We set out to investigate a rare form of keratinopathic ichthyosis caused by KRT1 mutation with two different optical imaging methods. We used a newly developed light emitting diode (LED) based device to analyze autofluorescence signal at 405 nm excitation and diffuse reflectance at 526 nm in vivo. Mean autofluorescence intensity of the hyperkeratotic palmar skin was markedly higher in comparison to the healthy control (162.35 vs. 51.14). To further assess the skin status, we examined samples from affected skin areas ex vivo by nonlinear optical microscopy. Two-photon excited fluorescence and second-harmonic generation can visualize epidermal keratin and dermal collagen, respectively. We were able to visualize the structure of the epidermis and other skin changes caused by abnormal keratin formation. Taken together, we were able to show that such imaging modalities are useful for the diagnosis and follow-up of keratinopathic diseases.Mastitis is a common disease in cows breeding. The milk quality will be significantly reduced with increased milk somatic cells, which often occurs in cows with mastitis. In this study, the influence of seasonal changes, age and lactation stages in the Dairy Herd Improvement (DHI) of cows was investigated. Then, the Dairy Herd Improvement (DHI) of cows with high somatic cell score (SCS) after melatonin treatment was systemically investigated. The results showed that melatonin significantly suppressed the milk somatic cell score under all of the tested conditions. The melatonin treatment also improved the milk nutritional value by reducing its fat but increasing its lactose and protein contents. The application of melatonin significantly improved the DHI. The beneficial effects of melatonin on DHI are likely attributed to the antioxidant and anti-inflammatory activities of melatonin.Egocentric activity recognition in first-person video (FPV) requires fine-grained matching of the camera wearer's action and the objects being operated. The traditional method used for third-person action recognition does not suffice because of (1) the background ego-noise introduced by the unstructured movement of the wearable devices caused by body movement; (2) the small-sized and fine-grained objects with single scale in FPV. Size compensation is performed to augment the data. It generates a multi-scale set of regions, including multi-size objects, leading to superior performance. We compensate for the optical flow to eliminate the camera noise in motion. We developed a novel two-stream convolutional neural network-recurrent attention neural network (CNN-RAN) architecture spatial temporal attention on compensation information (STAC), able to generate generic descriptors under weak supervision and focus on the locations of activated objects and the capture of effective motion. We encode the RGB features using a spatial location-aware attention mechanism to guide the representation of visual features. Similar location-aware channel attention is applied to the temporal stream in the form of stacked optical flow to implicitly select the relevant frames and pay attention to where the action occurs. The two streams are complementary since one is object-centric and the other focuses on the motion. We conducted extensive ablation analysis to validate the complementarity and effectiveness of our STAC model qualitatively and quantitatively. It achieved state-of-the-art performance on two egocentric datasets.Brassinin is a phytochemical derived from Chinese cabbage, a cruciferous vegetable. Brassinin has shown anticancer effects on prostate and colon cancer cells, among others. However, its mechanisms and effects on hepatocellular carcinoma (HCC) have not been elucidated yet. Our results confirmed that brassinin exerted antiproliferative effects by reducing proliferating cell nuclear antigen (PCNA) activity, a proliferation indicator and inducing cell cycle arrest in human HCC (Huh7 and Hep3B) cells. Brassinin also increased mitochondrial Ca2+ levels and depolarized the mitochondrial membrane in both Huh7 and Hep3B cells. Moreover, brassinin generated high amounts of reactive oxygen species (ROS) in both cell lines. The ROS scavenger N-acetyl-L-cysteine (NAC) inhibited this brassinin-induced ROS production. Brassinin also regulated the AKT and mitogen-activated protein kinases (MAPK) signaling pathways in Huh7 and Hep3B cells. Furthermore, co-administering brassinin and pharmacological inhibitors for JNK, ERK1/2 and P38 decreased cell proliferation in both HCC cell lines more than the pharmacological inhibitors alone.
