METHODS Human PC cells (DU145, PC-3, CWR22) and all docetaxel-resistant cells were maintained in Roswell Park Memorial Institute Medium (RPMI) 1640 media supplemented with 10% fetal bovine serum and 1% penicillin/streptomycin. ABCB1 was detected by using both parental and docetaxel-resistant CRPCs prepared for flow cytometry. For the evaluation of tumor-suppressive effects under each chemotherapeutic agent, subcutaneous xenografts of DU145 or DU145/TXR were implanted at the mouse flank. RESULTS The P-glycoprotein-encoding gene ABCB1 was distinctively upregulated in the resistant cells, and its overexpression played an essential role in docetaxel resistance in CRPC. When tested for the cytotoxicity of gemcitabine, another option for chemotherapy, the docetaxel-resistant cells were shown to become sensitive to the drug, implying additional phenotypic transformation in the docetaxel-resistant cells. Studies using xenograft animal models demonstrated that the growth of tumors composed of both docetaxel-sensitive and docetaxel-resistant cells was deterred most profoundly when docetaxel and gemcitabine were administered together. We suggest that increasing exposure to environmental pollutants due to industrial activity, acting over a lifetime, is also important. The combination of genetic mutations and pollutant exposure, with increased life expectancy, may account for the apparent variations in incidence of the disease in different countries and continents and even regionally within a given country. We focused on the function of the androgen receptor (AR) in HCC and tried to find new treatment strategy based on antiandrogen enzalutamide (Enz). Here, we found that olaparib, a FDA-approved PARP inhibitor, could enhance the cytotoxicity in HCC cells with a lower BRCA1 expression, and suppressing the AR with either Enz or AR-shRNA could further increase the olaparib sensitivity to better suppress the HCC cell growth via a synergistic mechanism that may involve suppressing the expression of BRCA1 and other DNA damage response (DDR) genes. Mechanism studies revealed that Enz/AR signaling might transcriptionally regulate the miR-146a-5p expression via binding to the Androgen Response Elements on its 5' promoter region, which could then lead to suppress the homologous recombination-related BRCA1 expression via direct binding to the mRNA 3'UTR. Preclinical studies using an in vivo mouse model also demonstrated that combining Enz plus olaparib led to better suppression of the HCC progression. Cox regressions were analyzed to assess the effects on LOS of patient demographics and the type of surgery performed. RESULTS Average hospital LOS from infection surgery to discharge was significantly lower in the fast-track cohort (3.8 vs 5.7 days; P = .012). There were no episodes of antibiotic mismatch in the fast-track group vs 1 recorded episode in the control group. No significant differences were noted comparing 90-day complications, reimplantation rate, or 12-month reimplant survival rates. CONCLUSION Through the utilization of an orthopedic-specific infectious disease physician, a fast-track PJI protocol can significantly shorten hospital LOS while remaining safe. Streamlining care pathways may help decrease the overall healthcare costs associated with treating PJI. Additionally, the 2-stage method is the preferred treatment for difficult to treat pathogens as well as in patients who have already undergone a previous revision procedure. An articulating prefabricated antibiotic spacer has entered the armamentarium of 2-stage revision knee surgery, and has demonstrated comparable results to custom and static spacers in terms of the primary goal of infection control. Importantly, the potential for enhanced mobility and function hold promise by safely providing a more "livable" knee during the convalescent period prior to definitive reimplantation. We reviewed the available literature and have found that the data at this point are equivocal with respect to whether antibiotic-impregnated cement intramedullary dowels augment the intra-articular antibiotic cement spacer in eradicating infection in total knee arthroplasty. Thus, we believe that the decision to use dowels can be left up to the surgeon preference. However, further research is warranted to review operative room efficiency and healthcare costs, and to validate the clinical efficacy of antibiotic-impregnated dowels in periprosthetic joint infection. Antibiotic-loaded bone cement (ALBC) spacer constructs for the treatment of periprosthetic joint infections of the knee continue to evolve from the original hockey puck designs. Countless techniques have since been described for augmentation of ALBC spacers with the use of intramedullary (IM) dowels. The use of IM dowels has become a vital addition to any knee spacer construct. ALBC IM dowels are an excellent vessel to provide targeted local antibiotic treatment to high-risk areas like the medullary canal while increasing the overall therapeutic antibiotic elution. In addition, IM dowels provide needed stabilization to the relatively unstable intra-articular spacer component, thereby reducing spacer-related complications like displacement and fracture. Therefore, we recommend regular use of IM dowel augmentation to ALBC spacer constructs. Two-stage exchange arthroplasty remains the gold standard for chronic total knee arthroplasty (TKA) and total hip arthroplasty infections in North America. Cement spacers impregnated with high-dose antibiotics have been successfully used in the interim period of the 2-stage exchange process. A number of spacers have been described; however, this article will focus on articulating spacers. In the presence of an intact extensor mechanism (for TKA), reasonable soft tissue envelope, and adequate bone, articulating antibiotic spacers provide several advantages. These include an infection eradication rate of approximately 90%, higher range of motion after reimplantation, and lower complication rates when compared with nonarticulating spacers. In the appropriate patient, articulating antibiotic spacers are an effective and a safe treatment for infected TKAs and total hip arthroplasties.

In this study, we implanted a window to the mouse uterus to visualize the developing embryo from embryonic day 9.5 to birth. In live mouse embryos, we observed transient neurotransmission and early vascularization of neural crest cell (NCC)-derived perivascular cells in the brain, autophagy in the retina, viral gene delivery, and chemical diffusion through the placenta. We combined the imaging window with in utero electroporation to label and track cell division and movement within embryos and observed that clusters of mouse NCC-derived cells expanded in interspecies chimeras, whereas adjacent human donor NCC-derived cells shrank. This technique can be combined with various tissue manipulation and microscopy methods to study the processes of development at unprecedented spatiotemporal resolution. The underlying principle is to convert a fluctuating, unbiased force into unidirectional motion. Here, we report the ratcheting of electrons at room temperature using a semiconductor nanowire with precisely engineered asymmetry. Modulation of the nanowire diameter creates a cylindrical sawtooth geometry with broken inversion symmetry on a nanometer-length scale. In a two-terminal device, this structure responded as a three-dimensional geometric diode that funnels electrons preferentially in one direction through specular reflection of quasi-ballistic electrons at the nanowire surface. The ratcheting effect causes charge rectification at frequencies exceeding 40 gigahertz, demonstrating the potential for applications such as high-speed data processing and long-wavelength energy harvesting. In this study, we implanted a window to the mouse uterus to visualize the developing embryo from embryonic day 9.5 to birth. In live mouse embryos, we observed transient neurotransmission and early vascularization of neural crest cell (NCC)-derived perivascular cells in the brain, autophagy in the retina, viral gene delivery, and chemical diffusion through the placenta. We combined the imaging window with in utero electroporation to label and track cell division and movement within embryos and observed that clusters of mouse NCC-derived cells expanded in interspecies chimeras, whereas adjacent human donor NCC-derived cells shrank. Here, we report the ratcheting of electrons at room temperature using a semiconductor nanowire with precisely engineered asymmetry. Modulation of the nanowire diameter creates a cylindrical sawtooth geometry with broken inversion symmetry on a nanometer-length scale. In a two-terminal device, this structure responded as a three-dimensional geometric diode that funnels electrons preferentially in one direction through specular reflection of quasi-ballistic electrons at the nanowire surface. The ratcheting effect causes charge rectification at frequencies exceeding 40 gigahertz, demonstrating the potential for applications such as high-speed data processing and long-wavelength energy harvesting. Except for the yellow index b of breast, II genotype individuals had the best growth characteristics, by comparison with the ID genotype and DD genotype individuals. Moreover, the mRNA expression of GOLGB1 was detected in the liver tissue of chicken with different GOLGB1 genotypes, where the DD genotype displayed high expression levels. The circular model, which is based on the partial replacement of primary raw materials by secondary ones, should, on the one hand, limit the depletion of natural resources and, on the other hand, reduce the amount of waste produced. The presented work focuses on the issue of sustainable development, which is closely related to the circular economy, and then explains the circular economy model, including the differences from the linear arrangement and possible obstacles to its implementation for the specific conditions of the Slovak republic. Hydrogeochemical understanding of groundwater is essential for the effective management of groundwater. This study has been carried out to have concrete data for the seasonal variations in hydrogeochemistry of groundwater in central Tamilnadu forming a complex geological terrain with a varied lithology. A total of 244 groundwater samples were collected during four different seasons, viz, southwest monsoon (SWM), summer (SUM), postmonsoon (POM), and northeast monsoon (NEM) from bore wells. The physical parameters such as pH, temperature, TDS, ORP, humidity, and electrical conductivity (EC) were measured insitu, whereas major ions were analyzed in the lab adopting standard procedures. Overall, higher EC and NO3 values were observed and exceeded the WHO permissible limit irrespective of seasons, except for NO3 in SWM. Na and HCO3 are the dominant cation and anion in the groundwater irrespective of seasons. The highest average values of Na (65.06 mg L-1) and HCO3 (350.75 mg L-1) were noted during SWM. We used the filter paper contact test to evaluate (i) the uptake of NPs in organisms exposed to the mixtures of NPs+Metals and NPs+CPF and (ii) the potential effects of the mixture of NPs+CPF on the CPF-induced inhibition of the biomarker enzymes acetylcholinesterase (AChE) and carboxylesterases (CES). We used the artificial soil test to deepen the study on joint effects of NPs+CPF. All compounds were applied separately and in binary mixtures. In the single exposure treatment, Fe levels decreased significantly in organisms exposed to NPs on filter paper, suggesting systemic effects aimed at eliminating Fe incorporated through NPs. Conversely, earthworms exposed to binary mixtures showed Fe levels similar (NPs+Metals) to or higher (NPs+CPF) than controls. The earthworms single exposed to NPs presented no changes in AChE and CES activities. In the artificial soil test, the only treatment that showed AChE inhibition after 72 h was single CPF exposure, while no significant changes were observed in CES activity. However, after 7-day exposure in artificial soil or 72-h exposure on filter paper, the mixture of NPs+CPF induced a similar degree of AChE and CES inhibition as single CPF exposure. All these suggested that NPs did not produce neurotoxic effects, and that the inhibition of the enzymes' activities in all cases was due to the presence of the pesticide. On the other hand, the differences in the pattern of Fe accumulation in the earthworms indicate that the presence of other contaminants in the exposure media can modify the uptake and/or the excretion of Fe and evidence the interactions that may be found in binary mixtures of metal oxide NPs and other pre-existing contaminants in the soil ecosystem.

In the single exposure treatment, Fe levels decreased significantly in organisms exposed to NPs on filter paper, suggesting systemic effects aimed at eliminating Fe incorporated through NPs. Conversely, earthworms exposed to binary mixtures showed Fe levels similar (NPs+Metals) to or higher (NPs+CPF) than controls. The earthworms single exposed to NPs presented no changes in AChE and CES activities. In the artificial soil test, the only treatment that showed AChE inhibition after 72 h was single CPF exposure, while no significant changes were observed in CES activity. Concurrent validity and discriminant validity were assessed using 2-sample t test assuming unequal variances. RESULTS The VELO was translated and optimized using cognitive interviews. The VELO-Nepali demonstrated excellent internal consistency, with Cronbach α coefficients of 0.93, 0.94, and 0.90 for VPI cases, guardians of VPI cases, and non-VPI controls, respectively. Recently newer synthetic DNA vaccines have been rapidly advanced to clinical study and have demonstrated an impressive degree of immune potency and tolerability. Improvements in DNA delivery over prior needle and syringe approaches include jet delivery, gene gun delivery, among others. Among the most effective of these new delivery methods, advanced electroporation (EP), combined with other advances, induces robust humoral and cellular immunity in both preventative as well as therapeutic studies. Advancements in the design of the DNA inserts include leader sequence changes, RNA and codon optimizations, improved insert designs, increased concentrations of DNA, and skin delivery, appear to complement newer delivery strategies. These advances also provide a framework for the in vivo production of synthetic DNA biologics. In this review, we focus on recent studies of synthetic DNA vaccines in the clinic for the prevention or treatment of infectious diseases with a focus on adaptive electroporation for delivery, and briefly summarize novel preclinical data advancing the in vivo delivery of DNA-encoded antibody-like biologics. The occurrence of legacy and novel per- and polyfluoroalkyl substances (PFASs) in multiple matrices from a farmland environment was investigated in the Beijing-Tianjin-Hebei core area of northern China. PFASs were ubiquitously detected in farmland soils, and the detection frequency of 62 chlorinated polyfluoroalkyl ether sulfonic acid (62 Cl-PFESA) was higher than that of perfluorooctane sulfonic acid (98% vs. 83%). Long-chain PFASs, including 62 Cl-PFESA, showed a centered distribution pattern around the metropolis of Tianjin, probably due to the local intensive industrial activity, while trifluoroacetic acid (TFA) showed a decreasing trend from the coast to the inland area. Other than soil, TFA was also found at higher levels than other longer-chain PFASs in dust, maize (Zea mays), poplar (Populus alba) leaf and locust (Locusta migratoria manilens) samples. Both poplar leaves and locusts can be used as promising biomonitoring targets for PFASs in farmland environments, and their accumulation potential corresponds with protein and lipid contents. Apart from being exposed to PFASs via food intake, locusts were likely exposed via uptake from soil and precipitated dust in farmland environments. The biomonitoring of locusts may be more relevant to insectivores, which is important to conducting a comprehensive ecological risk assessment of farmland environments. Although triphenyl phosphate (TPHP) has been reported to disrupt lipid metabolism, the effect of TPHP on lipid saturation remains unexplored. In this study, a lipidomic analysis demonstrated decreases in the levels of poly-unsaturated phosphatidylcholine (PC), phosphatidylethanolamine (PE), and phosphatidylserine (PS) in RAW264.7 murine macrophage cells exposed to 10 μM TPHP. The expression of the gene encoding lysophosphatidylcholine acyltransferase 3 (Lpcat3) was significantly downregulated by 0.76 ± 0.03 and 0.70 ± 0.08-fold in 10 and 20 μM TPHP exposure groups, relative to the control group. This finding explains the observed decrease in lipid saturation. Correspondingly, exposure to 10 and 20 μM TPHP induced endoplasmic reticulum (ER) stress and inflammatory responses, which have been linked to metabolic dysfunction such as insulin resistance and hypertriglyceridemia. Therefore, the treatment strategy needs to be plan very carefully in such case. CASE PRESENTATION We report a patient with 2 wide-neck aneurysms located in the feeding artery of the arteriovenous malformation and he, then, was treated by using Surpass stent (flow-diverter) to eliminate the aneurysms. CONCLUSION In our case, multiple wide-neck aneurysms in combination with low risk unruptured AVM, using long Surpass stent-diversion is an effective way to eliminate all the aneurysms in the main feeding artery while still preserving the AVM anatomy. Anterior chamber foreign bodies account for a considerable portion of all cases of all intraocular foreign bodies (up to 15%); however, they can be missed due to inconspicuous location. CASE REPORT We report two cases of retained intraocular foreign bodies in the iridocorneal angle that was missed at the first ophthalmic examination. They were only discovered when complications occurred, such as corneal edema and increased intraocular pressure. In the case whereby the foreign body was taken out early, corneal damages were reversible. However, in the case whereby the foreign body was taken out late, endothelial damage was irreversible and endothelial transplantation was needed. CONCLUSION Regarding trauma patients, a careful examination should be performed to discover foreign bodies in the iridocorneal angle. In Viet Nam, there was no report on the clinical and microbiological characteristics of pediatric posttraumatic endophthalmitis. Therefore, we conduct this study. AIM To describe clinical features, ultrasound results, gram stain and culture results of endophthalmitis in pediatric open globe injuries. METHODS Prospective non-controlled study. Case series of 30 eyes presenting with post-traumatic endophthalmitis between 2015 and 2016 were reviewed. RESULTS Mean age was 8.03 ± 3.99 years. Metallic and organic etiologies were the most common causes for injuries (n = 11). 27 cases had penetrating corneal trauma. Dense opaque vitreous was seen in 25 eyes. Retinal necrosis 1 quadrant were the most common fundus lesions. Dense vitreous opacity on ultrasound was most common (n = 28). Gram stain bacteria positivity was 93.3%, gram-positive was isolated in 63.3%. Vitreous samples were more often positive than aqueous (P = 0,002). CONCLUSION Posttraumatic endophthalmitis in children is more common in boys aged 6-10 years and most often caused by injury with metallic and organic matters. Culture results were very low. Vitreous samples were more often positive than aqueous. Gram-positive bacteria were the most common causative organism. Cataract surgery after PK has advantages that parameters of the eyes such as axial length, anterior chamber depth (ACD) as well as corneal curvature are stabilized after removing all sutures postoperatively, and intraocular lens (IOL) power can be calculated correctly. Therefore, postoperative VA will be improved significantly. In Vietnam, there have not been any study about cataract surgery after PK, therefore we conduct this research. AIM To evaluate the outcomes of phacoemulsification cataract surgery following primary PK. METHODS Non-randomized controlled intervention study. Ninteen eyes (19 patients) that underwent phacoemulsification plus IOL insertion after initial PK in Cornea department, Vietnam National Institute of Ophthalmology, from December 2013 to September 2014. RESULTS All patients presented with reduced VA, including 17 eyes (89.9%) with VA ≤ 20/200, mean astigmatism was 7.9 ± 1.0 D. Clear corneal grafts in 16 eyes while corneal opacity was seen in 3 eyes. All eyes with cataract were diagnosed from grade 2. After cataract surgery, improved VA > 20/200 was achieved in 72.22% of cases. There was a markable reduce of postoperative astigmatism with 1.8 ± 0.8 D (p less then 0.05). However, the immunologic graft reaction was presented in one eye, and two edematous corneas also reported after cataract surgery. After treatment, there was one cornea achieved its clarity. CONCLUSION Phacoemulsification cataract surgery following initial PK showed good outcomes with improved postoperative VA, reduced astigmatism, and the ultimate graft survival rate was high. Therefore, the treatment strategy needs to be plan very carefully in such case. CASE PRESENTATION We report a patient with 2 wide-neck aneurysms located in the feeding artery of the arteriovenous malformation and he, then, was treated by using Surpass stent (flow-diverter) to eliminate the aneurysms. CONCLUSION In our case, multiple wide-neck aneurysms in combination with low risk unruptured AVM, using long Surpass stent-diversion is an effective way to eliminate all the aneurysms in the main feeding artery while still preserving the AVM anatomy. Anterior chamber foreign bodies account for a considerable portion of all cases of all intraocular foreign bodies (up to 15%); however, they can be missed due to inconspicuous location. CASE REPORT We report two cases of retained intraocular foreign bodies in the iridocorneal angle that was missed at the first ophthalmic examination. They were only discovered when complications occurred, such as corneal edema and increased intraocular pressure. In the case whereby the foreign body was taken out early, corneal damages were reversible. However, in the case whereby the foreign body was taken out late, endothelial damage was irreversible and endothelial transplantation was needed. CONCLUSION Regarding trauma patients, a careful examination should be performed to discover foreign bodies in the iridocorneal angle. In Viet Nam, there was no report on the clinical and microbiological characteristics of pediatric posttraumatic endophthalmitis. Therefore, we conduct this study. AIM To describe clinical features, ultrasound results, gram stain and culture results of endophthalmitis in pediatric open globe injuries. METHODS Prospective non-controlled study. Case series of 30 eyes presenting with post-traumatic endophthalmitis between 2015 and 2016 were reviewed. RESULTS Mean age was 8.03 ± 3.99 years. Metallic and organic etiologies were the most common causes for injuries (n = 11). 27 cases had penetrating corneal trauma. Dense opaque vitreous was seen in 25 eyes. Retinal necrosis 1 quadrant were the most common fundus lesions. Dense vitreous opacity on ultrasound was most common (n = 28). Gram stain bacteria positivity was 93.3%, gram-positive was isolated in 63.3%. Vitreous samples were more often positive than aqueous (P = 0,002). CONCLUSION Posttraumatic endophthalmitis in children is more common in boys aged 6-10 years and most often caused by injury with metallic and organic matters. Culture results were very low. Vitreous samples were more often positive than aqueous. Gram-positive bacteria were the most common causative organism. Cataract surgery after PK has advantages that parameters of the eyes such as axial length, anterior chamber depth (ACD) as well as corneal curvature are stabilized after removing all sutures postoperatively, and intraocular lens (IOL) power can be calculated correctly. Therefore, postoperative VA will be improved significantly. In Vietnam, there have not been any study about cataract surgery after PK, therefore we conduct this research. AIM To evaluate the outcomes of phacoemulsification cataract surgery following primary PK. METHODS Non-randomized controlled intervention study. Ninteen eyes (19 patients) that underwent phacoemulsification plus IOL insertion after initial PK in Cornea department, Vietnam National Institute of Ophthalmology, from December 2013 to September 2014. RESULTS All patients presented with reduced VA, including 17 eyes (89.9%) with VA ≤ 20/200, mean astigmatism was 7.9 ± 1.0 D. Clear corneal grafts in 16 eyes while corneal opacity was seen in 3 eyes. All eyes with cataract were diagnosed from grade 2. After cataract surgery, improved VA > 20/200 was achieved in 72.22% of cases. There was a markable reduce of postoperative astigmatism with 1.8 ± 0.8 D (p less then 0.05). However, the immunologic graft reaction was presented in one eye, and two edematous corneas also reported after cataract surgery. After treatment, there was one cornea achieved its clarity. CONCLUSION Phacoemulsification cataract surgery following initial PK showed good outcomes with improved postoperative VA, reduced astigmatism, and the ultimate graft survival rate was high.
