The generation of bio-targetable photosensitizers is of utmost importance to the emerging field of photodynamic therapy and antimicrobial (photo-)therapy. A synthetic strategy is presented in which chelating dipyrrin moieties are used to enhance the known photoactivity of iridium(III) metal complexes. Formed complexes can thus be functionalized in a facile manner with a range of targeting groups at their chemically active reaction sites. Dipyrrins with N- and O-substituents afforded (dipy)iridium(III) complexes via complexation with the respective Cp*-iridium(III) and ppy-iridium(III) precursors (dipy=dipyrrinato, Cp*=pentamethyl-η5 -cyclopentadienyl, ppy=2-phenylpyridyl). Similarly, electron-deficient [IrIII (dipy)(ppy)2 ] complexes could be used for post-functionalization, forming alkenyl, alkynyl and glyco-appended iridium(III) complexes. The phototoxic activity of these complexes has been assessed in cellular and bacterial assays with and without light; the [IrIII (Cl)(Cp*)(dipy)] complexes and the glyco-substituted iridium(III) complexes showing particular promise as photomedicine candidates. We evaluated the performance of two commercially available lateral flow immunochromatographic assays (LFIC; Wondfo SARS-CoV-2 Antibody test and the INNOVITA 2019-nCoV Ab test) in comparison with a SARS-CoV-2 neutralization pseudotyped assay for coronavirus disease 2019 (COVID-19) diagnosis in hospitalized patients and investigate whether the intensity of the test band in LFIC associates with neutralizing antibody (NtAb) titers. Ninety sera were included from 51 patients with moderate to severe COVID-19. A green fluorescent protein (GFP) reporter-based pseudotyped neutralization assay (vesicular stomatitis virus coated with SARS-CoV-2 spike protein) was used. Test line intensity was scored using a 4-level scale (0 to 3+). The overall sensitivity of LFIC assays was 91.1% for the Wondfo SARS-CoV-2 Antibody test, 72.2% for the INNOVITA 2019-nCoV IgG, 85.6% for the INNOVITA 2019-nCoV IgM, and 92.2% for the NtAb assay. Sensitivity increased for all assays in sera collected beyond day 14 after symptoms onset (93.9%, 79.6%, 93.9%, and 93.9%, respectively). Reactivities equal to or more intense than the positive control line (≥2+) in the Wondfo assay had a negative predictive value of 100% and a positive predictive value of 96.4% for high NtAb50 titers (≥1/160). Our findings support the use of LFIC assays evaluated herein, particularly the Wondfo test, for COVID-19 diagnosis. We also find evidence that these rapid immunoassays can be used to predict high SARS-CoV-2-S NtAb50 titers.Green chemistry, which aims at the development of efficient methods for the synthesis of nanoparticles, is a relatively new emerging field of nanotechnology, which has economic and environment-friendly benefits over chemical and physical processes. The present work was carried out to develop silver nanoparticles (Ag-NPs) using the plant (Achillea millefolium or yarrow) aqueous extract as both a reducing and capping agent under the green synthesis method. Characterization of synthesized Ag-NPs was done using IR spectroscopy, scanning electron microscopy (SEM), X-ray diffraction (XRD), and ultraviolet-visible (UV-vis). The UV-vis spectrum showed the maximum absorbance at around 440-470 nm, which suggested the formation of green synthesized Ag-NPs. The morphological study demonstrated that the Ag-NPs were spherical in shape with an average size of 22.4 ± 7.4 nm. The antimicrobial activities of Ag-NPs against Fusarium and Aspergillus niger species of fungal and Escherichia coli species of bacteria were investigated through the disc diffusion and well-diffusion method using their zone of inhibition. The cytotoxicity effect of Ag-NPs on cell lines MOLT-4 was evaluated by using MTT assay. These nanoparticles showed remarkable antimicrobial activity against bacterias and fungus in low concentration. The cytotoxicity studies showed that IC50 of green synthesized Ag-NPs was 0.011 µm in comparison to 1.8 for Cisplatin which more active than anticancer drug for MOLT-4 cell line. The results showed that the green synthesized Ag-NPs are expected to have notable applications and can be potentially useful in pharmaceutical and biomedical applications.Heme-nitric oxide/oxygen binding (H-NOX) domains bind gaseous ligands for signal transduction in organisms spanning prokaryotic and eukaryotic kingdoms. In higher animals, soluble guanylyl cyclase (sGC) binds nitric oxide with an H-NOX domain to induce cyclase activity and regulate vascular tone, wound healing and memory formation. sGC also binds stimulator compounds targeting cardiovascular disease. The molecular details of stimulator binding to sGC remain obscure but involve a binding pocket near an interface between H-NOX and coiled-coil domains. Here, we report the full NMR structure for CO-ligated Sw H-NOX in the presence and absence of stimulator compound IWP-051, and its backbone dynamics. Nonplanar heme geometry was retained using a semi-empirical quantum potential energy approach. Although IWP-051 binding is weak, a single binding conformation was found at the interface of the two H-NOX subdomains, near but not overlapping with sites identified in sGC.
