The results show that the δ-Al2O3 phase contains three distinct AlO sites and three distinct AlT sites. This detailed Al site structural information offers a powerful way of analyzing the most complex γ-Al2O3 spectrum. It is found that the γ-Al2O3 phase consists of Al sites with local structures similar to those found in the δ-Al2O3 and θ-Al2O3 phases albeit with less ordering. Spin-lattice relaxation time measurement further confirms the disordering of the lattice. Collectively, this study uniquely assigns 27Al features in transition aluminas, offering a simplified method to quantify complex mixtures of aluminum sites in transition alumina samples.Carbonic anhydrases (CAs) catalyze the physiological hydration of carbon dioxide and are among the most intensely studied pharmaceutical target enzymes. A hallmark of CA inhibition is the complexation of the catalytic zinc cation in the active site. Human (h) CA isoforms belonging to different families are implicated in a wide range of diseases and of very high interest for therapeutic intervention. Given the conserved catalytic mechanisms and high similarity of many hCA isoforms, a major challenge for CA-based therapy is achieving inhibitor selectivity for hCA isoforms that are associated with specific pathologies over other widely distributed isoforms such as hCA I or hCA II that are of critical relevance for the integrity of many physiological processes. To address this challenge, we have attempted to predict compounds that are selective for isoform hCA IX, which is a tumor-associated protein and implicated in metastasis, over hCA II on the basis of a carefully curated data set of selective and nonselective inhibitors. Machine learning achieved surprisingly high accuracy in predicting hCA IX-selective inhibitors. The results were further investigated, and compound features determining successful predictions were identified. These features were then studied on the basis of X-ray structures of hCA isoform-inhibitor complexes and found to include substructures that explain compound selectivity. Our findings lend credence to selectivity predictions and indicate that the machine learning models derived herein have considerable potential to aid in the identification of new hCA IX-selective compounds.High-pressure carbon monoxide (HiPCO) single-walled carbon nanotubes (SWCNTs) were heat treated at high temperatures from 1700 to 3000 °C. During the heating below 2500 °C, the diameters of the SWCNTs gradually increase from ∼1.0 to >1.5 nm, and at the temperatures higher than 2500 °C, double-, triple-, multiwalled carbon nanotubes (MWCNTs) appear as a consequence of the coalescence of SWCNT bundles. It is surprising that most MWCNTs have odd number of walls, such as 3 or 5. The even-odd number effect agrees well with the mechanism of SWCNT bundle coalescence proposed by López M. J. [Phys. Rev. Lett.2002, 89, 255501], in which an SWCNT that templated the layer by layer coalescence of surrounding SWCNTs is responsible for the enrichment of MWCNTs with odd number of walls. This study confirms the mechanism of SWCNT bundle coalescence, discovers an interesting odd-even number of walls effect in the consequent MWCNTs, and suggests that it is possible to obtain structure-controllable MWCNTs via SWCNT bundle coalescence.A ≥ 200 °C fraction (CT200F) of low-temperature coal tar was prepared by a rotary film evaporator. The catalytic conversion experiments of CT200F and six model compounds were conducted on the pyrolysis gas chromatography-mass spectrometer. The yields of catalytic conversion products benzene, toluene, xylene, and naphthalene (BTXN) were analyzed by semi-quantitative analysis according to the chromatographic peak areas. Additionally, the possible formation pathways and mechanisms of the target products BTXN generated over different catalysts were investigated. The results show that the yield of aromatic hydrocarbons increases and the yield of acid compounds decreases during CT200F pyrolysis over ZSM-5, HY, USY, and β-zeolite compared with that of its non-catalytic pyrolysis, especially the yields of BTXN obtained over USY and β-zeolite increase by 128 and 108%, respectively. The pore structure of ZSM-5 is suitable to produce BTX, while the suitable acidity and pore structure of USY, HY, and β-zeolite are more bcan be effectively utilized. In this report, two chromotrope dyes, chromotropic acid (CA) and chromotrope 2R (CR), were explored as inhibitors against mild steel corrosion in 1.0 M sulfuric acid solutions at 303 K. Electrochemical, spectroscopic, chemical, and microscopic techniques, namely, potentiodynamic polarization (PDP), electrochemical impedance spectroscopy, mass loss, and scanning electron microscopy (SEM), have been employed to evaluate the inhibition efficiencies (%IEs) of the examined organic dyes. The %IEs were found to increase with the inhibitors' concentrations, while they decreased with rising temperature. The outcomes of the PDP technique displayed that the examined inhibitors operated as mixed-type inhibitors with anodic prevalence. The impedance spectra described by Nyquist and Bode graphs in the corrosive environment and in the presence of various concentrations of the examined inhibitors showed single depressed capacitive loops and one-time constants. This behavior signified that the mild steel corrosion was managed by the charge transfer process. The SEM micrographs of the surfaces of mild steel samples after adding the examined inhibitors revealed a wide coverage of these compounds on the steel surfaces. Thus, the acquired high %IEs of the examined inhibitors were interpreted by strong adsorption of the organic molecules on the mild steel surface. This constructed a shielding layer separating the alloy surface from the corrosive medium, and such adsorption was found to follow the Langmuir isotherm. Furthermore, the evaluated thermodynamic and kinetic parameters supported that the nature of such adsorption was mainly physical. Results obtained from all employed techniques were consistent with each other and revealed that the %IE of the CR inhibitor was slightly higher than that of CA under similar circumstances. https://www.selleckchem.com/products/Puromycin-2HCl.html Finally, the mechanisms of both corrosion of mild steel in sulfuric acid solutions and its inhibition by the tested organic dyes were also discussed.

Most ovarian cancers are infiltrated by prognostically relevant activated T cells1-3, yet exhibit low response rates to immune checkpoint inhibitors4. Memory B cell and plasma cell infiltrates have previously been associated with better outcomes in ovarian cancer5,6, but the nature and functional relevance of these responses are controversial. Here, using 3 independent cohorts that in total comprise 534 patients with high-grade serous ovarian cancer, we show that robust, protective humoral responses are dominated by the production of polyclonal IgA, which binds to polymeric IgA receptors that are universally expressed on ovarian cancer cells. Notably, tumour B-cell-derived IgA redirects myeloid cells against extracellular oncogenic drivers, which causes tumour cell death. In addition, IgA transcytosis through malignant epithelial cells elicits transcriptional changes that antagonize the RAS pathway and sensitize tumour cells to cytolytic killing by T cells, which also contributes to hindering malignant progression. Thus, tumour-antigen-specific and -antigen-independent IgA responses antagonize the growth of ovarian cancer by governing coordinated tumour cell, T cell and B cell responses. These findings provide a platform for identifying targets that are spontaneously recognized by intratumoural B-cell-derived antibodies, and suggest that immunotherapies that augment B cell responses may be more effective than approaches that focus on T cells, particularly for malignancies that are resistant to checkpoint inhibitors.Cognitive and behavioural flexibility permit the appropriate adjustment of thoughts and behaviours in response to changing environmental demands. Brain mechanisms enabling flexibility have been examined using non-invasive neuroimaging and behavioural approaches in humans alongside pharmacological and lesion studies in animals. https://www.selleckchem.com/products/MLN8237.html This work has identified large-scale functional brain networks encompassing lateral and orbital frontoparietal, midcingulo-insular and frontostriatal regions that support flexibility across the lifespan. Flexibility can be compromised in early-life neurodevelopmental disorders, clinical conditions that emerge during adolescence and late-life dementias. We critically evaluate evidence for the enhancement of flexibility through cognitive training, physical activity and bilingual experience.Altered functioning of GABAergic interneurons expressing parvalbumin (PV) in the basal ganglia-thalamo-cortical circuit are likely to be involved in several human psychiatric disorders characterized by deficits in attention and sensory gating with dysfunctional decision-making behavior. However, the contribution of these interneurons in the ability to acquire demanding learning tasks remains unclear. Here, we combine an operant conditioning task with local field potentials simultaneously recorded in several nuclei involved in reward circuits of wild-type (WT) and PV-deficient (PVKO) ****, which are characterized by changes in firing activity of PV-expressing interneurons. In comparison with WT ****, PVKO animals presented significant deficits in the acquisition of the selected learning task. Recordings from prefrontal cortex, nucleus accumbens (NAc) and hippocampus showed significant decreases of the spectral power in beta and gamma bands in PVKO compared with WT **** particularly during the performance of the operant conditioning task. From the first to the last session, at all frequency bands the spectral power in NAc tended to increase in WT and to decrease in PVKO. Results indicate that PV deficiency impairs signaling necessary for instrumental learning and the recognition of natural rewards. To date, little is known about the ecological significance of Comammox (COMplete AMMonia OXidizers) Nitrospira in the water column of freshwater lakes. Water samples collected along depth profiles were used to investigate the distribution of Comammox in 13 lakes characterized by a wide range of physicochemical properties. Several published primers, which target the α-subunit of the ammonia monooxygenase, generated non-specific PCR products or did not amplify target genes from lake water and other habitats. Therefore, a new primer set has been designed for specific detection of Comammox in lakes. The high specificity of the PCR assay was confirmed by sequencing analysis. Quantification of Comammox amoA genes in lake water samples based on droplet digital PCR (ddPCR) revealed very low abundances (not exceeding 85 amoA copies ml-1), which suggest that Comammox is of minor importance for the nitrification process in the water column of the study sites. Surprisingly, samples taken from the sediment/water-interface along an oxygen gradient in dimictic Piburger See showed Comammox abundances three to four magnitudes higher than in the pelagic realm of the lake, which indicates a preference of Comammox to a particle-attached lifestyle.Develop a rat model of hilar cholangiocarcinoma for detecting bile salt export pump (Bsep) expression in hilar cholangiocarcinoma tissues, in order to provide a new therapeutic target for the gene therapy of hilar cholangiocarcinoma. Sixty male Wistar rats (body weight, 190 ± 8 g) were randomly divided into three groups (the experimental group, the control group and the sham operation group, n = 20 each) as follows The three groups were fed a standard diet, the experimental group was injected by cholangiocarcinoma QBC939 cell suspension along the hilar bile duct into the bile duct bifurcation with microsyringe, the control group was injected by normal saline, the sham operation group did not inject anything. Every day assess the rats' mental state, diet, and motion by using Basso-Beattie-Bresnahan and combined behavioral score. At 4 weeks, one rat of the experimental group was sacrificed after it was administered anesthesia, and we recorded changes in hilar bile duct size, texture, and form. This procedure waerged from the bile ducts of rats in the experimental group. The Bsep/Gapdh mRNA ratio in hilar cholangiocarcinoma, control group and sham operation group differed markedly. Light microscopy revealed a granular pattern of Bsep protein expression which reacted with the anti-Bsep antibody. Each section was randomly divided into six regions, with 80 cells were observed in every region. Sections with > 10% positive cells were designated positive, Sections with less then 10% positive cells were designated negative. Each group included 4800 cells. In the experimental group, 1200 cells (25%) were positive, in the control group, 3648 cells (76%) were positive and in the sham operation group 3598 cells (75%) were positive, and this difference was statistically significant. Bsep expression significantly decreased in hilar cholangiocarcinoma of rats than those in control group and sham operation group, suggesting that drugs targeting Bsep are a new strategy for hilar cholangiocarcinoma.

This paper describes the life and work of Louis Ombrédanne, one of the founding fathers of the SICOT, a notorious French orthopaedic and paediatric surgeon with a strong interest in plastic surgery and reconstruction of post-traumatic defects and also with limb lengthening and treatment of sequalae. Born in Paris in 1871 as a son of a general practitioner doctor, Ombrédanne was a brilliant student enjoying anatomy and surgery since the early years. He was appointed as a Surgeon in 1902 in Paris and became Professor of Surgery in 1907. During the First World War, he worked in Verdun, one of the most exposed hospitals near the battlefield where over 700.000 people died and many cases were affected by head and neck burns and destructions, requiring surgical reconstruction. After the war, Ombrédanne developed Pediatric Surgery and it seems that his powerful drive and charisma explain why these specialties are studied together with orthopaedics in Europe and later in the World. Many innovations like the anaesthetic inhaler with face mask were authored by Ombrédanne and also techniques of limb lengthening, thoracic plasty. He described malignant hyperthermia in children and worked in the Sick Children Hospital in Paris, today Hôpital Necker. In 1929, Ombrédanne participated with a group of enthusiastic surgeons and teachers from many countries to the creation of SICOT during a historical meeting held in Hotel Crillon in Paris. This paper introduces Ombrédanne's work to the current readers and is a tribute to the work of our ancestors who made possible the development of our specialty. In vitro generation of hematopoietic cells and especially hematopoietic stem cells (HSCs) from human pluripotent stem cells (PSCs) are subject to intensive research in recent decades, as these cells hold great potential for regenerative medicine and autologous cell replacement therapies. Despite many attempts, in vitro, de novo generation of bona fide HSCs remains challenging, and we are still far away from their clinical use, due to insufficient functionality and quantity of the produced HSCs. The challenges of generating PSC-derived HSCs are already apparent in early stages of hemato-endothelial specification with the limitation of recapitulating complex, dynamic processes of embryonic hematopoietic ontogeny in vitro. Further, these current shortcomings imply the incompleteness of our understanding of human ontogenetic processes from embryonic mesoderm over an intermediate, specialized hemogenic endothelium (HE) to their immediate progeny, the HSCs. In this review, we examine the recent investigations of hemato-endothelial ontogeny and recently reported progress for the conversion of PSCs and other promising somatic cell types towards HSCs with the focus on the crucial and inevitable role of the HE to achieve the long-standing goal-to generate therapeutically applicable PSC-derived HSCs in vitro. The McMurdo Dry Valleys are the largest single ice-free area in Antarctica, and of considerable scientific and conservation value as an extreme polar desert. This is recognised through the McMurdo Dry Valleys Antarctic Specially Managed Area (ASMA), where management's goals focus on protection of its unique features, while facilitating science access. Using a mix of remote sensing and existing cartography, we have identified over 6000 lakes and ponds in the ASMA. This study develops a classification of those aquatic ecosystems to provide a framework for management. It uses a limited top-down, hierarchical classification to define 13 class separations based on physical attributes that could largely be ascribed from existing databases or remotely sensed information. The first hierarchical level was based on landscape position, separating coastal kettle holes (reflecting recent glacial history), from other "topographic" water bodies. https://www.selleckchem.com/products/s-gsk1349572.html The second level was based on endorheic vs exorheic drainage, the third on mid-summer ice condition (no-ice cap; ice capped; frozen to base) and the fourth on source of inflow (glacial or non-glacial). Kettles were sub-classed by mid-summer ice only. Classes were tested against a set of field observations and an expert workshop validation process considered management implications for the ASMA. This study shows how the classification assists our understanding of Dry Valley landscapes and addresses management issues faced by researchers, environmental managers and policy makers. The approach to classification, rather than the detailed classes that may be specific to the Dry Valleys, has potential for wider use in other polar landscapes.
The reported mortality for sepsis and septic shock varies between 15% and 59% in international comparison. For Germany, the number of studies is limited. Previous estimations of mortality in Germany are outdated or based on claims data analyses. Various authors discuss whether lacking quality initiatives and treatment standards in Germany could cause higher mortality for sepsis. This contrasts with the internationally well-recognized performance of the German intensive care infrastructure during the COVID-19 pandemic.
The objectives of this systematic review and meta-analysis were to estimate 30-day and 90-day mortality of patients with sepsis and patients with septic shock in Germany and to compare the mortality with that of other industrialized regions (Europe, North America).
Asystematic literature search included interventional and observational studies published between 2009 and 2020 in PubMed and the Cochrane Library that analyzed adult patients with sepsis, severe sepsis and septic shock in Europany is higher in international comparison. A higher mortality would not be obvious either, since intensive care, for example also during the COVID-19 pandemic, is regarded as exemplary in Germany and the structural quality, such as the number of intensive care beds per 100,000 inhabitants, is high in international comparison. Nevertheless, deficits could also exist outside intensive care medicine. A comparison of international individual studies should take greater account of the structure of healthcare systems, the severity of disease and the limitations resulting from the data sources used.

A comparison of international individual studies should take greater account of the structure of healthcare systems, the severity of disease and the limitations resulting from the data sources used.0 Reacties 0 aandelen 47 Views 0 voorbeeld -
Screening for prostate cancer using prostate-specific antigen (PSA) testing can lead to problems of underdiagnosis and overdiagnosis. Short, noncontrast magnetic resonance imaging (MRI) or transrectal ultrasonography might overcome these limitations.
To compare the performance of PSA testing, MRI, and ultrasonography as screening tests for prostate cancer.
This prospective, population-based, blinded cohort study was conducted at 7 primary care practices and 2 imaging centers in the United Kingdom. Men 50 to 69 years of age were invited for prostate cancer screening from October 10, 2018, to May 15, 2019.
All participants underwent screening with a PSA test, MRI (T2 weighted and diffusion), and ultrasonography (B-mode and shear wave elastography). The tests were independently interpreted without knowledge of other results. Both imaging tests were reported on a validated 5-point scale of suspicion. If any test result was positive, a systematic 12-core biopsy was performed. Additional image fusion-targetncer, without an increase in the number of men advised to undergo biopsy or overdiagnosed with clinically insignificant cancer. There was no evidence that ultrasonography would have better performance compared with PSA testing alone.
In this cohort study, when screening the general population for prostate cancer, MRI using a score of 4 or 5 to define a positive test result compared with PSA alone at 3 ng/mL or higher was associated with more men diagnosed with clinically significant cancer, without an increase in the number of men advised to undergo biopsy or overdiagnosed with clinically insignificant cancer. There was no evidence that ultrasonography would have better performance compared with PSA testing alone.Trichoderma harzianum is a widely used biocontrol agent in agriculture. Obtaining a full inventory of the small molecules that can be biosynthesized from the encoded biosynthetic gene clusters (****) is therefore useful for understanding associated plant-microbe and microbe-microbe interactions. Here we heterologously reconstituted a polyketide synthase-nonribosomal peptide synthetase (PKS-NRPS) encoding gene cluster from T. harzianum t-22 in Aspergillus nidulans A1145. Six new tetronate natural products trihazone A-F (1-6) were isolated and elucidated by HRESIMS and 1D and 2D NMR data. Three of the products contain an exocyclic olefin, which is derived from the oxidative decarboxylation of an α-ketoglutarate-dependent dioxygenase ThnC as shown by biochemical assays. This review covers the literature published in 2019 for marine natural products (MNPs), with 719 citations (701 for the period January to December 2019) referring to compounds isolated from marine microorganisms and phytoplankton, green, brown and red algae, sponges, cnidarians, bryozoans, molluscs, tunicates, echinoderms, mangroves and other intertidal plants and microorganisms. The emphasis is on new compounds (1490 in 440 papers for 2019), together with the relevant biological activities, source organisms and country of origin. Pertinent reviews, biosynthetic studies, first syntheses, and syntheses that led to the revision of structures or stereochemistries, have been included. https://www.selleckchem.com/products/cc-930.html Methods used to study marine fungi and their chemical diversity have also been discussed. The past decade has witnessed growing interest in developing soft wearable pressure sensors with the ultimate goal of transforming today's hospital-centered diagnosis to tomorrow's patient-centered bio-diagnosis. In this context, battery-free wireless antenna-based pressure sensors will be highly advantageous for ubiquitous real-time health monitoring. However, current wireless antennas are largely based on thin films from traditional bulk metallic films or novel nanomaterials with an air-cavity design, which can only be operated in a limited pressure range due to the rigidity of active films and/or inherent cavity dimensions. Herein we report a soft battery-free wireless pressure sensor that is based on a three-dimensional (3D) porous gold nanowire foam-elastomer composite and is fabricated by solution-based conformal electroless plating technology, followed by elastomer encapsulation. We observe a transducer trade-off point for our foam antenna, below which the inductive effect and capacitive effect function together and above which the capacitive effect dominates. When an external pressure is applied, initially the inductance and capacitance increase simultaneously but the capacitance decreases afterwards. This can be transformed into a variable resonant frequency that first decreases linearly and then increases (in the capacitance domination pressure range). Importantly, the linear detection range of the sensor can be tuned simply by adjusting the thickness of the sponge or the rigidity of the elastomer (PDMS). We can achieve a wide pressure range of 0-248 kPa, which is the largest linear detection range reported in the literature (typically from 0 to 30 kPa) to the best of our knowledge. As a proof of concept, we further demonstrated that our gold nanowire foam sensor can be used to weigh people under both static and dynamic conditions. The tetrahydrocarbazole (THC) motif is ubiquitous in natural products and biologically active compounds. THCs can serve as favorable synthetic intermediates or precursors en-route to desired complex natural products. Despite a considerable number of strategies for the synthesis of THCs that have emerged in the last two decades, only a handful of reviews have been published on the subject. Herein, we have summarized synthetic methodologies published in the last ten years for the benefit of organic chemists. The review focuses on non-enantioselective syntheses of THCs, and encompasses ring opening reactions of donor-acceptor cyclopropanes, metal-catalyzed C-C/C-N bond formation, cycloaddition, conjugate addition, and miscellaneous reactions employed for accessing the THC framework. We present a time-resolved study of the photodissociation dynamics of OCS after UV-photoexcitation at λ = 237 nm. OCS molecules (X1Σ+) were primarily excited to the 11A'' and the 21A' Renner-Teller components of the 1Σ- and 1Δ states. Dissociation into CO and S fragments was observed through time-delayed strong-field ionisation and imaging of the kinetic energy of the resulting CO+ and S+ fragments by intense 790 nm laser pulses. Surprisingly, fast oscillations with a period of ∼100 fs were observed in the S+ channel of the UV dissociation. Based on wavepacket-dynamics simulations coupled with a simple electrostatic-interaction model, these oscillations do not correspond to the known highly-excited rotational motion of the leaving CO(X1Σ+, J ≫ 0) fragments, which has a timescale of ∼140 fs. Instead, we suggest to assign the observed oscillations to the excitation of vibrational wavepackets in the 23A'' or 21A'' states of the molecule that predissociate to form S(3PJ) photoproducts.

The amounts of the major chorion proteins Rp30, Rp45, Rp100, and Rp200 were decreased in the ATG3-depleted chorions, as well as the readings for dityrosine cross-linking and sulfur, detected by fluorescence emission under ultraviolet excitation and X-ray elemental detection and mapping. Altogether, we found that ATG3 is important for the proper chorion biogenesis and, therefore, crucial for this vector reproduction.Inhibitors of apoptosis proteins (IAPs) are conserved regulators involved in cell cycle, cell migration, cell death, immunity and inflammation, should be due to the fact that they can assist with the ability to cope with different kinds of extrinsic or intrinsic stresses. Bivalve molluscs are well adapted to highly complex marine environments. As free-living filter feeders that may take toxic dinoflagellates as food, bivalves can accumulate and put up with significant levels of paralytic shellfish toxins (PSTs). PSTs absorption and accumulation could have a deleterious effect on bivalves, causing negative impact on their feeding and digestion capabilities. In the present study, we analyzed IAP genes (PyIAPs) in Yesso scallop (Patinopecten yessoensis), a major fishery and aquaculture species in China. Forty-seven PyIAPs from five sub-families were identified, and almost half of the PyIAP genes were localized in clusters on two chromosomes. Several sites under positive selection was revealed in the significantly expanded sub-families BIRC4 and BIRC5. After exposure to PST-producing dinoflagellates, Alexandrium catenella, fourteen PyIAPs showed significant responses in hepatopancreas and kidney, and more than eighty-five percent of them were from the expanded sub-families BIRC4 and BIRC5. The regulation pattern of PyIAPs was similar between the two tissues, with more than half exhibited expression suppression within three days after exposure. In contrast to hepatopancreas, more acute changes of PyIAPs expression could be detected in kidney, suggesting the possible involvement of these PyIAPs in tissue-specific PST tolerance. https://www.selleckchem.com/products/Cediranib.html These findings also imply the adaptive expansion of bivalve IAP genes in response to algae derived biotoxins.Urothelial cells have been implicated in bladder mechanosensory transduction, and thus, initiation of the micturition reflex. Cell deformation caused by tension forces at an air-liquid interface (ALI) can induce an increase in intracellular Ca2+ concentration ([Ca2+]i) and ATP release in some epithelial cells. In this study, we aimed to examine the cellular mechanisms underlying ALI-induced [Ca2+]i increase in cultured urothelial cells. The ALI was created by stopping the influx of the perfusion but maintaining efflux. The [Ca2+]i increase was measured using the Ca2+ imaging method. The ALI evoked a reversible [Ca2+]i increase and ATP release in urothelial cells, which was almost abolished by GdCl3. The specific antagonist of the transient receptor potential vanilloid (TRPV4) channel (HC0674) and the antagonist of the pannexin 1 channel (10panx) both diminished the [Ca2+]i increase. The blocker of Ca2+-ATPase pumps on the endoplasmic reticulum (thapsigargin), the IP3 receptor antagonist (Xest-C), and the ryanodine receptor antagonist (ryanodine) all attenuated the [Ca2+]i increase. Degrading extracellular ATP with apyrase or blocking ATP receptors (P2X or P2Y) with pyridoxalphosphate-6-azophenyl-2',4'-disulfonic acid (PPADS) significantly attenuated the [Ca2+]i increase. Our results suggest that both Ca2+ influx via TRPV4 or pannexin 1 and Ca2+ release from intracellular Ca2+ stores via IP3 or ryanodine receptors contribute to the mechanical responses of urothelial cells. The release of ATP further enhances the [Ca2+]i increase by activating P2X and P2Y receptors via autocrine or paracrine mechanisms. We compared the impact of two different, but commonly consumed, beverages on integrative markers of exercise recovery following a 2 h high intensity interval exercise (i.e., running 70-80% V̇O2max intervals and interspersed with plyometric jumps). Participants (n = 11 males, n = 6 females) consumed a chocolate flavored dairy milk beverage (CM 1.2 g carbohydrate/kg BM and 0.4 g protein/kg BM) or a carbohydrate-electrolyte beverage (CEB isovolumetric with 0.76 g carbohydrate/kg BM) after exercise, in a randomized-crossover design. The recovery beverages were provided in three equal boluses over a 30 min period commencing 1 h post-exercise. Muscle biopsies were performed at 0 h and 2 h in recovery. Venous blood samples, nude BM and total body water were collected before and at 0, 2, and 4 h recovery. Gastrointestinal symptoms and breath hydrogen (H2) were collected before exercise and every 30 min during recovery. The following morning, participants returned for performance assessment. In recovery, breath H2 reato target functionality and patency of the gastrointestinal tract as a prerequisite to assimilation of recovery nutrition, as well as restoration of immunocompetency.Unstable hemoglobinopathies (UHs) are rare anemia disorders (RADs) characterized by abnormal hemoglobin (Hb) variants with decreased stability. UHs are therefore easily precipitating, causing hemolysis and, in some cases, leading to dominant beta-thalassemia (dBTHAL). The clinical picture of UHs is highly heterogeneous, inheritance pattern is dominant, instead of recessive as in more prevalent major Hb syndromes, and may occur de novo. Most cases of UHs are not detected by conventional testing, therefore diagnosis requires a high index of suspicion of the treating physician. Here, we highlight the importance of next generation sequencing (NGS) methodologies for the diagnosis of patients with dBTHAL and other less severe UH variants. We present five unrelated clinical cases referred with chronic hemolytic anemia, three of them with severe blood transfusion dependent anemia. Targeted NGS analysis was performed in three cases while whole exome sequencing (WES) analysis was performed in two cases. Five different UH variants were identified correlating with patients' clinical manifestations. Four variants were related to the beta-globin gene (Hb Bristol-Alesha, Hb Debrousse, Hb Zunyi, and the novel Hb Mokum) meanwhile one case was caused by a mutation in the alpha-globin gene leading to Hb Evans. Inclusion of alpha and beta-globin genes in routine NGS approaches for RADs has to be considered to improve diagnosis' efficiency of RAD due to UHs. Reducing misdiagnoses and underdiagnoses of UH variants, especially of the severe forms leading to dBTHAL would also facilitate the early start of intensive or curative treatments for these patients.

SARS-CoV-2 is an international public health emergency; high transmissibility and morbidity and mortality can result in the virus overwhelming health systems. Combinations of social distancing, and test, trace, and isolate strategies can reduce the number of new infections per infected individual below 1, thus driving declines in case numbers, but may be both challenging and costly. These interventions must also be maintained until development and (now likely) mass deployment of a vaccine (or therapeutics), since otherwise, many susceptible individuals are still at risk of infection. We use a simple analytical model to explore how low levels of infection, combined with vaccination, determine the trajectory to community immunity. Understanding the repercussions of the biological characteristics of the viral life cycle in this scenario is of considerable importance. We provide a simple description of this process by modelling the scenario where the effective reproduction number [Formula see text] is maintained at 1. Since the additional complexity imposed by the strength and duration of transmission-blocking immunity is not yet clear, we use our framework to probe the impact of these uncertainties. Through intuitive analytical relations, we explore how the necessary magnitude of vaccination rates and mitigation efforts depends crucially on the durations of natural and vaccinal immunity. We also show that our framework can encompass seasonality or preexisting immunity due to epidemic dynamics prior to strong mitigation measures. Taken together, our simple conceptual model illustrates the importance of individual and vaccinal immunity for community immunity, and that the quantification of individuals immunized against SARS-CoV-2 is paramount. This study aims to estimate the material properties of the porcine vitreous while testing it in close to its natural physiological conditions. Eighteen porcine eyes were tested within 48 h post-mortem. A custom-built computer-controlled test rig was designed to support, load and monitor the behaviour of eye globes while being subjected to dynamic rotation cycles mimicking saccade eye movement. Specimens were glued to the base of a container, surrounded by gelatin, frozen and cut in half to expose the vitreous. After thawing, the container was subjected to concentric dynamic rotations of up to 5°, 10° or 15°, while taking 50 MP photos of the specimen every 2 ms. https://www.selleckchem.com/products/talabostat.html The images were analysed by a digital image correlation algorithm to trace the movement of marked points on the vitreous surface with different radii from the centre of the posterior chamber. The initial camera image was used in building a finite-element model of the test set-up, which was used in an inverse analysis exercise to estimate the material p4-4002. It provides a good contextual background on the development of the LTC system in China as well as the needs and preferences of LTC from family and older adults' perspectives. In addition, this issue covers the topic of evaluation of a recently developed long-term care nursing insurance and provides an example of family caregiving for persons with dementia within the Chinese context. The authors in this special issue also provided insights into the impact of the COVID-19 pandemic on older adults' life and LTC quality, and explored potential strategies to handle the challenges during and post-pandemic.
Functional food ingredients, such as dietary fiber, long-chain polyunsaturated fatty acids, and high-quality protein, have been shown to help control blood glucose concentration and lower high blood pressure (BP), as well as improving other cardiovascular disease risk factors. However, little research has assessed the impacts of consuming chia seeds, which are rich in these nutrients, on metabolic and physiological outcomes, and results are conflicting.
The study aimed to investigate the possible effects of chia seeds on fasting blood glucose, insulin, glycated hemoglobin, BP, lipid profile, body weight, and the inflammatory marker - high-sensitivity C-reactive protein - in people with type 2 diabetes (T2DM).
Adults with T2DM (
= 42) were randomly assigned equally to the chia seed group, which consumed 40 g/day chia seeds for 12 weeks, or a control group, which did not consume any supplement. Blood samples were collected at baseline and after a 12-week intervention period to assess the study outcomes, such as glycemic control, BP, cardiovascular risk parameters including lipid profile, inflammatory marker, and body weight.
Adjusted for gender and baseline values, the chia seed group had systolic BP (SBP) significantly reduced compared to control [
(1) = 2.867,
= 0.007,
= 0.174]. No differences were observed in any other parameter tested in the chia seed or control group.
People with T2DM and hypertension, maintaining usual dietary consumption, physical activity pattern, and medications, had significantly reduced SBP compared to the control group when having consumed 40 g/d of chia seeds for 12 weeks.

Existing quality of life questionnaires are either disease specific or generic in their assessment of themes which are perceived important to the quality of life in populations with disabilities. To be able to improve quality of life in a population with diverse disabilities there was a need for a cross-disability instrument. The Electronic Quality of Life (EQOL)-questionnaire was developed to meet this need. It is crucial that such an instrument is validated, easy to use, and interpret by, for example, clinicians and policy planners. This study aims to test the content validity of the EQOL questionnaire and to construct a user-friendly, cross-disability quality of life profile.
To further test the content validity of the EQOL-questionnaire, we conducted field test analyses on 318 individuals (aged 16-64) with self-reported disabilities. Comments on the questionnaire were scrutinised and sorted. A profile with six domains of quality of life was developed. Model fit was evaluated by confirmatory factor analysis and content validity was evaluated based on distributions.

Adults with T2DM ( = 42) were randomly assigned equally to the chia seed group, which consumed 40 g/day chia seeds for 12 weeks, or a control group, which did not consume any supplement. Blood samples were collected at baseline and after a 12-week intervention period to assess the study outcomes, such as glycemic control, BP, cardiovascular risk parameters including lipid profile, inflammatory marker, and body weight. Adjusted for gender and baseline values, the chia seed group had systolic BP (SBP) significantly reduced compared to control [ (1) = 2.867, = 0.007, = 0.174]. Model fit was evaluated by confirmatory factor analysis and content validity was evaluated based on distributions.0 Reacties 0 aandelen 33 Views 0 voorbeeld -
Among drugs in development and/or in market, there are poorly water-soluble and poorly lipid-soluble compounds. Rebamipide, classified into BCS class IV, is one of those drugs which provide very low bioavailability and/or the difficulty of formulation for oral administration. Because of its low solubility in available lipoidal excipients, it was impossible to prepare an adequate SNEDDS formulation of rebamipide. Then, we tried to increase the solubility of rebamipide in lipoidal excipients for preparing a more practical SNEDDS formulation by making the complex with its counter ion, tetrabutylphosphonium hydroxide (TBPOH) or NaOH. Rebamipide concentration in ethanol was proportionally increased with the increment of TBPOH or NaOH added, indicating that the formation of complex with a counter ion should contribute to the solubilization of rebamipide in ethanol. Both Rebamipide-TBPOH complex (Reb-TBPOH) and Rebamipide-NaOH complex (Reb-NaOH) obtained by lyophilization showed no endothermic peak in DSC and no dif complex successfully improved rebamipide absorption. The interaction between anticancer drugs and HSA may have a significant impact on the pharmacology and efficacy of drugs. Drugs change the binding properties of HSA by regulating the quenching mechanism, binding mode and binding affinity. In this study, the interactions of cisplatin (cDDP), HSA, and daphnoretin were elucidated by multi-spectroscopic analyses and docking simulation. Fluorescence quenching showed that cDDP could not change the static quenching mechanism of HSA-daphnoretin, but could enhance their binding affinity. Site competition experiments revealed that daphnoretin and cDDP both bound to site I, which was consistent with the results of molecular docking. Thermodynamic date indicated that cDDP and daphnoretin formed a more stable complex with HSA via hydrophobic, van der Waals interaction and hydrogen bond. Three-dimensional fluorescence and circular dichroism spectra showed that cDDP changed the conformation and micro-environment of HSA induced by daphnoretin. This work could provide valuable information for the binding properties and interaction among cDDP, daphnoretin and HSA, and put forward the possibility of using HSA as a multidrug carrier.
Ammoxetine is a novel selective serotonin and norepinephrine reuptake inhibitor. Preclinical studies have indicated the potential utility of ammoxetine for therapy in major depressive disorder.
To investigate the first-in-human safety, tolerability, and pharmacokinetics (PK) of ammoxetine in healthy subjects and evaluate the effect of CYP2C19 polymorphisms on metabolism of ammoxetine.
In this randomized, double-blind, placebo-controlled phase I study, healthy Chinese subjects were allocated to receive 2.5, 7.5, 15, 30, 45, 65, 100 mg ammoxetine or placebo in single-dose part and 15, 30, 45 mg ammoxetine or placebo twice daily for 8 days in multiple-dose part. Pharmacokinetic, safety and tolerability assessments were performed.
A total of 134 subjects were screened and 94 were enrolled. All the ammoxetine-related adverse events (AEs) were mild and resolved spontaneously. https://www.selleckchem.com/products/Puromycin-2HCl.html No hepatic AEs were reported during the study. Ammoxetine was well absorbed after oral administration with T
reached in 5.0-6.0 h. After single-dosing, C
and AUC increased proportionally with dose, except at 65 mg. After multiple-dosing, the exposures of ammoxetine at steady state increased slightly in a more-than-dose-proportional manner over the dose range studied, probably due to the saturated elimination. Steady state was achieved 6 days after multiple-dosing was initiated. The low extent of urinary excretion of ammoxetine (< 2%) indicated it is undergoing extensive metabolism. CYP2C19 polymorphisms had minimal effect on metabolism of ammoxetine.
Ammoxetine has a favorable pharmacokinetic profile after oral administration and good safety properties. The PK and safety profiles of ammoxetine could enable further clinical development in patients with major depressive disorder.

Once no topical formulation may be found in the market until now, SEC was incorporated in thermosensitive bioadhesive systems to extend the contact time in the mucosa and to avoid a systemic drug disposition. Formulations containing 20% poloxamer 407, 1% poloxamer 188 and 1 or 2.5% chitosan showed suitable sol-gel transition temperature (> 30 °C), presenting a fast gelation time (100-115 s). Rheological, dynamic light scattering and infrared spectroscopy analysis suggested molecular interactions among polymers. Chitosan increased the mucoadhesion strength of the formulations. In addition, hydrogels showed a tendency to decrease the drug transport rate through mucosa when compared to the control. Mucin was also added onto mucosa for a more realistic simulation of permeability/retention. In the presence of this agent, hydrogels containing chitosan reduced the permeability/retention of the drug in approximately 2.0-fold when compared to the control. Therefore, the hydrogels presented suitable characteristics to remain in the vaginal environment, which would result in effective local treatment of trichomoniasis.Stress-induced activation of locus coeruleus (LC)-norepinephrine (NE) projections to the prefrontal cortex are thought to promote cognitive responses to stressors. LC activation by stressors is modulated by endogenous opioids that restrain LC activation and facilitate a return to baseline activity upon stress termination. Sex differences in this opioid influence could be a basis for sex differences in stress vulnerability. Consistent with this, we recently demonstrated that μ-opioid receptor (MOR) expression is decreased in the female rat LC compared to the male LC, and this was associated with sexually distinct consequences of activating MOR in the LC on cognitive flexibility. Given that the LC-NE system affects cognitive flexibility through its projections to the medial prefrontal cortex (mPFC), the present study quantified and compared the effects of LC-MOR activation on mPFC neural activity in male and female rats. Local field potential (LFPs) were recorded from the mPFC of freely behaving male and female rats before and following local LC microinjection of the MOR agonist, DAMGO, or vehicle.

0 V vs. RHE with special emphasis on the formation of H2 gas and C1 products. Thus, the electrodes prepared at higher temperatures enable the suppression of competing H2 evolution due to the increased amount of high-index facets. Moreover, the formation rates of C1 products were inhibited as well at the electrodes with increased number of high-index facets. The drops in the formation rates of both H2 and C1 products indicate that they are consumed in the chemical reaction to commence the formation of multi-carbon products. However, further study is still required with superior attention on CO2RR towards the C2+ product formation at a range of applied potentials. The development of a simple and effective single constituent multifunctional nanotheranostic platform producing a multimodality diagnostic signal and curing effect is still a challenge. Herein, we synthesized simple and biodegradable FeWOx ternary oxide nanoparticles and modified their surface with RGD-PEG-NH2 (FeWOx-PEG-RGD NPs), whereby resulting NPs possessed a (T2/T1) switchable MRI/CT dual-modal imaging ability and synergistic photothermal therapy (PTT)/photodynamic therapy (PDT)/chemodynamic therapy (CDT) capacity. We showed that FeWOx-PEG-RGD NPs enabled tumor accumulation under a magnetic field drive and RGD-mediated tumor penetration and implemented PTT/PDT treatment under 980 nm laser irradiation. In an acidic tumor microenvironment (TME) with a high hydrogen peroxide (H2O2) expression, NPs degraded to release Fe3+ and Fe2+, triggering a Fenton reaction to generate ˙OH for CDT. The released Fe2+ led to T2/T1 signal conversion for tracing cancer therapy, while the high X-ray attenuation coefficient of W also made it a good CT contrast agent for guided therapy. Thus, the structurally simple FeWOx-PEG-RGD was capable of mediating (T2/T1-weighted) MR/CT two modal imaging-guided PTT/PDT/CDT synergic therapy with a high accuracy and superb anticancer efficiency. The simple, degradable, rapid-clearance, and multifunctional FeWOx-PEG-RGD NPs provide a novel, promising, and versatile nanotheranostic platform.Glycosaminoglycans (GAGs) are anionic, periodic, linear polysaccharides which are composed of periodic disaccharide units. They play a vital role in many biological processes ongoing in the extracellular matrix. In terms of computational approaches, GAGs are very challenging molecules due to their high flexibility, periodicity, predominantly electrostatic-driven nature of interactions with their protein counterparts and potential multipose binding. Furthermore, the molecular mechanisms underlying GAG-mediated interactions are not fully known yet, and experimental techniques alone are not always sufficient to gain insights into them. The aim of this study was to characterize protein-ion-GAG complexes for the systems where ions are directly involved in GAG binding. Molecular docking, molecular dynamics and free energy calculation approaches were applied to model and rigorously analyse the interactions between annexins (II and V), calcium ions (Ca2+) and heparin (HP). The computational data were examined and discussed in the context of the structural data previously reported by the crystallographic studies. The computational results confirm that the presence of Ca2+ has a tremendous impact on the annexin-HP binding site. https://www.selleckchem.com/products/s-gsk1349572.html This study provides a general computational pipeline to discover the complexity of protein-GAG interactions and helps to understand the role of ions involved at the atomic level. The limitations of the applied protocols are described and discussed pointing at the challenges persisting in the state-of-the-art in silico tools to study protein-ion-GAG systems.Correction for 'Quartz crystal microbalance for telomerase sensing based on gold nanoparticle induced signal amplification' by Haitang Yang et al., Chem. Commun., 2019, 55, 5994-5997, DOI .I have herein investigated the solvent-dependent photoluminescence quenching mechanism of [Ru(bpy)2(bpy-cc-AQ)]2+ using variable temperature emission spectroscopies. The photophysics of this complex are dominated by an excited-state thermal equilibrium between a photoluminescent 3MLCT state and a charge-separated state that lies higher in energy relative to the 3MLCT state in low polarity solvents and approximately isoenergetic in high polarity solvents. Furthermore, an unusual photoluminescence temperature-dependence in high polarity solvents is shown to arise from competition between enthalpic factors favouring the charge-separated state and entropic factors favouring the photoluminescent 3MLCT state, analogous to the molecular light-switch effect of [Ru(bpy)2(dppz)]2+. The solvent-dependent photoluminescence quenching of [Ru(bpy)2(bpy-cc-AQ)]2+ is attributed to two key solvent-dependent factors (1) the excited-state equilibrium position and (2) the rate of charge-recombination from the charge-separated state.Heterogeneous ice nucleation is a common process in the atmosphere, but relatively little is known about the role of different surface characteristics on the promotion of ice nucleation. We have used a series of iron oxides as a model system to study the role of lattice mismatch and defects induced by milling on ice nucleation activity. The iron oxides include wüstite (FeO), hematite (Fe2O3), magnetite (Fe3O4), and goethite (FeOOH). The iron oxides were characterized by X-ray diffraction (XRD) and Brunauer-Emmett-Teller (BET) surface area measurements. The immersion freezing experiments were performed using an environmental chamber. Wüstite (FeO) had the highest ice nucleation activity, which we attribute to its low lattice mismatch with hexagonal ice and the exposure of Fe-OH after milling. A comparison study of MnO and wüstite (FeO) with milled and sieved samples for each suggests that physical defects alone result in only a slight increase in ice nucleation activity. Despite differences in the molecular formula and surface groups, hematite (Fe2O3), magnetite (Fe3O4), and goethite (FeOOH) had similar ice nucleation activities, which may be attributed to their high lattice mismatch to hexagonal ice. This study provides further insight into the characteristics of a good heterogeneous ice nucleus and, more generally, helps to elucidate the interactions between aerosol particles and ice particles in clouds.

The FDA-approved drug ivermectin is applied for treatments of onchocerciasis and lymphatic filariasis. The anti-cancer and anti-viral activities have been demonstrated stressing possibilities for the drug repurposing and therefore new information on high dosage safety is on demand. https://www.selleckchem.com/CDK.html We analyzed in vivo tissue responses for high doses of ivermectin using Corydoras fish as animal model. We made intestinal histology and hematologic assays after oral administration of ivermectin transported with polyelectrolytes formulation. Histology showed any apparent damage of intestinal tissues at 0.22-170 mg of ivermectin/kg body weight. Immunofluorescence evidenced delocalization of Myosin-Vb at enterocytes only for the higher dose. Hematology parameters showed random variations after 7 days from administration, but a later apparent recover after 14 and 21 days. The study evaluated the potential of high doses of oral administration of ivermectin formulation, which could be an alternative with benefits in high compliance therapies.
L. have been utilized as traditional medicine in Indian subcontinent for the treatment of numerous illnesses such as inflammation, pain and skin infection. In this current study we sought to assess the anxiolytic, sedative and analgesic activity of
L. leaves extract.
We first performed a phytochemical screening test of the leaves extracts following standard phytochemical screening protocols. We next examined the anxiolytic and sedative activity of crude methanol (TIME), ethyl acetate (TIEAE) and n-Hexane (TIHE) extract of
L. leaves using mouse behavioral models such as elevated plus-maze test and pentobarbital-induced sleeping time test, respectively. Likewise, we evaluated the analgesic activity using acetic acid induced writhing test and formalin induced paw licking test. Additionally, we performed a quantitative analysis of heavy metals content of
L. leaves by overnight digestion in concentrated nitric acid (HNO
).
Phytochemical screening demonstrated that TIME, TIEAE and TIHE contain flavonoids, alkaloids, tannins, phenols, terpenoids and sterols. Administration of these extracts resulted in higher number of open arm entry, lower number of close arm entry and higher time spent in open arm compared to control treatment (p < 0.05). Moreover, these treatments decreased the onset of sleep time and increased the duration of sleep compared to control treated **** (all p < 0.05). Likewise, extracts treated **** exhibited decreased number of writhing as well as lower acute phase and late phase duration compared to control treatment (all p < 0.05). The average level of As and Fe in
L. leaves was 5.16 ± 0.012 ppm and 2.76 ± 0.015 ppm, respectively.
Results from this study support that
L. leaves extracts exhibit an anxiolytic, sedative and analgesic activity in ****.
Results from this study support that Tragia involucrata L. leaves extracts exhibit an anxiolytic, sedative and analgesic activity in ****.Various sealants have been developed to treat the exposure of dentinal tubules (DTs) and further dental hypersensitivity. Herein, the phosphate-terminated fourth generation polyamidoamine dendrimers (PAMAM-PO3H2) was successfully synthesized. Six duplicates of demineralized dentin and type I collagen matrix were incubated in artificial saliva solution with or without PAMAM-PO3H2 treatment at 37 °C for 2 weeks, respectively. The artificial saliva solution was replaced every day. These regenerated crystals on the surface of dentin and collagen matrix were confirmed to be hydroxyapatite (HAp). The approach thus demonstrates that PAMAM-PO3H2 can offer an efficient therapy for seal DTs in clinical dentistry.Nutritional aspects modulate the parasitological and immune response in infected sheep. The aim of the present study was to evaluate the parasitological and humoral immune response of Pelibuey sheep experimentally infected with Haemonchus contortus that were fed diets with two different energy (n = 12) and protein (n = 12) levels. Twenty-four Pelibuey lambs infected with H. contortus were evaluated over 12 weeks. An additional six animals were considered as a control group. Fecal egg count (FEC) was determined, in addition to packed cell volume (PCV), total plasma protein (TPP), and immunoglobulin levels (IgA, IgG, and IgM) by indirect enzyme-linked immunosorbent assay (iELISA). Data were analyzed by repeated measures over time. The lambs that received a high-energy (HighE) diet had the lowest FEC (P less then 0.01) regardless of whether they received a high (HighP) or low (LowP) level of protein. The effect of energy level was also observed over time FEC values decreased and PCV and TPP values increased. Higher immunoglobulin levels were obtained for females (P less then 0.05) than males yet, overall, the energy and protein levels of the diets did not affect the response of the immunoglobulins. Over time, however, an increase in IgG and IgM was observed, whereas the IgA level remained basal.
Fish is the important source of animal protein and regarded as the second food after rice in Bangladesh. Fish consumption is influenced by consumer socioeconomic characteristics.
The specific objectives of the current study are (i) to find information on consumers' fish consumption levels; (ii) to assess the relationships between consumer's preferences and their socioeconomic characteristics; and (iii) to investigate the factors affecting consumer fish consumption.
We surveyed a total of 128 randomly selected respondents from the Rangpur city corporation (RPCC), Bangladesh in 2019 using a semi-structural questionnaire.
The average consumption level of fish per quarterly was 1.45 kg/person. Rui (
), Pangas (Pangasius), Hilsha (
ilesha) and Tilapia (
) were the most frequently consumed fish species. Significant differences in consumption level were observed among the age categories, profession, gender, education, and income levels (p < 0.05). Most participants consumed fish more than once a week throughout the year and there was no seasonal impact on fish consumption. Fish consumption level was significantly and positively associated with education and income levels and negatively associated with age categories (p < 0.01). The stepwise multiple regression method elucidated 53.7% of the variance (p < 0.05) for fish consumption.
Fish consumption in the RPCC is lower than the average consumption level in Bangladesh particularly for lower income people. Lack of proper knowledge on fish consumption value and high price appeared as the important barrier to increase the fish consumption.

In this current study we sought to assess the anxiolytic, sedative and analgesic activity of L. leaves extract. We first performed a phytochemical screening test of the leaves extracts following standard phytochemical screening protocols. We next examined the anxiolytic and sedative activity of crude methanol (TIME), ethyl acetate (TIEAE) and n-Hexane (TIHE) extract of L. leaves using mouse behavioral models such as elevated plus-maze test and pentobarbital-induced sleeping time test, respectively. Likewise, we evaluated the analgesic activity using acetic acid induced writhing test and formalin induced paw licking test. Additionally, we performed a quantitative analysis of heavy metals content of L. leaves by overnight digestion in concentrated nitric acid (HNO ). Moreover, these treatments decreased the onset of sleep time and increased the duration of sleep compared to control treated mice (all p < 0.05). Likewise, extracts treated mice exhibited decreased number of writhing as well as lower acute phase and late phase duration compared to control treatment (all p < 0.05). The average level of As and Fe in L. leaves was 5.16 ± 0.012 ppm and 2.76 ± 0.015 ppm, respectively. Results from this study support that L. leaves extracts exhibit an anxiolytic, sedative and analgesic activity in mice. Results from this study support that Tragia involucrata L. leaves extracts exhibit an anxiolytic, sedative and analgesic activity in mice.Various sealants have been developed to treat the exposure of dentinal tubules (DTs) and further dental hypersensitivity. Rui ( ), Pangas (Pangasius), Hilsha ( ilesha) and Tilapia ( ) were the most frequently consumed fish species. Significant differences in consumption level were observed among the age categories, profession, gender, education, and income levels (p < 0.05). Most participants consumed fish more than once a week throughout the year and there was no seasonal impact on fish consumption.
