A bioinformatics analysis of the currently predicted GroEL-like proteins encoded by bacteriophage genomes was carried out in comparison with the phage double-ring EL and single-ring OBP chaperonins, previously described by us, as well as with the known chaperonins of group I and group II. A novel GroEL-like protein predicted in the genome of phage AR9 Bacillus subtilis was expressed in E. coli cells, purified and characterised by various physicochemical methods. As shown by native electrophoresis, analytical ultracentrifugation and single-particle electron microscopy analysis, the putative AR9 chaperonin is a single-ring heptamer. Like the EL and OBP chaperonins, the new AR9 chaperonin possesses chaperone activity and does not require co-chaperonin to function. It was shown to prevent aggregation and provide refolding of the denatured substrate protein, endolysin, in an ATP-dependent manner. A comparison of its structural and biochemical properties with those of the EL and OBP chaperonins suggests outstanding diversity in this group of phage chaperonins. V.A novel sulfonated chitosan-derived carbon-based catalyst was successfully prepared via isoamyl nitrite-assisted sulfanilic acid sulfonation, and its catalytic activity was examined using dehydration of fructose. The structural and chemical properties of sulfonated chitosan-derived carbon were characterized by SEM, FTIR, XRD, XPS, element analysis, N2 adsorption-desorption experiment, and acid-base titration experiment. KOH was used as activating agent in the synthesizing of carbon supports, and it was found that properly increasing the dose of KOH during activation stage had a positive effect on the subsequent sulfonation of prepared activated carbon. 4KSCC, with the highest sulfonation degree (2.04 mmol/g), exhibited high performance for the conversion of fructose to HMF in various solvent, and an optimal HMF yield of 80.9% was obtained at 140 °C in 40 min. In addition, the reusability of 4KSCC for fructose dehydration was fairly good. In recent years, the use of food grade natural biodegradable particles as Pickering emulsion stabilizer has attracted wide attentions. In this study, chitosan/gum Arabia (CS/GA) nanoparticles were prepared and their potential use in stabilizing Pickering emulsion and delivering curcumin were evaluated. It was found that CS and GA combined mainly through electrostatic interactions, and the obtained nanoparticles were about 100 nm of size and displayed higher surface activity than chitosan nanoparticles. Fluorescence microscopy showed that the nanoparticles accumulated at the oil-water interface. The environmental stability of Pickering emulsion got improved with the increase of nanoparticle concentration, and was sensitive to the changes of pH and ionic strength, while the emulsion remained stable under all test temperatures. The Pickering emulsion stabilized by 0.75% CS/GA nanoparticles displayed higher curcumin embedding rate of 94%, and also showed improved protection on curcumin during storage and controlled release during in vitro digestion. These results confirmed that the CS/GA nanoparticle stabilized-Pickering emulsion could be used as an effective deliver system for bioactive substances. Here, the complex structure of ulvan from Ulva pertusa was analyzed using specific ulvan lyase degradation, MS, and NMR detection. Retinal vasculitis and IOI were diagnosed at a mean of 30 days following brolucizumab IVI. Chronic hepatitis C virus (HCV) infection is associated with several hepatic and extrahepatic complications, including cancers and autoimmune disorders, whose frequency is reduced but not abolished after drug-induced viral clearance. We assessed, through a PCR real-time amplification assay, the transcription levels of the pol genes of HERV-H, -K, and -W, and of their repressor TRIM28 in white blood cells (WBCs) of vertically infected children, both before and after therapy with direct-acting antivirals (DAAs). The results documented significantly higher expressions of HERV-H-pol and HERV-K-pol, not of HERV-W-pol, in HCV-infected subjects as compared to age-matched controls. HERV RNA levels remained unchanged after DAA-driven viral clearance. No significant variations in transcription levels of TRIM28 were observed in infected subjects. Our findings demonstrate HERV-H-pol and HERV-K-pol overexpression in subjects with chronic HCV infection, without variations after a positive response to DAAs; this might justify their predisposition to cancers and autoimmune disorders that persist after a DAA-induced resolution of viremia.Ezetimibe reduces cardiovascular risk by lowering the levels of low-density lipoprotein cholesterol (LDL-C). However, there is limited information regarding the factors associated with ezetimibe-mediated LDL-C reduction. We investigated the factors associated with LDL-C reduction after ezetimibe administration in Japanese patients with or without type 2 diabetes mellitus (T2DM). This single-center retrospective observational study enrolled a total of 266 consecutive ezetimibe-naïve patients, of which 154 were excluded because of either switching from statin or fenofibrate to ezetimibe (n = 52) or ezetimibe discontinuation (n = 102). Finally, 112 patients were eligible for analysis. To identify the factors influencing LDL-C levels, univariate and multivariate linear regression analyses were performed after 52 weeks of ezetimibe treatment. Overall, advanced age, T2DM, and high baseline LDL-C were significantly associated with a greater decrease in LDL-C levels. In the non-T2DM group, advanced age and high baseline LDL-C were associated with greater decrease in LDL-C levels. In the T2DM group, baseline LDL-C was the only factor that influenced the change in LDL-C levels. Advanced age was significantly associated with higher LDL-C reduction in non-T2DM patients, but not in T2DM patients. Ezetimibe use might be beneficial in older patients without T2DM. The lack of association between age and the LDL-C lowering effect by ezetimibe in patients with T2DM may be due to yet unknown mechanism except low statistical power.Maternal high-fructose diets (HFD) impair the learning and memory capacity of adult female offspring via histone deacetylase 4 (HDAC4). Hippocampal adult neurogenesis is important for supporting the function of existing neural circuits. In this study, we investigated the effects of maternal HFD on hippocampal neural stem cell (NSC) proliferation and neuronal differentiation in adult offspring. Increased nuclear HDAC4 enzyme activity was detected in the hippocampus of HFD female offspring. The Western blot analyses indicated that the expressions of sex-determining region Y box2 (SOX2) and the transcription factor Paired Box 6 (PAX6), which are critical for the progression of NSC proliferation and differentiation, were downregulated. Concurrently, the expression of Ki67 (a cellular marker for proliferation) and doublecortin (DCX), which are related to NSC division and neuronal differentiation, was suppressed. Intracerebroventricular infusion with class II HDAC inhibitor (Mc1568, 4 weeks) led to the upregulation of these proteins. Environmental stimulation reversed the expression of Ki67 and DCX and the counts of Ki67- and DCX-positive cells in the hippocampi of HFD offspring as a result of providing the enriched housing for 4 weeks. Together, these results demonstrate that the suppressive effects of maternal HFD on hippocampal NSC proliferation and neuronal differentiation are reversibly mediated through HDAC4 and can be effectively reversed by environmental stimulation. Data were based on outpatient drug prescriptions collected from administrative databases. The study population included all patients with at least one drug prescription in 2018. Prevalence was used as a measure to estimate the degree of exposure to drugs. A total of 3,899,360 patients were treated with at least one drug (54.2% females). The number of prescriptions was higher in females than males (55.6% vs. 44.4%). Females recorded higher prevalence for the majority of therapeutic groups (ATC II-anatomical therapeutic chemical), as well as for anti-inflammatory and antirheumatic products drugs (M01) (25.6% vs. 18.7%, risk ratio (RR) 0.73), beta blocking agents (C07) (14.5% vs. 11.6%, RR 0.80), psychoanaleptics (N06) (7.1% vs. 3.7%, RR 0.52), and antianemic preparations (B03) (2.8% vs. 6.7%, RR 0.4). Higher prevalence was identified for males only for drugs used in diabetes (A10) (6.8% vs. 6.2%, RR 1.1), particularly for biguanides (A10BA). Conversely, treatment duration was longer among males, explaining the higher mean cost per treated patient. This real-world study showed substantial gender differences in terms of medication use and duration of treatment and costs. These results are relevant to promoting and supporting the emerging role of precision and personalized medicine.The S100A6 protein is present in different mammalian cells and tissues including the brain. It binds Ca2+ and Zn2+ and interacts with many target proteins/ligands. The best characterized ligands of S100A6, expressed at high level in the brain, include CacyBP/SIP and Sgt1. Research concerning the functional role of S100A6 and these two ligands indicates that they are involved in various signaling pathways that regulate cell proliferation, differentiation, cytoskeletal organization, and others. In this review, we focused on the expression/localization of these proteins in the brain and on their possible role in neurodegenerative diseases. Published results demonstrate that S100A6, CacyBP/SIP, and Sgt1 are expressed in various brain structures and in the spinal cord and can be found in different cell types including neurons and astrocytes. When it comes to their possible involvement in nervous system pathology, it is evident that their expression/level and/or subcellular localization is changed when compared to normal conditions.

The biosynthesis of AgNPs was visually validated through the appearance of reddish-brown color and further confirmed by the UV-spectra at 418 nm. The TEM and FE-SEM studies revealed the spherical shape of particles with size ranged between 10-50 nm. Face centered cubic crystalline nature of AgNPs was proved by XRD analysis. The negative value of zeta potential (-21.7) indicated the stability of AgNPs and elemental composition was confirmed by EDS. FT-IR analysis revealed the functional groups present in the plant extract trigger the biosynthesis of AgNPs. The AgNPs exhibited strong degradation of MO (86.68%), MB (93.60%), CR (92.20%) and 4NP (88.80%) by completing the reduction reaction within 20 min. The reaction kinetics followed the pseudo-first-order and displayed k-values (rate constant) 0.166 min-1, 0.138 min-1, 0.182 min-1 and 0.142 min-1 for MO, MB, CR and 4-NP respectively. Despite its widespread use, there is confusion arising from the recommended cutoff score for a positive diagnosis. This study aimed to identify the appropriate cutoff score for IGDS9-SF in a Chinese context. Methods The present study included a sample from clinical settings (n = 131) and another from universities (n = 3742). IGDS9-SF measurement and structured clinical interviews based on DSM-5 criteria for IGD were conducted in the sample from clinical settings. The cutoff score was determined using the receiver operating characteristics (ROC) curve. The validity of this cutoff score was further assessed in a sample from universities. Results Mathematical models suggest that the score of 32 is the optimal cutoff point (Youden's index, 96.2%; diagnostic accuracy, 96.1%; sensitivity, 98.0%; specificity, 91.9%; NPV, 91.9%; and NPY, 100%). The prevalence of IGD is 2.9% in this study. Conclusion This study suggested that the optimal cutoff score of IGDS9-SF is 32 for the positive diagnosis of IGD in a Chinese context.Pranayama refers to a set of yoga breathing exercises. Recent evidence suggests that the practice of pranayama has positive effects on measures of clinical stress and anxiety. This study explored the impact of a Bhastrika pranayama training program on emotion processing, anxiety, and affect. We used a randomized controlled trial design with thirty healthy young adults assessed at baseline and after 4 weeks of pranayama practices. Two functional magnetic resonance imaging (MRI) protocols were used both at baseline and post-intervention an emotion task as well as a resting-state acquisition. Our results suggest that pranayama significantly decreased states of anxiety and negative affect. The practice of pranayama also modulated the activity of brain regions involved in emotional processing, particularly the amygdala, anterior cingulate, anterior insula, and prefrontal cortex. Resting-state functional MRI (fMRI) showed significantly reduced functional connectivity involving the anterior insula and lateral portions of the prefrontal cortex. Correlation analysis revealed that changes in connectivity between the ventrolateral prefrontal cortex and the right anterior insula were associated with changes in anxiety. Although it should be noted that these analyses were preliminary and exploratory, it provides the first evidence that 4 weeks of B. pranayama significantly reduce the levels of anxiety and negative affect, and that these changes are associated with the modulation of activity and connectivity in brain areas involved in emotion processing, attention, and awareness. Methods A phenomenological qualitative approach was used to analyze self-reported experiences with 'Spice' (NPS) among users in prison. A semi-structured questionnaire was also disseminated among professionals working in these settings to better understand (a) the impact of NPS on their work; (b) perceived issues on safety in their working environment; (c) approaches used to tackle the phenomenon and best practices. Results Psychotic events resulting from the collected Spice accounts (5) were marked by hallucinations, depression, self-harm, and suicidal ideations. The Children's Depression Inventory was used to assess the participants' depressive symptoms. ST, FFs and SSBs consumption was measured by a self-reported questionnaire. The Bayesian multiple mediation model was used to analyze the mediation effect. We developed a simple method of analyzing the strontium (Sr) and calcium (Ca) content of intact eggshell samples in support of a broader study of how dietary Sr uptake impacts waterfowl eggshell quality. We used wavelength dispersive - x-ray fluorescence spectrometry (WD-XRF) to analyze eggshell pieces ranging in size from ∼6-mm2 fragments to intact half-shells. We verified this approach on a subset of reference shells by subjecting the same region and volume of shell material from which x-ray signals were measured to analysis by inductively-coupled plasma mass spectrometry (ICP-MS). An analysis of the sources of analytical uncertainty yielded total internal error estimates of ±0.3 and 5% relative for Ca and Sr, respectively, on the basis of which the chemistry of intact shell material analyzed by WD-XRF in this study is compared. The total external errors associated with the WD-XRF results of this study in relation to certified reference material (National Institute of Standards and Technology [NIST] 1400 [a bone ash]) are ±9 and 13.5% relative for Ca and Sr, respectfully (95% CL). Our results demonstrate this method is acceptably accurate and precise for many wildlife management applications. WD-XRF analysis is a quick and inexpensive alternative to traditional methods for determining eggshell Sr and Ca that require acid digestion, allowing for generation of larger datasets that might otherwise be cost-prohibitive, while preserving sample material intact.Contamination by per- and polyfluoroalkyl substances (PFASs) is of great concern in global environments. Due to strong regulation of legacy PFASs, emerging PFASs including alternatives and precursors have been introduced to the industrial market. In this study, legacy and emerging PFASs were measured in seawater, sediment, and bivalves collected along the Korean coast to investigate the occurrence, distribution, contamination sources, and bioaccumulation potential of PFASs. Wide concentration ranges of legacy PFASs were detected in multiple environmental samples, indicating widespread contamination. C8-based PFASs (e.g., PFOA and PFOS) were still major contaminants in all of the environmental samples. Some precursors, such as 82 fluorotelomer sulfonate (82 FTS) and N-ethyl-perfluorooctane sulfonamidoacetic acid (N-EtFOSAA), and perfluoro-2-propoxypropanoic potassium 9-chlorohexadecafluoro-3-oxanonane-1-sulfonate (F-53B), an alternative to PFOS, were detected in sediment or bivalve samples, implying a shift in consumption patterns from legacy to emerging PFASs. The highest concentrations of PFASs in environmental samples were found at the locations near industrial complexes, such as those for the semi-conductor, paper mill, automobile, and metal-plating industry. This result indicates that PFAS contamination is associated with intensive industrial activities in the coastal environment. Matrix-dependent contamination and profiles of PFASs were observed. Seawater was dominated by short-chained PFASs as a prompt reflection of regulation, while the sediment and bivalves were dominated by long-chained PFASs. Carbon-chain length was a major factor governing environmental behavior and bioaccumulation of PFASs. In this paper, we investigated for the first time the potential release of disinfection byproducts (DBPs) precursors when MPs are exposed to hydrolysis and/or degradation by simulated sunlight. Seventeen MPs with seven different polymer types were collected either as commercial products (e.g. drinking water bottles, shopping bags, recycled plastics, etc.) or pure/virgin polymers. PBDD/Fs, unlike their chlorinated counterparts (PCDD/Fs), have not been officially assigned toxic equivalence factors (TEFs) by the WHO therefore, we determined their relative potency towards AhR activation in both human and rodent cell-based DR CALUX® bioassays. This allowed us to compare GC-HRMS PBDD/F congener levels, converted to total Toxic Equivalents (TEQ) by using the PCDD/F TEFs, to CALUX Bioanalytical Equivalents (BEQ) levels present in contaminated plastic constituents from children's toys. Finally, an estimate was made of the daily ingestion of TEQs from PBDD/Fs-contaminated plastic toys by child mouthing habits. It is observed that the daily ingestion of PBDD/Fs from contaminated plastic toys may significantly contribute to the total dioxin daily intake of young children.

We studied the characteristics of acute blocking of supination of the distal radioulnar joint caused by triangular fibrocartilage complex injuries. Twenty-four patients who were treated for acute blocking of supination were retrospectively assessed. Supination was suddenly blocked after minor trauma to the wrist. Active and passive supination was severely restricted with a mean preoperative range of motion (11°), whereas pronation was almost normal. The cause was identified arthroscopically or at open operation. It was found to be a result of avulsion of the dorsal or palmar portion of the radioulnar ligament, which blocked movement of the ulnar head. Blocking was reduced manually in four cases, by arthroscopic surgery in eight cases and by open surgery in 12 cases. After treatment, forearm supination improved to 84° of the mean range of motion. Distal radioulnar joint blocking from a ruptured triangular fibrocartilage complex should be considered in the differential diagnosis of loss of forearm supination. Novel surgical approaches including minimally invasive procedures, sutureless aortic valve replacement, and transcatheter aortic valve implantation have been found to be acceptable alternatives to conventional surgeries. In addition, novel endovascular repair techniques and hybrid procedures have been introduced for the management of patients with thoracoabdominal aortic pathologies. However, these modalities are not readily available in every center, and such novel procedures impose a learning curve for surgeons and high costs for affected patients. In this review, we discuss the challenges of setting up an aortic service, having regard to the Iranian experience.A sound knowledge of the functional anatomy of the mitral valve and the alterations caused by different diseases is indispensable for surgeons treating patients with mitral valve disease. Rheumatic mitral valve disease remains the most common heart valvular disorder in developing countries, whereas mitral regurgitation due myxomatous degeneration of the valve is the most common in developed countries. The mitral valve should be repaired whenever possible, as long as the outcome is predictably better than that of replacement. The intraoperative decision to repair or replace is not always simple and depends on the experience of the surgeon and the pathological changes that caused mitral valve dysfunction.Sex workers are an 'at-risk' population facing stigmatisation, marginalisation and poorer health outcomes. Their multiple vulnerabilities include substance misuse, mental illness, homelessness, violence, unique health challenges and importantly, barriers to accessing healthcare. Little is known about the opinions of medical students towards sex workers. A 21-item questionnaire was electronically distributed to all medical students at the University of Sheffield in November 2019. The aim was to investigate the knowledge, perceptions, and attitudes of medical students towards sex workers in Sheffield, in the hope of identifying gaps in the medical curriculum and encouraging further research in the area. One hundred and seventy-seven students participated. Students showed a basic awareness of the occurrence of prostitution in Sheffield but lacked detailed knowledge. The vast majority hope that prostitution is legalised in the UK, primarily for the sake of the health and protection of sex workers involved. A significant number of medical students (29% and 23%, respectively) expressed that their attitudes towards treatment of sex workers would not be without pre-judgement or differential treatment. Clearly, this needs to be addressed. Victim behavior was identified in forensic case files of service members charged with sexual assault (N = 58) and referred for forensic evaluation or consultation. The identified victim behavior was coded and quantified for description. This study also examined the extent to which socio-demographic variables predicted formal help seeking among male victims of IPV. Data were drawn from 2009 and 2014 cycles of the Canadian General Social Survey on Victimization with a nationally representative sample of 52,400 respondents. It is estimated that about 655,400 men in Canada reported having experienced physical and/or sexual victimization due to IPV in married/common-law relationships at the time of the surveys between 2004 and 2014. The latent class analysis generated four types of IPV victimization among men. Among male victims of physical and/or sexual IPV, about 64,000 men experienced the most severe type of IPV characterized by chronic and severe physical and psychological violence with a high probability of injuries and negative emotional effects of IPV. Although most of the male victims of IPV did not seek formal help (e.g., did not contact the police and IPV services), the severity of experienced violence was associated with the increased use of formal services. Some structural factors, such as being unemployed and residing with young children, were found to be substantial barriers to contacting formal agencies for help. Our findings highlight the need for the development of gender-inclusive and gender-sensitive public policy and intervention programs that help all victims of IPV regardless of victim gender.

Rheumatic mitral valve disease remains the most common heart valvular disorder in developing countries, whereas mitral regurgitation due myxomatous degeneration of the valve is the most common in developed countries. The mitral valve should be repaired whenever possible, as long as the outcome is predictably better than that of replacement. The intraoperative decision to repair or replace is not always simple and depends on the experience of the surgeon and the pathological changes that caused mitral valve dysfunction.Sex workers are an 'at-risk' population facing stigmatisation, marginalisation and poorer health outcomes. Their multiple vulnerabilities include substance misuse, mental illness, homelessness, violence, unique health challenges and importantly, barriers to accessing healthcare. Little is known about the opinions of medical students towards sex workers. A 21-item questionnaire was electronically distributed to all medical students at the University of Sheffield in November 2019. The aim was to investigate the knowledge, perceptions, and attitudes of medical students towards sex workers in Sheffield, in the hope of identifying gaps in the medical curriculum and encouraging further research in the area. One hundred and seventy-seven students participated. Students showed a basic awareness of the occurrence of prostitution in Sheffield but lacked detailed knowledge. The vast majority hope that prostitution is legalised in the UK, primarily for the sake of the health and protection of sex workers involved. A significant number of medical students (29% and 23%, respectively) expressed that their attitudes towards treatment of sex workers would not be without pre-judgement or differential treatment. Clearly, this needs to be addressed. This study also examined the extent to which socio-demographic variables predicted formal help seeking among male victims of IPV. Data were drawn from 2009 and 2014 cycles of the Canadian General Social Survey on Victimization with a nationally representative sample of 52,400 respondents. It is estimated that about 655,400 men in Canada reported having experienced physical and/or sexual victimization due to IPV in married/common-law relationships at the time of the surveys between 2004 and 2014. The latent class analysis generated four types of IPV victimization among men. Among male victims of physical and/or sexual IPV, about 64,000 men experienced the most severe type of IPV characterized by chronic and severe physical and psychological violence with a high probability of injuries and negative emotional effects of IPV. Although most of the male victims of IPV did not seek formal help (e.g., did not contact the police and IPV services), the severity of experienced violence was associated with the increased use of formal services. Some structural factors, such as being unemployed and residing with young children, were found to be substantial barriers to contacting formal agencies for help. The impact of pod storage (PS) and roasting temperature (RT) on the quality parameters and the sensory profiles of dark chocolates were evaluated. Dark chocolates (70%) from ten liquors of different PS and RT combinations as well as six liquors of different origins (Ecuador, Ghana, Ivory Coast, Madagascar, Venezuela and Vietnam) with variable genetic groups were produced under identical conditions and compared. To a greater extent, the range of chocolate quality attributes underscored the generally minimal effects of PS, RT and origin of liquor on the processing conditions. Although with a few exceptions, generally, chocolate acidity (pH and TA) decreased with increasing PS and vice versa in the case of RT. Furthermore, results from a balanced incomplete block design (BIBD) involving a 16-member expert panel also revealed the impact of the applied treatments (PS and RT) on the final flavor profiles of the chocolates irrespective of the origin or genetic groups of the cocoa beans. The same was confirmed when instrumental aroma results were correlated with the sensory data using partial least squares (PLS) regression models. Thus, this study demonstrates the possibility of creating diverse flavor profiles (even towards 'fine' flavor) from 'bulk' cocoa beans through an optimized combination of PS and RT. The findings are therefore expected to challenge the status-quo, especially in the way 'bulk' cocoa is currently processed and consequently priced, thereby, possibly fostering a win-win situation between cocoa producers and industries. Indeed, tea flower extract (TFE) is a rich source of functional molecules, but its nutritional value remains unclear. This study, from the perspective of "whole food", aimed to investigate the composition of TFE and further explore its possible health-promoting effects on cyclophosphamide-induced ****. It was found that TFE was mainly composed of carbohydrates (34.02 ± 1.42%), phenolic compounds (11.57 ± 0.14%), crude proteins (27.72 ± 3.07%) and saponins (2.81 ± 0.00%). Supplementation of TFE at 200 mg/kg·BW/d regulated intestinal homeostasis by improving the intestinal barrier, alleviating dysbacteriosis (reverse 44 of 68 disordered genera), stimulated immunoreactions with significant enhancement of serum TNF-α, IFN-γ, IL-1β, IL-2 and IL-6. Furthermore, TFE could improve the liver function through decreasing the hepatic malondialdehyde and aminotransferase levels and increasing the levels of catalase, myeloperoxidase, superoxide dismutase and reduced glutathione. Assay concordance was assessed by overall percent agreement, positive percent agreement (PPA), negative percent agreement (NPA), and Cohen's kappa coefficient.Results Overall percent agreement between the assays was 84.0% (95% confidence interval [CI] 75.3 to 90.6%), PPA was 68.0% (95% CI 53.3 to 80.5%) and the kappa coefficient was 0.68 (95% CI 0.54 to 0.82). Sixteen specimens detected by the SHC-LDT were not detected by the Accula test, and showed low viral load burden with a median cycle threshold value of 37.7. Current tests that measure anti-streptolysin-O (ASO) and anti-DNaseB (ADB) titres require parallel processing, with predictive value limited by the slow rate of decay in antibody response. Accordingly, our objective was to develop and assess the diagnostic potential of a triplex bead-based assay, which simultaneously quantifies ASO and ADB together with titres for a third antigen, SpnA.Methods Our previous cytometric bead assay was transferred to the clinically appropriate Luminex platform by coupling streptolysin-O, DNaseB and SpnA to spectrally unique magnetic beads. Sera from over 350 subjects, including 97 ARF patients, were used to validate the assay and explore immunokinetics.Results Operating parameters demonstrate the triplex assay produces accurate and reproducible antibody titres which, for ASO and ADB, are highly correlative with existing assay methodology. When ARF patients were stratified by time (days following hospital admission) there was no difference in ASO and ADB between less then 28 and 28+ days groups. Biomarker levels were compared among those with no traumatic brain injury (TBI) (controls), 1-2 mTBIs, and repetitive (3 or more) mTBIs. Results Elevated exosomal and plasma levels of NfL were associated with repetitive mTBIs and with chronic PCS, PTSD, and depression symptoms. Performance was equivalent (P >0.48) to bulk stool for all targets when 50 μl of FecalSwab™ specimen was loaded onto the BD MAX™ assays. As stool specimens are often collected off-site from the clinical microbiology laboratory and require transport, we assessed the stability of stool specimens stored for up to 14 days at 40C, 220C, or 350C to account for varying transportation conditions. Molecular detection for the majority of viral targets (excluding astrovirus) was unaffected (ΔCT ≤1) by sample storage temperature over the two-week period; however, detection of enteric bacteria was variable if specimens were not refrigerated (220C or 350C). By demonstrating equivalent performance to matched bulk stool and maintaining molecular detection sensitivity when stored at 4°C, we suggest that the FecalSwab™ is a suitable specimen type for enteropathogen diagnostics on the BD MAX™ System.Objectives Streptococcal serology is a cornerstone in the diagnosis of acute rheumatic fever (ARF), a post-infectious sequalae associated with Group A Streptococcus infection. Current tests that measure anti-streptolysin-O (ASO) and anti-DNaseB (ADB) titres require parallel processing, with predictive value limited by the slow rate of decay in antibody response. Accordingly, our objective was to develop and assess the diagnostic potential of a triplex bead-based assay, which simultaneously quantifies ASO and ADB together with titres for a third antigen, SpnA.Methods Our previous cytometric bead assay was transferred to the clinically appropriate Luminex platform by coupling streptolysin-O, DNaseB and SpnA to spectrally unique magnetic beads. Sera from over 350 subjects, including 97 ARF patients, were used to validate the assay and explore immunokinetics.Results Operating parameters demonstrate the triplex assay produces accurate and reproducible antibody titres which, for ASO and ADB, are highly correlative with existing assay methodology. When ARF patients were stratified by time (days following hospital admission) there was no difference in ASO and ADB between less then 28 and 28+ days groups. We examined relationships between candidate biomarkers and symptoms of postconcussive syndrome (PCS), posttraumatic stress disorder (PTSD), and depression. Biomarker levels were compared among those with no traumatic brain injury (TBI) (controls), 1-2 mTBIs, and repetitive (3 or more) mTBIs. Results Elevated exosomal and plasma levels of NfL were associated with repetitive mTBIs and with chronic PCS, PTSD, and depression symptoms. Kushenol C (KC) is a prenylated flavonoid isolated from the roots of Sophora flavescens aiton. Little is known about its anti-inflammatory and anti-oxidative stress activities. Here, we investigated the anti-inflammatory and anti-oxidative stress effects of KC in lipopolysaccharide (LPS)-stimulated RAW264.7 macrophages, and tert-butyl hydroperoxide (tBHP)-induced oxidative stress in HaCaT cells. The results demonstrated that KC dose-dependently suppressed the production of inflammatory mediators, including NO, PGE2, IL-6, IL1β, MCP-1, and IFN-β in LPS-stimulated RAW264.7 macrophages. The study demonstrated that the inhibition of STAT1, STAT6, and NF-κB activations by KC might have been responsible for the inhibition of NO, PGE2, IL-6, IL1β, MCP-1, and IFN-β in the LPS-stimulated RAW264.7 macrophages. KC also upregulated the expression of HO-1 and its activities in the LPS-stimulated RAW264.7 macrophages. The upregulation of Nrf2 transcription activities by KC in the LPS-stimulated RAW264.7 macrophages was demonstrated to be responsible for the upregulation of HO-1 expression and its activity in LPS-stimulated RAW264.7 macrophages. The impact rising from the radius of reentrant cavities, as well as the Reynolds number on the heat transfer and the pressure drop, is also analyzed. The results indicate that, compared with straight microchannels, microchannels with reentrant cavities could enhance the heat transfer and, more importantly, reduce the pressure drop at the same time. For the ranges of parameters studied, increasing the radius of reentrant cavities could augment the effect of pressure-drop reduction, while the corresponding variation of heat transfer is complicated. It is considered that adding reentrant cavities in microchannel heat exchangers is an ideal approach to improve performance.

INTRODUCTION Cardiovascular disease (CVD) is the leading cause of death worldwide, and genetic investigations into the human lipidome may provide insight into CVD risk. The aim of this study was to estimate the heritability of circulating lipid species and their genetic correlation with CVD traits. Methods Targeted lipidomic profiling was performed on 4492 participants from the Busselton Family Heart Study to quantify the major fatty acids of 596 lipid species from 33 classes. We estimated narrow-sense heritabilities of lipid species/classes, and their genetic correlations with eight CVD traits - body mass index, high-density lipoprotein cholesterol (HDL-C), low-density lipoprotein cholesterol, triglycerides, total cholesterol, waist-hip-ratio, systolic blood pressure, and diastolic blood pressure. Results We report heritabilities and genetic correlations of new lipid species/sub-classes, including acylcarnitine, ubiquinone, sulfatide, and oxidized cholesteryl esters. The aim of this international survey was to assess the global variability in prevention, diagnostic and treatment strategies for AE-IPF. MATERIAL AND METHODS Pulmonologists with ILD expertise were invited to participate in a survey designed by an international expert panel. RESULTS 509 pulmonologists from 66 countries responded. Significant geographical variability in approaches to manage AE-IPF was found. Common preventive measures included antifibrotic drugs and vaccination. Diagnostic differences were most pronounced regarding use of KL-6 and viral testing, while HRCT, BNP and D-Dimer are generally applied. High dose steroids are widely administered (94%); the use of other immunosuppressant and treatment strategies is highly variable. Very few (4%) responders never use immunosuppression. Antifibrotic treatments are initiated during AE-IPF by 67%. Invasive ventilation or extracorporeal membrane oxygenation are mainly used as a bridge to transplantation. Most physicians educate patients comprehensively on the severity of AE-IPF (82%) and consider palliative care (64%). CONCLUSION Approaches to the prevention, diagnosis and treatment of AE-IPF vary worldwide. Global trials and guidelines to improve the prognosis of AE-IPF are needed. EVLP has also been explored as a platform for donor lung repair through injury specific treatments such as antibiotics or fibrinolytics. We hypothesised that actively expressed pathways shared between transplantation and EVLP may reveal common mechanisms of injury and potential therapeutic targets for lung repair prior to transplantation. MATERIALS AND METHODS A retrospective transcriptomics analyses were performed with peripheral tissue biopsies from "donation after brain death" lungs, with 46 pre/post-transplant pairs and 49 pre/post-EVLP pairs. Pathway analysis was used to identify and compare the responses of donor lungs to transplantation and to EVLP. RESULTS Twenty-one pathways were enriched predominantly in transplantation, including upregulation of lymphocyte activation and cell death, and downregulation of metabolism and protein synthesis.

We hypothesised that actively expressed pathways shared between transplantation and EVLP may reveal common mechanisms of injury and potential therapeutic targets for lung repair prior to transplantation. MATERIALS AND METHODS A retrospective transcriptomics analyses were performed with peripheral tissue biopsies from "donation after brain death" lungs, with 46 pre/post-transplant pairs and 49 pre/post-EVLP pairs. Pathway analysis was used to identify and compare the responses of donor lungs to transplantation and to EVLP.
