  • IgA nephropathy (IgAN) is achronic immuno-inflammatory progressive disease. Several systemic inflammatory indicators, mainly the neutrophil-to-lymphocyte ratio (NLR), are regarded as valuable markers for many diseases, such as IgA vasculitis and chronic kidney disease. Here, we investigated multiple peripheral blood indicators in a large IgAN registry with regular follow-up to evaluate their effects on IgAN phenotypes and progression.

    Totally, 1151 IgAN patients with regular follow-up, and 251 healthy volunteers were enrolled.     IgAN patients presented with increased WBC, NE, NLR and PLR levels and decreased LY levels compared with controls. In univariate survival analysis, WBC, NE and NLR showed significant associations with IgAN progression, and NLR had a higher area under the ROC curves than NE and WBC. When adjusted for well-known risk factors, NLR remained an independent risk factor for poor renal outcome in IgAN patients and performed better than NE. By using NLR 2.40 as cutoff point, IgAN patients were divided into two groups. IgAN patients in the high NLR group presented with lower eGFR, higher proteinuria, higher incidence of hypertension, and more severe pathological lesions, as well as lower event-free renal survival rate.

    Cigarette smoke is considered as a sterile inflammatory stimulus which triggers an innate immune response, accountable for vascular events. Previously, we reported smoking-induced NLRP3 inflammasome activation in the pathogenesis of atherosclerosis through caspase-1 activation and secretion of pro-cytokines (interleukin (IL)-1β and IL-18) in vitro and in vivo. Further, the levels of serum cotinine, oxidative stress markers (8-isoprostane and 8-oxo-2́'-deoxyguanosine), caspase-1 and pro-cytokines (IL-1β and IL-18) were also higher in smokers vs non-smokers. The optimized conditions included 30% aqueous ethanol, 62.66 mL/g ratio of liquid to solid, 3 mm centre hole diameter of hollow electrode, 123 mL/min flow velocity, length of serpentine pipe of 15 cm, 276 W ultrasonic power, 47 °C ultrasonic temperature, and discharge voltage was fixed at 14 kV. Under these conditions, it obtained a maximum saponins yield of 51.30 ± 0.08 mg/g with 10 min, which was higher than those of LPD (42.33 ± 0.98 mg/g) with 24 min, ultrasonic assisted extraction (UAE) (41.80 ± 1.31 mg/g) with 30 min and maceration (38.72 ± 1.13 mg/g) with 180 min. Meanwhile, the energy consumption of LPDU was 7560 kJ/kg, which was notably lower than those of LPD (8820 kJ/kg), UAE (25875 kJ/kg) and maceration (10248 kJ/kg). We found that the saponin constituents of LPDU were similar to LPD, UAE, ME by HPLC content detection method, and found that LPDU had the highest degree of tissue damage after scanning electron microscope (SEM) comparison, which verified the reason for its high extraction efficiency.

  • PURPOSE Lung cancer is one of the deadly diseases with limited treatment options available. The main aim of the current study was to investigate the antitumor effects of levopimaric acid - a naturally occurring diterpene, against cisplatin-resistant non-small cell lung carcinoma cells A-549 and normal **** cells. Effects of levopimaric acid on autophagy, reactive oxygen species (ROS), apoptosis and ERK/MAPK signalling pathways were also investigated in the current study. METHODS Proliferation rate was monitored by MTS assay. Apoptosis was detected by DAPI staining as well as western blot assay. Electron microscopy was used to investigate the autophagic effects of levopimaric acid. Effects on ROS and mitochondrial membrane potential (MMP) were evaluated by flow cytometry. Protein expression was examined by western blotting. RESULTS It was found that levopimaric acid exerts potent antiproliferative effects against the cisplatin-resistant lung cancer cells and exhibited an IC50 of 15 μM. However, the toxic effects of levopimaric acid were seen to be insignificant against the normal cells. The anticancer effects of levopimaric acid were due to induction of apoptosis which was also associated with modulation of apoptosis-related proteins (Bax and Bcl-2). Levopimaric acid also induced autophagy which was also associated with alterations of autophagy-related protein expressions (LC3I, II, and p62). Levopimaric acid caused ROS-mediated alterations in the MMP. It was also found that the molecule could induce drug-resistant lung cancer cell death by activating p38 MAPK and JNK signalling pathways while inhibiting ERK pathway. CONCLUSION The current results strongly indicate that levopimaric acid may prove to be a potential anticancer drug candidate provided further in depth studies are carried out.PURPOSE This study aimed to compare the efficacy of laparoscopic and conventional left hemicolectomy for treating colon cancer and their effects on stress response and quality of life of patients. The surgery, postoperative recovery, intraoperative and postoperative complications were compared between the two groups. The enzyme-linked immunosorbent assay (ELISA) was used to detect the levels of IL1β and IL-6. The quality of life of patients after surgery was analyzed by the Functional Assessment of Cancer Therapy-Lung (FACT-L). RESULTS The operation time and intraoperative blood loss of the study group were statistically lower than those of the control group (p less then 0.05). The postoperative exhaust time and hospitalization time of the study group were statistically shorter than those of the control group (p less then 0.05). Serum IL-1β and IL-6 levels in the study group were significantly lower than those in the control group (p less then 0.05). In the two groups, the overall scores of quality of life after surgery were significantly lower than those before surgery (p less then 0.05). After surgery, the overall score of quality of life in the study group was significantly higher than that in the control group (p less then 0.05). The cut-off value was determined as >8.5 fL for MPV (sensitivity 67.1%, specificity 54.5%) and ≤16% for PDW (sensitivity 66.7%, specificity 60.0%). RESULTS The study included 394 patients, 53.3% of which were male. Stage I, II, and III patients constituted 8.9%, 46.4%, and 44.7% of the study population, respectively. Among all patients, RFS and OS were significantly longer in patients with MPV≤8.5 fL and PDW>16 fL (p16% had significantly longer RFS than that in those with PDW ≤16 fL among stage III patients (p less then 0.001). In multivariate analysis, stage, perineural invasion, lymphovascular invasion, adjuvant treatment, CEA, CA19-9, PDW, and MPV were found the most significant factors affecting RFS. CONCLUSION Our study suggests that elevated MPV and decreased PDW appear to be unfavorable prognostic factors in early CC, especially in patients with stage III disease. There was overtly less intraoperative blood loss and shorter postoperative hospital stay in the CME group than those in the Traditional group (129.35±34.54 mL vs. 162.43±38.16 mL, p0.05). The number of lymph nodes dissected and the number of positive lymph nodes detected were clearly greater in the CME group than in the Traditional group (p less then 0.001). At the end of the follow-up, the overall survival rate and tumor-free survival rate were notably higher in the CME group than in the Traditional group (p=0.046, p=0.038). CONCLUSION In comparison with traditional radical operation for colon cancer, laparoscopic CME has higher yield of lymph nodes dissected, smaller intraoperative blood loss, no increase in perioperative complications, and higher overall survival and tumor-free survival of patients, demonstrating it as safe and applicable in the treatment of stage III colon cancer.

  • Taking into account its low cost, widespread availability and non-invasiveness, electroencephalography (EEG) would represent a candidate for tracking the prodromal phases of cognitive decline in routine clinical settings eventually in combination with other markers. In this scenario, the present paper provides an overview of epidemiology, genetic risk factors, neuropsychological, fluid and neuroimaging biomarkers in AD and describes the potential role of EEG in AD investigation, trying in particular to point out whether advanced analysis of EEG rhythms exploring brain function has sufficient specificity/sensitivity/accuracy for the early diagnosis of AD. OBJECTIVE A very high-resolution (70 MHz) ultrasound device (VHRUS) has recently been approved for use in humans. The aim of this study was to use VHRUS to collect data on healthy subjects to propose some reference values for the digital branches of the median nerves of the hand. METHODS A VHRUS with 70 MHz linear array transducer was used to measure the cross sectional area of the median nerve at the wrist (CSAw) and digital branches (CSAf), largest and smallest fascicles, the fascicles number (Nfasc), the fascicle density (FD), the flattening ratio (FR) and CSAw/CSAf. RESULTS Data from 20 healthy subjects were obtained for both hands. The median nerve at the wrist and digital branches were properly identified without anatomical alterations. No differences were found between the right and the left hand. In the dominant hand, CSAw was 9.35 mm2 (4.57-12.35) and Nfasc was 24 (18-38). FD and FR were respectively 2.94 (2.47-4.91) and 2.74 (1.70-4.90). CONCLUSION VHRUS technology can visualize the median nerves at the wrist, their internal structure and their small branches at the fingers, providing both a qualitative and quantitative assessment. Results from this study provide preliminary reference values in a young healthy sample. SIGNIFICANCE Most conventional ultrasound devices are not able to properly visualize the distal branches of the median nerve. A total of 216 samples from cattle and 154 samples from sheep were used to evaluate the ELISAs. The sensitivity and specificity results of the ELISA-rNcSRS2 were 91.5 % and 96.4 % for cattle, and 89.6 % and 96.3 % for sheep, respectively. For the ELISA-rNcSAG1, the sensitivity and specificity were 84.9 % and 97.3 % for cattle, and 89.6 % and 92.6 % for sheep, respectively. The sensitivity and specificity of the ELISA-rNcSRS2/rNcSAG1 was 98.1 % and 99.1 % for cattle, 100 % and 97.2 % for sheep, respectively. Manual phenotyping indicated that Arabidopsis lines over-expressing enzymes in the myo-inositol pathway have elevated AsA, accumulate more biomass of both aerial and root tissues and are tolerant to abiotic stresses including salt, cold, heat, and environmental pollutants. However, manual phenotyping is time consuming, low throughput, subjective, and limited to the resolution of the human eye. In contrast, high throughput phenotyping technologies are accurate, non-destructive, and more sensitive, allowing the detection of subtle phenotypes. Therefore, we used a phenomics platform to phenotype our high AsA Arabidopsis lines with visible, fluorescence, and near infrared cameras. Based on this approach, high AsA lines grew faster, accumulated more biomass, and displayed healthier chlorophyll fluorescence and water content profiles than controls. By studying abiotic stress in a high throughout fashion using optimized protocols, we have also shown that these high AsA lines are tolerant to salt and water limitation stresses. In addition, we developed open source algorithms to analyze images and by comparing results obtained with a widely used commercial software against our algorithms, here we show that our method achieved good accuracy for all phenotypic parameters of interest including projected leaf area, rosette diameter (caliper length), compactness, and color classification. EB was sprayed to strawberry plants every two days for two weeks. Then, the EB-treated plants were pre-treated with inhibitors of NR, tungstate, or NOS, L-NAME for 3 h. During the first three weeks, Fe was supplied as 100 μM EDTA-Fe or FeSO4 to Fe-sufficient or Fe-deficient plants, respectively. Thereafter, plants were subjected for further three weeks to control (100 μM EDTA-Fe) and Fe deficiency (ID; without Fe). ID reduced biomass, chlorophyll, and chlorophyll fluorescence, while increased oxidative stress parameters, ascorbate (AsA), glutathione (GSH), endogenous NO, and the activities of NR, NOS, and antioxidant enzymes.   • The severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) that is causing a global pandemic had a zoonotic origin in China. Considering the inter-connectedness between human, environment, and animal health, the One Health approach is the appropriate strategy to control and mitigate the effects of the ongoing coronavirus disease 2019 (COVID-19). This letter explains the benefits of the One Health approach and recommends specific measures that could be taken to accelerate the fight against COVID-19 and prevent the spread of newly emerging infectious diseases.

The purpose of this study was to provide a comprehensive overview of the reported medicinal plants used to manage TB symptoms, and to analyze scientific reports on their effectiveness and safety.

    A comprehensive literature search was performed in the major electronic databases regarding medicinal plants used in the management of TB in East Africa. A total of 44 reports were retrieved, and data were collected on various aspects of the medicinal plants such as botanical name, family, local names, part(s) used, method of preparation, efficacy, toxicity, and phytochemistry. The data were summarized into percentages and frequencies which were presented as tables and graphs.

    A total of 195 species of plants belonging to 68 families and 144 genera were identified. Most encountered species were from Fabaceae (42.6%), Lamiaceae (19.1%), Aste be effective in the management of symptoms of TB. More validation studies are required to promote the discovery of antimycobacterial drugs and to provide evidence for standardization of herbal medicine use.

We previously demonstrated that MT2 receptor knockout (MT2 -/- ) **** show a decreased non-rapid eye sleep over 24-hr and increased wakefulness during the inactive (light) phase. Here, we investigated the role of MT2 receptors in physiological light/dark cycle fluctuations in the activity of dorsal raphe nucleus (DRN) serotonin (5-HT) neurons and anxiety- and depression-like behaviour. We found that the 5-HT burst-firing activity was tonically reduced across the whole 24-hr in MT2 -/- ****. Importantly, the physiological changes in the spontaneous firing activity of DRN 5-HT neurons during the light/dark cycle were nullified in MT2 -/- ****, with an higher DRN 5-HT neural firing activity during the light phase in MT2 -/- ****. The role of MT2 receptors over DRN 5-HT neurons was confirmed by acute pharmacological studies in which the selective MT2 receptors agonist UCM1014 dose-dependently inhibited DRN 5-HT activity, mostly during the dark phase. Compared with WT, MT2 -/- **** displayed an anxiety-like phenotype in the novelty suppressed feeding and in the light/dark box tests; while anxiety levels in the light/dark box test were lower during the dark than the light phase in WT ****, the opposite was seen in MT2 -/- ****. No differences were observed for depression-like behavior in the forced swim and in the sucrose preference tests in MT2 -/- ****. These results suggest that MT2 receptor genetic inactivation impacts 5-HT neurotransmission and interferes with anxiety levels by perturbing the light/dark pattern. METHODS Twenty-seven families were screened for germline variants in eight XP-related genes. RESULTS All patients (N= 32) were diagnosed with bi-allelic germline pathogenic or potentially pathogenic variants, including nine variants previously undescribed. The c.2251-1G>C XPC pathogenic variant, reported as the founder mutation in Comorian and Pakistani patients, was observed in 15 cases in homozygous or compound heterozygous. Seven homozygous patients for POLH/XPV variants developed their symptoms by an average age of 7.7 years. ERCC2/XPD, DDB2/XPE, and ERCC5/XPG variants were found in a few patients. Aside from melanoma and non-melanoma skin tumors, a set of patients developed skin sebaceous carcinoma, leiomyosarcoma, angiosarcoma, mucoepidermoid carcinoma, gastric adenocarcinoma and serous ovary carcinoma. CONCLUSIONS We reported a high frequency of XPC variants in 32 XP Brazilian patients. In Experiment 1, we conducted fNIRS measurements to find the presence of a mapping of the McGurk effect in Japanese 8- to 9-month-old infants and to examine the difference between the activation patterns in response to own-race-face and other-race-face stimuli. We used two race-face conditions, own-race-face (East Asian) and other-race-face (Caucasian), each of which contained audiovisual-matched and McGurk-type stimuli. While the infants (N = 34) were observing each speech stimulus for each race, we measured cerebral hemoglobin concentrations in bilateral temporal brain regions. The results showed that in the own-race-face condition, audiovisual-matched stimuli induced the activation of the left temporal region, and the McGurk stimuli induced the activation of the bilateral temporal regions. No significant activations were found in the other-race-face condition. These results mean that the McGurk effect occurred only in the own-race-face condition. In Experiment 2, we used a familiarization/novelty preference procedure to confirm that the infants (N = 28) could perceive the McGurk effect in the own-race-face condition but not that of the other-race-face. While the infants (N = 34) were observing each speech stimulus for each race, we measured cerebral hemoglobin concentrations in bilateral temporal brain regions. The results showed that in the own-race-face condition, audiovisual-matched stimuli induced the activation of the left temporal region, and the McGurk stimuli induced the activation of the bilateral temporal regions. No significant activations were found in the other-race-face condition. These results mean that the McGurk effect occurred only in the own-race-face condition. For a broad clinical usage, protection of the coagulation activities may require further optimization. METHODS Otolaryngologists and resident trainees from across Canada were recruited to evaluate CardinalSim. Ethics approval and informed consent was obtained. A face and content validity questionnaire with questions categorized into 13 domains was distributed to participants following simulator use. The experimentation carried out in an off-the-self computer with the parallel execution enabled shows an improvement in run-time. This implementation may also lead to an important use of memory load. Results previously obtained with simulated and real data indicated that the procedure implemented in the package is robust and suitable for differentially expressed genes identification. CONCLUSIONS The new package, ORdensity, offers a friendly and easy way to identify differentially expressed genes, which is very useful for users not familiar with programming.   • This work reports for the first time a significantly improved and simplified electrochemical immunoassay to detect antibodies to tick-borne encephalitis virus (TBEV) using a 96-well microtiter plate as a platform for immobilization and silver nanoparticles (AgNPs) as electrochemical labels. The electrochemical assay is performed by detecting the elemental silver oxidation signal where the electroactive signalling silver species are released from the bioconjugates (Ab@AgNP, AbS@AgNP, and ProteinA@AgNP). For this purpose, AgNPs were synthesized and further tagged with biomolecules (antibodies to TBEV, cleaved antibodies to TBEV, and protein A). Signal is read by linear sweep anodic stripping voltammetry (LSASV) of silver ions (through the electrochemical stripping of accumulated elemental silver) on a graphite electrode (GE). AbS@AgNP was chosen as the best option for the new electrochemical immunoassay. The results of electrochemical measurements demonstrated that voltammetric signal increased with the increasing concentration of target antibodies to TBEV within the range from 100 to 1600 IU mL-1, with a detection limit of 90 IU mL-1. To verify the practical application of the novel electrochemical immunosensor, the quantity of immunoglobulins against TBEV in human serum was checked. Video-taped observations were scored using the Child and Maternal Behavior Rating Scales. No consistent group-level trend was found. Within singular interactions, parent's responsive behavior and child interactive engagement (attention and initiation) seem to be strongly related. Initial child initiation seems to positively predict parents' achievement orientation and directive behavior two years later. Many novel agents of different therapeutic strategies that aim to modulate LSCs self-renewal, proliferation, apoptosis, and differentiation are under investigation. In this review, we summarize the latest advances of different therapies in development based on the biological characteristics of LSCs, with particular attention on natural products, synthetic compounds, antibody therapies, and adoptive cell therapies that promote the LSC eradication. We also explore the causes of AML recurrence and proposed potential strategies with new dimensions for targeting LSCs in the future.The production of fuels and other valuable chemicals via biological routes has gained significant attention during last decades. Cyanobacteria are prokaryotes that convert solar energy to chemical compounds in vivo in direct processes. Intensive studies have been carried out with the aim of engineering cyanobacteria as microfactories for solar fuel and chemical production. Engineered strains of photosynthetic cyanobacteria can produce different compounds on a proof-of-concept level, but few products show titers comparable with those achieved in heterotrophic organisms. Efficient genetic engineering tools and metabolic modeling can accelerate the development of solar fuel and chemical production in cyanobacteria. This review addresses the most recent approaches to produce solar fuels and chemicals in engineered cyanobacteria with a focus on acetyl-CoA-dependent products.Radiologists work in conjunction with orthopedic surgeons to evaluate the progression of bone healing and identify potential problems during bone reconstruction. Accurate evaluation and identification of healing progression or complications are critical to optimizing successful patient outcomes with either distraction osteogenesis or bone grafting. Therefore, radiologists must understand the fundamental concepts behind these surgical reconstructive techniques in order to provide accurate postoperative radiographic assessments. The cases and discussion within this review aim to provide this foundational knowledge.Knowledge on the long-term interactive interplay between children with a significant cognitive and motor developmental delay and their parents is very scarce. We aimed to characterize the (in)variability and potential mutual influence of parent's interactional style and child interactive engagement throughout early childhood. Every six months over the course of two years, thirty-five parent-child dyads (children aged 6-59 months) living in Flanders (Belgium) or the Netherlands were video-taped during a 15-minute unstructured play situation. Video-taped observations were scored using the Child and Maternal Behavior Rating Scales. No consistent group-level trend was found. Within singular interactions, parent's responsive behavior and child interactive engagement (attention and initiation) seem to be strongly related. Initial child initiation seems to positively predict parents' achievement orientation and directive behavior two years later. Parental responsiveness might be an effective interactional strategy to increase child engagement and higher levels of engagement in children possibly can facilitate parental responsiveness within a concrete interaction. The more initiative children show, the more parents might have hope for developmental benefits resulting from a directive/achievement oriented approach. Further research is warranted applying more differentiated and dynamically evaluated outcome measures and a longer follow-up time frame, with specific attention to inter-individual differences.Purpose to elucidate the effects of multiple scattering and energy-loss straggling on electron beams slowing down in materials. Methods EGSnrc Monte Carlo simulations are done using a purpose-written user-code. Results Plots are presented of the primary electron's energy as a function of pathlength for 20 MeV electrons incident on water and tantalum as are plots of the overall distribution of pathlengths as the 20 MeV electrons slow down under various Monte Carlo scenarios in water and tantalum. The distributions range from 1 % to 135 % of the CSDA range in water and from 1 % to 186 % in tantalum. The effects of energy-loss straggling on energy spectra at depth and electron fluence at depth are also presented. Conclusions The role of energy-loss straggling and multiple scattering are shown to play a significant role in the range straggling which determines the dose fall-off region in electron beam dose vs depth curves and a significant role in the energy distributions as a function of depth.
