (GSH) and 19 other amino acids with high sensitivity. As expected, the colorimetric sensor can determine the Cys content in Cys supplement tablets due to its high stability and repeatability. Finally, the detection mechanism was studied.Herein, we have synthesized successfully a three-dimensional/two dimensional (3D/2D) core-shell Cu2O@Fe-Ni3S2 nanoflower on copper foam at room temperature. Remarkably, by virtue of rich active sites and vacancies, large surface area, high conductivity and close contact with the electrolyte, the Cu2O@Fe-Ni3S2 catalyst exhibits superior stability and oxygen evolution reaction performance.

  • Although the interaction between tumors and tumor-associated macrophages (TAMs) has been reported to facilitate the targeted drug resistance and progression of clear cell renal cell carcinoma (ccRCC), the related mechanisms remain unknown. Here, we report that SOX17 serves as a novel tumor suppressor in ccRCC and a positive regulatory loop, SOX17low/YAP/TEAD1/CCL5/CCR5/STAT3, facilitates the ccRCC-TAM interaction. SOX17 expression was commonly downregulated and negatively correlated with TAM infiltration in ccRCC specimens, and the integration of SOX17 and TAMs with the existing clinical indicators TNM stage or SSIGN score achieved better accuracy for predicting the prognosis of ccRCC patients. Mechanistically, SOX17 knockdown activated YAP signaling by promoting the transcription and nuclear distribution of YAP, which recruited TEAD1 to trigger CCL5 transcription. Then, CCL5 educated macrophages toward TAMs, which reciprocally enhanced ccRCC progression through CCL5/CCR5 and activated STAT3/SOX17low/YAP. However, SOX17 overexpression in ccRCC achieved the opposite effect. Thus, a positive regulatory loop, SOX17low/YAP/TEAD1/CCL5/CCR5/STAT3, was identified in the ccRCC-TAM interaction. Furthermore, targeting tumor-TAM interactions by blocking this positive regulatory network impaired the metastasis and targeted drug resistance of ccRCC in in vivo mouse models of lung metastasis and orthotopic ccRCC. These findings provide a new mechanism underlying the tumor-TAM interplay in ccRCC progression and present a potential target for inhibiting targeted drug resistance and metastasis in advanced ccRCC.
    Diabetic retinopathy (DR) is related to oxidative stress and insufficient intake of dietary antioxidants may be associated with the onset and progression of DR. This study aimed to detect the association between main dietary antioxidants intake and the risk for DR.

Four hundred and fifty-five subjects with type 2 diabetes were recruited and divided into diabetic patients without retinopathy (DWR) group and DR group based on their retinal status. CSMO (clinically significant macular oedema) was diagnosed by stereoscopic photography. Demographic and lifestyle characteristics were ascertained by questionnaire. General physical and ophthalmic examinations were completed for all subjects. Dietary antioxidants were assessed by 3-day food records. Subjects who have taken any type of vitamin supplements were excluded from the study. The association of dietary antioxidants with the risk for DR was analysed by logistic regression with adjustment of other factors. The dietary antioxidants levels of the CSMO subjects and non-CSMO subjects were compared using the Wilcoxon rank sum test.

    One hundred and nineteen subjects in DR group and 336 subjects in DWR group participated in the study. Only ten DR subjects had CSMO. The results showed that higher vitamin E (OR (95% CI)0.97 (0.95, 1.00), P = 0.036) and selenium (OR (95% CI)0.98 (0.96, 1.00), P = 0.017) intake appear to be the protective factors of DR. The dietary antioxidants levels of CSMO and non-CSMO subjects had no statistical differences (P > 0.05).

We therefore studied 937 patients with AML in complete remission transplanted using a reduced intensity conditioning regimen from an unrelated donor mismatched at a single allele. In a multivariate analysis, patient age, adverse karyotype and patient cytomegalovirus (CMV) seropositivity were correlated with decreased leukaemia free survival (LFS) and overall survival (OS). There was no significant difference in LFS or OS between patients transplanted from donors mismatched at HLA-A, -B, -C or -DRB1 in comparison to a HLA-DQB1 mismatched transplant. In a multivariate analysis, patients transplanted with a HLA-A mismatched donor had higher rates of acute graft-versus-host disease (GVHD) and non-relapse mortality (NRM) than patients transplanted with a HLA-DQB1 mismatched donor. Therefore, deep subsurface environments that combine these stressors, potentially including the subsurface of Mars, may be more habitable than previously thought.
    Although the interaction between tumors and tumor-associated macrophages (TAMs) has been reported to facilitate the targeted drug resistance and progression of clear cell renal cell carcinoma (ccRCC), the related mechanisms remain unknown. Here, we report that SOX17 serves as a novel tumor suppressor in ccRCC and a positive regulatory loop, SOX17low/YAP/TEAD1/CCL5/CCR5/STAT3, facilitates the ccRCC-TAM interaction. SOX17 expression was commonly downregulated and negatively correlated with TAM infiltration in ccRCC specimens, and the integration of SOX17 and TAMs with the existing clinical indicators TNM stage or SSIGN score achieved better accuracy for predicting the prognosis of ccRCC patients. Mechanistically, SOX17 knockdown activated YAP signaling by promoting the transcription and nuclear distribution of YAP, which recruited TEAD1 to trigger CCL5 transcription. Then, CCL5 educated macrophages toward TAMs, which reciprocally enhanced ccRCC progression through CCL5/CCR5 and activated STAT3/SOX17low/YAP. However, SOX17 overexpression in ccRCC achieved the opposite effect. Thus, a positive regulatory loop, SOX17low/YAP/TEAD1/CCL5/CCR5/STAT3, was identified in the ccRCC-TAM interaction. Furthermore, targeting tumor-TAM interactions by blocking this positive regulatory network impaired the metastasis and targeted drug resistance of ccRCC in in vivo mouse models of lung metastasis and orthotopic ccRCC. These findings provide a new mechanism underlying the tumor-TAM interplay in ccRCC progression and present a potential target for inhibiting targeted drug resistance and metastasis in advanced ccRCC. The association of dietary antioxidants with the risk for DR was analysed by logistic regression with adjustment of other factors.   • Biofilm formation is important for establishing plants-microbe associations. The role of calcium on biofilm formation has been studied in many bacteria except rhizobia. In this study, we investigated the role of calcium for biofilm formation in Azorhizobium caulindans, which forms nodules in the stem and root of its host plant Sesbania rostrata. We found that calcium is essential for A. caulindans biofilm formation, in addition to the presence of extracellular matrix components, eDNA and proteins. Also, calcium-mediated biofilm formation was tested with chemotaxis, motility, cyclic di-GMP synthesis, and quorum sensing mutants. Finally, calcium was found to promote S. rostrata root colonization of A. caulinodans. In total, these results show that calcium is essential for A. caulindans biofilm formation, and it affects the interaction between A. caulinodans and host plant.The intertidal zone often has varying levels of environmental stresses (desiccation, temperature, light) that result in highly stress-tolerant macrobiota occupying the upper zone while less tolerant species occupy the lower zone, but little comparative information is available for intertidal bacteria. As the soil microbial community is also affected by various environmental factors, such as pH, carbon availability, and soil moisture, we determined the total carbon (TC), total nitrogen (TN), and total phosphorus (TP) contents in the rhizoplane, rhizosphere, and bulk soil compartments to explore the associations between soil fungal communities and host plant characteristics. The differences between the soil fungal communities of TG and WT plants were detected. The alpha diversity of TG fungal communities was more correlated to environmental factors than the beta diversity. The abundance of operational taxonomic units (OTUs) enriched in TG root-related area was far more than that in the root-related area of WT plants. We first characterised the effect of C availability during free-living stage growth on fungal phenotype colonies growth and physiology (plasma membrane proton pumps, stable isotopic signatures, and potential extracellular decomposing enzymes). The effect of the C availability during the free-living stage of the PGPM was evaluated on wheat. We observed that C availability during the free-living stage regulated S. indica's growth, ultrastructure and physiology, resulting in two distinct colony phenotypes compact and explorer. The compact phenotype developed at low C, used peptone as the major C and N source, and displayed higher decomposing potential for C providing substrates; while the explorer phenotype developed at high C, used glucose and sucrose as major C sources and casein and yeast extract as major N sources, and displayed higher decomposing potential for N and P providing substrates. The C availability, or the C/N ratio, during the free-living stage left a legacy to the symbiosis stage, regulating S. indica's potential to promote plant growth wheat growth promotion by the explorer phenotype was ± 40% higher than that by the compact phenotype. Our study highlights the importance of considering microbial ecology in designing PGPM/biofertilizers. Further studies are needed to test the phenotypes under more extreme conditions, and to understand if the in vitro acquired characteristics persist under field conditions.

  • Good responder patients to therapy (R-RA) exhibited an increased CD39 but not CD73 expression on B cells after treatment, while most of the non-responder (NR) patients showed a reduction in ectoenzyme expression. The positive changes of CD39 expression on B cells exhibited a negative correlation with disease activity and rheumatoid factor levels. Our results suggest modulating the ectoenzymes/ADO pathway as a potential therapy target for improving the course of RA.
    Berotralstat (BCX7353) is an oral, once-daily inhibitor of plasma kallikrein in development for the prophylaxis of hereditary angioedema (HAE) attacks.

    Our aim was to determine the efficacy, safety, and tolerability of berotralstat in patients with HAE over a 24-week treatment period (the phase 3 APeX-2 trial).

    APeX-2 was a double-blind, parallel-group study that randomized patients at 40 sites in 11 countries 111 to receive once-daily berotralstat in a dose of 110 mg or 150 mg or placebo (Clinicaltrials.gov identifier NCT03485911). Patients aged 12 years or older with HAE due to C1 inhibitor deficiency and at least 2 investigator-confirmed HAE attacks in the first 56 days of a prospective run-in period were eligible. The primary efficacy end point was the rate of investigator-confirmed HAE attacks during the 24-week treatment period.

    A total of 121 patients were randomized; 120 of them received at least 1 dose of the study drug (n= 41, 40, and 39 in the 110-mg dose of berotralstat, 150-mg of dose berotralstat, and placebo groups, respectively). Berotralstat demonstrated a significant reduction in attack rate at both 110 mg (1.65 attacks per month; P= .024) and 150 mg (1.31 attacks per month; P< .001) relative to placebo (2.35 attacks per month). The most frequent treatment-emergent adverse events that occurred more with berotralstat than with placebo were abdominal pain, vomiting, diarrhea, and **** pain. No drug-related serious treatment-emergent adverse events occurred.

PGRPs can recognize pathogenic microorganism peptidoglycans (PGs) and play an important role in innate immunity. Twelve PGRPs have been identified in silkworms. However, the specific roles played by these PGPRs in the silkworm innate immune system have not been elucidated to date. In this study, we systematically investigated the biological functions of BmPGRP-S1 in silkworms. We observed that BmPGRP-S1 was highly expressed in silkworm immune-related organs and was upregulated in response to bacterial challenges. Furthermore, we determined that BmPGRP-S1 can bind to bacteria or PGs and activate antimicrobial peptide (AMP) expression. Inhibition of the expression of BmPGRP-S1 by siRNA reduced AMP gene expression in silkworms. Further experiments demonstrated that BmPGRP-S1 is involved in IMD pathway activation to induce AMP expression. Taken together, these results demonstrate that BmPGRP-S1 serves as a receptor to activate AMP gene expression through the IMD pathway to address bacterial challenges.

We evaluated the primary outcome of 12-month supply prescriptions during the four months before and after the change.   • In order to investigate the potential of various olive cultivars and leaf sampling times for phytochemical farming practice in Croatia, phenolic and mineral composition was determined in olive leaves of four Croatian cultivars and Italian cultivar Leccino collected at three occasions, in October 2017, January 2018, and March 2018. Istarska bjelica turned out to have the largest phytochemical potential among the investigated cultivars due to steady high oleuropein concentrations found in its leaves. The concentration of main phenolic components in Istarska bjelica leaves changed only slightly during the sampling period, suggesting the possibility of its higher capability for low air temperatures stress resistance and different metabolic response compared to the other studied cultivars. Low air temperatures increased the oleuropein level and antioxidant activity in leaves of Leccino, Oblica, Levantinka, and Drobnica cultivars, which may be of crucial phytochemical farming interest. Each of the investigated olive cultivars was characterized by a specific leaf mineral nutrient composition, which could have had a specific role in their interplay with phenols.Nowadays, analytical systems based on silver triangular nanoplates (AgTNPs) have been shown as good prospects for chemical sensing. The 30 day mean PM2.5 concentration decreased by 10.4% in 2020, which is contrasted with an average increase of 23.7% over the corresponding periods in the previous 5 years. The PM2.5 concentration decrease was city-wide and more prominent during daytime than at nighttime. The concentrations of carbon monoxide (CO) and nitrogen dioxide (NO2) decreased by 16.9% and 16.4%, respectively. These results show that social distancing, a weaker forcing toward reduced human activity than a strict lockdown, can help lower pollutant emissions. Chemical analysis with X-ray photoelectron spectroscopy (XPS) and Fourier-transform infrared spectroscopy (FTIR) showed the formation of oxygen-based functional groups on the surface, independently from the working gas used for the treatment. Laser scanning microscopy (LSM) examined the surface roughness and revealed that the two argon-containing working gases roughened the surface more than synthetic air. However, the contact angle for water was reduced significantly after treatment, revealing measurement artifacts for water and diiodomethane due to the severe changes in surface morphology.   • Shock from medical and traumatic conditions can result in organ injury and death. Limited data describe out-of-hospital treatment of shock. We sought to characterize adult out-of-hospital shock care in a national emergency medical services (EMS) cohort.

    This cross-sectional study used 2018 data from ESO, Inc. (Austin, TX), a national EMS electronic health record system, containing data from 1289 EMS agencies in the United States. We included adult (age ≥18 years) non-cardiac arrest patients with shock, defined as initial systolic blood pressure ≤80 mm Hg. We compared patient demographics, clinical characteristics, and response (defined as systolic blood pressure increase) between medical and traumatic shock patients, looking at systolic blood pressure trends over the first 90 minutes of care.

    Among 6,156,895 adult 911 responses, shock was present in 62,867 (1.02%; 95% confidence interval [CI] = 1.01%-1.03%); 54,239 (86.3%) medical and 5978 (9.5%) traumatic, and 2650 unknown. #link# Medical was more common than traumatic shock in women and older patients. The most common injuries associated with traumatic shock were falls (37.6%) and motor vehicle crashes (18.7%). Mean initial and final medical systolic blood pressure were 71 ± 10 mm Hg and 99 ± 24 mm Hg. Systolic blood pressure increased in 88.8% and decreased or did not change in 11.0%. Mean initial and final trauma systolic blood pressure were 71 ± 13 mm Hg and 105 ± 28 mm Hg; systolic blood pressure increased in 90.4% and decreased/did not change in 9.6%. On fractional polynomial modeling, systolic blood pressure changes were greater and faster for trauma than medical shock.

    Emergency department boarding is the practice of caring for admitted patients in the emergency department after hospital admission, and boarding has been a growing problem in the United States. Boarding of the critically ill has achieved specific attention because of its association with poor clinical outcomes. Accordingly, the Society of Critical Care Medicine and the American College of Emergency Physicians convened a Task Force to understand the implications of emergency department boarding of the critically ill. The objective of this article is to review the U.S. literature on (1) the frequency of emergency department boarding among the critically ill, (2) the outcomes associated with critical care patient boarding, and (3) local strategies developed to mitigate the impact of emergency department critical care boarding on patient outcomes.

Emergency physicians must be aware of the design, pitfalls, and management of this new prosthetic hip design.

    The patient exhibited no signs or complaints of dyspnea or hypoxia, but later collapsed and became hypoxic after sleeping comfortably with sedation for 12h in the psychiatric unit. When other resources are not immediately available, emergency physicians should feel confident in making timely decisions, guided by basic principles of medical ethics. We make the following recommendations (1) availability of a member of the hospital ethics committee to provide in-person or telephonic consultation concurrent with patient care; (2) appointment to the hospital ethics committee of an emergency physician who is familiar with bioethical principles and is available for consultation when other ethics consultants are not; and (3) development of educational tools by professional societies or similar organizations to assist emergency physicians in making reasoned and defensible clinical ethics decisions.

  • Within the hyperthermia community, consensus exists that clinical outcome of the treatment radiotherapy and/or chemotherapy plus hyperthermia (i.e. elevating tumor temperature to 40 - 44 °C) is related to the applied thermal dose; hence, treatment quality is crucial for the success of prospective multi-institution clinical trials. Currently, applicator quality assurance (QA) measurements are implemented independently at each institution using basic cylindrical phantoms. A multi-institution comparison of heating quality using magnetic resonance thermometry (MRT) and anatomical representative anthropomorphic phantoms provides a unique opportunity to obtain novel QA insights to facilitate multi-institution trial evaluation.

    Neuromuscular disorders are characterised by muscle weakness that limits upper extremity mobility, but can be alleviated with dynamic arm support devices. Current research highlights the importance and difficulties of evidence-based recommendations for device development. We aim to provide research recommendations primarily concerning upper extremity body functions, and secondarily activity and participation, environmental and personal factors.

    Evidence was synthesised from literature, ongoing studies, and expert opinions and tabulated within a framework based on a combination of the International Classification of Functioning, Disability and Health (ICF) model and contextual constructs.

    Current literature mostly investigated the motor capacity of muscle function, joint mobility, and upper body functionality, and a few studies also addressed the impact on activity and participation. In addition, experts considered knowledge on device utilisation in the daily environment and characterising the beneficiarructs within one study design. Second, include the influence of environmental and personal factors when developing and deploying a device. Third, include short-term and long-term measurements to monitor adaptations over time. Finally, include user satisfaction as guidance to evaluate the device effectiveness. IMPLICATIONS ON REHABILITATION Synthesized evidence will support future research and development of dynamic arm supports. Tabulated evidence stresses the importance of integrating ICF model components and contextual constructs to fill the knowledge gaps. Presented knowledge gaps and proposed steps guide the set up of future studies on dynamic arm supports.
    Intracardiac thrombosis is reported to occur frequently in cardiac amyloidosis (CA). However, data regarding arterial thrombo-embolic events (AEs) in CA are limited. We aimed at assessing prevalence, clinical characteristics and predictors of AEs in a large multicentric CA cohort.

    Four-hundred-six consecutive CA patients (134 AL, 73 ATTRm and 199 ATTRwt) from 5 Italian referral centres were retrospectively evaluated and followed-up for a median time of 19 months. Thirty-one patients (7.6%) suffered from an AE, of whom 10 (32.2%) were in sinus rhythm and had no history of AF. #link# There were no significant differences in terms of age, gender and type of CA between patients with or without AEs. Fourteen (7.6%) of 185 patients on anticoagulation had an AE despite therapy. Anticoagulation therapy did not appear to fully protect from the risk of events (HR 1.23, 95%CI 0.52-2.92,
     = .64). The only predictor of AEs, in particular among CA patients in sinus rhythm, was a CHA2DS2-VASC score ≥ 3 (HR 2.84, 95%CI 1.02-7.92,

    High sensitivity cardiac troponin-T (hs-TnT) has been associated with mortality in patients hospitalized with COVID-19. We aimed to determine if hs-TnT levels and their timing are independent predictors of adverse events in these patients.

    Retrospective chart review was performed for all patients hospitalized at our institution between 23 March 2020 and 13 April 2020 who were found to be COVID-19-positive. The results of the research indicate that most of the determinants of work ability identified at work are impacted, thus giving the opportunity to conduct some occupational health and preventive programmes in the workplace.Traumatic brain injury (TBI) produces microstructural damage to white matter pathways connecting neural structures in pre-frontal and striatal regions involved in self-regulation (SR). Dorsal and ventral frontostriatal pathways have been linked to cognitive ("cool") and emotional ("hot") SR, respectively. We evaluated the relation of frontostriatal pathway fractional anisotropy (FA) 2 months post-TBI on cool and hot SR assessed 7 months post-TBI. Participants were 8-15 years of age, including children with uncomplicated mild TBI (mTBI; n = 24), more severe TBI (complicated-mild, moderate, severe [cms]TBI; n = 60), and typically developing (TD) children (n = 55). Diffusion tensor tractography was used to map frontostriatal pathways. Cool SR included focused and sustained attention performance, and parent-reported attention, whereas hot SR included risk-taking performance and parent-reported emotional control. Multivariate general linear models showed that children with cmsTBI had greater parent-reported cool and hot SR difficulties and lower dorsal and ventral FA than TD children. Focused attention, risk taking, and emotional control correlated with FA of specific dorsal and ventral pathways; however, only the effect of TBI on focused attention was mediated by integrity of dorsal pathways. Results suggest that frontostriatal FA may serve as a biomarker of risk for SR difficulties or to assess response to interventions targeting SR in pediatric TBI and in broader neurodevelopmental populations.The COVID-19 pandemic presents a major challenge to societies all over the globe. Two measures implemented in many countries to curb the spread of the disease are (a) minimizing close contact between people ("physical distancing") and (b) wearing of face masks. In the present research, we tested the idea that physical distancing and wearing of face masks can be the result of a prosocial emotional process-empathy for people most vulnerable to the virus. In four preregistered studies (N = 3,718, Western population), we found that (a) empathy indeed relates to the motivation to adhere to physical distancing and to wearing face masks and (b) inducing empathy for people most vulnerable to the virus promotes the motivation to adhere to these measures (whereas merely providing information about the importance of the measures does not). In sum, the present research provides a better understanding of the factors underlying the willingness to follow two important measures during the COVID-19 pandemic.While it has been established that expression perception is rapid, it is unclear whether early appraisal mechanisms invoke holistic perception. Participants were shown a congruent (e.g. top angry/ bottom angry) or incongruent (e.g. top angry/ bottom happy) expression for 17, 50 or 250 ms and instructed to selectively attend to the cued expression depicted in the top (or bottom) half of the composite face and ignore the uncued portion. Compared to the isolated condition, a facilitation effect was found for congruent angry expressions, as well as an interference effect for incongruent happy and angry expressions at the shortest exposure duration of 17. Together these results provide evidence that the holistic gist perception of expression cannot be overridden by selective attention.The world remains cautiously optimistic about a COVID-19 vaccine that is relatively safe and efficacious and that offers sufficient long-lasting protection/immunity by neutralizing the virus infectivity. However, key technical hurdles pertaining to antigen-adjuvant formulation, delivery, and manufacturing challenges of lipid nanoparticles (LNPs) for mRNA vaccines and stability of formulations need to be addressed for successful product development and stockpiling. In addition, the dosage form, the dosage level and regimen for eliciting a protective immune response remain to be established. The high dependence of global supply chains and demand-supply to sourcing quality raw materials, glassware and other supplies, along with the stress on existing production capacities and platform-specific manufacturing challenges could impede vaccine development and access. This article provides critical analysis of vaccine development processes and unit operations that can derail the pandemic response, and also extends to other emerging infectious disease development efforts - issues that take on added significance given the global mandate for an accelerated and at-risk development path to tackle the COVID-19 pandemic.Social protection interventions (inclusive of cash grant receipt and care provision) have been found to be effective in response to some of the negative implications of the HIV epidemic on children and families. This study explores the impact of cash grant receipt and care provision (operationalised as good parenting) on child nutritional outcomes. In this cross-sectional study, 854 children and younger adolescents (5-15 years) and caregivers affected by HIV, attending community-based organisations in South Africa and Malawi, were interviewed. #link# Interviews comprised inventories on socio-demographic information, family data, cash grant receipt and child nutrition.
