Variation in the PE charge density (or degree of ionization) and grafting density leads to massive changes of the properties of the PE molecules (quantified by the changes in the height and the mobility of the PE brushes) as well as the local arrangement and distribution of the brush-supported counterions and water molecules within the brushes. The effect on the counterions is manifested by the corresponding variation of the counterion mobility, counterion concentration, extent of counterion binding to the charged site of the PE brushes, water-in-salt-like structure formation, and counterion-water-oxygen radial distribution function within the PE brushes. On the other hand, the effect on water molecules is manifested by the corresponding variation of water-oxygen-water-oxygen RDF, local water density, water-water and water-PE functional group hydrogen bond networks, static dielectric constant of water molecules, orientational tetrahedral order parameter, and water mobility. Enforcing such varying degree of ionization of weak polyelectrolytes is possible by changing the pH of the surrounding medium. Thus, our results provide insights into the changes in microstructure (at the atomistic level) of weak polyionic brushes at varying pH. We anticipate that this knowledge will prove to be vital for the efficient design of several nano-scale systems employing PE brushes such as nanomechanical gates, current rectifiers, etc.Dental clinicians have relied for centuries on traditional dental materials (polymers, ceramics, metals, and composites) to restore oral health and function to patients. Clinical outcomes for many crucial dental therapies remain poor despite many decades of intense research on these materials. Recent attention has been paid to biomolecules as a chassis for engineered preventive, restorative, and regenerative approaches in dentistry. Indeed, biomolecules represent a uniquely versatile and precise tool to enable the design and development of bioinspired multifunctional dental materials to spur advancements in dentistry. In this review, we survey the range of biomolecules that have been used across dental biomaterials. Our particular focus is on the key biological activity imparted by each biomolecule toward prevention of dental and oral diseases as well as restoration of oral health. Additional emphasis is placed on the structure-function relationships between biomolecules and their biological activity, the unique challenges of each clinical condition, limitations of conventional therapies, and the advantages of each class of biomolecule for said challenge. Biomaterials for bone regeneration are not reviewed as numerous existing reviews on the topic have been recently published. We conclude our narrative review with an outlook on the future of biomolecules in dental biomaterials and potential avenues of innovation for biomaterial-based patient oral care.The cyclometalated Pt(ii) acetylide derivative with a 1,3-bis(N-octyl-benzimidazol-2-yl)benzene (N^C^N) ligand and a free terpyridine (TPY) receptor has been successfully synthesized and characterized. #link# X-ray crystallography shows its inefficient conjugation degree between the [(N^C^N)Pt] and TPY planes. This bifunctional complex shows an enhanced 1MLCT/LLCT absorption band (ε = 3.30 × 104 dm3 mol-1 cm-1) centered at λmax = 365 nm, and the well-resolved vibronic-structured 3MLCT/LLCT emission bands (Φ = 0.08, τ = 3.43 μs) in the range of ca. 475-700 nm. Consecutive titrations show that added Zn2+ and Eu(HFA)3 bond to its free TPY receptor with 1  2 and 1  1 stoichiometry to form the heterotrinuclear Pt-Zn-Pt (Ka = 3.48 × 104 mol-1 dm3) and heterodinuclear Pt-Eu (Ka = 1.73 × 104 mol-1 dm3) complexes, respectively. A sensitizing effect of Zn2+ on the TPY unit, and the incomplete d → f energy transfer from the [(N^C^N)Pt(ii)] antenna donor to the Eu(iii) center with maximum efficiency of 51.8% are observed. Using an in situ mixed titration strategy, the R/G/B emission triads consisted of red [(TPY)Eu(HFA)3] and green [(N^C^N)Pt(ii)] dual phosphorescence and blue [(TPY)Zn(TPY)] fluorescence, which can be well balanced to realize the white-light-emission with CIE coordinates (x = 0.36, y = 0.36) by precisely controlling the molar ratio (9  1  2) of the parent complexes, Eu(HFA)3 and Zn(ClO4)2.In this work, we report the synthesis, unexpected glass-forming properties, molecular dynamics and conformational analysis of two thiacrown ethers 6-methyl-2,3-dihydro-1,4-benzodithiine (1), with a six-membered heterocyclic ring, and macrocyclic 2,3-(4'-methylbenzo)-1,4-dithia-7-oxacyclononane (2). Based on the calorimetric studies, we showed that compound 1 is a viscous liquid at room temperature undergoing vitrification at 192 K. Compound 2 is a crystalline solid at room temperature characterized by a melting point at 331 K; however, it can be vitrified with ease after being melted by cooling down to 224 K. This gave us the unique possibility to analyze the dielectric response and to follow the molecular dynamics in supercooled liquid and glassy states for each thiacrown ether. Two relaxation processes were found for compound 1, which are structural α-relaxation, connected with the collective rotational motions of molecules in a liquid, and a low-temperature secondary γ-process, resulting from conformational changes in the heterocyclic ring. Beside these two relaxation processes, an additional intermolecular β-process of JG type was detected in the case of compound 2. Finally, based on the analysis of the thermal evolution of the Kirkwood-Fröhlich factor, it has also been shown that thiacrown ethers may be characterized by a local ordering between neighboring molecules in the supercooled liquid state.Big data often has emergent structure that exists at multiple levels of abstraction, which are useful for characterizing complex interactions and dynamics of the observations. Here, we consider multiple levels of abstraction via a multiresolution geometry of data points at different granularities.   • Single image dehazing is a difficult problem because of its ill-posed nature. Increasing attention has been paid recently as its high potential applications in many visual tasks. Although single image dehazing has made remarkable progress in recent years, they are mainly designed for haze removal in daytime. In nighttime, dehazing is more challenging where most daytime dehazing methods become invalid due to multiple scattering phenomena, and non-uniformly distributed dim ambient illumination. While a few approaches have been proposed for nighttime image dehazing, low ambient light is actually ignored. In this paper, we propose a novel unified nighttime hazy image enhancement framework to address the problems of both haze removal and illumination enhancement simultaneously. Specifically, both halo artifacts caused by multiple scattering and non-uniformly distributed ambient illumination existing in low-light hazy conditions are considered for the first time in our approach. More importantly, most current daytime dehazing methods can be effectively incorporated into nighttime dehazing task based on our framework. Firstly, we decompose the observed hazy image into a halo layer and a scene layer to remove the influence of multiple scattering. After that, we estimate the spatially varying ambient illumination based on the Retinex theory. We then employ the classic daytime dehazing methods to recover the scene radiance. Finally, we generate the dehazing result by combining the adjusted ambient illumination and the scene radiance. Monocytes and macrophages are major cells that contribute to the initiation and progression of atherosclerotic plaques. During this process, an accumulation of LDL-laden macrophages (foam cells) and an alteration in the extracellular matrix (ECM) organization leads to a local vessel stiffening. Current in vitro models are carried out onto two-dimensional tissue culture plastic and cannot replicate the relevant microenvironments. To bridge the gap between in vitro and in vivo conditions, we utilized three-dimensional (3D) collagen matrices that allowed us to mimic the ECM stiffening during atherosclerosis by increasing collagen density. First, human monocytic THP-1 cells were embedded into 3D collagen matrices reconstituted at low and high density. Outdoor recreation, such as rural second home tourism, provides extensive exposure to the natural environment, but research around health impacts of this exposure is scattered. We review current research on health and well-being impacts of nature and discuss how the characteristics of rural second home environments and their use and users can affect these potential impacts in Finland. We discover four key issues affecting the impacts. First, health and well-being impacts depend on the users; urban people can especially benefit from rural second homes, while child development and the performance of elderly people can also be supported by contact with nature at second homes. Second, the regularity, length and season of second home visits influence the potential to receive benefits as they have an impact on the intensity of nature exposure. Third, the type and quality of second home environment affect contact with nature, such as exposure to health-supporting environmental microbes. Fourth, practices, motives and meanings modify activities and attachment and crucially affect both physical and mental well-being. We conclude that rural second homes have extensive potential to provide nature-related health and well-being benefits and further research is needed.The COVID-19 pandemic has taken the entire globe by storm. Currently the ultimate treatment is palliative; however, many modalities have been introduced with effective or minimal outcomes. Meanwhile, enormous efforts are ongoing to produce safe vaccines and therapies. Phototherapy has a wide range of clinical applications against various maladies. This necessitates the exploration of the role of phototherapy, if any, for COVID-19. This critical review was conducted to understand COVID-19 disease and highlights the prevailing facts that link phototherapy utilisation as a potential treatment modality for SARS-CoV-2 viral infection. The results demonstrated phototherapy's efficacy in regulating cytokines and inflammatory mediators, increasing angiogenesis and enhancing healing in chronic pulmonary inflammatory diseases. In conclusion, this review answered the following research question.

  • Hydrophobic photonic crystals (PCs) has been increasingly appreciated as a promising functional material due to their distinct surface characteristic of structural color and hydrophobicity. However, it remains a challenge to fabricate hydrophobic PCs via a one-step process. Inspired by the development of high-performance waterborne coatings, we propose an easy-to-perform and high-efficiency strategy to construct hydrophobic building blocks (diameter of 221, 247, 276 and 305 nm), where the umbelli-form hydrophobic long chain (veova10 Cn > 9) was loaded onto polystyrene (PS) colloidal particles in situ. Taking advantage of the hydrophobic driving force between the colloidal particles, large-scale colloidal photonic crystals (CPCs) film with crack-free morphology was obtained efficiently. The derived CPCs exhibit robust mechanical stability, prominent hydrophobicity and excellent optical properties. In addition, the colloidal latex holds great potential toward PCs coatings on a variety of substrates (glass, plastic and steel) with excellent adhesiveness. Furthermore, we contrive to construct angle-independent structural color films and supraballs, and explore their application in quantum dots (QDs) fluorescence enhancement, which achieved an enhancement effect by more than eight times.     Hydrophobic photonic crystals (PCs) has been increasingly appreciated as a promising functional material due to their distinct surface characteristic of structural color and hydrophobicity. However, it remains a challenge to fabricate hydrophobic PCs via a one-step process. Inspired by the development of high-performance waterborne coatings, we propose an easy-to-perform and high-efficiency strategy to construct hydrophobic building blocks (diameter of 221, 247, 276 and 305 nm), where the umbelli-form hydrophobic long chain (veova10 Cn > 9) was loaded onto polystyrene (PS) colloidal particles in situ. Taking advantage of the hydrophobic driving force between the colloidal particles, large-scale colloidal photonic crystals (CPCs) film with crack-free morphology was obtained efficiently. The derived CPCs exhibit robust mechanical stability, prominent hydrophobicity and excellent optical properties.     A total of 38 articles reporting 55 studies from 32 countries and one territory worldwide were included in the review. We found considerable heterogeneity in the populations studied, methods used and estimated prevalence of
    parasites with
    deletions. The derived prevalence of
    deletions ranged from 0% to 100%, including focal areas in South America and Africa. Only three studies (5%) fulfilled all seven criteria for study quality.

    The lack of representative surveys or consistency in study design impairs evaluations of the risk of false-negative results in malaria diagnosis due to
    deletions. Accurate mapping and strengthened monitoring of the prevalence of

    From the results of an initial feasibility study, we adapted the World Health Organization's criteria defining a maternal near miss to the Namibian health-care system. We visited most (27 out of 35) participating facilities before implementation and provided training on maternal near-miss identification and data collection. We visited all facilities at the end of the surveillance period to verify recorded data and to give staff the opportunity to provide feedback.

    During the 6-month period, we recorded 37 106 live births, 298 maternal near misses (8.0 per 1000 live births) and 23 maternal deaths (62.0 per 100 000 live births). We observed that obstetric haemorrhage and hypertensive disorders were the most common causes of maternal near misses (each 92/298; 30.9%). Of the 49 maternal near misses due to pregnancies with abortive outcomes, ectopic pregnancy was the most common cause (36/298; 12.1%). Fetal or neonatal outcomes were poor; only 50.3% (157/312) of the infants born to maternal near-miss mothers went home with their mother.

    Maternal near-miss surveillance is a useful intervention to identify within-country challenges, such as lack of access to caesarean section or hysterectomy. Knowledge of these challenges can be used by policy-makers and programme managers in the development of locally tailored targeted interventions to improve maternal outcome in their setting.

We identified clear potential routes for virus transmission to humans through local practices, including fruit consumed by bats and harvested by humans when Nipah virus is circulating, and palm juice production.   • The COVID-19 pandemic has placed extraordinary demands on healthcare services worldwide. Some have reported increased viral transmission to healthcare workers during aerosol-generating procedures such as intubation. We report our experience with universal preoperative and preprocedural screening for COVID-19 in children requiring general anaesthesia with low risk of having the infection.

    This was a data review involving children aged < 18years undergoing procedures under general anaesthesia in our institution from 18th March to 31st May 2020 and deemed low risk for COVID 19 infection, as defined by our institutional protocol. Confirmation of COVID-19 was by real time reverse-transcription polymerase chain reaction with confirmation by nucleic acid sequencing. All patients were followed up 14days post-swab.

    Of the 66 children (median age 4.5years, range 0-16.8) eligible for the study, 39 (60%) were male and infants (1-12months) formed the largest group (n = 24, 36%). None were positive for COVID-19. On 14day follow-up, none had symptoms related to COVID-19.

    Our study shows that low risk asymptomatic children with no history of contact with COVID-19 patients test negative on universal screening. Larger studies are required to ascertain the role of screening prior to procedures done under general anaesthesia.

    Midgut volvulus is associated with intestinal ischemia/reperfusion (IR) injury and can progress to severe intestinal damage. Remote ischemic conditioning (RIC) reduces IR-induced injury in distant organs. The aim of this study was to investigate whether RIC protects the intestine from IR injury.

    We investigated intestinal IR injury in 3weeks old SD rats. Animals underwent (i) sham laparotomy, (ii) intestinal IR injury, (iii) intestinal IR + RIC during ischemia, or (iv) intestinal IR + RIC after reperfusion. #link# Intestinal IR injury was achieved by 45min occlusion of superior mesenteric artery followed by de-occlusion. The aim of this study was to determine the potential causal effects of circulating SHBG concentrations on bone mineral density (BMD). Two-sample Mendelian randomization (MR) approach was applied in analyses. From summary-level data of genome-wide association studies (GWAS), we selected 11 single-nucleotide polymorphisms (SNPs) associated with SHBG levels as instrumental variable, and used summary statistics for BMD at forearm (FA) (n = 8143), femoral neck (FN) (n = 32,735), lumbar spine (LS) (n = 28,498) and heel (HL) (n = 394,929), and total-body BMD of different age-stages (15 or less, 15-30, 30-45, 45-60, 60 or more years old) (n = 67,358). Inverse causal associations was observed between SHBG levels and FA BMD (Effect = - 0.26; 95% CI - 0.49 to - 0.04; P = 0.022), HL eBMD (Effect = - 0.09; 95% CI - 0.12 to - 0.06; P = 3.19 × 10-9), and total-body BMD in people aged 45-60 years (Effect = - 0.16; 95% CI - 0.31 to - 2.4 × 10-3; P = 0.047) and over 60 years (Effect = - 0.19; 95% CI - 0.33 to - 0.05; P = 0.006). According to the molecular biology results, mostly well-known freshwater microorganisms, adapted to nutrient-poor conditions were found in the pelagic water column. The aim of this study was to determine the potential causal effects of circulating SHBG concentrations on bone mineral density (BMD). Two-sample Mendelian randomization (MR) approach was applied in analyses. From summary-level data of genome-wide association studies (GWAS), we selected 11 single-nucleotide polymorphisms (SNPs) associated with SHBG levels as instrumental variable, and used summary statistics for BMD at forearm (FA) (n = 8143), femoral neck (FN) (n = 32,735), lumbar spine (LS) (n = 28,498) and heel (HL) (n = 394,929), and total-body BMD of different age-stages (15 or less, 15-30, 30-45, 45-60, 60 or more years old) (n = 67,358). Inverse causal associations was observed between SHBG levels and FA BMD (Effect = - 0.26; 95% CI - 0.49 to - 0.04; P = 0.022), HL eBMD (Effect = - 0.09; 95% CI - 0.12 to - 0.06; P = 3.19 × 10-9), and total-body BMD in people aged 45-60 years (Effect = - 0.16; 95% CI - 0.31 to - 2.4 × 10-3; P = 0.047) and over 60 years (Effect = - 0.19; 95% CI - 0.33 to - 0.05; P = 0.006). Our study demonstrates that circulating SHBG concentrations are inversely associated with FA and HL eBMD, and total-body BMD in people aged over 45 years, suggesting that the role of SHBG in the development of osteoporosis might be affected by chronological age of patients and skeletal sites.ALDH2 inactivating mutation (ALDH2*2) is the most abundant mutation leading to bone morphological aberration. Osteoporosis has long been associated with changes in bone biomaterial in elderly populations. Such changes can be exacerbated with elevated ethanol consumption and in subjects with impaired ethanol metabolism, such as carriers of aldehyde dehydrogenase 2 (ALDH2)-deficient gene, ALDH2*2. So far, little is known about bone compositional changes besides a decrease in mineralization. Raman spectroscopic imaging has been utilized to study the changes in overall composition of C57BL/6 female femur bone sections, as well as in compound spatial distribution. Raman maps of bone sections were analyzed using multilinear regression with these four isolated components, resulting in maps of their relative distribution. A 15-week treatment of both wild-type (WT) and ALDH2*2/*2 mice with 20% ethanol in the drinking water resulted in a significantly lower mineral content (p  0.4). Highly localized islets of elongated adipose tissue were observed on most maps. Elevated fat content was found in ALDH2*2 knock-in mice consuming ethanol (p  less then  0.0001) and this effect appeared cumulative. This work conclusively demonstrates that that osteocytes in femurs of older female mice accumulate fat, as has been previously theorized, and that fat accumulation is likely modulated by levels of acetaldehyde, the ethanol metabolite.Lake Balaton is the largest European shallow lake, which underwent cultural eutrophication in the '70-80s. Therefore, strict pollution control measures were introduced and the water quality has become meso-eutrophic since the millennium. Due to the touristic significance and change in trophic levels of the lake, numerous ecological studies were carried out, but none of them was focused on both benthic and planktonic microbial communities at the same time. In our study, an attempt was made to reveal the spatial bacterial heterogeneity of the Lake Balaton and Zala River by 16S rDNA terminal restriction fragment length polymorphism fingerprinting and Illumina amplicon sequencing methods in the summer of 2017. According to the molecular biology results, mostly well-known freshwater microorganisms, adapted to nutrient-poor conditions were found in the pelagic water column. The LD12 subclade member Fonsibacter ubiquis, the cyanobacterial Synechococcus sp. and unknown Verrucomicrobia species were abundant in the less nutrient-dense basins, while the hgcI clade members showed various distribution.

Other studies aimed to further our understanding of the effects of endocrine disrupting chemicals, pharmaceuticals, and personal care products. Further research presented in this virtual issue examined the interactive effects of chemicals and pathogens, while the final set of manuscripts demonstrates continuing efforts to combine traditional biomonitoring, data from -omic technologies, and modeling for use in risk assessment and management. An additional goal of PRIMO meetings is to address the link between environmental and human health. Several articles in this issue of Aquatic Toxicology describe the appropriateness of using aquatic organisms as models for human health, while the keynote speakers, as described in the editorial below, presented research that highlighted bioaccumulation of contaminants such as PFOS and mercury from fish to marine mammals and coastal human populations such as the Gullah/GeeChee near Charleston, South Carolina, USA.Chemical contaminants can be discharged by vessel hull cleaning processes, such as scraping, jet spraying, and painting, all of which produce readily transportable contaminants into the marine environment, where they are referred to as 'hotspots' of contamination in coastal areas. However, many countries have not yet established effective evaluation methods for disposal of waste mixtures or management guidelines for areas of hull cleaning. To define the toxic effects of wastewater from vessel hull cleaning in dry docks on resident non-target organisms, we investigated the chemical concentrations and developmental toxicity on embryonic flounder, which is an organism sensitive to chemical contamination. In this study, the dominant inorganic metal discharged was zinc when cleaning Ship A (300 tons) and copper for Ship B (5,000 tons). The wastewater from high-pressure water blasting (WHPB) of Ship A (300 tons) and Ship B (5,000 tons) produced a largely overlapping suite of developmental malformations including pe inform WHPB effluent management practices to reduce impacts on non-target coastal organisms.The DNA repair system has evolved from the common ancestor of all life forms and its function is highly conserved within eukaryotes. In this study, to reveal the role of DNA double-strand break repair (DSB) genes in response to benzo[α]pyrene (B[α]P), we first identified DSB genes in relation to homologous recombination and non-homologous end joining events in four Brachionus rotifer spp. B. calyciflorus, B. koreanus, B. plicatilis, and B. rotundiformis. In all the Brachionus spp., 39 orthologous genes to human DSB repair genes were identified. Furthermore, three genes in B. koreanus, two genes in B. plicatilis, and one gene in B. calyciflorus and B. rotundiformis were present as duplicated genes, indicating that these genes were diversified over speciation in the genus Brachionus. Moreover, we compared DSB repair genes on the gene structures in four monogonont Brachionus rotifers and the bdelloid rotifer Adineta vaga, which possesses highly efficient DNA repair ability. The transcriptional responses of four monogonont Brachionus rotifers in response to B[α]P exposure showed how B[α]P exposure led to DSBs and subsequently recruited DNA DSB repair pathways in the rotifer B. koreanus. Taken together, this study provides a better understanding of the potential role of DSB repair genes in the monogonont rotifer Brachionus spp. in response to B[α]P.
    Recurrent dislocation after total hip arthroplasty (THA) using the dual mobility cup system can present challenges, while dual-mobility THA bearings can improve stability in both primary and revision total hip arthroplasties.

    A 72-year-old woman with a history of schizophrenia underwent a left primary THA using the G7 dual mobility system. Two postoperative posterior dislocations occurred within 2 months post-surgery. The patient underwent revision surgery in which the metal liner and dual mobility head were exchanged using the Freedom constrained liner system without revision of the cup and stem. As of this writing, 28 months after the revision surgery, no further dislocations have occurred. The implants are stable, and the patient has good range of motion.

    Dual-mobility bearings are utilized to improve stability in both primary and revision total hip arthroplasties, but even if the dual mobility system is chosen, it sometimes fails to prevent postoperative dislocation. Although a relatively high failure rate has been reported in THA using the constrained liner system, this patient's clinical course suggests that the G7 Freedom constrained liner system can be an efficacious option for some patients with unstable hip joints who undergo THA using the dual mobility system.

  • This review aims to summarize current knowledge and provide a basis for developing effective tailor-made formulations for cancer therapy in the future.In this study, we built and tested a contingency model linking leader daily empowering behaviors with employee daily job crafting. Drawing on the contingency leadership literature and the model of proactive motivation, we theorized employee daily work meaning and vigor as moderators of the above relationships. Daily data were collected from 103 Chinese employees for five consecutive days. Our findings suggest that leader dayT (a certain day) empowering behaviors are more strongly related to employee dayT+1 (next day after the certain day) job crafting when employee dayT work meaning is low and employee dayT+1 vigor is high. Our findings suggest that only under certain conditions can empowering leadership promote employee job crafting on a daily basis.Infectious diseases are caused by pathogenic microorganisms, whose transmission can lead to global pandemics like COVID-19. Contact with contaminated surfaces or objects is one of the major channels of spreading infectious diseases among the community. Nevertheless, safety and efficiency are the major concerns of the utilization of human labor in this process. The wall-following behavior of the two fuzzy systems was evaluated through simulations by considering heterogeneous wall arrangements. The simulation results validate the real-world applicability of the proposed FLSs for establishing the wall-following behavior for a wall disinfection robot. Furthermore, the statistical outcomes show that the IT2-FLS has significantly superior performance than the T1-FLS in this application.Antibiotic resistance is now considered a worldwide problem that puts public health at risk. The onset of bacterial strains resistant to conventional antibiotics and the scarcity of new drugs have prompted scientific research to re-evaluate natural products as molecules with high biological and chemical potential. A class of natural compounds of significant importance is represented by alkaloids derived from higher plants. In this review, we have collected data obtained from various research groups on the antimicrobial activities of these alkaloids against conventional antibiotic-resistant strains. In addition, the structure-function relationship was described and commented on, highlighting the high potential of alkaloids as antimicrobials.Interest in using harvest aids (defoliants or crop desiccants) such as paraquat, carfentrazone-ethyl, glyphosate, and sodium chlorate (NaClO3) have become increasingly important to assure harvest efficiency, producer profit, and to maintain seed quality. However, information on the effects of harvest aids on seed nutrition (composition) (protein, oil, fatty acids, sugars, and amino acids) in soybean is very limited. The objective of this research was to investigate the influence of harvest aids on seed protein, oil, fatty acids, sugars, and amino acids in soybean. Our hypothesis was that harvest aid may influence seed nutrition, especially at R6 as at R6 the seeds may still undergo biochemical changes.   • Accurate data on the Lassa virus (LASV) human case fatality rate (CFR) and the prevalence of LASV in humans, rodents and other mammals are needed for better planning of actions that will ultimately reduce the burden of LASV infection in sub-Saharan Africa. Even though, serine is an essential component of the sphingolipid biosynthesis pathway, its role in maintaining sphingolipid homeostasis has not been precisely studied. Here we show that serine uptake is an important factor for the regulation of sphingolipid biosynthesis in Saccharomyces cerevisiae. Using genetic experiments, we find the broad-specificity amino acid permease Gnp1 to be important for serine uptake. We confirm these results with serine uptake assays in gnp1Δ cells. We further show that uptake of exogenous serine by Gnp1 is important to maintain cellular serine levels and observe a specific connection between serine uptake and the first step of sphingolipid biosynthesis. Using mass spectrometry-based flux analysis, we further observed imported serine as the main source for de novo sphingolipid biosynthesis. Our results demonstrate that yeast cells preferentially use the uptake of exogenous serine to regulate sphingolipid biosynthesis. Our study can also be a starting point to analyze the role of serine uptake in mammalian sphingolipid metabolism.
    Non-alcoholic fatty liver disease (NAFLD) has been associated with small bowel bacterial overgrowth (SIBO) and cardiometabolic dysfunction. This cross-sectional study aimed to evaluate the cardio-metabolic parameters and SIBO in patients with different degrees of hepatic fibrosis estimated by NAFLD fibrosis score (NFS).

    Subjects (n = 78) were allocated to three groups Healthy control (n = 30), NAFLD with low risk of advanced fibrosis (NAFLD-LRAF, n = 17) and NAFLD with a high risk of advanced fibrosis (NAFLD-HRAF, n = 31). Anthropometrics, blood pressure, electrocardiogram and heart rate variability (HRV) were evaluated. Only the NAFLD-LRAF and NAFLD-HRAF groups were submitted to blood biochemical analysis and glucose hydrogen breath tests.

    The NAFLD-HRAF group had higher age and body mass index when compared to the control and NAFLD-LRAF groups. The prevalence of SIBO in the NAFLD group was 8.33%. The low frequency/high-frequency ratio (LF/HF ratio) was augmented in NAFLD-LRAF (p < 0.05) when compared with control group. NAFLD-HRAF group had a wide QRS complex (p < 0.05) and reduced LF/HF ratio (p < 0.05) compared to the control and NAFLD-LRAF groups. Serum levels of albumin and platelets were more reduced in the NAFLD-HRAF subjects (p < 0.05) than in the NAFLD-LRAF.

  • Enzyme immobilization in metal-organic frameworks (MOFs) as a promising strategy is attracting the interest of scientists from different disciplines with the expansion of MOFs' development. Different from other traditional host materials, their unique strengths of high surface areas, large yet adjustable pore sizes, functionalizable pore walls, and diverse architectures make MOFs an ideal platform to investigate hosted enzymes, which is critical to the industrial and commercial process. In addition to the protective function of MOFs, the extensive roles of MOFs in the enzyme immobilization are being well-explored by making full use of their remarkable properties like well-defined structure, high porosity, and tunable functionality. Such development shifts the focus from the exploration of immobilization strategies toward functionalization. Meanwhile, this would undoubtedly contribute to a better understanding of enzymes in regards to the structural transformation after being hosted in a confinement environment, particularly to the orientation and conformation change as well as the interplay between enzyme and matrix MOFs. In this Outlook, we target a comprehensive review of the role diversities of the host matrix MOF based on the current enzyme immobilization research, along with proposing an outlook toward the future development of this field, including the representatives of potential techniques and methodologies being capable of studying the hosted enzymes.Covalent adaptable networks (CANs) are covalently cross-linked polymers that may be reshaped via cross-linking and/or strand exchange at elevated temperatures. They represent an exciting and rapidly developing frontier in polymer science for their potential as stimuli-responsive materials and to make traditionally nonrecyclable thermosets more sustainable. CANs whose cross-links undergo exchange via associative intermediates rather than dissociating to separate reactive groups are termed vitrimers. Vitrimers were postulated to be an attractive subset of CANs, because associative cross-link exchange mechanisms maintain the original cross-link density of the network throughout the exchange process. As a result, associative CANs demonstrate a gradual, Arrhenius-like reduction in viscosity at elevated temperatures while maintaining mechanical integrity. In contrast, CANs reprocessed by dissociation and reformation of cross-links have been postulated to exhibit a more rapid decrease in viscosity with increasing teiderations.

    We divided participants from Alzheimer's Disease Neuroimaging Initiative at the 50th percentile of the predicted absolute risk of the polygenic hazard score (PHS). Outcome measures were the Alzheimer's Disease Assessment Schedule-Cognitive Subscale (ADAS-Cog), ADNI-Mem, Clinical Dementia Rating-Sum of Boxes (CDR SB), and Cognitive Function Composite 2 (CFC2). In addition to modeling, we use a power analysis compare numbers needed with each technique.

    Data from 188 cognitively normal and 319 mild cognitively impaired (MCI) participants were analyzed. Using the ADAS-Cog to estimate sample sizes, without stratification over 24 months, would require 930 participants with MCI, while using the CFC2 and restricting participants to those in the upper 50th percentile would require only 284 participants.

    Combining stratification by PHS and selection of a sensitive combined outcome measure in a cohort of patients with MCI can allow trial design that is more efficient, potentially less burdensome on participants, and more cost effective.
