The assessment of exergy efficiencies for each case was based on the transient systems simulation program TRNSYS, which was used for the simulation of energy use for space heating and cooling of the building, sanitary hot water production, and the thermal response of the seasonal heat storage and solar thermal system. The results show that an enormous waste of exergy is observed by the system based on an NG boiler (with annual overall exergy efficiency of 0.11) in comparison to the most efficient systems, based on HP water-water with a seasonal heat storage and solar thermal collector with the efficiency of 0.47. The same system with an added GW unit exhibits lower water temperatures, resulting in the exergy efficiency of 0.43. The other three systems, based on air-, water-, and ground-water HPs, show significantly lower annual source water temperatures (10.9, 11.0, 11.0, respectively) compared to systems with SHS and SHS + GW, with temperatures of 28.8 and 19.3 K, respectively.Redesigning and retrofitting cities so they become complex systems that create ecological and cultural-societal health through the provision of ecosystem services is of critical importance. Although a handful of methodologies and frameworks for considering how to design urban environments so that they provide ecosystem services have been proposed, their use is not widespread. A key barrier to their development has been identified as a lack of ecological knowledge about relationships between ecosystem services, which is then translated into the field of spatial design. In response, this paper examines recently published data concerning synergetic and conflicting relationships between ecosystem services from the field of ecology and then synthesises, translates, and illustrates this information for an architectural and urban design context. The intention of the diagrams created in this research is to enable designers and policy makers to make better decisions about how to effectively increase the provision of various ecosystem services in urban areas without causing unanticipated degradation in others. The results indicate that although targets of ecosystem services can be both spatially and metrically quantifiable while working across different scales, their effectiveness can be increased if relationships between them are considered during design phases of project development.Purpose This study was conducted to develop the 'College-based Lifestyle Modification Program' (College-based LMP) for young adult women with irregular menstruation and examine its effects after intervention. The primary outcome variables consisted of menstrual cycle index (MCI), sex hormone binding globulin (SHBG), and androgenic profile (testosterone-T, free androgen index-FAI), while the outcome variables included premenstrual symptoms (PMS), menstrual volume, body composition parameters, glycemic parameters (fasting blood sugar-FBS, insulin, HOMA-IR), sleep duration, perceived stress, and nutrient intake.. Results There were no significant differences in primary outcome variables (MCI, SHBG, T, and FAI). In the variables, there were no significant differences except for the partial domain of PMS (symptoms of depression and anxiety) and sleep duration. Conclusions The study was significant in that it demonstrated the importance of lifestyle, which could provide ordinary young adult women with healthy menstruation. The College-based LMP needs to be elaborated with further studies.
We aimed to provide a systematic review and meta-analysis of randomized controlled trials assessing the effect of oral vitamin supplementation on symptoms and disease activity in patients with rheumatoid arthritis (RA), spondyloarthritis (SpA) and psoriatic arthritis (PsA).
A systematic literature review and meta-analysis of randomized controlled trials including patients with inflammatory rheumatic diseases were performed using MEDLINE, EMBASE and abstracts from recent international rheumatology congresses. Studies were reviewed in accordance with PRISMA guidelines. We analysed clinical outcomes according to each type of vitamin supplementation.
The initial search yielded 606 articles. Of these, 13 studies were included in the qualitative synthesis eight studied vitamin D supplementation, two assessed vitamin E supplementation, two folic acid, and one vitamin K, all of them on RA patients. No studies on SpA or PsA were selected. Oral vitamin supplementations were not associated with a reduction in RA activity (DAS-28 or pain) or RA flares.

The Oswestry Disability Index (ODI) is one of the most common patient-reported outcome measures used for low **** pain (LBP). Since it was not available in Indonesian, this study aimed to perform a cross-cultural adaptation of ODI to Indonesian and evaluate its psychometric properties.
We performed a cross-cultural adaptation of ODI v2.1a into Indonesian language (ODI-ID) and determined its internal consistency, test-retest reliability, measurement error, confirmatory factor analysis, floor-ceiling effect, and construct validity by hypotheses testing of its correlation with Short Form-36 (SF-36) and visual analog scale (VAS). Adults (> 17years of age) diagnosed with low **** pain were included.

A subsample of 197 ewes was scored for body weight (BW), rump length (RL), rump width (RW), height at withers (HW), chest girth (CG), chest width (CW), chest depth (CD), cannon bone perimeter (CP), and body length (BL). Pearson's correlation analysis was performed on a sub-data set from 197 ewes to estimate the relationship between body weight and body measurements. Multiple linear regressions were fitted to obtain prediction equations of body weight from the eight body measurements, and prediction equations were obtained from the body measurement that showed the highest correlation with body weight using five nonlinear models allometric, saturation growth, exponential, and incomplete gamma. Data from an independent subsample of 196 ewes was used to validate the equation with the best goodness of fit using linear regression analysis. CG was the body measurement that showed the highest correlation with BW, and based on multiple stepwise regression, in the equation BW = - 60.622 + 1.233CG explained 79% of the body weight variation. Moreover, BW prediction was more accurate when other measurements such CW, BL, and RW were added to the model generating to the equation BW = - 68.875 + 0.845CG + 0.866CW + 0.195BL + 0.601RW (R2 = 0.85, MSE = 15.51). The results indicate that all the process responses are affected by changing the inlet air flow rate, temperature, and suspension spray rate. A design space (DS) in terms of failure probability was determined based on specifications for tablet moisture content ( less then 3.5%) and coating uniformity (tablet weight standard deviation less then 4 mg for tablet weight of 200 mg) using Monte Carlo simulations. Independent experiments were carried out and successfully validated the robustness and accuracy of the determined DS for the investigated tablet film coating process. All the data were generated using an industrial pilot-scale novel high-speed tablet coating unit from a continuous manufacturing line. The work facilitates the quality by design implementation of continuous pharmaceutical manufacturing.

To understand the molecular mechanisms underlying the triggering of abnormal liver metabolism by starvation, thus inducing hepatic lipid accumulation, we used zebrafish larvae to establish a starvation-induced hepatic steatosis model and conducted comparative transcriptome analysis by RNA-seq. We demonstrated that the incidence of larvae steatosis is positively correlated with starvation time. Under starvation conditions, the fatty acid transporter (slc27a2a and slc27a6-like) and fatty acid translocase (cd36) were up-regulated significantly to promote extrahepatic fatty acid uptake. Meanwhile, starvation inhibits the hepatic fatty acid metabolism pathway but activates the de novo lipogenesis pathway to a certain extent. More importantly, we detected that the expression of numerous apolipoprotein genes was downregulated and the secretion of very low density lipoprotein (VLDL) was inhibited significantly. These data suggest that starvation induces hepatic steatosis by promoting extrahepatic fatty acid uptake and lipogenesis, and inhibits hepatic fatty acid metabolism and lipid transport. Furthermore, we found that starvation-induced hepatic steatosis in zebrafish larvae can be rescued by targeting the knockout cd36 gene. In this paper, we investigate the experimental conditions to delaminate and recovery silicon in the recycling process, using a combination of mechanical, thermal, and chemical methods. The conditions of thermal treatment to remove the ethylene-vinyl acetate (EVA) layer were optimized to 30 min at 650 °C in the furnace. To separate silicon and metals, the composition of HF/HNO3 solution and the immersion time were adjusted considering environmental aspects and cost. The native species applicable for Cd phytoremediation were Ageratina glechonophylla, Bejaria sp., whereas Pernettya prostrata Achyrocline alata,Ageratina fastigiate, Baccharis alnifolia, Calceolaria tetragona, Arenaria digyna, Hypericum laricifolium, Brachyotum radula, and Nicotiana thyrsiflora were suitable for both Cd and Zn. None of the studied plants appeared to be suitable for phytoremediation of Pb, Cu, As and Ag.
Cats with neurologic feline infectious peritonitis (FIP) are difficult to diagnose. Aim of this study was to evaluate the diagnostic value of detecting feline coronavirus (FCoV) RNA and spike (S) gene mutations in cerebrospinal fluid (CSF).
The study included 30 cats with confirmed FIP (six with neurological signs) and 29 control cats (eleven with neurological signs) with other diseases resulting in similar clinical signs. CSF was tested for FCoV RNA by 7b-RT-qPCR in all cats. In RT-qPCR-positive cases, S-RT-qPCR was additionally performed to identify spike gene mutations.
Nine cats with FIP (9/30, 30%), but none of the control cats were positive for FCoV RNA in CSF. Sensitivity of 7b-RT-qPCR in CSF was higher for cats with neurological FIP (83.3%; 95% confidence interval (95% CI) 41.8-98.9) than for cats with non-neurological FIP (16.7%; 95% CI 6.1-36.5). Spike gene mutations were rarely detected.

To understand the molecular mechanisms underlying the triggering of abnormal liver metabolism by starvation, thus inducing hepatic lipid accumulation, we used zebrafish larvae to establish a starvation-induced hepatic steatosis model and conducted comparative transcriptome analysis by RNA-seq. We demonstrated that the incidence of larvae steatosis is positively correlated with starvation time. Under starvation conditions, the fatty acid transporter (slc27a2a and slc27a6-like) and fatty acid translocase (cd36) were up-regulated significantly to promote extrahepatic fatty acid uptake. Meanwhile, starvation inhibits the hepatic fatty acid metabolism pathway but activates the de novo lipogenesis pathway to a certain extent. More importantly, we detected that the expression of numerous apolipoprotein genes was downregulated and the secretion of very low density lipoprotein (VLDL) was inhibited significantly. These data suggest that starvation induces hepatic steatosis by promoting extrahepatic fatty acid uptake and lipogenesis, and inhibits hepatic fatty acid metabolism and lipid transport. Furthermore, we found that starvation-induced hepatic steatosis in zebrafish larvae can be rescued by targeting the knockout cd36 gene. It seems that the mechanical shaking method is a simple, fast, low-cost, and suitable alternative to the Soxhlet method for hydrocarbon extraction in oily sludge and petroleum sludge-contaminated soil.Available information on the bioaccumulation and trophodynamics of cyclic methylsiloxanes in aquatic food webs is insufficient for a reliable understanding of their toxicity and potential ecological harm. The concentrations of four cyclic methylsiloxanes in aquatic species collected from Lake Chaohu (China) were measured and the total concentration was in range of 2.01-36.1 ng/g dry weight. Dodecamethylcyclohexasiloxane (D6) represented 57.7% of the total measured methylsiloxane concentration. The distribution of these methylsiloxanes constitute the first tissue-specific record. The hexamethylcyclotrisiloxane (D3), octamethylcyclotetrasiloxane (D4) and decamethylcyclopentasiloxane (D5) tended to accumulate preferentially in fish muscles, while D6 was preferentially accumulated in fish liver and gills. In Lake Chaohu, significant trophic magnification of D3 (p 0.05) were observed within the food web. This study further confirmed the complexity of trophic transfer of the methylsiloxanes in the aquatic food web. The findings suggest that tissue-specific methylsiloxane distribution in aquatic species might also affect the observed TMFs. Additionally, Δpsntr-ANT206 was more sensitive to nitrobenzene and p-nitrophenol in the comparison of heat and hyperosmolarity, suggesting that psntr gene participated in the regulation of the tolerance against nitro-aromatic compounds (NACs). Further analysis was conducted by integrated gas chromatography-mass spectrometry (GC-MS), and several metabolites were identified. Among them, ethylbenzene, L-Alanine, citric acid, aniline, 4-aminophenol and other metabolites were different between the wild-type strain and Δpsntr-ANT206 under nitrobenzene and p-nitrophenol stress at different time periods under low temperature, respectively. Tb@TpPa-SO3H functionalized with regularly distributed sulfonic acid groups and terbium ions which could provide difunctional binding sites. Tb3+ sites could capture carboxylic acid group of FQs molecules according to the complexes coordination effect and sulfonic acid sites play a significant role in the adsorption of FQs molecules through electrostatic interaction with amine group. Tb@TpPa-SO3H with dual complementary function sites exhibited ultra-fast adsorption kinetics ( less then 2 min, average over 99% removing rate) and high adsorption capacities of 989, 956, and 998 mg g-1 for Norfloxacin (NOR), ciprofloxacin (CIP), enrofloxacin (ENR), respectively. Furthermore, Tb@TpPa-SO3H showed excellent selectivity for the adsorption of FQs in tanglesome system. Here, we investigated two treatment systems, namely ozonation (ozone) and sequential hydrodynamic cavitation and ozonation (HC+ozone), under varied conditions, to determine whether the extent of sludge disintegration can estimate a measurable removal of PAHs. The results showed that both the PAHs removal efficiency and kinetics highly depended on pH and ozone dose. Although no significant removal of PAHs occurred during HC treatment, the observed rate constants for Σ16 PAHs removal in HC+ozone system were 1.2-1.4-fold higher than those obtained in ozone system under the same condition. The accelerated PAHs removal could be attributed to the reduced flocs size (80% smaller) and increased soluble EPS concentration (5-fold higher) with HC pretreatment.

Hori's nevus is a common pigmented disorder on the face, preferentially in females. The clinical features have not been well characterized.
To characterize the clinical features of Hori's nevus in Chinese adults.
Data were collected from files of patients who visited our hospital from 2015 to 2018. https://www.selleckchem.com/products/gsk1070916.html Age- and disease duration-related characteristics were analyzed.
A total of 497 patients, including 486 females and 11 males, were included in this analysis. One fifth of the patients had a family history of Hori's nevus. Over 70% of patients were aged 21-30 years. Age at onset was comparable between males and females (20.64 ± 1.01 vs 18.99 ± 0.24). Out of 497 subjects, 218 subjects (44%) displayed yellow-brown lesions while blue-brown lesions wwere observed in 103 subjects (21%). The rest (176 cases, 35%) showed slate-grey lesions. Involvement in the zygomatic area was observed in 496/497 subjects. Involvement in a single area accounted for 74% of patients, while two areas were involved in 19% of patients. The number of involved areas correlated positively with disease duration. However, the proportion of subjects with yellow-brown lesions correlated negatively with disease duration, while the proportion of subjects with slate-grey lesions correlated positively with disease duration. The proportion of subjects with lesions involving the lower eyelids, the root of the nose, the temple and the outer frontal area correlated positively with age.

This review aims to summarize the effects of honey (based on unifloral and multi-floral nectar sources) on bone metabolism and each component of MetS. A literature search was performed using the PubMed and Scopus databases using specific search strings. Original studies related to components of MetS and bone, and the effects of honey on components of MetS and bone were included. Honey polyphenols could act synergistically in alleviating MetS by preventing oxidative damage and inflammation. Honey intake is shown to reduce blood glucose levels and prevent excessive weight gain. It also improves lipid metabolism by reducing total cholesterol, triglycerides and low-density lipoprotein, as well as increasing high-density lipoprotein. Honey can prevent bone loss by reducing the adverse effects of MetS on bone homeostasis, apart from its direct action on the skeletal system. In conclusion, honey supplementation could be integrated into the management of MetS and MetS-induced bone loss as a preventive and adjunct therapeutic agent.

Functional disruptions by large germline genomic structural variants in susceptible genes are known risks for cancer. We used deletion structural variants (DSVs) generated from germline whole-genome sequencing (WGS) and DSV immune-related association tumor microenvironment (TME) to predict cancer risk and prognosis.
We investigated the contribution of germline DSVs to cancer susceptibility and prognosis by silicon and causal inference models. DSVs in germline WGS data were generated from the blood samples of 192 cancer and 499 non-cancer subjects. Clinical information, including family cancer history (FCH), was obtained from the National Cheng Kung University Hospital and Taiwan Biobank. Ninety-nine colorectal cancer (CRC) patients had immune response gene expression data. We used joint calling tools and an attention-weighted model to build the cancer risk predictive model and identify DSVs in familial cancer. The survival support vector machine (survival-SVM) was used to select prognostic DSVs.

Copper is mostly absorbed in the upper gastrointestinal tract, particularly in the duodenum, but some Cu is absorbed in the stomach. One way to evaluate the efficacy of sources of Cu is to measure relative bioavailability where responses include tissue concentrations of Cu, concentrations of metalloproteins, and enzymatic activity of animals fed test diets containing graded levels of Cu. The requirement for Cu by pigs is 5 to 10 mg/kg diet, however, Cu can be included at growth-promoting levels (i.e., 75 to 250 mg/kg diet) in diets for weanling and growing pigs to reduce post-weaning diarrhea and improve growth performance. The consistently observed improvement in growth performance upon Cu supplementation is likely a result of increases in lipase activity, growth hormone secretion, and expression of genes involved in post-absorptive metabolism of lipids. The growth-promoting effects of dietary Cu have also been attributed to its bacteriostatic and bactericidal properties because Cu may change bacterial populations in the intestine, and thereby reduce inflammation caused by pathogens. However, further research is needed to determine potential interactions between Cu and non-nutritive feed additives (e.g., enzymes, probiotics, phytobiotics), and the optimum quantity of Cu as well as the optimum duration of feeding supplemental Cu in diets for pigs should be further investigated. These gaps needs to be addressed to maximize inclusion of Cu in diets to improve growth performance while minimizing diseases and mortality.
Non-caloric artificial sweeteners (NCAS) are widely used as a substitute for dietary sugars to control body weight or glycemia. Paradoxically, some interventional studies in humans and rodents have shown unfavorable changes in glucose homeostasis in response to NCAS consumption. To demonstrate feasibility of a music medicine intervention trial in pediatric intensive care and to obtain information on sedation and analgesia dose variation to plan a larger trial.
Pilot randomized controlled trial (RCT) was conducted at the Stollery Children's Hospital general and cardiac intensive care units (PICU/PCICU). The study included children 1 month to 16 years of age on mechanical ventilation and receiving sedation drugs. Patients were randomized in a 111 ratio to music, noise cancellation or control. The music group received classical music for 30 min three times/day using headphones. The noise cancellation group received the same intervention but with no music. The control group received usual care.
A total of 60 patients were included. Average enrollment rate was 4.8 patients/month, with a consent rate of 69%. Protocol adherence was achieved with patients receiving > 80% of the interventions. Overall mean (SD) daily Sedation Intensity Score was 52.4 (30.3) with a mean (SD) sedation frequency of 9.75 (7.21) PRN doses per day. There was a small but statistically significant decrease in heart rate at the beginning of the music intervention. There were no study related adverse events. Eighty-eight percent of the parents thought the headphones were comfortable; 73% described their child more settled during the intervention.
This pilot RCT has demonstrated the feasibility of a music medicine intervention in critically ill children. The study has also provided the necessary information to plan a larger trial.

Predicting specific abilities (e.g. walk and talk) to provide a functional profile six months after disabling stroke could help patients/families prepare for the consequences of stroke and facilitate involvement in treatment decision-making.
To develop new statistical models to predict specific abilities six months after stroke and test their performance in an independent cohort of patients with disabling stroke.
We developed models to predict six specific abilities (to be independent, walk, talk, eat normally, live without major anxiety/depression, and to live at home) using data from seven large multicenter stroke trials with multivariable logistic regression. We included 13,117 participants recruited within three days of hospital admission. We assessed model discrimination and derived optimal cut-off values using four statistical methods. We validated the models in an independent single-center cohort of patients (
= 403) with disabling stroke. We assessed model discrimination and calibration and reported the performance of our models at the statistically derived cut-off values.
All six models had good discrimination in external validation (AUC 0.78-0.84). Four models (predicting to walk, eat normally, live without major anxiety/depression, live at home) calibrated well. Models had sensitivities between 45.0 and 97.9% and specificities between 21.6 and 96.5%.
We have developed statistical models to predict specific abilities and demonstrated that these models perform reasonably well in an independent cohort of disabling stroke patients. To aid decision-making regarding treatments, further evaluation of our models is required.

The aim of this study is to analyse the effect of such an exoskeleton on shoulder muscle activity and perceived exertion, in six tasks of plasterers, each including multiple arm movements. The tasks of 'applying gypsum', 'screeding' and 'finishing' were performed at a ceiling and a wall, with exoskeleton (Exo) and without (NoExo). EMG was recorded of six muscles involved in upper arm elevation, four agonists and two antagonists, and plasterers rated their perceived exertion (RPE). In all tasks, the EMG amplitudes of three agonist muscles, Trapezius and Medial Deltoid, and Biceps Brachii, were lower in Exo vs NoExo, while the agonist, Anterior Deltoid, showed lower EMG values in Exo in most tasks. None of the antagonists (Triceps Brachii, Pectoralis Major) showed increased EMG values in the Exo condition. RPE's were lower in Exo condition for all tasks, except for 'applying gypsum to the wall'. Overall, the exoskeleton seems to reduce loads in realistic plastering tasks. These strategies are limited by many factors that lead to constitutive activation of ERα and consequently, resistance to treatment. Targeting the DNA binding domain (DBD) of ERα instead of its LBD with small-molecule inhibitors could be an alternative to impair ERα's signalling pathway. For this purpose, we conducted a structure based virtual screening of DrugBank against the crystal structure of ERα-DBD (PDB ID 1HCQ) using the Glide module in standard precision (SP) and extra precision (XP) mode of docking. Molecules with XP Gscore less than -8 kcal/mol were selected and visually inspected to keep only the reasonable docking poses. Subsequently, these molecules were clustered using structural interaction fingerprints analysis and the complexes of the top ranked molecules of each cluster based on XP Gscore were subjected to 200 ns molecular dynamics simulations followed by MM-GBSA binding free energy calculation for the last 100 ns of each complex. In this study, we identified three molecules from DrugBank namely DB03450, DB02593 and DB08001 showing significant stability and strong interaction with the key amino acids during MD simulation suggesting a potential inhibition of the target. To develop new statistical models to predict specific abilities six months after stroke and test their performance in an independent cohort of patients with disabling stroke. We developed models to predict six specific abilities (to be independent, walk, talk, eat normally, live without major anxiety/depression, and to live at home) using data from seven large multicenter stroke trials with multivariable logistic regression. We included 13,117 participants recruited within three days of hospital admission. We assessed model discrimination and derived optimal cut-off values using four statistical methods. We validated the models in an independent single-center cohort of patients ( = 403) with disabling stroke. We assessed model discrimination and calibration and reported the performance of our models at the statistically derived cut-off values. All six models had good discrimination in external validation (AUC 0.78-0.84). Four models (predicting to walk, eat normally, live without major anxiety/depression, live at home) calibrated well. Models had sensitivities between 45.0 and 97.9% and specificities between 21.6 and 96.5%. We have developed statistical models to predict specific abilities and demonstrated that these models perform reasonably well in an independent cohort of disabling stroke patients. The aim of this study is to analyse the effect of such an exoskeleton on shoulder muscle activity and perceived exertion, in six tasks of plasterers, each including multiple arm movements. The tasks of 'applying gypsum', 'screeding' and 'finishing' were performed at a ceiling and a wall, with exoskeleton (Exo) and without (NoExo). EMG was recorded of six muscles involved in upper arm elevation, four agonists and two antagonists, and plasterers rated their perceived exertion (RPE). In all tasks, the EMG amplitudes of three agonist muscles, Trapezius and Medial Deltoid, and Biceps Brachii, were lower in Exo vs NoExo, while the agonist, Anterior Deltoid, showed lower EMG values in Exo in most tasks. None of the antagonists (Triceps Brachii, Pectoralis Major) showed increased EMG values in the Exo condition. RPE's were lower in Exo condition for all tasks, except for 'applying gypsum to the wall'. Overall, the exoskeleton seems to reduce loads in realistic plastering tasks. These strategies are limited by many factors that lead to constitutive activation of ERα and consequently, resistance to treatment. Targeting the DNA binding domain (DBD) of ERα instead of its LBD with small-molecule inhibitors could be an alternative to impair ERα's signalling pathway. For this purpose, we conducted a structure based virtual screening of DrugBank against the crystal structure of ERα-DBD (PDB ID 1HCQ) using the Glide module in standard precision (SP) and extra precision (XP) mode of docking. Molecules with XP Gscore less than -8 kcal/mol were selected and visually inspected to keep only the reasonable docking poses. Subsequently, these molecules were clustered using structural interaction fingerprints analysis and the complexes of the top ranked molecules of each cluster based on XP Gscore were subjected to 200 ns molecular dynamics simulations followed by MM-GBSA binding free energy calculation for the last 100 ns of each complex. In this study, we identified three molecules from DrugBank namely DB03450, DB02593 and DB08001 showing significant stability and strong interaction with the key amino acids during MD simulation suggesting a potential inhibition of the target. In recent years, there have been major advances in the application of non-invasive techniques to predict pregnancy-related complications, for example by measuring cell-free RNA (cfRNA) in maternal blood. In contrast to cell-free DNA (cfDNA), which is already in clinical use to diagnose fetal aneuploidy, circulating RNA levels can correspond with tissue-specific gene expression and provide a snapshot of prenatal health across gestation. Here, we review the physiologic origins of cfRNA and its novel applications and corresponding challenges to monitor fetal and maternal health and predict pregnancy-related complications.Background Severe congenital neutropenia (SCN), also known as Kostmann syndrome, is a rare heterogeneous group of diseases characterized by arrested neutrophil maturation in the bone marrow. A 10-year-old boy had a history of recurrent, once a month, infection starting at 6 months of age. He had neutropenia for more than 9 years, as well as intellectual disability. He was homozygous for the exon 3 c.430dupG mutation of the HAX1 gene NM-006118. After treatment of antibiotics and G-CSF, his symtoms were relieved and was 3 months free of infection. The search revealed 29 articles related to Kostmann syndrome caused by HAX1 gene mutation; they were screened, and the main clinical features of 13 cases of Kostmann syndrome with neurological abnormalities were summarized and analyzed. Conclusions Kostmann syndrome has three main characteristics severe neutropenia ( less then 0.2 × 109/L), maturation arrest of granulopoiesis at the promyelocyte stage, and death due to infections. We focus on analyzing barriers to accessing genetic services, participating in genomic research, and other aspects such as concerns about data sharing, the handling of biospecimens, and the importance of capacity building.
The role of anticoagulants in chronic liver diseases is inconclusive. A meta-analysis was thus undertaken to evaluate treatment-related survival and antifibrotic effects in animal models of chronic liver diseases.
A systematic search of the literature took place (up to November 2020), screening for preclinical studies that evaluated anticoagulant effects in animal models of chronic liver diseases. We assessed the quality of methods and the certainty of evidence. Data on outcomes were extracted and pooled into random-effects models.
Sixteen studies proved eligible, each assessing anticoagulant use in animals with chronic liver diseases. Generally, the pooled evidence demonstrated that the administration of anticoagulants is preventive against fibrogenesis, as indicated by METAVIR fibrosis scores (risk ratio = 0.66, 95% confidence interval 0.47 to 0.94), portal pressure determinations (mean difference = -1.39, 95% confidence interval -2.33 to -0.44), inflammatory activity (mean difference = -169.69, 95% ce system, portal pressure, inflammatory activity, and serum indices of hepatocellular injury, without impacting survival. High-quality experimental studies are still required.
This study aimed to evaluate the risk factors of HCC development in patients with hepatitis B virus (HBV)-related DC and who underwent long-term antiviral therapy.
Data from 308 patients with HBV-related DC and long-term antiviral therapy were collected and retrospectively reviewed. Cox regression analysis was used to analyze independent risk factors of HCC development.
Data from 129 patients with definite records were analyzed. The median follow-up time was 5 years (range, 1 to 8 years). At the end of the follow-up, 41 (31.8%) patients developed HCC, and the time from DC diagnosis to HCC incidence who received antiviral therapy was 4.4 years (range, 1-7 years). The incidence of HCC was higher in males (30/78, 38.5%) than in females (11/51, 21.6%) (
= 0.04). Patients who developed HCC were significantly older than those who did not develop HCC (
< 0.01). The incidence of HCC in patients receiving nucleoside analogues, nucleotide analogues, and combination therapy was 34.7%, 38.1%, and 33.3%, respectively, and the difference showed no significant differences (

We aimed to provide a critical review and updates on the prevention and treatment of alcohol-related liver disease through regulating intestinal microbiota. A literature search was performed on the PubMed database for studies published in English about the therapeutic intervention with microbiota using animal models and patients with alcohol-related liver disease (1/2010-4/2020). The accumulating pieces of evidence suggest that the therapeutic use of probiotics, prebiotics, antibiotics, phages, or fecal microbial transplantation may have several influences on alcohol-related liver disease patients.

Therefore, we conducted this systematic review and meta-analysis to examine the burden of diabetes among TB patients and assess its impact on TB treatment in South Asia (Afghanistan, Bangladesh, Bhutan, Maldives, Nepal, India, Pakistan, and Sri Lanka). PubMed, Excerpta Medica Database (EMBASE), and CINAHL databases were systematically searched for observational (cross-sectional, case-control and cohort) studies that reported prevalence of diabetes in TB patients and published between 1 January 1980 and 30 July 2020. A random-effect model for computing the pooled prevalence of diabetes and a fixed-effect model for assessing its impact on TB treatment were used. The review was registered with PROSPERO number CRD42020167896. Of the 3463 identified studies, a total of 74 studies (47 studies from India, 10 from Pakistan, four from Nepal and two from both Bangladesh and Sri-uth Asia. Patients with TB-diabetes were at higher risk of treatment failure and mortality compared to TB alone. In this work, we applied both the quantum mechanical and classical atomistic molecular dynamics approaches to demonstrate the adsorption properties of HCQ/CQ on Ag, Au, AgAu, and Pt nanoparticles. We found the adsorption energies of HCQ/CQ towards nanoparticles have the following trend PtNP > AuNP > AuAgNP > AgNP. This shows that PtNP has the highest affinity in comparison to the other types of nanoparticles. However, it was poorly documented prior to its extinction ~ 1800 AD, and many of the small number of museum specimens attributed to it are taxonomically contentious. This places limitations on our understanding of its morphology, ecology, and the mechanisms responsible for its demise. We retrieved genetic information from ten of the sixteen putative blue antelope museum specimens using both shotgun sequencing and mitochondrial genome target capture in an attempt to resolve the uncertainty surrounding the identification of these specimens. We found that only four of the ten investigated specimens, and not a single skull, represent the blue antelope. This indicates that the true number of historical museum specimens of the blue antelope is even smaller than previously thought, and therefore hardly any reference material is available for morphometric, comparative and genetic studies.

Overall, "The Number Race" appears as a suitable tool to improve some aspects of numeracy in DS.We report a method to generate a 3D motor neuron model with segregated and directed axonal outgrowth. iPSC-derived motor neurons are cultured in extracellular matrix gel in a microfluidic platform. Neurons extend their axons into an adjacent layer of gel, whereas dendrites and soma remain predominantly in the somal compartment, as verified by immunofluorescent staining. Axonal outgrowth could be precisely quantified and was shown to respond to the chemotherapeutic drug vincristine in a highly reproducible dose-dependent manner. The model was shown susceptible to excitotoxicity upon exposure with excess glutamate and showed formation of stress granules upon excess glutamate or sodium arsenite exposure, mimicking processes common in motor neuron diseases. Importantly, outgrowing axons could be attracted and repelled through a gradient of axonal guidance cues, such as semaphorins. The platform comprises 40 chips arranged underneath a microtiter plate providing both throughput and compatibility to standard laboratory equipment. The model will thus prove ideal for studying axonal biology and disease, drug discovery and regenerative medicine.From the first month of the COVID-19 pandemic, the potential antiviral properties of hydroxychloroquine (HCQ) and chloroquine (CQ) against SARS-CoV-2 suggested that these drugs could be the appropriate therapeutic candidates. However, their side effects directed clinical tests towards optimizing safe utilization strategies. The noble metal nanoparticles (NP) are promising materials with antiviral and antibacterial properties that can deliver the drug to the target agent, thereby reducing the side effects. In this work, we applied both the quantum mechanical and classical atomistic molecular dynamics approaches to demonstrate the adsorption properties of HCQ/CQ on Ag, Au, AgAu, and Pt nanoparticles. We found the adsorption energies of HCQ/CQ towards nanoparticles have the following trend PtNP > AuNP > AuAgNP > AgNP. This shows that PtNP has the highest affinity in comparison to the other types of nanoparticles. The (non)perturbative effects of this drug on the plasmonic absorption spectra of AgNP and AuNP with the time-dependent density functional theory. The effect of size and composition of NPs on the coating with HCQ and CQ were obtained to propose the appropriate candidate for drug delivery. This kind of modeling could help experimental groups to find efficient and safe therapies.Native to southern Africa, the blue antelope (Hippotragus leucophaeus) is the only large African mammal species known to have become extinct in historical times. However, it was poorly documented prior to its extinction ~ 1800 AD, and many of the small number of museum specimens attributed to it are taxonomically contentious. This places limitations on our understanding of its morphology, ecology, and the mechanisms responsible for its demise. We retrieved genetic information from ten of the sixteen putative blue antelope museum specimens using both shotgun sequencing and mitochondrial genome target capture in an attempt to resolve the uncertainty surrounding the identification of these specimens. We found that only four of the ten investigated specimens, and not a single skull, represent the blue antelope.
