  • Since its introduction to clinical practice, preimplantation genetic testing (PGT) has become a standard of care for couples at risk of having children with monogenic disease, and for chromosomal aneuploidy to improve outcomes for patients with infertility. The primary objective of PGT is to reduce the risk of miscarriage and genetic disease and to improve the success of infertility treatment with the delivery of a healthy child. Until recently, the application of PGT to more common but complex polygenic disease was not possible, as the genetic contribution to polygenic disease has been difficult to determine, and the concept of embryo selection across multiple genetic loci has been difficult to comprehend. Several achievements, including the ability to obtain accurate, genome-wide genotypes of the human embryo, and the development of population level biobanks have now made PGT for polygenic disease risk applicable in clinical practice. With the rapid advances in embryonic polygenic risk scoring, diverse considerations beyond technical capability have been introduced.Experimental information from microscopy, structural biology, and bioinformatics may be integrated to build structural models of entire cells with molecular detail. This integrative modeling is challenging in several ways the intrinsic complexity of biology results in models with many closely packed and heterogeneous components; the wealth of available experimental data is scattered among multiple resources and must be gathered, reconciled, and curated; and computational infrastructure is only now gaining the capability of modeling and visualizing systems of this complexity. We present recent efforts to address these challenges, both with artistic approaches to depicting the cellular mesoscale, and development and application of methods to build quantitative models.Pulmonary arterial hypertension remains a progressive, life-limiting disease despite optimal medical therapy. Pulmonary artery denervation has arisen as a novel intervention in the treatment of pulmonary arterial hypertension, and other forms of pulmonary hypertension, with the aim of reducing the sympathetic activity of the pulmonary circulation. Pre-clinical studies and initial clinical trials have demonstrated that the technique can be performed safely with some positive effects on clinical, haemodynamic and echocardiographic markers of disease. The scope of the technique in current practice remains limited given the absence of well-designed, large-scale, international randomised controlled clinical trials. This review provides an overview of this exciting new treatment modality, including pathophysiology, technical innovations and recent trial results.Measles virus, a member of the genus Morbillivirus, is highly contagious and still shows considerable mortality with over 100000 deaths annually, although efficient attenuated vaccines exist. LAMP-2 may serve as a cholesterol reservoir, whereas LIMP-2, like NPC1, is able to transport cholesterol through a transglycocalyx tunnel. Contact sites and fusion events between lysosomes and other organelles mediate the distribution of cholesterol. Reductions in feeding efficiency and increases in energy expenditure were associated with body weight regulation in response to long-term disulfiram treatment. Loss of fat tissue and an increase in liver fenestrations were also observed in rats on disulfiram. Given the potent anti-obesogenic effects in rodents, repurposing disulfiram in the clinic could represent a new strategy to treat obesity and its metabolic comorbidities.

  • In this study, preparation of a nylon film-TiO2 composite sheet with both physical durability under UV irradiation and TiO2 photocatalysis functionality was investigated. First, a nylon film was directly prepared on paper by interfacial polymerization using ethylenediamine and terephthaloyl chloride in a cyclohexane-chloroform mixture (31, v/v). Next, the nylon-coated paper was treated with tetraethyl orthosilicate and 3-aminopropyltrimethoxysilane to prepare polysiloxane on its surface. This was followed by fixation of TiO2 powder via electrostatic interactions with the polysiloxane. Although nylon films on paper usually decompose under TiO2 photocatalysis, the nylon film-TiO2 composite sheets prepared using 0.5%-1.0% (w/v) TiO2 did not decompose under photocatalysis. The residual rate of strength of the sheet remained at almost 100% after 240 h, which could be attributed to protection of the sheet by the polysiloxane layer. The nylon film was fibrous and could effectively adsorb acetaldehyde gas. All of the nylon film-TiO2 composite sheets prepared using 0.5%-5.0% TiO2 photocatalytically removed acetaldehyde under UV irradiation and no acetaldehyde gas was detected after 240-300 min. These results show the nylon film-TiO2 composite sheet can effectively remove acetaldehyde gas by photocatalysis and adsorption and could be applied to removal of volatile organic compounds in indoor air.This study investigated the feasibility of co-treating H2S and CO2 in a biological trickling filter (BTF) inoculated with hydrogenotrophic methanogens (HMs) and nitrate-reducing, sulfur-oxidizing bacteria. This was accomplished by introducing a pure culture of Thiobacillus denitrificans in a BTF that was successfully upgrading a biogas mimic (6040 CH4CO2) to >97% methane using an enriched HM consortium. Nitrate was fed as the electron acceptor to oxidize H2S. The results revealed that a severe competition for hydrogen's electrons occurred between carbon dioxide and nitrate. Due to this competition, NS loading rates of 161 were required to achieve >98% H2S removal, a ratio which is four times greater than the theoretical NS ratio for complete sulfur oxidation. However, such high nitrate loading rates (>50 g N-NO3- m-3 h-1) had a negative impact on the BTF's biogas upgrading performance. An electron balance illustrated the increasing diversion of H2 electrons towards nitrate reduction as nitrate loading increased. Overall, this study showed that simultaneous biogas upgrading and H2S removal in a single bioreactor is possible, but that achieving high yields for both reactions requires further research in process and culture optimization.Accurate estimation of surface PM2.5 concentration is critical for the assessment of PM2.5 exposure and associated health impacts. Due to the limited spatial coverage of ground monitoring stations, most studies often use the satellite products to estimate surface PM2.5 concentration by constructing a comprehensive relationship between satellite-retrieved aerosol optical depth (AOD) and ground-based measured PM2.5 concentration with machine learning (ML) technologies. However, uncertainties of ML-based models may lead to considerable biases in PM2.5 estimation, which need carefully examined. Here we evaluate the accuracy of estimated PM2.5 concentration from two popular ML-models (i.e., Random Forest and the BP Neural Network) which were trained and tested using hourly data of satellite-retrieved AOD from HIMAWARI, ground-based measured PM2.5 from China National Environmental Monitoring Center, ERA5 meteorological conditions, and other auxiliary variables for a whole year of 2017 over China. We propose a new validation method considering the spatial pattern of the data during the validation. The results suggest that the traditional validation methods may overestimate the performance of the models on estimating the PM2.5 at the area with sparse in-situ measurements. Moreover, the spatial distribution pattern of the training data will largely affect the evaluation of models performance, which should be carefully considered. No product was identified in the reaction of Syr and OH radical at pH 3 due probably to the rapid mineralization of phenoxy radical formed from HAT, while products after OH additions were identified for a reaction at pH 6. On the other hand, reaction of Syr with SO4- starts by single-electron transfer (SET), forming Syr+, which can undergo hydrolysis, sulfation, and dimerization with Syr and other aromatic intermediates, etc. Dimerization products from the phenoxy-type radical were not found here.Chronic kidney disease is emerging as a novel risk factor for cerebrovascular disease, but this association remains largely unexplored in older adults. Cystatin C is a more accurate measure than creatinine of kidney function in the elderly. We evaluated cystatin C, cognitive function, and brain imaging in 193 participants from The 90+ Study neuroimaging component. The mean age was 93.9 years; 61% were women. Mean cystatin C was 1.62 mg/L with estimated glomerular filtration rate 39.2 mL/min/1.73 m2. Performance on measures of global cognition, executive function, and visual-spatial ability declined at higher tertiles of cystatin C (lower kidney function). Higher cystatin C was significantly associated with infratentorial microbleeds and lower gray matter volume. Adjusted risk of incident dementia was increased in the middle and high cystatin C tertile groups compared with the low group (hazard ratio in highest tertile 3.81 [95% confidence interval 1.14-12.7]), which appeared to be explained in part by the presence of cerebral microbleeds.

  • Despite the positive achievements attained, the treatment of male urethral strictures and hypospadiases still remains a challenge, particularly, in cases of severe urethral defects. Complication rate and need for additional interventions in such cases are the highest. Also, shortage of autologous tissue for graft harvesting and significant morbidity on the place of harvesting present a problem and often lead to staged treatment. Tissue engineering provides a promising alternative to the current sources of grafts for urethroplasty. Since the first experiments in urethral substitution with tissue engineered grafts, this topic in regenerative medicine has grown remarkably, as many different types of tissue engineered grafts and approaches in graft design have been suggested and tested in vivo. However, there have been only several clinical trials of tissue engineered grafts in urethral substitution with hardly more than hundred patients overall. This indicates that the topic is still in its inception, and the searching for the best graft design is continuing. The current review focuses on the state of art in urethral regeneration with tissue engineering technology. It gives comprehensive overview of components of the tissue engineered graft and overview of steps in the graft development. Different cell sources, types of scaffolds, assembling approaches, options for vascularization enhancement and preclinical models are considered.Constructing multicomponent electrode materials with a rational structure is an effective route to develop high-performance supercapacitors. We herein report a novel nickel-foam-supported hierarchical CoO@Ni(OH)2 nanowire-nanosheet core-shell heterostructure arrays synthesized by a facile hydrothermal-electrodeposition strategy. The core CoO nanowire arrays with good electrical conductivity and shell Ni(OH)2 nanosheets with thickness of ~ 2 nm synergistically contributes to increased active sites, fast mass transfer, and improved structural stability.     Despite the positive achievements attained, the treatment of male urethral strictures and hypospadiases still remains a challenge, particularly, in cases of severe urethral defects. Complication rate and need for additional interventions in such cases are the highest. Also, shortage of autologous tissue for graft harvesting and significant morbidity on the place of harvesting present a problem and often lead to staged treatment. Tissue engineering provides a promising alternative to the current sources of grafts for urethroplasty. Since the first experiments in urethral substitution with tissue engineered grafts, this topic in regenerative medicine has grown remarkably, as many different types of tissue engineered grafts and approaches in graft design have been suggested and tested in vivo. However, there have been only several clinical trials of tissue engineered grafts in urethral substitution with hardly more than hundred patients overall. This indicates that the topic is still in its inception, and the searching for the best graft design is continuing. The current review focuses on the state of art in urethral regeneration with tissue engineering technology. Los participantes refirieron que la aplicación contribuía a mejorar el acceso a material de inyección, a reducir el estigma de los drogodependientes, y a optimizar la planificación del usuario para adquirir la jeringa en el proceso ritual del consumo. El objetivo de este estudio es evaluar las diferencias en la sintomatología depresiva (TDM-P vs. TDM- IC) en los pacientes con TCC para mejorar su tratamiento. Métodos Se llevó a cabo un análisis secundario en una muestra transversal de 160 pacientes que presentaban TCC y algún TDM. La evaluación clínica, así como el diagnóstico diferencial entre TDM-P y TDM-IC, se realizó utilizando la entrevista PRISM. Resultados Los hombres representaron el 80% de la muestra con una edad media de 38,61 años y el 64,5% só** tenía estudios primarios. El diagnóstico de TDM-IC (61,3%) fue más frecuente que el de TDM-P (38,7%). Los pacientes con TDM-IC mostraron una edad de aparición más temprana para el TCC. El 79,4% de los pacientes cumplían criterios para otro trastorno por consumo de sustancias. Únicamente el criterio “Cambios en el peso o en el apetito” fue estadísticamente más prevalente (57,1%) en los pacientes con TDM-P. Adicionalmente se administraron el MULTICAGE-TIC y el Inventario de Síntomas Prefrontales (ISP). Se obtuvo una muestra de n=748 sujetos, 33% varones y 94% nacidos y residentes en España. El test obtuvo adecuados valores de consistencia interna, aplicando diferentes estimadores. Se realizó un análisis factorial confirmatorio sobre las escalas teóricas, alcanzando adecuados estimadores de ajuste. Se observó que los componentes obsesivo-compulsivos son de mayor magnitud a medida que se incrementa la implicación en el uso y su progresión al abuso del móvil. Las escalas del OCDUS-TIC mostraron correlaciones de gran magnitud con los síntomas de mal funcionamiento prefrontal, especialmente la de Pensamiento-Interferencia (r>0,80).   • BACKGROUND The relevance of formant-based measures has been noted across a spectrum of medical, technical, and linguistic applications. Therefore, the primary aim of the study was to evaluate the effect of ageing on vowel articulation, as the previous research revealed contradictory findings. The secondary aim was to provide normative acoustic data for all Czech monophthongs. METHODS The database consisted of 100 healthy speakers (50 men and 50 women) aged between 20 and 90. Acoustic characteristics, including vowel duration, vowel space area (VSA), fundamental frequency (fo), and the first to fourth formant frequencies (F1-F4) of 10 Czech vowels were extracted from a reading passage. In addition, the articulation rate was calculated from the entire duration of the reading passage. RESULTS Age-related changes in pitch were sex-dependent, while age-related alterations in F2/a/, F2/u/, VSA, and vowel duration seemed to be sex-independent. In particular, we observed a clear lowering of fo with age for women, but no change for men. With regard to formants, we found lowering of F2/a/ and F2/u/ with increased age, but no statistically significant changes in F1, F3, or F4 frequencies with advanced age. Although the alterations in F1 and F2 frequencies were rather small, they appeared to be in a direction against vowel centralization, resulting in a significantly greater VSA in the older population. The greater VSA was found to be related partly to longer vowel duration. CONCLUSIONS Alterations in vowel formant frequencies across several decades of adult life appear to be small or in a direction against vowel centralization, thus indicating the good preservation of articulatory precision in older speakers. INTRODUCTION Cavernositis is a rare acute infection of the corporal bodies of the penis. It might result in corporal cavernosal abscesses, fibrosis, and subsequently erectile dysfunction if the treatment is inadequate. Previous reports advocate penile aspiration when cavernosal abscesses were significantly present. AIM We report 2 cases of cavernositis treated by the standard antimicrobial therapy in addition to the daily use of phosphodiesterase type 5 inhibitors (iPDE-5). METHODS We include 2 patients who fulfill the criteria for the diagnosis of cavernositis. They were diagnosed clinically based on history and physical examination. Patient 1 had multifocal abscesses on initial penile contrast-enhanced magnetic resonance imaging (MRI). This patient showed rapid clinical improvement with the addition of phosphodiesterase inhibitor (PDEi) to the antimicrobial therapy without the need for surgical drainage of the abscesses. Patient 2 had persistent infection despite 3 weeks of oral second-generation cephalosporin (Cefuroxime).     BACKGROUND The relevance of formant-based measures has been noted across a spectrum of medical, technical, and linguistic applications. Therefore, the primary aim of the study was to evaluate the effect of ageing on vowel articulation, as the previous research revealed contradictory findings. The secondary aim was to provide normative acoustic data for all Czech monophthongs. METHODS The database consisted of 100 healthy speakers (50 men and 50 women) aged between 20 and 90. Acoustic characteristics, including vowel duration, vowel space area (VSA), fundamental frequency (fo), and the first to fourth formant frequencies (F1-F4) of 10 Czech vowels were extracted from a reading passage. In addition, the articulation rate was calculated from the entire duration of the reading passage. RESULTS Age-related changes in pitch were sex-dependent, while age-related alterations in F2/a/, F2/u/, VSA, and vowel duration seemed to be sex-independent. In particular, we observed a clear lowering of fo with age for women, but no change for men. With regard to formants, we found lowering of F2/a/ and F2/u/ with increased age, but no statistically significant changes in F1, F3, or F4 frequencies with advanced age. Although the alterations in F1 and F2 frequencies were rather small, they appeared to be in a direction against vowel centralization, resulting in a significantly greater VSA in the older population. The greater VSA was found to be related partly to longer vowel duration. CONCLUSIONS Alterations in vowel formant frequencies across several decades of adult life appear to be small or in a direction against vowel centralization, thus indicating the good preservation of articulatory precision in older speakers. AIM We report 2 cases of cavernositis treated by the standard antimicrobial therapy in addition to the daily use of phosphodiesterase type 5 inhibitors (iPDE-5). METHODS We include 2 patients who fulfill the criteria for the diagnosis of cavernositis. They were diagnosed clinically based on history and physical examination. Patient 1 had multifocal abscesses on initial penile contrast-enhanced magnetic resonance imaging (MRI). This patient showed rapid clinical improvement with the addition of phosphodiesterase inhibitor (PDEi) to the antimicrobial therapy without the need for surgical drainage of the abscesses. Patient 2 had persistent infection despite 3 weeks of oral second-generation cephalosporin (Cefuroxime).   • Micro ribonucleic acids (miRNAs) are short non-coding RNA molecules responsible for regulation of gene expression. They are involved in many pathophysiological processes of a wide spectrum of diseases. Recent studies showed their involvement in atrial fibrillation. They seem to become potential screening biomarkers for atrial fibrillation and even treatment targets for this arrhythmia. The aim of this review article was to summarize the latest knowledge about miRNA and their molecular relation to the pathophysiology, diagnosis and treatment of atrial fibrillation.This systematic literature review aims to point out sex-specific special features that are important in the bariatric treatment of women suffering from severe obesity. A systematic literature search was carried out according to Cochrane and Preferred Reporting Items for Systematic review and Meta-Analysis Protocols (PRISMA-P) guidelines. After the literature selection, the following categories were determined sexuality and sexual function; contraception; fertility; sex hormones and polycystic ovary syndrome; menopause and osteoporosis; pregnancy and breastfeeding; pelvic floor disorders and urinary incontinence; female-specific cancer; and metabolism, outcome, and quality of life. For each category, the current status of research is illuminated and implications for bariatric treatment are determined. A summary that includes key messages is given for each subsection. An overall result of this paper is an understanding that sex-specific risks that follow or result from bariatric surgery should be considered more in aftercare. In order to increase the evidence, further research focusing on sex-specific differences in the outcome of bariatric surgery and promising treatment approaches to female-specific diseases is needed. Nevertheless, bariatric surgery shows good potential in the treatment of sex-specific aspects for severely obese women that goes far beyond mere weight loss and reduction of metabolic risks.The identification of biomarkers for early diagnosis of Parkinson's disease (PD) is of pivotal importance for improving approaches for clinical intervention. The current study aimed at identifying protein candidates of deimination in plasma and plasma-EVs in a rat model of pre-motor PD, to assess putative contributions of such post-translational changes in the early stages of disease. Numerous observational studies show that not only hyperglycemia but also blood glucose levels near lower fasting limits (72 to 99 mg/dL) increase the incidence of AD, regardless of whether T2DM will develop in the future. As the comorbidity of these diseases and earlier development of AD in T2DM sufferers exist, new AD biomarkers are being sought for etiopathogenetic changes associated with early neurodegenerative processes as a result of carbohydrate disorders. The S100B protein seem to be interesting in this respect as it may be a potential candidate, especially important in early diagnostics of these diseases, given that it plays a role in both carbohydrate metabolism disorders and neurodegenerative processes. It is therefore necessary to clarify the relationship between the concentration of the S100B protein and glucose and insulin levels. The velocity of the RV decreased after ASD transcatheter closure (3.97 ± 1.48 to 3.56 ± 1.4, p = 0.024), especially in the RV inlet and outlet. The average strain decreased (-19.21 ± 5.79 to -16.87 ± 5.03, p = 0.002), as did the average SR (-2.28 ± 0.64 to -2.03 ± 0.61, p = 0.006). The average longitudinal displacement did not differ. With the VVI technique, we could clearly observe RV functional change immediately after transcatheter closure of the ASD.

A ddH2O control and a complex medium for Halobacterium were used as controls. The presence of organisms was tested by enumeration of colonists and sequencing of extracted DNA. At each time point, inoculation into fresh medium was used to test for growth of organisms. After 1 week, the "no salt" and saturated MgSO4 solutions were colonized. After 6 months, both the NaCl-saturated and Halobacterium solutions remained uninhabited, but all other samples were colonized. RESEARCH DESIGN AND METHODS Data from 37 patients with T1D during two clinical studies with identical data collection protocols were analyzed retrospectively four weeks of continuous glucose monitoring, carbohydrate intake, insulin injections, and PA (assessed via wearable activity tracker) were collected in free-living conditions. Five-hour glucose area under curves (GAUC) following the last-bolused meal of every day were computed to assess postprandial glucose excursions, and their relation with corresponding antecedent PA was analyzed using linear mixed effects regression models, accounting for meal, insulin, and current glycemic state. RESULTS Datasets yielded 845 days of data from 37 subjects (22.8±11.6 days/subject); post-meal GAUC was negatively associated with total daily PA measured by step count (p=.025), and total time spent performing higher than light intensity PA (p=.042). Patients with higher median total daily PA exhibited lower average postprandial GAUC (p less then .01). Additional analyses indicated that daily PA likely presents an immediate and delayed impact on glucose control. CONCLUSION Daily PA assessed by commonly available sensors is significantly associated with glycemic exposure after an evening meal, indicating that quantitative assessment of PA may be useful in mealtime treatment decisions.
