Multivariate and univariate analysis unveiled a panel of metabolites responsible for the separation between groups, mostly belonging to ketones, alcohols, alkanes and aldehydes classes. Some metabolites were found similarly altered for all RCC cell lines compared to non-tumorigenic cells, namely 2-ethylhexanol, tetradecane, formaldehyde, acetone (increased) and cyclohexanone and acetaldehyde (decreased). Furthermore, significantly altered levels of cyclohexanol, decanal, decane, dodecane and 4-methylbenzaldehyde were observed in all metastatic RCC cell lines when compared with the non-metastatic ones. This study assesses the potential impact of different control measures for mitigating the burden of COVID-19 in the UK. Methods We used a stochastic age-structured transmission model to explore a range of intervention scenarios, tracking 66·4 million people aggregated to 186 county-level administrative units in England, Wales, Scotland, and Northern Ireland. The four base interventions modelled were school closures, physical distancing, shielding of people aged 70 years or older, and self-isolation of symptomatic cases. We also modelled the combination of these interventions, as well as a programme of intensive interventions with phased lockdown-type rio resulted in a projected 120 000 cases (46 000-700 000) and 50 000 deaths (9300-160 000). Intensive interventions with lockdown periods would need to be in place for a large proportion of the coming year to prevent health-care demand exceeding availability. Interpretation The characteristics of SARS-CoV-2 mean that extreme measures are probably required to bring the epidemic under control and to prevent very large numbers of deaths and an excess of demand on hospital beds, especially those in ICUs. Patients and methods This retrospective study reviewed consecutive cases of head and neck cancer in which patients received a PET-CT scan within 3 weeks of undergoing a neck dissection performed by the Ascension Macomb-Oakland oral and maxillofacial surgery residency program from July 2013 through July 2018. Neck dissection specimens were orientated by the surgeon according to neck level and submitted for histopathologic examination. These histopathologic results served as the standard reference with which preoperative PET-CT results were compared. Results A total of 156 cases were reviewed in this study, 84 of which were included for data analysis.

Twelve healthy subjects (31.3 ± 5.6 years, eight males) underwent 45-60 min of continuous scanning during wakefulness and sleep, yielding one image set every 3.4 s. Concurrent electroencephalography (EEG) data were available in eight subjects. Mean values of the metabolic parameters measured during wakefulness were stable, with coefficients of variation below 7% (average values CMRO2 = 118 ± 12 µmol O2/min/100 g, SvO2 = 67.0 ± 3.7% HbO2, CBF = 50.6 ±4.3 ml/min/100 g). This between-group competition has evolutionary implications on adaptations like in-group cooperation even with non-kin. An emergent property of between-group competition is group dominance. Mechanisms of group dominance in wild animal populations are difficult to study, as they require long-term data on several groups within a population. Here, using long-term data on four neighbouring groups of wild western chimpanzees, we test the hypothesis that group dominance impacts the costs and benefits of between-group competition, measured by territory size and the pressure exerted by neighbouring groups. Larger groups had larger territories and suffered less neighbour pressure compared with smaller groups. Within-group increase in the number of males led to territory increase, suggesting the role of males in territory acquisition. However, variation in territory sizes and neighbour pressure was better explained by group size. This suggests that the bisexually-bonded social system of western chimpanzees, where females participate in territorial behaviour, confers a competitive advantage to larger groups and that group dominance acts through group size in this population. Individuals in the USA and the UK are self-isolating in response to the COVID-19 outbreak. Different motivations may drive their self-isolation some might see value in it (understood as the identified form of autonomous motivation), while others might feel forced into it by authorities or close others (family, friends, neighbourhoods, doctors; the external form of controlled motivation). People who typically prefer company will find themselves spending more time alone, and may experience ill-being uniformly, or as a function of their identified or external motivations for self-isolation. Self-isolation, therefore, offers a unique opportunity to distinguish two constructs coming from disparate literatures. This project examined preference and motivation (identified and external) for solitude, and tested their independent and interacting contributions to ill-being (loneliness, depression and anxiety during the time spent alone) across two weeks. Confirmatory hypotheses regarding preference and motivation were not supported by the data. A statistically significant effect of controlled motivation on change in ill-being was observed one week later, and preference predicted ill-being across two weeks. This between-group competition has evolutionary implications on adaptations like in-group cooperation even with non-kin. An emergent property of between-group competition is group dominance. Mechanisms of group dominance in wild animal populations are difficult to study, as they require long-term data on several groups within a population. Here, using long-term data on four neighbouring groups of wild western chimpanzees, we test the hypothesis that group dominance impacts the costs and benefits of between-group competition, measured by territory size and the pressure exerted by neighbouring groups. Larger groups had larger territories and suffered less neighbour pressure compared with smaller groups. Within-group increase in the number of males led to territory increase, suggesting the role of males in territory acquisition. However, variation in territory sizes and neighbour pressure was better explained by group size. This suggests that the bisexually-bonded social system of western chimpanzees, where females participate in territorial behaviour, confers a competitive advantage to larger groups and that group dominance acts through group size in this population. A well-functioning and trusting relationship between science, society and policy-makers offers a potent means to thwart and mitigate emergent global challenges.This multi-wave study examined the extent that both preference and motivation for time alone shapes ill-being during self-isolation. Individuals in the USA and the UK are self-isolating in response to the COVID-19 outbreak. Different motivations may drive their self-isolation some might see value in it (understood as the identified form of autonomous motivation), while others might feel forced into it by authorities or close others (family, friends, neighbourhoods, doctors; the external form of controlled motivation). People who typically prefer company will find themselves spending more time alone, and may experience ill-being uniformly, or as a function of their identified or external motivations for self-isolation. Self-isolation, therefore, offers a unique opportunity to distinguish two constructs coming from disparate literatures. This project examined preference and motivation (identified and external) for solitude, and tested their independent and interacting contributions to ill-being (loneliness, depression and anxiety during the time spent alone) across two weeks. Confirmatory hypotheses regarding preference and motivation were not supported by the data. A statistically significant effect of controlled motivation on change in ill-being was observed one week later, and preference predicted ill-being across two weeks. The null hypothesis investigated was that there was no correlation between Raman peak ratios (amide I phosphate ν1 ) and equivalent Knoop microhardness measurements. Ten freshly extracted human permanent teeth with carious dentine lesions were sectioned and examined using high-resolution Raman microscopy.   • Objectives To provide national estimates of the number and clinical and economic burden of medication errors in the National Health Service (NHS) in England. Methods We used UK-based prevalence of medication errors (in prescribing, dispensing, administration and monitoring) in primary care, secondary care and care home settings, and associated healthcare resource use, to estimate annual number and burden of errors to the NHS. Two options of the Centers for Medicare and Medicaid Services Kidney Care Choices model-the Kidney Care First option and the Comprehensive Kidney Care Contracting option-now include patient activation as a quality metric; both models specifically name the patient activation measure (PAM) as the patient-reported outcome to use when assessing activation in kidney disease. Because nephrology practices participating in these models will receive capitated payments according to changes in patients' PAM scores, it is time to more critically evaluate this measure as it applies to patients with kidney disease. In this review, we raise important issues related to the PAM's applicability to kidney health, review and summarize existing literature that applies this measure to patients with kidney disease, and outline key elements to consider when implementing the PAM into practice and policy. Our aim is to spur further dialogue regarding how to assess and address patient activation in kidney disease to facilitate best practices for supporting patients in the successful management of their kidney health.Cardiac hypertrophy is a common feature in patients with CKD. With decreasing renal function, elevated fibroblast growth factor-23 and decreased α-Klotho may contribute to cardiac hypertrophy by targeting the heart directly or by inducing systemic changes, such as vascular injury, hemodynamic disorders, and inflammation. However, several studies have demonstrated that disturbances in the fibroblast growth factor-23/α-Klotho axis do not lead to cardiac hypertrophy. In this review, we describe the cardiac effects of the fibroblast growth factor-23/α-Klotho axis and summarize recent progress in this field. In addition, we present not only the main controversies in this field but also provide possible directions to resolve these disputes.Background Uremic toxicity may play a role in the elevated risk of developing cognitive impairment found among patients with CKD. Some uremic toxins, like indoxyl sulfate, are agonists of the transcription factor aryl hydrocarbon receptor (AhR), which is widely expressed in the central nervous system and which we previously identified as the receptor of indoxyl sulfate in endothelial cells. Methods To characterize involvement of uremic toxins in cerebral and neurobehavioral abnormalities in three rat models of CKD, we induced CKD in rats by an adenine-rich diet or by 5/6 nephrectomy; we also used AhR-/- knockout **** overloaded with indoxyl sulfate in drinking water. We assessed neurologic deficits by neurobehavioral tests and blood-brain barrier disruption by SPECT/CT imaging after injection of 99mTc-DTPA, an imaging marker of blood-brain barrier permeability. Results In CKD rats, we found cognitive impairment in the novel object recognition test, the object location task, and social memory tests and an increase of blood-brain barrier permeability associated with renal dysfunction.

  • These data suggest that the antitumor response driven by antibody-cytokine fusions proceeds through an oligoclonal expansion and activation of tumor-infiltrating CD8+ T cells.Rotavirus infections have become one of the most common causes of infectious gastroenteritis in children. Although rotavirus infections have been intensively studied in infants and young children, the study in adults has been limited. As such, this study assessed the prevalence of rotaviruses and performed the molecular characterization of rotaviruses circulating in Thai adults experiencing acute gastroenteritis between January 2018 and December 2018. Group A human rotaviruses were detected in 100 feces samples by rapid immunochromatography. The peak incidence of infection occurred in February and began to decline in the summer months. From January 2018 to December 2018, there were 1344 acute gastroenteritis adult cases in the Hospital for Tropical Diseases, Bangkok, Thailand. Among these, 310 cases were rotavirus-suspected cases. Only 100 samples tested positive for rotavirus via an immunochromatography test. Twentynine out of the 100 rotavirus-positive samples were further characterized by real-time polymerase chain reaction. The G3[P8] strain was identified as the most prevalent (31.0%) followed by G1P[8], G8P[8] and G9P[8], and G2P[8], which accounted for 20.8%, 17.2%, and 13.8%, respectively. Because of the detection of rare rotavirus genotypes, such as G8, the surveillance of rotavirus epidemiology is crucial in monitoring new emergences of rotavirus strains, leading to a better understanding of the effects of strain variation for further vaccine development.Arabinogalactan-proteins (AGPs) are a family of plant extracellular proteoglycans implicated in many physiological events. AGP is decorated with type II arabinogalactans (AGs) consisting of a β-1,3-galactan backbone and β-1,6-galactan side chains, to which other sugars are attached. Based on the fact that a type II AG-specific inhibitor, β-Yariv reagent, perturbs growth and development, it has been proposed that type II AGs participate in the regulation of cell shape and tissue organization. However, the mechanisms by which type II AGs participate have not yet been established. Here, we describe a novel system that causes specific degradation of type II AGs in Arabidopsis, in which a gene encoding a fungal exo-β-1,3-galactanase that specifically hydrolyzes β-1,3-galactan backbones of type II AGs is expressed under the control of dexamethasone-inducible promoter. Dex treatment increased the galactanase activity, leading to a decrease in Yariv reagent-reactive AGPs in transgenic Arabidopsis. We detected the typical oligosaccharides released from type II AGs by Il3GAL in the soluble fraction, demonstrating that Il3GAL acted on type II AG in the transgenic plants. Here, we evaluate hypotheses relating to the neuroendocrine basis of sex-role reversal. We refute the most widely tested activational hypothesis for sex differences in androgen secretion; sex-role reversed females do not have higher levels of androgens in circulation than males. However, we find some evidence that the effects of androgens may be sex-specific; circulating androgen levels correlate with some competitive phenotypes in sex-role reversed females. We also review evidence that sex-role reversed females have higher tissue-specific sensitivity to androgens than males, at least in some species and tissues. Organizational effects may explain these relationships, considering that early exposure to sex steroids can shape later sensitivity to hormones, often in sex-specific ways. Moving forward, experimental and correlative studies on the ontogeny and expression of sex-role reversal will further clarify the mechanisms that generate sex-specific behaviors and sex roles.

We refute the most widely tested activational hypothesis for sex differences in androgen secretion; sex-role reversed females do not have higher levels of androgens in circulation than males. However, we find some evidence that the effects of androgens may be sex-specific; circulating androgen levels correlate with some competitive phenotypes in sex-role reversed females. We also review evidence that sex-role reversed females have higher tissue-specific sensitivity to androgens than males, at least in some species and tissues. Organizational effects may explain these relationships, considering that early exposure to sex steroids can shape later sensitivity to hormones, often in sex-specific ways. Moving forward, experimental and correlative studies on the ontogeny and expression of sex-role reversal will further clarify the mechanisms that generate sex-specific behaviors and sex roles.

A total of 1114 individuals, aged ≥19 years, with healthy phenotype in the third examination cycle of Tehran Lipid and Glucose Study were followed until the fifth examination. Dietary intakes were estimated using the food frequency questionnaires, collected during the study. The participants were grouped as normal weight and overweight/obese according to the baseline body mass index (BMI). Unhealthy metabolic status was determined as having more than one component of metabolic syndrome using the Joint Interim Statement. Regardless of BMI, higher intakes of magnesium, dairy products, poultry, apples/pears, citrus fruits, and tea/coffee decreased the risk of developing unhealthy phenotype, while higher intakes of fast foods, organ meats, and potatoes increased the risk. Food intake can influence the transition to unhealthy phenotype in metabolically healthy adults, irrespective of the BMI status.

  • Introduction The purpose of this study was to identify homogenous subgroups of adolescents with emotional and behavioural difficulties using latent profile analysis. The relationship between the latent profile classes and socio-emotional and school adjustment was also analysed. Materials and methods A total of 1,506 students, 667 men (44.3%) were selected by random cluster sampling. The mean age was 16.15 years (SD=1.36). The instruments used were the Strengths and Difficulties Questionnaire, the Personal Wellbeing Index-School Version, The 10-item Positive and Negative Affect Schedule for Children, the Prodromal Questionnaire Brief, the Paykel Suicide Scale, the Cyberbullying-Brief Questionnaire, the Penn Matrix Reasoning Test, and the school performance assessment. Results A total of 7.7% of adolescents obtained risk scores for mental health difficulties. We identified 3 mental health subgroups named Low risk, Externalizing, and Internalizing. These 3 latent classes demonstrated a differential pattern in terms of socio-emotional adjustment and academic performance. Conclusions To identify and validate risk groups of youths for emotional and behavioural problems allows implementing measures to promote emotional well-being and prevent mental health problems. Future studies should continue to analyse young people with emotional and behavioural problems longitudinally and to incorporate new forms of assessment and intervention.Recently implemented photon optimizer (PO) ****optimization algorithm is mandatory for RapidPlan modeling in Eclipse. This report quantifies and compares the dosimetry and treatment delivery parameters of PO vs its predecessor progressive resolution optimizer (PRO) algorithm for a single-dose of volumetric modulated arc therapy (VMAT) lung stereotactic body radiation therapy (SBRT). Clinical SBRT treatment plans for 12 early-stage non-small-cell lung cancer patients receiving 30 Gy in 1 fraction using PRO-VMAT were re-optimized using the PO-VMAT ****algorithm with identical planning parameters and objectives. Average planning target volume derived from the 4D CT scans was 13.6 ± 12.0 cc (range 4.3 to 41.1 cc) Patients were treated with 6 MV flattening filter free beam using Acuros-based calculations and 2.5 mm calculation grid-size (CGS). Both treatment plans were normalized to receive same target coverage and identical CGS to isolate effects of ****positioning optimizers. Original PRO and re-optimized PO plaaverage of 0.2 ± 0.1 minutes (up to 1.0 minutes) with PO 2.5 mm and 1.2 ± 0.39 minutes (maximum up to 3.22 minutes) with PO 1.25 mm plans compared to PRO 2.5 mm plans. PO-VMAT single-dose of VMAT lung SBRT plans showed slightly increased intermediate-dose spillage but boasted overall similar plan quality with less beam modulation and hence shorter beam-on time. However, PO 1.25 mm CGS had less intermediate-dose spillage and analogous plan quality compared to clinical PRO-VMAT plans with no additional cost of plan optimization. Further investigation into peripheral targets with PO-****algorithm is warranted. Methods Single-session 1H-/31P-Chemical Shift Imaging of thalamus, prefrontal (DLPFC) and anterior midcingulate (aMCC) cortices was applied to 69 UHR patients for psychosis and 61 matched healthy controls. N-acetylaspartate (NAA), glutamate/glutamine complex (Glx), energy (PCr, ATP) and phospholipid metabolites were assessed, analysed by ANOVA (or ANCOVA [with covariates]) and correlated with symptomatology (SCL-90R). Results The thalamus showed decreased NAA, inversely correlated with self-rated aggressiveness, as well as increased PCr, and altered phospholipid breakdown. While the aMCC showed a pattern of NAA decrease and PCr increase, the DLPFC showed PCr increase only in the close-to-psychosis patient subgroup. There were no specific findings in transition patients. Conclusion The results do not support the notion of a specific pre-psychotic neurometabolic pattern, but likely reflect correlates of an "at risk for mental disorders syndrome". This includes disturbed neuronal (mitochondrial) metabolism in the thalamus and aMCC, with emphasis on left-sided structures, and altered PL remodeling across structures.Introduction There is no consensus on how to perform acromioplasty, particularly regarding the level and extent of bone resection, which depend on scapular and humeral morphologies. Hypothesis We aimed to determine whether computer-assisted acromioplasty planning helps surgeons remove impinging bone, reduce unnecessary resections, and improve short-term outcomes of rotator cuff tears (RCR). Patients and methods We randomized 64 patients undergoing RCR of full-thickness supraspinatus tears into two groups 'guided acromioplasty' (GA) and 'freehand acromioplasty' (FA). The pre- and post-operative scapula models were reconstructed using computed-tomography scans to quantify impinging bone removal, unnecessary bone resections, and identify zones of acromial bone removal. All patients were evaluated preoperatively and at 6 months to assess their range of motion (ROM), functional scores and tendon integrity using ultrasound. Results The two groups did not differ in demographics, clinical or morphologic characteristics. Compared to FA, GA tended to lower impinging bone removal (55±26% vs. 43±27%, p=0.087) and to increase unnecessary resection of the total bone removed (49±22% vs. 57±27%, p=0.248). GA resulted in significant anterior under-resection, while FA resulted in significant medial over-resection. Clinical outcomes and ROM improved significantly for all patients, except for internal rotation in the GA group.
