Over the years, different therapeutic modalities have been adopted depending on tumor stage and patient characteristics, such as surgery, radiotherapy (RT), and chemotherapy. Recently, with the development of immune-checkpoint inhibitors (ICI), the treatment of metastatic and locally advanced non-small cell lung cancer (NSCLC) has experienced a revolution that has resulted in a significant improvement in overall survival with an enhanced toxicity profile. Despite this paradigm shift, most patients present some kind of resistance to ICI. In this setting, current research is shifting towards the integration of multiple therapies, with RT and ICI being one of the most promising based on the potential immunostimulatory synergy of this combination. This review gives an overview of the evolution and current state of the combination of RT and ICI and provides evidence-based data that can improve patient selection. The combination in lung cancer is a safe therapeutic approach that improves local control and progression-free survival, and it has the potential to unleash abscopal responses. Additionally, this treatment strategy seems to be able to re-sensitize select patients that have reached a state of resistance to ICI, further enabling the continuation of systemic therapy.The coupling of alternative splicing with the nonsense-mediated decay (NMD) pathway maintains quality control of the transcriptome in eukaryotes by eliminating transcripts with premature termination codons (PTC) and fine-tunes gene expression. Long noncoding RNA (lncRNA) can regulate multiple cellular processes, including alternative splicing. Previously, murine Morrbid (myeloid RNA repressor of Bcl2l11 induced death) lncRNA was described as a locus-specific controller of the lifespan of short-living myeloid cells via transcription regulation of the apoptosis-related Bcl2l11 protein. Here, we report that murine Morrbid lncRNA in hepatocytes participates in the regulation of proto-oncogene NRAS (neuroblastoma RAS viral oncogene homolog) splicing, including the formation of the isoform with PTC. We observed a significant increase of the NRAS isoform with PTC in hepatocytes with depleted Morrbid lncRNA. We demonstrated that the NRAS isoform with PTC is degraded via the NMD pathway. This transcript is presented almost only in the nucleus and has a half-life ~four times lower than other NRAS transcripts. Additionally, in UPF1 knockdown hepatocytes (the key NMD factor), we observed a significant increase of the NRAS isoform with PTC. By a modified capture hybridization (CHART) analysis of the protein targets, we uncovered interactions of Morrbid lncRNA with the SFPQ (splicing factor proline and glutamine rich)-NONO (non-POU domain-containing octamer-binding protein) splicing complex. Finally, we propose the regulation mechanism of NRAS splicing in murine hepatocytes by alternative splicing coupled with the NMD pathway with the input of Morrbid lncRNA.Ultrasonic testing using arrays is becoming widely used to test composite structures in the Aerospace industry. In recent years, the Full Matrix Capture (FMC) technique has been implemented to extract the signals for post-processing to form an image. The inherent anisotropy and the layering of the structure pose challenges for the interpretation of this FMC data. To overcome this challenge, modeling techniques are required that take into account the diffraction caused by finite-size transducers and the response of the structure to these bounded beams. Existing models either homogenize the entire structure, use computationally expensive finite difference time domain (FDTD) methods, or do not consider the shape of the bounded beam, which is used to test such structures. This paper proposes a modeling technique based on combining the Multi-Gaussian beam model with the recursive stiffness matrix method to simulate the FMC signals for layered anisotropic media. The paper provides the steps required for the modeling technique, the extraction of the system efficiency factor, and validation of the model with experimentally determined signals for aluminum as an isotropic material such as aluminum and Carbon Fiber Reinforced Plastic (CFRP) laminate as a layered material. The proposed method is computationally inexpensive, shows good agreement with the experimentally determined FMC data, and enables us to understand the effects of various transducer and material parameters on the extracted FMC signals.
