Between-groups comparisons were performed using a t test and Mann-Whitney U test. Correlations were determined utilizing Spearman rank correlation coefficient, and the influencing factors of the SDS scores were analyzed using logistic regression.The average score of the SDS among the 323 participants was 42.53 ± 8.51. We then analyzed patient, hospital-, and market-level factors associated with variation of colectomy. For each HRR, a Herfindahl-Hirschman index, a measure of market competition, was calculated.

    A total of 19,557 Medicare patients underwent an elective colon resection for diverticulitnd hospital level factors were strongly associated with this variation and may be the main drivers of colonic resection for diverticular disease. Further investigation and stronger national guidelines are needed to optimize patient selection for colectomy.

Variable importance measures were used to identify the factors with the strongest influence on survival, and partial dependence plots were used to determine the dependence of survival on the target variable while adjusting for all other variables.

    We identified 5309 liver transplants that met our criteria. The overall 1-year survival of high-acuity patients improved from 69% in 2001 to 87% in 2016. The strongest predictors of death within 1 year of transplant were patient on mechanical ventilator before transplantation, prior liver transplant, older recipient age, older donor age, donation after cardiac death, and longer cold ischemia.

    Liver transplant outcomes continue to improve even for patients with high medical acuity. Applying ensemble learning methods to recipient and donor factors available before transplant can predict survival probabilities for future transplant cases.     The NIH awarded 1101 new grants (total $389,006,782; median $313,030) between 2008 and 2018. Funding to DoS has doubled in the last 10 years ($22,983,500-2008 to $49,446,076-2018). Midwest/Southeast institutions and surgical oncologists accounted for majority of the grants (31.9% and 24.5%, respectively). This pilot study suggests that the time-frequency analysis of EEG signals could be a great tool to visualize the brain activities in response to different cognitive tasks.We analyze the efficiency of motor unit (MU) filter prelearning from high-density surface electromyographic (HDEMG) recordings of voluntary muscle contractions in the identification of the motor unit firing patterns during elicited muscle contractions. Motor unit filters are assessed from 10 s long low level isometric voluntary contractions by gradient-based optimization of three different cost functions and then applied to synthetic HDEMG recordings of elicited muscle contractions with dispersion of motor unit firings ranging from 13 ms to 1 ms. We demonstrate that the number of identified MUs and the precision of MU identification depend significantly on the selected cost function. Regardless the selected cost function and MU synchronization level, the median precision of motor unit identification in elicited contraction is ≥ 95 % and is comparable to the precision in voluntary contractions. On the other hand, median miss rate increases significantly from less then 1 % to ~ 3 % with the tested level of MU synchronization.Clinical Relevance-The identification of MU firings from HDEMG in elicited muscle contractions provides a new tool for in vivo investigation of motor excitability in humans.The absence of epileptiform activity in a scalp electroencephalogram (EEG) recorded from a potential epilepsy patient can cause delays in clinical care delivery. Here we present a machine-learning-based approach to find evidence for epilepsy in scalp EEGs that do not contain any epileptiform activity, according to expert visual review (i.e., "normal" EEGs). We found that deviations in the EEG features representing brain health, such as the alpha rhythm, can indicate the potential for epilepsy and help lateralize seizure focus, even when commonly recognized epileptiform features are absent. Hence, we developed a machine-learning-based approach that utilizes alpha-rhythm-related features to classify 1) whether an EEG was recorded from an epilepsy patient, and 2) if so, the seizure-generating side of the patient's brain. We evaluated our approach using "normal" scalp EEGs of 48 patients with drug-resistant focal epilepsy and 144 healthy individuals, and a naive Bayes classifier achieved area under ROC curve (AUC) values of 0.81 and 0.72 for the two classification tasks, respectively. These findings suggest that our methodology is useful in the absence of interictal epileptiform activity and can enhance the probability of diagnosing epilepsy at the earliest possible time.Brain-computer interface (BCI) systems enable humans to communicate with a machine in a non-verbal and covert way. The input data were modified using a bandpass filter and then applied to a convolutional neural network model. For five sleep stage classification, the classification performance 85.6% and 91.1% using the raw input data and the proposed input, respectively. This result also shows the highest performance compared to conventional studies using the same dataset. In this study, the effect of ADLs on the performance of EMG-based gesture recognition using a wearable EMG device was investigated. EMG data for 14 hand and finger gestures, as well as continuous activity during uncontrolled ADLs (>10 hours in total) were collected and analyzed. Results showed that (1) the cluster separability of 14 different gestures during ADLs was 171 times worse than during rest; (2) the probability distributions of EMG features extracted from different ADLs were significantly different (p less then ; 0.05). (3) of the 14 target gestures, a right angle gesture (extension of the thumb and index finger) was least often inadvertently activated during ADLs. These results suggest that ADLs and other non-trained gestures must be taken into consideration when designing EMG-based gesture recognition systems.

This pilot study suggests that the time-frequency analysis of EEG signals could be a great tool to visualize the brain activities in response to different cognitive tasks.We analyze the efficiency of motor unit (MU) filter prelearning from high-density surface electromyographic (HDEMG) recordings of voluntary muscle contractions in the identification of the motor unit firing patterns during elicited muscle contractions. Motor unit filters are assessed from 10 s long low level isometric voluntary contractions by gradient-based optimization of three different cost functions and then applied to synthetic HDEMG recordings of elicited muscle contractions with dispersion of motor unit firings ranging from 13 ms to 1 ms. We demonstrate that the number of identified MUs and the precision of MU identification depend significantly on the selected cost function. Regardless the selected cost function and MU synchronization level, the median precision of motor unit identification in elicited contraction is ≥ 95 % and is comparable to the precision in voluntary contractions. On the other hand, median miss rate increases significantly from less then 1 % to ~ 3 % with the tested level of MU synchronization.Clinical Relevance-The identification of MU firings from HDEMG in elicited muscle contractions provides a new tool for in vivo investigation of motor excitability in humans.The absence of epileptiform activity in a scalp electroencephalogram (EEG) recorded from a potential epilepsy patient can cause delays in clinical care delivery. Here we present a machine-learning-based approach to find evidence for epilepsy in scalp EEGs that do not contain any epileptiform activity, according to expert visual review (i.e., "normal" EEGs). We found that deviations in the EEG features representing brain health, such as the alpha rhythm, can indicate the potential for epilepsy and help lateralize seizure focus, even when commonly recognized epileptiform features are absent. Hence, we developed a machine-learning-based approach that utilizes alpha-rhythm-related features to classify 1) whether an EEG was recorded from an epilepsy patient, and 2) if so, the seizure-generating side of the patient's brain. We evaluated our approach using "normal" scalp EEGs of 48 patients with drug-resistant focal epilepsy and 144 healthy individuals, and a naive Bayes classifier achieved area under ROC curve (AUC) values of 0.81 and 0.72 for the two classification tasks, respectively. These findings suggest that our methodology is useful in the absence of interictal epileptiform activity and can enhance the probability of diagnosing epilepsy at the earliest possible time.Brain-computer interface (BCI) systems enable humans to communicate with a machine in a non-verbal and covert way. Results show that this new approach can decode attention with a high accuracy (>75%) and has a high information transfer rate (>10 bits/min) compared to other auditory BCI systems. This review discusses the relative dependency that AML cells have on mitochondrial function, and the ability to pivot this reliance to target important subsets of AML cells, including leukemia stem cells (LSCs). LSCs are tumor-initiating cells that are resistant to standard chemotherapy and promote the persistence and relapse of AML. Historically, LSCs have been targeted based on immunophenotype, but recent developments in the understanding of LSC metabolism has demonstrated unique abilities to target LSCs while sparing normal hematopoietic stem cells (HSCs) through inhibition of mitochondrial function. Here we highlight the use of small molecules that have been demonstrated to effectively target mitochondrial function. IACS-010759 and ME-344 target the electron transport chain (ETC) to inhibit oxidative phosphorylation (OXPHOS). Salt challenge, given at 7-8 weeks of age with either 2% saline drink (3 days) or 8% NaCl-containing chow (4 weeks), had little or no effect on systolic blood pressure (SBP) in female offspring, whereas the salt challenge significantly raised SBP in male offspring, with the magnitude of increase being greater in experimental, than control, rats. Furthermore, the salt challenge not only raised plasma AVP level more and caused greater depressor responses to V1a and V2 AVP receptor antagonists to occur in experimental, than control, males, but it also made GABA excitatory in a significant proportion of magnocellular AVP neurons of experimental males by depolarizing GABA equilibrium potential. The effect of the maternal salt loading on the salt challenge-elicited SBP response in male offspring was precluded by maternal conivaptan treatment (non-selective AVP receptor antagonist) during the salt-loading period, whereas it was mimicked by neonatal AVP treatment.     The Mutuelle générale de l'éducation nationale (MGEN), a provider of private health insurance in France, has developed the VIVOPTIM programme, a novel digital approach to healthcare based on individualized, multiprofessional, ranked management of cardiovascular risk factors.

    Between November 2015 and June 2016, eligible individuals (age 30-70 years) from two regions of France were invited to participate. Volunteers completed a questionnaire based on the Framingham Heart Study Risk Score and were assigned to one of three cardiovascular risk levels. VIVOPTIM comprises four components cardiovascular risk assessment, instruction on cardiovascular diseases and associated risk factors, personalized coaching (telephone sessions with a specially trained healthcare professional to provide information on risk factors and disease management, set individual health targets, monitor progress and motivate participants), and e-Health monitoring.

    Data from 2240 participants were analysed. Significant benefits were observed on mean systolic blood pressure (-3.4mmHg), weight (-1.5kg), smoking (-2.2 cigarettes/day) and daily steps (+1726 steps/day (all P<0.0001)), though not on weekly duration of exercise (-0.2hours/week, P=0.619).

    As a result of the positive mid-to-long-term results of the pilot programme on weight, smoking, blood pressure, and uptake of physical activity, the VIVOPTIM programme was extend to the whole of France in 2018 and has the potential to have a genuine impact on patient care and organization of the healthcare system in France.

  • Bilateral slipped capital femoral epiphysis (SCFE) is common. The management of the contralateral hip in unilateral SCFE remains controversial. The aim of this study was to report on the clinical outcomes using a posterior sloping angle (PSA) threshold of 14.5 degrees for prophylactic fixation in preventing contralateral SCFE.

    Having previously established through a retrospective study that PSA was predictive of future slip, the authors put in place a protocol where patients with unilateral SCFE who had a PSA ≥14.5 degrees on the contralateral side were offered prophylactic fixation. Those with unilateral SCFE presenting between January 2008 and December 2018 with a minimum of 12-month follow-up were included. Patients with renal or endocrine disorders were excluded. Primary outcomes were the number of slips prevented, the number needed to treat, and the complication rate.

    Of the 219 patients who were included, 114 (52.1%) underwent prophylactic fixation.A PSA threshold of 14.5 degrees prevented 77% of subsequent slips with a number needed to treat of 2.4 in our population. There were no cases of chondrolysis, avascular necrosis, or periprosthetic fracture associated with prophylactic pinning.

    Prophylactic fixation using a PSA of 14.5 degrees is safe, decreases unnecessary intervention, and reduces 77% of subsequent SCFE. The PSA can increase over time and the authors recommend that the protocol be applied for the duration of follow-up.

    Neurocysticercosis is the most common helminthic infection of the central nervous system caused by the larval stage of the pork tapeworm, Taenia solium. Endemic regions include Latin American countries, sub-Saharan Africa, and large regions of Asia, including the Indian subcontinent and is a global health problem. Seizures are the most common manifestation and approximately 30% of adult-onset seizures in endemic regions are attributable to NCC. Calcifications because of neurocysticercosis is the most common finding on imaging in endemic regions and are important seizure foci contributing to the burden of epilepsy.

    Scrub typhus, caused by Orientia tsutsugamushi, is a widely neglected disease which is gaining global momentum because of its resurgence patterns. The disease is now being reported in newer regions as well as areas previously endemic areas. In this review, we aim to comprehensively review the data available to assist physicians in making an accurate diagnosis and appropriate management of the disease.

    Several diagnostic tests have been developed for confirming scrub typhus. However, there is lack of clarity on which tests are most appropriate in a given clinical scenario. A recent study has demonstrated that in early disease (<7 days) when serological tests remain negative, the quantitative polymerase chain reaction is the most sensitive test. Among the serological tests, both IgM enzyme-linked immunosorbent assay as well as rapid diagnostic tests revealed excellent sensitivities and specificities.

Endemic regions include Latin American countries, sub-Saharan Africa, and large regions of Asia, including the Indian subcontinent and is a global health problem. Seizures are the most common manifestation and approximately 30% of adult-onset seizures in endemic regions are attributable to NCC.   • Deep neural networks were used to perform bite counting and food item recognition in an end-to-end manner. Experiments have shown that counting bites directly from video clips can reach 74.15% top-1 accuracy (classifying between 0-4 bites in 20-second clips), and a MSE value of 0.312 (when using regression). Our experiments on video-based food recognition also show that recognizing consumed food items is indeed harder than recognizing visible ones, with a drop of 25% in F1 score. In this work, we have presented a novel multi-modal approach, named MultiPredGO, for predicting protein functions by utilizing two different kinds of information, namely protein sequence and the protein secondary structure. Here, our contributions are threefold; firstly, along with the protein sequence, we learn the feature representation from the protein structure. Secondly, we develop two different deep learning models after considering the characteristics of the underlying data patterns of the protein sequence and protein 3D structures. Finally, along with these two modalities, we have also utilized protein interaction information for expediting the efficiency of the proposed model in predicting the protein functions. For the underlying modalities, we have utilized various variations of the convolutional neural network for extracting features from them. As the protein function classes are dependent on each other, we have used a neuro-symbolic hierarchical classification model, which resembles the structure of Gene Ontology (GO), for effectively predicting the dependent protein functions. Finally, to validate the goodness of our proposed method (MultiPredGO), we have compared our results with various uni-modal along with two well-known multi-modal protein function prediction approaches, namely, INGA and DeepGO. Results show that the overall performance of the proposed approach in terms of accuracy, F-measure, precision and recall metrics are better than those by the state-of-the-art methods. This study introduces a methodology design involving empirical mode decomposition (EMD) technique for diagnosis of SZ from EEG signals to perfectly handle the behavior of non-stationary and nonlinear EEG signals. In this study, each EEG signal is decomposed into intrinsic mode functions (IMFs) by the EMD algorithm and then twenty-two statistical characteristics/features are calculated from these IMFs. Among them, five features are selected as significant feature applying Kruskal Wallis test. The performance of the obtained feature set is tested through several renowned classifierson a SZ EEG dataset. Among the considered classifiers, theensemble bagged tree performed as the best classifier producing 93.21% correct classification rate for SZ, with an overall accuracy of 89.59% for IMF 2. These results indicate that EEG signals discriminate SZ patients from healthy control (HC) subjects efficiently and have the potential to become a tool for the psychiatrist to support the positive diagnosis of SZ.In monochrome-color dual-lens systems, the monochrome camera can capture images with higher quality than the color camera.   • Small mammals native to high altitude must sustain high rates of thermogenesis to cope with cold. Skeletal muscle is a key site of shivering and non-shivering thermogenesis, but the importance of mitochondrial plasticity in cold hypoxic environments remains unresolved. We examined high-altitude deer ****, which have evolved a high capacity for aerobic thermogenesis, to determine the mechanisms of mitochondrial plasticity during chronic exposure to cold and hypoxia, alone and in combination. Cold exposure in normoxia or hypoxia increased mitochondrial leak respiration and decreased phosphorylation efficiency and OXPHOS coupling efficiency, which may serve to augment non-shivering thermogenesis. Cold also increased muscle oxidative capacity, but reduced the capacity for mitochondrial respiration via complex II relative to complexes I and II combined. High-altitude **** had a more oxidative muscle phenotype than low-altitude ****. Therefore, both plasticity and evolved changes in muscle mitochondria contributeomplex II relative to the combined capacity of complexes I and II was consistently reduced in both cold environments.     We evaluated the efficacy and safety of laparoscopic ureterolithotomy (LPU) for the treatment of large proximal ureteric stone.

    A retrospective multicenter analysis for patients with solitary impacted proximal ureteric stone ≥15 mm who underwent LPU from 2016 to 2019 was performed. Primary outcome was to estimate the stone-free rate (SFR). SFR was defined as absence of residual stones on postoperative computed tomography scan. Secondary outcome was to assess the perioperative outcomes, as well as to review literature data of randomized controlled trials and meta-analyses comparing LPU to other treatment options.

    Forty-four patients were included in our study. Mean stone size was 22.9 ± 5.8 mm and median follow-up was 14 months. Three patients had previous abdominal surgery, one patient had severe degree of scoliosis and six patients failed primary therapy. All stones were extracted successfully (SFR = 100%) without need of auxiliary treatments. Mean operative time and estimated blood loss were 86.6 ± 14ral stones, LPU should be advised as the procedure that has the higher SFR, lower auxiliary treatments, and comparable complication rates to other treatments.The development of in vitro assays for 3D microenvironments is essential for understanding cell migration processes. Melt electrowriting (MEW) is used to fabricate the structural device with defined and intricate radial structures that are filled with Matrigel. Controlling the printing path is necessary to account for the distance lag in the molten jet, the applied electric field, and the continuous direct-writing nature of MEW. Circular printing below a diameter threshold results in substantial inward tilting of the MEW fiber wall. An eight-chamber radial device with a diameter of 9.4 mm is printed. Four different concentrations of Matrigel are dispensed into the radial chambers. Glioblastoma cells are seeded into the center and grow into all chambers within 8 days. The cell spreading area demonstrates that 6 and 8 mg mL-1 of Matrigel are preferred over 2 and 4 mg mL-1 . Furthermore, topographical cues via the MEW fiber wall are observed to promote migration even further away from the cell seeding depot. This work presents several innovations to reduce cryoprotectant (CPA) toxicity and improve tissue cryopreservation, including 1) improved tissue warming rates through radiofrequency metal form and field optimization and 2) an experimentally verified predictive model to optimize CPA loading and rewarming to reduce toxicity. CPA loading is studied by microcomputed tomography (µCT) imaging, rewarming by thermal measurements, and modeling, and viability is measured after loading and/or cryopreservation by alamarBlue and histology. Loading conditions for three common CPA cocktails (6, 8.4, and 9.3 m) are designed, and then fast cooling and metal forms rewarming (up to 2000 °C min-1 ) achieve ≥90% viability in cryopreserved 1-2 mm arteries with various CPAs. Despite high viability by alamarBlue, histology shows subtle changes after cryopreservation suggesting some degree of cell damage especially in the central portions of thicker arteries up to 2 mm.

  • The phycobilisome (PBS) serves as the major light-harvesting system, funnelling excitation energy to both photosystems (PS) in cyanobacteria and red algae. The picosecond kinetics involving the excitation energy transfer has been studied within the isolated systems and intact filaments of the cyanobacterium Anabaena variabilis PCC 7120. A target model is proposed which resolves the dynamics of the different chromophore groups. The energy transfer rate of 8.5 ± 1.0/ns from the rod to the core is the rate-limiting step, both in vivo and in vitro. The PBS-PSI-PSII supercomplex reveals efficient excitation energy migration from the low-energy allophycocyanin, which is the terminal emitter, in the PBS core to the chlorophyll a in the photosystems. The terminal emitter of the phycobilisome transfers energy to both PSI and PSII with a rate of 50 ± 10/ns, equally distributing the solar energy to both photosystems. Finally, the excitation energy is trapped by charge separation in the photosystems with trapping rates estimated to be 56 ± 6/ns in PSI and 14 ± 2/ns in PSII.Excitation energy transfer (EET) and trapping in Anabaena variabilis (PCC 7120) intact cells, isolated phycobilisomes (PBS) and photosystem I (PSI) complexes have been studied by picosecond time-resolved fluorescence spectroscopy at room temperature. Among these scaffold materials, environmentally responsive hydrogels have great prospects in repairing the microenvironment of cardiovascular tissues and vascular regeneration. They provide a new method for the treatment of cardiovascular tissue repair and space-time control for the release of various therapeutic drugs, including small-molecule drugs, growth factors, and stem cells. Herein, this article reviews the occurrence and current treatment of CVD, as well as the repair of cardiovascular injury by several environmental responsive hydrogels systems currently used, mainly focusing on the delivery of growth factors or the application of cell therapy to revascularization. In addition, we will also discuss the enormous potential and personal perspectives of environmentally responsive hydrogels in cardiovascular repair.PURPOSE Örebro County introduced an updated screening program 2016 with primary HPV test for women over 30 years and prolonged screening, increasing the cut-off age from 56-60 to 64-70. METHODS All women in Örebro County, born 1,946 (n = 1,968), were invited to a liquid-based cell sample with primary HPV screening. Samples were analyzed for hrHPV mRNA and positive samples were genotyped. hrHPV positive women were offered to do a conization. RESULTS Out of 809 participants, 31 (3.8%) were hrHPV positive, of these 22 did a conization. Histologically, 5/22 (23%) had LSIL and 5/22 (23%) had HSIL. Normal histology was found in 12/22 (55%). The most prevalent genotypes were HPV 16, 33, 52, 56, and 68. Of the women with HSIL, one case of cervical cancer was confirmed in a recone biopsy after 4 months.

However, the precise role and molecular mechanisms by which PTEN and CD38 are involved in airway remodeling throughout asthma pathogenesis remains poorly understood. This study aimed to elucidate the role of PTEN and CD38 in airway remodeling of asthma. Exposure to tumor necrosis factor-α (TNF-α) in airway smooth muscle (ASM) cells markedly decreased PTEN expression, and increased expression of CD38. Overexpression of PTEN suppressed the expression of CD38 and downregulated proliferation and migration induced by TNF-α stimulation, which was partially reversed by CD38 overexpression. PTEN/CD38 axis regulated Ca2+ levels and cyclic AMP response-element binding protein (CREB) phosphorylation in TNF-α-stimulated ASM cells. The in vitro knockdown of CD38 or overexpression of PTEN remarkably restricted airway remodeling and decreased Ca2+ concentrations and CREB phosphorylation in asthmatic ****. CD38 overexpression abolished the inhibitory effects of PTEN overexpression on airway remodeling. These findings demonstrate that PTEN inhibits airway remodeling of asthma through the downregulation of CD38-mediated Ca2+/CREB signaling, highlighting a key role of PTEN/CD38/Ca2+/CREB signaling in the molecular pathogenesis of asthma.We aimed to perform a pan-metastatic cancer analysis on survival and prognostic factors and to create a prognosis-based classification system. We selected distant metastasis patients from the Surveillance, Epidemiology, and End Results (SEER) database. The associations between the characteristics of the patients at admission and overall survival were determined. A prognosis-based metastatic cancer classification was established based on the identified prognostic factors. The differences in prognosis among these categories were tested. The survival rate and prognostic factors were not consistent across cancers. Three metastatic cancer categories were generated, each with different prognoses. The prognostic differences among the categories were satisfactorily validated. Different metastatic cancer types had homogeneous and heterogeneous survival rates and prognostic factors. A prognosis-based classification system for synchronous distant metastasis cancer patients at admission was created. This classification system reflects the grade of malignancy in metastatic cancers and may guide the prediction of survival and individualized treatment. Moreover, it may have important implications for the management of synchronous metastatic cancers and aid clinicians in properly allocating medical resources to metastatic patients.Long non-coding RNAs (lncRNAs) were found to play roles in various cancers, including nasopharyngeal carcinoma. In this study, we focused on the biological function of the lncRNA FAM133B-2 in the radio-resistance of nasopharyngeal carcinoma. The RNA-seq and qRT-PCR analysis showed that FAM133B-2 is highly expressed in the radio-resistant nasopharyngeal carcinoma cells. The following biochemical assays showed that FAM133B-2 represses the nasopharyngeal carcinoma radio-resistance and also affects the apoptosis and proliferation of nasopharyngeal carcinoma cells. Further investigations suggested that miR-34a-5p targets FAM133B-2 and also regulates the cyclin-dependent kinase 6 (CDK6). All these results suggested that the lncRNA FAM133B-2 might function as a competitive endogenous RNA (ceRNA) for miR-34a-5p in nasopharyngeal carcinoma radio-resistance, thus it may be regarded as a novel prognostic biomarker and therapeutic target in nasopharyngeal carcinoma diagnosis and treatment.Several epidemiologic, clinical and experimental reports indicate that nonsteroidal anti-inflammatory drugs (NSAIDs) could have a potential as anticancer agents. The aim of this study was the evaluation of cytotoxic potential in human glioblastoma cells of novel synthesized NSAID derivatives, obtained by linking, through a spacer, α-lipoic acid (ALA) to anti-inflammatory drugs, such as naproxen (AL-3, 11 and 17), flurbiprofen (AL-6, 13 and 19) and ibuprofen (AL-9, 15 and 21). The effects on the level of gene expression were also determined using quantitative real-time polymerase chain reaction (qRT-PCR) analysis. According to our results, NSAID derivatives exhibited concentration dependent cytotoxic effects on U87-MG cell line when compared with the control group. Moreover, treatment of the most active compounds (AL-3, AL-6 and AL-9) caused upregulation of tumor suppressor gene PTEN and downregulation of some oncogenes such as AKT1, RAF1 and EGFR. In conclusion, our results revealed that AL-3, AL-6 and AL-9 could be suitable candidates for further investigation to develop new pharmacological strategies for the prevention of cancer.

However, the precise role and molecular mechanisms by which PTEN and CD38 are involved in airway remodeling throughout asthma pathogenesis remains poorly understood. This study aimed to elucidate the role of PTEN and CD38 in airway remodeling of asthma. Exposure to tumor necrosis factor-α (TNF-α) in airway smooth muscle (ASM) cells markedly decreased PTEN expression, and increased expression of CD38. Overexpression of PTEN suppressed the expression of CD38 and downregulated proliferation and migration induced by TNF-α stimulation, which was partially reversed by CD38 overexpression. PTEN/CD38 axis regulated Ca2+ levels and cyclic AMP response-element binding protein (CREB) phosphorylation in TNF-α-stimulated ASM cells. The in vitro knockdown of CD38 or overexpression of PTEN remarkably restricted airway remodeling and decreased Ca2+ concentrations and CREB phosphorylation in asthmatic mice. CD38 overexpression abolished the inhibitory effects of PTEN overexpression on airway remodeling.
